The 6 x 6 x 4 analysis of variance of the percentages of theupper of two superposed ovules fertilized, after crosses betweengreen and variegated cultivars of Pelargonium x Hortorum Bailey,shows highly significant differences between female cultivarswith a highly significant maternal effect, and slight differencesbetween the plastid crosses which becomes highly significantin one cultivar. Differences between male cultivars and cultivarplastid interactions are barely significant. The differencesbetween cultivars and between plastids are attributed to additiveeffects corresponding to additive gene action. A comparisonbetween the mean frequency of upper embryos for mid-parentsand their reciprocal sets of progeny shows a significant linearregression which is equated with the narrow heritability. The binomial analysis of the distribution of upper and lowerembryos between flowers, shows that the likelihood of an upperor lower ovule being fertilized is solely dependent on the overallfrequency of the event in the population, indicating that eachcarpel behaves independently of other carpels in the same flower. A comparison between the three facets of fertilization - upperembryo frequency, twin frequency and mean fertilization, eachcontrolled by the genotype, shows that each cultivar has itsspecific and distinct pattern of behaviour. Polyembryony seems to be advantageous to mother plants in detriment of their siblings which face competition since the beginning of seed development. This competition may limit the turnover of embryos into seedlings and their survival ability. We analysed polyembryony frequency and embryo to seedling turnover in three Handroanthus species with sporophytic apomixis. We tested if the embryo number per seed affected seed and embryo morphometry, seedling survival ability and seed individual survival (i.e. survival of at least one seedling per seed). The number of embryos per seed was compared with seedling number at different developmental stages. All 14 populations showed high frequencies of polyembryonic seeds (21–91%). As the number of embryos per seed increased (up to eight embryos/seed), there was a reduction of mean embryo mass, area, seedling length, individual seedling survival ability, and embryo to seedling turnover. There was also an increase in embryo morphological anomalies. However, enhanced seed individual survival was also observed. Thus, the high frequency of polyembryonic seeds and the increase in seed individual survival support the idea that polyembryony represents an alternative reproductive mechanism which can favours these species.  相似文献

We investigated the fates of ovules following self- and cross-pollination of the variably self-sterile Epilobium obcordatum by observing thin sections of ovaries 4, 5, and 10 d postpollination (PP). Three types of unfertilized ovules were observed: apparently normal ovules not entered by pollen tubes, nonfunctional ovules with a nucellus but lacking an embryo sac, and empty ovules with integuments but lacking a nucellus. The average frequencies for all three types were not different for selfs and crosses. Fertilization frequencies did not differ among pollination treatments, averaging 61.4% of the total sample of 4 and 5 d PP ovules. The phenotypes of failing postfertilization ovules at 4 and 5 d PP included those containing preglobular and globular embryos lacking endosperm, ovules with endosperm but no embryo present, and failing embryos in the presence of endosperm. Self- and cross-fertilized 10 d PP ovules also possessed heart-stage and torpedo-stage failing embryos. Early embryo failures (5 d PP) were distributed throughout the length of the ovary, as expected if failures are the consequence of factors intrinsic to each ovule. The sizes of normally developing self- and cross-fertilized embryos were not different at 4 d PP. The 5 d PP cross embryos were larger than selfs. A comparison of 10 d samples was not possible due to attrition of self embryos. Although ovular position did not affect the probability of early embryo failure, it did affect the size of normally developing embryos at 4, 5, and 10 d PP, with embryos at the stylar end larger than those at the basal end.  相似文献

Measurements were made using GC/MS SIM1 of the effects of temperatureon cis,trans-ABA levels in developing ovules and embryos oftwo pea genotypes contrasted in seed size. These effects werethen related to differences in the growth of the pods, seeds,embryos, and testae. In both genotypes high temperatures hastenedthe onset and rate of logarithmic and then linear growth, greatlyshortening the duration of pod and seed development but withoutgreatly altering seed size. Cis,trans-ABA was most concentratedxin the ovules immediately after fertilization. It also accumulatedin the embryo, more rapidly in the larger-seeded line, duringseed maturation. The stage when accumulation in the embryo beganwas the same irrespective of temperature. Accumulation ceasedwhen the pods started to desiccate. The effects of differentconstant temperatures on the maximum levels of embryo cis,trans-ABAwere relatively small and confounded in one genotype by variationin ovule abortion and in the other by differences in the stagewhen cis,trans-ABA accumulation ceased. However, when plantswere transferred from 13 °C to 29 °C at two differentstages during seed maturation, further seed growth was greatlyinhibited coincident with a substantial increase in embryo cis-trans-ABA.The results suggested a role for cis,trans-ABA in the controlof cotyledon enlargement during the linear phase of seed growth.  相似文献

Embryos excised from fresh seed had a moisture content of about37% (fr. wt.), 7% above that of the whole seed: mainly as aresult of the dryness of the seed coat. When cultured for 14d at 26°C embryo growth ranged from unorganized to fullyorganized. Embryos which had been directly surface-sterilizedexhibited lower fully organized growth than aseptically excisedembryos. During desiccation of the whole seed at 15°C and 15% r.h.moisture loss from the embryo was slow until the embryo reacheda mean critical moisture content for fully organized growthin vitro of about 30%. After this point the rate of moistureloss increased, quickly diminishing the moisture content differentialbetween the embryo and the whole seed. No such shoulder in theembryo desiccation curve was observed when using a low viability(7%) seed batch. When dried below 30% moisture content, suchaseptically excised embryos increasingly exhibited unorganizedgrowth in vitro, with only root meristem survival (46%) at 13%moisture content. In contrast, a much higher level of root meristem survival (84%of control) was observed when aseptically excised embryos wererapidly desiccated to 13% moisture content, using a flow ofsterile air. However, the benefits of rapid drying were lostif the embryos were directly surface-sterilized prior to desiccation.Furthermore, 80% of aseptically excised embryos which survivedrapid desiccation to 20% moisture content exhibited root meristemsurvival following subsequent cryopreservation. Winter rapeseed (Brassica napus, cv. Samouraï) flowersearly in spring and, under field conditions, short freezingperiods can occur. Unacclimatized plants were freeze-stressed(–3°C for 4 h) at different developmental stages ofbuds, open flowers and seeds. The dissection of pistils from stressed plants showed that freezingresults in shrivelled ovules. We assessed freezing injury onthe basis of per cent of shrivelled ovules: ovule sensitivitybegins early (8 d before anthesis) but increases up to anthesis.Crosspollination of stressed pistils with non-stressed pollenshowed that recording of freeze-injured ovules is a good methodfor early estimation of the effect of stress on seed yields. On the other hand, stress does not reduce the viability of pollen,except when it was applied at the binucleate pollen stage. Useof frozen pollen x nonstressed pistils has little effect onseed yields. Freeze injury on seeds was assessed by seed filling:seeds are very susceptible just after fertilization until 20d after fertilization (DAF). Freezing seems to inhibit seedfilling. A germination test of stressed seeds during their developmentindicated that embryo viability is not affected if the stressoccurs after 35 DAF. As the embryos develop, resistance to stressincreases. Apomixis and adventitious polyembryony have been reported for several species of Bombacoideae, including Eriotheca pubescens, a tree species of the Neotropical savanna (Cerrado) areas in Brazil. However, the origin of polyembryonic seeds and their importance for the reproduction of the species remained to be shown. Here, we analyzed the early embryology of this species to establish the apomictic origin of extranumerary embryos. We also observed the geographic distribution of polyembryony in E. pubescens, and tested if apomixis was related to the source of pollen (self or cross) and population density. Moreover, we tested if polyembryonic apomictic embryos would develop normally into seedlings. In the observed seed primordia, after a relatively long quiescent period, the zygote developed into a sexual embryo concurrently with adventitious apomictic embryos which developed from nucellus cells. Adventitious embryos develop faster than sexual ones and are morphologically similar, so that 44 days after anthesis it was virtually impossible to distinguish and trace the fate of the sexual embryo. Polyembryony is widely distributed in populations some 400 km distant, and only one strictly monoembryonic individual was observed during the study. The number of embryos per seed varied between fruits and individuals but was significantly higher in seeds from cross-pollinations than from selfs, although fruit and seed set after crosses were much lower than after selfs. Embryo development into seedlings depended on their weight at germination, but polyembryonic seeds germinated and produced up to seven seedlings per seed in greenhouse conditions. Adventitious embryony and apomictic seedlings would explain the mostly clonal populations suggested by molecular studies.  相似文献

Dun well, J. M. and Thurling, N. 1985. Role of sucrose in microsporeembryo production in Brassica napus ssp. oleifera.—J.exp. Bot. 36: 1478–1491. One cultivar of winter oil seed rape (Brassica napus ssp. oleifera)and three cultivars of spring rape were used in a study of theeffects of sucrose on microspore survival and embryo inductionin cultured anthers. A preliminary study on the winter cultivar(Fiona) revealed that the osmotic pressure of the supernatantof anther homogenates was equivalent to a solution of 17% sucrose.A study of microspore survival and embryo induction in thiscultivar on media containing either 8 %, 12%, 16% or 20% sucroserevealed the highest survival (after 16 d) and the greatestnumber of anthers with induced embryos (after 42 d) occurredon the highest sucrose concentration. A subsequent study on three spring cultivars (Willi, Duplo andTower) examined microspore survival at 8 d and embryo induction(42 d) on media containing either 8 % or 16 % sucrose and againrevealed much higher survival and induction at the higher concentration.The variation in response between the cultivars was also reducedby culture at the higher sucrose concentration. The beneficialeffect of the 16% level occurred regardless of the growth environmentof the donor plants and of the stage of pollen development atthe start of culture. The ovules and seeds of Canna show some striking differencesto those in other zingiberalean families. The pachychalazaldevelopment of the ovule results in a seed of which only a smallpart of the testa is of tegumentary origin. A silicified endotesta,characteristic of the order, is lacking. The mechanical layerof the seed is formed by a continuous exotesta of Malpighiancells. The intact seed coat is impermeable. The seed is ableto absorb water after the raising of a preformed imbibitionlid on the raphe. During imbibition the extotesta loses itshardness, allowing the embryo to emerge. The special structuralfeatures of the seed are discussed in relation to records onthe extreme longevity of Canna seeds. Embryogenic cultures were initiated from undeveloped ovules of several polyembryonic Citrus species on a basal medium supplemented with either malt extract, 2,4-D alone, or 2,4-D in combination with BA or daminozide. Primary embryos of all responsive cultivars were harvested directly from ovule cultures; secondary embryo harvests were made from Handin orange ovule cultures and long-term embryogenic callus. Differences were observed among cultivars and treatments in percentage of responsive ovules and total number of embryos produced. The most effective treatment for embryo production varied among cultivars. Embryo germination and plant establishment frequencies were determined for this plant regeneration system. Differences among cultivars with respect to regenerate survival percentage were minimal. Plant regeneration via secondary or long-term callus-derived embryos was as efficient as from primary embryos. Critical factors influencing plant production and survival were the production of normal viable embryos, balanced germination, and successful acclimatization to the external environment.  相似文献

The water status and water content of embryos of Quercus roburhave been determined during development. Using psychrometers,changes in osmotic potential at full imbibition and the proportionof water at zero reciprocal water potential, R0, were estimatedfrom curves of embryo relative water content and reciprocalwater potential. The specific moisture content of embryos decreasedduring development, with a concomitant increase in the osmoticpotential. However, R0 also increased such that, during thelast four weeks of development, there was little net changein the content of osmoticum per dry weight. Evidence is presentedthat starch stored in the cotyledon may be the major site ofmatrix-bound water, R0. Isolated embryonic axes had a slightly higher osmotic potentialthan the whole embryo but the R0 was much lower. The differencesbetween whole embryo and axis water relations parameters emphasizethe need to take account of R0 when discussing seed water statusand survival. Hypocotyl segments ofEleutherococcus senticosuscultured on Murashigeand Skoog's (MS) medium with 4.5 µM2,4-D produced somaticembryos directly from the surface of explants without interveningcallus formation. When these somatic embryos were subculturedto the same MS medium with 4.5 µM2,4-D, friable embryogeniccalli were formed mainly from radicle tips of somatic embryos,but at a low frequency (5%). Selected embryogenic calli weremaintained on MS agar or liquid medium with 4.5 µM2,4-D.To induce somatic embryo development, embryogenic calli andcell clumps were transferred to MS medium lacking 2,4-D. Thefrequency of somatic embryo formation differed between culturetypes with 1570 embryos formed per Petri dish from callus cultureand 5514 embryos formed per flask from cell suspension cultures.Somatic embryos formed on agar medium had larger cotyledonsthan those of embryos formed in liquid medium. GA3treatmentwas necessary to induce germination from somatic embryos. Therate of plant conversion was 97% in somatic embryos from callusculture and 76% in embryos from liquid culture. Johnson, M. A., Carlson, J. A., Conkey, J. H. and Noland, T.L. 1987. Biochemical changes associated with zygotic pine embryodevelopment.—J. exp. Bot. 38: 518–524. At intervals during the period from pre-fertilization to nearmaturation, pine (Pinus resinosa Ait. and Pinus strobus L.)ovules were analysed for several biochemical constituents, andthe results were expressed on a fresh weight basis. Lipid accumulatedin parallel with the growth of the developing seeds. Solubleprotein also accumulated but only in the initial stages of development.ATP content peaked approximately 2 weeks after fertilization,followed about one week later by the energy charge; these peakswere associated with maximal growth stages of the developingembryos. Likewise, peaks of glutathione (GSH) and ascorbic acid(AA), two water-soluble reductants, preceded or coincided withthe ATP maximum. At late stages of seed development, dissectionof the more mature ovules into embryos and gametophytes foranalysis revealed that the ATP, GSH, and AA were more concentratedin the embryonic tissue. On the other hand, this segregationshowed that virtually no proanthocyanidin was located in thedeveloping embryos proper, although they contained other reductants,some of which were probably phenolics. Also, general stainingwith reagents for phenolics and thiols indicated that the formeroccurred primarily in non-embryonic tissue, whereas GSH wasin the embryo per se. These findings are consistent with rolesfor ATP, GSH, and AA in the growth and development of zygoticpine embryos; however, it would appear that lipid and proteinare being stored for subsequent germination events and thatmuch of the phenolic component is segregated from the developingembryo. In chickpeas (Cicer arietinum L.) flowering and pod developmentproceed acropetally. In plants grown under normal field conditionsat Hyderabad, in peninsular India, and at Hissar in north India,at successively apical nodes of the branches there was a declinein pod number per node, weight per pod, seed number per podand/or weight per seed. The percentage of nitrogen in the seedswas the same in earlier and later-formed pods at Hyderabad;at Hissar the later-formed seeds contained a higher percentage.Earlier- and later-formed flowers contained similar numbersof ovules. The decline in seed number and/or weight per seedin the later-formed pods of 28 out of 29 cultivars indicatedthat pod-filling was limited by the supply of assimilates orother nutrients. By contrast, in one exceptionally small-seededcultivar there was no decline in the number or weight of seedsin later-formed pods, indicating that yield was limited by ‘sink’size. The 27 lemon cultivars analysed could be considered slightly or moderately polyembryonic, with 25 to 43% of seeds being polyembryonic and from 1.3 to 1.6 embryos per seed. On this basis, it is necessary to rescue zygotic embryos at an immature stage. Rescue and in vitro embryo development have been studied in two Citrus limon polyembryonic cultivars. Sucrose (50 and 70 g/l) was combined with Murashige and Skoog and Gamborg’s B5 media and tested for optimal growth response. An important effect of genotype was observed: embryos from cultivar ‘Eureka’ had greater survival, germination percentage, and radical development. While the sucrose concentration in the medium did not have an effect on germination, the medium affected the embryo survival and root development of the seedlings, Gamborg’s B5 medium giving the best results. The ability to form plants in vitro was affected by an increase of embryo developmental stage. The germination and seedling height were greater with embryos of seeds collected 135–150 days after anthesis.  相似文献

Development of multiple embryos from a single zygote, the phenomenon called monozygotic polyembryony, is a widespread reproductive strategy found in higher plants and especially in gymnosperms. The enigma of plant monozygotic polyembryony is that only one embryo in a polyembryonic seed usually survives while the others are eliminated at an early stage. Here we report that programmed cell death (PCD) is the major mechanism responsible for elimination of subordinate embryos in a polyembryonic seed. Using post-fertilized pine (Pinus sylvestris) ovules, we show that once the dominant embryo is selected and, subsequently, the entire female gametophyte is affected by PCD, the cells of subordinate embryos initiate an autolytic self-destruction program. The progression of embryonic PCD follows a rigid basal-apical pattern, first killing the most basally situated cells, adjacent to the suspensor, and then proceeding towards the apical region until all cells in the embryonal mass are doomed. Our data demonstrate that during polyembryony, PCD serves to halt competition among monozygotic embryos in order to ensure survival of one embryo.  相似文献

Using a combination of observations of fate of ovules in matured fruits and of fluorescence techniques to study pollen tube growth and fertilization of ovules, we examined patterns of seed formation within pods in natural populations of two species of Lathyrus, L. sylvestris and L. latifolius. We also examined variation in these patterns within and among populations and between two consecutive years. In both species, only a portion of the ovules were fertilized. Fertilization occurs over a period of several days and ovules at the stigmatic end of the fruit are the first to be fertilized. Fertilized ovules farthest from the stigma are closest to the maternal nutrition. The pattern of embryo abortion is interpreted as a balance between the early start and genetic quality of embryos near the stigma, on the one hand, and the nutritional advantage of proximity to maternal nutrients, on the other. Differences between the two species in patterns of seed maturation are postulated to be related to differences in breeding system. In L. sylvestris, a higher frequency of selling leads to less genetic diversity of pollen deposited on the stigma, lower competition among potential sires, and a more nearly random pattern of ovule fertilization and maturation within the pod.  相似文献

An ovule culture technique has been established for barley that allows the regeneration of plants from zygotes. An average of 1.3 plantlets per ovule could be regenerated from more than 60% of the cultured ovules and about 75% of the regenerated plantlets developed into normal, fertile plants. The same regeneration frequencies were obtained in intact ovules and in ovules where the two integuments had been removed from the micropylar region. Unfertilized ovules and ovules where the fertilized eggs had been destroyed by a microinjection needle did not give rise to embryo-like structures. Plants could be regenerated from the zygote at the same frequency at developmental stages from immediately after fertilization until the formation of bicellular embryos. This tissue culture system appeared to be largely independent of genotype since similar regeneration frequencies were obtained in two different barley cultivars, Igri and Alexis, that in anther and microspore culture behave differently. The same technique has also been applied successfully in the wheat cultivar Walter.  相似文献

The breeding system ofDalbergia miscolobiumBenth. was investigatedby means of controlled pollinations and studies of post-pollinationevents in pistils. This species does not have homomorphic self-incompatibility,but crossed flowers always have higher fruit set than selfs,despite the fact that embryo development may be initiated inselfed pistils.
