This review commemorates and examines the significance of the work of Isobel Gallaud more than 100 years ago that first established the existence of distinct structural classes (Arum-type and Paris-type) within arbuscular mycorrhizal (AM) symbioses. We add new information from recent publications to the previous data last collated 10 years ago to consider whether any patterns have emerged on the basis of different fungal morphology within plant species or families. We discuss: (1) possible control exerted by the fungus over AM morphology; (2) apparent lack of plant phylogenetic relationships between the classes; (3) functions of the interfaces in different structural classes in relation to nutrient transfer in particular; and (4) the occurrence of plants with both of the major classes, and with intermediate AM structures, in different plant habitats. We also give suggestions for future research to help remove uncertainties about the functional and ecological significance of differences in AM morphology. Lastly, we urge retention of the terms Arum- and Paris-type, which are now well recognised by those who study AM symbioses.  相似文献

Purified basic chitinase or #-1,3-glucanase or a combination of the two enzymes were applied to hyphae of the arbuscular mycorrhizal fungus Glomus mosseae grown in vitro. Chitinase applied to the hyphal tip produced an inhibition of hyphal extension, lysis of the apex and alterations of the growth pattern of the fungus. No effect was observed, however, when chitinase was applied to subapical parts of the hyphae or when glucanase was applied to any part of the hyphae. Application of a combination of the two enzymes to the hyphal tip produced an effect similar to that of chitinase alone.  相似文献

Arbuscular mycorrhizal fungal (AMF) communities were established in pots using fungal isolates from a single field in Switzerland. It was tested whether multispecies mixtures provided more phosphorus and supported greater plant growth than single AMF species. Two host plants, medic (Medicago truncatula) and leek (Allium porrum), were inoculated with three AMF species (Glomus mosseae, G. claroideum and G. intraradices), either separately or in mixtures. The composition of the AMF communities in the roots was assessed using real-time PCR to determine the copy number of large ribosomal subunit genes. Fungal communities in the roots were usually dominated by one AMF species (G. mosseae). The composition of the communities depended on both plant identity and the time of harvest. Leek colonized by a mixture of G. claroideum and G. intraradices acquired more P than with either of the two AMF separately. Direct evidence is provided for functional complementarity among species within the AMF community colonizing a single root system. Competition among the species poses a major challenge in interpreting experiments with mixed inoculations, but this is greatly facilitated by use of real-time PCR.  相似文献

This review presents the point of view that arbuscular mycorrhizal fungi (AMF) do not play a vital role in the nutrition and growth of plants in many production-orientated agricultural systems. Highly available soil P often limits AM colonisation and causes the C-costs to the host to outweigh any benefits from colonisation. Even when P availability is low and AM colonisation levels are high, as may occur in organic and biodynamic agricultural systems, AMF may not always contribute to plant growth for reasons not yet understood. AM fungal activity may also be greatly limited by soil fumigation, non-responsive plant varieties, or rotations based primarily on non-mycorrhizal crops or crops of low AM dependency. Thus, profitability may sometimes be enhanced by management practices, such as tillage and P-fertilisation, which limit AM colonisation. Manipulation of agricultural systems to favour AMF must occur only if there is clear evidence that AMF make a positive contribution to yield or are vital for maintenance of ecosystem health and sustainability. A crucial role for AMF in soil structural stability or in enhancing micronutrient concentrations in produce may be sufficient evidence and may eventually compel consideration of AMF responsiveness when breeding new crop varieties.  相似文献

Several available models of arbuscular mycorrhizal infection are based on fitting % infection to a logistic curve and then relating the various parameters to biological functions. I suggest here that this direction is misleading. Percent infection is a value derived from the growth of two interdependent but distinct organisms, each of which is seeking to maximize its own growth and survival. I suggest that two-organism models, such as those derived from Lotka-Volterra equations, are more useful for understanding the biology and functioning of mycorrhizae. The identification and quantification of cyclohexenone glycoside derivatives from the model legume Lotus japonicus revealed far higher levels than expected according to the stoichiometric relation to another, already determined carotenoid cleavage product, i.e., mycorradicin. Mycorradicin is responsible for the yellow coloration of many arbuscular mycorrhizal (AM) roots and is usually esterified in a complex way to other compounds. After liberation from such complexes it has been detected in AM roots of many, but not of all plants examined. The non-stoichiometric occurrence of this compound compared with other carotenoid cleavage products suggested that carotenoid biosynthesis might be activated upon mycorrhization even in plant species without detectable levels of mycorradicin. This assumption has been supported by inhibition of a key enzyme of carotenoid biosynthesis (phytoene desaturase) and quantification of the accumulating enzymic substrate (phytoene). Our observations suggest that the activation of carotenoid biosynthesis in AM roots is a general phenomenon and that quantification of mycorradicin is not always a good indicator for this activation.  相似文献

Activated carbon (AC) is widely used in ecological studies to elucidate the role of allelopathic substances in interspecific plant competition. However, by adsorbing chemical signalling compounds AC may also have negative effects on plants with symbiosis partners such as arbuscular mycorrhizal fungi and rhizobia. Here we test whether addition of AC has detrimental effects on the mycorrhizal root colonization of the native forb Plantago lanceolata and the exotic legume Lupinus polyphyllus, the nodulation of L. polyphyllus, and the nutrient uptake and growth of the plants growing in intra- and interspecific competition. Allelopathic effects probably occurred in the germination and seedling establishment phase when P. lanceolata suffered from the presence of L. polyphyllus. However, this negative effect of L. polyphyllus on P. lanceolata was not ameliorated by AC addition. AC negatively affected L. polyphyllus root biomass in week 4, and root and shoot biomass of P. lanceolata in week 9 of the experiment; both effects were independent of the presence and absence of the competing plant species. Mycorrhizal root colonization of both plant species was reduced in the presence of AC, although the effect tended to be stronger for L. polyphyllus. No significant effect of AC on the nodulation of L. polyphyllus was detected. P. lanceolata was the superior competitor and led to reduced biomasses of L. polyphyllus in interspecific competition. We conclude that AC can reduce the mycorrhization and performance of plants which may lead to changes in interspecific competition without the involvement of allelopathy. Contrary to former studies the AC used in our study did not enhance the nutrient availability for the plants, but reduced plant growth and mycorrhization. We suggest that the nutrient properties of the used AC are of crucial importance for the direction and the mechanisms of the effects and should always be reported.  相似文献

A very wide range of plant organ movements have been described and yet it is not clear how each of them is related to the others. This uncertainty has had two undesirable consequences. Firstly, some workers have accepted the idea of a vague unity in the area and have subsequently been misled by information obtained in one system and applied without adequate justification to a study of a quite different system. Secondly, some researchers have evoked a possible diversity to explain why a particular mechanistic explanation may continue to be valid even when the model fails to explain events of a very similar nature in a slightly different system. We argue that this confusion has resulted from a classification of organ movements which has been based on functional rather than on mechanistic considerations. Mechanistic unity is to be expected on evolutionary grounds. This unity, however, may apply only to certain elements of the stimulus-response chain, at certain levels of organization. It follows from this that in seeking this unity, comparisons should be made between equivalent elements of the stimulus-response chain at the same level of organization in different systems. Only when this is done will theories built around the concept of unity provoke meaningful discussion.  相似文献

Symbioses involving arbuscular mycorrhizal fungi (AMF) are among the most important ecological associations for many plant species. The diversity of AMF associated with ferns, however, remains poorly studied. Using recently designed Glomus-specific primers, we surveyed the AMF community associated with ferns from deciduous, broad-leaved second-growth forest habitats at the eastern edge of the piedmont region of central Virginia, USA. Results indicate that this molecular approach may be a useful tool for detecting AMF in ferns compared to traditional techniques based on morphology. Over 30 potential fungal ribotypes were identified from eight fern species using denaturing gradient gel electrophoresis. Fungal ribotypes were found to differ widely in terms of (1) the number of fern partners with which they interact and (2) their relative frequency within each fern. Sequence analysis of fungal isolates from three species of fern indicated that the primers were generally highly specific for Glomus species but some non-target DNA was also amplified. Cloned polymerase chain reaction (PCR) products from Polystichum acrostichoides and Osmunda regalis revealed several phylogenetically distinct Glomus species. A single Glomus species was identified in the cloned PCR products from Botrychium virginianum. These findings challenge the hypothesis that the extent or degree of fern–fungal symbiosis is somehow tied to root complexity. Environmental factors appear to influence the suite of AMF that form partnerships with ferns. Some species of fern from similar habitats associated with dissimilar fungal partners (e.g., P. acrostichoides and Athyrium filix-femina var. asplenioides), whereas others harbored uniform fungal communities (e.g., Asplenium platyneuron). The significance of these data in terms of ecological and evolutionary dynamics of the AMF–fern symbiosis is discussed. Aims Our study quantified the combined effects of fertilization and inoculation with arbuscular mycorrhizal fungi (AMF) on grain yield and allocation of biomass and nutrients in field-grown rice (Oryza sativa L.).Methods A two-factor experiment was conducted at a field site in northeast of China (in Shuangcheng, Heilongjiang Province, Songhua River basin): six nitrogen–phosphorus–potassium fertilizer levels were provided (0, 20, 40, 60, 80 and 100% of the local norm of fertilizer supply), with or without inoculation with Glomus mosseae. At maturity, we quantified the percentage of root length colonization by AMF, grain yield, shoot:root ratios, shoot N and P contents and nutrients allocated to panicles, leaves and stems.Important findings As expected, inoculation resulted in greatly increased AMF colonization, which in turn led to higher shoot:root ratios and greater shoot N contents. Shoot:root ratios of inoculated rice increased with increasing fertilization while there was a significant interaction between fertilization and inoculation on shoot:root ratio. Additionally, AMF inoculation increased panicle:shoot ratios, panicle N:shoot N ratios and panicle P:shoot P ratios, especially in plants grown at low fertilizer levels. Importantly, inoculated rice exhibited higher grain yield, with the maximum improvement (near 62%) at the lower fertilizer end. Our results showed that (i) AMF-inoculated plants conform to the functional equilibrium theory, albeit to a reduced extent compared to non-inoculated plants and (ii) AMF inoculation resulted in greater allocation of shoot biomass to panicles and increased grain yield by stimulating N and P redistribution to panicles.  相似文献

Consistency of response to arbuscular mycorrhizal (AM) inoculation is required for efficient use of AM fungi in plant production. Here, we found that the response triggered in plants by an AM strain depends on the properties of the soil where it is introduced. Two data sets from 130 different experiments assessing the outcome of a total of 548 replicated single inoculation trials conducted either in soils with a history of (1) high input agriculture (HIA; 343 replicated trials) or (2) in more pristine soils from coffee plantations (CA; 205 replicated trials) were examined. Plant response to inoculation with different AM strains in CA soils planted with coffee was related to soil properties associated with soil types. The strains Glomus fasciculatum-like and Glomus etunicatum-like were particularly performant in soil relatively rich in nutrients and organic matter. Paraglomus occultum and Glomus mosseae-like performed best in relatively poor soils, and G. mosseae and Glomus manihotis did best in soils of medium fertility. Acaulospora scrobiculata, Diversispora spurca, G. mosseae-like, G. mosseae and P. occultum stimulated coffee growth best in Chromic, Eutric Alluvial Cambisol, G. fasciculatum-like and G. etunicatum-like in Calcaric Cambisol and G. manihotis, in Chromic, Eutric Cambisols. Acaulospora scrobiculata and Diversispora spurca strains performed best in Chromic Alisols and Rodic Ferralsols. There was no significant relationship between plant response to AM fungal strains and soil properties in the HIA soil data set, may be due to variation induced by the use of different host plant species and to modification of soil properties by a history of intensive production. Consideration of the performance of AM fungal strains in target soil environments may well be the key for efficient management of the AM symbiosis in plant production.  相似文献

As the newest plant hormone, strigolactone research is undergoing an exciting expansion. In less than five years, roles for strigolactones have been defined in shoot branching, secondary growth, root growth and nodulation, to add to the growing understanding of their role in arbuscular mycorrhizae and parasitic weed interactions.1 Strigolactones are particularly fascinating as signaling molecules as they can act both inside the plant as an endogenous hormone and in the soil as a rhizosphere signal.2-4 Our recent research has highlighted such a dual role for strigolactones, potentially acting as both an endogenous and exogenous signal for arbuscular mycorrhizal development.5 There is also significant interest in examining strigolactones as putative regulators of responses to environmental stimuli, especially the response to nutrient availability, given the strong regulation of strigolactone production by nitrate and phosphate observed in many species.5,6 In particular, the potential for strigolactones to mediate the ecologically important response of mycorrhizal colonization to phosphate has been widely discussed. However, using a mutant approach we found that strigolactones are not essential for phosphate regulation of mycorrhizal colonization or nodulation.5 This is consistent with the relatively mild impairment of phosphate control of seedling root growth observed in Arabidopsis strigolactone mutants.7 This contrasts with the major role for strigolactones in phosphate control of shoot branching of rice and Arabidopsis8,9 and indicates that the integration of strigolactones into our understanding of nutrient response will be complex. New data presented here, along with the recent discovery of phosphate specific CLE peptides,10 indicates a potential role for PsNARK, a component of the autoregulation of nodulation pathway, in phosphate control of nodulation.  相似文献

Some host individuals tend to acquire parasites at a much faster rate than do others--a consequence of heterogeneities in susceptibility and/or exposure. This is termed 'overdispersion' and, as for many other statistical phenomena, the degree of overdispersion often conforms to a 20/80 rule, where 20% of the host population is responsible for approximately 80% of the parasite transmission. But which are the hosts driving so much of the dynamics of an infectious disease? If host individuals at the tail of the frequency distribution can be identified by some common label, controlling parasitic diseases would be much easier. In two recent papers, Perkins et al. and Ferrari et al. have shown that male hosts are much more important than female hosts in the transmission of parasites.  相似文献

To study whether the biomass of soil microorganisms in a boreal Pinus sylvestris-Vaccinium vitis-idaea forest was limited by the availability of carbon or nitrogen, we applied sucrose from sugar cane, a C4 plant, to the organic mor-layer of the C3–C dominated soil. We can distinguish between microbial mineralization of the added sucrose and respiration of endogenous carbon (root and microbial) by using the C4-sucrose as a tracer, exploiting the difference in natural abundance of 13C between the added C4-sucrose (13C –10.8) and the endogenous C3–carbon (13C –26.6 ). In addition to sucrose, NH4Cl (340 kg N ha–1) was added factorially to the mor-layer. We followed the microbial activity for nine days after the treatments, by in situ sampling of CO2 evolved from the soil and mass spectrometric analyses of 13C in the CO2. We found that microbial biomass was limited by the availability of carbon, rather than nitrogen availability, since there was a 50% increase in soil respiration in situ between 1 h and 5 days after adding the sucrose. However, no further increase was observed unless nitrogen was also added. Analyses of the 13C ratios of the evolved CO2 showed that increases in respiration observed between 1 h and 9 days after the additions could be accounted for by an increase in mineralization of the added C4–C.  相似文献

Ryanodine receptors (RyRs) are colossal membrane protein complexes that reside in the endoplasmic reticulum of skeletal and cardiac muscle myocytes and neurons, in addition to many non-excitable cells. They comprise high-conductance ion channels that mediate the massive release of Ca2+ ions from the endoplasmic reticulum into the cytoplasm. This is the trigger for contraction during each muscle excitation-contraction coupling cycle. Individual RyRs are believed to network with other RyRs indirectly, through diffusion of released Ca2+ ions, namely the Ca2+-induced Ca2+ release phenomenon. However, RyRs can intrinsically organize into a regular array resembling a distinctive checkerboard pattern, with each square-shaped receptor appearing to abut four neighbours at each corner. In this opinion article, we describe recent data showing structural interactions between RyR oligomers in reconstituted arrays, and we suggest that this provides strong evidence for direct inter-RyR communication through a novel, allosteric regulatory mechanism.  相似文献

The size of plant cells is determined by genetic, structural and physical factors as well as by internal and external signals. Our knowledge of the molecular mechanisms of these controls is still rudimentary. Recent studies indicate that ploidy level exerts an important control on cell size. By increasing ploidy, endoreduplication may allow cells to reach extraordinary sizes. This process is widespread in plants and may provide a means to manipulate the cell volume.  相似文献
