We used DNA microsatellites to study colony kin structure and breeding patterns in the primitively eusocial wasp Polistes biglumis. P. biglumis inhabits cool areas at high altitudes and, as a consequence, has a reduced colony cycle compared to more temperate Polistes. P. biglumis colonies are always founded and controlled by a single foundress, but nest failure is common and foundresses losing their
nests do not have time to start new ones due to the short season. Instead, nests are characterized by frequent female turnover,
in the form of females taking over (usurpation) other con-specific nests. Our results showed that most nests had offspring
from multiple unrelated females, including some where multiple females were not observed in monitoring. Reconstruction of
behavioural events from the genetic data revealed three types of multiple matriline nests: (a) nests that were usurped by
another female, where the original nest owner disappeared following the usurpation event, (b) nests that were joined by another
female, where the original nest owner stayed following the joining event, (c) nests that were both usurped and joined by other
females. We also found, for the first time, a clear indication of multiple mating by Polistes females. Moreover, males mating with the same female were related, which may be explained by the lek mating system of P. biglumis. Finally, we analysed the nest sex ratios and how it changed during the season and found that sexes were produced sequentially,

Summary Polistes paper wasp species vary in their nest site selection, with some nesting in relatively exposed areas and others in protected cavities. Locating the nest sites of cavity dwelling species in natural habitat can prove difficult, and most behavioral studies on these species are therefore conducted using human-built structures. Since Hungerford and Williams (1912) and Rau (1929) noted the location of several P. fuscatus (Fabricius) nests in rodent burrows, there have been no published accounts of ground nesting Polistes species. Identifying the sex of larvae is important in social Hymenoptera. Until now for Polistes wasps it has been necessary to genotype larvae at microsatellite loci, and assign their sex based on homozygosity at these
loci. In our study on the paper wasp Polistes dominulus we have found morphological differences between larval sexes that can be used for larvae from the 3rd instar on to easily and cheaply identify larval sex: the external gonopore and the shape and size of larval gonads. The robustness
of these indicators was supported by genotype data at four microsatellite loci. Using gonopore and gonad features for sex
assignment will assign diploid males as males, unlike techniques based on genetic loci or chromosomes.

Summary Interspecific facultative social parasitism is well known in ants and in bumble-bees, but it is rarer in wasps. This form of parasitism is traditionally considered to be an intermediate stage in the evolution of obligate interspecific parasitism, where the parasites are no longer able to nest alone. We report field and experimental observations of a newly discovered facultative parasitic relationship between two closely related free-living Polistes species: P. nimphus and P. dominulus. P. nimphus foundresses sometimes usurp the nests of the larger P. dominulus before worker emergence. The invading queen takes over the nest with abundant abdomen stroking on the nest surface and is accepted by workers if they emerge 6 or more days after usurpation. Morphometric comparisons show that the usurper species, though smaller than its victims, has morphological adaptations consisting of larger heads, mandibles and front femora relative to their body size that may give it an advantage during nest invasion. This strategy is likely to be taken only after the foundress loses her original nest because invading P. nimphus queens have lower reproductive success than they would have had on their own nest. Overall, we found that P. nimphus usurpers use strategies of invasion similar to those of two obligate parasites, suggesting that this may be an example of one of the pathways by which social parasitism evolved.Received 4 April 2003; revised 8 August 2003; accepted 14 September 2003. 相似文献

In simple societies, activity of the workers can be regulated in a centralized way by a sole individual. Polistes wasps are simple in some respects because they have small, open colonies, with a single queen who is not morphologically differentiated from the workers. Worker activity in Polistes is episodic. Pause periods are followed by periods of intense activity, when most individuals are in motion. One way to investigate the regulation of worker activity is to see if activity periods are initiated by one specific individual. The queen was found regulating the resumption of activity in P. fuscatus in a centralized way, but recently, a decentralized regulation was found in P. instabilis and P. dominula. Thus, what is the pattern of regulation of worker activity in Polistes? To solve this question, broader studies of other species are desirable. Therefore, we investigated whether activity of P. versicolor workers are regulated by one individual. We tested for two mechanisms: triggering periods of colonial activity or actually physically stimulating workers to forage. The results demonstrated that no single individual was responsible to start periods of activity or physically stimulate worker’s departure. P. versicolor appears to be an example of a society with an intermediary organization of work, where the queen controls reproduction but does not organize work. 相似文献

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes. 相似文献

The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate
larvae Galleria

Social insects are excellent invaders that have had negative impacts on native species and humans. Many invasive species move
from warmer to cooler climates. For these species, thermal adaptations may both be important for their ability to invade and
to limit their invasion range. The invasion of Polistes dominulus into North America provides an example of a primitively eusocial invader from a warmer climate. We studied the differences
in thermoregulation between P. dominulus and the native P. fuscatus. We found that, during flight, thorax temperature in P. fuscatus was less affected by ambient temperature than thorax temperature of P. dominulus. We also found that P. dominulus and P. fuscatus showed different patterns of warming after removal from a cold environment. Unlike P. dominulus, live P. fuscatus never fully cooled down in a cold environment. P. fuscatus also reached their relative minimum flight temperatures earlier than P. dominulus, but P. dominulus maintained higher elevated temperatures for longer. These differences in thermoregulatory ability suggest that the lower
winter survival of P. dominulus could be offset by a greater thermal tolerance during flight, while the lower thermal tolerance of P. fuscatus in flight is offset somewhat by better thermoregulatory ability. 相似文献

A genetic transformation system has been developed for callus cells of Crataegus
aronia using Agrobacterium
tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with
5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different
types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red
colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this

mercedesae is a serious ectoparasite of Apis
mellifera in China. The aim of this study was to investigate the infestation rates and intensity of T. mercedesae in A. mellifera in China, and to explore the relative importance of climate, district, management practices and beekeeper characteristics
that are assumed to be associated with the intensity of T. mercedesae. Of the 410 participating apiaries, 379 apiaries were included in analyses of seasonal infestation rates and 352 apiaries
were included in multivariable regression analysis. The highest infestation rate (86.3%) of T. mercedesae was encountered in autumn, followed by summer (66.5%), spring (17.2%) and winter (14.8%). In autumn, 28.9% (93) of the infested
apiaries were in the north (including the northeast and northwest of China), 71.1% (229) were in the central and south (including
east, southeast and southwest China), and 306 apiaries (82.9%) were co-infested by both T. mercedesae and Varroa. Multivariable regression analysis showed that geographical location, season, royal jelly collection and Varroa infestation were the factors that influence the intensity of T. mercedesae. The influence of beekeeper’s education, time of beekeeping, operation size, and hive migration on the intensity of T. mercedesa was not statistically significant. This study provided information about the establishment of the linkage of the environment

We investigated colony foundation behavior of three species of the Lasius claviger group, L. latipes, L. interjectus and L. claviger, using field observations and laboratory experiments. Laboratory studies included observing gyne behavior prior to interaction
with host colonies (e.g. grouping, overwintering, feeding) and experiments in which gynes were introduced to putative host
colonies. Observations of gyne parasitic behavior immediately after mating flights in the field and the discovery of mixed
colonies both confirm the parasitic behavior of these species and point toward a wide host species range including two cases
of hyperparasitism. L. interjectus and L. latipes enter the host colony immediately after the nuptial flight, while L. claviger may hibernate and enter in the spring. Laboratory experiments showed that L. interjectus gynes prefer to group and suffer lower mortality when in a group, which is consistent with field observations of L. interjectus gynes entering host colonies in large numbers. The maT clade of transposons is a group of transposable elements intermediate in sequence and predicted protein structure to mariner and Tc transposons, with a distribution thus far limited to a few invertebrate species. We present evidence, based on searches of publicly available databases, that the nematode Caenorhabditis briggsae has several maT-like transposons, which we have designated as CbmaT elements, dispersed throughout its genome. We also describe two additional transposon sequences that probably share their evolutionary history with the CbmaT transposons. One resembles a fold back variant of a CbmaT element, with long (380-bp) inverted terminal repeats (ITRs) that show a high degree (71%) of identity to CbmaT1. The other, which shares only the 26-bp ITR sequences with one of the CbmaT variants, is present in eight nearly identical copies, but does not have a transposase gene and may therefore be cross mobilised by a CbmaT transposase. Using PCR-based mobility assays, we show that CbmaT1 transposons are capable of excising from the C. briggsae genome. CbmaT1 excised approximately 500 times less frequently than Tcb1 in the reference strain AF16, but both CbmaT1 and Tcb1 excised at extremely high frequencies in the HK105 strain. The HK105 strain also exhibited a high frequency of spontaneous induction of unc-22 mutants, suggesting that it may be a mutator strain of C. briggsae. 相似文献

Summary The biology of the invasive Polistes dominulus and the native P. fuscatus was compared at a field site in Rochester, Michigan over a two-year period. Colonies nesting semi-naturally in plywood nestboxes were studied using videography, extensive surveys, and colony-specific marking of gynes.Both single- and multiple-foundress colonies of P. dominulus were significantly more productive than comparable colonies of P. fuscatus. The disparity in productivity was significantly more pronounced in single-foundress colonies than in multiple-foundress colonies. P. dominulus had significantly shorter larval and pupal development times than P. fuscatus, which allowed P. dominulus to produce its first workers about a week earlier than P. fuscatus. P. dominulus had a number of additional advantages over P. fuscatus that contributed to its productivity including (1) significantly less parasitism by Strepsiptera, (2) significantly greater probability of renesting after predation by raccoons, (3) significantly lower usurpation pressures, and (4) possibly longer foraging days . The recovery of colony-marked foundresses indicated that gynes of P. dominulus suffered significantly greater mortality than gynes of P. fuscatus during winter diapause and that foundresses of both species were equally, strongly philopatric.P. dominulus is likely replacing P. fuscatus in many areas of southeastern Michigan via indirect or exploitative competition. A pea rust fungus, Uromyces viciae-fabae, has been classified into two varieties, var. viciae-fabae and var. orobi, based on differences in urediniospore wall thickness and putative host specificity in Japan. In principal component analyses, morphological features of urediniospores and teliospores of 94 rust specimens from Vicia, Lathyrus, and Pisum did not show definite host-specific morphological groups. In molecular analyses, 23 Uromyces specimens from Vicia, Lathyrus, and Pisum formed a single genetic clade based on D1/D2 and ITS regions. Four isolates of U. viciae-fabae from V. cracca and V. unijuga could infect and sporulate on P. sativum.
