Fabrication of protein a-viologen hetero Langmuir-Blodgett film for fluorescence immunoassay

Protein A molecular thin film was fabricated as a platform of antibody-based biosensor. For the immobilization of the protein A thin film, a viologen multilayer was built up using the Langmuir-Blodgett (LB) technique, and then, protein A was adsorbed on the viologen LB film by an electrostatic interaction force, which was formed as a hetero-film structure. For the deposition of viologen, surface pressure area (π-A) isotherm was investigated. The fabricated protein A-viologen hetero LB film was investigated using atomic force microscopy (AFM). Using the developed molecular film, antibody immobilization and fluorescence measurement was carried out.

Application of x-ray analysis for the study of the orientation in DNP films]

A planar texture in dried film samples of DNP was found by X-ray study. Axes of DNA macromolecules are located in the film plane and the 77 A reflection is directed along this axis. Periodic structures corresponding to 55 and 34 A are packed parallel to the film plane. The results obtained do not contradict with a supercoil DNP model, however they all allow other possible packing models of DNP molecules, in particular, histone layers alternating with DNA layers in oriented films.     

A stereocomplex platform efficiently detecting antigen-antibody interactions

Ultrathin poly(methyl methacrylate) (PMMA) stereocomplex films with macromolecularly double-stranded regular nanostructures were prepared by layer-by-layer assembly of isotactic and syndiotactic PMMAs on solid surfaces. Antibodies were immobilized through the Fc region-capturing protein A, which had been physically adsorbed on the complex film, and the binding of antigens to immobilized antibodies was quantitatively investigated by the quartz crystal microbalance technique. Greater amounts of protein A with native forms were adsorbed on the complex film than those on conventional single-component PMMA films. Antibodies with high target-binding activities were also immobilized on the complex film. A greater amount of antigens could be detected on the complex film. The activity of protein A was maintained on the complex for a long time even within a dried state. The mechanism for the preservation of protein native forms on the complex surface was speculated by analyzing the physical adsorption of proteins with various secondary structures. Stereocomplex films can be utilized as novel coating nanomaterials for efficiently detecting protein-protein interactions.

Increased Hydrophobicity of Carboxymethyl Starch Film by Conjugation with Zein

A carboxymethyl starch (CMS) film was prepared by a process in which gelatinized CMS was dried, and subsequently treated with water-soluble carbodiimide in the presence of zein in 70% ethanol or 70% acetone to form acid-amide cross-linkages in order to increase the hydrophobicity of the surface of the film. A small amount of zein protein was found to be present on the surface of the zein-CMS conjugate (Zein-CMS) film, resulting in its insolubility in hot water, low water vapor permeability, and resistance to digestion with α-amylase and β-amylase. Digestion of the Zein-CMS film with protease rendered the film readily water-soluble, suggesting conjugation with zein as an effective means of increasing the hydrophobicity of biodegradable starch-based articles.

A simplified X-ray film method for detecting bacterial production of Hgo by Escherichia coli was developed. There was excellent agreement of results between this X-ray film method and radioactive mercury volatilization assays. Although not quantitative, the X-ray film method has the advantages of being rapid, simple to perform, and inexpensive.

A constant-depth laboratory model film fermentor

A laboratory model constant-depth film fermentor was developed. Film grew on the surface of polytetrafluoroethylene (PTFE) plugs and was limited to a predetermined depth by mechanically removing excess film. Six film-forming organisms were isolated from river water and used to assess the operating characteristics of the fermentor. Film accumulation was logarithmic, and a steady state was maintained. Electron micrographs show early film development. The fermentor enables film to be grown on any substratum and allows discrete, reproducible, and representative samples to be taken.

Studies on film production by mycoplasmas revealed that film was produced by completely disintegrated mycoplasma cells on Noble agar in the presence of horse serum. Film production was due to an enzymic reaction between mycoplasma lipase, possibly phospholipase A, and phospholipid in serum.     

Electropolymerization molecularly imprinted polymer (E-MIP) SPR sensing of drug molecules: pre-polymerization complexed terthiophene and carbazole electroactive monomers

A novel chemosensitive ultrathin film with high selectivity was developed for the detection of naproxen, paracetamol, and theophylline using non-covalent electropolymerized molecular imprinted polymers (E-MIP). A series of monofunctional and bifunctional H-bonding terthiophene and carbazole monomers were compared for imprinting these drugs without the use of a separate cross-linker. A key step is the fast and efficient potentiostatic method of washing the template, which facilitated enhanced real-time sensing by surface plasmon resonance (SPR) spectroscopy. Various surface characterizations (contact angle, ellipsometry, XPS, AFM) of the E-MIP film verified the templating and release of the drug from the cross-linked conducting polymer film.
