Rhizosolenia mat abundance, distribution and chemical compositionwere studied on two cruises in the central North Pacific gyrein order to determine large-scale distribution patterns andcontribution to upward nitrogen (N) flux. These macroscopicdiatom mats are composed of multiple species of Rhizosoleniathat exploit subsurface nitrate pools by vertically migratingbelow the euphotic zone. Although numerically dominated by thesmall-diameter species, R.fallax (73–95% of total numbers),mat biovolume was dominated by large-diameter (>50 µmdiameter) Rhizosolenia spp. (85–99% of total volume).Integrated mat abundance was substantially higher when matsaccumulated at the surface during calm weather (  相似文献

The presence of nitrogen–Fixing cyanobacterial endophytes, such as Richelia intracellularis Schmidit commonly observed within several species of Rhizosolenia, in nitrogen-limited oceanic waters has obvious implications both for the host organism(s) and for the entire planktonic assemblage. Recently, epi-fluorescent examination of pllankton samples collected off Oahu, Hawaii, in september 1982 revealed R. intracellularis within the diatoms Hemiaulus membranaceus Cleve and H. hauckii Grunow as well as its expected presence within Rhizosolennia spp. Richelia intracellularis coccurred with within Hemiaulus spp. at frequenccies (ca.80%)comparable to those noted for Rhizosolenia spp. Standard bright field or phaseconrast microsocopy could not reliably distinguish the cyanobacteria within Hemiaulus spp. If this association is common, the occurrence of R.intracellularis (and its significance in the nitrogen dynamics of the ocean)may have been greatly underestimated in previous studies.  相似文献

The warm oligo-eurytherm diatomsRhizosolenia robusta Norman andRhizosolenia imbricata Brightwell were cultured to determine the temperature range for the best competitive position by growth. Comparison of their generation times with those of other diatoms indicate thatR. robusta reaches this position around 20 °C andR. imbricata above 25 °C. The temperature ranges for growth were 12 °C up to 28 °C forR. robusta and 12 °C to above 30 °C forR. imbricata. The use of both species as indicator species for warm water currents is discussed on account of their lower temperature limit. The cold oligo-eurytherm diatomRhizosolenia shrubsolei had a temperature range for growth of below –1.0 °C to 25 °C. Our experimental results demonstrate thatR. imbricata andR. shrubsolei can be considered separate species.  相似文献

We present a numerical model of nutrient uptake and photosynthesisduring migrations of the marine diatom Rhizosolenia that wasdeveloped to estimate fluxes of carbon and nitrogen due to thesemigrations in the open ocean. The predicted specific rate ofincrease of Rhizosolenia was 0.11–0.15 day1, whereas thetotal time for one migration cycle ranged between 3 and 5 days.Using published estimates of Rhizosolenia abundance, we estimatethat new primary production due to Rhizosolenia migrations rangesbetween 0.018 and 0.033 mmol N m–2 day–1. Thesevalues represent up to 17% of new production due to turbulentdiffusive fluxes of nitrate into the euphotic zone and are ofthe same order of magnitude as new production due to nitrogenfixation in tropical oceans. Large-scale contributions of Rhizosoleniato oceanic new production are limited by their relatively lowstanding crop. Net-collected Ethmodiscus spp. were examined and found to becapable of positive buoyancy with ascent rates averaging 1.4m h–1 on individually measured cells, and 0.3 m h–1using a homogeneous settling technique. These data, togetherwith published reports of positive buoyancy in Rhizosolenia,indicate that upward movement is a widespread phenomenon inlarge oceanic diatoms. These observations also suggest thatpublished size-rate relationships for marine diatoms requirerevision to include the effects of positive buoyancy.  相似文献

Empty packets were isolated from Anabaena-free Azolla (Azollapinnata var. imbricata R. Br.) leaves treated with the enzymesolution. Bot. 38: 1–12 Glasshouse grown, symbiotically-dependent seedlings of Acaciaalata R.Br., .A. extensa Lindl., and A. pulchella R.Br. wereexamined for acetylene reduction in closed assay systems usingundisturbed potted plants, excavated whole plants, nodulatedroots or detached nodules. Nitrogenase activity declined sharplyover the first hour after exposure of detached nodules to acetylene(10% v/v in air), less steeply or not at all over a 3 h periodin assays involving attached nodules. Using detached nodules,rates of acetylene reduction, nitrogen (15N2) fixation, andhydrogen evolution in air (15N2) and acetylene-containing atmosphereswere measured in comparable 30 min assays. Total electron flowthrough nitrogenase in air was determined from rates of nitrogen(15N2) fixation ( ? 3) plus hydrogen evolution, that in thepresence of acetylene from rates of acetylene reduction andhydrogen evolution in air: acetylene. Values for the ratio ofelectron flow in air: acetylene to that in air ranged from 0?43to 0?83 in A. pulcheila, from 0?44 to 0?66 in A. alala and from0?37 to 0?70 in A. extensa, indicating substantial inhibitionof electron flow through nitrogenase of detached nodules byacetylene. Relative efficiencies of nitrogenase functioningbased on hydrogen evolution and acetylene reduction were from0?15 to 0?79, those based on nitrogen (15N2) fixation and hydrogenevolution from 0?53 to 0?87. Molar ratios of acetylene reducedto nitrogen (15N2) fixed were 2?82 ? 0?24, 201 ? 0?15, and 1?91? 0?11 (?s.e.; n = 7) for A. pulcheila,A. extensa and A. alata respectively A standard 5–10 min acetylene reduction assay, conductedon freshly detached unwashed nodules in daytime (12.00–14.00h), was calibrated for field use by comparing total N accumulationof seedlings with estimated cumulative acetylene reduction overa 7-week period of glasshouse culture. This represents the first study of nuclear DNA content in alarge sample (135 spp.) from a tropical arboreal genus, in whicha large proportion of the species were examined (42 spp., 31.1%).Somatic chromosome numbers and 4C-DNA values for 51 taxa ofLonchocarpus are reported. All taxa were diploid with 2 n =22,but their DNA content ranged from 1.92 to 2.86 pg 4C nucleus,corresponding to a 48.95% variation in genome size. In the 74collections studied, no correlation was observed between DNAcontent and habitat altitude. Variation in nuclear DNA contentwas analysed at the level of genus, subgenus, section and subsection.Variation in genome size was also studied within some species,either among widely separated populations or among differentintraspecific taxa. Very little variation in genome size wasdetected between populations, subspecies, and varieties of thesame species. The dried leaves of 31 species and two varieties of the genusErythroxylum were quantitatively analysed for cocaine and bothcis- and trans-cinnamoylocaine, using a stable-isotope dilutionmethod and gas chromatography-mass spectrometry selected-ionmonitoring. Both cocaine and cinnamoylcocaine were detectedin all four varieties of cultivated coca (E. coca var. coca,E. coca var. ipadu, E. novogranatense var. novogranatense andE. novogranatense var.truxillense). Cinnamoylcocaines (cis andtrans) were found in much higher concentrations in both varietiesof E. novogranatense than in either variety of E. coca. Amazoniancoca (E. coca var. ipadu) contained consistently lower cocainelevels than the montane variety (E. coca var. coca). Twenty-ninewild species of Erythroxylum, selected to represent morphological,ecological and taxonomic diversity, were analysed for the samealkaloids; cocaine was detected in 13 neotropical species, representingfive sections of the genus. No cocaine was detected in the OldWorld species. Two wild species from Venezuela, E. recurrensand E. steyermarkii, contained cocaine levels comparable tothose found in the commercially cultivated species. The vegetative anatomy of 18 (19) Uncinia spp. (Cyperaceae-Cariceae)including representatives of most taxonomic subdivisions wasstudied to determine the range of variation in certain anatomicaland morphological characters of the vegetative organs withinthe genus. The two South American species U. erinacea and U. kingii differfrom all others, the former in having a closed cylinder of sclerenchymain the culm, and the latter a grooved culm. The three tall SouthAmerican spp., U. brevicaulis, U. hamata, and U. phleoides var.trichocarpa, are characterized by adaxial intercostal fibrestrands in the leaves, and in this respect show affinities withtall New Zealand species, U. sinclairii and U. uncinata. Thelast species does not have the intercostal strands. The muchsmaller U. tenuis from S. America resembles, in the size andshape of transverse sections of leaf and culm, a group of speciesfrom New Zealand comprising U. angustifolia, U. egmontiana,and U. rupestris. To these could be added U. banksii, U. hookeri,and perhaps also U. tenella, although the last two spp. exhibitsome distinctive characters in the transverse section of theleaf and also in the leaf epidermis in surface view. No exactcounterpart to the Australian and New Zealand spp. U. divaricata,U. riparia, U. rubra and U. scabra was found amongst the S.American material. This group of spp. is distinctive, for example,because of the triangular or irregularly triangular shape oftransverse sections of the culm and the large amount of sclerenchymain transverse sections of the leaf. The range of structural variation appears to be particularlywide in the S. American spp., which represent two extreme typesin the shape of the leaf in transverse section. The other talland small species have their respective counterparts in eacharea, although they are more profusely represented in New Zealandthan in America, and therefore show a correspondingly greaterrange of structure.  相似文献

Physico-chemical parameters, hydrological conditions, and microbial interactions can affect the growth and persistence of cyanobacteria, but the interacting effects among these bloom-forming factors are still poorly known. This hampers our capacity to predict the occurrence of cyanobacterial bloom accurately. Here, we studied the relationship between temperature, N and P cycles, and the microbial community abundance and diversity at 0.5 m under the surface of West Lake (China) from January 21 to November 20, 2015, in order to better understand the key factors regulating temporal changes in the cyanobacterial community. Using high throughput sequencing of the 16S rRNA gene V3-V4 region, we studied the diversity and abundance of bacteria. In parallel, we measured physico-chemical parameters and followed the abundance of key genes involved in N fixation, denitrification, and nutrient uptake. Multivariate analyses suggest that P concentration and water temperature are the key factors controlling the outbreak of summer cyanobacterial bloom. RT-qPCR analyses of the bacterial community and measurements of the copy number of denitrification-related gene (nirK, nosZ, nirS) show that denitrification potential and denitrifying bacteria relative abundance (Pseudomonas and Bacillus) increased in concert with diazotrophic cyanobacterial genera (Anabaena, Nostoc, Aphanizomenon flos-aquae) and the common bloom-forming non-diazotrophic cyanobacterium genus Microcystis. The present study brings new insights on the complex interplay between physico-chemical parameters, heterotrophic bacterial community composition, nitrogen cycle, and cyanobacteria dominance in a eutrophic lake.

Bot. 38: 13–25 Growth, nitrogen accumulation, nodulation and nitrogenase activity(C2H2 reduction activity) were monitored in a dense stand ofseedlings of Acacia pulchella R.Br. and A. alata R.Br. duringtwo growing seasons after fire at a jarrah (Eucalyptus marginataDonn ex Sm.) forest site at Illawarra, 35 km south-east of Perth.Nitrogen fixation, estimated by a previously calibrated C2H2reduction assay, was essentially restricted to the winter andspring months (July to October) and was estimated to contribute37% and 9% respectively of the total N accumulated by A. pulchelladuring the first and second seasons of growth. Comparable valuesfor A. alata were 29% and 2%. Comparisons with fully symbioticplants raised in a glasshouse in supposedly non-limiting growthconditions in minus nitrogen sand culture indicated that waterstress rather than high temperatures was responsible for lossof nodules, cessation of symbiotic activity and attenuationof growth during summer in the field. By 19 months glasshouseplants had gained 130–230 times the dry weight and accumulated110–160 times the total N of similarly aged, field grownplants. Growth, nodulation and N2 fixation of symbiotically-dependentsand cultured plants of A. pulchella and A. alata respondedmarkedly to phosphate, and additions of water and nutrientsto 3-year plants of A. pulchella during a winter growing seasonin the field indicated that periods of reduced soil moistureand low availability of nutrients, particularly phosphorus,might limit symbiotic performance under natural conditions. 15N2 studies and acetylene reduction assays of leaves and shootsof Douglas fir and other forest trees do not confirm previousreports that extensive nitrogen fixation occurs on leaf surfacesand it is concluded that the importance of nitrogen fixationin the canopy of forest trees has been exaggerated. The presenceof nitrogen-fixing bacteria on the leaves of trees is confirmed,however, and they have been identified as Enterobacter agglomerans,Clostridium butyricum and Bacillus sp. Abstract: Four microbial mat-forming, non-axenic, strains of the non-heterocystous, filamentous, cyanobacterial genus Microcoleus were maintained in culture and examined for the ability to fix atmospheric nitrogen (N2). Each was tested for nitrogenase activity using the acetylene reduction assay (ARA) and for the presence of the dinitrogenase reductase gene ( nifH ), an essential gene for N2 fixation, using the polymerase chain reaction (PCR). The Microcoleus spp. cultures were incapable of growth without an exogenous nitrogen source and never exhibited nitrogenase activity. Attempts to amplify a 360-bp segment of the nifH gene using DNA purified from the cyanobacterial cultures did not produce any cyanobacteria-specific nifH sequences. However, several non-cyanobacterial homologous nifH sequences were obtained. Phylogenetic analysis showed these sequences to be most similar to sequences from heterotrophic bacteria isolated from a marine microbial mat in Tomales Bay (California, USA), and bulk DNA extracted from a cryptobiotic soil crust in Moab (Utah, USA). Microcoleus spp. dominated the biomass of both systems. Cyanobacteria-specific 16S rDNA sequences obtained from the cultured cyanobacterial strains demonstrate that the lack of cyanobacteria-specific nifH sequences was not due to inefficiency of extracting Microcoleus DNA. Hence, both the growth and genetic data indicate that, contrary to earlier reports, Microcoleus spp. appear incapable of fixing N2 because they lack at least one of the requisite genes for this process. Furthermore, our study suggests epiphytic N2-fixing bacteria form a diazotrophic consortium with these Microcoleus spp. and are likely key sources of fixed N2 generated within soil crusts and marine microbial mats.  相似文献

The eutrophication of freshwaters is a global health concern as lakes with excess nutrients are often subject to toxic cyanobacterial blooms. Although phosphorus is considered the main element regulating cyanobacterial biomass, nitrogen (N) concentration and more specifically the availability of different N forms may influence the overall toxicity of blooms. In this study of three eutrophic lakes prone to cyanobacterial blooms, we examined the effects of nitrogen species and concentrations and other environmental factors in influencing cyanobacterial community structure, microcystin (MC) concentrations and MC congener composition. The identification of specific MC congeners was of particular interest as they vary widely in toxicity. Different nitrogen forms appeared to influence cyanobacterial community structure leading to corresponding effects on MC concentrations and composition. Total MC concentrations across the lakes were largely explained by a combination of abiotic factors: dissolved organic nitrogen, water temperature and ammonium, but Microcystis spp. biomass was overall the best predictor of MC concentrations. Environmental factors did not appear to affect MC congener composition directly but there were significant associations between specific MC congeners and particular species. Based on redundancy analyses (RDA), the relative biomass of Microcystis aeruginosa was associated with MC-RR, M. wesenbergii with MC-LA and Aphanizomenon flos-aquae with MC-YR. The latter two species are not generally considered capable of MC production. Total nitrogen, water temperature, ammonium and dissolved organic nitrogen influenced the cyanobacterial community structure, which in turn resulted in differences in the dominant MC congener and the overall toxicity.  相似文献

The sexual phase of three species of Jamesonia (J. imbricata, J. rotundifolia and J. scammaniae) has been studied, from spore germination to gametangia formation, with special attention to the morphological development of prothalli. This is the first work to deal with gametophytes of species in this genus. All three species have trilete, tetrahedrical spores that germinate following the Vittaria type. The developmental pattern of J. imbricata and J. rotundifolia is intermediate between the Adiantum and Ceratopteris types, as the initial mitotic activity is assumed by an obconical apical cell, substituted later by a lateral meristem. In J. scammaniae, no apical cell or lateral meristem is formed, so the prothalli remain ameristic throughout the developmental process. Adult gametophytes are naked and variable in shape, in the same species ranging from typical cordate prothalli to irregularly lobed forms. Gametangia of normal shape and size was observed in both J. imbricata and J. rotundifolia, but not in J. scammaniae. Apogamy could be expected to occur in the genus, a phenomenon to be examined in the future. Comparisons are made with known species of related genera and results are discussed from an ecological perspective in the paramo environment.  相似文献

Responses of the leaves of five species of azalea to environmentalstresses, such as freezing, dehydration, high temperature andsalt spray, were measured in terms of water proton NMR relaxationtimes (T1), supercooling ability, and water content. Three subtropicalspecies (R. scabrum cv. Shounoshin, R. eriocarpum and R. tashiroivar. lasiophyllum) and two northern species (R. indicum cv.Kumano-satsuki and R. yedoense f. poukhanense), which originatedin different ecological habitats, showed characteristic behaviorsin terms of T1 relaxation times. In general, a species witha large change in T1 is more stress-sensitive than a speciesshowing the opposite tendency. The relative sensitivity to variousstresses of each species appears to be related to the severityof conditions in its natural habitat. It seems possible thatthose species of azalea with higher sensitivity to a particularsingle stress may also exhibit higher sensitivity to severalor even most stresses, and vice versa. Isolations of the rhizobia from nodules of field plants of Ulexeuropaeus and Medicago lupulina have been examined for effectivenessin fixation of nitrogen by inoculating them into host plantsgrowing in a medium free of combined nitrogen. The results indicatethat vigorous fixation may be presumed to occur in associationwith field plants of the Ulex species, but that fixation inMedicago is probably decreased by the presence of ineffectiverhizobia in some nodules. The general importance of fixationin wild legumes is discussed, and it is pointed out that dueattention must be paid to non-legume genera with root nodules.These genera are locally abundant, and in former periods werestill more prominent in some regions of the world.  相似文献

The transport of nitrogen from the roots and nodules of chickpea(Cicer anetinum L.), lentil (Lens culinaris Medic), faba bean(Vicia faba L.) and pea (Pisum sativum L.) was examined in glasshouse-grownplants supplied either with nitrate-free nutrients or with nutrientssupplemented with 1,2,4 or 8 mol m-3153N-nitrate. A sixth treatmentcomprised uninoculated plants supplied with 8–0 mol m-31513N-nitrate. For each species, more than 75% of the nitrogenwas exported from the nodules as the amides, asparagine andglutamine. In fully symbiotic plants, the amides also dominatednitrogen transport to the shoot When N2 fixation activity wasdecreased by the addition of nitrate to the rooting medium,the N-composition of xylem exudate and stem solutes changedconsiderably. The relative concentrations of asparagine tendedto increase in the xylem whilst those of glutamine were reduced;the levels of nitrate increased in both xylem exudate and thesoluble nitrogen pool of the stem with a rise in nitrate supply.The changes in relative nitrate contents reflected generallythe contributions of root and shoot to overall nitrate reductaseactivity at the different levels of nitrate used. The relationshipsbetween the relative contents of xylary or stem nitrate andamino nitrogen and the plants' reliance on N2 fixation (determinedby the 15N isotope dilution procedure) were examined. Data suggestthat compositional relationships based on nitrate may be reasonableindicators of symbiotic dependence for all species under studyexcept faba bean when greater than 25% of plant nitrogen wasderived from N2 fixation.
