The majority of bioassay methods used for determining the susceptibility of the flour mite Acarus siro L. to pesticides provide data which are difficult to assess. In this new method, the mites are confined on treated, moulded filter papers held in position by two glass sheets to form an escape-proof exposure chamber. The method is simple and uses materials that are likely to be readily available. The dose–response data obtained from laboratory and field strains of A. siro to three organophosphorus-based pesticides are presented. The method provided repeatable results and no significant variation when used by different operators. A discriminating dose for the detection of pirimiphos-methyl resistance is suggested.  相似文献

Two types of trap, the BT Trap and PCTM pitfall cone trap, were evaluated against the most widely used method of sampling and sieving for their ability to detect low numbers of storage mites (Acarus siro, Tyrophagus longior and Lepidoglyphus destructor) in bulk oilseed rape, under UK maritime conditions. In addition, a synthetic lure (2-nonanone) was incorporated into the two types of trap to assess whether its presence improved trapping performance. The BT traps and PC traps were significantly more effective in detecting low numbers of mites of all species compared with sampling and sieving. The PC trap with synthetic lure was the most effective method. As the PC trap is already widely used for monitoring insects in cereal bulks in the UK, extending its application to include oilseed rape, and to detect mites, would be a logical and cost effective extension of its use. A method is described for the transfer to microslides of large numbers of mites extracted from house dust, in a single operation. Mites were frozen in water in a watch glass at –20°C. The ice was then everted onto a siliconeized microslide and the meltwater evaporated off on a hotplate at 55°C. Mounting medium and coverslip were then added in the normal way. The technique avoids the difficulty and tedium of transferring individual mites to a microslide and the haphzard and messy process of transfer in liquid medium via a pipette.  相似文献

Previous research has demonstrated that legume proteins have insecticidal activity against stored-product pests, but activity against stored-product mites has not been tested. A study was therefore conducted to explore the potential of bean, Phaseolus vulgaris L., flour as novel botanical acaricide against five species of storage and dust mites: Acarus siro L., Aleuroglyphus ovatus (Troupeau), Caloglyphus redickorzevi (Zachvatkin), Lepidoglyphus destructor (Schrank), and Tyrophagus putrescentiae (Schrank). The effect of wheat, Triticum aestivum L., grain enriched with bean flour to eight concentrations (0, 0.01, 0.1, 0.5, 1, 2.5, 5, and 10%) on population growth initiating from the density of 50 mites per 100 g of wheat was recorded for 21 d under laboratory conditions (grain moisture 14.6% moisture content and 25 degree C in darkness). The enrichment of grain with bean flour suppressed the population growth of all tested species: 0.01% concentration reduced population growth of all tested species to >50% in comparison with the control population. The most sensitive species were A. siro and L. destructor, followed by T. putrescentiae and C. redickorzevi. The least sensitive species was A. ovatus. The terminal (i.e., after 21 d) density of mites positively correlated with bean flour concentration. The suppressive effect of bean flour was not linear but rather asymptotic. The results of this study are discussed in the context of the application of bean flour in integrated control of stored-product mites and the elimination of stored-product mite allergens.  相似文献

The determination of mite numbers in dust samples depends on the effectiveness of the sampling procedure. We report on mite isolation from dust samples by flotation distinguishing between living and dead mites. The results are varied and unreproducible; ageing processes are of relevance in the case of dead mites. Living mites can be determined more reliably when using mite mobility. Two methods based on this principle, the mobility test and the heat escape method, are described and discussed. In the case of a natural analogous mite development on carpets, mite numbers ranging from 80 000 to 200 000 mites per m2 are found after approx. 3 months. It is shown that the mobility of different mite species varies. The mobility test allows the detection of mites in textile objects, and shows how mites are distributed over these objects. It is demonstrated how the infestation of clothes by mites can be determined.  相似文献

Abstract: The model introduced here is an open test which produces experimental results of the same quality as a former test model and at the same time achieves considerable savings in both time and materials. The characteristic feature of the present method is that the mites are kept on a treated glass surface floating on water. We describe methods for interactive visualization and analysis of density maps available in the UCSF Chimera molecular modeling package. The methods enable segmentation, fitting, coarse modeling, measuring and coloring of density maps for elucidating structures of large molecular assemblies such as virus particles, ribosomes, microtubules, and chromosomes. The methods are suitable for density maps with resolutions in the range spanned by electron microscope single particle reconstructions and tomography. All of the tools described are simple, robust and interactive, involving computations taking only seconds. An advantage of the UCSF Chimera package is its integration of a large collection of interactive methods. Interactive tools are sufficient for performing simple analyses and also serve to prepare input for and examine results from more complex, specialized, and algorithmic non-interactive analysis software. While both interactive and non-interactive analyses are useful, we discuss only interactive methods here.  相似文献

Stored product mites are pests of serious economic and medical importance. Recently, it has been shown that the biological control of these pests based on the use of natural compounds and predators had a high potential for success. In this study, we investigated the suppression of three pest mites by combination of the predator Cheyletus malaccensis Oudemans, 1913 and bean flour (Phaseolus vulgaris), which is an antifeedant. Wheat grain (100 g) was contaminated with 100 individuals (Acarus siro (Linnaeus, 1758), Aleuroglyphus ovatus (Troupeau, 1878) or Tyrophagus putrescentiae (Schrank, 1781)). We added bean flour in concentrations of 0, 1, 5 and 10 g kg−1 (wt/wt) and/or the predator with initial predator/prey ratios 0, 1/50, 1/25 and 1/10. The experiment was carried out under abiotic conditions that are optimal for pest development. After 21 days, pest and predators numbers were recorded. Application of flour significantly reduced populations of A. siro, slightly reduced populations of T. putrescentiae, the efficiency increased with increasing concentration. A. ovatus was not affect by bean flour. Populations of all mite species were successfully reduced by the sole addition of C. malaccensis, with a higher efficiency at higher Cheyletus ratios. The additive effect of the predator and flour was mainly apparent in the reduction of T. putrescientae. The population of C. malaccensis was not affected by the presence of bean flour. It is therefore recommended to use bean flour for the reduction of A. siro, a combination of bean flour and Cheyletus for the reduction of T. putrescientae, and only Cheyletus for the reduction of A. ovatus. An easily constructed apparatus for extraction of VAM fungal spores from soil samples by a modification of Gerdemann and Nicolson's wet sieving and decanting method is described. For the soils employed in this study, it proved considerably more effective and more precise than either the original wet sieving and decanting method or differential water/sucrose centrifugation. The apparatus gave results that were less subject to interference from extraneous particles than the wet sieving and decanting method. The effect of prior soil dispersion is also reported.  相似文献

Western flower thrips, Frankliniella occidentalis (Pergande), is the principal insect pest of spring flower crops grown in the northeastern United States for use as bedding plants. Neoseiulus (=Amblyseius) cucumeris (Oudemans) is a predacious mite reared commercially that is recommended for control of western flower thrips in various vegetable and flower crops at a rate of ca 53 mites/m2/week. Efficacy on spring flower crops, however, is not well demonstrated, reports being either from other crops or extension demonstration trials. In two trials (each replicated), we compared suppression of western flower thrips in spring bedding plants provided by (1) N. cucumeris at the recommended rate, (2) spinosad (at the labeled rate), the most widely used thrips-control pesticide, and (3) both combined. Trial No. 1 was run in mixed bedding plants in commercial greenhouses and Trial No. 2 in impatiens monocultures in University greenhouses. We found that in commercial greenhouses, variation in species composition of crops and movement of plants during crop production made it difficult to detect any significant effects. In an impatiens monoculture (Trial No. 2), we found better evidence of partial suppression of thrips larvae and adults by treatments. Spinosad alone provided the best control, with mites alone usually providing control intermediate to that of spinosad alone and the untreated control. Control from spinosad plus mites was not significantly different from that of spinosad alone (all treatments evaluated as counts of thrips per plant, in flowers). In another University-based trial (Trial No. 3), we compared the commercially recommended rate of N. cucumeris (53 mites/m2/week) to a 3- to 4-fold higher rate (190 mites/m2/week) in impatiens monocultures. This trial was replicated twice in the fall of 2004 and once in spring of 2005 in Amherst, Massachusetts. We found that the higher release rate, while not resulting in statistically significantly more mites per plant (in flowers) than the standard rate, did suppress thrips larvae per plant (in flowers) by 50–75%, a higher level than that achieved by the recommended standard rate. No reductions, however, were found in counts of adult thrips, either as numbers per plant (in flowers) or as numbers caught per yellow sticky card, except for one replication in which thrips counts were lowered compared to controls by mites (at both release rates). We conclude that N. cucumeris, especially at the higher rate, provides partial control of western flower thrips in impatiens bedding plants, but that control from spinosad is better. Biological control of western flower thrips with this predator is not a complete thrips IPM program, but may be used together with spinosad or other materials to prevent development of pesticide resistance. This approach is most likely to be of value in crops grown as continuous relay plantings or a series of different, but thrips-susceptible, crop species.  相似文献

A simple method is described for separating fungus gnat eggs from soilless growing media. The flotation/extraction method primarily involves the use of a MgSO4 solution (density 1.065 g cm(-3)) and a series of sieving procedures. Bradysia sp. nr. coprophila Lintner eggs were collected in an isolation chamber containing adult fungus gnats. Three soilless growing media were used: Metro-Mix 560 with Scott's Coir, Sunshine LC1 Mix, and Universal SB 300 Mix. Each growing medium was inoculated with 100 fungus gnat eggs. Growing medium samples were processed using separatory funnels and a MgSO4 solution. Eggs were washed through a 250-microm sieve and collected on a 53-microm sieve, and then the eggs were rinsed into glass petri dishes. The sieving process separated fungus gnat eggs from the larger growing medium particulates (perlite, bark, and peat), providing a sample consisting of water, eggs, and fine growing medium particulates (peat). Using a vortex for 60 s increased the number of fungus gnat eggs recovered (37.4 +/- 3.7-67.1 +/- 1.9 [mean +/- SEM]) for all three growing media. The mean number of fungus gnat eggs recovered from the three soilless growing media was significantly different, with the growing medium containing coir (Metro-Mix 560) having the highest recovery rate of 61.0 +/- 2.1. For all three growing media, there was > 50% recovery of fungus gnat eggs. In addition to fungus gnat eggs, this method also may prove useful for quantitative recovery of shore fly (Scatella sp.) eggs and the eggs of other arthropods from soilless growing media.  相似文献

A simple method is described for a quick determination of thesize structure of zooplankton communities. This is based onsieving of a live sample, using one or two sieves of known meshsize, and determining the dry weight of the size fractions.Size separation by live sieving is validated statistically onDaphnia galeata, a common planktonic cladoceran, by comparingsize-sieved fractions against direct microscopic measurementsof individuals. A non-linear model was used for assessment ofsieving statistics. The sieving technique facilitates the gatheringof size-structured biomass data for zooplankton for routinemonitoring or long-term studies.  相似文献

Debris often hampers the detection of mites in washed leaf samples. We describe in detail a method for the extraction of mites from leaf samples, based on the adherence of mite cuticles to liquid paraffin, at the interface of paraffin and ethanol in a so-called mite-counting channel. We demonstrate its efficacy by comparing the mite numbers in samples before and after extraction. We illustrate the method's reliability by extracting known numbers of a taxonomic variety of plant-inhabiting mites, manually added to mite-free debris: for 13 of the 15 taxa all mites were retrieved. This method can also be used to extract small non-mite arthropods such as scales, whiteflies, thrips, cicadellids, hymenopteran parasitoids and psyllids.  相似文献

We observed the number of predatory mites (Phytoseiidae:Typhlodromus caudiglans) on the foliage of 20 North American species of grapes (Vitis spp) plus the domesticated EuropeanVitis vinifera, all grown in a common garden. We found relatively few phytophagous mites. The numbers of phytophagous mites were not correlated with the plant characteristics that we measured. We found approximately five times as many predatory mites as phytophagous mites and the numbers of these phytoseiid predators were not affected by the availability of prey. Similarly, numbers of phytoseiids were unaffected by plant gender and, hence, the availability of pollen, another source of food. The numbers of phytoseiids were not clustered according to the taxonomic grouping of the tested plant species. Leaf surface characteristics explained over 25% of the variance in the numbers of phytoseiids. Numbers of phytoseiids were positively associated with the density of vein hairs, the density of bristles in leaf axils, and the presence of leaf domatia. These results suggest that sheltered habitats rather than food availability may limit the numbers of phytoseiid mites on grapevines.  相似文献

Aerial dispersal of European red mite, Panonychus ulmi (Koch), in commercial apple orchards was estimated by trapping windborne mites. Studies were conducted at four orchards in eastern New York during 1989 and 1990 and at three orchards in western New York during 1989. In each orchard mites were trapped in three locations; the interior of the orchard, at the border of the orchard and in a field or woodlot beyond the orchard. Large numbers of mites were captured, even when the numbers of mites on apple foliage were well below levels where mite injury to leaves was visible (less than five per leaf). The log numbers of mites trapped were linearly related to the log density of mites on leaves and this relationship was consistent for each year and region the study was conducted. The trap captures among the three locations in and outside an orchard were highly correlated. The implications these findings may have on metapopulation dynamics and resistance to acaricide dynamics are discussed.  相似文献

Abstract 1. Ecological theory suggests that density‐dependent regulation of organism abundance will vary from exogenous to endogenous factors depending on trophic structure. Changes in abundance of soil arthropods were investigated at three trophic levels, springtails (Collembola), predaceous mites (Acari), and macro‐arthropods (spider, adult and larval beetles, centipedes). Predictions were that springtails are predator regulated and mites are food limited according to the Hairston et al. (1960) model, which predicts alternating regulation by competition and predation from fungi to springtails to mites to macro‐arthropods. The alternate hypothesis was based on the bottom‐up model of trophic dynamics, which predicts that each trophic level is regulated by competition for resources. 2. The relative contributions to springtail and mite population dynamics of endogenous (i.e. density‐dependent population growth related to food availability) and exogenous (i.e. predation and weather) factors were tested using time‐series analysis and experimental manipulation of water conditions. Box patterns were distributed within an aspen forest habitat located in the Canadian prairies and surveyed weekly from May to September 1997–1999. Each box depressed the leaf litter, creating a microhabitat island for soil arthropods that provided counts of invertebrates located immediately beneath the boxes. 3. Strong evidence was found for endogenous control of springtail and mite numbers, indicated by a reduction in population growth related to density in the previous week. Contrary to predictions, no evidence was found for regulation of springtail numbers by mites, or for regulation of mite numbers by macro‐arthropods. Springtail population growth rate was related positively to current springtail density (8 and 23% variation explained) and related negatively to 1‐week lagged density (85 and 58%), and related negatively to temperature (5 and 5%) for time‐series data and for experimental addition of water respectively. Mite population growth rate was related positively to current mite density (54%) and temperature (4%), and negatively to 1‐week lagged mite density (20%) and precipitation (6%) for time‐series analysis. For experimental addition of water, mite growth rate was related positively to current mite density (44%) and temperature (5%), and negatively to 1‐week lagged density (11%). Results differed from the Hairston et al. (1960) model predictions but were consistent with a bottom‐up view that springtail and mite populations were regulated intrinsically by competition for food and secondarily by temperature as a function of reproduction.  相似文献

There is a wide and strong allergenic background in Moscow, which is determined by relatively high population density and regular frequency of the allergenic mites in various premises. 34 species have been identified, 5 of them are allergen producers. The mites of the family Pyroglyphidae are the main component of the house dust fauna. Dermatophagoides pteronyssinus and D. farinae distributed all over the world are predominate. A comparative evaluation of the fauna structure, numbers and frequency of the allergenic mites in the flat dust of the allergic patients and healthy people as well as premises is given. Possible importance of the premises in prevalence and circulation of the allergenic mites in the modern town is estimated for the first time.  相似文献

Microvessels were isolated from autopsied human brain using a simple procedure involving disruption, sieving, and centrifugation on a sucrose density gradient. The present procedure is characterized by isolation, from frozen autopsied brain, of materials either from the cerebral cortex or white matter, and subsequent separation of the capillary fraction from the large vessel fraction. The preparation appears highly purified under phase-contrast microscopic examination. The purity was also established by the enrichment of gamma-glutamyl transpeptidase activity and by the nearly negligible cerebroside content in the vessel fractions as compared to the brain homogenate.  相似文献

Summary The present study investigated recolonization patterns of selected soil and litter arthropods following replanting of pasture with rainforest species in the Mary River catchment of eastern subtropical Australia. While extensive research has been conducted in rehabilitated mined sites in Australian dry sclerophyll forests, very little attention has been paid to rainforest restoration on previously pastoral land. We examined the utility of soil and litter arthropod groups for monitoring the progress of restoration, and the relationship between arthropod assemblage patterns and environmental factors potentially under the control of those doing the replanting. Leaf litter was extracted from 20 sites: five remnant rainforest, five pasture, and 10 sites that had been revegetated (from 1 to 12 years previously) with a diversity of indigenous species. Ants (identified to genus), centipedes, millipedes, isopods, amphipods and mites were enumerated and their assemblages described with multi- and univariate methods. Ant genera alone proved unable to distinguish pasture from rainforest, and mites (identified only to oribatid or non-oribatid) proved of limited use because these two groups were present in all samples in extremely high numbers that overwhelmed the contributions of other arthropod groups. However, a coarse taxonomic approach using five arthropod groups (ants, centipedes, millipedes, isopods, amphipods) clearly discriminated between pasture and rainforest, and also arranged the revegetated sites between these two extremes. Simple frequency scores based on the presence/absence of these arthropod groups in each of three replicate subplots were sufficient to achieve this separation of site types. Habitats created by close planting of trees and mulching may accelerate invertebrate colonization and, thus, promote the rapid establishment of processes and functions characteristic of developing rainforests.  相似文献
