N-Acetylglutamate synthase, an early enzyme of the arginine pathway, provides acetylglutamate for ornithine synthesis in the so-called "acetylglutamate cycle." Because acetylglutamate is regenerated as ornithine is formed, the enzyme has only a catalytic or anaplerotic role in the pathway, maintaining "bound" acetyl groups during growth. We have detected this enzyme in crude extracts of Neurospora crassa and have localized it to the mitochondria along with other ornithine biosynthetic enzymes. The enzyme is bound to the mitochondrial membrane. The enzyme has a pH optimum of 9.0 and Km values for glutamate and CoASAc of 6.3 and 1.6 mM, respectively. It is feedback-inhibited by L-arginine (I0.5 = 0.16 mM), and its specific activity is augmented 2-3-fold by arginine starvation of the mycelium. Mutants of the newly recognized arg-14 locus lack activity for the enzyme. Because these mutants are complete auxotrophs, we conclude that N-acetylglutamate synthase is an indispensible enzyme of arginine biosynthesis in N. crassa. This work completes the assignment of enzymes of the arginine pathway of N. crassa to corresponding genetic loci. The membrane localization of the enzyme suggests a novel mechanism by which feedback inhibition might occur across a semipermeable membrane.  相似文献

The arg-12(s) mutation of Neurospora causes a partial block in the ornithine transcarbamylase (OTC) reaction. Strains carrying this mutation will use endogenous ornithine, but not exogenous ornithine, as a precursor of arginine. Certain strains carrying arg-12(s) may be used for direct selection of variants able to use exogenous ornithine as an arginine precursor. Among eight such derivatives, six lacked the catabolic enzyme ornithine transaminase (OTA). All six mutations were alleles of a single gene, designated ota, on linkage group III. No mutation affected arginase, the first enzyme in the catabolic pathway with OTA. Strains carrying ota mutations alone are vigorous prototrophs, but, unlike wild-type Neurospora, fail to use ornithine efficiently as a sole nitrogen source. The selection method by which ota mutants arose suggests that OTA is severely competitive with OTC for exogeneous ornithine.  相似文献

Sixty-five independent, N2 fixation-defective (Nif-) vector insertion (Vi) mutants were selected, cloned, and mapped to the ORS571 genome. The recombinant Nif::Vi plasmids obtained in this way were used as DNA hybridization probes to isolate homologous phages from a genomic library of ORS571 constructed in lambda EMBL3. Genomic maps were drawn for three ORS571 Nif gene loci. Forty-five Nif::Vi mutants in genomic Nif locus 1 defined two gene clusters separated by 8 kilobase pairs (kb) of DNA. In the first cluster, 36 Nif::Vi mutants mapped to a 7-kb DNA segment that showed DNA homology with Klebsiella pneumoniae nifHDKE and encoded at least two Nif operons. In the other cluster, nine Nif::Vi mutants mapped to a 1.5-kb DNA segment that showed homology with K. pneumoniae and Rhizobium meliloti nifA; this DNA segment encoded a separate Nif operon. Fifteen Nif::Vi mutants mapped to a 3.5-kb DNA segment defined as Nif locus 2 and showed DNA homology with the R. meliloti P2 fixABC operon. Nif locus 2 carries a second nifH (nifH2) gene. Four Nif::Vi mutants mapped to a 2-kb DNA segment defined as Nif locus 3 and showed DNA homology with K. pneumoniae nifB. DNA from lambda Nif phages comprising all three genomic Nif loci was subcloned in plasmid vectors able to stably replicate in ORS571. These plasmid subclones were introduced into ORS571 strains carrying physically mapped Nif::Vi insertions, and genetic complementations were conducted. With the exception of certain mutants mapping to the nifDK genes, all mutants could be complemented to Nif+ when they carried plasmid subclones of defined genomic DNA regions. Conversely, most nifDK mutants behaved as pseudodominant alleles.  相似文献

Six schemes were used to identify 80 independent recessive lethal deficiencies of linkage group (LG) II following X-ray treatment of the nematode Caenorhabditis elegans. Complementation tests between the deficiencies and ethyl methanesulfonate-induced recessive visible, lethal and sterile mutations and between different deficiencies were used to characterize the extents of the deficiencies. Deficiency endpoints thus helped to order 36 sites within a region representing about half of the loci on LG II and extending over about 5 map units. New mutations occurring in this region can be assigned to particular segments of the map by complementation tests against a small number of deficiencies; this facilitates the assignment of single-site mutations to particular genes, as we illustrate. Five sperm-defective and five oocyte-defective LG II sterile mutants were identified and mapped. Certain deficiency-by-deficiency complementation tests allowed us to suggest that the phenotypes of null mutations at two loci represented by visible alleles are wild type and that null mutations at a third locus confer a visible phenotype. A segment of LG II that is about 12 map units long and largely devoid of identified loci seems to be greatly favored for crossing over.  相似文献

arg-13为精氨酸代谢途径里的一个渗露型突变。经研究发展了该突变的严格选择方法。该法省略了基本培养基的氮源而加上相似浓度的鸟氨酸与赖氨酸。此法在严紧山梨糖/葡萄糖条件下能强烈抑制arg-13突变株生长,但在斑点试验条件下允许arg-13突变株生长。由于鸟氨酸是通过线粒体合成和由细胞质至线粒体的过膜转运而积累,我们构建了arg-4,arg-13双突变株,其中arg-4阻断了线粒体鸟氨酸合成。在斑点试验条件下,arg-4,arg-13双突变株能利用鸟氨酸作为唯一氮源与精氨酸合成前体,但受赖氨酸与刀豆氨酸强烈抑制。具正常鸟氨酸转运功能的arg-4单突变株在鸟氨酸基本培养基的生长只受微弱的赖氨酸抑制。已有报道arg-13为嘧啶合成代谢途径里pyr-3(CPSACT~ )突变的部分抑制基因,序列分析表明arg-13编码一线粒体转运酶。本文数据提示arg-13在线粒体鸟氨酸过膜转运过程中起主要作用。arg-13突变株仍携带一定的线粒体鸟氨酸转运功能并受碱性氨基酸赖氨酸、刀豆氨酸抑制,可能为另一线粒体碱性氨基酸转运酶介导。  相似文献

arg-13 is a leaky mutation involved in arginine metabolism. A tight selection is developed using similar amount of lysine and ornithine replacing other nitrogen source in minimal medium. This selection strongly inhibits the growth of arg-13 under stringent sorbose/glucose condition but allows arg-13 to grow under spot test conditions. As ornithine is build up through mitochondrial ornithine biosynthesis and transport from cytoplasm to mitochondria, arg-13 is combined in genetic crosses with arg-4 which blocks mitochondrial ornithine synthesis. Under spot test conditions, double mutant arg-4, arg-13 is able to use ornithine as sole nitrogen source and arginine biosynthesis precursor, but subject to strong lysine and canavanine inhibition. While the usage of ornithine in arg-4 single mutant with intact ornithine transport function is only slightly inhibited by lysine. All available data suggest arg-13 plays a major role in mitochondrial ornithine transport. The strain carrying the mutation at the arg-13 locus allows inefficient mitochondrial ornithine trafficking, possibly mediated by another distinct basic amino acid carrier.  相似文献

Summary The arg-7 locus is the structural gene for the argininosuccinate lyase (ASL). Interallelic complementation was previously found to occur between several mutants of the locus: this is indicative for the homomultimeric nature of ASL.Two complementing (arg-7-5 and arg-7-7) and two non-complementing (arg-7-1 and arg-7-6) mutants of the arg-7 locus were crossed to the pab-2 strain (which is wild-type for the arg-7 locus). In each cross, heterozygote phenotypically wild-type strains were isolated; their diploid pattern was demonstrated by various criteria: mating type, cell volume, nuclear size.The four heterozygotes were compared to the haploid wild-type and in some experiments, to the diploid strain arg-1xpab-2 homozygous for the arg-7 locus. No difference was found in growth rate and in the Michaelis constant values for ASL. The specific activity of the enzyme produced in the heterozygotes was about 50 percent of the activity found in haploid or diploid wild-type. The heat sensitivity of ASL was also investigated in the different strains: two (containing the complementing mutations arg-7-5 and arg-7-7) of the four heterozygotes produce ASL varieties different from the wild-type enzyme as far as the thermolability is concerned.These results suggest that hybrid ASL can be formed by interaction between the products of wild-type and mutant genes. A clear dominance of the wild-type allele is expected only when the mutant allele has no product of the gene: this could be the case for arg-7-1 and arg-7-6.  相似文献

Candidate genes for human type II gonadotropin-releasing hormone receptor (GnRH-RII) reside on two separate loci, 1q12-q21 and 14q21-23, yet neither locus generates functional GnRH-RII. Instead, their opposite DNA strands encode functional RNA-binding motif protein 8 (RBM8s), which is also encoded by another locus, 5q13-q14. To elucidate the mechanism through which such multiple human GnRH-RII/RBM8 loci arose, here we have defined an RBM8 locus in a comparative model species, the medaka Oryzias latipes. The medaka RBM8, which exists as a single copy gene, is linked to, but does not overlap with, GnRH-R2 on linkage group (LG) 16, demonstrating the ancient origin of the physical linkage between GnRH-R and RBM8. The medaka LG 16 contains orthologous segments to the human chromosome 1 and therefore the 1q12-q21 locus would be an originating human GnRH-RII/RBM8 segment. Furthermore, like the human RBM8s on 1q12-q21 and 5q13-q14 but not that on 14q21-q23, the medaka RBM8 is a multiexon gene, indicating that the 14q21-q23 and 5q13-q14 loci were generated by retrotransposition and segmental genomic duplication, respectively, of the originating 1q12-q21 locus.  相似文献

New mutations of the polyamine pathway of Neurospora crassa fell into three categories. The majority affected ornithine decarboxylase and lay at the previously defined spe-1 locus. One mutation, JP100, defining the new spe-2 locus, eliminated S-adenosyl-methionine decarboxylase and led to putrescine accumulation. Revertants of this mutation suggested that the locus encodes the enzyme. Two other mutations, LV105 and JP120, defined a third locus, spe-3. Strains with these mutations also accumulated putrescine and were presumed to lack spermidine synthase activity, which catalyzes the formation of spermidine from putrescine and decarboxylated S-adenosylmethionine. The three spe loci lay within about 20 map units of one another on the right arm of Linkage Group V in the order: centromere-spe-2-spe-1-spe-3. The requirement for spermidine for growth was much less in spe-2 and spe-3 mutants than in spe-1 mutants, which do not accumulate putrescine. This suggested that putrescine fulfills many, but not all, of the functions of spermidine, or that high levels of putrescine render spermidine more effective in its essential roles.  相似文献

Optic morphology (Om) mutants associated with insertions of the tom transposable element at each of three tested loci are neomorphs as defined by the phenotypic equivalence of +/+/Om with +/Om and of +/Om/Om with Om/Om. Mutants behaving as suppressors of Om mutants and mapping to at least six loci are recovered from the same source and in similar frequency as Om mutants. The semidominant and nonpleiotropic suppressors at four of the six loci display defective eye phenes themselves, and the phenotypically normal mutants at a fifth locus are suspected alleles of a gene represented by recessive furrowed eye mutants. These and other properties imply that the suppressors, like suppressible Om mutants, are neomorphic due to insertion of the tom element into a hypothetical sequence they share with other members of a set of genes involved in development of the eye. Concurrently premature expression of both the suppressor and suppressed mutants would allow interaction of their products just as in normal development.  相似文献

Scab, caused by the fungal pathogen Venturia inaequalis, is the most common disease of cultivated apple (Malus xdomestica). The fungal races 6 and 7 have now overcome the major resistance gene Vf, which is widely used in apple breeding programmes. New breeding strategies to achieve durable resistance are thus necessary. The aim of this study was to determine the genetic basis of quantitative resistance of the apple cultivar 'Dülmener Rosenapfel', known to be scab resistant under different environmental conditions. An F1 progeny derived from the cross between the susceptible cultivar 'Gala' and 'Dülmener Rosenapfel' was tested in a greenhouse with a multi-isolate inoculum of V. inaequalis. Rvi14, a new major gene that conditions a chlorotic-type reaction, was mapped on linkage group (LG) 6 in a genomic region not known to be involved in disease resistance. A further three quantitative trait loci (QTL) for resistance were identified. One co-localized with Rvi14 on LG6, whereas the remaining two were detected on LG11 and LG17, in genomic regions already reported to carry broad-spectrum QTL in other genetic backgrounds. Since a selective genotyping approach was used to detect QTL, an expectation-maximization (EM) computation was used to estimate the corrected QTL contributions to phenotypic variation and was validated by entire progeny genotyping.  相似文献

In Pseudomonas aeruginosa arginine can be degraded by the arginine "dihydrolase" system, consisting of arginine deiminase, catabolic ornithine carbamoyltransferase, and carbamate kinase. Mutants of P. aeruginosa strain PAO affected in the structural gene (arcB) of the catabolic ornithine carbamoyltransferase were isolated. Firt, and argF mutation (i.e., a block in the anabolic ornithine carbamoyltransferase) was suppressed specifically by a mutationally altered catabolic ornithine carbamoyltransferase capable of functioning in the anabolic direction. The suppressor locus arcB (Su) was mapped by transduction between hisII and argA. Second, mutants having lost suppressor activity were obtained. The Su- mutations were very closely linked to arcB (Su) and caused strongly reduced ornithine carbamoyltransferase activities in vitro. Under aerobic conditions, a mutant (PA0630) which had less than 1% of the wild-type catabolic ornithine carbamoyltransferase activity grew on arginine as the only carbon and nitrogen source, at the wild-type growth rate. When oxygen was limiting, strain PA0630 grown on arginine excreted citrulline in the stationary growth phase. These observations suggest that during aerobic growth arginine is not degraded exclusively via the dihydrolase pathway.  相似文献

J. Bacteriol. 92:35-42. 1966.-Fluorophenylalanine-resistant mutants (fpa-r) of Pseudomonas aeruginosa have been isolated. By cotransduction analysis, the mutations were shown to have at least two chromosomal locations. One locus (fpaA) showed linkage to three other markers, str, try-3bi, and arg-3, and the order of these four linked markers was found to be try-3bi, arg-3, fpaA, str. The linkage relationships of the other fpa loci are not yet known. The phenotypic expression of resistance at the fpaA locus can be suppressed by mutation of the str locus from str-s to str-r, whereas that at an unlinked fpa locus cannot.  相似文献

Three lysine auxotrophs, strains AU363, 7305d, and 8201-7A, were investigated genetically and biochemically to determine their gene loci, biochemical lesions, and roles in the lysine biosynthesis of Saccharomyces cerevisiae. These mutants were leaky and blocked after the alpha-aminoadipate step. Complementation studies placed these three mutations into a single, new complementation group, lys14. Tetrad analysis from appropriate crosses provided evidence that the lys14 locus represented a single nuclear gene and that lys14 mutants were genetically distinct from the other mutants (lys1, lys2, lys5, and lys9) blocked after the alpha-aminoadipate step. The lys14 strains, like lys9 mutants, accumulated alpha-aminoadipate-semialdehyde and lacked significant amounts of saccharopine reductase activity. On the bases of these results, it was concluded, therefore, that LYS9 and LYS14, two distinct genes, were required for the biosynthesis of saccharopine reductase in wild-type S. cerevisiae.  相似文献

A human ornithine aminotransferase (OAT) locus has been mapped to the Xp11.2, as has the Norrie disease locus. We used a cDNA probe to investigate a 3-generation UCLA family with Norrie disease; a 4.2-kb RFLP was detected and a maximum lod score of 0.602 at zero recombination fraction was calculated. We used the same probe to study a second multigeneration family with Norrie disease from Utah. A different RFLP of 7.5 kb in size was identified and a recombinational event between the OAT locus represented by this RFLP and the disease loci was observed. Linkage analysis of these two loci in this family revealed a maximum load score of 1.88 at a recombination fraction of 0.10. Although both families have affected members with the same disease, the lod scores are reported separately because the 4.2- and 7.5-kb RFLPs may represent two different loci for the X-linked OAT.  相似文献

We describe the identification of two mutations in bacterial genes, designated as fipB and fipC, which resulted in temperature-sensitive morphogenesis of bacteriophage f1. These mutations mapped at separate loci but had to be present simultaneously to block f1 production at 41.5 degrees C. One mutation defined the locus fipB at 85.3 min on the Escherichia coli linkage map; the other defined the locus fipC, which mapped very close to rpsL at 73 min. Since these mutations did not appear to affect phage DNA replication, gene expression, or protein localization, they probably interfered with the its life cycle at the level of assembly. fipB mutants were partially deficient in adsorption of bacteriophage lambda, and fipB and fipC mutants leaked beta-lactamase into the medium, suggesting that the mutations affect outer-membrane structure or function.  相似文献

A new series of pyrimidine-requiring mutants of Neurospora has been isolated and all enzymes involved in pyrimidine biosynthesis are represented by at least one mutant. Among these mutants is included a single isolate for a new locus, pyr-6. This mutant is deficient in dihydroorotase (DHOase) and represents the only enzymatic step in orotate synthesis for which no mutant previously had been found. This mutant, which mapped genetically on the right arm of linkage group V, is unlinked to any of the other pyrimidine mutants. The DHOase-deficient mutant is also characterized by an unexpected growth behavior. The pyr-1 locus has been specifically associated with a lack of dihydroorotate dehydrogenase (DHOdehase). Mutants isolated in this series for other pyrimidine loci have been related to previously isolated mutants by allelism, recombination, and accumulation studies.  相似文献
