The self-directing optimization technique was employed to determine the optimal conditions for -1,3-glucanase production by Trichoderma harzianum in batch culture. A maximum -1,3-glucanase production of 0.910 U was obtained at a pH (controlled) of 4.9, an aeration rate of 0.9 1/(1)(min), and an agitator speed of 220 rev/min which were found to be the most suitable.  相似文献

Response surface methods were used to determine the optimum concentration of medium nutrients for extracellular -1,3-glucanase production by Trichoderma harzianum in shakeflask culture. A Plackett-Burman design was used to screen the important variables which were then grouped and studied by a central composite design. Studies on the constitutive β-1,3-glucanase were conducted in submerged as well as in the stationary culture conditions, in the presence and in the absence of lactose and glucose as main carbon sources. In the absence of lactose or glucose, expression of β-1,3-glucanase was observed at 96?h in extracellular, periplasmic, cell wall bound and internal fractions during submerged fermentation. In shake flask culture, enzyme was found in all subcellular fractions using optimal glucose concentration. When Trichoderma harzianum was grown on media containing 55?kg lactose/m3 in submerged culture, activity was found in extracellular, cell wall bound and in the periplasmic fractions. The relative distribution of the enzyme in the cell is independent of the nature of the carbon source and its concentration.  相似文献

Trichoderma harzianum, a soil-borne filamentous fungus, is capable of parasitizing several plant pathogenic fungi. Secretion of lytic enzymes, mainly glucanases and chitinases, is considered the most crucial step of the mycoparasitic process. The lytic enzymes degrade the cell walls of the pathogenic fungi, enabling Trichoderma to utilize both their cell walls and cellular contents for nutrition. We have purified a 110 kDa novel extracellular β-1,3-exoglucanase from T. harzianum, grown with laminarin or in dual cultures with host fungi. The corresponding gene, lam1.3, and its cDNA were isolated and their nucleotide sequences determined. The deduced amino-acid sequence predicted a molecular mass of 110.7 kDa of a mature protein excluding a signal peptide. LAM1.3 showed high homology to EXG1, a β-1,3-exoglucanase of the phytopathogenic fungus Cochliobolus carbonum, and a lower homology to BGN13.1, a β-1,3-endoglucanase isolated from T. harzianum. However, it contains a unique C-terminal embodying cysteine motifs. The expression of lam1.3 in growth with laminarin, but not with glucose, was found to be a result of differential accumulation of the corresponding mRNA.  相似文献

Uracil, acting as a precursor of UDP-glucose, served as an activator on the production of curdlan with Agrobacterium sp. (ATCC 31750). The time of adding uracil was important to improve curdlan production. When uracil was added after ammonium depletion (at 26 h), it was used as a nitrogen source for cell growth. Although the cell concentration increased, the curdlan production was decreased. If uracil was added at 46 h, then uracil was degraded slowly but still activated curdlan production. With the addition of both sucrose (200 g) and uracil (1.5 g), the curdlan production was increased up to 93 g l–1 after 160 h fermentation.  相似文献

Trichoderma species have become the important means of biological control for fungal diseases. This research was carried on to access the high β-1,3-glucanase and β-1,4-glucanase enzyme producer of Trichoderma species isolates using two different carbon sources for finding a method to obtain more concentrate culture filtrates. Therefore, 14 Trichoderma isolates belonging to species: Trichoderma ceramicum, T. virens, T. pseudokoningii, T. koningii, T. koningiosis, T. atroviridae, T. viridescens, T. asperellum, T. harzianum1, T. orientalis, T. harzianum2, T. brevicompactum, T. viride and T. spirale were cultured in Wiendling’s liquid medium plus 0.5% glycerol or 0.5% Phytophthora sojae-hyphe as the carbon source in shaking and non-shaking (stagnant) statuses. Enzyme activity rate and total protein were evaluated in raw, acetony and lyophilized concentrated culture filtrates and the specific enzyme activity of β-1,3-glucanase and β-1,4-glucanase were measured by milligramme glucose equivalent released per minute per milligramme total protein in culture filtrates. The results showed that using Phytophthora – hyphe in medium increased the enzyme activities as compared to glycerol at all Trichoderma species which suggested that these substrates can also act as inducer for synthesis of lytic enzymes, in addition the most enzymes activity was observed in the lyophilised concentrated culture filtrate. The most successful species in β-1,3-glucanase and β-1,4-glucanase enzymes activities were T. brevicompactum and T. virens and these species can be used for mass production of these enzymes which are supposed to be used in commercial formulation and also will be able to control P. sojae directly.  相似文献

Summary An Aspergillus sp. was isolated which secreted high levels of -glucosidase in growth medium. The maximum activity(10 IU/ml of -glucosidase and 22.6 IU/ml of cellobiase) was obtained in cellulose medium supplemented with wheat bran. Summary Examination of the relationship between the rate of oxygen transfer and the rate of polymer production revealed an unexpectedly high requirement for oxygen. At a cell density of about 3 g (dry wt)/L, the threshhold value for OTR for optimal synthesis of polymer is about 50 mmoles O2/L.hr. Whereas Rushton turbines are effecient at transfering oxygen to solution, their use reduces the quality of the recovered polymer. Although better quality polymer can be produced in a reactor employing an agitator which causes less shear stress, the productivity can be compromised due to the inefficiency in OTR. The present study describes operating conditions for the provision of sufficient OTR in a system compatible with the production of high-quality polymer whereby turbine impellers were replaced with a marine-type propeller and mass transfer was assisted by means of a gas dispersion device.  相似文献

The design of scaffolds for solubilizing/dispersing poorly water-soluble bioactive molecules in neutral aqueous media is a major challenge of functional food, pharmaceuticals, and cosmetics development, as highlighted by the plethora of corresponding solubilization/dispersion strategies. Herein, renatured β-1,3-1,6-glucan (r-glucan) nanoparticles prepared by neutralization of alkali-denatured β-1,3-1,6-glucan and subsequent centrifugation are used as a host to disperse water-insoluble bioactive molecules (curcumin, all-trans-retinoic acid, and rebamipide) by simple mixing of host and guest solutions. Curcumin in the r-glucan cavity is found to be stacked in the form of J-aggregates and twisted along the helix, and is demonstrated to be retained for significantly longer than curcumin in the corresponding γ-cyclodextrin (γ-CD) complex. Specifically, curcumin incorporated in γ-CD is released within 5.5 hours, whereas that in the r-glucan complex is released very slowly, with 12% of curcumin in the latter complex retained after 31-day incubation at 37°C. Thus, inclusion protocol simplicity and slow release ability make r-glucan nanoparticles a potential carrier scaffold for various applications.  相似文献

β-1,3-Glucan (curdlan) is a water-insoluble polysaccharide composed exclusively of β-1,3 linked glucose residues. Extracellular curdlan was mostly synthesized byAgrobacterium species andAlcaligenes faecalis under nitrogen-limiting conditions. In this study, we screened the microorganisms capable of producing extracellular curdlan from soil samples. For the first time, we reported Gram-positive bacteriumBacillus sp. SNC 107 capable of producing extracellular curdlan in appreciable amounts. The effect of different carbon sources on curdlan production was studied and found that the yield of curdlan was more when glucose was used as carbon source. It was also found that maximum production was achieved when the initial concentration of ammonium and phosphate in the medium was 0.5 and 1.9 g/L respectively. In this study the curdlan production was increased from 3 to 7 g/L in shake flask cultures.  相似文献

Background

Unsatisfactory surgical handiness is a commonly known disadvantage of implantable granular bioceramics. To overcome this problem, β-1,3-glucan, biotechnologically derived polysaccharide, has been proposed as a joining agent to combine granular ceramics into novel compact and elastic composite. Hydroxyapatite/glucan elastic material was processed and evaluated as a potential bone void filler.

Methodology

The procedure of composite formation was based on gelling properties of glucan. Its properties were studied using X-ray microtomography, SEM-EDS, FTIR spectroscopy, compression test and ultrasonic method. Sorption index was determined in phosphate buffered saline; bioactivity in simulated body fluid; sterility in growth broth and human blood plasma; implantation procedure in dog model.

Results

HAp/glucan composite is sterilizable, flexible and self-adapting to defect shape. It exhibits bioactivity, good surgical handiness, high sorption index and profitable mechanical properties, resembling those of spongy bone. Results of pilot clinical experiment on animal (dog) patients of a local clinic of animal surgery suggested good healing properties of the composite and its transformation into new bone tissue within critical-size defect.

Conclusions

The results obtained in this study confirm that flexible HAp/glucan composite has potential as a bone-substituting material. Promising results of pilot clinical experiment suggest that further in vivo experiments should be performed.  相似文献

Surface and submerged culture studies were carried out for the production of extracellular -1,3-glucanase and carboxymethylcellulase from Trichoderma harzianum, NCIM 1185. The different parameters which influence the enzyme production, viz., spore age in the slant growth, seed age, and pH (initial) of the production medium have been optimized. The ranges of these parameters studied were: Spore age of T. harzianum between 2 and 10 days, seed age between 12 h and 48 h, and pH (initial) between 3 and 7. The other conditions of fermentation were: Temperature 30°C, gyratory shaker speed 160 rev. min–1. It was observed that the activities of -1,3-glucanase and carboxymethylcellulase varied during the entire fermentation cycle which had a distinct effect on protoplast generation from T. reesei mycelium. The optimum values of the above-mentioned parameters for lytic enzymes production were: Spore age of T. harzianum in the slant growth 5 days, seed age 36 h, and pH (initial) 5.0 for both surface and submerged culture processes.Mr. S. The fragmentation of the biologically active 1,3;1,6-beta-D-glucan Antivir by endo-1,3-beta-D-glucanase LIV from crystalline styles of the marine mollusk Spisula sachalinensis was carried out. It was found that low molecular mass oligomers possessing a stabilizing effect on membranes and anti-viral activity against tobacco mosaic virus appeared in the process of enzymatic hydrolysis of Antivir. Biological activity of 1,3;1,6-beta-D-glucooligo- and polysaccharides was found to be associated with molecular mass (polymerization degree (n) not less than 14) and with presence of intralinked beta-1,6-connected monosaccharide residues. Probably, decrease in molecular mass is compensated by increase in number of intralinked beta-1,6-connected monosaccharide residues.  相似文献

From the dikaryotic mycelium ofSchizophyllum commune ATCC 38548 several monokaryotic strains were obtained by isolating the two types of monokaryotic protoplasts and their reversion to hyphal growth. The dikaryoticS. commune ATCC 38548 produced about 10 g/liter of extracellular β-1,3-/β-1,6-glucan (schizophyllan) after 96 h of growth, while the monokaryons excreted much less of this polysaccharide. During growth of strains of both types of monokaryons indigo and β-1,3-glucanase activities were excreted. Two selected monokaryons were mated with other monokaryoticS. commune strains and some of the dikaryotic mycelia obtained produced about 12 g/liter of extracellular β-1,3-/β-1,6-glucan after 120 h of cultivation.  相似文献

6-pentyl-alpha-pyrone (6PP) production by Trichoderma harzianum, in an extractive fermentation system, was elicitated by Rhizoctonia solani. The extent of 6PP elicitation was related to the state of Rhizoctonia and to the Trichoderma inoculum type. The use of non-viable Rhizoctonia solani mycelium in mycelium-inoculated Trichoderma harzianum culture, yielded the maximal 6PP production (474 mg l(-1)) compared to control cultures (147 mg l(-1)) and decreased the process time from 192 to 96 h.  相似文献

Trichoderma species are readily isolated from Brazilian cerrado soil by conventional methods and some of them were characterized as Trichoderma koningii. The effect of carbon source on the production of β-1,3-glucanases in the culture filtrates of a specific Trichoderma koningii strain (ALL 13) was investigated. Enzyme activity was detected in all carbon sources tested and only one band of β-1,3-glucanase was detected in non-denaturing PAGE. This enzyme was purified by Sephacryl S-200 gel filtration and Phenyl Sepharose CL 4B chromatography. A typical procedure provided 105-fold purification with 13.4% yield. The molecular weight of the purified enzyme was 75 kDa as estimated by SDS-PAGE. The enzyme hydrolyzed laminarin in an endo-like fashion to form small oligosaccharides and glucose. The Km and Vmax values for β-1,3-glucanase, using laminarin as substrate, were 0.148 mg.mL−1 and 0.159 U.min−1, respectively. The pH optimum for the enzyme was pH 4.6 and maximum activity was obtained at 50°C. Hg2+ inhibited the purified enzyme.  相似文献
