The influence of NaCl on senescence-related parameters (proteinand chlorophyll concentrations, membrane permeability and chlorophyllfluorescence) was investigated in young and old leaves of fiverice cultivars differing in salt resistance. NaCl hastened thenaturally-occurring senescence of rice leaves which normallyappears during leaf ontogeny: it decreased chlorophyll and proteinconcentrations and increased membrane permeability and malondialdehydesynthesis. Such an acceleration of deteriorative processes affectedall leaves in salt-sensitive cultivars while it was more markedin oldest than in youngest leaves of salt-resistant genotypes.NaCl-induced senescence also involved specific modifications,such as an increase in basal non-variable chlorophyll fluorescence(F 0) recorded in all cultivars or a transient increase in solubleprotein concentration recorded in salt-resistant genotypes only.Alteration of membrane permeability appeared as one of the firstsymptoms of senescence in rice leaves and allowed discriminationamong cultivars after only 7 d of stress. In contrast, F v/F mratio (variable fluorescence/maximal fluorescence) was thesame for all cultivars during the first 18 d of stress and thuscould not be used for identifying salt-resistant rice exposedto normal light conditions. Relationships between parametersinvolved in leaf senescence are discussed in relation to salinityresistance of rice cultivars. The regulation of senescence by oxygen-concentration, lightirradiance and H2O2 has been studied in leaf segments of Avenasativa L. cv. Suregrain. The development of the components of the senescence process,for example chlorophyll breakdown, proteolysis (as soluble aminoacids), hydroperoxides (as malondi-aldehyde) and permeability(as conductivity) is accelerated in light as the O2-tensionincreases. In darkness, 0.3% O2 accelerates increases in hydroperoxides,permeability and proteolysis and delays the chlorophyll break-down,but 0.0005% O2 delays all the components studied. In every casethe hydroperoxide content, permeability and proteolysis areclosely related. Any treatment inducing an increase in membranepermeability causes chlorophyll bleaching (photo-oxidation)if leaf segments are then treated with light in an atmospherecontaining oxygen. Light has a modulating effect on the senescenceprocess. An irradiance lower or higher than 40 W m–2 hasan accelerating effect on the senescence process. Net photosynthesis rate (Pn), stomatal conductance to CO2 andresidual conductance to CO2 were measured in the last six leaves(the sixth or flag leaf and the preceding five leaves) of Triticumaestivum L. cv. Kolibri plants grown in Mediterranean conditions.Recently fully expanded leaves of well-watered plants were alwaysused. Measurements were made at saturating photosynthetic photonflux density, and at ambient CO2 and O2 levels. The specificleaf area, total organic nitrogen content, some anatomical characteristics,and other parameters, were measured on the same leaves usedfor gas exchange experiments. A progressive xeromorphic adaptation in the leaf structure wasobserved with increasing leaf insertion levels. Furthermore,mesophyll cell volume per unit leaf area (Vmes/A) decreasedby 52·6% from the first leaf to the flag leaf. Mesophyllcell area per unit leaf area also decreased, but only by 24·5%.However, nitrogen content per unit mesophyll cell volume increasedby 50·6% from the first leaf to the flag leaf. This increasecould be associated to an observed higher number of chloroplastcross-sections per mm2 of mesophyll cell cross-sectional areain the flag leaf: values of 23000 in the first leaf and 48000in the flag leaf were obtained. Pn per unit leaf area remainedfairly constant at the different insertion levels: values of33·83±0·93 mg dm–2 h–1 and32·32±1·61 mg dm–2 h–1 wereobtained for the first leaf and the flag leaf, respectively.Residual conductance, however, decreased by 18·2% fromthe first leaf to the flag leaf. Stomatal conductance increasedby 41·7%. The steadiness in Pn per unit leaf area across the leaf insertionlevels could be mainly accounted for by an opposing effect betweena decrease in Vmes/A and a more closely packed arrangement ofphotosynthetic apparatus. All panicles were removed from rice plants (Oryza sativa L.)at anthesis and the effects of this treatment on photosyntheticgas-exchange rates and the underlying biochemical propertiesof the blade of the flag leaf were examined during senescence.Panicle removal retarded the decrease in photosynthesis in theflag leaf during senescence. Similarly, the levels of ribulose-l,5-bisphosphatecarboxylase (Rubisco), chlorophyll and Cyt f, and the activitiesof sucrose-phosphate synthase (SPS) and ADP-glucose pyrophosphorylasein panicle-free plants remained relatively high during leafsenescence and decreased more slowly than those in the controlplants. Regression analysis showed no difference between thetreated and untreated plants in the relationship between CO2-limitedphotosynthesis (measured at an intercellular CO2 pressure of15 Pa) and Rubisco content, or in the relationships betweenCO2-saturated photosynthesis (measured at an intercellular CO2pressure above 60 Pa), Cytf content and SPS activity. Theseresults indicated that the change in photosynthesis caused bypanicle removal at anthesis could be explained by the changesin the amounts and/or activities of the major biochemical participantsin photosynthesis. The temporary accumulation of starch and sucrose in the flagleaf was observed in the panicle-free plants, but the accumulationwas not significant. However, in the panicle-free plants, theweight of shoots, excluding panicles, increased by 200% andthat of roots increased by 150% at the harvesting stage. Thus,it appears that, since the effective translocation of photosynthateto other organs, such as late tillers and roots, can occur,the removal of panicles has no effect on the relationship betweenthe rate of photosynthesis and the levels of the major biochemicalparticipants in photosynthesis. The effects of nitrate supply on the composition (cell numbers,protein and chlorophyll contents) of flag leaves of winter wheatgrown with two amounts of N fertilizer and of spring wheat grownin the glasshouse under controlled nitrate supply are describedand related to photosynthesis. Nitrogen deficiency decreasedthe size of leaves, mainly by reducing cell number and, to asmaller extent, by decreasing cell volume. Protein content perunit leaf area, per cell and per unit cell volume was largerwith abundant N. Total soluble protein, ribulose bisphosphatecarboxylase-oxygenase (RuBPc-o) protein and chlorophyll changedin proportion irrespective of nitrogen supply and leaf age.Photosynthesis per unit area of flag leaf and carboxylationefficiency in both winter and spring wheat were proportionalto the amount of total soluble protein up to 7.0 g m–2and to the amount of RuBPc-o protein up to 4.0 g m–2.However, photosynthesis did not increase in proportion to theamount of total soluble or RuBPc-o protein above these amounts.In young leaves with a high protein content the measured ratesof photosynthesis were lower than expected from the amount andactivity of RuBPc-o. Carboxylation per unit of RuBPc-o protein,measured in vitro, was slightly greater in N-deficient leavesof winter wheat but not of spring wheat. RuBPc-o activity perunit of RuBPc-o protein was similar in winter and spring wheatleaves and remained approximately constant with age, but increasedin leaves showing advanced senescence. RuBPc-o protein fromN-deficient leaves migrated faster on polyacrylamide gels thanprotein from leaves with high N content. Regulation of the rateof photosynthesis in leaves and chloroplasts with a high proteincontent is discussed. The conductance of the cell to the fluxof CO2 from intercellular spaces to RuBPc-o active sites iscalculated, from cell surface areas and CO2 fluxes, to decreasethe CO2 partial pressure at the active site by less than 0.8Pa at an internal CO2 partial pressure of 34 Pa. Thus the decreasein partial pressure of CO2 is insufficient to account for theinefficiency of RuBPc-o in vivo at high protein contents. Otherlimitations to the rate of photosynthesis are considered. A remarkable difference was found in the survival of leavesof Mesembryanthemum crystallinum with plants grown in the C3versus the CAM mode. With excised leaves (petiole in solution)of C3-mode plants subjected to 6 days of darkness, there wasa large reduction in the chlorophyll content of the leaf andleaf turgor had decreased. By day 9, the chlorophyll had disappeared,except at the major veins, and the leaf tip had dried and turnedbrown. In contrast, the leaf tissue in the CAM mode showed onlya partial loss of chlorophyll during the same period, and evenafter 17 days of darkness, the tissue at the base was stillalive. Similarly, intact plants grown in the C3 mode deterioratedmuch faster during 20 days of darkness than did plants grownin the CAM mode. Chlorophyll content, chlorophyll a/b ratio,phosphoenolpyruvate carboxylase, NADP-malic enzyme, malate andstarch content were measured. In both C3- and CAM-mode plants,the starch content decreased rapidly during the dark periodand was nearly depleted after two days. In the CAM-mode tissue,there was a relatively high level of malate during prolongeddarkness (up to 17 days), with a transitory rise early in thedark period. In contrast, the malate content was low and rapidlydepleted in the C3-mode leaves kept in darkness. These findingssuggest that malate may be an important source of carbon forsustaining leaves of CAM-mode M. crystallinum during prolongeddarkness. 小麦品种的更新换代是小麦产量不断提高的重要因素,阐明小麦品种演替过程中不同生理特性的变化对新品种选育具有重要参考价值.旗叶衰老速率快慢是影响小麦产量水平的关键因素,目前对于不同小麦品种衰老过程中旗叶光系统Ⅱ功能的变化规律尚不清楚.本试验选用1941-2014年间河南地区不同时期种植的31个品种,通过黑暗诱导离体叶片衰老,测定旗叶叶绿素荧光诱导动力学参数、叶绿素相对含量的变化,分析了光系统Ⅱ功能的变化规律.结果表明:品种演替过程中旗叶的叶绿素含量逐渐提高,衰老过程中近代品种叶绿素的降解速率低于较早年代品种;旗叶衰老过程中,近代品种荧光诱导动力学曲线的J点上升幅度小于I点;品种更替过程中光系统Ⅱ最大光化学效率和单位面积有活性反应中心数目逐渐增加,但是近代品种降低速率低于较早年代品种.叶绿素含量的变化与未衰老叶片中Fv/Fm没有显著相关性,但是随着衰老程度增加,相关性逐渐增大,且趋势线斜率逐渐提高;光系统Ⅱ单位面积有活性反应中心数目与品种育成时间呈显著正相关,且随着衰老程度增加,相关程度和趋势线斜率均显著提高.综上,小麦品种演替过程中,旗叶叶绿素含量逐渐升高,降解速率逐渐减缓,光合电子传递过程中QA到QB电子传递的抗衰老能力得到改善,从而减缓了衰老过程中光系统Ⅱ最大光化学效率和有活性反应中心的衰减速率,同时,叶绿素含量的提高和旗叶光系统Ⅱ抗衰老能力的增强也是品种更替过程中产量逐渐提高的重要因素.  相似文献

The effects of leaf age and of shading on photosynthetic rateand on other leaf parameters of potato (Solanum tuberosum L.)were studied using a portable gas exchange system. A rapid decreasein the rate of photosynthesis during leaf senescence was observed.This was accompanied by an increase in stomatal resistance,and as a result a fairly constant level of sub-stomatal CO2concentration was maintained at all leaf ages. The reductionin the photosynthetic rate in older leaves was therefore assumedto be essentially mesophyllic in origin, whereas the stomatalresponse was probably secondary. Canopy density significantly affected the rate of photosyntheticreduction with leaf age. Leaves maintained under high radiationintensities manifested a slower decline in their photosyntheticrate, especially in the early stages of their senescence, thanleaves kept under shade conditions. Leaf senescence is a developmentally regulated process that contributes to nutrient redistribution during reproductive growth and finally leads to tissue death. Manipulating leaf senescence through breeding or genetic engineering may help to improve important agronomic traits, such as crop yield and the storage life of harvested organs. Here, we studied natural variations in the regulation of plant senescence among 16 Arabidopsis thaliana accessions. Chlorophyll content and the proportion of yellow leaves were used as indicator parameters to determine leaf and plant senescence respectively. Our study indicated significant genotype effects on the onset and development of senescence. We selected three late- and five early-senescence accessions for further physiological studies. The relationship between leaf and plant senescence was accession-dependent. There was a significant correlation between plant senescence and the total number of leaves, siliques and plant bolting age. We monitored expression of two senescence marker genes, SAG12 and WRKY53 , to evaluate progression of senescence. Our data revealed that chlorophyll content does not fully reflect leaf age, because even fully green leaves had already commenced senescence at the molecular level. Integrating senescence parameters, such as the proportion of senescent leaves, at the whole plant level provided a better indication of the molecular status of the plant than single leaf senescence parameters.  相似文献

Kinetin retarded the decrease in chlorophyll content in leafdiscs from 5 species of plants with amphistomatous leaves, wherethe upper surface was exposed to air, but not in Rumex acetosera.When leaf discs were floated so that the lower surface was exposed,the effect of kinetin was less evident. Kinetin also stimulatedtranspiration in leaf discs from Nicotiana tabacum (amphistomatous),but not in leaf discs from Paederia chinensis (hypostomatous).Nor kinetin did retard chlorophyll breakdown in this specieswhen leaf discs were floated so that the stomatal surface wasin contact with the solution. The ineffectiveness of cytokininsin chlorophyll retention in leaf discs from hypostomatous leaveswas not due to reduced uptake of benzylaminopurine-14C. Chlorophyll retention was severely inhibited by coating theleaf surface with vaseline either in the presence or absenceof kinetin. Leaf discs floated on a solution exposed to CO2-lessair retained more chlorophyll than those in normal air. Thereis thus a close relationship between stomatal opening (as measuredby stimulation of transpiration) and chlorophyll retention,as influenced by cytokinins. It is suggested that cytokinin-induced chlorophyll retentionand odier effects on leaf tissues could be mediated throughits effects on stomatal opening. (Received January 22, 1976; )  相似文献

The role of photosynthesis in inducing adventitious bud formationon leaf segments of Heloniopsis orientalis was investigated.The effect of white light reached a maximum at about l25 J?m–2?sec–1.White, red, blue and far-red light were effective in inducingbud formation, but green light was not. In darkness, bud formationwas induced if sugar was added to the nutrient medium. The photosyntheticinhibitors DCMU and AT blocked the effect of light. Bud formationwas inhibited in CO2-free air. The requirement of sucrose forbud formation in darkness could be replaced by citrate. Flag leaf removal at any stage of grain growth hastened senescence (reduction in chlorophyll content) of the sterile glumes whereas a removal of the latter did not alter senescence of the former. Kernel mass, grain mass per ear, harvest index and sink activity reduced more conspicuously by the removal of glumes than by the flag leaf removal. Removal of grains hastened senescence of the glumes only, although protein content increased at a later grain removal in the majority of source organs. Moisture contents of the developing grains were mostly reduced by the removal of either the flag leaf or the glumes, though flag leaf removal at anthesis affected it the most. Protein levels of the developing grains reduced at harvest only when the glumes were removed at anthesis but sugar levels in later phases always decreased irrespective of the time of removal and nature of the source organs.  相似文献

A large decrease was observed in the chlorophyll content ofthe primary leaves of Phaseolus vulgaris during senescence.Chloroplasts isolated from mature and senescent leaves gavevery similar light saturation curves for electron transportreactions involving either PS I or PS II, indicating that theaverage number of chlorophyll molecules associated with eachreaction centre did not change during senescence. It is concludedthat the reaction centres ceased to function at the same timeas, or perhaps before, their antenna chlorophylls were lostfrom the thylakoid membrane, and that the percentage decreasein the number of functional reaction centres per leaf was atleast as great as the percentage decrease in the leaf chlorophyllcontent. The chlorophyll-protein composition of thylakoid membrane preparationswas examined by electrophoresis of samples treated with sodiumdodecyl sulphate. In older leaves a smaller proportion of thechlorophyll applied to polyacrylamide gels was associated withthe P700- chlorophyll a-protein complex. There was also a declinein emission at 734 nm in the 77 °K fluorescence spectrumof intact leaf tissue during senescence. These results indicatethat older leaves contained a smaller proportion of chlorophyllsassociated with PS I, and this is consistent with the decreaseobserved in the leaf chlorophyll a/b ratio during senescence.The effect of these changes in chlorophyll content on the capacityof the chloroplast to carry out photosynthetic electron transportis discussed.  相似文献

Acclimation of fully developed leaves of Bischofia javanicaBlume to shadelight was examined. Seedlings were grown undersimulated daylight (1000 µmol m–2 s–1), thentransferred to a simulated shadelight (40 µmol m–2s–1). When a high-light leaf was transferred to low light, large negativenet photosynthetic rates (Pm) were recorded. This decrease wasrapid, but within 7 d the rate increased and became equal tothe low-light control leaf. These changes in photosynthesisdid not follow the pattern of changes in stomatal conductance(gs). Transfer to the low light resulted in a dramatic decreasein leaf weight per unit area (Lw), and most of the decreasesin Lw occurred within 3 d of transfer when the Pm of the transferredleaf was well below that of the low-light control leaf. There was a significant decrease in chlorophyll a in the transferredleaf without an appreciable change in chlorophyll b resultingin a large decrease in the chlorophyll a to chlorophyll b ratio.Leaf chlorophylls per unit area were higher in the transferredleaf than the low-light control leaf. Maximum photosyntheticrate in the transferred leaf was decreased by 40% compared tothat for the high-control leaf, but was almost at the same extenthigher than the low-light control leaf The results are discussedin the context of carbon gain capacity of its seedlings underlight-limiting forest understorey habitats. Rates of net photosynthesis of the flag leaves of 15 genotypesof wheat and related species were measured throughout theirlife, using intact leaves on plants grown in the field. At thestage when rates were maximal, they were in general highestfor the diploid species, intermediate for the tetraploidspeciesand lowest for Triticum aestivum (means of 38, 32 and 28 mgCO2 dm–2 h–1 respectively). Rates were stronglynegatively correlated with leaf area, leaf width and the meanplan area per mesophyll cell and positvely correlated with stomatalfrequency and number of veins per mm of leaf width. The differencesamong species in these attributes were mainly related to ploidylevel. It was not possible to determine the relative importanceof each anatomical feature, though the changes in stomatal frequencyhad only slight effects on stomatal conductance and the observeddifferences in rates of photosynthesis were much greater thanwould be expected from those in stomatal conductance alone. There was genetic variation in rates of light dependent oxygenevolution of isolated protoplasts and intact chloroplasts butno difference attributable to ploidy. The mean rate, 91 µmolO2 mg–1 chlorophyll h–1, equivalent to 3.9 mg CO2mg-1chlorophyll h-1 was considerably less than the rate of photosynthesisin comparable intact leaves, which was 7.2 mg CO2 mg–1chlorophyll h–1. The total above-ground dry matter yields were least for thewild diploids T. urartu and T. thauodar and the wild tetraploidT. dicoccoides, but the other wild diploids produced as muchdry matter as the hexaploids. The prospects of exploiting differences in photosynthetic ratein the breeding of higher yielding varieties are discussed. When leaf discs of Xanthium strumarium L. a C3 plant, or Zeamays L. a C4 plant, are incubated in 1-aminocyclopropane-l-carboxylicacid (ACC) in closed flasks, ethylene is released. The rateof ethylene release appears to be dependent on the levels oflight and CO2 available for photosynthesis in the tissues. In Xanthium the rate of ethylene release is lower in the lightthan in the dark regardless of the presence or absence of addedbicarbonate as a source of CO2. The inhibition of ethylene releaseis most apparent in the absence of added bicarbonate (i.e. atthe CO2 compensation point), and at light intensities sufficientto saturate photosynthesis (had the CO2 level in the test flaskbeen maintained). In contrast, light dramatically promotes therate of ethylene release from Zea leaf tissue when the CO2 levelis maintained above the CO2 compensation point. The rate ofethylene release from either Xanthium or Zea, incubated withor without added bicarbonate, does not appear to be alteredby further increasing the light intensity above the minimallevels sufficient to saturate photosynthesis. In the closed system used in these studies and at a light intensitysufficient to saturate photosynthesis, Xanthium and Zea leaftissue both appear to release comparable amounts of ethylenefrom ACC when the data is expressed on a chlorophyll basis.However, in Xanthium the rate of ethylene release is similarin light and dark, while in Zea the rate in the light is muchgreater than in the dark when the data is expressed either ona leaf area or on a chlorophyll basis. It is suggested thatthe different responses of these tissues to light/dark transientsmay reflect differences in their ability to metabolize ACC and/ordifferences in their ability to retain and metabolize ethyleneitself.  相似文献

Nitrogen mobilization and the pattern of proteolytic enzymeswere investigated in leaves and glumes of field-grown winterwheat (Triticum aestivum L.) during maturation. Source/sinkrelations were changed by removal of the ear, the flag leafor the lower leaves shortly after anthesis. Removal of the earwas most effective, resulting in delayed senescence of the flagleaf with the chlorophyll, aminopeptidase and carboxypeptidaseactivities remaining high in contrast to the control, whereasneutral endopeptidase activity increased more slowly. No majorchanges were observed in the second leaf from the top in plantswith either ears or flag leaves removed. Nitrogen mobilizationand proteolytic activities in glumes and the remaining leaveswere influenced only slightly by leaf removal. In earless plants,nitrogen was transported from the second leaf into the leafsheath and stem, but in the flag leaf the total reduced nitrogenremained high and free amino groups increased. The increase in endopeptidase activity was influenced by thesource/sink relations. However, the accumulation of amino groupsand the increasing endopeptidase activity in the flag leaf ofearless plants suggest that the nitrogen sink capacity did notgreatly control protein degradation; it remains to be seen whetherphytohormones, accumulated amino acids or other factors delayedthe increase in endopeptidase activity. Chlorophyll synthesis in the regreening photoheterotrophic calluscultures of Arachis hypogaea and Kalanchoe fedtschenkoi wassaturated at a light intensity of 5,000 lux. However, the optimaldevelopment of photosynthetic functions (O2 evolution, CO2 fixation,photosystem activities) required more irradiance (12,000–14,000lux). Two Phaseolus vulgaris L. cultivars were grown at 20/15, 25/20,and 30/25 °C day/night temperatures in growth chambers witha 16 h thermoperiod corresponding to the photoperiod. When thefirst trifoliolate leaf was fully expanded rates of CO2 exchange(CER) were measured at 27 °C and saturating light usinginfrared gas analysis. Stomatal (rs) and mesophyll resistances,CO2 compensation points, activities of the enzymes ribulosebisphosphate carboxylase (RuBPCase), glycolate oxidase (GAO),malate dehydrogenase (MDH), and fructose-1, 6 diphosphate (FDP),chlorophyll content, Hill activities, and leaf anatomy at boththe light and electron microscope level were also investigatedin these leaves. Rates of CO2 exchange in the light, transpiration rate, andchlorophyll content increased with increasing growth temperaturewhile leaf thickness, specific leaf weight, RuBPCase activity,compensation point, and stomatal resistance decreased. Mesophyllresistance also decreased when calculated assuming zero chloroplastCO2 concentration (rm, o), but not when calculated assuminga chloroplast CO2 concentration equal to the CO2 compensationconcentration (rm, g). Average leaf size was maximal in 25/20°C plants while dark respiration, MDH activity, stomataldensity, and starch were minimal. The activities of GAO andFDP and Hill activity were not affected by temperature pretreatment.  相似文献
