Summary Two methods were evaluated to determine the concentration of sulphite in sandy loam soils. Use of p-rosaniline-formaldehyde reagent and sodium tetrachloromercurate as extractant proved effective only if a wide soil to extractant ratio was employed. Use of Ellman's reagent in conjunction with this extractant was however, unsatisfactory. Soils exposed to heavy atmospheric pollution differed in sulphite concentration only marginally compared with relatively unpolluted soils. The effect of soil water content on the stability of sulphite in soil was also determined. 相似文献

Spatial variation in carbon isotope discrimination across the thalli of several lichen species 总被引:3,自引:0,他引:3
Variations in stable carbon isotope discrimination (δ) were investigated across the thalli of several lichen species possessing different photobiont associations. Lichens containing (i) green algae (phycobiont), (ii) green algae in association with cyanobacteria confined in cephalodia, or (iii) cyanobacteria (cyanobiont) as the photobiont partner were studied. Carbon isotope discrimination was analysed in different thallus sections, which varied in distance from the margin and in age. The marginal thallus region is considered to be youngest, while the central region is thought to be oldest. This analysis showed a clear variation in δ across the thallus related to distance from the growing margin. In most of the species examined, the highest δ values were found in marginal regions (younger), while the central and basal regions (older) showed significantly lower δ. To investigate the effects of the historical increase in atmospheric CO2 concentration and the concurrent decrease in the 13C content of atmospheric CO2 on the δ of lichens, experiments were carried out on herbarium samples of Lobaria pulmonaria collected from the mid 19th Century to 1953. The results obtained showed a pattern of variation of δ consistent with that of freshly collected samples; δ decreased substantially with increasing distance from the thallus margin, irrespective of the collection date. Moreover, no consistent variation of discrimination was found among different collection dates. These results demonstrate that the observed variation in δ is caused by age-related changes in the physiological behaviour of different thallus sections, and that the past 150 years of increasing CO2 concentration have not had significant effects on A in L. pulmonaria. Photosynthesis measurements, chlorophyll analysis and observations using optical microscopy, performed on freshly collected lichens, showed significant changes in morphological and physiological characteristics across the thallus. Particularly, remarkable variations in thickness were found across the thallus. These anatomical changes may be responsible for the variation in δ, through variations in CO2 transfer resistance and, consequently, in CO2 availability across the thallus. The lack of age-dependent variation in δ in cyanobiont lichens is possibly attributable to the operation of a CO2-coneentrating mechanism and, therefore, to a more constant CO2 environment across the thallus in this lichen group. 相似文献

Three fresh water microalgal isolates [Phormidium ambiguum (Cyanobacterium), Pseudochlorococcum typicum and Scenedesmus quadricauda var quadrispina (Chlorophyta)] were tested for tolerance and removal of mercury (Hg2+), lead (Pb2+) and cadmium (Cd2+) in aqueous solutions as a single metal species at conc. 5–100 mg / L under controled laboratory conditions. The obtained results showed that Hg2+ was the most toxic of the three metal ions to the test algae even at low concentration (< 20 mg/L). While lower concentration of Pb2+ and Cd2+ (5–20 mg / L) enhanced the algal growth (chlorophyll a and protein), elevated concentrations (40–100 mg / L) were inhibitory to the growth. The results also revealed that Ph. ambiguum was the most sensitive alga to the three metal ions even at lower concentrations (5 and 10 mg / L) while P. typicum and S. quadricauda were more tolerant to high metal concentrations up to 100 mg / L. The bioremoval of heavy metal ions (Hg2+, Pb2+ and Cd2+) by P. typicum from aqueous solution showed that the highest percentage of metal bioremoval occurred in the first 30 min of contact recording 97% (Hg2+), 86% (Cd2+) and 70% (Pb2+). Transmission electron microscopy (TEM) was used to study the interaction between heavy metal ions and P. typicum cells. At ultrastructural level, an electron dense layers were detected on the algal cell surfaces when exposed to Cd, Hg and Pb. At the same time, dark spherical electron dense bodies were accumulated in the vacuoles of the algal cells exposed to Pb. Sterols were extracted from the lichens Lobaria pulmonaria, Lobaria scrobiculata and Usnea longissima with chloroform-methanol (2:1) (solvent-extractable fraction) followed by saponification of the residual lichen material to give a tightly-bound sterol fraction. Photosynthetic parameters of the lichen Ramalina duriael were investigated in vivo, by the photoacoustic method using intensity—modulated exciting light (frequency range 5–300 Hz). The photoacoustic signal in intact lichens was similar in its general characteristics to that obtained from intact leaves of higher plants (Poulet et al., Biochim. Biophys. Acta 724, 433–446, 1983). It included two components interpreted to be due to modulated heat and modulated oxygen evolution. The quantum yield of the oxygen evolution component was maximal in the red spectral region and exhibited the red drop decrease at wavelengths larger than 680 nm, similar to observations in higher plants. In contrast to those however, there was a pronounced decrease in this yield in the region below about 600 nm, indicating that pigments absorbing at shorter wavelengths are inefficient energy transfer agents. Similar results were observed for the quantum yield spectrum of photochemical energy storage. Analysis of oxygen diffusion in the symbiont alga, from the modulation frequency dependence of the ratio of oxygen evolution to photothermal signal component is consistent with an average diffusion path of about 4 m, compared to a smaller, 1 m, average diffusion path obtained in green leaves. 相似文献

Sulphite at a concentration of 1 mM did not strongly affect the growth of mycelium. Higher concentrations of 5–20 mM almost
completely inhibited the growth of mycelium and superoxide dismutase (SOD) [EC.1.15.1.1.] activity. The activity of this enzyme
was not detectable on polyacrylamide gels. The lack of induction of SOD and the resulting oxidative stress may in part be
responsible for the growth inhibition caused by high concentrations of sulphite.

Aim The global distribution of the European species of Melanelia Essl. (Lichenes: Parmeliaceae) was investigated in order to understand their distribution patterns against the background of ecogeographical and historical factors. Location The location of the study is global, with a local emphasis on Europe. Methods Geographical distribution and ecology of the species were investigated on the basis of herbarium studies and data from the literature as well as field observations. Distribution maps were created with ArcView GIS. The distribution patterns are expressed as three‐dimensional ‘areal formulas’, regarding zonal distribution, altitudinal range and oceanicity, using a previously described method. The observed distribution patterns are discussed on the basis of their analogies with vascular plants and with respect to the ecogeographical vs. historical factors reflected by them. Results With the exception of one species that is endemic to Europe, all species studied occur in both northern continents. A number of species have tropical‐alpine outposts, and two species occur also in extratropical zones of the southern hemisphere. Arctic and boreal distributions are circumpolar, while in the southern holarctic zones an affinity to the western sides of both northern continents is frequent. Main conclusions The distribution patterns appear to be mainly determined by contemporary ecogeographical factors. Most species probably have largely filled their potential distribution, at least within the Holarctic. Thus, the geographical origin and dispersal history of a species cannot reliably be reconstructed; they can be dissimilar in different species with similar distributions. 相似文献

Following a forest fire (27 500 ha) in 1994, post-fire recolonization of Quercus hypoeleucoides by epiphytic lichens was documented as changes in lichen cover, number of small thalli, specific factors that affected reestablishment of lichens, and modes of dispersal. Three sites in the Chiricahua Mountains (Arizona, USA) were chosen according to the severity of fire damage—unburned, moderately burned, and severely burned. From 1994 through 1997, the amount of dead lichen cover significantly increased at the moderately burned site. For the same time period, the amount of live lichen cover significantly increased at the severely burned site. Numbers of new thalli increased significantly at the severely burned site each year but only in the last year (1996–1997) for the moderately burned site. Bark texture and proximity to trees with lichens were among the most important physical factors for recolonization. The most important means of dispersal for Flavopunctelia praesignis was fragmentation. For Punctelia hypoleucites, the primary means of dispersal was spores. Increases in live lichen cover and numbers of new thalli occur faster in severely burned areas probably due to the loss of lichens on tree trunks, which provides space and a lack of competition. 相似文献

Cadmium is suspected to exert its toxic action on cells through oxidative damage. However, the transition metal is unable to directly generate reactive oxygen species (ROS) via redox reactions with molecular oxygen in a biological environment. Here, we show that bright yellow-2 (BY-2) tobacco cells exposed to millimolar concentrations of CdCl(2) developed cell death within 2-3 h. The death process was preceded by two successive waves of ROS differing in their nature and subcellular localization. Firstly, these consisted in the transient NADPH oxidase-dependent accumulation of H(2)O(2) followed by the accumulation of O(2) (-*) in mitochondria. A third wave of ROS consisting in fatty acid hydroperoxide accumulation was concomitant with cell death. Accumulation of H(2)O(2) was preceded by an increase in cytosolic free calcium concentration originating from internal pools that was essential to activate the NADPH oxidase. The cell line gp3, impaired in NADPH oxidase activity, and that was unable to accumulate H(2)O(2) in response to Cd(2+), was nevertheless poisoned by the metal. Therefore, this first wave of ROS was not sufficient to trigger all the cadmium-dependent deleterious effects. However, we show that the accumulation of O(2) (-*) of mitochondrial origin and membrane peroxidation are key players in Cd(2+)-induced cell death. 相似文献

利用植物修复污染的土壤已受到广泛的关注.采用土壤盆栽试验,比较了超累积植物遏蓝菜与3种高生物量植物印度芥菜、烟草和向日葵对长期施用含镉有机、无机肥料污染的土壤(总Cd,2.87mg·kg-1)的提取效率.研究结果表明,遏蓝菜富集镉的能力明显高于其他3种植物,其地上部镉含量可达43.7mg·kg-1,分别是烟草、印度芥菜和向日葵(叶)的10、27和56倍;而地上部生物量最高的植物烟草,其生物量干重为24.8g· pot-1,分别是遏蓝菜、印度芥菜、向日葵的35倍、3倍、2倍.4种植物提取镉最多的是烟草,每盆可以提取117μg,遏蓝菜和印度芥菜提取镉量分别为35μg·pot-1和30μg·pot-1,向日葵提取量最少,每盆仅为10μg左右.植物对土壤中镉的提取效率分别为:烟草 1%,遏蓝菜0.6%,印度芥菜 0.5%,向日葵0.08%.4种植物种植后,土壤总镉和有效态镉含量没有显著的变化. 相似文献

Abstract The formate oxidizing capacity of Wolinella recta ATCC 33238 was studied in relation to growth under anaerobic and microaerobic conditions. Three distinct activities could be recognized: (a) cyanide-insensitive H2 O2 -producing oxidation of formate; (b) peroxidation of formate (H2 O2 -consuming); (c) oxidation of formate via an electron transport chain with oxygen as the electron acceptor. The contribution of these different formate oxidizing components during the growth of W. recta was dependent on the extent of aeration. It is suggested that due to the relative increase in overall H2 O2 formation at higher oxygen tensions growth of W. recta appears possible only under anaerobic and microaerobic conditions. 相似文献

Sulphur-heterotrophic growth exhibited a dual response to the expression of sulphate-assimilating enzymes. The level of ATP-sulphurylase (EC 2.7.7.4) appeared repressed while sulphite reductase (EC 1.8.7.1) and O-acetyl-l-serine sulphhydrylase (EC 4.2.99.8) were derepressed and coordinated in their occurrence. The capability of the cells to reduce adenylylphosphosulphate or 3-phospho adenylylphosphosulphate to cysteine coincided with the activity of sulphite reductase. 重金属的生物不可降解性使其在环境中长期存在,导致严重的环境污染,对人类健康和生态系统构成威胁。与传统的物化修复技术相比,微生物修复具有成本低廉、环境友好和高效等特点。在面对重金属胁迫或营养不均衡时,微生物会被激发以分泌合成胞外多糖(exopolysaccharides, EPS)。由此可见,EPS的产生是微生物对抗重金属胁迫的重要策略之一。EPS不仅能保护微生物在低温、高温、高盐等极端环境或受毒性化合物胁迫的条件下存活,并且在细胞内外进行信息和物质的交流与传递,既作为保护屏障限制重金属离子进入细胞,又作为介质进行交流。EPS结构中含有多个带负电荷的官能团,能够与重金属离子发生络合、离子交换、氧化还原等反应,从而降低重金属的生物有效性并减轻其毒性。微生物EPS在重金属胁迫环境中的修复具有重要意义。然而,目前缺乏关于微生物EPS合成过程、与重金属互作机制及其在重金属胁迫环境中应用现状的系统综述。本文概述了微生物EPS及其分类,详细阐述了细菌EPS胞内及胞外的生物合成机制,并探讨了微生物EPS与重金属互作机制,以及微生物EPS修复水、土环境中重金属污染方面的研究进展。最后,展望了EPS合成及其在重金属修复中的作用机制研究,可为微生物EPS进一步应用于环境重金属污染修复提供支持。 相似文献

1. The NADPH-oxidizing activity of a 100 000 × g particulate fraction of the postnuclear supernatant obtained from guinea-pig phagocytosing polymorphonuclear leucocytes has been assayed by simultaneous determination of oxygen consumption, NADPH oxidation and O?2 generation at pH 5.5 and 7.0 and with 0.15 mM and 1 mM NADPH.2. The measurements of oxygen consumption and NADPH oxidation gave comparable results. The stoichiometry between the oxygen consumed and the NADPH oxidized was 1 : 1.3. A markedly lower enzymatic activity was observed, under all the experimental conditions used, when the O?2 generation assay was employed as compared to the assays of oxygen uptake and NADPH oxidation.4. The explanation of this difference came from the analysis of the effect of superoxide dismutase and of cytochrome c which removes O?2 formed during the oxidation of NADPH.5. Both superoxide dismutase and cytochrome c inhibited the NADPH-oxidizing reaction at pH 5.5. The inhibition was higher with 1 mM NADPH than with 0.15 mM NADPH.6. Both superoxide dismutase and cytochrome c inhibited the NADPH-oxidizing reaction at pH 7.0 with 1 mM NADPH but less than at pH 5.5 with 1 mM NADPH.7. The effect of superoxide dismutase at pH 7.0 with 0.15 mM NADPH was negligible.8. In all instances the inhibitory effect of cytochrome c was greater than that of superoxide dismutase.9. It was concluded that the NADPH-oxidizing reaction studied here is made up of three components: an enzymatic univalent reduction of O2; an enzymatic, apparently non-univalent, O2 reduction and a non-enzymatic chain reaction.10. These three components are variably and independently affected by the experimental conditions used. For example, the chain reaction is freely operative at pH 5.5 with 1 mM NADPH but is almost absent at pH 7.0 with 0.15 mM NADPH, whereas the univalent reduction of O2 is optimal at pH 7.0 with 1 mM NADPH. 相似文献

A comparative study of the three gastropod species Nucella lapillus (L.), Ocenebra erinacea (L.) and Hinia (Nassarius) reticulata (L.) reveals that Nucella is the most TBT sensitive species while Hinia is the least sensitive. Of the two imposex indices VDSI and RPSI, good interspecies correlations were obtained only for VDSI.
The three species can be considered as complementary not only in terms of their ecology but also for their levels of sensitivity.
Indeed, Nucella and Ocenebra are useful test species at TBT concentrations below 2 ng Sn 1−1 while Hinia is the more appropriate species at higher TBT levels. For the first time, two sterilised Hinia females are recorded. This sterilization does not seem to be due to proliferation of vas deferens tissue in the vaginal opening
and further investigation is needed to find intermediary VDS stages between stage 4+ and sterilization. Studies using Nucella show that the use of narcotization in imposex analysis leads to an underestimation of RPSI compared with non-narcotization
methods. Indeed, narcotization straightens the penis and increases its length when compared with non-narcotized animals, but

Detention ponds and constructed wetlands have proven to be effective in reducing peak stormwater runoff volume and flow, and recent interest has extended to utilizing them to improve stormwater runoff quality. A review of stormwater runoff studies indicated that lead, zinc, copper, cadmium, phosphorus, and chloride are contaminants of primary concern. In laboratory settings, the uptake of contaminants by three wetland plant species, Glyceria grandis, Scirpus validus, and Spartina pectinata, was examined and removal rates from nutrient solutions inflow and nonflow reactors were measured. The removal rates varied by plant species and target contaminant, and no one species was the best accumulator of all six contaminants. Belowground tissues of all three species accumulated higher concentrations of the four heavy metals and aboveground tissues accumulated higher concentrations of phosphorus and chloride. Plants grown in flow reactors showed significantly higher accumulation rates than those grown in nonflow reactors. Also, plants grown hydroponically accumulated higher concentrations of the six target contaminants than those grown in sand reactors. However, those grown in sand had a much greater increase of biomass and removed a greater mass of the six target contaminants. Removal rates measured in these experiments can be used to design detention ponds to maximize stormwater remediation. 相似文献

Population structure and history is poorly known in most lichenized ascomycetes. Many species display large-scale infraspecific disjunctions, which have been explained alternately by range fragmentation in species of high age and widespread long-distance dispersal. Using the lichen Cavernularia hultenii, which is widely disjunct across North America and Europe, Pleistocene and Holocene population history was inferred. The internal transcribed spacer (ITS) and part of the the intergenic spacer (IGS) region of the nuclear ribosomal DNA were sequenced in 300 individuals representing 62 populations across the range of the species. While four ancestral haplotypes are found in all areas, none of the observed tip haplotypes is present in more than one of the three part ranges. Although this is evidence for a past fragmentation event, nested clade analysis (NCA) remains equivocal in the choice between allopatric fragmentation and long-distance dispersal. Mismatch distributions indicate exponential population growth, probably during postglacial invasion of C. hultenii into formerly glaciated areas of western North America. The presence of one southern and at least one northern glacial refugium in South Central Alaska is inferred. Evidence for another refugium in the Queen Charlotte Islands or Alexander Archipelago is inconclusive because of sparse sampling. However, a range expansion was not confirmed unambiguously by NCA. The limited power of NCA to infer past range fragmentations and expansions is due apparently to the shallow haplotype network and widespread ancestral haplotypes. This can be explained by slow genetic drift causing incomplete removal of ancestral haplotypes from the postfragmentation and postexpansion areas. 相似文献
