新疆北疆棉田立枯丝核菌菌丝融合群及其营养亲和群研究   总被引:1,自引:0,他引:1  
从新疆北疆棉区采集了典型的棉花立枯病病苗及棉田土标样686份,按常规分离方法分离得到399个分离物,从中鉴定出272个纯化的立枯丝核菌(Rhizotonia solaniKühn)菌株,经玻片吉姆萨氏染剂(Gimsa′sstain)染色程序观察细胞核数目,全部测试菌株均属于多核。用标准菌株,通过载玻片菌丝融合实验测定,将纯化的272个菌株划归为3个菌丝融合群:AG-2、AG-4和AG-5,分别占总菌株的6.24%、84.2%和1.1%。另有23个菌株不与任何标准菌株融合,占8.46%,说明新疆北疆棉田立枯丝核菌的优势菌系是多核丝核菌的AG-4融合群。通过从10种不同配方的培养基中筛选的效果好的麦芽蛋白胨(MPDA)配方培养基(Ⅱ)进行对峙培养,以建立标准菌株,将纯化获得的272个丝核菌菌株,划分为6个不同的营养亲和群,研究说明新疆北疆地区棉田立枯丝核菌各菌丝融合群内确有不同程度的分化。  相似文献

对立枯丝核菌(Rhizoctonia solani giihn)和禾谷丝核菌(R.E erealis Vande r Hoeven)的16个菌丝融合群或亚群的标准菌株及来自}工苏大麦纹枯病的9个菌丝融合群或亚群共68个菌株进行了聚丙烯酰胺凝胶电泳,测定酯酶同工酶。结果表明:1.立枯丝核菌主酶带数的变幅范围比禾谷丝核菌大;2.无论禾谷丝核菌,还是立枯丝核菌,各菌丝融合群或亚群之间的电泳图谱都有显著差异,但与各自对应的融合群或亚群的标准菌株的图谱则相似;3.44个大麦禾谷丝核菌CAG一1群菌株间的图谱也有差异,但都有一条共同的主酶带(E.);4.据主副酶带数目和位置,将禾谷丝核菌CAG一1群菌株再分为2个类型(1型和II型)和5个亚型(1a、Ib、Ic和Iia、Iib);酶谱类型与菌株的采集品种和致病性无关,但与采集地点似有一定的联系。  相似文献

【目的】了解立枯丝核菌(Rhizoctonia solani) AG1 IA对日本结缕草(Zoysia japonica steud.)的侵染过程及其引起的病害症状,为进一步从分子水平研究该菌的病理学侵染机制和草坪草抗病分子育种提供理论基础。【方法】通过根部接种法促使立枯丝核菌与日本结缕草无菌苗建立侵染关系,从而对其病叶率、病株率及病情指数进行统计分析,同时结合组织染色透明技术及植物组织石蜡切片对立枯丝核菌的侵染过程、感染方式进行研究。【结果】立枯丝核菌AG1 IA的侵染过程主要为:菌丝吸附在植物组织表面,并沿组织表面定向生长,形成侵染结构——侵染垫与组织建立密切的侵染关系。菌丝通过细胞间隙侵入植物组织内部,主要侵染植物皮层细胞及除木质部导管以外的整个维管束系统。结缕草的地上部分与地下部分组织对立枯丝核菌的侵染显现不同的寄主反应。【结论】立枯丝核菌的侵染过程主要包括吸附、定向生长、渗透、定殖4个部分;立枯丝核菌的侵染主要引起结缕草叶片病症;结缕草病变与菌丝直接侵染无直接联系,表明该菌具有复杂的侵染机制。  相似文献

藓类提取物对植物病原真菌的抑菌性研究   总被引:3,自引:0,他引:3  
以立枯丝核菌(Rhizoctonia)、尖孢镰孢菌(Fusarium oxysporum)、苹果轮纹病菌(Macrophoma kawatsukai)、梨黑星病菌(Fusicladium pirina)、草莓灰霉菌(Botrytis cinerea)等七种植物病原真菌为供试菌,通过生长速率法测定了24种藓类的乙醇提取物对病原真菌的抑制作用.结果表明,在提取物浓度为1.2mg干样/mL,大镰刀藓提取物对立枯丝核菌的抑制作用最强,抑菌率为67.8%.藓类提取物不仅对植物病原真菌有抑菌活性,而且还有多种藓类提取物对病原真菌菌丝有促进生长活性,如锐尖匍灯藓对立枯丝核菌菌丝生长的促进率为95.1%.选出大镰刀藓、扁灰藓、紫萼藓、山地水灰藓、疣小金发藓、橙色净口藓、锐尖匍灯藓和羊角藓对立枯丝核菌和灰霉菌的抑菌性或菌丝生长促进性进行了进一步研究,藓类提取物对病原菌的抑菌性或菌丝生长促进性在一定范围内随浓度的增加而增强,并且抑菌性随时间的延长呈下降趋势.  相似文献

利用系统聚类分析方法,通过对丝核菌菌丝融合一群Rhizoctonia solani (AG-1)的23个菌株在七个同功酶电泳所显示的50个表现型进行了遗传分析。结果显示,23个丝核菌菌株可聚类为三大类,他们分别代表了丝核菌菌丝融合一群中的三个亚群AG-I A,AG-IB,AG—IC;其中第一聚类又可进一步分为四个亚类。同功酶的分析证实了丝核菌菌丝融合1群中的三个亚群的分类概念是具有遗传背景的,并可作为丝核菌菌丝融合群之间及群内分类的重要手段。  相似文献

以重要植物病原菌为特征的丝核菌是一类在土壤中广泛分布的丝状真菌,通常不产孢,以菌丝或菌核的形式存在,多样性非常丰富。本文基于国内外最新研究进展,对依据菌丝体的细胞核数目、菌丝融合、有性生殖和系统进化等方面的基本特征展开的丝核菌分类体系及分类现状进行了综述。基于菌丝的细胞核数目,丝核菌被分为单核、双核和多核丝核菌三大类群。自然界中单核丝核菌数量极少,多核和双核丝核菌在全球分布广泛,占丝核菌的绝大多数。基于菌丝融合试验的结果,目前多核丝核菌被分为13个菌丝融合群,双核丝核菌被分为18个菌丝融合群。部分融合群内又根据一些稳定的特征分了亚群,但亚群的建立标准并不统一。目前的分子系统学研究结果基本支持丝核菌的菌丝融合群及亚群的分类。基于部分有性世代被发现的菌株的形态特征,多核和双核丝核菌分别被鉴定为亡革菌属和角担菌属。此外,目前已有分属重要植物病原菌和兰科菌根菌类群的至少9个融合群或亚群的17个菌株完成了基因组测序,比较基因组学和线粒体组学开始在丝核菌分类和进化研究中发挥作用。丝核菌分类系统特殊且复杂,作者在文末提出了目前丝核菌分类学研究面临的问题和今后研究的趋势,期待更多的学者参与到这个重要菌...  相似文献

研究了节丛孢Arthrobotrys、单顶孢Monacrosporium和隔指孢Dactylella三个捕食线虫丝孢菌属16个菌株,对水稻立枯丝核菌RhizoctoniasolaniAG1、大豆核盘菌Sclerotiniasclerotiorum、茄科镰刀菌Fusariumsolani和恶疫霉Phytophthoracactorum四种常见土壤植物病原真菌的菌寄生性。结果表明供试菌可以通过弹簧式菌丝圈缠绕、类附着胞结构吸附、简单的菌丝缠绕或者贴附寄主菌丝生长四种方式寄生病原菌。其中,绝大多数菌株对立枯丝核病菌有寄生作用,一些供试真菌对其它三种病原真菌有寄生现象。利用孢子液浸泡法测定了其中5种捕食线虫真菌对核盘菌菌核的寄生能力,显示有较高寄生率。  相似文献

【背景】贝莱斯芽胞杆菌(Bacillus velezensis) HN-Q-8菌株是本实验室前期获得的能有效拮抗立枯丝核菌(Rhizoctonia solani)的生防细菌。【目的】明确贝莱斯芽胞杆菌HN-Q-8菌株的抑菌活性物质。【方法】采用菌丝生长速率法检测其发酵液对5种马铃薯病原菌的拮抗能力以及稳定性,利用基质辅助激光解吸电离飞行时间质谱技术对HN-Q-8菌株活性物质进行鉴定。【结果】HN-Q-8菌株对立枯丝核菌(Rhizoctonia solani)、尖镰孢菌(Fusarium oxysporum)、茄链格孢(Alternaria solani)和致病疫霉(Phytophthora infestans)具有良好的抑菌活性,抑菌率可达50%-90%。发酵液分别经紫外照射35min、自然光照射10 h以及100°C高温处理后,相对抑菌率分别为74%、92%和98%;发酵液的抑菌活性不受胰蛋白酶和蛋白酶K的影响;但不耐强酸、强碱,其适宜pH为4.0-10.0,表明HN-Q-8菌株活性物质具有良好的稳定性。活性成分能使立枯丝核菌菌丝形态畸形扭曲,从而抑制立枯丝核菌的生长,经鉴定活性物质...  相似文献

AIM: To study the antagonistic activity by Pseudomonas fluorescens strain 96.578 on the plant pathogenic fungus Rhizoctonia solani. METHODS AND RESULTS: Strain 96.578 produced a new cyclic lipopeptide, tensin. High tensin production per cell was detected in liquid media with glucose, mannitol or glutamate as growth substrate while fructose, sucrose and asparagine supported low production. Tensin production was nearly constant in media with different initial C levels, while low initial N contents reduced production. When applied to sugar beet seeds, strain 96.578 produced tensin during seed germination. When challenged with strain 96.578 or purified tensin, Rhizoctonia solani reduced radial mycelium extension but increased branching and rosette formation. CONCLUSION: The antagonistic activity of strain 96.578 towards Rhizoctonia solani was caused by tensin. SIGNIFICANCE AND IMPACT OF THE STUDY: When coated onto sugar beet seeds, tensin production by strain 96.578 could be of significant importance for inhibition of mycelial growth and seed infection by Rhizoctonia solani.  相似文献

两种秸秆醋液对尖孢镰刀菌和立枯丝核菌的抑菌作用   总被引:1,自引:0,他引:1  
本研究以玉米秸秆醋液、水稻秸秆醋液为供试材料,研究不同稀释度的秸秆醋液对尖孢镰刀菌(Fusarium oxysporum)和立枯丝核菌(Rhizoctonia solani)的抑制作用,测定其可溶性蛋白和可溶性糖。结果表明,50倍稀释液对2种病原菌的抑制率为100%,即2种病原菌在50倍稀释处理的秸秆醋液中不能生长。玉米秸秆醋液对尖孢镰刀菌中可溶性蛋白的影响不显著,对立枯丝核菌中可溶性蛋白的影响表现为先升高后降低。水稻秸秆醋液对尖孢镰刀菌中可溶性蛋白的影响是先升高后降低,对立枯丝核菌中可溶性蛋白的影响是逐渐累积。高浓度的水稻秸秆醋液可提高2种菌体内可溶性糖含量,100倍稀释的玉米秸秆醋液可提高尖孢镰刀菌体内的可溶糖含量,玉米秸秆醋液对立枯丝核菌的可溶性糖含量基本不会产生影响。本研究为秸秆醋液在农业生产上的推广应用等提供科学依据。  相似文献

Two isolates of binucleate Rhizoctonia spp., previously selected for efficacy in suppression of Rhizoctonia solani and Pythium spp., as well as plant growth promotion, were incorporated into various solid substrate formulations. These formulated products were assayed at three doses in three glass-house experiments for biocontrol of damping-off diseases in Capsicum annuum. R. solani anastomosis group 4 or Pythium ultimum var. sporangiiferum were incorporated into pasteurized potting medium with each formulated binucleate Rhizoctonia product. All formulations were effective against both pathogens in at least two experiments, but some formulations of one isolate of binucleate Rhizoctonia did not give consistent control of R. solani in one experiment. The most consistent formulation, which provided control of both pathogens at all doses of binucleate Rhizoctonia, was the simple substrate of rice hulls. The implications for commercialization of a biocontrol product are discussed.  相似文献

以施药作为干扰因子,研究干扰条件下水稻纹枯病菌的时间和空间生态位变化.结果表明,各施药处理与对照纹枯病菌的空间生态位存在差异,其中孕穗期施药+齐穗期施药和孕穗期施药+灌浆期施药两处理的空间生态位宽度指数分别为0.520、0.572,与对照的空间生态位宽度指数0.8563相差最大;分蘖盛期一次施药处理的空间生态位宽度指数为0.8577,与对照相差最小.各施药处理和对照的时间生态位宽度无显著差异,表明施药对纹枯病菌的时间生态位宽度影响不大.空间生态位宽度指数、病叶(鞘)率、病情指数及防治效果等计测结果表明,两次施药对病害的控制好于一次施药,其中两次施药中以孕穗期施药+齐穗期施药和孕穗期施药+灌浆期施药对病害的控制作用最强.施药不但压缩了水稻纹枯病菌的空间生态位宽度,限制了其在产量形成较为重要的上部叶位的扩张,而且压缩了水稻纹枯病菌的时间生态位宽度,限制了其在水稻产量形成时期的扩张.但施药只改变了病菌生态位的分布,调整压缩了水稻产量形成期的病害生态位,病菌生态位势的总和不变.  相似文献

6-pentyl-alpha-pyrone (6PP) production by Trichoderma harzianum, in an extractive fermentation system, was elicitated by Rhizoctonia solani. The extent of 6PP elicitation was related to the state of Rhizoctonia and to the Trichoderma inoculum type. The use of non-viable Rhizoctonia solani mycelium in mycelium-inoculated Trichoderma harzianum culture, yielded the maximal 6PP production (474 mg l(-1)) compared to control cultures (147 mg l(-1)) and decreased the process time from 192 to 96 h.  相似文献

Field isolates of Rhizoctonia solani obtained from three discrete bare patches in a wheat field in Western Australia were characterized by pectic zymogram grouping. The genetic background of pectic enzymes was analysed by comparing the zymograms of asexual homokaryons and sexual progenies derived from field isolates. The 170 field isolates obtained from the field site produced indistinguishable pectic zymograms. However, variations among field isolates of the same zymogram group were detected, on the basis of zymograms of their resultant protoplast-regenerated cultures. Asexual sibling homokaryons derived from each of the field isolates were heterogeneous for their pectic enzymes. Homokaryons with a common heterokary on incompatibility factor, obtained from a field isolates were homogeneous for pectic enzymes. Basidiospore progenies of a field isolate segregated widely in pectic zymograms. It appeared that the expression of pectic enzymes by field isolates involved multiple genetic factors. The variation of zymograms among homokaryotic strains suggests that each field isolate of R. solani contains two types of nuclei, although cells of vegetative hyphae are multinucleate.  相似文献

The soil fungus Rhizoctonia solani produces phytotoxic phenylacetic acid (PAA) and hydroxy (OH-) and methoxy (MeO-) derivatives of PAA. However, limited information is available on the specific role that these compounds play in the development of Rhizoctonia disease symptoms and concentration(s) required to induce a host response. Reports that PAA inhibits the growth of R. solani conflict with the established ability of the fungus to produce and metabolize PAA. Experiments were conducted to clarify the role of the PAA metabolic complex in Rhizoctonia disease. In this study the concentration of PAA and derivatives required to induce tomato root necrosis and stem canker, in the absence of the fungus, and the concentration that inhibits mycelial growth of R. solani were determined. The effect of exogenous PAA and derivatives of PAA on tomato seedling growth also was investigated. Growth of tomato seedlings in medium containing 0.1-7.5 mM PAA and derivatives induced necrosis of up to 85% of root system. Canker development resulted from injection of tomato seedling stems with 7.5 mM PAA, 3-OH-PAA, or 3-MeO-PAA. PAA in the growth medium reduced R. solani biomass, with 50% reduction observed at 7.5 mM. PAA, and derivatives were quantified from the culture medium of 14 isolates of R. solani belonging to three distinct anastomosis groups by GC-MS. The quantities ranged from below the limit of detection to 678 nM, below the concentrations experimentally determined to be phytotoxic. Correlation analyses revealed that isolates of R. solani that produced high PAA and derivatives in vitro also caused high mortality on tomato seedlings. The results of this investigation add to the body of evidence that the PAA metabolic complex is involved in Rhizoctonia disease development but do not indicate that production of these compounds is the primary or the only determinant of pathogenicity.  相似文献

为了开发新型植物源杀菌剂和充分利用菊芋(Helianthus tuberosus L.)资源,本文尝试用石油醚、乙醚、乙酸乙酯和水等4种溶剂对菊芋叶片进行平行提取,采用生长速率法测定菊芋叶片提取物对水稻纹枯菌(Rhizoc-tonia solaniKühn)、小麦赤霉菌[Gibberella zeae(Schw.)Petch]、番茄早疫菌[Alternaria solani(Ellis et Martin)Jones et Grour]和番茄灰霉菌(Botrytis cinerea Pers.)生长量的抑制活性;并通过试管法和滤纸法对菊芋叶片内化学成分进行初步预试。抑菌实验结果表明:(1)菊芋叶片各溶剂提取物处理与对照处理相比差异显著;(2)菊芋叶片水提取物处理与各有机溶剂提取物处理差异也基本显著;(3)菊芋叶片各溶剂提取物同浓度处理对水稻纹枯菌、番茄早疫菌和番茄灰霉菌抑制效果较好;(4)菊芋叶片乙酸乙酯提取物抑菌效果最为显著,浓度为20mg/mL时对番茄早疫菌和番茄灰霉菌已达到完全抑制,对水稻纹枯菌抑制率也达到77.91%。初步化学预试结果说明,菊芋叶片中含有蛋白质、氨基酸、还原糖类、有机酸、酚类和鞣质、黄酮类、内酯类、强心甙以及油脂等化学成分。  相似文献

Two isolates of Trichoderma koningii were evaluated for efficacy in control of damping-off diseases in seedlings of Capsicum annuum grown in pasteurized potting medium in a glasshouse. A selected isolate of binucleate Rhizoctonia and two fungicides were also included as standards for control of Rhizoctonia solani and Pythium ultimum var. sporangiiferum. Both isolates of T. koningii reduced seedling death caused by R. solani in one of two experiments, and by P. u. sporangii-ferum in two of three experiments. Neither isolate of T. koningii suppressed damping-off caused by either pathogen as consistently as the binucleate Rhizoctonia or fungicides. The implications of these results for commercial disease management are discussed.  相似文献

The aim of two year investigation was the valuation the effect of biopreparate Polyversum (B.A.S. Pythium oligandrum) and preparate Biochikol 020 PC (B.A.S. chitosan) used in control of potato against Phytophthora blight on the tuber infestation during storage by Helminthosporium solani and tuber infestation by sclerotia of Rhizoctonia solani. As the standard fungicide Vitavax 200 FS (B.A.S. karboxin and thiuram) was used. After harvesting 100 tubers from each plots was collected and put in storage. The analysis of tuber infestation by Rhizoctonia solani and Helminthosporium solani was made after harvesting (September) and later every 3 months during storage period (December, March). The percent of diseased tubers in tested sample and also infestation degree of bulbs using 5-degree scale was estimated. The received results of investigations ascertained, that all tested preparations during potato vegetation influenced on lower (in comparison with control) degree of bulbs infestation by sclerots of Rhizoctonia solani and the mean degree of infestation by Helminthosporium solani. Moreover the percent of diseased tubers infected by pathogens with tested preparations combination was significant lower than in control.  相似文献
