Nanoindentation has been widely used as a means to measure the micro-mechanical properties of bone and to predict the macroscopic properties. The role of indent depth and indenter tip geometry in measuring the hierarchical properties of bone tissue was explored experimentally using a range of spherical indenter tips of R=5, 25, 65, and 200 μm. Nanoindentation arrays, not targeted to fall on specific structures or locations, enabled statistical sampling of osteons within PMMA-embedded, bovine, cortical bone on a single sample to a range of maximum displacements (minimum of 100 nm and maximum of 2000 nm). Elastic finite element models were then utilized to isolate the contributions of indenter tip radius, contact area, and position within the lamellar structure in comparison to the experimental results. For a small, R=5 μm indenter tip, indentation modulus consistently increased with contact depth and increased plastic deformation, resulting in an artificial increase in elastic properties. While larger radius tips (R=25, 65, and 200 μm) did not enable evaluation of a high spatial resolution on the surface, they produced data that was representative of the lower load and contact depth measurements with the smaller tip. However the sensitivity to mechanical property variations across the 2-D surface of the material was lost with increase in indenter tip size. Correspondingly, measurement variance was also decreased as the volume contributing to the indent response represented an average of surface roughness, varying mineral content, defects, and underlying tissue type and structure.  相似文献

The structure of mechanoreceptors at the base of labeilar taste hairs of the blowfly Phaenicia serricata were examined in stimulated and unstimulated conditions (i.e. with the hair bent or unbent). Physiological recordings from the mechanoreceptor showed that the receptors responded when the hair is bent dorsally or ventrally and when the hair is bent at extreme angles. These conditions are the same as those placed on hairs in the anatomical studies. Bending the hair toward the ventral labellar surface caused the hair base to compress and indent the tubular body and its surrounding membrane and sheath at the distal end of the mechanoreceptor dendrite. In compressed tubular bodies, microtubules oriented longitudinally were bent and separated a greater distance from each other. Separation as much as 70 nm was observed in compressed tubular bodies as compared with a maximum of 26 nm between microtubules in tubular bodies of unbent hairs. The dense amorphous material between microtubules of compressed tubular bodies formed prominent bridges 18 nm thick connecting the microtubules at intervals of 48–74 nm. Thin 10 nm filaments were also evident in the spaces between microtubules. When the hair was bent toward the proximal end of the proboscis, the tip of the tubular body was bent about 15 °. The tubular body appears to function as a firm but resilient structure over which the dendritic membrane can be stretched during mechanostimulation. Comparison of morphology of bent and unbent hairs suggests a means by which mechanical force from the movement of the hair is transferred to the receptors by structures in the hair socket region. No differences were found in ciliary structures of stimulated and unstimulated receptors.  相似文献

The fracture toughness of hard biomaterials, such as nacre, bovine hoof wall and beetle cuticle, is associated with fibrous or lamellar structures that deflect or stop growing cracks. Their hardness and reduced modulus were measured by using a nanoindenter in this paper. Micro/nanoscale cracks were generated by nanoindentation using a Berkovich tip. Nanoindentation of nacre and bovine hoof wall resulted in pile-up around the indent. It was found that the fracture toughness (Kc) of bovine hoof wall is the maximum, the second is nacre, and the elytra cuticle of dung beetle is the least one.  相似文献

Summary Several crosses werer made between some races of peas frequently grown in Holland for culinary purposes. Of these Blauwpeul is a large brown seeded pea with indent seeds. It has pigment in its testa, but does not possess the factor for spots.Krombek, a pea with curved pods, yellow cotyledons and pigmentless testa possesses this factor for spots; thus the cross Krombek x Blauwpeul gives spotted seeds in the F1.Kroonerwt, a green pea with big globular seeds has also the spto factor, but American Wonder, green with wrinkled seeds, has not. Low Cape Capucin has small spotted seeds of indent shape. Crossed with American Wonder it gives rise to all kinds of peas, but a remarkable fact in all crosses mentioned is, that where pigment is present in the testa, the form of the seeds always is indent. Pure wrinkled and globular seeds have colourless testa.The factor for indent is hyperstatic over globular.  相似文献

Cracks in articular cartilage are a common sign of joint damage, but failure properties of cartilage are poorly understood, especially for damage initiation. Cartilage failure may be further complicated by rate-dependent and depth-dependent properties, including the compliant surface layer. Existing blunt impact methods do not resolve local cartilage inhomogeneities and traditional fracture mechanics tests induce crack blunting and may violate underlying assumptions of linear elasticity. To address this knowledge gap, we developed and applied a method to indent cartilage explants with a sharp blade and initiate damage across a range of loading rates (strain rates 0.5%/s–500%/s), while recording local sample deformation and strain energy fields using confocal elastography. To investigate the importance of cartilage’s compliant surface, we repeated the experiment for samples with the surface removed. Bulk data suggest a critical force at which the tissue cuts, but local strains reveals that the deformation the sample can sustain before reaching this force is significantly higher in the surface layer. Bulk and local results also showed significant rate dependence, such that samples were easier to cut at faster speeds. This result highlights the importance of rate for understanding cracks in cartilage and parallels recent studies of rate-dependent failure in hydrogels. Notably, local sample deformation fields were well fit by classical Hookean elasticity. Overall, this study illustrates how local and global measurements surrounding the initiation of damage in articular cartilage can be combined to reveal the importance of cartilage’s zonal structure in protecting against failure across physiologically relevant loading rates.  相似文献

The role ofmyosin light chain phosphorylation in regulating the mechanicalproperties of the cytoskeleton was studied in NIH/3T3 fibroblastsexpressing a truncated, constitutively active form of smooth musclemyosin light chain kinase (tMK). Cytoskeletal stiffness determined byquantifying the force required to indent the apical surface of adherentcells showed that stiffness was increased twofold in tMK cells comparedwith control cells expressing the empty plasmid (Neo cells).Cytoskeletal stiffness quantified using magnetic twisting cytometryshowed an ~1.5-fold increase in stiffness in tMK cells compared withNeo cells. Electronic volume measurements on cells in suspensionrevealed that tMK cells had a smaller volume and are more resistant toosmotic swelling than Neo cells. tMK cells also have smaller nuclei,and activation of mitogen-activated protein kinase (MAP kinase) andtranslocation of MAP kinase to the nucleus are slower in tMK cells thanin control cells. In tMK cells, there is also lessbromodeoxyuridine incorporation, and the doubling time isincreased. These data demonstrate that increased myosin light chainphosphorylation correlates with increased cytoskeletal stiffness andsuggest that changing the mechanical characteristics of thecytoskeleton alters the intracellular signaling pathways that regulatecell growth and division.

Indirect surveys may prove to be useful tools in complementing classical direct counts when monitoring ungulate populations and may also promote better understanding of the precise structure and functioning of the rich ungulate communities of African savannas. However, the identification of faecal pellets can be difficult where several sympatric species occur. This study develops simple field criteria for distinguishing between pellets among ten sympatric West African ungulates. A discriminant analysis was performed, using the mean of measurements of pellet groups from different species to pinpoint and characterize the most useful morphological criteria for separation between them. The mean diameter of pellets within each pellet group proved to be the most valuable variable for species segregation, whilst the second axis separated species by mean indent depth. The pellet groups of six of the ten designated species could be identified with a minimum misclassification error. However, no simple morphological variables emerged to permit discrimination between hartebeest and topi, or between bushbuck and Bohor reedbuck pellets. Once pellet groups have been identified, their density and spatial distribution may provide useful information on the use of space and habitat of sympatric species, over given periods.  相似文献

There are two major kinds of fisheries for the chub mackerel in Japan. The purse seine net fishery exploits young and adult fish during the foraging season, while the dip net fishery exploits fish before and during the spawning season. To compare the damage to the chub mackerel stock caused by purse seine net and dip net fisheries, we introduced impact factors of the two kinds of fisheries on the two kinds of fisheries and estimated number of eggs spawned in 1982. We defined the impact factor as the ratio of the average reproductive value of individuals caught by each fishery to the reproductive value at maturity. The number of individuals caught by the purse seine net fishery was 66 times larger than the number caught by the dip net fishery, while the impact factor of the former was 84% of that of the latter. Thus, the total damage caused by the purse seine net fishery was much higher than the damage caused by the dip net fishery. We can evaluate the damage caused by mortality factors on endangered species using this method.  相似文献

The detachment of biomass from suspended biofilm pellets in three-phase internal loop airlift reactors was investigated under non-growth conditions, and in the presence of bare carrier particles. In the experiments the size of biofilm pellets and bare carrier particles was varied. Results show that an increase in particle size drastically increases the abrasion rate caused by particle collisions. This increase is larger than predicted by conventional collision theory, which accounts for changes in collision frequency and collision impact. However, collision theory was formulated for neutrally buoyant particles which follow the liquid flow. This condition does not hold for biofilm pellets and carrier particles. The difference might therefore be caused by differences in particle responses to flow fluctuations. An empirical relationship, including this flow response, was formulated. The collision impact is also strongly affected by the roughness of a bare carrier particle: sharp and edgy particles cause much more damage than smoother ones. (c) 1997 John Wiley & Sons, Inc. Goal, Scope and Background The goal of this study is to determine the environmental impact of using one cubic metre of water in the Walloon Region. The whole anthropogenic water cycle is analysed, from the pumping stations to the wastewater treatment plants. The functional unit has been defined as one cubic metre of water at the consumer tap. This study was carried out in the context of the EU Water Framework Directive. It is part of a programme called PIRENE launched by the Walloon Region to fulfil the requirements of this Directive. Methods A model of the whole anthropogenic water cycle in the Walloon Region was developed. The model is mainly based on site-specific data given by the companies working in the field of water production and wastewater treatment. It was used to assess the environmental impact from the pumping station to the wastewater treatment plant using the Eco-Indicator 99 methodology. Eco-Indicator 99 has been adapted in order to better take into account environmental impact of acidification and eutrophication. Characterisation factors have been calculated for COD, nitrogen and phosphate emissions. From the reference model, different scenarios have been elaborated. Results and Discussion On the basis of the inventory, the environmental impact of five scenarios has been evaluated. Acidification and eutrophication is the most important impact category. It is mainly caused by the wastewater that is discharged without any treatment, but also by the effluent of the wastewater treatment plant. So, this impact category has the lowest environmental load when the wastewater treatment rate is high. For the other impact categories, the impact generally increases with the wastewater treatment rate. During wastewater treatment, energy and chemicals are indeed consumed to improve the quality of the final outputs, and thus to reduce the environmental impact related to acidification and eutrophication. A comparison between the scenarios has also shown that the building of the sewer network has a significant contribution to the global environmental load and that the stages before the tap contribute less to the environmental impact than the stage after the tap. Conclusions The three stages that contribute significantly to the global environmental load are: water discharge, wastewater treatment operation and, to a lesser extent, the sewer system. The results show that the wastewater treatment rate must be as high as possible, using either collective or individual wastewater treatment plants. Even a small water discharge without any treatment has a significant environmental impact. Operation of the wastewater treatment plants must also be improved to reduce the environmental impact caused by the effluent of the plants. For new wastewater treatment plants, building plants treating nitrogen and phosphorus should be encouraged. A sensitivity analysis was conducted and showed that the results of the study were not very affected by a modification of key parameters. Impact assessment using the CML methodology has confirmed the results obtained with Eco-Indicator 99.  相似文献

Pollen analytical results from a littoral profile taken in Lake Constance compared with pollen profiles from small kettle holes nearby form the basis for conclusions concerning human population density, the economy and environment from the Neolithic period to the Middle Ages. Early Neolithic human impact is implicated in a lime decline and also the expansion of beech. The late Neolithic lakeshore settlements caused a decline of elm, beech and lime and, by shifting cultivation, considerably changed the forest cover. The settlements were abandoned after less than 100 years. There were long periods without distinct human impact in the middle and towards the end of the late Neolithic period. Since at least the Late Bronze Age there has been permanent habitation in the region. Human impact was greatest in the High Medieval period and later, and was also substantial in the late La Tène and Roman periods. Distinct declines in human impact can be observed between the La Tène and Roman periods and in the Migration and Merovingian periods. In these intervals, open land and grazed oak forest were replaced by birch and later on by beech forests. The decreases in human impact are not of the same intensity in all diagrams.  相似文献

The forestry sector is experiencing an increasing demand for documentation about its environmental performance. Previous studies have revealed large differences in environmental impact caused by forestry operations, mainly due to differences in location and forestry practice. Reliable information on environmental performance for forestry operations in different regions is thus important. This article presents a case study of forestry operations in Norway. Environmental impact and value added of selected operations were assessed. This was done with a hybrid life cycle assessment (LCA) approach. Main results, including a sensitivity analysis, are presented for a set of four impact categories. The production chain assessed included all processes from seedling production to the delivery of logs to a downstream user. The environmental impact was mainly caused by logging, transport by forwarders, and transport by truck. These three operations were responsible for approximately 85% of the total environmental impact. The contribution to value added and total costs were more evenly distributed among the processes in the value chain. The sensitivity analysis revealed that the difference in environmental impact between the worst case scenario and the best case scenario was more than a factor of 4. The single most important process was the transport distance from the timber pile in the forest to the downstream user. The results show that the environmental impact from forestry operations in boreal forests was probably underreported in earlier studies.  相似文献

Abstract:  The haemogram of Rhynocoris kumarii Ambrose and Livingstone comprises prohaemocytes, plasmatocytes, granular haemocytes, cystocytes and oenocytoids. The impact of five insecticides, viz. monocrotophos, dimethoate, methylparathion, quinalphos and endosulfan on the total haemocyte count (THC) and differential haemocyte counts (DHC) was studied. All of the insecticides except endosulfan initially reduced both prohaemocytes and plasmatocytes, increased the granular haemocytes, altered the percentage of cystocytes and oenocytoids and increased the total haemocyte count (THC). On the contrary, endosulfan initially increased the prohaemocytes and plasmatocytes, decreased the granular haemocytes and also the THC. The highest impact on the DHC and THC was caused by methylparathion and monocrotophos and the least impact by endosulfan. Hence, endosulfan is considered as the safest insecticide followed by dimethoate and quinalphos among these five insecticides to use with R. kumarii .  相似文献

The juvenile shell of living discinid brachiopods is composed of valvular mosaics of rhombic, micrometric–sized siliceous tablets. The tablets are shed in adult growth stages leaving shallow imprints on the primary layer of the organophosphatic mature shell, which occur on an upper Silurian discinoid. Imprints also indent the first–formed shells of over 100 lingulate genera, including all acrotretides and Paterula . No micrometric bodies that could have made these imprints are known, but their structure and composition can be inferred from imprint morphology. Diagnostic features of imprints as casts include constancy of shape and size, and rigidity of the indenting bodies relative to the rheology of the polymerizing primary layer of chitin, glycosaminoglycans (GAGs) and apatite, in which they were embedded. Three kinds of discrete structures are distinguishable. Nanometric–sized, basinal pits that may be compound or deformed, are assumed to be casts of mucinous vesicles. Larger, flat–based and hemispherical imprints were almost certainly made by biomineralized discoids and spheroids. Additional evidence that discoids and spheroids were more soluble than apatite suggests that they were calcitic. Mineralized mosaics could have protected pelagic juveniles from solar radiation in the early Palaeozoic when the ozone layer was more rarified than today.  相似文献

Abstract

Results from two sets of molecular dynamics simulations are reported. In the first set of simulations a nanoscale tip was used to indent single-crystal gold lattices subjected to external strains. These were carried out to explore possible relationships between nanoindentation curves and elastic properties of uniformly strained films. The changes in the slope of the loading curves reflect the stress state of the sample. In the second set of simulations the use of shallow nanoindentation for mapping nonuniform residual surface stress near a dislocation intersecting a surface was tested. Correlation between the maximum force on the tip and the initial local stresses at the point of indentation were observed. Preliminary atomistic simulations indicate that atomic-force microscopy can be used as a nondestructive, nanoscale probe of the surface stress distributions.  相似文献

Four neocatastrophist claims about mass extinction are currently being debated; they are that: 1, the late Cretaceous mass extinction was caused by large body impact; 2, as many as five other major extinctions were caused by impact; 3, the timing of extinction events since the Permian is uniformly periodic; and 4, the ages of impact craters on Earth are also periodic and in phase with the extinctions. Although strongly interconnected the four claims are independent in the sense that none depends on the others. Evidence for a link between impact and extinction is strong but still needs more confirmation through bed-by-bed and laboratory studies. An important area for future research is the question of whether extinction is a continuous process, with the rate increasing at times of mass extinctions, or whether it is episodic at all scales. If the latter is shown to be generally true, then species are at risk of extinction only rarely during their existence and catastrophism, in the sense of isolated events of extreme stress, is indicated. This is line of reasoning can only be considered an hypothesis for testing. In a larger context, paleontologists may benefit from a research strategy that looks to known Solar System and Galactic phenomena for predictions about environmental effects on earth. The recent success in the recognition of Milankovitch Cycles in the late Pleistocene record is an example of the potential of this research area.  相似文献

The pharmacokinetic (PK) behavior of monoclonal antibodies in cynomolgus monkeys (cynos) is generally translatable to that in humans. Unfortunately, about 39% of the antibodies evaluated for PKs in cynos have fast nonspecific (or non-target-mediated) clearance (in-house data). An empirical model relating variable region (Fv) charge and hydrophobicity to cyno nonspecific clearance was developed to gauge the risk an antibody would have for fast nonspecific clearance in the monkey. The purpose of this study was to evaluate the predictability of this empirical model on cyno nonspecific clearance with antibodies specifically engineered to have either high or low Fv charge. These amino acid changes were made in the Fv region of two test antibodies, humAb4D5-8 and anti-lymphotoxin α. The humAb4D5-8 has a typical nonspecific clearance in cynos, and by making it more positively charged, the antibody acquires fast nonspecific clearance, and making it less positively charged did not impact its clearance. Anti-lymphotoxin α has fast nonspecific clearance in cynos, and making it more positively charged caused it to clear even faster, whereas making it less positively charged caused it to clear slower and within the typical range. These trends in clearance were also observed in two other preclinical species, mice and rats. The effect of modifying Fv charge on subcutaneous bioavailability was also examined, and in general bioavailability was inversely related to the direction of the Fv charge change. Thus, modifying Fv charge appears to impact antibody PKs, and the changes tended to correlate with those predicted by the empirical model.  相似文献

Exposure to Lead -causes testicular dysfunction through oxidative stress, inflammation, and apoptosis; however, naringenin (NGN) therapeutic impact against lead-evoked testicular dysfunction remains elusive. Herein, the point of the study was to examine the defensive impact of NGN on testicular dysfunction initiated by lead. Seventy-Two male Wistar rats were allotted into nine groups; control group, drug control groups, lead acetate group, as well as NGN treated groups (10, 25, and 50 mg/kg) respectively, given 5 days before lead acetate treatment. The result showed clearly the impact of lead on reduced sperm count, sperm motility as well as serum testosterone and LH levels. Additionally, it caused a significant rise in testicular inflammatory markers TNF-α, IL-1β, and TGFβ, effects that were accompanied by a reduction of AKT and mTOR levels. Lead acetate also caused degenerative changes in the testis, atrophy, and loss of spermatogenic series. Our findings revealed that NGN in a dose-dependent manner improved spermiotoxicity induced by lead acetate via restoration of the testicular function, preservation of spermatogenesis, halting inflammatory cytokines along with the enhancement of germ cell survival using upregulation of AKT/mTOR expressions. The present study discloses that NGN suppresses lead acetate toxicity that is involved in the antioxidant effect in a dose-dependent manner, besides its anti-inflammatory property.  相似文献

The objective of this study is to investigate the impact of trauma on erythrocyte caused by long term in vitro pumping using roller pump. Ten bags of human blood (400 ml each) were provided by a local blood bank and they were divided into two groups with five bags in each group. Each blood bag was subject to pumping in a closed circuit, which was composed of silica gel tubes and a roller pump. Polystan and COBE pumps were used for the two groups, respectively. The blood was pumped for 16 h in vitro. Free hemoglobin (FHb), platelets (PLT), erythrocyte fragility (EF), and morphological analysis of erythrocytes observed under scanning electron microscope were measured to evaluate the impact of trauma on erythrocytes. A small amount of blood was collected for analysis before pumping, at the end of the 4th hour and then every 2 h till the end of the 16th hour. Some blood samples were also collected for electron microscope scanning before pumping and every 4 h during pumping. It was found that FHb and PLT linearly increased with the pumping time. There was a significant correlation between the two parameters (r=0.7745, p<0.001). The hemolysis indexes of the two groups were 0.296 and 0.3993 mg/L/h, respectively, with no significant difference. During the pumping process, EF changed slightly. The observation of scanning electron microscopy showed various deformed erythrocytes after pumping, including the distortion of cell membrane and the appearance of echinocytes, which increased with pumping time. This study demonstrated that long term pumping using roller pump not only caused the immediate rupture of red blood cells, i.e. the immediate hemolysis, but also caused sub-trauma to a large number of erythrocytes, which led to the delayed hemolysis. The change of erythrocyte morphology was the basis of the delayed hemolysis.  相似文献
