Using whole cottonseed as a lipid source, silage-based diets that were isocaloric and isonitrogenous yet varied in lipid level were fed to multiparous cows. In Experiment 1, 48 cows (n = 12 per treatment) were allotted to 1 of 4 treatments where diets were formulated to supply 3.9, 4.3, 5.3 and 6.3% of total lipid. In Experiment 2, 66 cows (n = 22 per treatment) were allotted to 1 of 3 treatments where diets were formulated to supply 3.1, 5.5 and 8.3% of total lipid. Length of the first ovarian cycle, length of the first normal estrous cycle, postpartum intervals to onset of ovarian luteal activity and to first estrus were not affected by diet (P>0.10) in either experiment. Mean progesterone (P(4)) concentrations for first normal estrous cycles were not different (P>0.10) in either experiment. Anestrous periods were divided into 3 phases for analyses: Phase I) parturition to onset of ovarian luteal activity, Phase II) first ovarian luteal activity and Phase III) first normal estrous cycle. No differences were observed in P(4) concentrations during any phase of the postpartum period. In conclusion, isocaloric and isonitrogenous diets with increasing levels of lipid had no effect on reproductive performance in suckled beef in these experiments. 相似文献

The effect of melatonin treatment on intervals from calving to first postpartum estrus and ovulation was determined in Shorthorn cows which calved May 8 to June 14. Melatonin (500 mg in beef tallow) was injected subcutaneously (s.c.) into 20 cows on June 15 (4 to 38 d postpartum). Ovulation was determined from progesterone concentrations in jugular venous blood collected weekly from June to August. Mean intervals to first estrus and first ovulation were significantly longer in primiparous than in multiparous cows (85 +/- 4 vs 55 +/- 3 d and 83 +/- 4 vs 52 +/-3 d). Melatonin treatment caused a significant increase in the intervals to first postpartum estrus (68 +/- 4 vs 58 +/- 5d) and ovulation (68 +/- 4 vs 55 +/- 5 d). Mean plasma melatonin concentrations during the daytime were significantly higher in treated than in control cows one and two weeks after melatonin injection and were within the lower range of nighttime values reported previously for cows. Thus melatonin treatment raised daytime plasma concentrations of melatonin and delayed the onset of estrus and ovulation. These results support the possibility of a role of photoperiod through melatonin secretion in the onset of postpartum ovarian activity in cattle. 相似文献

The effects of purified human interferons IFN-α and IFN-β on immunocompetent cells were compared. Both types of IFN inhibited blast transformation to the T-cell-dependent mitogens phytohemagglutinin, concanavalin A and pokeweed mitogen with IFN-α being more potent than IFN-β. B-Cell mitogensis induced by Staphylococcus Cowan I was enhanced by both IFN preparations with IFN-β being more potent than IFN-α. Both types of IFN enhanced natural killer cell activity with differences in the dose-response curves. The expression of the IFN-induced modulation of the mitogenic responses was shown to be associated with human chromosome 21. No such association could be demonstrated for the effect of IFN on natural killer cell activity. 相似文献

Effect of prolonged maternal fasting on the fetal liver and heart glycogen and triglyceride content and on concentration of glucose, urea, uric acid and alpha amino-nitrogen in the amniotic fluid has been studied in rats. The animals were divided into four groups: fed (control), fasted for one day (from 20 to 21 day of pregnancy), fasted for two days (from 19 to 21 day) and fasted for three days (from 18 to 21 day). Maternal fasting for two and three days resulted in reduction in fetal growth. The fetal liver glycogen content was reduced already after one day of fasting, stabilized after two days and then further decreased after three days. The fetal heart glycogen content was reduced only after three days of fasting. The fetal liver triglyceride content increased gradually during the first two days of fasting and then stabilized. The content of triglycerides in the heart was elevated after two and three days of food deprivation. The amniotic fluid glucose concentration decreased after one day of fasting and then stabilized. Fasting did not effect the concentration of the nitrogenous compounds in the amniotic fluid. It is concluded that maternal fasting affects markedly metabolism of energy substrates stored in the fetal liver and the heart and the composition of the amniotic fluid. 相似文献

Protection against Brucella abortus induced abortion and infection provided by strain 19 (S19) vaccination was evaluated in American bison (Bison bison). Forty-eight pregnant bison were manually inoculated (MI) with S19 vaccine, 44 were ballistically inoculated (BI) with an absorbable hollow pellet containing lyophilized S19, and 46 were manually injected with buffered saline as non-vaccinated controls (NVC). All bison were Brucella spp. seronegative prior to the experiment, in the second trimester of pregnancy, and were randomly assigned to experimental groups. Approximately 60 days post-vaccination, abortions were observed in the vaccinated bison. Brucella abortus strain 19 was recovered from a bison that had recently aborted, her fetus, and from 11 of 12 other aborted fetuses. Fifty-eight percent (53 of 92) of vaccinated bison aborted, and no abortions were observed in the NVC bison. One cow aborted during her second post-vaccinal pregnancy and S19 was identified from the dam and fetus indicating that chronic S19 infections can occur in bison. Positive antibody titers were present 10 mo post-vaccination in 73% (66 of 91) of the bison. Thirteen mo post-vaccination, 30 MI vaccinates, 27 BI vaccinates, and 30 NVC bison were challenged during the second trimester of pregnancy with 1 x 10(7) CFU of B. abortus strain 2308 via bilateral conjunctival inoculation. Protection against abortion was 67% (P less than or equal to 0.0001) for vaccinated bison compared to 4% in NVC. Protection against B. abortus infection was determined to be 39% (P greater than or equal to 0.001) for vaccinates and 0% (zero of 30) for NCV. Persistent antibody titers, vaccine induced abortions, and chronic S19 infections indicate that the S19 vaccine doses used in this study are not suitable for pregnant bison. 相似文献

Potassium iodide (KI), the specific treatment for sporotrichosis, apparently does not have a direct action on Sporothrix schenckii. The spontaneous healing and the variability of the clinical presentation in the disease have strengthened the idea that the KI rather interacts with the immune response of the host. The phagocytic process is inefficient in individuals with sporotrichosis in whom the microbicidal mechanism of halogenation fails to control the disease. There is evidence that blocking of free radicals decreases in the presence of KI. Humoral and cellular immunity are present in sporotrichosis but its participation is uncertain; it is yet to be determined if in this mycosis the KI influences other processes or factors of immune response. 相似文献

Prolonged postpartum acyclicity in suckled beef cows is a source of economic loss to beef cattle producers. Duration of postpartum acyclicity is influenced by suckling status, nutritional status, calving season, age, and several other factors. Although uterine involution begins and ovarian follicular waves resume soon after parturition, dominant follicles of these waves fail to ovulate, due to a failure to undergo terminal maturation. As a result, postpartum anovulatory dominant follicles are smaller than the ovulatory follicles in cyclic cows. Failure of postpartum dominant follicles to undergo terminal maturation is due to absence of appropriate LH pulses, a prerequisite for follicular terminal maturation prior to ovulation. Absence of LH pulses early post partum is primarily due to depletion of anterior pituitary LH stores, although GnRH pulses are also absent during this period due to suckling. Following replenishment of LH stores between Days 15 and 30 post partum, absence of LH pulses is due to continued sensitivity of the hypothalamic GnRH pulse-generator to the negative feedback effect of ovarian estradiol-17beta, which results in absence of GnRH pulses. This negative feedback effect of estradiol-17beta is modulated by suckling which stimulates release of endogenous opioid peptides from the hypothalamus. As the postpartum interval increases, sensitivity of the GnRH pulse-generator to the negative feedback effect of ovarian estradiol-17beta decreases. This is followed by an increasing frequency of GnRH discharges and LH pulses, terminal follicular maturation, ovulation, and continued cyclicity. The first ovulation post partum is usually followed by a short cycle due to premature luteolysis because of premature release of PGF2alpha from the uterine endometrium, which is possibly intensified by the suckling-induced oxytocin release from the posterior pituitary. This study was designed to evaluate in suckling early pregnant beef cows with and without eCG-pre-stimulation: (i) the influence of day gestation (from 40 to 101 days) and the consecutive eCG treatments on the follicular growth induced by means of ultrasound-guided transvaginal follicle ablation (FA; all follicles ≥ 5 mm) and the number and quality oocytes recovered by ovum pick-up (OPU) and (ii) the possible effects of repeated hormonal stimulation and FA/OPU on pregnancy outcome. Twelve suckling early pregnant Angus cows (40 days post fixed-time artificial insemination) were randomly assigned to each of two groups (n=6 group(-1)). Group 1 treatments included: FA (Day 0), eCG (1600 IU; Day 1) and OPU (Day 5). Group 2: as cited Group 1 with no eCG treatment. In both groups, OPU was repeated five times (Days 45, 59, 73, 87 and 101 of gestation). The numbers (mean ± SEM) of class II (5-9 mm; 4.3 ± 0.9) and class III (≥10 mm; 2.5 ± 0.4) follicles visualized per cow per OPU session in eCG-treated cows were greater (P<0.05) than for non-treated cows (0.9 ± 0.1 and 0.9 ± 0.1, respectively). In contrast, the number (mean ± SEM) of class I (<5mm) follicles per cow per OPU session was lower for cows with eCG treatment (2.8 ± 0.4) than for non-treated cows (5.7 ± 0.5). The mean number of aspirated follicles was not significantly different (P<0.05) between eCG-treated cows and non-treated cows at 45 and 59 days of pregnancy. However, the mean number of aspirated follicles was greater (P=0.03) in eCG-treated cows than non-treated cows from 73 day of pregnancy onwards. The numbers (mean ± SEM) of recovered oocytes and viable oocytes/cow/session were greater (P<0.05) for eCG-treated cows (2.2 ± 0.2 and 1.6 ± 0.4, respectively) than for non-treated cows (1.0 ± 0.2 and 0.9 ± 0.2, respectively). No donor pregnancies were lost either during or following OPU procedure. We can conclude that (1) eCG-treated pregnant suckled cows can be a source of oocytes for IVF at least to 100 days of gestation and (2) repeated FA/eCG treatment/OPU procedures did not affect the pregnancy outcome. 相似文献

In three experiments, we examined endogenous opioid inhibition of luteinizing hormone (LH) secretion during the bovine estrous cycle. An increase in serum LH in response to the opioid antagonist naloxone (Na; 1 mg/kg i.v.) was the criterion for opioid inhibition. Estrous cycles were synchronized via prostaglandin administration. In Experiment 1, mean serum LH was not different during the luteal phase in yearling heifers (n = 6/group) at Hour 1 after Nal (2.1 ng/ml) compared to controls (1.8 ng/ml). However, LH peak amplitude was increased (p less than 0.05) in the Nal compared to the control group. Serum LH was increased (p less than 0.01) during the follicular phase in heifers at Hour 1 post-Nal compared to controls (4.7 and 3.5 ng/ml, respectively). Again, Nal administration was followed by increased (p less than 0.05) LH pulse amplitude compared to control. In Experiment 2, no effect of Nal upon serum LH was detected in cows (n = 9) during proestrus, metestrus, midluteal and late luteal portions of the estrous cycle. In Experiment 3, the LH response to Nal was examined simultaneously in yearling heifers and cows (n = 5/group) during the luteal and follicular phases. Serum LH increased (p less than 0.001) during Hour 1 post-Nal in heifers compared to cows during the follicular (3.4 vs. 1.7 ng/ml) but not during the luteal phase. Thirty-two postpartum (PP) cows were used to investigate the effect of suckling on secretion of luteinizing hormone (LH). Calves remained with their dams (suckled; S), or they were removed within 24 h of birth (nonsuckled; NS). To evaluate the relationship between suckling and negative feedback regulation of LH, cows were ovariectomized on Day 5 PP, then injected intravenously with estradiol-17 beta (E) or vehicle (V) on Day 10 PP. To investigate the influence of suckling on the gonadotropin-releasing hormone (GnRH)-induced release of LH, cows were injected with 80 micrograms of GnRH on a single day varying from 18 to 85 days PP. Suckling inhibited the postcastration rise in LH, as LH concentrations increased at a faster rate in NS compared with S cows [0.031 +/- 0.02 ng/(ml X day) LH: P less than 0.05]; this was not influenced by basal amounts of E since amounts did not differ between S and NS cows at ovariectomy (5.37 +/- 0.36 vs. 5.34 +/- 0.48 pg/ml E; P greater than 0.05). Serum concentrations of LH were negatively related to total follicular E only in S cows (r = -0.71; P less than 0.01). Estradiol-17 beta caused a decrease not only in the level but also the variability in LH concentrations in both S and NS cows: LH in S cows was less variable after E than in NS cows (P less than 0.001), but the magnitude of LH suppression was not influenced by suckling (P greater than 0.25). The regression of LH response on days PP was essentially the same over time for both S (P greater than 0.25) and NS (P greater than 0.25) cows, indicating that LH response to a GnRH injection was not influenced by suckling or days PP. Suckled cows had a tendency to release more LH relative to their baseline in response to GnRH as time PP increased (P less than 0.10), but NS cows did not. These results indicate that even though ovarian secretions inhibit LH release from the pituitary, other inhibitory influences may have a major effect in S cows. Concentrations of LH were lower in S cows than NS cows on Day 10 PP, following removal of the ovaries on Day 5, suggesting that suckling had a direct effect on the hypothalamic-pituitary axis. 相似文献

To study whether dietary restriction (DR; 70% of ad lib. feeding)-elicited immunosuppression results from the trade-off between the costs of mounting an immune response and the metabolic costs of maintenance, we subjected mice from two divergent lines selected for high basal metabolic rate (H-BMR) and low BMR (L-BMR) to 4 wk of DR and then challenged them with keyhole limpet hemocyanin (KLH) antigen. Those line types differ genetically with respect to BMR and to the mass of metabolically expensive internal organs, which are larger in H-BMR mice. In mice of both line types, DR resulted in a significant reduction of body mass, an immune response, and the downsizing of spleen, lymph nodes, thymus, heart, and kidneys but not small intestines. DR resulted in a greater reduction of the spleen and lymph nodes in mice of the H-BMR line type, whereas the thymus was more affected in L-BMR line type. In contrast, immunization resulted in an increase of liver mass in DR mice of both line types. A comparison of the results of current and earlier studies on the same mouse line types suggests that metabolic trade-offs involving the costs of an immune response are more apparent when animals are forced to increase energy demands (e.g., by cold exposure) compared to when energy demands are decreased through DR. Our findings also suggest that divelrgent selection on BMR resulted in between-line-type differences in T-cell- and B-cell-mediated types of an immune response. More generally, our results indicate that production of a wide repertoire of antibodies is not correlated with high BMR. 相似文献
