Formation of Nodular Structures on Rice Seedlings by Rhizobia   总被引:5,自引:0,他引:5  
Nodular structures have been induced on rice roots by treatingthe roots of 2-d-old seedlings with a cell wall degrading enzymemixture consisting of 1.0% cellulase YC, 0.1% pectolyase Y23,and 8% mannitol, followed by inoculation with rhizobia in thepresence of polyethylene glycol. Rhizobia were located bothwithin the degenerating cytoplasm of cells and between the cellsof spherical and elongate nodular structures. Although nitrogenaseactivity in such structures was at the limit of sensitivityof the assay procedure, this first report of the induction ofnodular structures on rice by rhizobia opens up the possibilityof extending effective nodulation to non-legumes, includingcereals, by facilitating the entry of rhizobia through cellwall degradation coupled with polyethylene glycol treatment. This investigation was designed to examine whether or not deionizedwater could be acidified by roots of intact rice seedlings.Roots of intact rice seedlings caused significant acidificationof the deionized water in which they were immersed and thisacidification could be repeated after replacement of acidifiedwater with fresh deionized water. The addition of K+, Na+, andMg2+ to the deionized water significantly increased the rateand extent of acidification. However, no such increase was foundwhen Ca2+ was present in the water. The inhibition of acidificationby vanadate and its promotion by fusicoccin indicated that theacidification of water by roots of intact rice seedlings originatedfrom an ATP-driven proton pump located in the plasmalemma. Ferricyanide was effectively reduced by the roots of intactrice seedlings. This reduction was associated with the acidificationof the bathing solution. 8-Hydroxyquinoline and p-nitrophenyl-acetateinhibited both the reduction of ferricyanide and ferricyanide-inducedacidification. Vanadate, although it slightly inhibited thereduction of ferricyanide, did not inhibit the ferricyanide-stimulateddecrease in pH. It seems that the involvement of redox activityassociated with the plasmalemma in the acidification of deionizedwater cannot be excluded. (Received August 30, 1989; Accepted April 5, 1990)  相似文献

A survey of 51 strains of Bradyrhizobium japonicum was performedwith respect to the composition of extracellular polysaccharide(EPS), the production of rhizobitoxine and the hydrogenase phenotype.A good correlation was found among these three different characteristics.Thirty-six strains producing an EPS composed of glucose, mannose,galactose, 4-O-methyl galactose and galacturonic acid did notsynthesize rhizobitoxine, whereas 14 strains producing an EPScomposed of rhamnose and 4-O-methyl glucuronic acid were allfound to synthesize rhizobitoxine. Hydrogen-uptake positive(Hup+) strains were confined exclusively to the former groupof strains which produced an EPS composed of glucose, mannose,galactose, 4-O-methyl galactose and galacturonic acid. Theseresults suggest that the phenotype with respect to rhizobitoxineproduction and hydrogen uptake is involved in the phylogenyof Bradyrhizobium japonicum as well as in the productivity ofnodulated soybeans. Several cultivars of Solanum tuberosum L., the potato, weregrown on tissue culture media and their roots inoculated withstrains of rhizobia known to infect legumes at root junctionsor between epidermal cells. Infection incidence and severityshowed considerable cultivar/bacterial strain interaction. Bacteriaspread through intercellular spaces and invaded cells in a non-structuredway: some infections penetrated to the root xylem. There wasno evidence that potato root cells produced nod-inducing factorsand nitrogenase activity was not detected. In some host/rhizobialcombinations outgrowths were formed on roots. These varied fromloose infected callus tissue at the junctions of lateral roots,to modified lateral roots of limited growth which showed somecolonization by rhizobia. Previous analysis of a transgenic tobacco line (BN1) that over-expressedrice phytochrome A (PhyA) indicated that the introduced PhyAwas spectrally and biologically active [Kay et al. (1989) PlantCell 1: 775, Nagatani et al. (1991) Proc. Natl. Acad. Sci. USA88: 5207]. In the present study, we have further investigatedresponses of the BN1 plants to light. Fluence rate dependenceanalysis of the inhibition of hypocotyl elongation indicatedthat the response is biphasic. The amplitude of the low fluencerate component increased by 2 to 3 fold in the BN1 plants comparedto the wild type. In contrast, the presence of rice PhyA didnot alter the level of chlorophyll in the BN1 seedlings grownunder the same light conditions. Ultrastructure studies showedthat chloroplasts in the BN1 plants were not significantly differentfrom those in the wild type plants, except that chloroplastsin the guard cells of the BN1 plants appeared to be more developedthan those of the wild type plants. Fructans are water-soluble fructose oligomers and polymers thatare based on sucrose, and have been implicated in protectingplants against water stress. Rice (Oryza sativa L.) is highlysensitive to chilling temperatures, and is not able to synthesizefructans. Two wheat fructan-synthesizing enzymes, sucrose:sucrose1-fructosyltransferase, encoded by wft2, or sucrose:fructan6-fructosyltransferase, encoded by wft1, were introduced intorice plants, and rice transformants that accumulate fructanswere successfully obtained. The mature leaf blades of transgenicrice lines with wft2 or wft1 accumulated 16.2 mg g–1 FWof oligo- and polysaccharides mainly composed of inulin oligomersof more than DP7, and 3.7 mg g–1 FW of oligo- and polysaccharides,mainly composed of phlein oligomers of more than DP15, respectively.The transgenic rice seedlings with wft2 accumulated significantlyhigher concentrations of oligo- and polysaccharides than non-transgenicrice seedlings, and exhibited enhanced chilling tolerance. Theoligo- and polysaccharide concentrations of seedlings expressingwft1 were obviously lower than those of lines expressing wft2,and no correlation between oligo- and polysaccharide concentrationsand chilling tolerance was detected in wft1-expressing ricelines. The results suggest that transgenic rice lines expressingwheat-derived fructosyltransferase genes accumulated large amountsof fructans in mature leaf blades and exhibited enhanced chillingtolerance at the seedling stage. This is the first report owingthat fructan accumulation enhanced tolerance to non-freezinglow temperatures. Cyclic adenosine 3',5'-monophosphate (cAMP) content of variouscultured rhizobia strains and tissues of legumes and non-leguminousplants was measured by enzyme immunoassays. Most rhizobia, culturedfor 44 to 165 h, contained cAMP ranging from 0.6 to 5 pmol mg-1proteinexcept forAzorhizobium caulinodansORS571. The culture mediaalso contained varying amounts of cAMP depending on the strainof rhizobia.Azorhizobiumcells and their media contained no detectablecAMP. Nodules from most legumes and non-legumes had cAMP contentsranging from 2–70 pmol g-1f.wt. However, nodules fromSesbaniarostrata,Crotalaria spectabilisandParasponia andersoniishowedundetectable cAMP levels, and those fromGlycine maxandVignaangularisoccasionally showed levels below the detection limit.The leaves of non-legumes mostly had cAMP levels below detectionlimit (approx. 1.0 pmol g-1 f.wt), while the leaves ofa few legumes occasionally had detectable cAMP. Endogenous levels of gibberellins in shoots and ears of twodwarf rice (Oryza sativa L.) cultivars, Tan-ginbozu (dx mutant)and Waito-C (dy mutant), were analyzed and compared with thoseof normal rice cultivar, Nihonbare. The endogenous levels of13-hydroxylated gibberellins in Tan-ginbozu were much lowerthan those in Nihonbare. In Waito-C, the levels of GA19 andGA20 in the shoots were higher but that of GA1 was lower thanthe levels of these gibberellins in Nihonbare. These resultssupport the hypothesis that the dy gene controls the 3ß-hydroxylationof GA20 to GA1 while the dx gene controls a much earlier stepin the gibberellin biosynthesis. Our results indicate that GA1is the active gibberellin that regulates the vegetative growthof rice. The endogenous levels of GA4 in the ears of the twodwarf cultivars of rice were higher than the level of GA4 inthe ears of the normal cultivar, Nihonbare suggesting that thebiosynthesis of gibberellin is not blocked in the anthers ofthe dwarf rice. A nitrate reductase inactivating factor was found in extractsof leaf blades, leaf sheaths, and roots of rice seedlings. Thefactor was nondialyzable, precipitable with (NH4)2SO4, and heatlabile. The factor from rice roots inactivated NADH nitratereductase, FMNH2 nitrate reductase, and NADH cytochrome c reductasefrom rice shoots, but had no effect on the activities of NADHdiaphorase and nitrite reductase. The factors from rice shoots,rice roots, and maize roots inactivated NADH nitrate reductaseprepared from cultured rice cells. The factor from culturedrice cells also inactivated rice shoot NADH nitrate reductase. The activity of the inactivating factor showed a diurnal changein shoots of rice seedlings grown with NO3– medium, althoughthe fluctuation was not large compared to that of NADH nitratereductase activity. When the seedlings were placed in darkness,the activity of the factor did not change during 20 hr withNO3– medium. However, the activity of the factor fluctuatedwith NO3– -free medium in light; its activity startedto increase at the 8th hour after transfer. The curling of root hairs and the deformation response wereobserved when white clover was infected with homologous strainsof Rhizobium leguminosarum biovar trifolii 4S and 0403. In thecase of Rhizobium meliloti NZ and Rhizobium leguminosarum biovarviciae 128C53, however, curling was only induced when thesebacteria were pretreated with flavonoids: luteolin in the caseof R. meliloti and naringenin for R.I. viciae. The same resultswere obtained with oat, a monocotyledonous non-leguminous plant.The two flavonoids mentioned are secreted from the host plantsand induce the expression of genes for root hair curling (Hac)on Sym plasmid in homologous rhizobia, therefore, the curlingresponse in both white clover and oat appears to be correlatedwith the activation of the Hac genes. These results suggestthat a factor(s) that activates the Hac genes, such as 7,4'-dihydroxyflavonewhich is known as the factor required by R. I. trifolii, issecreted from the oat roots. The IAA biosynthetic pathway from tryptophan to IAA via IAM(IAM pathway) was investigated in Rhizobium spp. (fast-growingrhizobium). Southern hybridization with the bam gene, a structuralgene for IAM hydrolase (the enzyme that converts IAM to IAA)cloned from Bradyrhizobium japonicum J1063, indicated that homologoussequences exist among wild-type Rhizobium spp. However the IAMpathway has not been detected biochemically in free-living bacteria.When 5-methyltryptophan-resistant strains were screened forRhizobium leguminosarum biovar viciae K5 which has DNA sequenceswith high homology to the bam gene, spontaneous mutants showingIAM hydrolase activity were isolated. The results suggest thepossibility that the activity of IAM hydrolase is suppressedin free-living state in Rhizobium leguminosarum biovar viciaeK5. In addition we detected the peak at the same tR of IAM byHPLC analysis using two columns when a large amount of L-tryptophanwas added to the suspension of 5-methyltryptophan-resistantvariants. The xyloglucan present in the 24% KOH extract of the cell wallsof suspension-cultured rice cells was characterized by fragmentationanalysis with Trichoderma viride cellulase and Aspergillus oryzaeß-D-glucosidase. The xyloglucan is composed mainlyof the following oligosaccharide units: Results showed that the xyloglucan of suspension-cultured ricecells is more extensively branched than is that of rice seedlings.Another structural characteristic of the former xyloglucan isthe presence of D-galactosyl-D-xylosyl side chains that arenot found in the latter. The microscopic events leading to nodulation in normally nodulatingsoybean [Glycine max (L.) Merr.] genotypes, and the effectsof Bradyrhizobium strain and inoculum dose on nodulation, wereexamined in the NN5 non-nodulating mutant derived from cv. Williams.The NN5 mutant possesses the recessive genes rj5 and ,rj6. BradyrhizoblumJaponicum strain USDA 110 cells attached normally to the rootsurface of NN5, many in a polar manner as in its wild-type parent,but failed to induce root hair curling and sub-epidermal celldivision in the root. Co-culturing NN5 and Williams did notmodify nodulation of either genotype. Hydroponically-grown NN5seedlings did not nodulate at a high inoculum dose (1 x 1010cells seedling–1) of any B. japonicum strain tested (USDA110, USDA 26, USDA 136, and the tryptophan metabolic variantsB-14075 and ta 11 Nod+). A higher inoculum dose of 3 x 10 USDA136 cells seedling also failed to induce nodulation in NN5 andnod139 (a non-nodulating mutant of cv. Bragg). The lack of nodulationof NN5 at any inoculum dose is contrary to previous observationsof sparse nodulation of other non-nodulating mutants at highinoculum dose. The attachment of Rhizobiumjaponicum 61A89 to the roots of wheat and rice seedlings is an equilibrium process that follows a Langmuir adsorption isotherm. This model predicts a maximum of 8 × 109 viable bound bacteria per g of root. Different strains of bound rhizobia have both characteristic appearances and surface densities on the root surface. The inoculation of soybean (Glycine max L.) roots with Bradyrhizobium japonicum produces a regulatory response that inhibits nodulation in the younger regions of the roots. By exposing the soybean roots to live homologous bacteria for only a short period of time, the question of whether or not early interactions of rhizobia with root cells, prior to infection, elicit this regulatory response has been explored. B. japonicum cells mixed with infective bacteriophages were applied to the roots and then 6 or 24 hours later roots were again inoculated with phage-resistant rhizobia. Mixing of the rhizobia and bacteriophages caused bacterial lysis in 6 to 8 hours and allowed the bacteria to act as live symbionts on the root for only a few hours. However, the interaction of live homologous bacteria with the soybean roots for a few hours did not cause inhibition of nodulation in the younger regions of the roots. Results of these experiments indicate that the self-regulatory response in soybean is not rapidly produced by the early, pre-infection, interactions between rhizobia and the root cells.  相似文献

Exposing dark-grown seedlings for 3 d to oxygen deficiency (0or 5 kPa) or to additions of carbon dioxide (10 kPa) or ethylene(0·1 Pa) slowed shoot extension in Echinochloa oryzoides,while in rice it was promoted by these treatments, except that5 kPa oxygen was without effect. In E. oryzoides this was dueto reduced growth of the mesocotyl, and in rice to enhancedgrowth of the coleoptile. These responses to carbon dioxideand oxygen deficiency were not consequences of increased ethyleneproduction, since this remained unchanged by carbon dioxideand depressed by oxygen shortage in both species. Furthermore,exogenous ethylene and the ethylene action inhibitor 2,5-norbornadieneeach failed to influence extension in anoxic seedlings, indicatingno regulatory role for ethylene in the absence of oxygen. However,concentrations of the ethylene precursor 1 -aminocyclopropane-1-carboxylic acid (ACC) were increased by carbon dioxide and0 kPa or 5 kPa oxygen, although after 72 h without oxygen totalACC production (i.e. changes in ethylene + ACC + MACC) was suppressedin both species. There was little effect on bound ACC [putativemalonyl-ACC (MACC)] formation. Transferring anaerobic (0 kPa)seedlings to oxygenated conditions (21 kPa) resulted in abnormallyfast rates of ethylene formation, possibly due to the accumulationof ACC under anoxia. This post-anoxic ethylene may have contributedto the faster extension by rice coleoptiles and slower extensionby mesocotyls of E. oryzoides compared with those of seedlingsmaintained continuously in air. Chamaecrista mimosoides is an annual herb legume widely distributed in tropical and subtropical Asia and Africa. It may have primitive and independently-evolved root nodule types but its rhizobia have not been systematically studied. Therefore, in order to learn the diversity and species affinity of its rhizobia, root nodules were sampled from C. mimosoides plants growing in seven geographical sites along the coast line of Shandong Peninsula, China. A total of 422 rhizobial isolates were obtained from nodules, and they were classified into 28 recA haplotypes. By using multilocus sequence analysis of the concatenated housekeeping genes dnaK, glnII, gyrB, recA and rpoB, the representative strains for these haplotypes were designated as eight defined and five candidate novel genospecies in the genus Bradyrhizobium. Bradyrhizobium elkanii and Bradyrhizobium ferriligni were predominant and universally distributed. The symbiotic genes nodC and nifH of the representative strains showed very similar topology in their phylogenetic trees indicating their co-evolution history. All the representative strains formed effective root nodules in nodulation tests. The correlation between genospecies and soil characteristics analyzed by CANOCO software indicated that available potassium (AK), organic carbon (OC) and available nitrogen (AN) in the soil samples were the main factors affecting the distribution of the symbionts involved in this current study. The study is the first systematic survey of Chamaecrista mimosoides-nodulating rhizobia, and it showed that Chamaecrista spp. were nodulated by bradyrhizobia in natural environments. In addition, the host spectrum of the corresponding rhizobial species was extended, and the study provided novel information on the biodiversity and biogeography of rhizobia.  相似文献

Salinity and high temperature stresses adversely affect growthand development of rice plants. To investigate the responseof rice cells to these stresses, we have analysed short-termstress-induced subcellular alterations in undifferentiated leafcells of rice seedlings by transmission electron microscopy.Perturbations noted particularly with respect to plasma membrane,mitochondrial membranes, endoplasmic reticulum, polyribosomesand dictyosomes are highlighted. The subcellular changes evokedby both stresses after 4 h were lysis of the cytoplasm, accumulationof electron-dense granules in the cytoplasm, distension in theER membranes, enhanced association of ribosomes with the endoplasmicreticulum, reduction in the number of mitochondrial cristae,as well as disorganization of cell wall fibrillar material.Certain changes were found to be unique to either the salinityor high temperature stress. Rice seedling germination and developmental behaviour were examinedin the presence of six different barbituric acid derivatives.The relationship between liquid/aqueous partition coefficientsof barbiturates and their interference with, but not completeinhibition of, normal mitochondria1 oxidative metabolism observedin animal mitochondria in vitro also appear to be factors influencingthe effects of these compounds on the germination and developmentalbehaviour of the rice seedlings described in this investigation.Evidence is presented here which points to normal oxidativemetabolism as being a necessary requisite for normal light responseand normal morphogenetic development in germinating rice seedlings.  相似文献
