Human growth hormone (HGH) response to arginine (25 gm IV in 30 min) and to insulin (0.1 U/kg B.W.) was studied in 12 male patients (mean age 36 +/- 2 years), with normal glucose tolerance and normal body weight, affected with Fredrickson's Type IV primary hyperlipemia. The patients were examined both when plasma triglycerides (TG) were elevated and following clofibrate (2 gm/die for 30-60 days) induced TG reduction. No variations in glucose or FFA behaviour or in body weight were observed after clofibrate. HGH response to arginine was absent, while that to insulin was only inhibited, when plasma TG were elevated. A significant increase in HGH peaks after arginine (from 1.99 +/- 0.59 to 9.34 +/- 1.58 ng/ml) and a slight increment in HGH peaks after insulin (from 23.09 +/- 7.19 to 31.46 +/- 7.95 ng/ml) were observed following reduction in plasma TG. Arginine test was carried out in 7 normal subjects during saline infusion and at the 3rd hour of lipid infusion (Intralipid 20%). HGH response to arginine was absent in all of the subjects during lipid infusion. The HGH response to insulin test, carried out in 9 other normal subjects during saline infusion and at the 3rd hour of lipid infusion (Lipiphysan 15%) was significantly inhibited during lipid infusion. Since lipid infusion provoked an increment, not only in plasma TG but also in FFA, the inhibition of HGH release could be correlated with the elevated plasma levels of both TG and FFA. The results obtained in both spontaneous and experimental hyperlipemia not only confirm the role played by FFA in the regulation of HGH secretion, but also support the hypothesis that elevated TG levels could inhibit HGH response to some stimuli.  相似文献

Ethanolamine kinase, CTP:ethanolaminephosphate cytidylyltransferase (ECT), and ethanolaminephosphotransferase, which sequentially catalyze the primary pathway for phosphatidylethanolamine synthesis, were measured in castor bean (Ricinus communis L. var Hale) endosperm for 6 d after the onset of imbibition. Ethanolamine kinase (EC 2.7.1.82) activity was cytosolic, increasing slowly during the first 5 d and then declining. Total ECT (EC 2.7.7.14) activity increased until the 4th d, but the endoplasmic reticulum fraction of the activity peaked at d 3, and the mitochondrial activity peaked at d 4. Diacylglycerol:CDPethanolamine ethanolaminephosphotransferase (EC 2.7.8.1) increased during the first 2 d after imbibition began, after which it declined. The lowest activity of ethanolamine kinase during postgermination was more than 5-fold higher than the maximum activity of ECT, and the total activity of diacylglycerol:CDPethanolamine ethanolaminephosphotransferase at d 2 was at least triple that of ECT of the endoplasmic reticulum. We have partially purified ECT from mitochondrial fractions of postgermination castor bean endosperm starting with mitochondria purified by sucrose (Suc) density gradient centrifugation and broken by osmotic shock and homogenization. The membrane-bound ECT was solubilized with 1.5% 3-[(3-cholamidopropyl)dimethylammonio]-2-hydroxy-1-propanesulfonate and purified approximately 118-fold by polyethylene glycol precipitation, chromatography on Sephacryl S-200, and then Suc gradient centrifugation. The continuous presence of both salt (0.5 M NaCl) and detergent (1% [w/v] 3-[(3-cholamidopropyl)dimethylammonio]-2-hydroxy-1-propanesulfonate) was necessary to prevent aggregation. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of the final activity peak resulted in a prominent protein band at 35 kD, which correlated with bands from peak ECT activity fractions after both Suc gradient centrifugation and gel filtration on Sephacryl S-200. The activity of this enzyme was enhanced by the addition of several phospholipids.  相似文献

目的:观察头颅局部行亚低温治疗脑梗死后患者血清一氧化氮、内皮素以及细胞间粘附分子含量的变化,探讨亚低温治疗对脑组织发挥保护作用的机制。方法:搜集2012年5月至2013年2间,哈尔滨医科大学附属第四医院神经内科收治的62例脑梗死患者,并随机分为亚低温治疗组(32例)和常规治疗对照组(30例),治疗组在药物治疗脑梗死的同时加用局部亚低温治疗技术,在治疗前、治疗后3、7、14d进行一氧化氮、内皮素及细胞间粘附分子的测定;两组患者分别于治疗前以及治疗后第30d,对神经功能缺损采用NIHSS量表进行评分,对日常生活活动能力采用Bathel指数评分,对运动功能采用FuglMeyer法进行评价。结果:在治疗后第3、7、14天,亚低温治疗组一氧化氮含量较对照组显著升高(均为P〈0.01),在治疗后第7天亚低温治疗组内皮素含量低于对照组(P〈0.05),第3、14天亚低温组内皮素含量显著低于对照组(P〈0.01);治疗前两组NIHSS评分、Bamel指数和FuglMeyer评分差异均无显著性意义(P〉0.05)。在治疗组治疗后的第3、7、14天,亚低温治疗组细胞间粘附分子与治疗前相比,均出现降低(P〈0.05);而且在第3天,治疗组和对照组有显著性差异(P〈0.05)。治疗1个月后,亚低温治疗组NIHss评分比常规治疗组显著降低,亚低温治疗组Bathel指数和FuglMeyer评分均明显高于对照组,差异具有显著性。结论:局部亚低温治疗对血管内皮细胞功能有较好的保护作用,可改善脑梗死患者神经功能缺损及预后。  相似文献

Acute renal failure (ARF) was produced by the single intraperitoneal injection of 3 mg/kg mercuric chloride (HgCl2) in male Wistar rats. Immediately after, and in the 1st, 3rd, 6th, 24th and 48th hour after HgCl2 administration the following variables were monitored: plasma renin concentration (PRC), renal renin concentration (RRC) blood-urea nitrogen (BUN), plasma sodium (PNa), plasma creatinine (PCr) concentrations and haematocrit (Ht). Haematocrit and PNa increased during the first hour and returned to the control value in the 3rd hour. Thereafter, their level remained unchanged. Plasma renin concentration increased threefold during the first six hours after the HgCl2 injection, however, by the 48th hour it returned to the control value. In the first 24 hours of ARF, RRC remained unchanged. However, by the 48th hour its level increased four times the control value. After mercury injection BUN and PCr increased progressively. We were not able to establish any significant correlation between the changes of PRC and BUN. A gradual increase of RRC was observed in the course of ARF.  相似文献

To evaluate the clinical significance of human growth hormone (HGH) dynamics during transsphenoidal microsurgery for acromegaly, serial HGH levels during the surgery were compared with the post-operative basal HGH levels in 15 acromegalic patients, retrospectively. In all patients, in whom the HGH level immediately after tumor resection was reduced below 5 ng/ml or the decreasing rate which stood for the magnitude of decrease of HGH during resection procedure was above 80%, postoperative permanent HGH levels fell to below 5 ng/ml, namely, within the normal range. The rapid radioimmunoassay (RIA) of HGH was developed by the use of high affinity antibody in order to inform the surgeons of plasma HGH levels quickly during the surgery. It is now possible to know HGH levels about 1 hour after submitting the samples. It was verified that HGH levels measured in the rapid RIA correlated well with those obtained in the conventional way. The rapid RIA method was applied to 6 patients and the correlation between HGH levels during the surgery and post operative levels was also studied prospectively. The results were essentially identical to those in a retrospective study. It was suggested that the HGH level immediately after tumor resection and the extent of the decrease in HGH during the resection procedure were good indicators of the prognosis of surgical results in acromegaly and the rapid RIA method was useful in order to judge HGH levels quickly during the surgery.  相似文献

Twelve male, sedentary volunteers (22.0 +/-) were submitted to three weeks of a bicycle ergometer training, consisting of 45 min exercise (at 70% VO2max), 4 times in the first week and 3 times in the next 2 weeks. They performed four incremental exercise tests with the power output increased by 50 W every 3 min until volitional exhaustion: two before training (C1 and C2), and after one (T1) and three (T3) weeks of training. Before and after each load the plasma noradrenaline (NA), adrenaline (A) and blood lactate (LA) concentrations were determined in venous blood samples as well as plasma growth hormone (HGH) and cortisol concentrations before and at the end of exercise. A decrease in NA concentration was found already after 1 week of training at power output of 100 W (p<0.01) and 200 W (p<0.05). Similar decline was maintained after 3 weeks of training. No significant training-induced differences in plasma A concentration were found, however, the thresholds for both catecholamines were significantly shifted towards higher values after 3 weeks of training. One week of training caused a decrease in the pre-exercise (p<0.01), as well as post-exercise (p<0.05) plasma cortisol and HGH concentrations. It was concluded that endurance training induced a decrease in HGH, cortisol and NA concentration already after one week of training. A decline of pre-exercise plasma HGH and cortisol levels with time of experiment may, in part, indicate familiarization to exercise protocol.  相似文献

目的:探讨抗CCL21单克隆抗体处理对小鼠急性心肌梗死后心室重构和心功能的影响。方法:C57BL/6小鼠随机分为假手术组、模型组和CCL21单抗干预组,并进一步分为1、3、7和21 d亚组。采用结扎冠状动脉左前降支的方法构建小鼠急性心肌梗死模型,在冠状动脉结扎后5 min和第3天,模型组小鼠静脉注射isotype-IgG 1.0 mg,CCL21单抗干预组小鼠静脉注射山羊抗小鼠CCL21单克隆抗体1.0 mg。建模后,Western blot法检测急性心肌梗死后第1、3、7天心肌组织CCR7表达,检测急性心肌梗死后第7天心肌组织MMP-2和MMP-9表达;建模后第1、3、7天,ELISA法检测各组小鼠血清TNF-α和IL-6水平,每组检测8只小鼠。在建模后第7天和21天,超声心动图法评估左心室功能变化。结果:与假手术组比较,模型组小鼠急性心肌梗死后血清CCL21、TNF-α和IL-6及心肌组织CCR7、MMP-2、MMP-9明显升高(P<0.05);与模型组比较,CCL21单抗干预组小鼠血清TNF-α和IL-6及梗死区心肌组织MMP-9水平明显降低(P<0.05)。结论:抗CCL21单克隆抗体处理,通过抑制梗死后炎症反应及MMP-9表达水平发挥防止小鼠心脏重构和保护左心室功能的效应。  相似文献

Fecal bacteria from a healthy individual were screened for the specific bacteria involved in the metabolism of dietary isoflavonoids. Two strains of bacteria capable of producing primary and secondary metabolites from the natural isoflavone glycosides daidzin and genistin were detected. The metabolites were identified by comparison of their HPLC/mass, 1H NMR and UV spectra with those of standard and synthetic compounds. Both Escherichia coli HGH21 and the gram-positive strain HGH6 converted daidzin and genistin to the their respective aglycones daidzein and genistein. Under anoxic conditions, strain HGH6 further metabolized the isoflavones daidzein and genistein to dihydrodaidzein and dihydrogenistein, respectively. The reduction of a double bond between C-2 and C-3 to a single bond was isoflavonoid-specific by strain HGH6, which did not reduce a similar bond in the flavonoids apigenin and chrysin. Strain HGH6 did not further metabolize dihydrodaidzein and dihydrogenistein. This is the first study in which specific colonic bacteria that are involved in the metabolism of daidzin and genistin have been detected.  相似文献

目的研究一年内Xive种植体周常见致病菌的变化情况,为种植体的定期维护提供理论基础。方法临床选取32名种植患者的44枚种植体,记录了修复后1个月,修复后3个月,修复后6个月,修复后12个月四个时段,入选种植牙的菌斑指数(PLI),牙龈指数(G1),牙龈出血指数(GBI),探诊深度(PD);采用产黑菌、放线菌、具核梭杆菌及厌氧菌的选择性培养基对龈下菌斑标本进行了分离培养。结果随时间延长,种植体周几种龈下菌的检出量除牯放菌外不断增加,1个月到3个月时增加的趋势最为明显,到6个月左右趋于稳定。厌氧菌总数、产黑菌、核梭菌、粘放菌的统计值在1个月和3个月、3个月和6个月比较差异有显著性,但在6个月和12个月比较差异无显著性。各临床指标和X-线指标随时问的变化差异无显著性。结论在临床工作中,应注意加强修复后3个月时种植患者的口腔维护,防止种植体周围炎的发生,提高种植成功率。  相似文献

目的:观察电休克对大鼠空间记忆和海马磷酸化细胞外调节蛋白激酶(p-ERK)活性的影响。方法:大鼠随机分为电休克组和伪电休克组,每组12只。电休克组每天给予电痉挛刺激,伪电休克组每天给予假电痉挛刺激,共10天;第11天用水迷宫检测各组大鼠的空间学习记忆,然后每组大鼠再随机分为两组,每组6只。一组于学习后1小时处死取海马用Western blot法检测p-ERK活性,另一组于48小时后行水迷宫空间位置探寻实验检测大鼠的存储记忆。结果:电休克组的潜伏期显著长于伪电休克组(P0.01)。电休克组在隐匿平台周围区域/相反区域的搜寻时间无显著性差异(P0.05);伪电休克组在隐匿平台周围区域/相反区域的搜寻时间有显著性差异(P0.05)。电休克组海马p-ERK活性较伪电休克组显著下降(P0.01)。结论:电休克可导致大鼠显著空间记忆障碍,海马p-ERK活性的降低可能是其机制之一。  相似文献

The research involved comparison of characteristics of the power spectrum, index, coherence of the EEG at preterm and full-term infants of the first month of life. 6 groups of children are surveyed: (1) 30-32, (2) 33-34, (3) 35-36, (4) 37-38, (5) 39-40, (6) 41-42 weeks of gestation; each group consisted of 10 children. Capacity of delta frequencies was high in the 5th group. Capacity of theta frequencies was high in the 3rd group. The index of theta frequencies was high in the 2nd, 3rd and 4th groups. Capacity of alpha frequencies was hagh in the 2nd group. The index of alpha frequencies was high in the 1st, 2nd and 3rd groups. Indexes and of beta1- and beta2-frequencies were higher in the 1st, 2nd and 4th groups. The maximum quantity of coherent delta, theta and alpha frequencies was in the 6th group. In the 2nd group, in comparison with 5th group, were more coherent delta frequencies in the occipital and occipital-central areas. The results obtained show that the spectrum of capacity, indexes and coherence of the EEG depend on term of gestation at the moment of birth but this dependence has nonlinear character.  相似文献

目的:观察血栓通对急性脑梗死(actue cerebral infarction,ACI)患者血清D-二聚体和超敏C反应蛋白(high sensitivity C-reactive protein,hs-CRP)水平的影响。方法:将68例ACI患者随机分为观察组和对照组。两组进行常规治疗,观察组加用血栓通0.5 g/d,静滴。于治疗前、治疗后3 d、7 d和14 d检测患者血清D-二聚体和hs-CRP浓度变化,比较两组治疗前后改良爱丁堡-斯堪的纳维亚神经功能评分(MESSS)及日常生活能力Barthel指数(BI)的变化。结果:观察组在治疗后3 d、7 d和14 d,血清D-二聚体和hs-CRP浓度较对照组明显降低(P0.05),MESSS和BI评分在治疗后7 d和14 d较对照组明显改善(P0.05)。观察组的显效率和总有效率高于对照组(P0.05)。结论:血栓通可降低ACI患者血清D-二聚体和hs-CRP水平,改善神经功能,是一种有效治疗急性脑梗塞的方法。  相似文献

目的 采用牙周洁治和强化的口腔健康教育方法,通过检测牙周菌群和牙周临床指数的动态变化,探讨口腔卫生预防措施对固定正畸患者牙周菌群的影响。方法 选择正畸初诊患者20例,平均分为试验组(T组)和对照组(C组)。于正畸治疗开始前检查右上颌第一磨牙16和右下颌中切牙41牙周状况,并采集其近中颊轴嵴处龈缘菌斑做细菌分离培养。T组于治疗前进行全口洁治,每月复诊加力时均给予口腔卫生检查,强调口腔卫生的重要性;C组仅在初诊时进行口腔卫生指导,其余不作处理。2组患者分别于矫治器安装后1、3和6个月进行临床及细菌学检查。结果 随观察时间的延长,T组颊侧菌斑指数和牙龈指数在第1、3、6个月时较基线降低;C组颊侧菌斑指数第6个月时较基线降低,舌侧探诊深度则升高(P<0.05)。细菌检出率和检出量的变化在T组可见韦荣球菌属降低而弯曲杆菌属和Gn产黑色素厌氧杆菌(BPAR)升高,C组消化链球菌属和BPAR升高(P<0.05);BPAR在第3个月、消化链球菌属在第6个月时T组检出率低于C组(P<0.05),而细菌检出量和牙周临床指数在2组间没有观测到处理因素的作用(P>0.05)。结论 正畸前即存在牙龈炎的患者,建议进行预防性牙周洁治;牙周洁治必须和口腔卫生教育、正确的日常菌斑控制措施结合进行。  相似文献

Sixty-four blood samples, obtained from 25 pituitary dwarfs treated with KABIHGH preparation, were checked for serum HGH concentration by a double antibody radioimmunoassay and for antibodies against HGH by the method of propylethyleneglycol separation. Antibodies to HGH were detected in the samples whose HGH concentration was falsely estimated to be more than 5 ng/ml, but not in those whose HGH concentration was 5 ng/ml or less. The measurement of HGH concentration by a double antibody technique in the serum obtained from pituitary dwarfs 3-4 days after the last injection can be used to screen the presence of antibodies to HGH peparation used.  相似文献

以扬麦16和徐麦30为试验材料,利用人工气候室模拟低温逆境,研究拔节期-3 ℃和-5 ℃低温胁迫对小麦植株受冻率、叶片内源激素含量和抗氧化酶活性的影响.结果表明: 随着处理温度的降低、胁迫时间的延长,小麦植株冻害等级与冻害指数增加,-5 ℃处理72 h两品种五级冻害率均为100%.低温处理结束当天,小麦叶片中内源激素脱落酸(ABA)、玉米素核苷(ZR)含量、抗氧化酶超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性随胁迫程度加重呈先升高后降低的趋势;处理结束后3 d,ABA、ZR含量及抗氧化酶活性较处理结束当天升高;至处理结束后6 d,与自然生长的对照处理接近.低温胁迫叶片中赤霉素(GA3)含量下降,处理结束后3和6 d,扬麦16叶片中GA3含量呈上升趋势,徐麦30则表现为先升高后下降.-5 ℃ 72 h重度胁迫处理叶片中ABA、ZR、GA3含量和SOD、POD、CAT活性均较对照显著下降.相关分析表明,较高的ABA、ZR含量、SOD、POD、CAT活性以及较低的GA3含量可减缓低温胁迫对小麦植株的伤害.  相似文献

At the 1st, 2nd and 3rd days after birth, subcutaneous injections of 6-oxydopamine in physiological solution containing 0.1% of ascorbic acid have been made to rat puppies in a dose 100 mg/kg per one injection. Within first three weeks, electromyographic studies were made on outbursts of autogenic periodic motor activity which is typical of animals at this age. It was shown that at the 3rd day of postnatal life, after 6-oxydopamine administration, total duration of motor excitation is significantly lower, whereas mean duration of the outbursts of continuous activity is lower even at the 7th day. Beginning from the 10th day, these parameters undergo opposite changes at the background of the increase in the number of outbursts and the decrease of intervals between the latter. The data obtained are discussed in relation to the role of catecholaminergic systems in regulation and realization of age dynamics of autogenic motor activity.  相似文献

Serum levels of luteinizing hormone (LH) were determined in fetal and prepubertal pigs from Day 49 postcoitum to 25 weeks of age, and also in unilaterally cryptorchid, freemartin and castrated pigs of similar ages. Serum LH was undetectable in the second trimester of pregnancy, but then gradually increased up to 2 weeks after birth in both sexes. 2-week-old pigs showed irregular LH peaks exceeding 2 ng/ml. Serum LH concentrations decreased during the 3rd and 4th weeks of life. Mean LH concentrations were approximately 2 ng/ml in castrated pigs and 1.3 + or -.7 ng/ml in freemartins. The differentiation and functional activity of the testis appeared to be well correlated with the changes in serum LH concentrations. Changes in serum LH could not be correlated with normal and abnormal gubernacular development.  相似文献

The aim of this study was the search of permanent proteins of the urinary proteome during a 520-day isolation experiment at the Institute of Biomedical Problems (IBMP) Ground-Based Test Facility in controlled conditions, using an autonomous life support system. The object of the study was urine sampled from 6 normal male subjects aged 25 to 37. The biological material samples (the second morning urine fractions) were collected for proteomic investigations against the background, on the 50th, 93rd, 124th, 153rd, 180th, 251st, 274th, 303rd, 330th, 371st, 400th, and 427th days of isolation, and on the 7th day after its completion. The samples were analyzed using chromatography–mass spectrometry, while the obtained results were analyzed using bioinformatics resources. The following seven permanent proteins were identified and observed during the entire period of urine investigations: epidermal growth factor, polymeric immunoglobulin receptor, plasma serine protease inhibitor, Alpha 1 microglobulin/bikunin precursor (AMBP), keratin (type II cytoskeletal 1), collagen alpha-1 (VI) chain, and serum albumin.  相似文献

Serum levels of free and total cholesterol and phospholipids were st udied in 84 women (average age 43.8 years) treated with Ovosiston for 1 to more than 9 months. Controls were 19 untreated women (average age 45 years). Total cholesterol and phospholipid levels were significantly greater in women treated for more than 3 months, but no significant change in free cholesterol levels was observed. Serum lipid levels were examined continuously in 6 women during several treatment cycles, and a trend toward increasing phospholipid levels was seen in 5 women; this trend reversed when treatment was interrupted. It is concluded that the characteristic increases in total cholesterol and phospholipid levels occur after the 2nd or 3rd treatment cycle.  相似文献

Effects of RU 486 (10 mg.kg-1, per.os) were assessed at mid-pregnancy in the rat. One hour after RU 486 treatment, myometrial electrical activity stayed low. It increased from the 3rd hour after administration of RU 486 and a perfect synchronization of the bursts of the action potentials was observed from the 6th h to the 24th h. Tissular steroid hormones and PGF2 alpha, evaluated at hour 6 after RU 486 administration, showed a decrease of progesterone concentrations in both myometrium and uterus. Estradiol levels decreased in uterus whereas, PGF2 alpha levels increased in both myometrium and uterus. These results show for the first time that RU 486 strongly increases the myometrial electrical activity in the rat at mid-pregnancy. This action was closely related to E2, P4 and PGF2 alpha concentrations.  相似文献
