The comparative biologies of two hymenopterous parasites of the chaff scale,Parlatoria pergandii

Résumé La biologie des aphelinidesAphytis hispanicus (Mercet) etProspaltella inquirenda Silvestri, respectivement ecto et endoparasite deParlatoria pergandii Comstock, a été étudiée comparativement en laboratoire.A. hispanicus accomplit son cycle en 34 jours environ à 20 °C et en 16 jours à 28 °C; il produit en moyenne 23,8 descendants, tandis que pourP. inquirenda ces durées sont de 62 et 26 jours et le nombre moyen de descendants 20,6. Le comportement pendant l'oviposition et les interactions diverses entre l'h?te et le parasite ont été notés. Le nectar d'agrumes a prolongé la vie des deux parasites autant que le miel.A. hispanicus a généralement une vie plus prolongée queP. inquirenda. Le premier parasite a mieux survécu dans des conditions de forte humidité relative (61%, 80%), maisP. inquirenda s'est trouvé favorisé dans des conditions de faible humidité relative, ainsi qu'après une exposition à des températures extrêmes. En conclusion,A. hispanicus a un taux plus rapide de croissance, tandis queP. inquirenda est mieux adapté aux conditions naturelles.

La spécificité deDiadromus pulchellus [Hyménoptère: Ichneumonidae] vis-à-vis de son hôteAcrolepiopsis assectella,la teigne du poireau

Résumé La spécificité deDiadromus pulchellus vis-à-vis de son h?te habituel,Acrolepiopsis assectella, est testée à l'aide de 3 autres espèces de Lépidoptère,Plutella xylostella, Ephestia kuehniella etPlodia interpunctella. D. pulchellus parasite spontanémentA. assectella etP. xylostella, que les chrysalides soient nues ou avec leur cocon et que les larves soient élevées ou non sur leur plante h?te. Les 2 autres espèces ne sont parasitées que si elles sont introduites dans un cocon d'A. assectella ou deP. xylostella. Le cocon est donc un des éléments de la spécialisation deD. pulchellus. Il pourrait s'agir de facteurs physiques liés à la structure du cocon, mais également de composés chimiques insolubles présents dans la soie et les glandes labiales mais absents des glandes mandibulaires. La spécificité deD. pulchellus, essentiellement éthologique, semble renforcée par des facteurs physiologiques nutritionnels, la mortalité deD. pulchellus étant plus élevée dansP. unterpunctella que dans les autres espèces h?tes, en particulierA. assectella etE. kuehniella. La base trophique des larves est donc plus large que la base de ponte des femelles. Les parasito?des entièrement élevés surE. kuehniella ouP. interpunctella étant incapables de pondre spontanément dans les chrysalides des espèces dont ils sont issus, l'hypothèse d'Hopkins est ici infirmée.      

Réponses électroantennographiques de deux parasitoïdes Ichneumonidés,Diadromus pulchellus et D. collaris,aux odeurs de végétaux,du phytophage-hôte Acrolepiopsis assectella et du partenaire sexuel

Résumé Chez Diadromus pulchellus WSM et D. collaris Gr., Hyménoptères Ichneumonidés parasitoïdes spécialistes des nymphes d'A. assectella lépidoptère inféodé aux Allium, des EAG sont enregistrés en réponse à l'odeur du poireau sain ou attaqué par les larves du phytophage, odeurs actives par ailleurs sur leur comportement locomoteur (cinèses). Le thiosulfinate de dipropyle (Ti-P₂) très spécifique et abondant dans les Allium, et instable, est le mieux perçu par les deux parasitoïdes mais n'a aucun effet locomoteur chez D. pulchellus contrairement aux disulfures et thiosulfonates, plus stables et moins spécifiques. Les propriétés de l'acétate d'éthyle, actif sur l'antenne des deux parasitoïdes, et sur la locomotion de D. pulchellus expliqueraient en partie les activités biologiques du poireau attaqué sur ces parasitoïdes. Des EAG sont aussi obtenus chez les deux espèces avec l'odeur du chou et de composés soufrés plus ou moins spécifiques des Crucifères. Le thiocyanate de méthyle (TCN-M), le plus actif sur l'antenne et seul efficace sur le comportement locomoteur (de D. pulchellus), expliquerait en partie l'installation reconnue de D. collaris sur Plutella maculipennis spécialiste des Crucifères et rend celle de D. pulchelles vraisemblable.L'odeur des larves de dernier stade d'A. assectella est seule efficace sur l'antenne et la locomotion des femelles de D. pulchellus. Les mâles de ce dernier réagissent (EAG, locomotion) à des odeurs de cocons de l'hôte et particulièrement à ceux occupés par une femelle de leur espèce.

Role of futC slipped strand mispairing in Helicobacter pylori Lewisy phase variation

The O antigen of the Helicobacter pylori lipopolysaccharide is composed of repeating units of fucosylated Lewis (Le) antigens. The alpha(1,2)-fucosyltransferase (futC) of H. pylori, which catalyzes the conversion of Le(x) to Le(y) by addition of fucose, is subject to slipped-strand mispairing involving a homonucleotide (poly-C) tract. To explore the distribution of Le phenotypes within H. pylori cells grown in vitro, 379 single colonies of strain J166 were examined for Le expression. Two major populations with reciprocal Le(x)/Le(y) phenotypes were identified. Phenotypes correlated with futC frame status, suggesting that strain J166 represents a mixed population with respect to futC poly-C tract length, which was confirmed by a translational reporter. After hundreds of generations in vitro, phenotypes did not change significantly, indicating that the observed J166 Le diversity reflects the founding population. Since slipped-strand mispairing in the futC poly-C tract was postulated to explain the Le(y) phenotypic change observed in J166 derivative strain 98-169 isolated 10 months after rhesus monkey challenge, in trans complementation with in-frame futC was performed. Le(y) synthesis was restored and Le(x) expression was reciprocally lowered. From these studies, we confirmed the principal role of futC slipped-strand mispairing in Le antigenic variation in vitro and in vivo.
