Carbon replicas of germinating sporangiospores of Linderinapennispora show the outer wall complex to break open basally,during the phase of swelling, and the surface of the germ tubeto be smooth. Chemical treatment reveal the microfibrillar wallof the germ tube to be continuous with the microfibrillar innerwall complex of the spore. Microfibrils of the germ tube arerandornly arranged and appear to be finer than those of thespore wall. Ultra-thin sections reveal the wall of the germtube to consist of an outer electron-dense layer and an innermicrofibrillar electron-transparent layer and both layers originatein the basal region of the spore between the plasmalemma andthe inner wall complex of the spore.  相似文献

Sporangiospore structure in Piptocephalis unispora Benjamin was studied using light microscopy, freeze-etching, scanning and transmission electron microscopy, and compared with that of other members of the Mucorales. A merosporangial wall, plasmalemmal invaginations, and wall protuberances were demonstrated in sections and their possible significance discussed.  相似文献

Infection of the mucoraceous host Cokeromyces recurvatus by Piptocephalis unispora was studied ultrastructurally, using a new technique involving yeast-phase cells of the host to obtain large numbers of infection sites for thin-sectioning. Morphologically, the haustorial apparatus was similar to that of fungi parasitic on higher plants, and comprised an appressorium, a neck region with a collar and a neck ring, and a lobed region surrounded by a sheath matrix enclosed in an extra-haustorial membrane. Penetration of the host by the infection peg probably involved both enzymatic degradation and physical pressure. Reaction of the host to infection is described and the results related to the theory of host infection by haustorial fungal parasites.  相似文献

Summary In germinated sporangiospores of Gilbertella persicaria, negatively contrasted fibrils, 20–70 Å diam, are seen in thin sections of the inner vegetative wall that is continuous with the germ tube wall. The fibrils are randomly oriented in a loose network in this wall and in the germ tube wall. Germ tubes have an additional surface layer of fine, positively contrasted fibrils which appear as a nap-like coating on the hyphae. Patterns of wall fibril orientation are not revealed by transverse sections of spore and germ tube walls, whereas oblique and tangential sections are favorable for examining cell wall architecture in situ. Staining patterns show textural and compositional differences among various wall layers.  相似文献

A mycoparasite identified as Thamnocephalis quadrupedata (Mucorales) was observed on cultures of the frog dung fungus, Basidiobolus ranarum. The parasitic fungus, T. quadrupedata possessed infection hyphae with appressoria and penetrating hyphae to attack their host prey and adhere firmly to the surface. The invasion was often by slender infection hyphae or infecting pegs which grew from the appressoria and penetrated the chitin-protein cuticle by both mechanical pressure and exocellular enzymes. The host fungus, B. ranarum, possessing primary conidia, capilliconidia, hyphal bodies, vegetative mycelia and zygospores, were infected by means of direct penetration and intrahyphal growth, resulting in host cell death. T. quadrupedata may also grow as a saprophyte on damp filter paper in a Petri dish, manifesting facultative necrosis.  相似文献

The toxigenicity of representatives of 15 species of Mucorales (Absidia glauca, Actinomucor elegans, Cunninghamella elegans, Helicostylum piriforme, Mortierella isabellina, Mortierella (Mucor) rammaniana, Mucor hiemalis, Mucor mucedo, Mucor spinosus, Phycomyces blakesleeanus, Rhizopus oligosporus, Rhizopus stolonifer, Syncephalastrum racemosum, Thamnidium elegans, Zygorhynchus moelleri) towards the larvae of brine shrimp (Artemia salina) and the growth of pea seedlings (Pisum sativum) and tobacco plants (Nicotiana tabacum) was evaluated. The fungi were cultivated on malt extract agar and aqueous solutions of the cultures were tested.Thamnidium elegans showed a marked toxic action towards brine shrimp (mortality: 74.1%) andPhycomyces, Actinomucor andSyncephalastrum were only weakly toxic. Length and weight of stems of pea seedlings were moderately reduced by extracts ofAbsidia, Cunninghamella, Zygorhynchus andThamnidium and to a lesser degree byMucor spinosus. Cunninghamella andMucor spinosus also inhibited the development of pea hypocotyls. The length of tobacco stems was reduced byMortierella ramanniana, Rhizopus stolonifer andCunninghamella elegans. Wilting or other toxic phenomena were never observed with both test plants. Considering the present results and data from literature it is suggested that species of Mucorales have only a weak toxigenicity.  相似文献

Two basic types of spore wall structures were observed in ultrastructural examination of Schizoplasmodiopsis micropunctata, Schizoplasmodiopsis pseudonendospora, Schizoplasmodipsis reticulata, and Schizoplasmodipsis vulgare. The spores of S. micropunctata, S. pseudoendospora, and S. vulgare had walls bearing electron-dense protrusions, while those of S. reticulata had walls with irregular thickenings. Prominent cytoplasmic features included 2 types of vesicles and electron-dense bodies studded with ribosomes. The ameboid phase of all species lacked the electron-dense bodies and, unlike the dormant phases, did not have mitochondrial surfaces covered with ribosomes.  相似文献

Appressorium development in the mycoparasite Piptocephalis unispora was studied by means of scanning electron microscopy using the techniques of critical point drying, sputter coating and light microscopy. The germ tube which contacts both the young host hypha or a germinating spore swells at the tip to form an appressorium closely adpressed to the surface of the host. Lateral proliferation of hyphae may occur from the mature appressorium. Factors affecting the sites of appressorium development are suggested and their significance discussed.  相似文献

During studies on Mucorales in semiarid and littoral dune areas in the northeast of Brazil, two cultures of an Absidia-like species were isolated from soil. They were characterized based on morphological, physiological and molecular data (5.8S and LSU rDNA sequences). The phylogenetic analyses of the isolates revealed that they belong to the Lichtheimiaceae and are closely related to species of Lichtheimia. The two isolates produced simple or branched, erect and circinate sporophores, occasionally with a septum under the sporangia, characteristics also common in Lichtheimia species. However, different from the described Lichtheimia species, the columellae of our isolates were mainly short hemispherical, never spatulate or elliptical and without projections. Sometimes, a long conical or bell shaped apophysis was found. Both isolates grew better at 30–35 °C, with no development at 42 °C, and giant cells were not observed. Based on the evidence of the analyzed datasets a new species of Lichtheimia is proposed.  相似文献

The ultrastructure of a marine, free-living heterotrophic kinetoplastid Cruzella marina was investigated with special attention being paid to the mitochondrion and flagellar organization. The flagellates have a polykinetoplastidal mitochondrion. Two flagella emerge from the pocket; one of these turns anteriorly being forward-directed, while the other is posteriorly directed to be adjacent to the ventral cell surface. The transition zone of both the flagella includes central filaments. The cytostome opens on the tip of the rostrum. The cytostome leads to the channel of cytopharynx, which penetrates the rostrum and proceeds into the flagellate body cytoplasm. The comparison of the relevant morphological and molecular data suggest that C. marina may arise early in the Kinetoplastidea lineage, before divergence of the majority taxa of the kinetoplastid flagellates.  相似文献

Summary The ultrastructure of the flagellar apparatus of aPleurochrysis, a coccolithophorid was studied in detail. Three major fibrous connecting bands and several accessory fibrous bands link the basal bodies, haptonema and microtubular flagellar roots. The asymmetrical flagellar root system is composed of three different microtubular roots (referred to here as roots 1,2, and 3) and a fibrous root. Root 1, associated with one of the basal bodies, is of the compound type, constructed of two sets of microtubules,viz. a broad sheet consisting of up to twenty closely aligned microtubules, and a secondary bundle made up of 100–200 microtubules which arises at right angles to the former. A thin electron-dense plate occurs on the surface of the microtubular sheet opposite the secondary bundle. The fibrous root arises from the same basal body and passes along the plasmalemma together with the microtubular sheet of root 1. Root 2 is also of the compound type and arises from one of the major connecting bands (called a distal band) as a four-stranded microtubular root and extends in the opposite direction to the haptonema. From this stranded root a secondary bundle of microtubules arises at approximately right angle. Root 3 is a more simple type, composed of at least six microtubules which are associated with the basal body. The flagellar transition region was found to be unusual for the classPrymnesiophyceae. The phylogenetic significance of the flagellar apparatus in thePrymnesiophyceae is discussed.  相似文献

The liberation of asexual propagules in the Mucorales was investigated by means of wind-tunnel experiments. Some species do not liberate propagules into an air stream, others liberate single spores or groups of spores.
