Fumonisin B1 toxin is produced by the fungusFusarium moniliforme Sheldon, which is systemic to maize (Zea mays L.) and maize seeds. The effects of zero to 100 parts per million fumonisin B1 on the germination process of maize seeds was determined. The presence of fumonisin had no effect on percent seed germination, but fumonisin inhibited radicle elongation by up to 75% after 48 hours of imbibition. An analysis of amylase secretion in the maize endosperm indicated that fumonisins inhibited amylase production in the germinating seed. Isoelectric focusing of endosperm extracts indicated that secretion of the low pI class of amylases was affected more that other amylase isozymes. The results suggested that the presence of high levels of fumonisin in maize seed may have deleterious effects on seedling emergence.  相似文献

Enhancement of synthetic seed conversion to seedlings in hybrid rice   总被引:3,自引:0,他引:3  
Somatic embryos of hybrid rice (Oryza sativa L.) were encapsulated in sodium alginate matrix provided with MS nutrients, growth regulators and protectants as synthetic endosperm to enhance germination and conversion capacity. The synthetic seeds with synthetic endosperm constituents of MS nutrient; sucrose (3%), IAA (0.5 mg l–1), NAA (0.5 mg l–1), BA (0.5 mg l–1) and charcoal (1.25%) gave a maximum germination (30%) and conversion (27%) of synthetic seeds. The inclusion of protectants, bavistin and streptomycin as constituents of synthetic endosperm had no effect on germination and conversion. The application of self-breaking gel beads technology increased the germination (52%) and conversion (47%) of synthetic seeds.  相似文献

Extracts prepared from resting seeds of Scots pine, Pinus sylvestris L., rapidly hydrolysed two peptides, Leu–Tyr and Ala–Gly, at pH 8.6 and 7.8, respectively. In gel chromatography on Sephadex G-100 the two activities were eluted as separate peaks, which indicates that they are due to two different peptidases. The seeds were allowed to germinate at 20°C, the activities of the two enzymes were assayed separately on extracts from the endosperm and seedling tissues at different stages of germination, and compared with corresponding changes in dry weight and total nitrogen. Both enzyme activities were relatively high in the endosperm of resting seeds, and they increased about 2- and 3-fold during germination (expressed as enzyme units per seed), the increases coinciding with the time of rapid reserve protein mobilization. Both enzymes were also abundant in the embryos of resting seeds, and during germination their activities increased even more rapidly than those in the endosperm. The possible role of these two “alkaline peptidases” in reserve protein hydrolysis is discussed.  相似文献

Extracts prepared from endosperms of germinating seeds of Scots pine, Pinus sylvestris L., rapidly hydrolysed the β-naphthylamides of L-phenylalanine and L-leucine optimally at pH 6.5 and that of L-arginine at pH 7.7. Disc electrophoresis followed by activity staining showed that the activities were due to two naphthylamidases (aminopeptidases) with different substrate specificities. Seeds were allowed to germinate at 20°C on agar gel in the dark and the activities on the three substrates were assayed from separated endosperms and seedlings at various stages of germination. The activities in the endosperm of resting seeds were relatively high and they remained unchanged throughout the period of reserve protein mobilization (seedling length up to 50 mm), after which they began to decrease. The activities of the naphthylamidases are rather small compared with those of the two alkaline peptidases of pine, contributing about 17% of the total amino-peptidase activity in the endosperm of germinating seeds. The total aminopeptidase activity is sufficient to account for the rate of storage protein mobilization during germination. In the seedlings the naphthylamidase activities (per seedling) increased continuously during germination, and activities per g dry weight were higher than those in the endosperm.  相似文献

Electron microscopic observations of the endosperm of tomato ( Lycopersicon esculentum Mill.) seeds revealed that changes in the cell wall structures along with the vacuolation of protein bodies occurred in the micropylar portion of the endosperm prior to germination. No changes were detected at that time in the rest of the endosperm. Endo‐β‐mannanase activity was restricted to the micropylar region of the endosperm prior to germination. Cell wall digestion by this pregerminative mannanase seemed to be associated with the changes in cell wall structures occurring in the micropylar region prior to germination. The protein content in the micropylar part of the endosperm began to decrease shortly after imbibition and attained about 40% of the initial level by the time of radicle protrusion (38 h after imbibition). On the other hand, only slight changes in the content were detected in the lateral endosperm during the same time; the protein content in the lateral endosperm decreased only after germination started. In conformity with the results on protein contents, proteolytic activity began to develop first in the micropylar portion prior to germination, and then in the lateral portion after germination. Thus, the timing of the biochemical activation of the endosperm after imbibition differed between the micropylar and the lateral region. Some qualitative differences in patterns of polypeptides synthesized in vivo were detected, as analyzed by pulse‐labeling and fluorography, between the micropylar and the lateral portions of the endosperm of seeds imbibed for 25 h. This suggests that processes of the biochemical activation of the endosperm may be qualitatively, as well as quantitatively, different depending on the regions of the endosperm.  相似文献

Summary Cotyledonary Quercus robur L. somatic embryos from two cell lines were encapsulated in 4% (w/v) sodium alginate. An artificial endosperm was provided by the addition of P24 medium plus 3% (w/v) sucrose. Oak somatic embryos and oak synthetic seeds were germinated on P24 medium plus 0.1 μM indole-3-butyric acid and 0.9 μM 6-benzylaminopurine or were dehydrated prior to germination. The highest conversion rates (26%) were obtained with encapsulated somatic embryos as well as artificial endosperm-coated somatic embryos. Encapsulation improved the regeneration into oak plantlets in one of the two cell lines tested. The artificial endosperm had no additional beneficial effect on conversion frequency, but increased germination rate in one cell line tested. Significant higher conversion could be attributed to slow desiccation compared to the non-encapsulated control. Cold storage as a post-maturation treatment had no influence on the germination ability of oak synthetic seeds. Differences in the response of the cell lines with respect to conversion frequencies and timing of germination were observed. Fifty-six well-developed plantlets regenerated 12 wk after germination, and 29 plants were transferred to the greenhouse, where they have been successfully established in substrate.  相似文献

Several isoforms of endo-1,4-D-mannanase (EC3.2.1.78) are produced in the endosperm and embryo of tomato (Lycopersicon esculentum Mill.) seed prior to the completion of germination. Other isoforms appear in the embryo and in the lateral endosperm following germination. This occurs in seeds removed from the fruit prior to completion of development at 45 d after pollination and placed directly on water, or following drying. Hence desiccation is not required to induce either germination- or post-germination-related mannanase activity. Incubating seeds in abscisic acid or osmoticum results in a reduction of both germination and total mannanase activity, but the isoforms that are produced in the embryo and micropylar region of the endosperm are identical to those produced in water-imbibed seeds prior to germination. Incubation of seeds in a high concentration of abscisic acid prevents all enzyme production. Only after the completion of germination does mannanase increase in the lateral regions of the endosperm. In contrast, mannanase is produced in the micropylar region regardless of whether the seed germinates or not. The isoforms produced in the two regions of the endosperm are different, those in the lateral endosperm being more similar to those produced in the cotyledons and axes of the embryo. The mechanism of seed dormancy at low temperatures (15-9°C) was investigated in the seeds of Syringa josikaea, S. reflexa and S. vulgaris. Low temperature dormancy in Syringa species was mainly imposed by endosperm embedding the radicle. Different degrees of embryo dormancy may occur in S. reflexa seeds. In most cases the low temperature dormancy was broken completely by removing the endosperm around the radicle. The endosperm did not seem to contain significant quantities of germination inhibitors, and the results indicate that it prevents germination mainly due to its mechanical resistance. The mechanical resistance of endosperm did not change during chilling or during induction of dormancy by high temperature incubation. The strength of the endosperm decreased rapidly in non-dormant seeds before visible germination. Similar changes were not observed in dormant seeds. Generally, the strength of the endosperm was lower in the non- (or less) dormant species S. josikaea and S. vulgaris than in the more dormant S. reflexa seeds. The growth potential of the embryos, measured as their ability to germinate in osmotic solutions (mannitol or polyethylen glycol 4000), was increased by chilling and by GA3-treatment. The growth potential of untreated S. josikaea and S. vulgaris embryos was generally higher than that of S. reflexa embryos. Acid ethyl acetate fractions of methanol extracts from embryos of all three species contained substances with GA3-like activity in the lettuce hypocotyl test. The activity was found at Rf 0.9–1.0 on paper chromatograms run in distilled water. No significant changes in the activity were detected during chilling or prior to visible germination.  相似文献

利用常规石蜡制片技术对北柴胡胚和胚乳的发育及对其种子萌发的影响进行了观察。结果表明北柴胡胚的发育属于茄型,基细胞进行一次横分裂后不再分裂,因而胚柄不发达,且很早解体。胚乳的发育属于核型,初生胚孔核的分裂远远早于受精卵的分裂。对果实采收时期胚发育状况进行统计发现,在被测采收期果实中有20%的果实的胚处于球形胚阶段,70%处于心形胚,只有10% 处于鱼雷胚,说明北柴胡种子采收时胚处于不同的发育阶段,存在形态后熟现象,这是北柴胡种子萌发难、萌发率低且出苗不整齐的主要因素。  相似文献

In higher plants, mannans are found as dominant reserve material in the endosperm of Arecaceae seeds and also in some species from Apiaceae, Rubiaceae and Asteraceae. A linear β(1 → 4)-d-mannan was now isolated from the endosperm of Schizolobium parahybae, family Caesalpiniaceae, a native of Southern Brazil. Its seeds were germinated and the consumption of polysaccharides from the endosperm, namely galactomannans and β(1 → 4)-d-mannan, was analysed at differents stages of germination. At the 6th day after germination no residual 3:1 Man:Gal galactomannan was found, indicating that complete degradation of galactomannan had been reached. However, after 12 days of germination, the mannan was recovered from the remaining endosperm. Its presence in the endosperm after germination demonstrated that it is not a reserve material as described for seeds of other species.  相似文献

Salicylhydroxamic acid (SHAM) stimulated germination of photosensitive lettuce (Lactuca sativa L. cv Waldmann's Green) seeds in darkness. To determine whether SHAM acts on the embryo or the endosperm, we investigated separately effects of SHAM on growth potential of isolated embryos as well as on endosperm strength. Embryo growth potential was quantified by incubating decoated embryos in various concentrations of osmoticum and measuring subsequent radicle elongation. Growth potential of embryos isolated from seeds pretreated with 4 millimolar SHAM was equal to that of untreated controls. Rupture strength of endosperm tissue excised from seeds pretreated with SHAM was 33% less than that of controls in the micropylar region. To determine if the embryo must be in contact with the endosperm for SHAM to weaken the endosperm, some endosperms were incubated with SHAM only after dissection from seeds. Rupture strength of SHAM-treated, isolated endosperms in the micropylar region was 25% less than that of untreated controls. There was no difference in rupture strength in the cotyledonary region of endosperm isolated from seeds treated with SHAM in buffer or buffer alone. SHAM therefore stimulates germination not by enhancing embryo growth potential, but by weakening the micropylar region of the endosperm enclosing the embryo.  相似文献

Decoated pepper (Capsicum annuum L. cv Early Calwonder) seeds germinated earlier at 25°C, but not at 15°C, compared to coated seeds. The seed coat did not appear to impose a mechanical restriction on pepper seed germination. Scarification of the endosperm material directly in front of the radicle reduced the time to germination at both 15°C and 25°C.

The amount of mechanical resistance imposed by the endosperm on radicle emergence before germination was measured using the Instron Universal Testing Machine. Endosperm strength decreased as imbibition time increased. The puncture force decreased faster when seeds were imbibed at 25°C than at 15°C. The reduction in puncture force corresponded with the ability of pepper seeds to germinate. Most radicle emergence occurred at 15°C and 25°C after the puncture force was reduced to between 0.3 and 0.4 newtons.

Application of gibberellic acid4+7 (100 microliters per liter) resulted in earlier germination at 15°C and 25°C and decreased endosperm strength sooner than in untreated seeds. Similarly, high O2 concentrations had similar effects on germination earliness and endosperm strength decline as did gibberellic acid4+7, but only at 25°C. At 15°C, high O2 concentrations slowed germination and endosperm strength decline.

The effect of various abiotic stresses on germination rate, growth and soluble sugar content in Sorghum bicolor (L.) Moench cv. CSH 6 seed embryos and endosperm during early germination was investigated. Under stress conditions germination, water potential and tissue water content decreased markedly. Subsequently, this reduction resulted in marked decreases in fresh weight both in embryos and endosperm. Conversely, a substantial increase in dry weight was observed. Furthermore, a considerable increase in the sugar contents in both embryo and endosperm was detected. The fructose level was always higher than glucose and sucrose in response to various stresses. However, as compared to the control the level of glucose and sucrose was higher in embryos and endosperm after stress treatments. Based upon these results a possible physiological role of sugars in the germination of sorghum seeds is discussed.  相似文献

A lot of pepper seeds having 87 % germination were subjected to X-ray inspection using a non lethal dose of radiation. Seeds with less than 2.7 % (on the basis of total seed area) of free space area, i.e. the spaces between embryo and endosperm, were classified as highly viable seeds (97–100 % germination) with the lowest level of abnormal seedlings. Seeds X-ray classified as good were subjected to a computerised image analysis to study seed imbibition and radicle elongation. The patterns of seed area increase, chosen as the most accurate indicator of seed swelling, resembled the triphasic curve of water uptake. The first phase was completed at 9 h followed by a second phase that varied widely in time until completion of germination between 52 and 96 h. The proportion of seeds with radicle protrusion between 52–56 h and 64–72 h assessed with the image analysis was significantly higher than that recorded using a conventional germination test. In addition, the rate of increase of seed area during the third phase of imbibition, mostly due to protrusion of the radicle tip and its growth, was highly correlated with the corresponding radicle elongation rate.  相似文献

东北刺人参(Oplopanax elatus Nakai)种子透水性良好,休眠后萌发不受其影响.种皮和胚乳的水提取物中存在萌发抑制物质,胚乳中提取物对白菜种子萌发的抑制效果比种皮更明显.种子自然脱落时胚尚未分化完全,处于心形胚阶段.种子需要先温暖层积以完成胚的分化与生长,然后转入低温层积完成生理后熟.同批种子胚的发育不完全同步,变温层积处理7个月有极少数种子萌发,连续变温层积处理17个月大部分种子萌发.不同年份受气候条件影响,种子产量和发芽率差异较大.种子耐贮性较强,贮藏2年的种子生活力变化不大,仍具有较高的萌发潜力.  相似文献

ABSTRACT

Endosperm weakening is a key event for completion of seed germination in plants such as tomato and tobacco. Weakening is related to the action of endo-β-mannanases able to hydrolyse the mannose polymers typically stored in the wall of the endosperm cells. In this study, we determined the presence and the localisation of endo-β-mannanases in Phacelia tanacetifolia seeds during the early phases of germination. In endosperm cells of dry seeds, and of seeds incubated in the light for 16 h, a similar distribution of endo-β-mannanases, mainly localised in protein bodies, was revealed by immunolocalization. In contrast, under conditions of permissive germination (seeds incubated for 16 h in the dark), these enzymes appeared localised near the cell walls, and were no longer detectable in protein bodies. Western blot analyses showed the presence of three isoforms of endo-β-mannanases in the endosperm and one isoform in the embryo. All these isoforms had similar molecular weights (approx 38 kDa). A possible role of endo-β-mannanases during early phases of germination is suggested.  相似文献

A current hypothesis is that endo--mannanase activity in the endosperm cap of tomato (Lycopersicon esculentum Mill. cv. Moneymaker) seeds is induced by gibberellin (GA) and weakens the endosperm cap thus permitting radicle protrusion. We have tested this hypothesis. In isolated parts, the expression of endo--mannanase in the endosperm after germination is induced by GAs, but the expression of endo--mannanase in the endosperm cap prior to radicle protrusion is not induced by GAs. Also, abscisic acid (ABA) is incapable of inhibiting endo--mannanase activity in the endosperm cap, even though it strongly inhibits germination. However, ABA does inhibit enzyme activity in the endosperm and embryo after germination. There are several isoforms in the endosperm cap and embryo prior to radicle protrusion that are tissue-specific. Tissue prints showed that enzyme activity in the embryo spreads from the radicle tip to the cotyledons with time after the start of imbibition. The isoform and developmental patterns of enzyme activity on tissueprints are unaffected when seeds are incubated in ABA, even though germination is inhibited.
