Aporocotylids (fish blood flukes) are emerging as pathogens of fishes in both marine and freshwater aquaculture. Efforts to control these parasites are hampered by a lack of life cycle information. Here we report on the life cycle of Cardicola forsteri, which is considered a significant pathogen in southern bluefin tuna, Thunnus maccoyi, ranched in South Australia. We surveyed polychaetes, bivalves and gastropods from sites close to tuna pontoons. Infections consistent with the Aporocotylidae were found in terebellid polychaetes, a single Longicarpus modestus and five individuals of Reterebella aloba. All infections were comprised of hundreds of sporocysts in the body cavity of the host, each filled with developing and mature cercariae. Sequences of ITS-2 and lsrDNA from the infection from L. modestus were a perfect match with those of adult C. forsteri from T. maccoyi. This life cycle link is considered confirmed but it is possible that additional terebellid species are infected in South Australia; equally, other species of intermediate host are likely to be involved in other parts of the range of this cosmopolitan trematode. Sequences of the species from R. aloba did not match a known adult but phylogenetic analysis of lsrDNA suggests that it is also a species of Cardicola Short, 1953. These findings show that terebellid polychaetes are a major host group for marine aporocotylids, especially given that Cardicola is the largest marine aporocotylid genus. The two cercarial types are among the smallest known for the family and are unusual, but not unique, in having short, simple tails. We speculate that the form of the tail means that these cercariae are not active swimmers and are thus heavily dependent on currents for dispersal. Control of this parasite might be effected by moving the tuna pontoons appropriate distances to avoid encounter with current-dispersed cercariae, or by increasing the separation of the nets from the sea floor, either by raising the nets or moving to deeper water.  相似文献

Fish blood flukes (Aporocotylidae) are important pathogens of farmed finfish around the world. Among them, Cardicola spp. infecting farmed tuna are considered to be serious threats to tuna farming and have received tremendous attention. We conducted periodical samplings at a tuna farming site in Japan between January and May, 2015 to determine the life cycle of Cardicola spp. We collected over 4700 terebellid polychaetes from ropes, floats and frames of tuna culture cages and found nearly 400 infected worms. Sporocysts and cercariae found in Nicolea gracilibranchis were genetically identified as Cardicola orientalis by 28S and ITS2 ribosomal DNA sequences. This was the first discovery of the intermediate host for this parasite species. Infection prevalence and the abundance of N. gracilibranchis significantly varied between sampling points and the highest number of infected terebellids were collected from ropes. We also demonstrated morphologically and molecularly that asexual stages found in a single Amphitrite sp. (Terebellidae) and adult worms isolated from farmed juvenile tuna were Cardicola forsteri. This is the first report of C. forsteri in Pacific bluefin tuna (PBT) Thunnus orientalis in Japan. Our results demonstrated that all three species of Cardicola orientalis, C. forsteri and Cardicola opisthorchis exist in Japanese farmed PBTs and that they all use terebellid polychaetes as the intermediate hosts.  相似文献

The diets of 1219 southern bluefin tuna, Thunnus maccoyii, from inshore (shelf) and offshore (oceanic) waters off eastern Tasmania were examined between 1992 and 1994. Immature fish (< 155 cm fork length) made up 88% of those examined. In all, 92 prey taxa were identified. Inshore, the main prey were fish (Trachurus declivis and Emmelichthys nitidus) and juvenile squid (Nototodarus gouldi). Offshore, the diversity was greater, reflecting the diversity of micronekton in these waters. Interestingly, macrozooplankton prey (e.g. Phronima sedentaria) were prevalent in tuna > 150 cm. The offshore tuna, when in subantarctic waters, ate relatively more squid than when in the East Australia Current. In the latter, fish and crustacea were more important, although there were variations between years. No relationship was found between either prey type or size with size of tuna. Feeding was significantly higher in the morning than at other times of the day. The mean weight of prey was significantly higher in inshore-caught tuna than in those caught offshore. We estimated that the mean daily ration of southern bluefin tuna off eastern Tasmania was 0.97% of wet body weight day−1. However, the daily ration of inshore-caught tuna was ∼ 3 times higher (2.7%) than for tuna caught offshore (0.8%) indicating that feeding conditions on the shelf were better than those offshore. Our results indicate that the inshore waters of eastern Tasmania are an important feeding area for, at least, immature southern bluefin tuna. Near reef abundance and coast–offshore distribution of tuna larvae around Minicoy Island were studied based on three cruises carried out on board FORV Sagar Sampada during 2014–15. The samples were collected by oblique tows using bongo twin nets (300 micron) for three seasons (Fall inter-monsoon (FIM), Spring inter-monsoon (SIM) and Summer monsoon (SM)) in six on–offshore stations ranging from 1 to 20?km off-shore of the Minicoy. Results indicated that the tuna larvae aggregate in the near reef waters and total tuna larvae abundance (all the species combined) shows a decreasing trend towards off-shore. Tuna larvae are found to be more abundant during SM and FIM. Comparison of the species-wise variation in tuna larvae for one season (SM) indicated that the larvae of Euthunnus affinis (32.5%) dominated among the tuna species, followed by Auxis spp. (31.6%), Thunnus spp. (3.9%) and Katsuwonus pelamis (2.7%). A pattern of highest larvae abundance in the near reef areas was observed except for Thunnus larvae. GAM indicated that mean seawater temperature and mean seawater salinity are the major environmental factors that significantly influence the larvae abundance, particularly for Thunnus followed by Katsuwonus pelamis, E. affinis and Auxis spp. The study confirms that the environmental parameters show high variation between stations and among transects as a result of the complex oceanographic characteristics around the islands. Further studies with better understanding of the larvae dynamics and their association with the environmental parameters including physical processes are essential to elucidate these relationships more clearly.  相似文献

In this study, adaptive immune response was investigated in farmed southern bluefin tuna, Thunnus maccoyii, infected with a sanguinicolid Cardicola forsteri. A cohort (Cohort2005) of southern bluefin tuna was sampled between March 2005 and August 2006. Samples were taken at the transfer of wild caught tuna to sea cages and then at regular intervals. Parasite intensity, abundance and prevalence data were recorded. An ELISA was developed to detect and quantify an antibody response against the blood fluke in southern bluefin tuna serum. Intensity and prevalence of the blood fluke were shown to peak in May 2005 at 10.9 flukes per infected fish (SE = 1.72) and 97.5% prevalence and then decreased to low prevalence (10%) and intensity (1.0). There were no significant changes in prevalence or intensity in 2006. Antibody titres and seroprevalence increased from 1.37 U μl−1 and 10% at transfer in March 2005 to reach a peak in December 2005 of 25.86 U μl−1 (SE = 6.26 U μl−1) and 66.66%. No significant changes were observed in antibody titres for the same cohort of fish during 2006. Parasitological and serological values from Cohort2005 were compared to a 2006 cohort (Cohort2006) in March 2006 and August 2006 to determine if prior infection in Cohort2005 elicited any protection against infection in 2006. Although significant differences were not observed in intensities between cohorts it was shown that Cohort2005 had significantly lower abundances and prevalences of blood fluke infection than Cohort2006. Although there was no significant difference in mean antibody titres between cohorts in March 2006, the mean antibody titre of Cohort2006 was significantly greater than that of Cohort2005 in August 2006. No significant differences were observed in seroprevalence. This is one of the few studies to demonstrate the development of acquired resistance in fish against a parasite in an aquaculture environment under natural infection conditions.  相似文献

Natural Caligus elongatus Nordmann infections of wild coastal fishes on the Norwegian south east coast were monitored at various times of the year from 2002 to 2004. The prevalence for all coastal fish (n = 4427) pooled was 15%, and there were great differences between fish species and seasons. Lumpfish Cyclopterus lumpus L. spawners were the most infected fish, with a prevalence of 61% and a median intensity of 4 lice fish(-1), whereas gadids had a mean prevalence of 19% and a median infection of 1 to 2 lice fish(-1). Sea trout Salmo trutta L. and herring Clupea harengus L. carried C. elongatus at prevalence values of 29 and 21%, respectively. The results were compared with infection data for immature North Sea lumpfish. Lumpfish spawners caught on the coast in March to April had fewer lice than North Sea lumpfish in July. Spawners carried mostly adult lice, as did coastal fish hosts in May to June. The low development rates of lice at low spring temperatures and new genetic data suggest that the May to June adult lice could not have been offspring of the March to April lice, indicating transfer of adult lice to coastal fish. Most coastal fish species appeared to acquire new C. elongatus infections between May to June and September. The relatively high numbers of chalimii on North Sea lumpfish suggest that offshore fish sustain an oceanic population of this louse species.  相似文献

Synopsis Four species of nototheniid fish were sampled from below the sea ice near Cape Armitage, McMurdo Sound:Pagothenia borchgrevinki from just below the ice 1.5 km offshore,Trematomus bernacchii, Trematomus hansoni andTrematomus centronotus from off the bottom in about 20 m of water near the shore. Scale worms and isopods were conspicuous non-planktonic prey, and present in the three benthic fish species. The planktonic pteropod molluscLimacina helicina was numerically common in all four species of fish. The planktonic hyperiid amphipodHyperiella dilatata was also found in all fish species. WhereasP. borchgrevinki is planktivorous in accord with its pelagic habit, theTrematomus spp. clearly also feed on plankton from the water column.T. hansoni is particularly planktivorous, taking smaller copepods thanP. borchgrevinki.  相似文献

Infestations of blood flukes of the genus Cardicola have been observed in juvenile Pacific bluefin tuna (PBT) cultured in Japan. Infected fish harbor large numbers of parasite eggs in their gills. Although the link between blood fluke infection and juvenile mortality is not clear, accumulation of parasite eggs appears to be pathogenic to the fish. We investigated the origins, general morphology/distribution, and histopathology of these eggs in artificially produced 0 yr old PBT. Dead and live fish were sampled on several occasions from two culture facilities in Wakayama prefecture, Japan. The number of eggs in each gill filament was enumerated under a microscope. In addition, we estimated the total number of eggs by dissolving the gills in a weak NaOH solution. We observed two morphologically distinct egg types in the gill filaments, smaller, oval shaped eggs in the gill lamellae and larger, crescent shaped eggs that occurred primarily in the filamentary arteries. Based on the ITS2 sequence, the ovoid and crescent shaped eggs were identified as C. orientalis and C. opisthorchis, respectively. Eggs of the former species were more abundant (maximum: 6400 per filament) than the latter (maximum: 1400), but the number was highly variable among filaments. The eggs of the latter species were relatively evenly distributed among the filaments. In a heavily infected individual, we estimated a total of >4.5 million eggs were present in the gills on one side of the fish. The number of eggs from the two species was positively correlated to each other and the dead fish tended to harbor more eggs than the live fish. Histological observation revealed host responses around the eggs, including encapsulation by fibroblasts and nodule formation, as seen in response to other aporocotylid eggs. In addition, we observed widespread fusion of gill lamellae and blockage of the filamentary arteries in some instances. Our results provide information that can be used for routine diagnosis of Cardicola blood flukes in cultured tuna and suggest they represent a risk to juvenile PBT.  相似文献

Ranched southern bluefin tuna Thunnus maccoyii were fed baitfishes supplemented with vitamins (predominantly E and C) or vitamins and immunostimulants, nucleotides and β-glucans, over 12 weeks after transfer and monitored for enhancement in immune response, health and performance through their 19 week grow-out period. Fish from two different tows were sampled separately at three different sampling points: at transfer to grow-out pontoons, at 8 weeks post-transfer and at harvest, 19 weeks post-transfer. Lysozyme activity was enhanced during vitamin supplementation compared to control fish. Performance (i.e. survival, condition index and crude fat), health (i.e. blood plasma variables including pH, osmolality, cortisol, lactate and glucose) and alternative complement activity were not commonly improved through diet supplementation. There were some tow-specific improvements in performance through vitamin supplementation including survival, selected parasite prevalence and intensity, and alternative complement activity. Immunostimulant supplementation also showed a tow-specific improvement in plasma cortisol level. Tow-specific responses may suggest that life history, previous health condition and husbandry can affect the success of vitamin and immunostimulant enhancement of immune response, health and performance of ranched T. maccoyii.  相似文献

In some years, large numbers of Caligus chiastos have been observed on the external surfaces of southern bluefin tuna, particularly on the head and eyes, in some sea cages in Spencer Gulf, Australia. As no epidemiological data were available, we monitored sea lice on tuna (N = 130) in 4 research cages sampled at 6 wk intervals during the 2005 farming season. No lice were observed on a sample of 10 wild-caught tuna when the cohort was transferred to cages in early April. By late May more than half the sampled tuna (22 of 40) were infected, with up to 42 parasites; we also recorded one unidentified Caligus sp. at this time. In early July the number of tuna infected with lice declined to 10%; in the final sample in late August none were detected. Prevalence in May was significantly higher than on other dates (p < or = 0.001), whereas mean abundances did not differ significantly (p > 0.05). The decline in prevalence corresponded with a seasonal fall in temperature, from ca. 17 degrees C in May to 14 degrees C in August. Counts of lice at the peak of infection were associated with the severity of eye damage (Spearman's rank correlation coefficient, r(S,38df) = 0.654, p < 0.001); this may be because lice graze on the cornea or because tuna injure their eyes when flashing (rubbing against objects). Counts at this time were also strongly and inversely correlated with the condition index (r(S,38df) = -0.707, p < 0.001). It appears that tuna become infested with adult sea lice via wild teleosts and elasmobranchs attracted to sea cages.  相似文献

Larval nematode infections were investigated in 236 fish from 17 estuarine and near-shore species in southwestern Australia. Larvae of the genus Contracaecum were found in 4 species of fish (Acanthopagrus butcheri, Sillaginodes punctata, Mugil cephalus and Aldrichetta forsteri). The prevalence and intensity of infection was highest in the 2 species of mullet: 81% infected and 9.8 +/- 1.5 larvae fish(-1) for M. cephalus; 100% and 12.7 +/- 3.7 larvae fish(-1) for A. forsteri. There was no evidence of post-mortem migration of worms from the visceral organs to the musculature of the fish.  相似文献

A study was conducted to determine the transfer of eicosapentaenoic acid (EPA, 20:5) and docosahexaenoic acid (DHA, 22:6) from fish oil into goats’ milk. Goats were sequentially offered three diets: control (C) pellets (lucerne hay-oat grain: 60/40 w/w), C plus tuna oil protected against ruminal biohydrogenation (PTO pellets), and C plus unprotected tuna oil (UTO pellets). In supplemented diets, tuna oil constituted 3% of total dry matter (DM), and each supplement was fed for 7 days, with 12 days allowed between the two fish oil feeding periods to minimise carry-over effects. Dry matter intake, milk yield, protein and fat yield were reduced by feeding UTO, but not PTO, pellets. Goats produced ω-3 enriched milk (0.3–0.5% EPA and 1.01–1.12% DHA) when fed either supplement. The rate of transfer of dietary EPA and DHA to milk ranged from 3.5 to 7.6%. Significant transfer of EPA and DHA from tuna oil into goat milk, without deleterious effects on intake or milk yield is possible, provided that the oil supplement is substantially protected against ruminal biohydrogenation.  相似文献

The antioxidant enzyme glutathione peroxidase 4 (GPx4) is capable of reducing complex lipid hydroperoxides in addition to hydrogen peroxide and organic hydroperoxides. Mammals express three GPx4 isoforms that are targeted to nucleoli, mitochondria or cytosol via variable amino termini. To better understand the role of this important antioxidant enzyme in marine finfish, we determined the subcellular localisation of a GPx4 homologue from southern bluefin tuna (Thunnus maccoyii; SBT). We created constructs for the expression of the selenocysteine-to-cysteine mutant of SBT GPx4 (GPx4C) tagged with enhanced green fluorescent protein (EGFP), including or lacking a putative amino-terminal signal peptide, and expressed the fusion proteins in a fish cell line. Fluorescence microscopy revealed that the full-length GPx4C-EGFP fusion protein localised to the trans-Golgi, suggesting that tuna GPx4 may be directed to the secretory pathway. Anti-GFP immunoblotting of cell lysates and proteins from culture media showed that the secretion of SBT GPx4 into the culture medium required an amino-terminal signal peptide. According to available sequence data, the SBT GPx4 isoform studied here is representative of other piscine GPx4 isoforms, suggesting that the secretion of at least one GPx4 isoform may be common amongst teleost fish.  相似文献

The occurrence of 12 helminth and 4 crustacean parasites on young whiting 0–2 years old from an inshore (Port Erin Bay) and an offshore (off Bradda Head) area of the Isle of Man were compared. No parasites were found on whiting 4.5–15 mm long and the intestinal digenean Lecithaster gibbosus Rudolphi together with the larvae of the copepod Caligus rapax M. Edwards were the first parasites to appear in whiting 15–42 mm long. This appears to be the only time during the life history of the whiting that C. rapax is of importance as a parasite. In whiting 6 months-2 years old from off Bradda Head such helminths as the acanthocephalan Echinorhynchus gadi Muller and the digeneans Podocotyle atomon Muller and Cryptocotyle lingua Creplin (metacercariae) were found in significantly fewer fish than in an equivalent group from Port Erin Bay. The differences are correlated with the availability of intermediate hosts and the different food spectra of the two groups of fish and are used to support the theory that inshore and offshore whiting of these age groups form separate populations and that the offshore fish may be more important than the inshore fish in replenishing stocks of older whiting.  相似文献

Beach seines, gill nets and otter trawls were used during 1979–81 to sample extensively the fish fauna of the Peel-Harvey estuarine system. Approximately 145,000 individuals were caught, representing 29 familes and 55 species. The Clupeidae, Teraponidae, Mugilidae, Apogonidae, Atherinidae and Gerreidae were the dominant families, each contributing at least 8.9% to the total catch and together contributing 86.2%. Seasonal catch data and length-frequency distributions were used to help clarify the way in which the estuary was utilised by fish. Nine of the 15 most abundant fish species were marine species which entered the estuary for variable periods while the other six were represented by populations in which the individuals were capable of passing through the whole of their life cycle within the estuary. In order of abundance, the nine marine species were Hyperlophus vittatus, Pelates sexlineatus, Aldrichetta forsteri, Mugil cephalus, Torquigener pleurogramma, Favonigobius lateralis, Pranesus ogilbyi, Gymnapistes marmoratus and Sillago schomburgkii , while the six species with estuarine populations were Apogon rueppellii, Atherinosoma elongata, Nematalosa vlaminghi. Atherinosoma wallacei, Cnidoglanis macrocephalus and Amniataba caudavittatus. The extent and seasonality of the distribution of the species within the estuary and associated river systems varied considerably. Evidence is also presented to show that fish tend to move further away from the shallow banks near the shore with increasing age and size. The fish fauna of the Peel-Harvey is compared with those of Cockburn Sound and the estuaries of the Swan-Avon and Blackwood rivers in Western Australia and with Botany Bay in eastern Australia.  相似文献

Sea lice, especially Lepeophtheirus salmonis and Caligus spp., have the greatest economic impact of any parasite in salmonid fish farming and are also a threat to wild salmonids. Here, I review how the biology and ecology of various louse and host species influence their pathogenicity and epidemiology. Recent discoveries of new species and genotypes emphasize the need for more basic research on louse taxonomy and host preferences. Louse development rates are strongly dependent on temperature, and increasing mean sea temperatures are likely to increase infestation pressure on farms and wild fish, as well as affecting the geographical distribution of hosts and parasites. Despite progress in finding L. salmonis larvae in the plankton and in modelling louse production in several countries, more data on larval behaviour and distribution are required to develop dispersal and transmission models for both L. salmonis and Caligus spp. This knowledge could be used to take measures to reduce the risks of lice affecting farmed and wild fish.  相似文献

The European shelf seas can be divided into regions which have tidally mixed waters and thermally stratified waters. The tidally mixed near shore environments support zooplankton communities dominated by smaller copepods and having large meroplankton contributions. These small copepods (Centropages spp., Temora spp., Acartia spp., Paral Pseudo/Microcalanus spp.) together with the microzooplankton component form a different and more complex food web than the larger copepod/diatom link associated with thermally stratified waters. The copepods Calanus finmarchicus and C. helgolandicus account for over 90% of the copepod dry weight biomass in stratified waters. Although occurring in lower numbers in mixed waters they can still make significant contributions to the biomass. A 31 year time series from the European shelf shows the inter- and intea-annual variability of these species. The basic biology and food web that these two systems support, and the transfer of energy, can result in marked differences in quantity and quality of particulates available as food for fish larvae. Calanus dominated systems allow the primary production to be directed straight through the trophic food chain (diatoms/Calanus/fish larvae) while the near shore communities of smaller copepods limit the amount of energy being transferred to the higher trophic levels. Eighty-two Longhurst Hardy Plankton Recorder hauls were used as the data base for this study. In all cases the zooplankton was dominated by copepods both in numbers and biomass accounting for > 80% of total zooplankton dry weight in the Irish Sea, Celtic Sea, shelf edge of the Celtic Sea and the northern and southern North Sea in Spring.  相似文献

Beluga whales (Delphinapterus leucas) are the most abundant odontocetes in Arctic waters and are thus thought to influence food web structure and function. The diet of the Beaufort Sea beluga population is not well known, partly due to the inherent difficulty of observing feeding behaviour in Arctic marine cetaceans. To determine which prey items are critical to the Beaufort Sea beluga diet we first examine and describe the Mackenzie Delta and Beaufort Sea food web using fatty acid analyses. Fatty acid profiles effectively partitioned prey items into groups associated with their habitat and feeding ecology. Next, the relative contribution of various prey items to beluga diet was investigated using fatty acids. Finally, beluga diet variability was examined as a function of body size, a known correlate of habitat use. Beluga appeared to feed predominantly on Arctic cod (Boreogadus saida) collected from near shore and offshore regions. Size related dietary differences suggested larger sized beluga preferred offshore Arctic cod given the shared high levels of long chain monounsaturates, whereas smaller sized beluga appeared to feed on prey in their near shore habitats that included near shore Arctic cod. The presence of Arctic cod groups in shallow near shore and deep offshore habitats may facilitate the behavioural segregation of beluga habitat use as it relates to their size and resource requirements. Given Arctic cod are a sea ice associated fish combined with the accelerated sea ice loss in this region, beluga whales may need to adapt to new dietary regimes.  相似文献

Knowledge of spawning behaviour and fecundity of fish is important for estimating the reproductive potential of a stock and for constructing appropriate statistical models for assessing sustainable catch levels. Estimates of length-based reproductive parameters are particularly important for determining potential annual fecundity as a function of fish size, but they are often difficult to estimate reliably. Here we provide new information on the reproductive dynamics of southern bluefin tuna (SBT) Thunnus maccoyii through the analysis of fish size and ovary histology collected on the spawning ground in 1993–1995 and 1999–2002. These are used to refine previous parameter estimates of spawning dynamics and investigate size related trends in these parameters. Our results suggest that the small SBT tend to arrive on the spawning ground slightly later and depart earlier in the spawning season relative to large fish. All females were mature and the majority were classed as spawning capable (actively spawning or non-spawning) with a very small proportion classed as regressing. The fraction of females spawning per day decreased with fish size, but once females start a spawning episode, they spawned daily irrespective of size. Mean batch fecundity was estimated directly at 6.5 million oocytes. Analysis of ovary histology and ovary weight data indicated that relative batch fecundity, and the duration of spawning and non-spawning episodes, increased with fish size. These reproductive parameter estimates could be used with estimates of residency time on the spawning ground as a function of fish size (if known) and demographic data for the spawning population to provide a time series of relative annual fecundity for SBT.  相似文献

Southern bluefin tuna (SBT) were heavily depleted in the mid-1980s, and the fishing quota has been restricted since 1985. As a result of this restriction and protection of immature individuals, spawning stock biomass (SSB) recently has shown a slight increase. The Commission for the Conservation of Southern Bluefin Tuna (CCSBT) has the target of recovering SSB to the 1980 level by 2020. We investigated whether SBT populations will recover and reach the target level set by CCSBT. Our projection shows that the SSB of the SBT will temporarily decrease again after 1999. This temporary decrease of SSB does not always mean failure of the recovery plan, because the SSB trend is highly vulnerable to age-composition dynamics. The SBT is an example of this. The number of mature SBT was small during the 1980s because of overfishing. Thus, the number of eggs that were spawned by these mature SBT was small in these years, and when these small numbers of immature fish become mature, the SSB will decrease again. We call this effect the inverse baby-boom effect. The inverse baby-boom effect may be common for managed bioresources that have once been overexploited.
