The review contains information on modern approaches to the development of antituberculosis vaccines and remedies. Data on the comparative effectiveness of different subunit and DNA vaccines against tuberculosis are presented. The use of comparative and structural genomics for the search and characterization of new Mycobacterium tuberculosis genes, whose products may prove to be important antigens for the development of vaccines or target proteins for remedies against tuberculosis, is considered.  相似文献

Pathogenic Candida species remain a significant medical problem despite the availability of antifungal therapies. Two key issues must be addressed to improve the treatment of life-threatening systemic Candida infections. First, advanced diagnostic tools are required to facilitate the early identification of these infections, when therapeutic intervention is more likely to be effective. Second, improved antifungal therapies are needed. These therapies, which might include combinations of antifungals, need to be less toxic to the patient and more potent in killing a broader range of Candida species. Recent advances in unravelling the genomics of these species should facilitate efforts to achieve these goals. We discuss the contribution of genomics to the development of novel antifungals and new diagnostic tools.  相似文献

Herein we review theoretical and methodological considerations important for finding and delimiting cryptic species of parasites (species that are difficult to recognize using traditional systematic methods). Applications of molecular data in empirical investigations of cryptic species are discussed from an historical perspective, and we evaluate advantages and disadvantages of approaches that have been used to date. Developments concerning the theory and practice of species delimitation are emphasized because theory is critical to interpretation of data. The advantages and disadvantages of different molecular methodologies, including the number and kind of loci, are discussed relative to tree-based approaches for detecting and delimiting cryptic species. We conclude by discussing some implications that cryptic species have for research programmes in parasitology, emphasizing that careful attention to the theory and operational practices involved in finding, delimiting, and describing new species (including cryptic species) is essential, not only for fully characterizing parasite biodiversity and broader aspects of comparative biology such as systematics, evolution, ecology and biogeography, but to applied research efforts that strive to improve development and understanding of epidemiology, diagnostics, control and potential eradication of parasitic diseases.  相似文献

Vector systematics research is being transformed by the recent development of theoretical, experimental and analytical methods, as well as conceptual insights into speciation and reconstruction of evolutionary history. We review this progress using examples from the mosquito genus Anopheles. The conclusion is that recent progress, particularly in the development of better tools for understanding evolutionary history, makes systematics much more informative for vector control purposes, and has increasing potential to inform and improve targeted vector control programmes.  相似文献

Although progress in the development of an antimalarial vaccine has been mostly obtained through the study of P. falciparum, significant advances have recently been made in the study of P. vivax, the other major human malarial parasite. Antigens which have been shown to be important in P. falciparum have been characterized and in some cases cloned in P. vivax. Other studies have examined some of the more specific biological characteristics of P. vivax. Among these are studies on components present in caveolae-vesicle complexes of the infected erythrocyte, on the occurrence of delayed hepatic development leading to relapse, or on the Duffy erythrocyte antigen as a key receptor for parasite invasion. Although progress has been made in the short-term in vitro maintenance of P. vivax, the inability to maintain the parasite in continuous culture led to the investigation of wild parasite populations in patients; occurrence of extensive antigenic and karyotype polymorphism was detected in this way, as was a double-blocking and enhancing activity of human antibodies on parasite development in the vector. The association of monoclonal antibodies with DNA recombinant technology allowed the characterization of a number of P. vivax antigens to be made. Among these, an antigen shared between sexual and asexual stages was shown to constitute a target for transmission-blocking immunity. The cloning of an antigen involved in transmission-blocking immunity, along with that of the surface antigen of the sporozite (CSP) and of a major surface antigen of the invasive merozoite (PV200) constitutes a significant step towards the development of a multivalent recombinant vaccine against P. vivax.  相似文献

Due to social demands with regard to sustainability and ecological considerations, an aim of current research in short-rotation coppices (SRC) is to find effective candidates for biological control of pest species. The great red poplar leaf beetle, Chrysomela populi L. (Coleoptera: Chrysomelidae), is one of the most harmful pests in the SRC. The endoparasitic mite Linobia coccinellae (Scopoli) (Acari: Astigmata, Hemisarcoptidae) lives under the elytra of C. populi and feeds on the beetles' hemolymph. The population dynamics of the mites and their impact on the fecundity and longevity of female beetles were examined in two experiments. To study the occurrence of the mites, beetles were collected from two poplar SRC sites in Germany on three dates. Infestation rates of collected male and female C. populi individuals increased over the course of the year and reached 78–97% in the last survey. Females were parasitized more often than males at any time. For the second part of the study, beetles were reared under laboratory conditions to study the development of the mites and their impact on the hosts. For this purpose, half of the females were actively seeded with five adult females of L. coccinellae and the other half was used as a control. There was no significant impact on longevity or reproductive traits of the beetles. Despite the high rates of infestation, it appears that the mites do not have a serious effect on female beetles. Thus L. coccinellae, by itself, is not an effective biological control agent against C. populi. However, it remains to be investigated whether the mites increase the beetles' susceptibility to other biotic and abiotic factors, and so may contribute to an integrated pest management regime with multiple antagonists.  相似文献

Within a single organism, numerous parasites often compete for space and resources. This competition, together with a parasite’s ability to locate and successfully establish in a host, can contribute to the distribution and prevalence of parasites. Coinfection with trematodes in snail intermediate hosts is rarely observed in nature, partly due to varying competitive abilities among parasite taxa. Using a freshwater snail host (Biomphalaria glabrata), we studied the ability of a competitively dominant trematode, Echinostoma caproni, to establish and reproduce in a host previously infected with a less competitive trematode species, Schistosoma mansoni. Snails were exposed to S. mansoni and co-exposed to E. caproni either simultaneously or 1 week, 4 weeks, or 6 weeks post S. mansoni exposure. Over the course of infection, we monitored the competitive success of the dominant trematode through infection prevalence, parasite development time, and parasite reproductive output. Infection prevalence of E. caproni did not differ among co-exposed groups or between co-exposed and single exposed groups. However, E. caproni infections in co-exposed hosts took longer to reach maturity when the timing between co-exposures increased. All co-exposed groups had higher E. caproni reproductive output than single exposures. We show that although timing of co-exposure affects the development time of parasite transmission stages, it is not important for successful establishment. Additionally, co-exposure, but not priority effects, increases the reproductive output of the dominant parasite.  相似文献

Molecular variation is widespread in parasite populations, and its analysis has important implications for studying aspects relating to the function and organisation of genes, and the taxonomy, phylogeny and population genetics of parasites. This article reviews some PCR-based mutation scanning techniques that have advantages over currently used DNA methods for the analysis of genetic variation in parasites. The review is technical and describes briefly the principles of relevant techniques, examines some of their advantages and disadvantages and gives several examples for possible applications.  相似文献

The behaviour of boron (B) and its fate in the environment remains in many respects obscure. The major B sources and fluxes seem to have been identified, although there are many uncertainties about the magnitude and importance of each; published evaluations vary greatly. Little is known about B speciation of living matter-bearing formations, i.e. soils, natural waters, and sediments. Many authors suppose that B in nature occurs only as oxo-compounds, although, natural inorganic and organic B compounds and complexes have been described. Moreover, the most common approach to B biogeochemistry in terms of inorganic B compounds (as boric acid and borates) seems to oversimplify the matter. Data on the physiological functions of B are fragmentary and often contradictory. Some data exists on endemic symptoms and diseases among humans and grazing cattle that are supposedly triggered by B abundance in the natural environment. Unfortunately, the published data are scarce and vague. In this paper, we review the up-to date references on B sources, turnover, and speciation in the environment and the element potential impact on health.  相似文献

Recent efforts of researchers to elucidate the molecular mechanisms of biological systems have been revolutionized greatly with the use of high throughput and cost-effective techniques such as next generation sequencing (NGS). Application of NGS to microbial genomics is not just limited to predict the prevalence of microorganisms in food samples but also to elucidate the molecular basis of how microorganisms respond to different food-associated conditions, which in turn offers tremendous opportunities to predict and control the growth and survival of desirable or undesirable microorganisms in food. Concurrently, NGS has facilitated the development of new genome-assisted approaches for correlating genotype and phenotype. The aim of this review is to provide a snapshot of the various possibilities that these new technologies are opening up in area of food microbiology, focusing the discussion mainly on lactic acid bacteria and yeasts associated with fermented food. The contribution of NGS to a system level understanding of food microorganisms is also discussed.  相似文献

The implications of parasite regulation of host endocrinology for successful biological control have not been fully appreciated. Insect parasites regulate host metamorphosis in a number of different ways. For a given host (pest) situation, each form of host regulation has its own advantages and disadvantages. Careful selection of a parasite based upon its mode lf host regulation can enhance the potential for biological control success. A basic knowledge of the endocrine basis for parasite regulation of its host will enable prediction of whether a parasite can regulate, and survive in, a host to which it has not been previously exposed.  相似文献

This article discusses different methods for the recycling of chiral catalysts, including heterogenization of the soluble catalyst on an insoluble inorganic or organic support, membrane filtration of homogeneously soluble catalysts, precipitation, and two-phase systems. In principle, all the methods presented enable the repeated use of a chiral catalyst without loss of activity and/or enantioselectivity. Examples will be given from laboratory and industrial processes, incuding hydrogenations, ketone reductions, epoxidations, dihydroxylations, diethylzinc additions and Diels-Alder reactions catalyzed by chemocatalysts or biocatalysts. Different approaches for cyanation, hydrogenation and epoxidation are compared. Data from industrial processes include the production of metalochlor, the production of (-)-menthol and the production of L-tert-leucine.  相似文献

Cluster-flowering camellias are among the most promising types of camellia due to both their ornamental characteristics and their resistance capacity, especially to heat and alkaline soils. Because of global climate change, researchers at the Shanghai Botanical Garden are attempting to identify more heat tolerant plants; camellia is such a plant, and it has been promoted for more than 20 years. Morphological and physiological approaches were used in this study to evaluate the heat tolerance of two new cultivars, and new methods were also explored. The results indicate that the heat tolerance of the new cultivar Camellia hybrid ‘Pink Cascade’ is higher than that of Camellia hybrid ‘Sweet Gem’. In addition, infrared thermography can further be used to evaluate the heat resistance of plants in combination with their stomatal and photosynthetic characteristics.  相似文献

The present study investigates the isotopic composition of anthropogenic boron (B) and its potential affects on the environment. The isotopic ratios of B in synthetic products from the main ores in the world have been measured by negative thermal ionization mass spectrometry. The data show that the isotopic compositions of Na-borate products and washing powders overlap with those of natural Na-borate minerals. In contrast, the11B/10B ratios of synthetic Ca-borate and Na/Ca borate products are significantly lower (by 15 permil) and overlap with those of the natural Ca-borate minerals. Consequently, the original isotopic signature of natural borate minerals is not modified during the manufacturing process of synthetic products. The B isotopic composition of domestic wastewater from Israel and Riverside, California suggests that B in sewage is derived from Na-borate components used in detergents. Since B, like other inorganic ions, is not removed during conventional sewage treatment, it accumulates in domestic wastewater. Although the B concentration in pristine groundwaters is generally low (<0.05 mg/L), contaminant sources (e.g., wastewater) are relatively enriched in B (0.5–1 mg/L). The isotopically distinguished signature of borate compounds is used to trace groundwater contamination.  相似文献
