In this study, we compared the chemical forms and subcellular distribution of Cd in high-Cd (X16) and low-Cd (N88) sweet potato cultivars through hydroponic experiments and examined the Cd distribution in their roots by histochemical staining. The results showed that inorganic and pectate/protein-integrated Cd predominated in the leaves, and Cd concentrations were significantly higher in X16 than in N88. However, in the roots, Cd was mostly integrated with pectate and protein, and Cd concentration was higher in N88 than in X16. It was mainly stored through vacuolar sequestration and cell wall binding. In the leaves and stems, Cd concentrations in all subcellular fractions were higher in X16 than in N88; the opposite was observed in the roots. In X16, Cd was mostly accumulated in the root stele, and its Cd translocation factor was higher than that of N88. Overall, the subcellular fractions of X16 roots retained less Cd than N88 roots, and more Cd entered the root stele of X16 and subsequently moved to the shoots. The higher amounts of inorganic, water-soluble, and pectate/protein-integrated Cd with high mobility in the shoots of X16 than in N88 might facilitate Cd remobilization to other tissues, but this needs to be further studied.

Contamination by heavy metals is one of the most serious environmental problems generated from human activities. Because phytoremediation utilizes plants to uptake contaminants, it could potentially be used to remediate metal-contaminated areas. A pot culture experiment with four levels of cadmium (Cd) (0, 20, 40, and 80 mg of Cd/kg dry soil) was conducted to investigate Cd accumulation and tolerance of roots, shoots, and leaves of Lagerstroemia indica and Lagerstroemia fauriei as well as their potential for phytoremediation. Experimental results indicated that Cd inhibited seedling growth only at the higher Cd exposure concentration (40 and 80 mg/kg). The tolerance index revealed that on average L. indica is more tolerant of Cd than L. fauriei. Moreover, plants in the experiment accumulated Cd differentially. In comparisons between L. indica and L. fauriei, the leaves of the former had higher concentrations of Cd, while the roots of latter had higher concentrations of Cd. Furthermore, the roots, shoots, and leaves had very high bioaccumulation factors that markedly exceeded 1.0 (exceptional only in shoots of 80 mg/kg for L. fauriei), indicating that the seedlings extracted Cd from the soil. The leaves' translocation factor of L. indica was greater than 1.0, being significantly higher than that of L. fauriei. Chlorophyll a, Chlorophyll b and total declined in both species significantly as Cd concentrations exceeded 40 mg/kg in the soil. In contrast, lipid peroxidation and proline content was found to increase with increasing Cd concentration. From the assessments of biomass production, Cd tolerance and uptake L. indica and L. fauriei could stand as excellent species for remediating Cd-contaminated soils.

The objective of this study was to assess the effect of different Cd and Si concentrations on the maize plants. The following Cd and/or Si treatments were used: 5 Cd; 10 Cd; 100 Cd; 5 Cd + 0.08 Si; 10 Cd + 0.08 Si; 100 Cd + 5 Si treatments (Cd concentration in μM, Si concentration in mM). The plant growth, photosynthetic pigments content, antioxidant enzymes activities (POX, SOD, CAT), Cd and Si accumulation, translocation and cell wall deposition of the maize plants was observed. Changes in the endodermal cell walls development and late metaxylem elements lignification due to Cd and/or Si treatment were also evaluated. The negative effect of Cd (5 and 10 μM) on the growth parameters was alleviated by Si at 0.08 mM. The positive effect of Si was not observed at higher Cd and Si concentrations. This indicates that the alleviating effect of Si on Cd toxicity depends on the Cd and Si concentrations. Plants responded to Cd toxicity by an increase of antioxidant enzyme activity. Silicon addition in Cd + Si treatment stimulated an increase in the activity of antioxidant enzymes in comparison with the Cd treatment. Chlorophyll and carotenoid content in the Cd treated plants was not significantly affected by Si. The young maize plants retained much more Cd in their roots as they translocated into the shoots. 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments correlated with an increase in Cd concentration in the roots and shoots, and in the cell walls. Silicon caused a slight decrease of the Cd translocation into the shoots in 5 Cd + 0.08 Si and 10 Cd + 0.08 Si treatments. Negative correlation between the root Cd cell wall deposition and Cd translocation was observed. Cadmium and/or Si altered root anatomy. Cadmium enhanced suberin lamellae development and late metaxylem lignification; silicon in Cd + Si treatments accelerated suberin lamellae deposition and enhanced the tertiary endodermal cell walls formation in comparison with Cd treatments. Negative correlation between the endodermal cell walls development and Cd translocation was observed.

We evaluated the potential of Salix viminalis (5027) and Populus × generosa for the phytoextraction of heavy metals (HM) inoculated or not with an arbuscular mycorrhizal (AM) fungus Glomus intraradices during a second year of growth in a randomized complete block field trial on a slightly contaminated site. Both plant clones produced high aboveground biomass yields, however P. × generosa produced significantly more biomass than S. viminalis. The two plant clones accumulated high concentrations of Cd and Zn in their shoots, while Cu and Pb were stored in their roots. In general, S. viminalis accumulated higher concentrations of HM. The inoculation of G. intraradices in the previous year did not influence plant clones’ biomass yields during the second growing season. However, Cu and Cd translocation to shoots was limited, and Cu was preferentially concentrated in roots of inoculated plants, compared to non-inoculated plants, which were also colonized by native AM fungi taxa. Efficiency of S. viminalis and P. × generosa for Cd and Zn rehabilitation in slightly contaminated soil has been demonstrated, but mycorrhizal inoculation did not significantly increase HM extraction.

The potential suitability of Eucalyptus camaldulensis for Cd phytoextraction was tested in a hydroponic study. Saplings were exposed to 4.5 and 89 μM Cd for one month, with and without EDTA and s,s-EDDS at 0.1, 1, and 5 mM. The saplings’ growth was not affected at the 4.5 μM Cd concentration, yet it decreased 3-fold at 89 μM, and almost all the Cd taken up was immobilized in the roots, reaching 360 and 5300 mg Cd kg?1, respectively (approximately 75% of which was non-washable in acid). The respective Cd root-to-shoot translocation factors were 0.14 and ≈5*10?4. At 0.1 mM concentration, EDTA and EDDS had no effect or even a positive effect on the saplings growth. This was reversed at 1 mM, and the chelants became lethal at the 5 mM concentration. At 89 μM Cd in the growth medium, 0.1 mM EDTA increased Cd translocation into the shoots by almost 10-fold, however it strongly reduced Cd content inside the roots. This hydroponic study indicates the feasibility of E. camaldulensis use for cleanup Cd-contaminated soils at environmental concentrations, both for site stabilization (phytostabilization) and gradual remediation (phytoextraction). EDTA was shown to be much more efficient in enhancing Cd translocation than s,s-EDDS.

In order to study the effect of mycorrhizal fungi (inoculated and non-inoculated) and heavy metals stress [0, Pb (150 and 300 mg/kg) and Cd (40 and 80 mg/kg)] on pot marigold (Calendula officinalis L.), a factorial experiment was conducted based on a randomized complete block design with 4 replications in Research Greenhouse of Department of Horticultural Sciences, University of Tehran, Iran, during 2012–2013. Plant height, herbal and flower fresh and dry weight, root fresh and dry weight and root volume, colonization percentage, total petal extract, total petal flavonoids, root and shoot P and K uptakes, and Pb and Cd accumulations in root and shoot were measured. Results indicated that with increasing soil Pb and Cd concentration, growth and yield of pot marigold was reduced significantly; Cd had greater negative impacts than Pb. However, mycorrhizal fungi alleviated these impacts by improving plant growth and yield. Pot marigold concentrated high amounts of Pb and especially Cd in its roots and shoots; mycorrhizal plants had a greater accumulation of these metals, so that those under 80 mg/kg Cd soil?1 accumulated 833.3 and 1585.8 mg Cd in their shoots and roots, respectively. In conclusion, mycorrhizal fungi can improve not only growth and yield of pot marigold in heavy metal stressed condition, but also phytoremediation performance by increasing heavy metals accumulation in the plant organs.

Lithium (Li) is a trace element that is essential in the human diet due to its importance for health and proper functioning of an organism. However, the biological activity of this metal in crop plants, which are the primary dietary sources of Li, is still poorly understood. The aim of the presented study was to comparatively analyse two Li chemical forms on the growth, as well as the l-ascorbic acid content, the Li accumulation and translocation in butterhead lettuce (Lactuca sativa L. var. capitata) cv. Justyna. The plants were grown in a nutrient solution enriched with Li in the form of LiCl or LiOH at the following concentrations: 0, 2.5, 20, 50 or 100 mg?Li?dm?3. The obtained results indicate that the presence of Li+ ions in the root environment reduced the yield of edible parts of the lettuce if the Li concentration in a nutrient solution had reached 20 mg?Li?dm?3. However, a yield reduction under these conditions was found to be significant only for LiOH. In plants exposed to 50 mg?Li?dm?3, both shoot and root fresh weights (FW) significantly decreased, regardless of the supplied Li chemical form. On the other hand, under the lowest LiOH dose, a significant increase in the root FW was noted, suggesting beneficial effects of Li on the growth of lettuce plants. However, applied Li concentrations and forms did not affect the l-ascorbic acid content in the lettuce leaves. Regardless of which Li form was used, Li accumulated mainly in the root tissues. An exception was the higher concentration of this metal in the shoots than in the roots of plants supplied with 100 mg?Li?dm?3 in LiCl, and there were almost the same Li concentrations in both examined organs of plants supplied with 100 mg?Li?dm?3 in LiOH. The effectiveness of Li translocation from roots to shoots rose with increasing Li concentrations in the growth medium, and this suggests a relatively ready translocation of this metal throughout the plant. Moreover, these results suggest that Li toxicity in lettuce plants is related to a high accumulation of this element in the root and shoot tissues, causing a drastic reduction in the yield, in the presence either of LiCl or LiOH, but not affecting the l-ascorbic acid accumulation in the leaves.

The effects of Cd, in combination with salicylic acid (SA) and sodium nitroprusside (SNP), on ryegrass seedlings were studied. Exposure of plants to 0.1 mM CdCl2 for 2 weeks resulted in toxicity symptoms such as chlorosis and necrotic spots on leaves. The addition of 0.2 mM SA or 0.1 mM SNP slightly alleviated the toxic effects of Cd. After application of both SA and SNP, these symptoms significantly decreased. Treatment with Cd resulted in a decrease of dry weight of roots and shoots, chlorophyll content, net photosynthetic rate (P n), transpiration rate (T r), and the uptake and translocation of mineral elements. In Cd-treated plants, levels of lipoxygenase activity and malondialdehyde, hydrogen peroxide (H2O2), and proline contents significantly increased, whereas the activities of antioxidant enzymes, such as superoxide dismutase, guaiacol peroxidase, catalase, and ascorbate peroxidase, decreased in both roots and shoots. The results indicated that Cd caused physiological stresses in ryegrass plants. The Cd-stressed plants exposed to SA or SNP, especially to SA + SNP, exhibited improved growth compared with Cd-stressed plants. Application of SA or SNP, especially the combination SA + SNP, considerably reduced root-to-shoot translocation of Cd and increased the activities of antioxidant enzymes in both roots and shoots of Cd-stressed plants. The interaction of SA and SNP increased chlorophyll content, P n and T r in leaves, and the uptake and translocation of mineral elements, and decreased lipid peroxidation and H2O2 and proline accumulation in roots and shoots. These results suggest that SA or SNP, and, in particular, their combination counteracted the negative effects of Cd on ryegrass plants.

The effects of Cd on the growth and Cd uptaking in corn (Zea mays L.) were explored under different Cd stress. The results showed that no reduction in shoot and root dry matter yields were noted when the plants were grown at Cd supply levels ≤100 μmol l?1 nutrient solution. The Cd concentration in the shoots and roots of corn increased sharply with increasing external Cd supply levels, peaked at 50 μmol l?1, and then decreased slowly with further increasing Cd levels due to high Cd toxic effects on root growth. The concentrations of chlorophyll a, chlorophyll b and chlorophyll a + b in the leaf of corn decreased slowly with increasing external Cd supply levels. Proline concentrations of corn increased when the plants were grown under the external Cd influence. The lower concentration of Cd treatments did not influenced the growth of corn significantly, and increased the uptake of Cd, the higher levels of Cd supply caused significantly physiological resposes and decreased the Cd uptaking.

Some plants can tolerate and even detoxify soils contaminated with heavy metals. This detoxification ability may depend on what chemical forms of metals are taken up by plants and how the plants distribute the toxins in their tissues. This, in turn, may have an important impact on phytoremediation. We investigated the impact of arbuscular mycorrhizal (AM) fungus, Glomus intraradices, on the subcellular distribution and chemical forms of cadmium (Cd) in alfalfa (Medicago sativa L.) that were grown in Cd-added soils. The fungus significantly colonized alfalfa roots by day 25 after planting. Colonization of alfalfa by G. intraradices in soils contaminated with Cd ranged from 17% to 69% after 25–60 days and then decreased to 43%. The biomass of plant shoots with AM fungi showed significant 1.7-fold increases compared to no AM fungi addition under the treatment of 20 mg·kg−1 Cd. Concentrations of Cd in the shoots of alfalfa under 0.5, 5, and 20 mg·kg−1 Cd without AM fungal inoculation are 1.87, 2.92, and 2.38 times higher, respectively, than those of fungi-inoculated plants. Fungal inoculation increased Cd (37.2–80.5%) in the cell walls of roots and shoots and decreased in membranes after 80 days of incubation compared to untreated plants. The proportion of the inactive forms of Cd in roots was higher in fungi-treated plants than in controls. Furthermore, although fungi-treated plants had less overall Cd in subcellular fragments in shoots, they had more inactive Cd in shoots than did control plants. These results provide a basis for further research on plant-microbe symbioses in soils contaminated with heavy metals, which may potentially help us develop management regimes for phytoremediation.

The cadmium (Cd) uptake characteristics by Sorghum bicolor cv. Nengsi 2# and Cowley from the acidic sandy loam soil (pH = 6.1) during the entire growth period (100 days) were investigated in pot outdoors in a tropical district of southern China, Hainan Island. The Cd-spiked levels in soil were set as 3 and 15 mg/kg. Correspondingly, the available Cd levels in soil extracted by Mehlich III solution were 2.71 and 9.41 mg/kg, respectively. Basically, two varieties in a full growth period (100 days) did not show a significant difference in their growth and Cd uptake. Under high Cd stress, the plant growth was inhibited and its biomass weight and height decreased by 38.7–51.5% and 27.6–28.5%, respectively. However, S. bicolor showed higher bioaccumulation capability of Cd from soil to plant [bioconcentration factor (BCF)>4], and higher transfer capability of Cd from roots to shoots [translocation factor (TF)>1] under high Cd stress; Cd contents in the roots, stems, and leaves of S. bicolor reached 43.79–46.07, 63.28–70.60, and 63.10–66.06 mg/kg, respectively. S. bicolor exhibited the potential phytoextraction capability for low or moderate Cd-contamination in acidic sandy loam soil.

Siegesbeckia orientalis L. is a promising species for cadmium (Cd) phytoextraction with large biomass and fast growth rate, while little information about their intracellular mechanisms involved in Cd tolerance and detoxification has been explored. A soil pot experiment with total target Cd concentrations of 0, 10, 50, 100, and 150 mg kg?1 were designed to investigate the subcellular distribution, chemical forms and thiol synthesis characteristics of Cd in S. orientalis. More than 90% of Cd was bound to the soluble fractions (48.4–76.5%) and cell walls (19.9–46.3%). Increasing soil Cd concentrations enhanced Cd sequestration into the cell walls. Most of the Cd (69.8–82.7%) in the plant organ was mainly in the forms of pectate and protein integrated Cd and undissolved Cd phosphate, while a minor portion (6.8–20.9%) was in the forms of the inorganic Cd and the water soluble Cd. Nonprotein thiols and phytochelatins significantly increased with increasing soil Cd treatment levels, while glutathione concentrations had no obvious change trends. Therefore, intracellular detoxification mechanisms of Cd in S. orientalis mainly rely on formation of less toxic Cd chemical forms, store of a large amount of Cd in cell wall and synthesis of thiol compounds.

A wide range of cadmium concentrations (from 4 to 200 μM for seedlings and up to 2 mM for germinating kernels) was used to assess Cd toxic effects on maize (Zea mays L.) plants at the different developmental stages: germinating kernels, seedlings (4–9 days), and juvenile plants (34 days). Cd accumulation in plant organs was followed, and its lethal concentration was elucidated. In maize, cadmium was accumulated predominantly in roots; in shoots it was mainly accumulated in the lower leaves, and the higher was leaf position the lower was Cd content in it. At high concentrations (80 and 200 μM), kernels became the substantial cadmium depot. Germinating kernels manifested the lowest sensitivity to cadmium; seedlings were more sensitive; the inhibition of juvenile plant growth attained 90% and more. In the tested range of concentrations, cadmium suppressed shoot mass accumulation harder than that of roots. In 34-day-old plants, water content in shoots was stronger reduced than in roots. Plant death was also manifested earlier in shoots. It was concluded that maize plant sensitivity to cadmium increases with plant growing and that, under conditions of normal mineral nutrition, cadmium inhibits shoot growth more severe than root growth.

Among the technologies used to recuperate cadmium (Cd) contaminated soils, phytoextraction are particularly important, where the selection of suitable plants is critical to the success of the soil remediation. Thus, the objectives of this study were to evaluate the responses of jack-bean and sorghum to Cd supply and to quantify Cd accumulation by these species grown in hydroponic culture. The plants were subjected to 0, 15, 30, or 60 μmol Cd L?1 in the nutrient solution, and gas exchange, plant growth and Cd accumulation were measured at 25 days after starting Cd treatments. The Cd supply severely reduced growth of shoots and roots in both species. In jack-bean, Cd decreased photosynthesis by 56–86%, stomatal conductance by 59–85% and transpiration by 48–80%. The concentrations and amounts of Cd accumulated in the plant tissues were proportional to the metal supply in the nutrient solution. Sorghum was more tolerant than jack-bean to Cd toxicity, but the latter showed a greater metal concentration and accumulation in the shoot. Therefore, jack-bean would be more suitable than sorghum for use in Cd phytoremediation programs based on phytoextraction.

Subcellular distribution and chemical forms of cadmium in Morus alba L.
Morus alba L. (mulberry) is a perennial woody tree and a species with great potential for Cd phyremediation owing to its large biomass and extensive root system. The mechanisms involved in Cd detoxification were investigated by analyzing the subcellular distribution and chemical forms of Cd in mulberry in the present study. These results indicated that 53.27–70.17% of Cd mulberry accumulated was stored in the root and only about 10% were in the leaves. Lots of the Cd was located in the cell wall of the mulberry root and in soluble fraction of the mulberry leaf. Moreover, in roots, the largest amount of Cd was in the form of undissolved Cd-phosphate. While in mulberry leaves and stems, most of the Cd was extracted by 2% Acetic acid and 0.6 M HCl, representing Cd-phosphate and Cd-oxalate. It could be concluded that the Cd combination with peptides and organo-ligands in vacuole of leaf or complexed with proteins or cellulose in the cell wall of root might be contributed to the tolerance of mulberry to Cd stress. The mulberry could be used to remediate the Cd polluted farmland soils.

Pot culture experiments were established to determine the effects of colonization by arbuscular mycorrhizal fungi (AMF) (Glomus mosseae and G. sp) on maize (Zea mays L.) grown in Pb, Zn, and Cd complex contaminated soils. AMF and non-AMF inoculated maize were grown in sterilized substrates and subjected to different soil heavy metal (Pb, Zn, Cd) concentrations. The root and shoot biomasses of inoculated maize were significantly higher than those of non-inoculated maize. Pb, Zn, and Cd concentrations in roots were significantly higher than those in shoots in both the inoculated and non-inoculated maize, indicating the heavy metals mostly accumulated in the roots of maize. The translocation rates of Pb, Zn, and Cd from roots to shoots were not significantly difference between inoculated and non-inoculated maize. However, at high soil heavy metal concentrations, Pb, Zn, and Cd in the shoots and Pb in the roots of inoculated maize were significantly reduced by about 50% compared to the non-inoculated maize. These results indicated that AMF could promote maize growth and decrease the uptake of these heavy metals at higher soil concentrations, thus protecting their hosts from the toxicity of heavy metals in Pb, Zn, and Cd complex contaminated soils.

Remediation of soils is vital to mitigate the negative effects of heavy metals in ecosystems. There is little information available about the metals’ phytostabilization potential of old man saltbush plants [Atriplex nummularia]. A pot experiment in a randomized complete block design was conducted to study the accumulation of heavy metals by old man saltbush plants, as affected by the application of compost and biochar. The cultivation of A. nummularia is an effective tool in immobilizing metals in the contaminated soils. The cultivation of metal-contaminated soil with A. nummularia reduced the availability of Zn, Cu, Cd, and Pb by 20%, 4%, 21%, and 28%, respectively, in comparison to the non-cultivated soil. Zn, Cu, Cd, and Pb concentrations in the aboveground parts of old man saltbush plants were 70–100, 50–80, 4–5, and 50–90 mg/kg of dry biomass. The higher Zn, Cu, Cd, and Pb concentrations were accumulated in the roots, and the lower concentrations were transferred to the shoots of old man saltbush plants. Compost reduced the concentration of Zn, Cu, Cd, and Pb in the shoots by 10%, 19%, 20%, and 6%, respectively, compared to the control soil. Biochar reduced the concentrations of Zn, Cu, and Pb in the shoots by 30%, 38%, and 44%, respectively, compared to the control. Compost had a lower effect in reducing the metals uptake as biochar. Biochar reduced the uptake of Zn, Cu, and Pb in the shoots of the tested plant by 22%, 23%, and 41%, respectively, in comparison to compost. Based on the obtained results, old man saltbush has good characteristics to be a promising candidate for phytostabilization strategies of metal-contaminated soils. Moreover, biochar is a good tool to enhance metals’ phytostablization.

Seedlings of wheat (Triticum aestivum L.) cultivars Jing 411, Jinmai 30 and Yangmai 10 were exposed to 0, 10, 20, 30, 40 or 50 μM of CdCl2 in a solution culture experiment. The effects of cadmium (Cd) stress on wheat growth, leaf photon energy conversion, gas exchange, and Cd accumulation in wheat seedlings were investigated. Gas exchange was monitored at 3, 9, 24 days after treatment (DAT). Growth parameters, chlorophyll content, leaf chlorophyll fluorescence, and Cd concentration in shoot and root were measured at 24 DAT. Seedling growth, gas exchange, chlorophyll content, chlorophyll fluorescence parameters were generally depressed by Cd stress, especially under the high Cd concentrations. Cd concentration and accumulation in both shoots and roots increased with increasing external Cd concentrations. Relationships between corrected parameters of growth, photosynthesis and fluorescence and corrected Cd concentrations in shoots and roots could be explained by the regression model Y = K/(1 + exp(a + bX)). Jing 411 was found to be Cd tolerant considering parameters of chlorophyll content, photosynthesis and chlorophyll fluorescence in which less Cd translocation was from roots into shoots. The high Cd concentrations were in shoots and roots in Yangmai 10 which has been found to be a relative Cd tolerant cultivar in terms of most growth parameters.
