对菜蛾盘绒茧蜂Cotesia plutellae多分DNA病毒的特性及其对寄主小菜蛾Plutella xylostella幼虫的生理效应进行了研究。结果表明:菜蛾盘绒茧蜂雌蜂输卵管萼中含有大量的多分DNA病毒(polydnavirus, PDV);一个PDV内含多个核衣壳,最多可达16个;核衣壳长40~168 nm,直径39~40 nm;PDV仅在输卵管萼细胞内复制;雌蜂产卵时,随蜂卵将PDV注入寄主血腔,并扩散到寄主的许多组织中;PDV可能先通过脱膜再侵染寄主组织。雌蜂经Co60辐射处理后再寄生(即假寄生)小菜蛾2龄、3龄和4龄初期的幼虫,被寄生后的寄主幼虫几乎全部不能化蛹,但末龄(即4龄)幼虫期显著延长,并在寄生后期,幼虫胸部有褐色的短翅芽出现;即将化蛹的4龄末小菜蛾幼虫被假寄生后,即使每头寄主被过寄生9次,依然能正常化蛹,但不能羽化。假寄生与正常寄生后寄主的脂肪体数量和形态结构有明显的不同,推测在正常寄生的情况下蜂卵孵化时释放的畸形细胞及随后的幼蜂可能对脂肪体的结构产生了作用。  相似文献

Females of insect endoparasitoids inject substances along with the egg at oviposition, which can regulate or induce changes in a number of host physiological processes to benefit the developing parasitoid. These changes can be caused by substances such as venoms, calyx fluids, or symbiotic‐associated virus particles (polydnavirus), which are injected by females along with their eggs, and by substances secreted by parasitoid‐derived tissues (teratocytes) or the developing parasitoid larvae. Teratocytes (dissociated cells of the serosal membrane after parasitoid eclosion) release substances that have roles (i) in parasitoid nutrition, (ii) in the digestion of host tissues, and (iii) in the regulation of host development. Teratocytes of Toxoneuron nigriceps (Viereck) (Hymenoptera: Braconidae) have been implicated in the arrestment of the host development and in the regulation of circulating levels of host ecdysteroids. Here we describe the cDNA of a teratocyte‐secreted chitinase and its expression during parasitoid development, and discuss its putative role in this host–parasitoid association.  相似文献

Crude venom and calyx fluid from Cotesia plutellae (Hymenoptera Braconidae) were assayed for biological activity toward hemocytes of Plutella xylostella (Lepidoptera Plutellidae). Venom from C. plutellae displayed high activity toward the spreading of plasmatocytes of P. xylostella early in the incubation period, and the inhibition was more severe as the concentration of venom increased. However, most inhibited hemocytes spread normally after being incubated for 4h. No effects were found toward granular cells from the host. Additionally, the venom from C. plutellae had some lethal effects on hemocytes of P. xylostella at high concentrations. In contrast, when incubated with different concentrations of calyx fluid, the spreading of some hemocytes was inhibited, some began to disintegrate, and some were badly damaged with only the nucleus left. After 4h, the majority of hemocytes died. The same results were observed when hemocytes were incubated in calyx fluid together with venom. These results show that calyx fluid from C. plutellae may play a major role in the suppression of the host immune system, whereas venom from C. plutellae has a limited effect on hemocytes and probably synergizes the effect of calyx fluid or polydnavirus.  相似文献

过寄生、寄生时寄主龄期和寄生后寄主饥饿处理影响菜蛾盘绒茧蜂Cotesia plutellae(Kurdj.)幼蜂及畸形细胞的发育。显微解剖和观察表明,4龄小菜蛾Plutella xylostella L.幼虫被寄生后,其体内菜蛾盘绒茧蜂幼蜂发育不整齐、假寄生比例增高。过寄生后,每头被寄生的寄主血腔中畸形细胞数量明显增多,但直径变小;随着过寄生程度的加剧,幼蜂发育严重受阻。寄主营养显著影响体内幼蜂及畸形细胞的发育,被寄生的小菜蛾经饥饿处理62 h后,体内畸形细胞的数量、活性明显降低,与此同时,幼蜂的发育也受到明显抑制,寄主发育与寄生蜂和畸形细胞的发育呈正相关性。由此可见,寄主不同龄期、过寄生及寄主营养状况均对寄主体内幼蜂和畸形细胞发育产生影响。  相似文献

We studied the development of teratocytes derived from two congeneric gregarious endoparasitic species, Cotesia chilonis and C. flavipes, parasitizing two congeneric novel hosts, Diatraea saccharalis and D. grandiosella. The host-parasitoid combinations studied allowed us to investigate relationships between host suitability and teratocyte development. D. saccharalis was a suitable host for both parasitoids, whereas D. grandiosella was suitable for C. chilonis development but often encapsulated C. flavipes progeny. Encapsulation of C. flavipes by D. grandiosella commenced around the time of parasitoid egg hatch, when teratocytes were released into the host's hemolymph. The gregarious parasitoids studied here released about 200 teratocytes per egg. Both absolute and normalized (teratocytes/parasitoid) numbers decreased over time. D. saccharalis supported more C. flavipes-derived teratocytes than D. grandiosella, possibly because of the unsuitability of the latter host. On intermediate assay days the number of C. flavipes-derived teratocytes was greater than for C. chilonis. However, C. chilonis-derived teratocytes grew larger than C. flavipes. Teratocytes in all host-parasitoid combinations doubled in size during parasitoid development. Teratocytes generally grew larger in D. grandiosella, which was a less suitable host.  相似文献

Zoophthora radicans (Zygomycetes: Entomophthorales), Diadegma semiclausum (Hymenoptera: Ichneumonidae), and Cotesia plutellae (Hymenoptera: Braconidae) are all natural enemies of the diamondback moth, Plutella xylostella (Lepidoptera: Yponomeutidae). Adult C. plutellae are not susceptible to Z. radicans infection but the pathogen can infect and kill adult D. semiclausum. Infection of adult D. semiclausum prior to exposure to P. xylostella host larvae significantly reduced the number of parasitoid cocoons subsequently developing from the host larvae. Although Z. radicans infection of P. xylostella larvae prior to parasitism by D. semiclausum or C. plutellae always resulted in the death of the immature parasitoids, neither species discriminated between healthy and Z. radicans-infected host larvae in an oviposition choice experiment. However, host larvae recently killed by Z. radicans were always rejected by D. semiclausum but sometimes accepted by C. plutellae. At 20 degrees C, egg to pupa development took 6.7 and 7.8 days for D. semiclausum and C. plutellae, respectively. C. plutellae parasitism significantly increased host instar duration but D. semiclausum parasitism did not. Cadavers of P. xylostella larvae parasitized 1 day prior to fungal infection showed no reduction in Z. radicans conidia yield. However, cadavers of larvae parasitized 3 days prior to fungal infection demonstrated a marked decrease in Z. radicans conidia yield. Z. radicans infection of P. xylostella larvae < or = 4 days after parasitism resulted in 100% parasitoid mortality; thereafter, the reduction in parasitoid cocoon yield decreased as the time between parasitism and initiation of fungal infection increased. The extended duration of the host larval stage induced by C. plutellae parasitism increased the availability of the parasitoid to the pathogen. Estimates of interspecific competition indicated a similar pattern for the interaction between Z. radicans and each species of parasitoid.  相似文献

Summary A method for in vitro cultivation of teratocytes from the egg parasitoidTelenomus heliothidis (Hymenoptera: Scelionidae) is described. Parasitoid eggs, from which teratocytes were derived, were collected from 24-h-oldHeliothis virescens (Lepidoptera: Noctuidae) eggs previously parasitized byT. heliothidis females. Optimal culture conditions, including species and concentration of serum, were determined experimentally. Thirty percentManduca sexta hemolymph or 10% chicken serum in Hink’s TNH-FH medium were found to generate the most satisfactory number of teratocytes per parasitoid larva. Teratocytes cultivated in vitro showed similar development and morphology to those produced in vivo. However, cultured teratocytes lived approximately 10 times longer than teratocytes in natural hosts and were not dependent upon the presence of the parasitoid larva for normal development.  相似文献

Teratocytes originating from the serosal membrane of an endoparasitic wasp embryo, which is one of the species found in the Hymenopteran families Braconidae, Platygasteridae and Socelionidae, act an important role in coordinating the relationship between the larval wasp and its host. It may be concluded that inhibiting the immune system of the host, regulating development of the host and nourishing the parasitoid larva are three major functions of the teratocytes. By means of releasing inhibitor of phenol oxidase, a fundamental ingredient of the immune system of the host, the teratocytes prevent the host from encapsulation, which is fatal to the parasitoid larva. In the process of parasitism, the teratocytes reduce ecdysteroid titer and inhibit the metabolism of JH in the hemoymph of the host, and subsequently result in the developmental arrest of the host larva. On the other hand, the teratocytes secrete proteins or present themselves as food to the parasitoid. Although all those above are generally acknowledged functions of the teratocytes, there are other putative functions of the teratocytes, such as secreting histolytic enzymes and mycostatic material, and protecting the host from overharmed by the parasitoid before the parasitoid emerges from its host. The studies in this area can provide us more understanding and knowledge about the relationship between a parasitoid and its host.  相似文献

寄主小菜蛾Plutella xylostella被内寄生蜂菜蛾盘绒茧蜂Cotesia plutellae寄生后,其取食、发育及营养代谢在各种寄生因子的作用下伴随幼蜂的发育而发生很大的变化,畸形细胞作为调节因子之一也发挥了重要的作用。本实验通过比较被寄生和未被寄生小菜蛾血淋巴蛋白浓度以及两种血淋巴对菜蛾盘绒茧蜂幼蜂进行体外培养的培养液的蛋白浓度,发现被寄生小菜蛾血淋巴比未被寄生小菜蛾血淋巴的蛋白浓度略低但差异不显著,而未被寄生小菜蛾血淋巴幼蜂培养液的蛋白浓度显著低于被寄生小菜蛾血淋巴幼蜂培养液的蛋白浓度,证明畸形细胞的蛋白质分泌功能。被寄生后期, 小菜蛾体重明显大于未被寄生的小菜蛾体重,而脂肪体重量相比正好相反;通过显微染色观察,在小菜蛾念珠状脂肪体表面粘附有畸形细胞,对脂肪体进行分解破坏而使其成颗粒状; 蛋白含量和脂滴浓度测定也表明,脂肪体的可溶性蛋白含量和脂滴浓度也迅速降低,同比低于未被寄生小菜蛾。而与此同时,幼蜂正处在快速生长阶段,中肠酯酶的活性逐步上升,幼蜂得以快速消化吸收小菜蛾体内的营养直到完成幼虫发育,整个幼蜂的脂滴浓度也达到了最大值。因此寄生后期,推测在畸形细胞的协助下,幼蜂吸收了寄主小菜蛾体内的营养为自身生长发育所用。  相似文献

应用高效液相色谱法检测施用于寄主幼虫的多杀菌素可否传递到在其体内发育的寄生蜂幼虫。以对多杀菌素高抗的小菜蛾Plutella xylostella幼虫作为菜蛾绒茧蜂Cotesia plutellae的寄主,待绒茧蜂发育到1龄幼虫时,将浓度为50 mg/L的多杀菌素点滴到寄主幼虫背板上,随后让寄主幼虫取食经50 mg/L多杀菌素处理过的甘蓝叶片,寄主幼虫和其体内的蜂幼虫再发育3天后,将寄主幼虫解剖取出蜂幼虫,用高效液相色谱法对经多杀菌素处理的小菜蛾幼虫体液以及绒茧蜂幼虫匀浆液进行检测,结果多杀菌素的2个活性成分spinosyn A和spinosyn D均被检测到,两者的多杀菌素残留浓度分别是2.79 mg/L和0.94 mg/L。这表明,通过寄主幼虫体壁接触和取食进入其体内的多杀菌素,可通过寄生蜂幼虫体壁浸透、蜂幼虫对寄主血淋巴的取食,或这两种途经一起进入蜂幼虫的体内,对蜂幼虫产生作用。  相似文献

The braconid Cotesia plutellae is an important larval parasitoid of the diamondback moth (Plutella xylostella), a major pest of crucifers in the tropics and sub-tropics. The in-flight searching behaviour of C. plutellae was investigated in a wind tunnel and the close-range attack behaviour observed in cages. The relative importance of volatile stimuli emanating from the plant-host-complex, oilseed rape (Brassica napus) – P. xylostella, in the long-range attraction of C. plutellae was investigated. Plants that were mechanically damaged, or damaged by P. xylostella larvae, were attractive to the parasitoid. Host-damaged leaves remained attractive to the parasitoid after removal of the host larvae. These results indicate that C. plutellae predominantly uses plant derived stimuli in its in-flight searching behaviour. An oviposition experience or contact with a host-damaged leaf prior to the bioassay significantly increased the response to these volatile cues. The foraging behaviour of C. plutellae is compared with other braconid larval parasitoids attacking lepidopteran hosts on crucifers.  相似文献

Abstract.  1. Little is known about underlying mechanisms by which plants indirectly affect parasitism success in hymenopteran endoparasitoids. The hypothesis that host-plant effects can challenge the innate immune system of an insect host was experimentally tested in this study using a model tritrophic, crucifer – lepidopteran [ Plutella xylostella (L.)] – parasitoid [ Cotesia plutellae (Kurdjumov)], system.
2. The effects of host-plant suitability on herbivore performance and parasitism were examined. The bottom-up effect of plant suitability on host-parasitoid immune responses was then evaluated using measures of cellular and humoral effectors.
3. Host-plant quality showed a significant effect on the encapsulation response of P. xylostella to first instar but not to second instar parasitoid larvae. Encapsulation was never sufficient to prevent parasitoid emergence.
4. Poor host-plant suitability suppressed phenoloxidase activity in the absence of the parasitoid. The suppressive effect of C. plutellae on phenoloxidase activity was much greater and no plant effects were detectable after insects had been parasitized.
5. Despite strong plant effects on parasitism, those on immune effectors of the host were transitory or overwhelmed by the effect of the parasitoid.
6. These results demonstrated that plant-mediated variation in parasitism success by C. plutellae were not as a result of plant nutritional status or other attributes affecting the immune function of P. xylostella , nor to host-plant effects on superparasitism.
7. In these experiments, P. xylostella was a fully permissive host to C. plutellae and host-plant-mediated effects on the innate immune response appeared to play no part in parasitoid survival within hosts.  相似文献

Botanical preparations, usually from non-host plants, can be used to manipulate the behaviour of insect pests and their natural enemies. In this study, the effects of extracts of Chrysanthemum morifolium, a non-host plant of the diamondback moth, Plutella xylostella (Linnaeus), on the olfactory and oviposition responses of this phytophagous insect and on levels of parasitism by its specialist parasitoid Cotesia plutellae (Kurdjumov) were examined, using Chinese cabbage Brassica campestris L. ssp. pekinensis as the test host plant. Olfactometer tests showed that volatiles of chrysanthemum extract-treated host plants were less attractive to P. xylostella females than those from untreated host plants; and in contrast, volatiles of the chrysanthemum extract-treated host plants were more attractive to females of its parasitoid C. plutellae than those from untreated host plants. Oviposition preference tests showed that P. xylostella females laid only a small proportion of their eggs on chrysanthemum extract-treated host plants, while ovipositing parasitoid females parasitized a much higher proportion of host larvae feeding on the treated host plants than on untreated host plants. These results suggest that certain non-host plant compounds, when applied onto a host plant, may render the plant less attractive to a phytophagous insect but more attractive to its parasitoids. Application of such non-host plant compounds can be explored to develop push-pull systems to reduce oviposition by a pest insect and at the same time enhance parasitism by its parasitoids in crops.  相似文献

Endoparasitic wasps are able to develop inside permissive host insects due to their ability to overcome or evade the host's immune system. In the present study, we provide experimental evidence that ovarian calyx fluid of the ichneumonid endoparasitoid Venturia canescens has the potential to alter host haemocyte spreading and inhibit host haemolymph melanisation due to the presence of a putative serine protease inhibitor (serpin) activity. The existance of a serpin-like activity in the calyx fluid is also supported by experiments where the synthetic protease inhibitor p-APMSF had effects on cellular and cell-free immune reactions similar to ovarian calyx fluid. In addition, based on proteolytic digestion patterns of a wasp egg surface protein, we predict an Arg-specific trypsin-like protease activity in the host haemolymph which is possibly affected by calyx fluid components as well. Our data suggest that ovarian calyx fluid, deposited into the host together with the parasitoid egg, contains serpin activity which might transiently inactivate host defence reactions until other means of protection are established on the egg surface.  相似文献

The developing embryo of the braconid, Microplitis croceipes (Braconidae : Hymenoptera), is encased in an extraembryonic serosal membrane. Hatching of the parasitoid within the larva of its habitual host, Hehothis virescens (Noctuidae : Lepidoptera), is initiated about 40 hr after oviposition when held at 25 ± 2°C. At this time, the monolayered serosal membrane begins to dissociate into individual cells (teratocytes). After dissociation, teratocytes become dispersed in the hemolymph of the host. The average number of teratocytes released from each parasitoid embryo is 914 ± 43. Teratocytes average 14.1 ± 2.4 μm in diameter when first released, and reach a maximum average diameter of 68.1 ± 4.6 μm 6 days after liberation. Newly released teratocytes have ovoid nuclei, simple mitochondria and a limited number of profiles of the endoplasmic reticulum, all of which indicate relative metabolic inactivity. The ramified nuclei, extensive endoplasmic reticulum, polymorphic mitochondria and accumulation of glycogen granules and lipid droplets observed in older teratocytes provide circumstantial evidence that protein synthesis is occurring. Within hours after dissociation, microvilli begin to cover the surface of the teratocytes. Anatomical deformation (blebs) that occurred on some older (8-day-old) teratocytes probably resulted from enlargement or expansion of microvilli.  相似文献

茉莉酸是植物体内重要的伤信号分子,向植物施用外源茉莉酸后, 可诱导植物产生各种防卫反应, 如挥发物组成发生改变等, 进而影响植食性昆虫及其天敌。该文报道用不同浓度外源茉莉酸处理白菜和甘蓝后,诱导植物反应所产生的挥发物对菜蛾绒茧蜂搜索及寄生选择行为的影响。外源茉莉酸处理白菜和甘蓝后,处理植株的挥发物对菜蛾绒茧蜂的引诱力增强;与在对照植株上相比,该蜂对经茉莉酸处理后白菜植株上的小菜蛾幼虫的寄生数显著要高。表明茉莉酸处理白菜及甘蓝后,植物诱导反应导致其挥发物的作用发生变化,进而可提高该蜂的搜索和寄生效率。  相似文献

The calyx fluid in the lateral oviduct of a gregarious parasitoid, Apanteles glomeratus contained ellipsoid particles of ca. 130 × 200 nm. These calyx fluid particles did not appear to be embedded in a fibrous outer layer on the surface of eggs in the lateral oviduct. They were not observed on the surfaces of the eggs 3 to 4 hr after being deposited into the host haemocoele. Oviposition experiments indicated that the occurrence of haemocytic defence reactions of the late 2nd instar larvae of the Pieris rapae crucivora against 1 st instar larvae of the parasitoid increased with a decreasing number of the parasitoid eggs introduced into a host, and that more than 5 to 9 parasitoid eggs were needed for suppressing the ability of the host to encapsulate its parasitoid larvae immediately after hatching. When eggs with calyx fluid obtained from egg reservoir were injected into the host, they were found to be encapsulated 1 to 2 days after the injection. They could not start their embryonic development. When calyx fluid-free 3-hr-old eggs were injected in a number of more than 5 eggs into a 5th instar larva of Pieris, 58% of 31 eggs injected had normally hatched without evoking encapsulation reactions by the host. Both electron microscopic observations of parasitoid eggs in the host haemocoele and the experimental results suggested that calyx fluid or calyx fluid particles of the parasitoid might not be involved in the encapsulation-inhibiting activity of the parasitoid eggs. Rather it was anticipated that a substance (or substances) might be secreted by the parasitoid eggs into the haemocoele of the host, which suppressed defence reactions of the host.  相似文献

Teratocytes originate from the dissociation of the extraembryonic serosal membrane in some Braconidae and Scelionidae. Methods used to culture teratocytes in vitro are described and the yield of teratocyte secreted proteins (TSP) was measured. Although 90% are viable after 6 days, in vitro teratocytes reached only half the diameter (32&mgr;m) of the same age teratocytes obtained in vivo. Teratocytes cultured in vitro secrete as much as 0.7&mgr;g of protein per day per larval equivalent ( approximately 900 cells). Presence of parasitoid larvae enhanced teratocyte viability while periodic exchange of medium did not. However, medium exchange significantly increased the total amount of protein secreted. Size and viability were improved with the addition of 10% FBS to the Ex-cell 400 culture medium. Non-denaturing PAGE showed at least 15 proteins with molecular sizes estimated to be between 24 to 347kDa in medium containing teratocytes. An in vitro fat body assay was developed to measure the effect of TSP on protein synthesis and juvenile hormone esterase (JHE) activity. Crude TSP inhibited in vitro incorporation of [(35)S]-methionine into protein synthesized by the fat body. The amount of JHE released from in vitro fat body treated with crude TSP was significantly less than controls, most likely caused by the inhibition of general protein synthesis. The active fraction of TSP passed through a 30kDa molecular weight cutoff filter but was retained by a 3kDa filter. SDS-PAGE revealed four proteins with molecular weights between 8 and 20kDa not present in control medium incubated without teratocytes.  相似文献

Teratocytes, cells which originate from the serosal membrane of some Braconidae and Scelionidae, can be found in the hemocoel of permissive hosts during part or all of the developmental time of the parasitoid larva. Teratocytes from Microplitis croceipes are known to secrete biologically active proteins, which contribute to developmental arrest and failure to pupate of Heliothis virescens larvae. One such protein, which has a molecular weight of approximately 14 kDa is called TSP14. The presence of parasitoid larvae is essential to maintain teratocytes under in vitro conditions with protein-free EX-CELL 400. The teratocyte viability was maintained in vitro for at least 12 days in the presence of larvae when medium was exchanged every three days. Western blots show that TSP14 was secreted during the entire period of exchanges. In the absence of parasitoid larvae, teratocyte viability was only 30% by day 6 and no TSP14 had been secreted. In the absence of parasitoid larvae, teratocytes maintained in vitro in EX-CELL 400 medium supplemented with 10% FBS remained viable for at least nine days and secreted TSP14 for at least six days. This suggests that parasitoid larval secretions are sufficient but not uniquely essential to maintain teratocyte viability. Parasitoid larvae maintained in the absence of teratocytes did not secrete TSP14 and their secretory products did not inhibit pupation of H. virescens larvae.  相似文献
