Low molecular (7.2 kD) cold shock protein D (CspD) from Bacillus thuringiensis was isolated in our laboratory in the late 1990s It was shown that CspD induced nonspecific resistance in plants to viral and fungal phytopathogens. It was suggested to explore the opportunity to express the gene of this elicitor in tobacco plant and evaluate the resistance ofthe transgenic plants to various pathogens. Several transgenic tobacco lines were obtained. Enhanced resistance of the transgenic plants was confirmed both to tobacco mosaic virus and to fungus Alternaria longipes.  相似文献

Polygalacturonases (PGs), enzymes that hydrolyze the homogalacturonan of the plant cell wall, are virulence factors of several phytopathogenic fungi and bacteria. On the other hand, PGs may activate defense responses by releasing oligogalacturonides (OGs) perceived by the plant cell as host-associated molecular patterns. Tobacco (Nicotiana tabacum) and Arabidopsis (Arabidopsis thaliana) plants expressing a fungal PG (PG plants) have a reduced content of homogalacturonan. Here, we show that PG plants are more resistant to microbial pathogens and have constitutively activated defense responses. Interestingly, either in tobacco PG or wild-type plants treated with OGs, resistance to fungal infection is suppressed by exogenous auxin, whereas sensitivity to auxin of PG plants is reduced in different bioassays. The altered plant defense responses and auxin sensitivity in PG plants may reflect an increased accumulation of OGs and subsequent antagonism of auxin action. Alternatively, it may be a consequence of perturbations of cellular physiology and elevated defense status as a result of altered cell wall architecture.  相似文献

Insect herbivore-induced plant volatile emission and the subsequent attraction of natural enemies is facilitated by fatty acid-amino acid conjugate (FAC) elicitors, such as volicitin [N-(17-hydroxylinolenoyl)-L-glutamine], present in caterpillar oral secretions. Insect-induced jasmonic acid (JA) and ethylene (E) are believed to mediate the magnitude of this variable response. In maize (Zea mays) seedlings, we examined the interaction of volicitin, JA, and E on the induction of volatile emission at different levels of nitrogen (N) availability that are known to influence E sensitivity. N availability and volicitin-induced sesquiterpene emission are inversely related as maximal responses were elicited in N-deficient plants. Plants with low N availability demonstrated similar volatile responses to volicitin (1 nmol plant(-1)) and JA (100 nmol plant(-1)). In contrast, plants with medium N availability released much lower amounts of volicitin-induced sesquiterpenes compared with JA, suggesting an alteration in volicitin-induced JA levels. As predicted, low N plants exhibited greater sustained increases in wound- and volicitin-induced JA levels compared with medium N plants. N availability also altered volicitin-E interactions. In low N plants, E synergized volicitin-induced sesquiterpene and indole emission 4- to 12-fold, with significant interactions first detected at 10 nL L(-1) E. Medium N plants demonstrated greatly reduced volicitin-E interactions. Volicitin-induced sesquiterpene emission was increased by E and was decreased by pretreatment the E perception inhibitor 1-methylcyclopropene without alteration in volicitin-induced JA levels. N availability influences plant responses to insect-derived elicitors through changes in E sensitivity and E-independent JA kinetics.  相似文献

Content of endogenous abscisic acid (ABA) increased in rice plants under salt stress. Pre- or post-treatment by jasmonic acid (JA) mostly further increased ABA content. In the presence of salt stress also content of gibberellins (GAs) mostly increased more after treatment by JA. The gene encoding a cowpea trypsin inhibitor (CpTI), which confers insect resistance in trangenic tobacco, was introduced into rice. Expression of the CpTi gene driven by the constitutively active promoter of the rice actin 1 gene (Act1) leads to high-level accumulation of the CpTI protein in transgenic rice plants. Protein extracts from transgenic rice plants exhibit a strong inhibitory activity against bovine trypsin, suggesting that the proteinase inhibitor produced in transgenic rice is functionally active. Small-scale field tests showed that the transgenic rice plants expressing the CpTi gene had significantly increased resistance to two species of rice stem borers, which are major rice insect pests. Our results suggest that the cowpea trypsin inhibitor may be useful for the control of rice insect pests.  相似文献

 The truncated chimeric Bt gene, cryIA(b) of Bacillus thuringiensis, driven by two constitutive promoters, 35S from CaMV and Actin-1 from rice, and two tissue-specific promoters, pith tissue and pepcarboxylase (PEPC) for green tissue from maize, was introduced into several varieties of rice (indica and japonica) by microprojectile bombardment and protoplast systems. A total of 1800 putative transgenic Bt rice plants could be produced. Southern analysis revealed that more than 100 independently transformed plants could be confirmed for integration of the cryIA(b) gene. High levels of CryIA(b) proteins were obtained in the green tissue (leaves and stem) of many plants using the PEPC promoter. There was little difference in Bt protein level in leaves and stems from transgenic plants with the 35 S or Actin-1 promoter. Out of 800 Southern-positive plants that were bioassayed, 81 transgenic plants showed 100% mortality of insect larvae of the yellow stem borer (Scirpophaga incertulas). The transgene, cryIA(b), driven by different promoters showed a wide range of expression (low to high) of Bt proteins stably inherited in a number of rice varieties with enhanced yellow stem borer resistance. This first report of transgenic indica Bt rice plants with the PEPC or pith promoter either alone or in combination should provide a better strategy for providing rice plants with protection against insect pest resistance, minimizing the expression of the CryIA(b) protein in seeds and other tissues. The phytohormone jasmonic acid (JA) plays a core role in plant defence against herbivores. When attacked by herbivores, JA and its bioactive derivatives are accumulated at the damage site, and subsequently perceived by the jasmonate co-receptors COI1 and JAZ proteins. The (+)-7-iso-jasmonoyl-L-isoleucine (JA-Ile) is known to be the main active JA derivative controlling vascular plant responses to herbivores as well as other JA-regulated processes. However, whether other endogenous JA-amino acid conjugates (JA-AAs) are involved in herbivore-induced defence responses remain unknown. Here, we investigated the role of herbivore-elicited JA-AAs in the crop plant rice. The levels of five JA-AAs were significantly increased under the armyworm, leaf folder and brown planthopper attack. Of the elicited JA derivatives, JA-Ile, JA-Val and JA-Leu could serve as ligands to promote the interaction between rice COI1 and JAZs, inducing OsJAZ4 degradation in vivo. JA-Val or JA-Leu treatment increased the expression of JA- and defence-related pathway genes but not JA-Ile levels, suggesting that these JA-AAs may directly function in JA signalling. Furthermore, the application of JA-Val or JA-Leu resulted in JA-mediated plant growth inhibition, while enhancing plant resistance to herbivore attack. This study uncovers that JA-Val and JA-Leu also play a role in rice defence against herbivores.  相似文献

The pattern recognition receptor AtRLP23 from Arabidopsis thaliana recognizes the epitopes (nlp24s) of necrosis and ethylene‐inducing peptide 1‐like proteins (NLPs) and triggers pattern‐triggered immunity (PTI). Here, we established methods for studying the early events of PTI in the hybrid poplar cultivar Shanxin (Populus davidiana × Populus bolleana) in response to the flagellin epitope. We confirmed that wild‐type Shanxin cannot generate PTI responses on nlp24 treatment. Four NLP homologues were characterized from two common fungal pathogens of Shanxin, namely Marssonina brunnea f. sp. monogermtubi (MbMo) and Elsinoë australis (Ea), which cause black leaf spot and anthracnose disease, respectively, and the nlp24s of three of them could be responded to by Nicotiana benthamiana leaves expressing AtRLP23. We then created AtRLP23 transgenic Shanxin lines and confirmed that the heterologous expression of AtRLP23 conferred on transgenic Shanxin the ability to respond to one nlp24 of each fungal pathogen. Consistently, infection assays with MbMo or Ea showed obviously lower levels of disease symptoms and significantly inhibited the growth of fungi on the transgenic poplar compared with that in wild‐type poplar. Overall, our results indicated that the heterologous expression of AtRLP23 allowed transgenic Shanxin to generate a PTI response to nlp24s, resulting in increased broad‐spectrum fungal disease resistance.  相似文献

Nitric oxide (NO) is known for its role in the activation of plant defense responses. To examine the involvement and mode of action of NO in plant defense responses, we introduced calmodulin-dependent mammalian neuronal nitric oxide synthase (nNOS), which controls the CaMV35S promoter, into wild-type and NahG tobacco plants. Constitutive expression of nNOS led to NO production and triggered spontaneous induction of leaf lesions. Transgenic plants accumulated high amounts of H2O2, with catalase activity lower than that in the wild type. nNOS transgenic plants contained high levels of salicylic acid (SA), and they induced an array of SA-, jasmonic acid (JA)-, and/or ethylene (ET)-related genes. Consequently, NahG co-expression blocked the induction of systemic acquired resistance (SAR)-associated genes in transgenic plants, implying SA is involved in NO-mediated induction of SAR genes. The transgenic plants exhibited enhanced resistance to a spectrum of pathogens, including bacteria, fungi, and viruses. Our results suggest a highly ranked regulatory role for NO in SA-, JA-, and/or ET-dependent pathways that lead to disease resistance.  相似文献

The antagonistic action between jasmonic acid (JA) and salicylic acid (SA) in plant defence responses has been well documented. However, their relationship in secondary metabolite production is largely unknown. Here, we report that PB90, a protein elicitor from Phytophthora boehmeriae , triggers JA generation, SA accumulation and flavonol glycoside production of Ginkgo biloba cells. JA inhibitors suppress not only PB90-triggered JA generation, but also the elicitor-induced flavonol glycoside production. However, the elicitor can still enhance flavonol glycoside production even though the JA generation is totally inhibited. Over-expression of SA hydrolase gene NahG not only abolishes SA accumulation, but also suppresses the elicitor-induced flavonol glycoside production when JA signalling is inhibited. Interestingly, expression of NahG does not inhibit the elicitor-induced flavonol glycoside accumulation in the absence of JA inhibitors. Moreover, JA levels are significantly enhanced when SA accumulation is impaired in the transgenic cells. Together, the data suggest that both JA and SA are involved in PB90-induced flavonol glycoside production. Furthermore, we demonstrate that JA signalling might be enhanced to substitute for SA to mediate the elicitor-induced flavonol glycoside accumulation when SA signalling is impaired, which reveals an unusual complementary relationship between JA and SA in mediating plant secondary metabolite production.  相似文献
