Although Saccharomyces cerevisiae can form petite mutants with deletions in mitochondrial DNA (mtDNA) (ρ) and can survive complete loss of the organellar genome (ρo), the genetic factor(s) that permit(s) survival of ρ and ρo mutants remain(s) unknown. In this report we show that a function associated with the F1-ATPase, which is distinct from its role in energy transduction, is required for the petite-positive phenotype of S. cerevisiae. Inactivation of either the α or β subunit, but not the γ, δ, or ɛ subunit of F1, renders cells petite-negative. The F1 complex, or a subcomplex composed of the α and β subunits only, is essential for survival of ρo cells and those impaired in electron transport. The activity of F1 that suppresses ρo lethality is independent of the membrane Fo complex, but is associated with an intrinsic ATPase activity. A further demonstration of the ability of F1 subunits to suppress ρo lethality has been achieved by simultaneous expression of S. cerevisiae F1α and γ subunit genes in Kluyveromyces lactis– which allows this petite-negative yeast to survive the loss of its mtDNA. Consequently, ATP1 and ATP2, in addition to the previously identified AAC2, YME1 and PEL1/PGS1 genes, are required for establishment of ρ or ρo mutations in S. cerevisiae. Summary Using a direct Monte Carlo simulation, population growth of helper T-cells (N H) and viral cells (N v) is studied for an immune response model with an enhanced spatial inter-cellular interaction relevant to HIV as a function of viral mutation. In the absence of cellular mobility (P mob=0), the helper T-cells grow nonmonotonically before reaching saturation and the viral population grows monotonically before reaching a constant equilibrium. Cellular mobility (P mob=1) enhances the viral growth and reduces the stimulative T-cell growth. Below a mutation threshold (P c), the steady-state density of helper T-cell (p H) is larger than that of the Virus (p v); the density difference Δp o(=pV−pH) remains a constant at P mob=1 while −Δp o→0 as P mutP c at P mob=0. Above the mutation threshold, the difference Δp o in cell density, grows with ΔP=P mutP c monotonically: ΔP o ∞ (ΔP)β ≃ with β≈0.574±0.016 in absence of mobility, while Δp o≈6(ΔP) with P mob=1.  相似文献

By simultaneously analyzing the chlorophyll a fluorescence transient and light absorbance at 820 nm as well as chlorophyll fluorescence quenching, we investigated the effects of different photon flux densities (0, 15, 200 μmol m−2 s−1) with or without 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) on the repair process of cucumber (Cucumis sativus L.) leaves after treatment with low temperature (6°C) combined with moderate photon flux density (200 μmol m−2 s−1) for 6 h. Both the maximal photochemical efficiency of Photosystem II (PSII) (F v/F m) and the content of active P700 (ΔI/I o) significantly decreased after chilling treatment under 200 μmol m−2 s−1 light. After the leaves were transferred to 25°C, F v/F m recovered quickly under both 200 and 15 μmol m−2 s−1 light. ΔI/I o recovered quickly under 15 μmol m−2 s−1 light, but the recovery rate of ΔI/I o was slower than that of F v/F m. The cyclic electron transport was inhibited by chilling-light treatment obviously. The recovery of ΔI/I o was severely suppressed by 200 μmol m−2 s−1 light, whereas a pretreatment with DCMU effectively relieved this suppression. The cyclic electron transport around PSI recovered in a similar way as the active P700 content did, and the recovery of them was both accelerated by pretreatment with DCMU. The results indicate that limiting electron transport from PSII to PSI protected PSI from further photoinhibition, accelerating the recovery of PSI. Under a given photon flux density, faster recovery of PSII compared to PSI was detrimental to the recovery of PSI or even to the whole photosystem.  相似文献

The lower oxygen limit (LOL) in plants may be identified through the measure of respiratory gases [i.e. the anaerobic compensation point (ACP) or the respiratory quotient breakpoint (RQB)], but recent work shows it may also be identified by a sudden rise in dark minimum fluorescence (F o). The interrelationship between aerobic respiration and fermentative metabolism, which occur in the mitochondria and cytosol, respectively, and fluorescence, which emanates from the chloroplasts, is not well documented in the literature. Using spinach (Spinacia oleracea), this study showed that Fo and photochemical quenching (q P) remained relatively unchanged until O2 levels dropped below the LOL. An over-reduction of the plastoquinone (PQ) pool is believed to increase F o under dark + anoxic conditions. It is proposed that excess cytosolic reductant due to inhibition of the mitochondria’s cytochrome oxidase under low-O2, may be the primary reductant source. The maximum fluorescence (F m) is largely unaffected by low-O2 in the dark, but was severely quenched, mirroring changes to the xanthophyll de-epoxidation state (DEPS), under even low-intensity light (≈4 μmol m−2 s−1). In low light, the low-O2-induced increase in F o was also quenched, likely by non-photochemical and photochemical means. The degree of quenching in the light was negatively correlated with the level of ethanol fermentation in the dark. A discussion detailing the possible roles of cyclic electron flow, the xanthophyll cycle, chlororespiration and a pathway we termed ‘chlorofermentation’ were used to interpret fluorescence phenomena of both spinach and apple (Malus domestica) over a range of atmospheric conditions under both dark and low-light.  相似文献

Brush border membrane vesicles, BBMV, from eel intestinal cells or kidney proximal tubule cells were prepared in a low osmolarity cellobiose buffer. The osmotic water permeability coefficient P f for eel vesicles was not affected by pCMBS and was measured at 1.6 × 10−3 cm sec−1 at 23°C, a value lower than 3.6 × 10−3 cm sec−1 exhibited by the kidney vesicles and similar to published values for lipid bilayers. An activation energy E a of 14.7 Kcal mol−1 for water transport was obtained for eel intestine, contrasting with 4.8 Kcal mol−1 determined for rabbit kidney proximal tubule vesicles using the same method of analysis. The high value of E a , as well as the low P f for the eel intestine is compatible with the absence of water channels in these membrane vesicles and is consistent with the view that water permeates by dissolution and diffusion in the membrane. Further, the initial transient observed in the osmotic response of kidney vesicles, which is presumed to reflect the inhibition of water channels by membrane stress, could not be observed in the eel intestinal vesicles. The P f dependence on the tonicity of the osmotic shock, described for kidney vesicles and related to the dissipation of pressure and stress at low tonicity shocks, was not seen with eel vesicles. These results indicate that the membranes from two volume transporter epithelia have different mechanisms of water permeation. Presumably the functional water channels observed in kidney vesicles are not present in eel intestine vesicles. The elastic modulus of the membrane was estimated by analysis of swelling kinetics of eel vesicles following hypotonic shock. The value obtained, 0.79 × 10−3 N cm−1, compares favorably with the corresponding value, 0.87 × 10−3 N cm−1, estimated from measurements at osmotic equilibrium. Antioxidant enzymes in chloroplasts and chlorophyll fluorescence parameters of leaves of Puccinellia tenuiflora (Turcz.) scribn.et Merr. under isotonic Na2CO3 and NaCl stresses were studied. Ascorbate peroxidase (APX) and superoxide dismutase (SOD) activities showed a similar increasing trend and then decreased with the decreasing osmotic potential of culture solution, peaking at −4.74 × 105 Pa under NaCl stress and at −3.40 × 105 Pa under Na2CO3 stress. APX, glutathione transferase and SOD activities were higher under NaCl stress than those under Na2CO3 stress, and higher activities of antioxidant enzymes in chloroplasts were accompanied by lower MDA content under NaCl stress. F v/F m, F v/F o and F v′/F m′ all initially increased and then decreased with the decreasing osmotic potential of culture solution, while Φ PSII, qNP and HDR showed a constant increase. F v/F m, F v/F o, Φ PSII and qNP under NaCl stress were also shown to be higher than those under Na2CO3 stress. The present study suggested that acidity played an important role in the hurt toPuccinellia tenuiflora seedlings, which was due to higher activities of antioxidant enzymes, qNP and Φ PSII, and the Na2CO3 resistance to Puccinellia tenuiflora was also supposed to be less than NaCl resistance in present work.  相似文献

The symbiotic association of endophyte fungus, Neotyphodium lolii, and ryegrass improves the ryegrass resistance to drought. This is shown by a 30 % increase in the number of suckers in infected plants (E+), compared to plants lacking endophyte (E−), and by a higher water potential in the E+ than E− plants. The E+ plants have higher stomatal conductance (g s), transpiration rate, net photosynthetic rate (P N), and photorespiratory electron transport rate than the E− plants. The maximal photochemical efficiency (Fv/Fm) and the actual photochemical efficiency (ΦPS2) are not affected by the endophyte fungus. The increase in P N of the E+ plants subjected to water stress was independent from internal CO2 concentration. An increased P N was observed in E+ plants also in optimal water supply. Hence the drought resistance of E+ plants results in increased g s, P N, and photorespiratory electron transport rate. Due to anthropogenic influences, solar UV-B irradiance at the earth’s surface is increasing. To determine the effects of enhanced UV-B radiation on photosynthetic characteristics of Prunus dulcis, two-year-old seedlings of the species were submitted to four levels of UV-B stress, namely 0 (UV-Bc), 4.42 (UV-B1), 7.32 (UV-B2) and 9.36 (UV-B3) kJ m−2 d−1. Effects of UV-B stress on a range of chlorophyll (Chl) fluorescence parameters (FPs), Chl contents and photosynthetic gas-exchange parameters were investigated. UV-B stress promoted an increase in minimal fluorescence of dark-adapted state (F0) and F0/Fm, and a decrease in variable fluorescence (Fv, Fv/Fm, Fv/F0 and F0/Fm) due to its adverse effects on photosystem II (PSII) activity. No significant change was observed for maximal fluorescence of dark-adapted state (Fm). Enhanced UV-B radiation caused a significant inhibition of net photosynthetic rate (P N) at UV-B2 and UV-B3 levels and this was accompanied by a reduction in stomatal conductance (g s) and transpiration rate (E). The contents of Chl a, b, and total Chl content (a+b) were also significantly reduced at increased UV-B stress. In general, adverse UV-B effects became significant at the highest tested radiation dose 9.36 kJ m−2 d−1. The most sensitive indicators for UV-B stress were Fv/F0, Chl a content and P N. Significant P<0.05 alteration in these parameters was found indicating the drastic effect of UV-B radiation on P. dulcis.  相似文献

The changes in photosynthetic efficiency and photosynthetic pigments during dehydration of the resurrection plantSelaginella lepidophylla (from the Chiuhahuan desert, S.W. Texas, USA) were examined under different light conditions. Changes in the photosynthetic efficiency were deduced from chlorophyll a fluorescence measurements (Fo, Fm, and Fv) and pigment changes were measured by HPLC analysis. A small decrease in Fv/Fm was seen in hydrated stems in high light (650 μmol photons·m−2·s−1) but not in low light (50 μmol photons·m−2·s−1). However, a pronounced decline in Fv/Fm was observed during dehydration in both light treatments, after one to two hours of dehydration. A rise in Fo was observed only after six to ten hours of dehydration. Concomitant with the decrease in photosynthetic efficiency during dehydration a rise in the xanthophyll zeaxanthin was observed, even in low-light treatments. The increase in zeaxanthin can be related to previously observed photoprotective non-photochemical quenching of fluorescence in dehydrating stems ofS. lepidophylla. We hypothesize that under dehydrating conditions even low light levels become excessive and zeaxanthin-related photoprotection is engaged. We speculate that these processes, as well as stem curling and self shading (Eickmeier et al. 1992), serve to minimize photoinhibitory damage toS. lepidophylla during the process of dehydration.  相似文献

A yellow leaf colouration mutant (named ycm) generated from rice T-DNA insertion lines was identified with less grana lamellae and low thylakoid membrane protein contents. At weak irradiance [50 μmol(photon) m−2 s−1], chlorophyll (Chl) contents of ycm were ≈20 % of those of WT and Chl a/b ratios were 3-fold that of wild type (WT). The leaf of ycm showed lower values in the actual photosystem 2 (PS2) efficiency (ΦPS2), photochemical quenching (qP), and the efficiency of excitation capture by open PS2 centres 1 (Fv′/Fm′) than those of WT, except no difference in the maximal efficiency of PS2 photochemistry (Fv/Fm). With progress in irradiance [100 and 200 μmol(photon) m−2 s−1], there was a change in the photosynthetic pigment stoichiometry. In ycm, the increase of total Chl contents and the decrease in Chl a/b ratio were observed. ΦPS2, qP, and Fv′/Fm′ of ycm increased gradually along with the increase of irradiance but still much less than in WT. The increase of xanthophyll ratio [(Z+A)/(V+A+Z)] associated with non-photochemical quenching (qN) was found in ycm which suggested that ycm dissipated excess energy through the turnover of xanthophylls. No significant differences in pigment composition were observed in WT under various irradiances, except Chl a/b ratio that gradually decreased. Hence the ycm mutant developed much more tardily than WT, which was caused by low photon energy utilization independent of irradiance.  相似文献

The concept of critical nitrogen concentration(%N c) has been proposed as the minimum%N in shoots required to produce the maximum aerial biomassat a given time. Several authors have shown that%N c declines as a function of aerial biomassaccumulation (W) and the %N cW relationship has been proposed as a diagnostic tool of N statusin different crops, excluding maize. From data obtained in five nitrogenfertilisation experiments in irrigated maize crops, 26 critical data-pointswere selected with a precise statistical procedure. An allometric relationwas fitted and a critical %NW relationshipmodel is proposed in maize as: If W < 1 t ha-1%N c = 3.40 If 1 t ha-1W ≤ 22 t ha-1%N c = 3.40(W)−0.37 The model is applicable to maize crop development between emergenceand silking + 25 days. The model was tested and validated with dataobtained in a network of 17 N fertilisation experiments conducted inFrance under contrasting pedoclimatic conditions. In only nineout of 280 data-points (3.2%), the plant N status was mispredictedwhen ±5% error around %N c wasallowed. A critical N uptake model (Nuc, kg Nha-1) is proposed as Nuc = 34 (W)0.63 A comparison between Nuc and N uptake observedin N treatments giving the maximal grain yields has shown that maizecrops assimilate at least 30 kg N ha-1 in a storage N poolat the silking stage. The significance of the critical%NW and Nu−W relationships is discussed in relation to theoretical models proposed inwhole plant ecophysiology. Different relationships calculated betweenleaf area index and aerial biomass accumulation, and between N uptakeand leaf area were consistent with previous results for other crops.This strengthens the interest of the critical%NW relationship for use as diagnostictool of nitrogen status in maize crops. Experimental study of a glow discharge with electrostatic confinement of electrons is carried out in the vacuum chamber volume V ≈ 0.12 m3 of a technological system “Bulat-6” in argon pressure range 0.005–5 Pa. The chamber is used as a hollow cathode of the discharge with the inner surface area S ≈ 1.5 m2. It is equipped with two feedthroughs, which make it possible to immerse in the discharge plasma interchangeable anodes with surface area S a ranging from ∼0.001 to ∼0.1 m2, as well as floating electrodes isolated from both the chamber and the anode. Dependences of the cathode fall U c = 0.4−3 kV on the pressure p at a constant discharge current in the range I = 0.2−2 A proved that aperture of the electron escape out of the electrostatic trap is equal to the sum S o = S a + S f of the anode surface S a and the floating electrode surface S f . The sum S o defines the lower limit p o of the pressure range, in which U c is independent of p. At p < p o the cathode fall U c grows up dramatically, when the pressure decreases, and the pressure p tends to the limit p ex, which is in fact the discharge extinction pressure. At pp ex electrons emitted by the cathode and the first generation of fast electrons produced in the cathode sheath spend almost all their energy up to 3 keV on heating the anode and the floating electrode up to 600–800°C and higher. In this case the gas in the chamber is being ionized by the next generations of electrons produced in the cathode sheath, their energy being one order of magnitude lower. When S a < (2m/M)1/2 S, where m is the electron mass and M is the ion mass, the anode may be additionally heated by plasma electrons accelerated by the anode fall of potential U a up to 0.5 kV.  相似文献

Photosynthetic parameters of phytoplankton and sea ice algae from landfast sea ice of the Chukchi Sea off Point Barrow, Alaska, were assessed in spring 2005 and winter through spring 2006 using Pulse Amplitude Modulated (PAM) fluorometry including estimates of maximum quantum efficiency (F v/F m), maximum relative electron transport rate (rETRmax), photosynthetic efficiency (α), and the photoadaptive index (E k). The use of centrifuged brine samples allowed to document vertical gradients in ice algal acclimation with 5 cm vertical resolution for the first time. Bottom ice algae (0–5 cm from ice–water interface) expressed low F v/F m (0.331–0.426) and low α (0.098–0.130 (μmol photons m−2s−1)−1) in December. F v/F m and α increased in March and May (0.468–0.588 and 0.141–0.438 (μmol photons m−2s−1)−1, respectively) indicating increased photosynthetic activity. In addition, increases in rETRmax (3.3–16.4 a.u.) and E k (20–88 μmol photons m−2 s−1) from December to May illustrates a higher potential for primary productivity as communities become better acclimated to under-ice light conditions. In conclusion, photosynthetic performance by ice algae (as assessed by PAM fluorometry) was tightly linked to sea ice salinity, temperature, and inorganic nutrient concentrations (mainly nitrogen).  相似文献

Miconia albicans, a common evergreen cerrado species, was studied under field conditions. Leaf gas exchange and pre-dawn leaf water potential (Ψpd) were determined during wet and dry seasons. The potential photosynthetic capacity (P Npmax) and the apparent carboxylation efficiency (ε) dropped in the dry season to 28.0 and 0.7 %, respectively, of the maximum values in the wet season. The relative mesophyll (Lm) and stomatal (Ls) limitations of photosynthesis increased, respectively, from 24 and 44 % in the wet season to 79 and 57 % at the peak of the dry season when mean Ψpd reached −5.2 MPa. After first rains, the P Npmax, ε, and Lm recovered reaching the wet season values, but Ls was maintained high (63 %). The shallow root system growing on stonemason limited by lateral concrete wall to a depth of 0.33 m explained why extreme Ψpd was brought about. Thus M. albicans is able to overcome quickly the strains imposed by severe water stress.  相似文献

Negative effects of osmotically-induced dehydration of two foliose lichen species, Lasallia pustulata and Umbilicaria hirsuta, was studied at physiological (22 °C), low (5 °C) and freezing temperature (−10 °C), using chlorophyll (Chl) fluorescence. In both species, exposure to increasing sucrose concentrations led to a pronounced decrease in potential (FV/FM), and actual (Φ2) quantum yields of photochemical processes in photosystem 2. L. pustulata was more sensitive to osmotic stress, because comparable osmotic dehydration inhibited FV/FM and Φ2 more than in U. hirsuta. Critical concentration of sucrose that fully inhibited photochemical processes of photosynthesis was 2.5 M, which represented water potential (Ψw) of −18.8 MPa. Decrease in background Chl fluorescence (F0) and increase in non-photochemical quenching (qN) revealed two phases of osmotic stress in lichens: phase I with no change (Ψw 0 to −6.6 MPa) and phase II (Ψw −11.3 to −18.8 MPa) typical by substantial change in Chl fluorescence parameters. Effects of thallus anatomy on species-specific response to osmotic dehydration is discussed and attributed to the results obtained by optical microscopy and Chl fluorescence imaging technique.  相似文献

We show that rabbit skeletal RyR channels in lipid bilayers can be activated or inhibited by NO, in a manner that depends on donor concentration, membrane potential and the presence of channel agonists. 10 μm S-nitroso-N-acetyl-penicillamine (SNAP) increased RyR activity at −40 mV within 15 sec of addition to the cis chamber, with a 2-fold increase in frequency of channel opening (F o ). 10 μm SNAP did not alter activity at +40 mV and did not further activate RyRs previously activated by 2 mm cis ATP at +40 or −40 mV. In contrast to the increase in F o with 10 μm SNAP, 1 mm SNAP caused a 2-fold reduction in F o but a 1.5-fold increase in mean open time (T o ) at −40 mV in the absence of ATP. 1 mm SNAP or 0.5 mm sodium nitroprusside (SNP) induced ∼3-fold reductions in F o and T o at +40 or −40 mV when channels were activated by 2 mm cis ATP or in channels activated by 6.5 μm peptide A at −40 mV (peptide A corresponds to part of the II–III loop of the skeletal dihydropyridine receptor). Both SNAP-induced activation and SNAP/SNP-induced inhibition were reversed by 2 mm dithiothreitol. The results suggest that S-Nitrosylation or oxidation of at least three classes of protein thiols by NO each produced characteristic changes in RyR activity. We propose that, in vivo, initial release of NO activates RyRs, but stronger release increases [NO] and inhibits RyR activity and contraction. An amiloride-sensitive, Ca2+-activated nonselective cation (NSC) channel in the apical membrane of fetal rat alveolar epithelium plays an important role in stimulation of Na+ transport by a beta adrenergic agonist (beta agonist). We studied whether Ca2+ has an essential role in the stimulation of the NSC channel by beta agonists. In cell-attached patches formed on the epithelium, terbutaline, a beta agonist, increased the open probability (P o ) of the NSC channel to 0.62 ± 0.07 from 0.03 ± 0.01 (mean ±se; n= 8) 30 min after application of terbutaline in a solution containing 1 mm Ca2+. The P o of the terbutaline-stimulated NSC channel was diminished in the absence of extracellular Ca2+ to 0.26 ± 0.05 (n= 8). The cytosolic Ca2+ concentration ([Ca2+] c ) in the presence and absence of extracellular Ca2+ was, respectively, 100 ± 6 and 20 ± 2 nm (n= 7) 30 min after application of terbutaline. The cytosolic Cl concentration ([Cl] c ) in the presence and absence of extracellular Ca2+ was, respectively, 20 ± 1 and 40 ± 2 mm (n= 7) 30 min after application of terbutaline. The diminution of [Ca2+] c from 100 to 20 nm itself had no significant effects on the P o if the [Cl] c was reduced to 20 mm; the P o was 0.58 ± 0.10 at 100 nm [Ca2+] c and 0.55 ± 0.09 at 20 nm [Ca2+] c (n= 8) with 20 mm [Cl] c in inside-out patches. On the other hand, the P o (0.28 ± 0.10) at 20 nm [Ca2+] c with 40 mm [Cl] c was significantly lower than that (0.58 ± 0.10; P < 0.01; n= 8) at 100 nm [Ca2+] c with 20 mm [Cl] c , suggesting that reduction of [Cl] c is an important factor stimulating the NSC channel. These observations indicate that the extracellular Ca2+ plays an important role in the stimulatory action of beta agonist on the NSC channel via reduction of [Cl] c . In sunflower (Helianthus annuus L.) grown under controlled conditions and subjected to drought by withholding watering, net photosynthetic rate (P N) and stomatal conductance (g s) of attached leaves decreased as leaf water potential (Ψw) declined from −0.3 to −2.9 MPa. Although g s decreased over the whole range of Ψw, nearly constant values in the intercellular CO2 concentrations (C i) were observed as Ψw decreased to −1.8 MPa, but C i increased as Ψw decreased further. Relative quantum yield, photochemical quenching, and the apparent quantum yield of photosynthesis decreased with water deficit, whereas non-photochemical quenching (qNP) increased progressively. A highly significant negative relationship between qNP and ATP content was observed. Water deficit did not alter the pyridine nucleotide concentration but decreased ATP content suggesting metabolic impairment. At a photon flux density of 550 μmol m−2 s−1, the allocation of electrons from photosystem (PS) 2 to O2 reduction was increased by 51 %, while the allocation to CO2 assimilation was diminished by 32 %, as Ψw declined from −0.3 to −2.9 MPa. A significant linear relationship between mean P N and the rate of total linear electron transport was observed in well watered plants, the correlation becoming curvilinear when water deficit increased. The maximum quantum yield of PS2 was not affected by water deficit, whereas qP declined only at very severe stress and the excess photon energy was dissipated by increasing qNP indicating that a greater proportion of the energy was thermally dissipated. Leaves ofNerium oleander L. plants, which had been previously kept in a shaded glasshouse for at least two months, were fed 1 mM dithiothreitol (DTT) through their petioles, either for 12h in darkness (overnight) or for 2h in low light (28 μmol photons·m−2·s−1), in each case followed by a 3-h exposure to high light (1260 μmol photons·m−2·s−1). During exposure to high light, violaxanthin became converted to zeaxanthin in control leaves, to which water had been fed, whereas zeaxanthin did not accumulate in leaves treated with DTT. Total carbon gain was not reduced by DTT during the photoinhibitory treatment. Exposure to high light led to a decrease in the photochemical efficiency of photosystem II, measured as the ratio of variable over maximum fluorescence emission,F v/F M, at both 298 K and 77K. The decrease was much more pronounced in the presence of DTT, mainly owing to a sustained increase in the instantaneous fluorescence,F o. By contrast, in the control leaves,F o determined immediately after the high-light treatment showed a transient decrease below theF o value obtained before the onset of the photoinhibitory treatment (i.e. after 12 h dark adaptation), followed by a rapid return (within seconds) to this original level ofF o during the following recovery period in darkness. Incubation of leaves with DTT led to large, sustained decreases in the photon-use efficiency of photosynthetic O2 evolution by bright light, whilst the capacity of photosynthetic O2 evolution at light and CO2 saturation was less affected. In the control leaves, only small reductions in the photon yield and in the photosynthetic capacity were observed. These findings are consistent with previous suggestions that zeaxanthin, formed in the xanthophyll cycle by de-epoxidation of violaxanthin, is involved in protecting the photosynthetic apparatus against the adverse effects of excessive light.  相似文献

Six months old in vitro-grown Anoectochilus formosanus plantlets were transferred to ex-vitro acclimation under low irradiance, LI [60 μmol(photon) m−2 s−1], intermediate irradiance, II [180 μmol(photon) m−2 s−1], and high irradiance, HI [300 μmol(photon) m−2 s−1] for 30 d. Imposition of II led to a significant increase of chlorophyll (Chl) b content, rates of net photosynthesis (P N) and transpiration (E), stomatal conductance (g s), electron transfer rate (ETR), quantum yield of electron transport from water through photosystem 2 (ΦPS2), and activity of ribulose-1,5-bisphosphate carboxylase/ oxygenase (RuBPCO, EC 4.1.1.39). This indicates that Anoectochilus was better acclimated at II compared to LI treatment. On the other hand, HI acclimation led to a significant reduction of Chl a and b, P N, E, g s, photochemical quenching, dark-adapted quantum efficiency of open PS2 centres (Fv/Fm), probability of an absorbed photon reaching an open PS2 reaction centre (Fv′/Fm′), ETR, ΦPS2, and energy efficiency of CO2 fixation (ΦCO2PS2). This indicates that HI treatment considerably exceeded the photo-protective capacity and Anoectochilus suffered HI induced damage to the photosynthetic apparatus. Imposition of HI significantly increased the contents of antheraxanthin and zeaxanthin (ZEA), non-photochemical quenching, and conversion of violaxanthin to ZEA. Thus Anoectochilus modifies its system to dissipate excess excitation energy and to protect the photosynthetic machinery.  相似文献
