Branch bending has been practiced for decades in China to induce flower buds in ‘Fuji’ apple. However, the optimum bending angle is yet to be elucidated. The main objectives of this study were to compare the effect of branch bending angles (70°, 90° and 110°) on the flowering and nutrient accumulation of 1-year-old shoots of ‘Fuji’ and ‘Gala’ apples and to determine the optimum branch bending angle for each cultivar. In both cultivars, the production of spurs and terminal flower buds, and the total sugar concentration and the carbon-to-nitrogen (C/N) ratio in the shoot terminals increased, whereas the N concentration decreased with increasing bending angles. The nutrient concentration was significantly higher in spurs than in medium and long shoots. The distinction between the changing patterns of C and N concentrations in the bent shoots during the growing season in our study suggested the competition of these two nutrients caused by vegetative and reproductive growth at different growing times. In ‘Fuji’ apple, the proportion of flowering buds appeared to increase more rapidly with the increase of bending angle from 70° to 110° than that in ‘Gala’ apple, and particularly a higher proportion of spurs was observed on ‘Fuji’ branches bent at a larger angle. The increase in the total sugar concentration and the C/N ratio in the shoot terminals of the bent branches might be involved in inducing floral buds after bending. The optimum bending angle was about 90° for ‘Gala’ apple and 110° for ‘Fuji’ apple, respectively. Bending could help farmers to reduce the severity of biennial fruiting in ‘Fuji’ apple.  相似文献

A survey of the reproductive features of the angiosperm flora of the Juan Fernández Archipelago (Chile) is presented to provide a species-based review of reproduction and pollination, to identify generalizations associated with these systems, to understand the evolution of these features, and to utilize these data to promote conservation. The collection of original data was extensive, based on our own fieldwork, and was combined with data from existing literature. Data recorded include habit, sexual system, flower size, shape, and color, and the hypothesized pollination system of the first colonizers. In addition, the data on compatibility, presence and type of dichogamy, observed floral visitors, presence of floral rewards, and currently known pollination systems are summarized. Pairwise comparisons of different features are tested for statistical association. The flora is typically composed of perennials. The majority of the species have very small or small flowers. Inconspicuous flowers (i.e., a shape character describing flowers with no optical attraction) are widespread, as are dish-shaped flowers. Green is the most frequent flower color, followed by white and yellow. Most species are hermaphroditic, 9% are dioecious, and 9% are monoecious. Some 30% of the species are protandrous and 7% protogynous. Detailed studies of compatibility of about 14% of the flora indicate that 85% of these species are self-compatible (SC). Although most species studied are SC, their level of autogamy is low. Nevertheless, selfing mediated via geitonogamy is the most frequent mechanism of pollen transfer. Outcrossing is mainly achieved through dioecy and self-incompatibility (SI), promoted by dichogamy in the hermaphroditic flowers, and facilitated by wind pollination. About 55% of the species offer nectar rewards, and only 2% offer pollen rewards. Floral visitors are rare to uncommon. Two hummingbird species, one of them endemic, are considered as pollinators for 14 plant species. Flies, moths, and beetles are the native insect visitors to flowers, but they have been documented on only 11 plant species (7%). Even insect visits to these few species were rarely observed. Given the infrequent, irregular, and imprecise nature of native insect association with flowers, there is no certainty that any of the species are truly insect pollinated. Two species of introduced ants and a new endemic bee were recorded as well; however, neither is likely currently important to the pollination of the native flora species. About 9% of the extant flora is currently bird pollinated, and we hypothesize that 47% is wind pollinated. However, we propose that most of the colonizers were ancestrally either insect or wind pollinated. There is association between a number of current floral features and the hypothesized pollination of colonizers. Therefore, to a large extent the flower color, shape, and size of the extant flora may express the pollination syndromes of colonizers rather than representing extant pollination. In addition, the presence of nectar in many species of extant flora does not necessarily indicate biotic pollination. Thus, studies of the reproductive biology on oceanic island plants need to be conducted species by species before broad generalizations can be made, because the observed features can be misleading. Possible changes in the pollination system were assessed by comparison of species for which there are reliable data with the hypothesized pollination of their colonist progenitors. The wind-or bird-pollinated species have retained the pollination system of the colonizers. In other instances, species seem to bear a different pollination system: from ancestral insect systems to current hummingbird-or wind-pollination systems. The lack of alternative means of biotic pollination seems to have led in a number of instances to anemophily—in essence a default pollination system. The lack of strong selection pressure for wind pollination and the relative youth of the archipelago may help explain why the features associated with wind pollination in these species are not so obvious. Because there are many recorded extinctions of vascular plants from islands versus those from continental areas, it is imperative to invest additional effort in protecting the remaining island species. Conservation or restoration programs cannot be effective without a deep and broad understanding of the reproductive biology of the plants. In order to conserve these plants, programs must involve a combination of reproductive and environmental measures. The ultimate fate of some species may depend on preserving the plant-hummingbird relationship, including the web of organisms that affect both plant and pollinator. The populations of introduced animals and weeds must be controlled. Experimentally produced allogamous seeds would enhance diversity in restoration programs. In addition, the preservation of habitat seems to be the central challenge to indirectly protect the unique island species.  相似文献

  • The association between plants and flower visitors has been historically proposed as a main factor driving the evolutionary change of both flower and pollinator phenotypes. The considerable diversity in floral morphology within the tribe Antirrhineae has been traditionally related to pollinator types. We used empirical data on the flower visitors from 59 Antirrhineae taxa from the literature and our own field surveys, which provide an opportunity to test whether flower phenotypes are reliable predictors of visitors and pollinator niches.
  • The degree of adjustment between eight key floral traits and actual visitors was explored by testing the predictive value of inferred pollinator syndromes (i.e. suites of floral traits that characterise groups of plant species related to pollination). Actual visitors and inferred pollinator niches (categorisation of visitors’ association using a modularity algorithm) were also explored using Linear Discriminant Analysis (LDA).
  • The bee pollinator niche is correctly classified for flowers with dull corolla colour, without nectar guides, as the most important predictor. Both predictive value and statistical classification prove useful in classifying Antirrhineae taxa and the bee pollinator niche, mostly as a consequence of the high proportion of genera and taxa with occluded corollas primarily visited by bees. Our predictive approach rendered a high Positive Predictive Value (PPV) of floral traits in the diagnosis of visitors/pollinator niches. In particular, a high PPV was found for bees as both visitors and forming pollinator niches. In addition, LDA showed that four pollinator niches are well defined based on floral traits.
  • The large number of species visited by bees irrespective of pollinator syndromes leads us to hypothesise their generalist pollinator role, despite the phenotypically specialised flowers of Antirrhineae.

Since its discovery in the early 1990s, aptamer technology has progressed tremendously. Automated selection procedures now allow rapid identification of DNA and RNA sequences that can target a broad range of extra- and intracellular proteins with nanomolar affinities and high specificities. The unique binding properties of nucleic acids, which are amenable to various modifications, make aptamers perfectly suitable for different areas of biotechnology. Moreover, the approval of an aptamer for vascular endothelial growth factor by the US Food and Drug Administration highlights the potential of aptamers for therapeutic applications. This review summarizes recent developments and demonstrates that aptamers are valuable tools for diagnostics, purification processes, target validation, drug discovery, and even therapeutic approaches.  相似文献

In order to improve our understanding of floral size control we characterised three mutants of Antirrhinum majus with different macroscopic floral phenotypes. The recessive mutant compacta ?hnlich has smaller flowers affected mainly in petal lobe expansion, the dominant mutant Grandiflora has overall larger organs, whilst the semidominant mutation Nitida exhibits smaller flowers in a dose-dependent manner. We developed a cell map in order to establish the cellular phenotypes of the mutants. Changes in organ size were both organ- and region-specific. Nitida and compacta ?hnlich affected cell expansion in proximal and distal petal regions, respectively, suggesting differential regulation between petal lobe regions. Although petal size was smaller in compacta ?hnlich than in wild type, conical cells were significantly bigger, suggesting a compensation mechanism involved in petal development. Grandiflora had larger cells in petals and increased cell division in stamens and styles, suggesting a relationship between genes controlling organ size and organ identity. The level of ploidy in petals of Grandiflora and coan was found to be equivalent to wild type petals and leaves, ruling out an excess of growth via endoreduplication. We discuss our results in terms of current models about control of lateral organ size.  相似文献

Mitochondrial dysfunction is considered to play an important role in the development of diabetic retinopathy. Recent evidence has also shown many similarities between diabetic retinopathy and a low grade chronic inflammatory disease. The aim of this study is to understand the interrelationship between proinflammtory mediator, IL-1β and mitochondrial dysfunction in the accelerated loss of capillary cells in the retina. Using IL-1β receptor gene knockout (IL-1R1?/?) diabetic mice, we have investigated the effect of regulation of IL-1β on mitochondrial dysfunction and mtDNA damage, and increased retinal capillary cell apoptosis and the development of retinopathy. Retinal mitochondrial dysfunction and mtDNA damage were significantly ameliorated in IL-1R1?/? mice, diabetic for ~10 months, compared to the wild-type diabetic mice. This was accompanied by protection of accelerated capillary cell apoptosis and the development of acellular capillaries, histopathology associated with diabetic retinopathy. Thus, mitochondrial damage could be one of the key events via which increased inflammation contributes to the activation of the apoptotic machinery resulting in the development of diabetic retinopathy, and the possible mechanism via which inflammation contributes to the development of diabetic retinopathy includes continuous fueling of the vicious cycle of mitochondrial damage, which could be disrupted by inhibitors of inflammatory mediators.  相似文献

The α-, β-, and γ-crystallins, proteins characteristic for the vertebrate eye lens, have been localized in the developing lens of Notophthalmus viridescens, the eastern spotted newt. Using the immunofluorescence technique, antibodies to the α-, β-, and γ-crystallin classes were applied to tissue sections through the eye region of developing N. viridescens embryos, Harrison (external) Stages 30 to 46+. β-Crystallins were the first of the crystallins to appear in a few cells of the lens vesicle even before the lengthening of the prospective primary fiber cells. γ-Crystallins were first detectable at a slightly more advanced stage in the prospective primary fibers, and α-crystallins in a few cells of the beginning primary fiber area. The external layer/epithelium was negative for β-crystallins until late in lens morphogenesis, and α- and γ-crystallins could not be detected in these cells at any time. This, the first use in amphibia of homologous antibodies specific for the crystallin classes, makes clear that phylogenetic differences exist as to the primacy and relevance of specific crystallins to events during morphogenesis of the eye lens.  相似文献

In this article, we attempt to estimate the contemporary phytoplankton species pool of a particular lake, by assessing the rate of floral change over a period of 15 years. Phytoplankton time series data from Lake Stechlin, an oligo-mesotrophic lake in the Baltic Lake District (Germany) were used. Of the 254 algal species recorded during the 15-year of studies with roughly biweekly sampling, 212 species were planktonic. In the individual plankton years, the recorded total number of species changed between 97 and 122, of which the number of dominants (>1% contribution to the annual average of total biomass) was only 10–19. The 15-year cumulative number of species exhibited an almost linear increase after an initial saturation phase. This increase was attributed to two reasons: increase of sample size and immigration of species new to the flora. Based on a probabilistic model developed in this study, we estimated the number of co-existing planktonic species of the lake as some 180, and the rate of floral change as 1–2 species per year. Of these co-existing species, only few maintain the matter–energy processing ecosystem functions in any particular plankton year. Selection of these dominants is probably driven by mesoclimatic cycles, coupled with human-induced forcing, like eutrophication. All others are hiding as an ecological memory, in the sense of the capacity or experiences of past states to influence present or future responses of the community. Data analyses suggest that selection of the ‘memory species’ that show temporary abundance increases over shorter (several years) periods are largely dependent upon the dominants. These results show that interspecific interactions and the particular autecological features of the dominants, together with their effects on the whole ecosystem, act as a major organizing force. Some phytoplankton species, like Planktothrix rubescens, are efficient ecosystem engineers with cascading effects of both a top-down and bottom-up nature. Historical scientific data on Planktothrix blooms in Lake Stechlin suggest cyclic patterns in long-term development of phytoplankton which, as the legend of the Red Cock suggests, dates back much further than scientific archives.  相似文献

In a recent work on the Tzotzil (Maya) of Chiapas, Mexico, George Collier has suggested that indigenous groups frequently employ agricultural practices which are in obvious disequilibrium with their environment. As a result, he claims, such groups bring about the permanent destruction of their lands and forests. In this article, historical and demographic evidence is presented to demonstrate that the development of commencal agriculture outside of native communities, not overpopulation or technological conservatism within them, lies at the heart of such destruction. Finally, it is argued that anthropologists must consider the evolution of social classes in rural areas if they are to understand the difficulties which economic development entails.  相似文献

This study describes the early expression and function of β-catenin in the gastropod, Crepidula fornicata. In other bilaterians β-catenin functions in cell adhesion, gastrulation, and cell signaling, which is related to the establishment of the dorso-ventral axis and mesendoderm. Here, we studied the distribution of β-catenin mRNA and protein in C. fornicata via whole mount in situ hybridization and by expressing GFP-tagged β-catenin in vivo. During early cleavage, β-catenin mRNA and protein appear to be broadly localized to all cells in the early embryo. The mRNA tends to be concentrated at inter-phase centrosomes in these cells. At later stages, the mRNA is predominantly in the vegetal macromeres, and subsequently in the rudiment of the hindgut, stomodeum, and velar lobes. Expression of full-length GFP-tagged protein suggests that there is no active mechanism to degrade β-catenin within cells of the early embryos prior to the 25-cell stage. However, by the second day of development, when the fourth quartet micromeres have formed, β-catenin becomes selectively stabilized in the progeny of the 4d mesentoblast (e.g., ML and MR and their daughters) and is missing from most other blastomeres, including vegetal macromeres. Over the next 2 days of development, during subsequent divisions of 4d, β-catenin protein becomes progressively degraded, along the proximo-distal axes, within the progeny of the paired mesendodermal bands. The cells located at the tips of the mesodermal bands (2?mL2 and 2?mR2) are the last to contain this protein, which is no longer detected after 4 days of development. In animals like C. fornicata, which undergo a spiral cleavage program (e.g., molluscs, annelids, nemerteans, and polyclad flatworms), the mesentoblast or 4d cell represents the progenitor of endomesoderm (forming hindgut, internal and external kidneys, and various muscles). Therefore, the selective stabilization of β-catenin in the progeny of 4d in C. fornicata is consistent with arguments that a basic, ancestral role of β-catenin lies in the formation of endomesodermal fates. Experiments using a truncated β-catenin clone show that the regions located in the C-terminus, distal to the 11th armadillo repeat, are required for normal stabilization/degradation of β-catenin protein within the embryo. Microinjection of translation blocking β-catenin morpholinos into zygotes led to the down-regulation of β-catenin expression. This resulted in the subsequent failure of gastrulation, but did not interfere with the formation and early cleavage of 4d, although there were no discernable differentiated cell fates in these defective embryos. Green fluorescent protein (GFP)-based video microscopy can provide profound insight into biological processes by generating information on the ‘history,’ or dynamics, of the cellular structures involved in such processes in live cells. A crucial limitation of this approach, however, is that many such structures may not be resolved by light microscopy. Like more recent super-resolution techniques, correlative video-light–electron microscopy (CLEM) was developed to overcome this limitation. CLEM integrates GFP-based video microscopy and electron microscopy through a series of ancillary techniques, such as proper fixation, hybrid labeling and retracing, and so provides sufficient resolution as well as, crucially, cellular ‘context’ to the fluorescent dynamic structures of interest. CLEM ‘multiplies’ the power of video microscopy and is having an important impact in several areas cell and developmental biology. Here, we discuss potential, limitations and perspectives of correlative approaches aimed at integrating the unique insight generated by video microscopy with information from other forms of imaging.  相似文献

The vertical inflorescences of the Mediterranean annual Salvia viridis carry many small, colorful flowers, and are frequently terminated by a conspicuous tuft of colorful leaves ('flag') that attracts insect visitors. Insects may use the flags as indicators of food rewards in the inflorescences below, as long-distance cues for locating and choosing flowering patches, or both. Clipping of all flags from patches of inflorescences in the field reduced the number of arriving insects, but not the total number of inflorescences and flowers visited by them. The number of flowers visited per inflorescence increased with inflorescence size, and inflorescence and flower visits rates significantly increased with patch size. Six percent of the plants in the study population did not develop any flag during blooming, yet suffered no reduction in seed set as compared to flag-bearing neighboring individuals. Removal of flags from all inflorescences in a patch reduced seed set in comparison with untreated controls, while flag clipping from ten randomly selected inflorescences in a patch did not decrease seed production. These results suggest that flags signal long-distance information to potential pollinators (possibly indicating patch location or size), while flower-related cues may indicate inflorescence quality.
Plants that do not develop flags probably benefit from the flag signals displayed by their neighbors, without bearing the costs of signal production. Greenhouse-grown S. viridis plants allocated a low proportion of their biomass to flags. Plants grown under water stress did not reduce biomass allocation to flags as compared to irrigated controls. Water loss rates of picked flags were lower than those of picked leaves. These findings suggest that the expenses of flag production and maintenance are modest, reducing the selective advantage of individuals that do not carry flags. We discuss additional potential evolutionary mechanisms that may select for flag production.  相似文献

Diverse pollinator assemblages may impose complex selection and thus limit specialisation to particular pollinators. Previous work has concentrated on how visitation rates of different pollinators vary in space and time and how pollinators may vary in efficiency. In this study I quantify variation in visitation rates and foraging behaviour of different insect types (1) in space and time and (2) in relation to variation in floral design (flower size and form) and floral display (number of open flowers) for the distylous clonal shrub Jasminum fruticans. Mean visitation rate showed a significant interaction between insect type and population for seven populations in one year, and between insect types and years for a single population over 3 years. There was also a significant interaction between insect type and population for the proportion of flowers visited. In general the number of visits was positively related to the number of open flowers in a patch, but analyses by insect type showed that this was only true for bee flies and butterflies. Short-tongued bees showed a positive relationship between visitation rate and the number of open flowers on the focal stem, and hawkmoths and butterflies made more visits to plants with larger flowers. Hawkmoths were the only insect type to show a positive relation between the number of flowers visited per foraging bout and flower size. The significant differences between different insect types in patterns of variation in visitation rates in response to floral design and display may act to diversify selection on floral traits, and thereby constrain specialisation of the plant to particular pollinators.  相似文献

Anther and ovule development of the theaceous Ternstroemioideae is reported for the first time on the basis of eight specles of three generaAdinandra, Cleyera andEurya. Anthers of these three genera are similar and can be characterized by the following traits: tapetum of glandular type, anther dehiscing latrorse-introrse, both connective and anther epidermis heavily tanniniferous, and connective and even anther wall layers having abundant druses. Their ovules are also very similar in being bitegmic and tenuinucellate, and in having a micropyle formed by the inner integument only, three cell-layered integuments, a raphe bundle terminating at chalaza, usually amphitropous or less often campylotropous ovule, embryo sac formation of Polygonum type, ephemeral antipodal cells, and tanniniferous ovule epidermis. Such ovules are readily distinguishable from those of Camellioideae and all other families. It is suggested that the three genera studied are closely related, and that the degree of embryological specialization is highest inAdinandra and lowest inEurya. On the basis of the significant embryological discrepacies, the Ternstroemioideae seem to have diverged rather distantly from the other core-subfamily Camellioideae of the Theaceae.  相似文献
