Hydroquinoline acyclonucleosides 2, 4, 6a,b, 8a,b, 9a,b, and their corresponding N-alkyl derivatives (10–12) were obtained by the reaction of 1a,b with acetoxybutylbromide, (2-acetoxyethoxy)methyl bromide, 3-chloropropanol, 1,3-dichloro-2-propanol, epichlorohydrin, propargyl/allyl bromides in the presence of K2CO3 in dry dimethylformamide (DMF). In a similar manner, reaction of 1a,b with glycosyl/galactosyl and lactosyl bromide afforded the corresponding N-nucloside derivatives 13a,b, 15a,b, and 17, respectively. Deacetylation of the N-nucleosides derivatives in the presence of Et3N/MeOH and few drops of water gave the deprotected derivatives 3, 5, 7a,b, 14a,b, 16a,b, and 18 in good yields, respectively. All the newly synthesized compounds are elucidated by infrared, 1H, 13C NMR and elemental analyses. Some of these compounds were screened for antimicrobial activities.  相似文献

Glycosylation of 2-pyridonesulfonamide 1a,b with glycosyl/galactosyl bromide gave the corresponding glycosides 2a,b, 3a,b, 6a,b, and 7a,b, respectively. Deacetylation of the resulting glycosides gave the corresponding glycosides 4a,b, 5a,b, 8a,b, and 9a,b, respectively, in good yields. Furthermore, reaction of 2-pyridonesulfonamide 1b with lactosyl bromide gave a mixture the corresponding N, O-lactosides 10 and 11, which were deacetylated to give the corresponding glycosides 12 and 13, respectively. The structures of the new synthesized compounds were characterized by using IR, 1H, 13C NMR spectra, and microanalysis. Selected members of these compounds were screened for antimicrobial activity.  相似文献

The solvent-free 1,3-dipolar cycloaddition reaction of dimethylacetylene dicarboxylate (1) with 2-chlorophenyl azide (2) afforded 1,2,3-triazole diester 3 that upon hydrazinolysis, furnished the corresponding bis-acid hydrazide 4. The treatment of compound 4 with carbon disulfide in a refluxing potassium hydroxide solution furnished the desired bis-1,3,4-oxadiazole-2-thione 5 tethered to a 1,2,3-triazole moiety. The respective SOx-glycosides 9–11 were obtained by glycosylation of bis-oxadiazole 5 with 2,3,4,6-tetra-O-acetyl-α-d-glucopyranosyl bromide (6), 2,3,4,6-tetra-O-acetyl-α-d-galactopyranosyl bromide (7), and 2-acetamido-3,4,6-tri-O-acetyl-2-deoxy-α-d-glucopyranosyl chloride (8) in dry acetone in the presence of Et3N, which acted as a base. However, alkylation of 5 with halogeno-alkanol 12 or 13, chloroglycerol 14, bromoethers 20 or 21, and epichlohydrin 22 in the presence of K2CO3 in DMF yielded the corresponding acyclonucleoside analogs 16–18 and 23–25. The isopropylidenes 19 and acetyl derivatives 26–28 of the products were also prepared. The newly synthesized compounds were characterized by 1H NMR, 13C NMR, 2D NMR, and mass spectra. The compounds were screened for their antibacterial and antifungal activities. A number of the tested compounds exhibited significant antimicrobial activity compared to the reference drugs.  相似文献

The synthesis of a novel series of 4-arylhydrazono-5-methyl-1,2-dihydropyrazol-3-ones 4a–h, and their N 2-alkyl and acyclo, glucopyranosyl, and ribofuranosyl derivatives is described. K2CO3 catalyzed alkylation of 4a–h with allyl bromide, propargyl bromide, 4-bromobutyl acetate, 2-acetoxyethoxymethyl bromide, and 2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl bromide proceeded selectively at the N 2-position of the pyrazolinone ring. Glycosylation of 4a with 1,2,3,5-tetra-O-acetyl-β-D-ribofuranose under Vorbruggen glycosylation conditions gave the corresponding N 2-4-arylhydrazonopyrazolone ribofuranoside 9a in good yield. Conventional deprotection of the acetyl protected nucleosides furnished the corresponding 4-arylhydrazonopyrazolone nucleosides in good yields. Selected numbers of the newly synthesized compounds were screened for antimicrobial activity. Compounds 4b, 12a, and 14d showed moderate activities against Aspergillus flavus, Penicillium sp., and Escherichia coli.  相似文献

Reaction of ethyl 4-thioxo-3,4-dihydropyrimidine-5-carboxylate derivatives 1a,b and ethyl 4-oxo-3,4-dihydropyrimidine-5-carboxylate 1c with 2,3,4,6-tetra-O-acetyl-α-D-glucopyranosyl bromide in KOH or TEA afforded ethyl 2-aryl-4-(2′,3′,4′,6′-tetra-O-acetyl-β-D-glucopyranosylthio or/ oxy)-6-methylpyrimidine-5-carboxylate 6a-c. The glucosides 6a and 6b were obtained by the reaction of 1a and 1b with peracetylated glucose3 under MW irradiation. Mercuration of 1a followed by reaction with acetobromoglucose gave the same product 6a. The reaction of 1a-c with peracetylated ribose 4 under MW irradiation gave ethyl 2-aryl-4-(2′,3′,5′-tri-O-acetyl-β-D-ribofuranosylthio)-6-methylpyrimidine-5-carboxylate 8a–c. The deprotection of 6a–c and 8a–c in the presence of methanol and TEA/H2O afforded the deprotected products 7a–c and 9a–c. The structure were confirmed by using 1H and 13CNMR spectra. Selected members of these compounds were screened for antimicrobial activity.  相似文献

Abstract

A convenient synthesis of 2′-deoxy-2-fluoro-adenosine (1) is described. Deaminative fluorination of 2-aminoadenosine (2) followed by silylation of the 3′, 5′-hydroxyl groups gave the corresponding 2-fluoroadenosine derivative 4 in good yield. Thiocarbonylation of 4 to thiocarbonylimidazolyl derivative 5a followed by treatment with an excess of tris(trimethylsilyl)silane (TTMSS) and tert-butyl peroxide in toluene at 80 [ddot]C was found to affect an efficient deoxygenation to the corresponding 2′-deoxy derivative 6. Desilylation of 6 by Et4NF in CH3CN afforded 1 in high yield.  相似文献

This study reports a novel and efficient method for the synthesis of the first reported novel class of triazole thioglycosides. These series of compounds were designed through the reaction of potassium cyanocarbonimidodithioate 2 with hydrazine derivatives 3a-d in EtOH at room temperature to give the corresponding potassium 5-amino-1H-1,2,4-triazole-3-thiolates 4a-d. The latter compounds were treated with tetra-O-acetyl-α-D-glucopyranosyl bromide 6a and tetra-O-acetyl-α-D-galactopyranosyl bromide 6b in DMF at room temperature to give in high yields the corresponding triazole thioglycosides 7a-h. Treatment of triazole salts 4a–d with hydrochloric acid afforded the corresponding 3-mercaptotriazoles 5a-d. Compounds 5a-d were then reacted with bromoperacetylated sugars 6a,b in sodium hydride-DMF at ambient temperature to afford the thioglycosyl compounds 7a-h. Ammonolysis of the triazole thioglycosides 7a-h afforded the corresponding free thioglycosides 8a-h. The scope and limitation of the method is demonstrated. The structure of the reaction products was confirmed on the basis of their elemental analysis and spectral data (IR, 1H NMR, MS and 13C NMR).  相似文献

Metathesis reaction between equimolar amount of [Et4N][GaCl4] and Na2edt in methanol resulted in the formation of the dichloro complex [Et4N][Ga(edt)Cl2] (1), whereas reaction of [Et4N][GaCl4] with two equivalents of Na2edt in methanol gave the complex [Et4N][Ga(edt)2] (2) which can act as a metalloligand. Treatment of 2 with M(PPh3)2NO3 in DMF/CH2Cl2 afforded the heterobimetallic complexes [Ga(edt)2M-(PPh3)2] (M = Cu 3, Ag 4) in moderate yields. The structures of 1-4 were determined by single-crystal X-ray diffraction analyses. Both [Ga(edt)Cl2] and [Ga(edt)2] anions have a distorted tetrahedral geometry. The former consists of one five-membered ring formed by chelating dithiolate and two terminal chloride atoms while the latter consists of two five-membered rings formed by two the chelating dithiolates. Complexes 3 and 4 consist of metalloligand [Ga(edt)2] anion chelated to [M(PPh3)2]+via the sulfur atoms. Both tetrahedrally coordinated Ga and Cu(Ag) atoms are bridged by two sulfur atoms, forming a planar “GaS2M” (M = Cu, Ag) core. Thermogravimetry analysis revealed that heterobimetallic complexes 3 and 4 decomposed to give the corresponding ternary metal sulfide materials.  相似文献

Reaction of pyridin-2(1H)-one 1 with 4-bromobutylacetate (2), (2-acetoxyethoxy)methyl bromide (3) gave the corresponding nicotinonitrile O-acyclonucleosides, 4 and 5, respectively. Deacetylation of 4 and 5 gave the corresponding deprotected acyclonucleosides 6 and 7, respectively. Treatment of pyridin-2(1H)-one 1 with 1,3-dichloropropan-2-ol (8), epichlorohydrin (10) and allyl bromide (12) gave the corresponding nicotinonitrile O-acyclonucleosides 9, 11, and 13, respectively. Furthermore, reaction of pyridin-2(1H)-one 1 with the propargyl bromide (14) gave the corresponding 2-O-propargyl derivative 15, which was reacted via [3+2] cycloaddition with 4-azidobutyl acetate (16) and [(2-acetoxyethoxy)methyl]azide (17) to give the corresponding 1,2,3-triazole derivatives 18 and 19, respectively. The structures of the new synthesized compounds were characterized by using IR, 1H, 13C NMR spectra, and microanalysis. Selected members of these compounds were screened for antibacterial activity.  相似文献

A series of 2-(chloro-substituted-1H-benzoimidazol-2-yl)-6-(1-aryliminoethyl)pyridines (L1-L6) was synthesized and fully characterized. They reacted with NiCl2 or (DME)NiBr2 to form the corresponding N^N^N tridentate nickel dichlorides (C1-C6) or dibromides (C7-C12). All nickel complexes were characterized by elemental analysis and infrared spectroscopy, while the X-ray diffraction study reveals the distorted octahedral geometry of representative nickel dichloride complex C3 and nickel dibromide complex C7. All nickel complexes, activated with either Et3Al2Cl3 or Et2AlCl, showed good activities in ethylene oligomerization, and notably remarkably high selectivity of α-olefins was achieved in the presence of Et3Al2Cl3. The chloro-substituted ligands enhanced the activities of complexes in ethylene polymerization.  相似文献

Abstract

The addition reaction of either 3-bromo-5-lithiopyridine (2a) or 3-cyano-5-lithiopyridine (2b) to 2,3:4,5-di-O-isopropylidene-aldehydo-D-arabinose (1) or 2,4:3,5-di-O-benzylidene-aldehydo-D-lyxose (8) gave respectively a D-gluco/D-manno mixture of 3-bromo- and 3-cyano-5-(2,3:4,5-di-O-isopropylidene-pentitol-1-yl)pyridine (3a,b) or a D-galacto/D-talo mixture of respectively 3-bromo- and 3-cyano-5-(2,4:3,5-di-O-benzylidene-pentitol-1-yl)pyridine (9a,b). Mesylation of C-1′ followed by reaction with CF3COOH/H2O resulted in the formation of the corresponding D-arabino- or D-lyxofuranosyl pyridine C-nucleosides. The cyano group of (5b) and (11b) was converted into a carbamoyl group using Amberlite IRA 400 (OH?). 3-Cyano-5-D-arabinofuianosylpyridine (5b) was converted into 3-thiocarbamoyl-5-D-arabinofuranosyl-pyridine (7) using H2S and triethylamine.

None of the test compounds showed a marked cytostatic or antiviral activity in vitro.  相似文献

The ditopic tris(2-mercaptoimidazol-1-yl)borate ligand K2[(mtEt)3B-B(mtEt)3] cannot be prepared from B2(NMe2)4/4 HmtEt/2 KmtEt, because the stable intramolecular diadduct (mtEt)B(μ-mtEt)2B(mtEt) is generated instead (HmtEt = 2-mercapto-1-ethylimidazole). Introduction of a meta- or para-phenylene spacer between the two boron atoms precludes the 2-mercaptoimidazol-1-yl groups from adopting a bridging position so that the potassium salts K2[(mtEt)3B-(m-C6H4)-B(mtEt)3] and K2[(mtEt)3B-(p-C6H4)-B(mtEt)3] become readily accessible. These ligands react with [(p-cym)RuCl2]2 to give the dinuclear RuII complexes [(p-cym)Ru(mtEt)3B-(m-C6H4)-B(mtEt)3Ru(p-cym)]Cl2 and [(p-cym)Ru(mtEt)3B-(p-C6H4)-B(mtEt)3Ru(p-cym)]Cl2 (p-cym = p-cymene). After the exchange of the Cl counterions for [PF6], both complexes have been crystallized and structurally characterized by X-ray diffraction.  相似文献

The synthesis and crystal and molecular structure of the first zinc(II) mixed ligand chelate containing a dithiolene ligand (maleonitriledithiolate) and N,N- diethyldithiocarbamate are reported. The compound Ph4As[Zn(mnt)(Et2dtc)] crystallizes monoclinic, space group P21/c with four molecules in the unit cell; a=17.834(3), b=12.056(2), c=16.171(4) Å, β=93.73(2)°.The coordination geometry of the ZnS4 unit is nearly tetrahedral, with a dihedral angle of 87.6° between the chelate rings. The structure is compared with those of both the patent compounds (Ph4- As)2[Zn(mnt)2] and Zn2(Et2dtc)4. Ph4As[Zn(mnt)(Et2dtc)] could be used as the host lattice in single- crystal ESR investigations of the planar Cu mixed ligand complex. The rhombic spin-Hamiltonian parameters g and ACu are indicative of a low symmetry of the incorporated [Cu(mnt)(Et2dtc)] complex anions. In order to substantiate the experimental findings about the actual structure of the copper molecules, the principal values of g and ACu were recalculated by means of Extended Hückel MO calculations. However, the calculations performed for the dihedral angles between the ligand planes varying between 0° and 90° suggest that the rotation of the ligands is not larger than 10°  相似文献

Abstract

5-O-tert-Butyldimethylsilyl-1,2-O-isopropylidene-3(R)-(nicotinamid-2-ylmethyl)-α-D-ribofuranose (11a) and ?3(R)-(nicotinamid-6-ylmethyl)-α-D-ribofuranose (11b) were prepared by condensation of 5-O-tert-butyldimethylsilyl-1,2-O-isopropylidene-α-D-erythro-3-pentulofuranose (10) with lithiated (LDA) 2-methylnicotinamide and 6-methylnicotinamide, respectively, and then deprotected to give 1,2-O-isopropylidene-3-(R)-(nicotinamid-2-ylmethyl)-α-D-ribofuranose(12a) and 1,2-O-isopropylidene-3(R)-(nicotinamid-6-ylmethyl)-α-D-ribofuranose (12b). Benzoylation as well as phosphorylation of compounds 12 afforded the corresponding 5-O-benzoate (13b) and 5-O-monophosphates (14a and 14b). Treatment of 13b with CF3COOH/H2O caused 1,2-de-O-isopropylidenation with simultaneous cyclization to the corresponding methylene-bridged cyclic nucleoside - 3′,6-methylene-1-(5-O-benzoyl-β-D-ribofuranose)-3-carboxamidopyridinium trifluoro-acetate (8b) - restricted to the “anti” conformation. In a similar manner compounds 14a and 14b were converted into conformationally restricted 2,3′-methylene-1-(β-D-ribofuranose)-3-carboxamidopyridinium-5′-monophosphate (9a - “syn”) and 3′,6-methylene-1-(β-D-ribofuranose)-3-carboxamido -pyridinium-5′monophosphate (9b - “anti”) respectively. Coupling of derivatives 12a and 12b with the adenosine 5′-methylenediphosphonate (16) afforded the corresponding dinucleotides 17. Upon acidic 1,2-de-O-isopropylidenation of 17b, the conformationally restricted P1-[6,3′-methylene-1-(β-D-ribofuranos-5-yl)-3-carboxamidopyridinium]-P2-(adenosin-5′-yl)methylenediphosphonate 18b -“anti” was formed. Compound 18b was found to be unstable. Upon addition of water 18b was converted into the anomeric mixture of acyclic dinucleotides, i. e. P1-[3(R)-nicotinamid-6-ylmethyl-D-ribofuranos-5-yl]-P2-(adenosin-5′-yl)-methylenediphosphonate (19b). In a similar manner, treatment of 17a with CF3COOH/H2O and HPLC purification afforded the corresponding dinucleotide 19a.

Regioselectively fluorinated 1-(naphth-2-ylmethyl)imidazoles 1ah have been synthesized starting from the corresponding (naphth-2-yl)methanols (2). 2ad have been obtained by LiAlH4-promoted reduction of fluorinated 1-methyl-2-naphthaldehydes. The latter were easily prepared in fairly good overall yields by ceric ammonium nitrate (CAN)-promoted oxidative addition of the suitable 3-(fluoroaryl)-1-trimethylsilyloxy-1-butenes to ethyl vinyl ether in methanol followed by cyclization of the resulting acetals in strongly acidic medium in the presence of DDQ. 2eh were prepared by LiAlH4-promoted reduction of the corresponding fluorinated methyl 2-naphthoates. The latter were more profitably obtained by reacting the suitable benzyl bromide with the sodium salt of dimethyl 2-(2,2-dimethoxyethyl)malonate in DMF followed by demethoxycarbonylation and acid catalysed cyclization of the resulting acetals. Compared with the non-fluorinated parent compounds 1il, fluorinated 1-(naphth-2-yl)methylimidazoles 1ah turned out to be potent inhibitors of CYP17 and CYP19 enzymes. The most active inhibitor of CYP17 is 1c, whereas CYP19 is strongly inhibited by 1b, 1e, and 1g. Interestingly, 1g is a potent dual inhibitor also being very active towards CYP19.  相似文献

Abstract

The oxidation of methyl 5–0-benzyl-3-deoxy-3-fluoro-α-D-arabi-nofuranoside (1) with DMSO/Ac2o afforded a ~ 2:1 mixture of 2-keto derivatives with erythro and threo configuration resulting from isomerization at C3. Successive treatment of the above mixture with MeONH2, LiA1H4, and S-ethyl trifluoroacetate followed by silica gel chromatography afforded methyl 5–0-benzyl-2, 3-dideoxy-3-fluoro-2-(trifluoroacetamido)-α-D-ribofuranoside (6b) and its lyxo isomer 7b in a total yield of 25% and 5%, respectively. The arabino analogue 25 was prepared from 6b. Compounds 6b, 7b and 25 were converted to the corresponding 5–0-benzoyl derivatives 8a, 9 and 26. A series of 2′-amino-2′, 3′-dideoxy-3′-fluoro-β-D-ribo- and-α-D-lyxofuranosides of natural heterocyclic bases have been synthesized starting from 8a and 9. None of the test compounds had any antiviral activity. 3′-Fluoro-2′-amino-2′, 3′-dideoxycytidine (16) was the only compound showing inhibition of murine L1210 and human Molt/4F cell proliferation (50% effective concentration: 39–42μg/m1).  相似文献

Two hitherto unknown mixed-ligand tris chelated complexes containing 2-aminothiophenolate, [Et4N]2[MIV(NH-(C6H4)-S)(mnt)2] (M = Mo, 1a; W, 2a) and two mixed-ligand tris chelate complex containing N,N-diethyldithiocarbamate, [Et4N]2[MIV(Et2NS2)(mnt)2] (M = Mo, 1b; W, 2b) have been synthesized and characterized structurally. Although these complexes are supposed to be quite similar to the well-known symmetric tris chelate complexes of maleonitriledithiolate (mnt), [Et4N]2[MIV(mnt)3] (M = Mo, 1c; W, 2c), but display both trigonal prismatic and distorted trigonal prismatic geometry in their crystal structure indicating the possibility of an equilibrium between these two structural possibilities in solution. Unlike extreme stability of 1b, 2b, 1c and 2c, both 1a and 2a are highly unstable in solution. In contrast to one reversible reduction in case of 1b and 2b, 1a and 2a exhibited no possible reduction up to −1.2 V and two sequential oxidation steps which have been further investigated with EPR study. Differences in stability and electrochemical behavior of 1a, 1b, 2a and 2b have been correlated with theoretical calculations at DFT level in comparison with long known 1c and 2c.  相似文献

A comparative study for selective glucosylation of N-unsubstituted 4-hydroxyquinolin-2(1H)-ones into 4-(tetra-O-acetyl-β-d-glucopyranosyloxy)quinolin-2(1H)-ones is reported. Four glycosyl donors including tetra-O-acetyl-α-d-glucopyranosyl bromide, β-d-glucose pentaacetate, glucose tetraacetate and tetra-O-acetyl-α-d-glucopyranosyl trichloroacetimidate were tested, along with different promoters and reaction conditions. The best results were obtained with tetra-O-acetyl-α-d-glucopyranosyl bromide with Cs2CO3 in CH3CN. In some cases the 4-O-glucosylation of the quinolinone ring was accompanied by 2-O-glucosylation yielding the corresponding 2,4-bis(tetra-O-acetyl-β-d-glucopyranosyloxy)quinoline. Next, 4-(tetra-O-acetyl-β-d-glucopyranosyloxy)quinolin-2(1H)-ones were deacetylated into 4-(β-d-glucopyranosyloxy)quinolin-2(1H)-ones with Et3N in MeOH. In some instances the deacetylation was accompanied by the sugar-aglycone bond cleavage. Structure elucidation, complete assignment of proton and carbon resonances as well as assignment of anomeric configuration for all the products under investigation were performed by 1D and 2D NMR spectroscopy.  相似文献

The new aminoalkyl-substituted derivatives of known CK2 inhibitors 4,5,6,7-tetrabromo-1H-benzimidazole (TBBi) and 4,5,6,7-tetrabromo-1H-benzotriazole (TBBt) were synthesized, and their influence on the activity of recombinant human CK2 α, CK2 holoenzyme and PIM1 kinases was evaluated. All derivatives inhibited the activity of studied kinases and the most efficient were aminopropyl-derivatives 8b and 14b. These compounds also exerted inhibition of cancer cell lines – CCRF-CEM (acute lymphoblastoid leukemia), MCF-7 (human breast cancer), and PC-3 (prostate cancer) proliferation and their EC50 is comparable with the value for clinically studied CK2 inhibitor CX-4945. Preliminary structure activity relationship analysis indicated that the spacer length affected antitumor potency, and two to three methylene units were more favorable. The complex of CK2 α1-335/8b was crystallized, both under high-salt conditions and under low-salt conditions giving crystals which diffracted X-rays to about 2.4 Å resolution, what enabled the determination of the corresponding 3D-structures.  相似文献

Abstract

Reaction of 2′,3′,5′-O-silylated inosine derivative 1 with 2, 3-O-isopropylidene-5-O-tritylribosyl chloride (3) in a two-phase (CH2Cl2-aq. NaOH) system in the presence of Bu4NBr gave three products, i. e., 6-O-α-, 6-O-β-, and N 1-β-isomers of glycosides 4, 5a, and 5b. A similar PTC reaction of 1 with 2, 3, 5-tri-O-benzylribosyl bromide (9) gave four regio- and stereo-isomers involving the N1-β-glycoside 10. Reaction of 1 with 2, 3, 5-tri-O-benzoylribosyl bromide (11) afforded three products involving the desired N1-β-glycoside 12b, which could be deprotected to give N 1-ribosylinosine (15b) as a useful intermediate for the synthesis of cIDPR.
