The initial stage of bacterial infection is characterized by an increase in the level of total lipids and polyenoic fatty acids in membrane phospholipids of blood lymphocytes of carp (Cyprinus carpio L.). In the fish infected with aeromonads, changes in the ratios of fatty acids in phospholipids are similar for either lymphocytes, liver, or whole blood. The extent to which these changes are pronounced depends on the original physiological status of the fish.  相似文献

This study was conducted to investigate the effect of pistachio green hull extract (PGHE) on hematological and serum biochemical changes in common carp, Cyprinus carpio. Three hundred common carp (11.65 ± 1.65 g) were fed one of five different dietary treatments (with three replications) containing 0, 0.5, 1.5, 4.5 or 9 g PGHE kg?1diet for ten continuous weeks. Each tank had a 90‐L capacity and water flow rate of about 500 ml min_1.Total phenolic compounds of the different diets differed significantly (P < 0.001) according to the amount of PGHE. At the end of experiment, six fish were removed randomly from each treatment. Blood samples were taken for hematological and serum biochemical analyses at room temperature. Liver tissue samples were processed for histology and stained by H&E. The results indicated that all doses of tested PGHE induced no significant changes in hematocrit, hemoglobin, or erythrocytes, nor alkaline phosphatase, alanine transaminase, lactate dehydrogenase, total protein, albumin, globulin, triglycerides, low‐density lipoprotein, high‐density lipoprotein, glucose or cholesterol in the serum. Leukocytes were higher (P < 0.01) in fish fed a 1.5, 4.5, or 9 g PGHE kg?1 diet when compared to the 0.5 g PGHE kg?1 diet group or the control. Serum aspartate transaminase in treatments containing a 4.5 or 9 g PGHE kg?1 diet was significantly (P < 0.001) higher in comparison with the control. Liver histology showed focal necrosis, cytoplasm degeneration, lateral nuclei and an increase in Kupffer cells following PGHE administrations. The results of this trial indicated that although there were no significant changes in most hematological and biochemical parameters, PGHE could induce some adverse pathological effects on liver tissue.  相似文献

Changes in blood ion concentration, oxygen absorption, blood haemoglobin concentration and blood urea-ammonia nitrogen level were measured in a group of mirror carp, Cyprinus carpio (L.) infected with measured numbers of Ichthyophthirius multifiliis under standardized conditions. The disease was characterized by a drop in serum sodium and magnesium and increase in serum potassium. The ability of infected fish to absorb oxygen was reduced as was their ability to tolerate dissolved oxygen concentrations under 5 ppm. Blood ureaammonia level was elevated during the disease but blood haemoglobin level remained unchanged.  相似文献

In this study, we investigate the toxic effects of cobalt chloride on some hematological factors of the carp Cyprinus carpio, such as white blood cell count, red blood cell count, hemoglobin, hematocrit, mean corpuscular volume, mean corpuscular hemoglobin, and mean corpuscular hemoglobin concentration. At first, LC50 of cobalt in C. carpio was measured during 96 h after exposure. Also, physicochemical parameters of water including pH, dissolved oxygen, viscosity, temperature, and conductivity were monitored, continuously. The results showed that LC50 values of cobalt in C. carpio were 327 and 328 mg/L in two replicates, respectively. Then, the changes in some hematological factors in the five treatment groups placed under concentration of 100, 200, 300, 400, and 500 mg/L cobalt were compared with the control group. Based on hematological tests conducted in this research, exposure of carp to 500- and 300-mg/L concentrations of cobalt in 48 h showed significant difference (p?<?0.05) in white blood cell count. The concentration of 500 mg/L cobalt in 24 h showed a significant difference in the amount of hemoglobin, number of red blood cells, and hematocrit level as compared with the control group. The concentration of 100 mg/L cobalt in 48 h did not show a significant difference in comparison with the control group (p?>?0.05). Also, the concentration of 500 mg/L cobalt in 24 h showed a significant difference in the amount of mean corpuscular volume and mean corpuscular hemoglobin as compared with the control group and other treatments. Also, the percentage of mean corpuscular hemoglobin concentration in a concentration of 200 mg/L cobalt in 24 h showed a significant difference as compared with the control group and other treatments.  相似文献

Cysteine is important for protein synthesis, detoxification, and diverse metabolic functions. However, cysteine metabolism has been poorly described in fish, and the role of the therapeutic effect in pesticide toxicology on aquatic organisms is unknown. The aim of this study was to determine the effects of regular cysteine treatment on the hematology, biochemistry, apoptosis, oxidative DNA damage, and antioxidant parameters in fish blood after chemical application. Therefore, fish were exposed to cypermethrin for 2 weeks. Then two different concentrations of N‐acetylcysteine (NAC) were applied for a 4‐day treatment period and compared with the group of the self‐healing process. At the end of the treatment, the hematological index, blood biochemical parameters, paraoxonase (PON), arylesterase (ARE), and myeloperoxidase (MPO) activities in the fish blood samples were investigated. With regard to the hematological parameters, statistical differences were obtained except for mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) (P < 0.05). Enzyme activities (ARE, PON, and MPO), as well as some biochemical parameters (creatinin [Cre], alanine amino transferase, total glyceride, alkaline phosphatase, iron, calcium, low density lipoprotein‐cholesterol [LDL‐C], sodium, and potassium), were found to be importantly different among all groups at the P < 0.05 level, while 8‐hydroxydeoxyguanosine and caspase‐3 levels were determined to be high in the pesticide group but decreased significantly in NAC‐treated groups ( P < 0.05). According to the results of the study, acute cysteine treatment showed an ameliorative effect on the hematological index, biochemical parameters, PON, MPO, and ARE in the blood in the all the treatment group fish. The positive effect of NAC on protein synthesis, detoxification, and diverse metabolic functions against cypermethrin toxicity was more effective in 1.0 mM NAC. NAC has an important therapeutic effect on pesticide‐induced hematoxicity for fish in terms of all the data.  相似文献

In this study, the effects of tributyltin (TBT) on biochemical parameters (antioxidant responses and Na+‐K+‐ATPase) in different tissues were investigated by using juvenile common carp (Cyprinus Carpio) as well as growth and ion regulation–related genes expression and stress‐related proteins profiling in fish liver. Oxidative stress indices and Na+‐K+‐ATPase showed tissues‐specific responses in fish exposed to different TBT concentrations. All tested genes related to GH/IGF‐I axis and ion‐regulation were significantly induced in the TBT group with lower concentrations (except for the igfbp3 in 10 μg/L) and were inhibited in 20 μg/L. In addition, the profiling of two proteins Hsp 70 and MT were increasing in a dose‐dependent manner under TBT stress. In short, TBT‐induced biochemical and molecular responses in different tissues were reflected in the measured parameters in the test. On the basis of TBT residue levels in the natural environment, more long‐term experiments at lower concentrations will be necessary in the future.  相似文献

The present study investigated the potential changes in hematologic parameters and histology of fish Heteropneustes fossilis when exposed to industrial waste water of different concentrations. The toxicant enter into the fish body from different rout, inhalation, dermal oral, and other several rout for entering in the fish body. The blood parameters of fish H. fossilis control group and treatment were investigated on 1st, 5th, 10th, and 20th days of experiment, hematologic variation was observed in RBC, WBC, MCV, MCH, and MCHC count. Histopathologic changes in liver, intestine, gill, muscle, and heart showed increasing degrees of damage in the tissues in correlation with the accumulation pattern of pesticides, while liver, intestine, gill, muscle, and heart of control group exhibited a normal architecture. Toxic effects of pesticides vary in different organs of the fish H. fossilis significant. The fish exposed to higher concentration showed uncoordinated alterations in behavioral responses, especially erratic and jerky swimming, physiologic, malformation, histologic, hematologic and biochemical changes, frequent surfacing and in gulping, mucus secretion, an increase in opercular movement, and copious secretion of mucus of all over the body.  相似文献

The author investigated the presence of various carotenoids in the different parts of the body of Cyprinus carpio L. by means of columnar and thinlayer chromatography.The material in the form of the fertilized eggs, week-old fry, month-old fry, four-months old fry, one-year old fry and marketable carp were investigated.Yugoslavian carp brought to Poland as fry, and hybrid the wild carp crossed with of the cultivation carp, were also taken for analysis. The carotenoid content of Cyprinus carpio bred in waters polluted by municipal wastes and specimens suffering from diseases of the airbladder was also determined.The content of carotenoids, the provitamin of vitamin A, in Cyprinus carpio depends of many factors existing during the cultivation of these fish.  相似文献

Thermal discharge from power stations can affect normal environmental conditions and change in heat shock proteins expression of native fish with increasing temperature. In this study, we investigated levels of Hsp70 in the heart, kidney, brain and gill of the common carp Cyprinus carpio both in long-term heat discharge environment and after 24 h acute heat shock exposure. In laboratory exposure experiments, fish acclimated at 10 °C were exposed to various elevated temperatures (20, 24 and 28 °C). Hsp70 concentrations were determined in tissues by Western blotting analysis after one dimensional SDS-PAGE separation. In the field study, the level of Hsp70 in the gill of the carp remained at control values, and Hsp70 expression in the heart, kidney and brain underwent a 2.8 to 3.7-fold increase. A lower thermal sensitivity of the Hsp70 response of the brain, compared with the heart, kidney and gill, was observed in the laboratory experiments. Our data show that these tissues had different levels of Hsp70 responses to thermal influence both in acute exposure and long-term acclimation. The pattern of tissue Hsp70 expression may have a close relationship with the thermal tolerance of the carp and allows the fish to survive long-term thermal pollution.  相似文献

Histological and protein pattern changes in the gill of the common carp, Cyprinus carpio were investigated following exposure to sublethal concentrations of the organochlorine insecticide, endosulfan. Hyperplasia, lamellar fusion, curling of the tips of primary lamellae, exudation of erythrocytes and bulging of the tips of primary lamellae were the histological lesions observed. Protein pattern of the gill varied at different sampling periods characterised by disappearance of certain fractions and occurrence of additional fractions. It was concluded that the severity of histological changes and protein pattern variations seen in the gill was dependent on both the duration of exposure and the concentrations of endosulfan used (over the range 5—25 % of the 96 hr LC50 value, 5·199ppb).  相似文献

Morphological analyses of 183 specimens of Japanese common carp Cyprinus carpio (171 from Lake Biwa and 12 from nursery ponds) using genetic hybrid indices demonstrated that the typical native Japanese strain of C. carpio has a more elongate body, more branched dorsal‐fin rays, fewer and shorter gill rakers, more developed pneumatic bulb, more coiled pneumatic duct, longer posterior swimbladder and shorter intestine than the typical introduced C. carpio. These results provide a basis for a better understanding of the ecological characteristics and taxonomic status of the endangered Japanese strain of C. carpio.  相似文献

The present study was to investigate the interactive effect of ammonia and crowding stress on ion-regulation and expression of immune-related genes in juvenile turbot (Scophthalmus maximus). The fish were exposed to four total ammonia nitrogen (0, 5, 20, 40 mg/L TAN) and two stocking density. After 96 h of exposure, blood, gill, and liver samples were collected to measure biochemical parameters and mRNA levels of immune-related genes. The results showed that co-exposure to high TAN (20 and 40 mg/L) and high density significantly increased plasma sodium (Na+), gill Na+/K+-ATPase activity and mRNA levels. Following individual and combined exposure to high TAN and high density, the heat shock protein 70 (HSP 70), HSP 90, tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β) genes expression were obviously higher than their control. Conversely, the lysozyme (LZM) and hepcidin mRNA levels were down-regulated in liver of fish exposed to high TAN alone and combination of high TAN-density. Moreover, glutathione S-transferase (GST) mRNA levels significantly increased in treatments with individual high TAN and high density, but decreased in high TAN-density co-exposed fish. Overall, ion homeostasis and immune status were adversely influenced in high exposed turbot under high density.  相似文献

Only single cells in the carrier fish species Carassius carassius (Linnaeus, 1758) for koi herpesvirus (KHV) are infected in contrast to large numbers in the susceptible species common carp Cyprinus carpio (Linnaeus 1758). Several species of the family Cyprinidae have been described as virus carrier species, showing no clinical signs of a KHV disease but able to transmit the virus to other susceptible fish. In this study, 72 common carp Cyprinus carpio (Linnaeus, 1758), 36 tench Tinca tinca (Linnaeus, 1758), 36 crucian carp Carassius carassius (Linnaeus, 1758) and 36 common roach Rutilus rutilus (Linnaeus, 1758) were experimentally infected with KHV (isolate “Israel”) by immersion and kept at 20°C. The fish were euthanized at 12 timepoints over a period of 90 days and virus DNA was quantified in tissues by a real‐time TaqMan PCR. Whereas KHV‐DNA was found in Cyprinus carpio for up to 90 days, the virus DNA was detectable only in single individuals of Rutilus rutilus, Tinca tinca and Carassius carassius for up to 25 days after experimental virus exposure. Tissue samples of Cyprinus carpio and Carassius carassius were screened by in‐situ hybridization. Positive signals were found in various organs of the common carp tested crucian carp. In the latter species a much smaller number of virus‐positive stained cells was detected compared to the infected carp.  相似文献

This study investigated the hypoglycemic effect of the methanol extract of Shorea roxburghii leaves (SRL) in high fat diet/high fructose solution (HFDHF) and streptozotocin (STZ) induced type 2 diabetes mellitus (T2DM) in rats as well as evaluating its ameliorative potentials in altered biochemical and hematological parameters in the treated rats. T2DM was induced in Sprague Dawley (SD) rats by feeding with HFDHF for 4 weeks and administering STZ (35 mg/kg, i. p.). Diabetic rats were given SRL extract at doses of 100 and 400 mg/kg for 30 days. The food and water intake were monitored on a daily basis, while the fasting blood glucose (FBG) levels and body weight were measured weekly. Biochemical and hematological parameters as well as histopathological studies of the pancreas were also evaluated. SRL significantly decreased FBG and improved the body weight, food and water intake of treated diabetic rats. Furthermore, biochemical and hematological parameters including liver and kidney function enzymes, lipid profiles, white blood and red blood cells parameters were markedly ameliorated by SRL. Histopathological analyses of the pancreas indicated reconstitution of β‐cells architecture in SRL treated rats. The results of this study suggest that SRL has antidiabetic potential and can be considered for the treatment of T2DM.  相似文献

Cyprinid fish of different mature age classes (3+ -4+) and stocks (100, 300 and 500 kg/ha) were introduced into each of three experimental ponds with area of 0.3 ha (average depth ca 1.7 m) while the fourth pond was left free of fish. Bream (Abramis brama L.), white bream (Blicca bjoerkna L.) and roach (Rutilus rutilus L.) made up 75% of the total cyprinid biomass, with wild carp (Cyprinus carpio L.) as the remaining 25%. The introduced fish spawned successfully. The high (above 300 kg/ha) planktivorous and benthivorous fish stocks resulted in several qualitative and quantitative alterations of the food chain structure in our simulation pond experiments. These alterations must primarily be assigned to changes caused by both the zooplanktivory and benthivory nature of the stocked fish populations. At the higher levels of fish biomass, Secchi depth was influenced significantly by chlorophyll-a concentration. Most of the variance in suspended solids concentration could be explained by the biomass ratio of the mature benthivorous fish. There was a clear shift in algal cell size in the ponds with the higher fish stocks: ponds with more fish had larger cells later in the summer. The relative influence of young cyprinid fish on crustaceans species composition and biomass, and mature populations on benthic fauna abundance and biomass, was sufficiently greater at higher (300–500 kg/ha) fish stock rates.  相似文献

Cyprinus carpio (Linnaeus, 1758) caught in Sidi Salem Dam in 2011 were analysed for the levels of organochlorine pesticides (OCPs), including hexachlorobenzene (HCB), aldrin, heptachlor, lindane, dieldrin, endrin, and pp-DDT and its two main metabolites pp-DDD and pp-DDE, in order to evaluate the extent of contamination of the dam. Fish were sampled monthly within the dam during 2011. Muscle, liver and gill tissues were analysed by gas chromatography for the determination of OCPs. Samples had variable concentrations of OCPs, ranging from 2.26 to 17.93 ng g?1, 0.59 to 42.67 ng g?1 and 3.30 to 84.27 ng g?1, which were isolated from gills, muscle and liver, respectively. The organochlorine pesticide levels found posed no threat to human health. The highest OCP levels were found in the liver tissues. The pp-DDE concentrations and their ratios to ΣDDTs indicate aged DDT pollution and suggests that there has been no recent input of technical DDT from the agricultural areas into Sidi Salem Dam.  相似文献

Smallmouth bass (Micropterus dolomieu) were collected to quantify the nature and prevalence of biomarker responses, including biochemical indices, toxicopathic lesions and general health indices, among fish collected from polychlorinated biphenyl (PCB)-contaminated and nearby uncontaminated reaches of the Kalamazoo River, Michigan, USA. Blood and tissue samples (gill, liver, spleen, head kidney, trunk kidney, thyroid and gonads) were collected and preserved at necropsy for biochemical and histological analyses. The body condition factor and liver somatic index were significantly lower in fish collected from the downstream, contaminated site. Plasma vitellogenin was not detected in male fish collected from either site. Liver ethoxyresorufin-O-deethylase activity and liver and spleen superoxide dismutase activity were significantly depressed in fish collected from the downstream site. Significant toxicopathic lesions such as glycogen depletion, enhanced macrophage aggregates, hepatic foci of cellular alteration (i.e. preneoplastic lesions) and neoplasia were also detected in the liver of fish collected from the downstream site. This study indicates that many of the biochemical and histopathological biomarker responses were associated with liver and body tissue PCB concentrations. Taken together, the biomarkers of exposure and effect strongly suggest that fish within the downstream site are adversely affected by PCBs and other chemical stressors.  相似文献

The immunosuppressive effects of bath exposure to a sub lethal concentration of the synthetic pyrethroid alpha-permethrin (3.05 x 10(-4) mg l(-1)) in the Indian Major carp, Labeo rohita was studied after 45 days' exposure. In some groups, the effects of alpha-permethrin on non-specific defences and serum enzymes of carp were investigated after challenge with Aeromonas hydrophila. Several nonspecific immune responses and serum enzymes were reduced after exposure of alpha-permethrin. Bactericidal activity of rohu serum was reduced significantly in pesticide and bacteria treated fish. The Glutamic Oxaloacetate Transaminase (GOT) and Glutamic Pyruvate Transaminase (GPT) activity were increased in immunosuppressed fish. Blood glucose level was elevated significantly and Hb% was reduced significantly in pesticide and bacteria treated fishes as compared to the negative control.  相似文献
