Quercus floribunda Lindl. (tilonj oak), a late successionalspecies, and Cupressus torulosa D. Don. (surai), an early successionalspecies, were compared in terms of seed germination and seedlinggrowth. In Petri dish germination tests in which light quality,temperature and osmotic water stress were varied, the germinationof both species was reduced by continuous dark or far-red lightand by osmotic water stress. These effects were greater on thegermination of surai than on that of tilonj oak. The growthof seedlings in soil was measured over a twelve-month periodduring which different levels of shading and different wateringregimes were applied. Surai was less affected than tilonj oakby reduced soil moisture, but the effect of deep shade treatmentswas greater on surai than with tilonj oak. At higher soil moisturestress seedlings of surai attained greater height then tilonjoak, but the latter achieved higher dry weight and a higherroot: shoot and lower leaf-weight ratio. In a series of experiments the desiccation-sensitive seeds ofQuercus robur were exposed to drying conditions both beforeand after a period of moist storage. Viability loss occurredat higher moisture contents in stored seed than in newly harvestedseeds. Measurements were made at intervals during desiccation.In both stored and unstored seeds viability loss was precededby an increase in the rate of ethane evolution, a commonly usedindicator of lipid peroxidation, and by an increase in electrolyteleakage indicative of membrane damage. Jasmonic acid (JA), itsmethyl ester (MeJA) and ABA were quantified in the same extractsfrom both cotyledonary and axis tissues. The concentration ofall three hormones was higher in the axis than in the cotyledonsof untreated seeds and were within the range of concentrationsquantified elsewhere in seed tissues from other species. Theconcentration of JA, MeJA and ABA progressively increased duringdrying in both cotyledons and axes of whole seeds and in excisedaxes prior to viability loss and then subsequently declined.The concentration of these hormones increased earlier duringdrying in stored seeds in line with their enhanced desiccationsensitivity. Exogenous JA, MeJA and ABA were shown to inhibit germination.However, none of these substances promoted ethylene evolution,which also inhibits germination of Q. robur seeds, or inducedsenescence-like deterioration. The results presented are discussed in relation to the natureof desiccation sensitivity and viability loss in Q. robur seeds. It has been suggested that rate of desiccation can influencethe expression of recalcitrant behaviour in seeds, thus complicatingthe task of determining which seeds are truly recalcitrant.The objective of this study was to see if variable rates ofdesiccation influenced such behaviour inQuercus nigra L., atree seed known to be recalcitrant. Acorn moisture content, leachate conductivity, and germinationwere determined at various times during desiccation at threerates at 27 and 40°C. Moisture contents and germinationdecreased as the severity of desiccation increased. Leachateconductivity increased slightly but was not a sensitive indicatorof loss of viability. The critical (lethal) moisture contentfor these acorns was 10–15%, although rehydration within48 h of reaching this level prevented death in about 25% ofthe acorns. At 27°C any desiccation treatment that producedlosses of 30–50mg of moisture per g of acorn dry weightper day should be suitable as a test for recalcitrance in thegenus. Apparent physiological or fungal damage at 40°C rulesout the higher temperature for such a test. Seeds of Araucaria hunsteinii K. Schum. dried quicker at 29°Cthan at 19°C and quicker with the seed-coat removed thanwhen intact; seeds enclosed in polyethylene bags increased inmoisture content. At 15°C, seeds in a flow of air driedquicker than seeds in a box with silica gel, which in turn driedquicker than seeds in a box with no desiccant. No loss of germinationability occurred on drying fresh seed from 53 to about 32 percent moisture content (fresh weight basis); during further desiccationthe percentage germination was related to percentage moisturecontent in the form of a sigmoid curve, culminating in a completefailure to germinate at approximately 14 per cent moisture content.A consistent relationship was observed for all treatments andthe mean critical moisture content for seed death (failure togerminate) was near 23 per cent. Excised embryos grew on 1 percent agar but died if previously subjected to 14 h of desiccationat 15°C. In contrast, no relationship was found between germination andmoisture content of A. cunninghamii D. Don on desiccation from21 to 7 per cent moisture content. Possible causes for the observeddifference in response to desiccation are discussed and methodsfor seed storage are considered. Background and Aims

Several widespread tree species of temperate forests, such as species of the genus Quercus, produce recalcitrant (desiccation-sensitive) seeds. However, the ecological significance of seed desiccation sensitivity in temperate regions is largely unknown. Do seeds of such species suffer from drying during the period when they remain on the soil, between shedding in autumn and the return of conditions required for germination in spring?

Methods

To test this hypothesis, the Mediterranean holm oak (Quercus ilex) forest was used as a model system. The relationships between the climate in winter, the characteristics of microhabitats, acorn morphological traits, and the water status and viability of seeds after winter were then investigated in 42 woodlands sampled over the entire French distribution of the species.

Key Results

The percentages of germination and normal seedling development were tightly linked to the water content of seeds after the winter period, revealing that in situ desiccation is a major cause of mortality. The homogeneity of seed response to drying suggests that neither intraspecific genetic variation nor environmental conditions had a significant impact on the level of desiccation sensitivity of seeds. In contrast, the water and viability status of seeds at the time of collection were dramatically influenced by cumulative rainfall and maximum temperatures during winter. A significant effect of shade and of the type of soil cover was also evidenced.

Conclusions

The findings establish that seed desiccation sensitivity is a key functional trait which may influence the success of recruitment in temperate recalcitrant seed species. Considering that most models of climate change predict changes in rainfall and temperature in the Mediterranean basin, the present work could help foresee changes in the distribution of Q. ilex and other oak species, and hence plant community alterations.  相似文献

Effects of 2 °C chilling on the threshold moisture contentsand water potentials for various physiological processes wereestimated forAesculus hippocastanumL. seed. Seed harvested atthe time of maximum seed fall exhibited a dual response to drying:partial drying from near 50% to 32–40% moisture contentprogressively increased germination percentage (at 16 °C)up to various peak values; further desiccation was detrimental,confirming that the seeds are ‘recalcitrant’. Themoisture content for optimum germination was increased by atleast 10% as the chilling period was raised from 0 to 9 weeks.A negative linear relationship was found between log10mean timeto germinate and probit final germination, regardless of pre-treatment,indicating that partial desiccation and chilling are interchangeablein promoting germination of hydrated seed. For nearly fullyhydrated seeds, increasing the chilling period from 6 to 26weeks increased the viability-loss onset point for desiccationinjury from near 40% to about 48% moisture content without alteringthe drying rates of seed tissues. Extending moist chilling invarious seed lots from 0 to 26 weeks decreased subsequent longevityat 16 °C. For 26-week-chilled seeds longevity (the periodto lose one probit of germination) differed above and belowa threshold moisture content of 48%. It remained constant inthe moisture-content range 48–38%, but increased progressivelyas moisture content was raised above 48%. This threshold moisturecontent coincided with the value above which chilled seed pre-germinatedin storage. Question: Lower montane treeline ecotones such as the inner Alpine dry valleys are regarded as sensitive to climate change. In the dry Valais valley (Switzerland) the composition of the widespread, low altitude Pinus forests is shifting towards a mixed deciduous state. The sub‐boreal P. sylvestris shows high mortality rates, whereas the deciduous sub‐mediterranean Quercus pubescens is spreading. These species may act as early indicators of climate change. We evaluate this hypothesis by focusing on their differences in drought tolerance, which are hardly known, but are likely to be crucial in the current forest shift and also for future forest development. Methods: We used dendroecological methods to detect species‐specific patterns in the growth response to drought. The relationship between radial growth of 401 trees from 15 mixed stands and drought was analysed by calculating response functions using yearly tree‐ring indices and monthly drought indices. PCA was applied to the response ratios to discover spatial patterns of drought response. Results: A species‐specific response to moisture as well as a sub‐regional differentiation of the response patterns were found. While Quercus showed a response mainly to the conditions of the previous autumn and those of current spring, Pinus did not start responding before May, but showed responses throughout the whole summer. Quercus may restrict physiological activity to moist periods; growth of Pinus was much more dependent on prior growth. Conclusions: Given that the climate is changing towards (1) longer summer drought periods, (2) higher mean temperatures and (3) shifted seasonally of moisture availability, Quercus may benefit from adapting better to drier conditions. Pinus may increasingly face problems related to drought stress as it depends on summer moisture and has a smaller adaptive capacity due to its long‐lived photosynthetic tissue.  相似文献

The effect of water stress in three Quercus species belongingto different habitats is investigated. Chlorophyll concentrationdecreased in all Quercus species with the greatest decreaseoccurring in Quercus robur. An increase of total carotenoidcontent was observed in Quercus coccifera and Quercus ilex,while in Q. robur water stress effected a carotenoid contentreduction. Anthocyanins increased with decreasing relative watercontent in the three Quercus species, the largest increase occurringin Q. coccifera. Immature Glycine max (L.) Merr. seeds initially at 50–70mg fresh weight were successfully grown and matured in vitroin detached pods. Surface sterilized pods were floated in aliquid medium containing 5 per cent sucrose, minerals, and glutaminein 125 ml Erlenmeyer flasks and incubated at 25 °C under350–400 µE m–1 s–1 white light. Seedswhich were matured in vitro increased tenfold in dry weight,were visually similar to commercial seeds of the same size,were tolerant to desiccation and germinated with normal seedlinggrowth. Excised pods transported dye from the pedicel to thegrowing seed within 120 min. Seed growth characteristics of Aesculus hippocastanum were examinedin detail during development from about 70 to 140 d after anthesis(DAA), mainly in 1988 and 1989. Mean fresh and dry weights increasedfor both the axis and the whole seed up to the time of peakseed fall at 135 DAA with no cessation before fruit abscission.Water per seed increased up to 100 DAA, after which no furtherincrease occurred; moisture content declined for the embryonicaxis and whole seed respectively from above 75 and 65% at 95DAA to 65 and 50% at 130 DAA. At fruit shedding in 1990 waterpotential values of -1·2, -2·6 and -1·1MPa were observed for the testa, cotyledon and axis tissuesrespectively; relevant sorption isotherms are presented. Decreases in seed moisture content during development were accompaniedby increases in desiccation tolerance and in germinability,both reaching their maximum at the time of peak seed fall. Atmaturity, only about 10% viability was retained on drying seedto 20% moisture content; it is confirmed that the seeds are'recalcitrant'. The exact relationship between moisture contentand germination during development was dependent on the deptof dormancy, as judged by the period of chilling required; eachduration of chilling at 2°C within the range 3-12 weeksyields a curve of sigmoid shape. No germination occurred at26°C without chilling, but nearly full germination can beobserved for samples collected at 6 weeks before maximum seedfall with 12 weeks chilling. As pea plants (Pisum sativum L.) cv. Finale and Solara developin the field, pods located at the second flowering node, increasein fresh and dry weight until 75 d after planting (DAP); thereafter,dry weight and moisture content decrease rapidly. Seed developmentconsists of three phases, all limited by low moisture content(MQ. The first phase (PI) corresponds to the formation of theembryo and surrounding structure (MC is stable at 85%). Duringthe second phase (P2) the cotyledons are filled (MC decreasesfrom 84% to 55%). The third phase (P3) entails desiccation onthe mother plant. Fresh and dry weight increase until 75 DAP(55% MC or physiological maturity, PM) and rapidly decreaseduring the third phase until 14% MC. Leakage conductivity ofimmature seeds reflects sap arrival in the testa and accumulationin the apoplast it is important during PI and the onset of P2,but during P2 it decreases slowly until physiological maturityand rapidly thereafter. Immature seed germination is noted from62 DAP, increases until 67 DAP, then decreases with a maximumat physiological maturity; some days after physiological maturitygermination is complete (up to 100%). Seedling growth from immatureseeds during P2 is low, but slowly increases until physiologicalmaturity. When germination (up to 100%) is attained seedlinggrowth is normal. Hydration of the immature seed indicates that55% MC is a highly specific value at which a transition in thehydration rate is occurring. Moisture loss from pea seeds duringthe P2 filling stage appears to be necessary for seed adaptationto the abrupt desiccation which occurs at physiological maturity.Physiological maturity is attained when disruption of the vascularconnection between the pod and the mother-plant occurs. Merr.] develop, they undergoa change in seed moisture. When excised prematurely from thepod and planted, seeds do not exhibit seedling growth until63 d after flowering (DAF) when the seed moisture has fallenbelow 60%. In contrast, seed germination (radicle protrusion)can occur when seeds as young as 35 DAF (68–79% moisture)are excised, but this germination docs not lead to comparableseedling growth frequencies unless seeds are first given a moistureloss treatment to artificially reduce their moisture below 60%.A moisture loss treatment applied at 35 DAF thus enables seedto undergo the transition from germination (cell expansion)to seedling growth (cell division and expansion) to the extentthat treated immature seed have a vigour index comparable toseeds matured on the plant (100%). The pattern of protein synthesisin vivo was examined in 35 DAF seed using [35S]-methionine incorporation.When moisture loss treatment was applied for 24 h to 35 DAFseeds, seeds synthesized several new polypeptides when comparedwith untreated seeds at the same developmental stage. The sameseed samples showed 0% seedling growth in the absence of moistureloss treatment and 80% seedling growth when the treatment hadbeen applied. Moisture loss from soybean seeds appears to bea prerequisite for the synthesis of new proteins which may bepart of the metabolic process or processes that allow the soybeanseed to undergo the transition from seed germination to seedlinggrowth. Seeds of four cultivars of arabica coffee (Coffea arabica L.)were tested for germination following hermetic storage for upto 12 months at several different combinations of temperaturesbetween –20 °C and 15 °C and moisture contentsbetween 5% and 10% (wet basis). Most of the seeds from one cultivarwithstood desiccation to between 5% and 6% moisture content,a seed water potential of approximately –250 MPa, butthose of the remaining three cultivars were much more sensitiveto desiccation damage. Moreover, in all four cultivars, seedlongevity at cool and sub-zero temperatures, and at low moisturecontents did not conform with orthodox seed storage behaviour:viability was lost more rapidly under these conditions thanat either warmer temperatures or higher moisture contents. Theresults confirm that coffee seeds fail to satisfy the definitionsof either typical orthodox or recalcitrant seed storage behaviour.These results, therefore, point to the possibility of a thirdcategory of storage behaviour intermediate between those oforthodox and recalcitrant seeds. The relationship between modern pollen assemblages and modern vegetation along two elevational transects within the Transverse and Peninsular Ranges of southern California, USA, is demonstrated using cluster analysis of the pollen data. Cluster analysis separates the Sonora Desert vegetation, Valley grassland/agricultural land and chaparral vegetation types on the San Jacinto Mountains transect. Chaparral is not easily separated on the San Bernardino Mountains transect, probably due to the presence of Quercus dumosa (scrub oak) there. The lower montane QuercusPinus (oak – pine) community is distinct from other forest types, and can be subdivided palynologically based upon relative importance of Quercus, Pinus and Cupressaceae [primarily Calocedrus decurrens (incense cedar)] pollen. Subdivisions include QuercusPinus – Cupressaceae, Quercus – Cupressaceae – Pinus and QuercusPinus assemblages. Higher elevation PinusAbies (pine – fir) and Pinus-dominated communities are also differentiated from one another, although the subalpine vegetation type only occurs on the San Bernardino Mountains transect. Though the study area presently straddles a transition between winter-wet and summer-wet climatic regimes, differences between the pollen assemblages in the two mountain ranges are minimal. Pollen assemblages from lower elevations document the effects of human activities, primarily agriculture, on the modern pollen rain of the region, with the occurrence of introduced citrus (Citrus sp.) and shade (Eucalyptus sp.) trees and weedy disturbance indicators (e.g., Brassicaceae).  相似文献

Seeds with low vigour as a result of ageing showed a large improvementin seed quality following a 2 h soak in water, reflected inan increased rate of germination, higher emergence in soil,and the retention of high germination after 24 h rapid ageingin a controlled deterioration vigour test. Similar, but smallerimprovements were seen after a priming treatment using polyethyleneglycol. Seeds dried to their initial moisture content aftertreatment retained these beneficial effects. The treatmentshad little effect on high vigour seed. These findings suggestthat seed invigoration by hydration treatments results fromthe repair of previously sustained deterioration. Abstract Seed germination, and survival and growth of seedlings of four dominant tree species, Quercus dealbata, Quercus griffithii, Quercus glauca and Schima khasiana were studied in the treefall gaps and forest understorey of an undisturbed mature-phase humid subtropical broadleaved forest in northeast India. Three important microenvironmental factors namely photosynthetically active radiation (PAR), soil moisture and litter depth, were also measured in the forest understorey and gaps and correlated with seedling mortality. Seed germination of S. khasiana was significantly higher in the treefall gaps than in the understorey; among the tree species studied, it had the highest germination. Quercus seedlings were abundant in the understorey and small gaps, while S. khasiana seedlings were more numerous in the large gaps. The survivorship curves for the seedling populations revealed that the three Quercus species survived better in the understorey, while S. khasiana did so in the gaps. PAR and soil moisture were positively correlated with tree seedling mortality, which occurred mainly during the winter months. The Quercus seedlings grew better in the forest understorey and small gaps and S. khasiana seedlings in the large gaps. The differential performance of the tree seedlings to the conditions prevailing in the understorey and gaps of two sizes indicates that different species were adapted to different light environments depending upon their optimum requirements. This could be an effective mechanism for promoting species coexistence in the forest community.  相似文献

Embryos excised from fresh seed had a moisture content of about37% (fr. wt.), 7% above that of the whole seed: mainly as aresult of the dryness of the seed coat. When cultured for 14d at 26°C embryo growth ranged from unorganized to fullyorganized. Embryos which had been directly surface-sterilizedexhibited lower fully organized growth than aseptically excisedembryos. During desiccation of the whole seed at 15°C and 15% r.h.moisture loss from the embryo was slow until the embryo reacheda mean critical moisture content for fully organized growthin vitro of about 30%. After this point the rate of moistureloss increased, quickly diminishing the moisture content differentialbetween the embryo and the whole seed. No such shoulder in theembryo desiccation curve was observed when using a low viability(7%) seed batch. When dried below 30% moisture content, suchaseptically excised embryos increasingly exhibited unorganizedgrowth in vitro, with only root meristem survival (46%) at 13%moisture content. In contrast, a much higher level of root meristem survival (84%of control) was observed when aseptically excised embryos wererapidly desiccated to 13% moisture content, using a flow ofsterile air. However, the benefits of rapid drying were lostif the embryos were directly surface-sterilized prior to desiccation.Furthermore, 80% of aseptically excised embryos which survivedrapid desiccation to 20% moisture content exhibited root meristemsurvival following subsequent cryopreservation. In constrast to seeds of orthodox species, those of recalcitrantspecies do not acquire desiccation tolerance during their developmentand are shed from the parent plant at high water contents. Dehydrinproduction in seeds of recalcitrant species was examined duringdevelopment and germination, in response to abscisic acid (ABA),and following the imposition of various water-deficit-relatedstresses, including desiccation, water stress, high salt, highosmolarity, and low temperature. Two tropical species exhibiteda differential capacity to produce dehydrin-related proteinsduring seed maturation. Dehydrins were present in axes and cotyledonsof Castanospermum australe seeds during mid-maturation and atmaturity. In Trichilia dregeana, no dehydrin-related polypeptideswere detected in the mature seed. During the development ofC. australe seeds, the nature of the dehydrin related polypeptidesaccumulated in the cotyledons and axis changed and new polypeptideswere detected in the mature seeds that were not present duringmid-maturation. The dehydrins present in cotyledons of matureseeds (31, 37 and 40 kDa) were still detectable after germination(i.e. in untreated seedlings). These dehydrins became less abundantin the cotyledons of C. australe seedlings following ABA andall stress treatments except cold, although most of the dehydrinswere still detectable. An exception was the desiccation-treatedseedlings, in which no dehydrins were detected. In the rootsof C. australe seedlings, no dehydrins were found after germinationnor were they induced in the root by ABA or any of the stresstreatments imposed on seedlings. Seedlings of Trichilia dregeanadid not produce dehydrins in the roots or cotyledons when exposedto ABA or water-deficit-related stresses. Freshly-harvested and dried seeds of Bromus sterilis L. withrespectively 52-54% and less than 10% moisture contents, differedmarkedly in their rates of germination and in their responsesto light. Dried seeds attained full germination in darknessat 15 ?C by 4 d from sowing and were inhibited by the formationof Pfr. In contrast, freshly-harvested seeds required 3-4 weeksto attain maximum germination and were promoted by Pfr. Themodified photoresponse of the seeds was induced only when theseeds were dried in an alternating temperature regime. Preservation of genetic diversity within germplasm repositories represents an important tool for plant conservation. However, seeds must tolerate extreme levels of post-harvest desiccation and cold to realize benefits of ex situ storage. Factors including local climate and habitat impact expression of desiccation and freezing tolerance especially for widely distributed species. Our aim here was to understand the influence of a latitudinal gradient on seed desiccation and cryo-freezing tolerance. We sampled mature U. paniculata seeds from two geographically and genetically distinct populations then examined seed-water relations and germination following desiccation via equilibrium drying assays (0.5 to 91% RH; ?797 to ?12.9 MPa). Germination ability after drying and subsequent cryo-freezing treatments (?196?°C, 1 to 1440 min) was also evaluated. Seeds of both populations displayed similar reverse sigmoid moisture sorption isotherms characteristic of desiccation tolerant tissues. Furthermore, initial seed water potential (?63 and ?90 MPa) was considerably lower than the lethal limit (?20 MPa) identified for desiccation sensitive tissues. Final germination (range 58–93%) and temporal patterns differed significantly between populations following desiccation and cryo-freezing stress, but these germination responses were similar to initial germination. A higher proportion of non-germinated, yet viable seeds remained for the northern compared to southern population. Location does influence germination response, but differential germination is related to seed dormancy rather than desiccation or cryo-freezing sensitivity. Ex situ conservation of U. paniculata is therefore feasible across the latitudinal gradient studied here.  相似文献
