Reproductive isolation among biological strains can be detected by analyzing reproductive barriers between populations. The Anastrepha fraterculus Wiedemann (Diptera: Tephritidae) cryptic species complex comprises eight morphotypes defined by morphometric characteristics. The present study analyzed the existence of pre‐ and post‐zygotic isolation among populations of A. sp.1 aff. fraterculus (A. sp.1) and A. sp.3 aff. fraterculus (A. sp.3) determined by tests of sexual compatibility, analyses of the temporal pattern of calling behavior, and the reproductive performance of the offspring of homotypic and heterotypic crosses. Pre‐zygotic tests indicated asymmetric matings with preferences for homotypic crosses. The A. sp.3 population demonstrated distinct characteristics, including low copulation percentages and a high proportion of homotypic copulations under crowded conditions. The females of the A. sp.1 population demonstrated lower copulation latency. The analyzed morphotypes demonstrated two peaks in pheromone emission activity, at the beginning and at the end of the morning and were differentiated in the abundance of males. The calling behavior of the hybrids between A. sp.1 male and A. sp.3 female crosses had an intermediate pattern compared to either parental type. Post‐zygotic compatibility tests indicated low viability of the cross between A. sp.3 males and A. sp.1 females. Pronounced asymmetries were found in the sex ratios of the offspring of heterotypic crosses, and only hybrids from A. sp.1 male vs. A. sp.3 female crosses produced descendants. The observed pre‐ and post‐zygotic incompatibilities reflect the probable occurrence of incipient speciation between A. sp.1 and A. sp.3. A more adequate taxonomic classification of this species complex that considers the distinct characteristics of each morphotype will be needed to improve environmentally wise control methods against this insect pest.  相似文献

Glomerella cmgulata is a homothallic species but produces a ridge of fertile perithecia at a frontier between certain wild-type strains on agar. To account for the presence or absence of perithecia earlier workers suggested that alleles at A and B loci control the formation of perithecia at mycelial frontiers in + and – strains. We propose that G. cingulata actually demonstrates “relative heterothallism”. Of 7 induced nutritionally deficient mutants (auxotrophs) in 2 wild-type strains from apple, only one methionine (met-1) and one arginine (arg) mutant in only one wild-type strain gave a heavy ridge of perithecia at their junctures. Neither the met-l nor arg mutations have been identified as those in the A or B locus. The perithecia were either homozygous (selfs) for met-1 or arg, or heterozygous (hybrids). Paired met-1 and arg segregants from hybrid perirhecia as well as diauxotrophic strains from met-l or arg mutants also gave hybrids of selfs. Specific nutritional deficiencies in certain wild-type strains which can direct sexuality are not yet known. Genetic studies are now feasible in G. cingulata to define enzymatic factors responsible for pathogenicity.  相似文献

Biochemical and genetic studies on the arginine-requiring auxotrophs derived from a Serratia marcescens strain were carried out. The arg mutants were classified into seven biochemical groups based on their growth response to five precursors of arginine biosynthesis and enzyme deficiency. Reciprocal transduction tests among those arg mutants divided them into three linkage groups, and the fine mapping in each of the groups by two- or three-point crosses revealed the following arrangement of loci. (1) arg44–thy11–lys1; (2) met1–glt2–argE–(arg19–arg51)–arg120–argG–argH; (3) arg33–pyr4. Five of the seven biochemically distinct arg mutants belonged to the second linkage group, and they constituted an arg-gene cluster. A characteristic feature of the arg-gene cluster of S. marcescens is that it involves argG, which was previously reported only in the Proteus group of Enterobacteriaceae.  相似文献

The two tropical estuarine dinoflagellates, Alexandrium tamiyavanichii Balech and A. minutum Halim, were used to determine the ecophysiological adaptations in relation to their temperate counterparts. These species are the two main causative organisms responsible for the incidence of paralytic shellfish poisoning (PSP) in Southeast Asia. The effects of light (10, 40, 60, and 100 μmol photons·m?2·s?1) and temperature (15, 20, and 25°C) on the growth, nitrate assimilation, and PST production of these species were investigated in clonal batch cultures over the growth cycle. The growth rates of A. tamiyavanichii and A. minutum increased with increasing temperature and irradiance. The growth of A. tamiyavanichii was depressed at lower temperature (20°C) and irradiance (40 μmol photons·m?2·s?1). Both species showed no net growth at 10 μmol photons·m?2·s?1 and a temperature of 15°C, although cells remained alive. Cellular toxin quotas (Qt) of A. tamiyavanichii and A. minutum varied in the range of 60–180 and 10–42 fmol PST·cell?1, respectively. Toxin production rate, Rtox, increased with elevated light at both 20 and 25°C, with a pronounced effect observed at exponential phase in both species (A. tamiyavanichii, r2=0.95; A. minutum, r2=0.96). Toxin production rate also increased significantly with elevated temperature (P<0.05) for both species examined. We suggest that the ecotypic variations in growth adaptations and toxin production of these Malaysian strains may reveal a unique physiological adaptation of tropical Alexandrium species.  相似文献

Wide crosses have been used for decades as a method for transferring novel genetic material and traits in plant breeding. Historically, many products of wide crosses require tedious and inefficient surgical embryo rescue prior to embryo abortion to recover single plantlets. We have utilized transgenic switchgrass (Panicum virgatum L. cv Alamo) as a pollen donor in conjunction with antibiotic or herbicide selection for recovery of intra‐and interspecific F1 crosses by using developing ovules from the female parent and selecting for embryogenic cultures derived from the in situ immature embryo. Using this approach, several intravarietial crosses were generated between transgenic Alamo and the switchgrass varieties Kanlow, Blackwell and Cave ‐ in ‐ Rock as well as an interspecific cross with Atlantic coastal panicgrass. This procedure selected F1 embryogenic callus produced from the developing embryo contained within isolated immature ovules. Several clonal plants were successfully regenerated from each cross. Southern blot, PCR, phenotypic analyses and genomic analysis confirmed F1 hybrids. Using genotyping‐by‐sequencing shows the hybridization of the recovered plants by determining the ratio of transgressive markers to total compared markers between parents and their potential offspring. The ratio of transgressive markers to total compared markers was significantly lower between parents and their predicted offspring than between parents and offspring unrelated to them. This approach provides the possibility to move useful transgenes into varieties that are recalcitrant to direct transformation which can be optionally segregated thus useful to create new hybrids, as well as recovery of wide crosses that are either difficult or impossible using traditional techniques.  相似文献

Summary Primary and secondary spore clones were analyzed from two- and three-factor crosses involving the mitochondrial markers conferring resistance to antimycin (A R ), chloramphenicol (C R ), and erythromycin (E R ). As in zygote clones (Seitz-Mayr et al., 1978), transmission of markers is higher in two-factor trans-crosses than in cis-crosses. Except transmission of C R in the cross A R C R E R xA S C S E S , no significant differences between cis- and trans-configuration were observed in three-factor crosses. In contrast to zygote clones, in spore clones transmission rates of the two or three markers in a given cross are roughly equal. 18 out of 20 secondary spore clones of different mitochondrial phenotypes appeared to be homoplasmic, whereas 2 still continued to segregate. One of these spore isolates was analyzed, and segregation was found to continue for more than 150 generations after spore germination. Whereas up to more than 80% of zygote clones in certain crosses were uniform, only 2 out of 91 tetrads were uniform, i.e. all four spores were homoplasmic for the same mitochondrial genotype. Presence or absence of recombinant mitochondrial phenytypes among secondary spore clones from tetrads indicated, whether, cytoplasmic mixing had occurred in the original zygote or not. Within an ascus, the number of spores containing recombinant genotype(s) is a direct measure for the extent of cytoplasmic mixing in the zygote. In 82 tetrads analyzed, the number of tetrads with 0, 1, 2, 3, and 4 spores containing recombinant genotype(s) were 25, 37, 14, 5, and 1, respectively. In conclusion, the extent of cytoplasmic mixing at the cell stage before forespore membrane formation is highly variable.  相似文献

Summary In addition to the generec-4, other genetic factors affect the frequency of allelic recombination in thehis-3 locus. One dominant factor, designated asrec-6 +, in association withrec-4 + causes greater reduction in prototrophic frequency than obtained withrec-4 + alone. The action ofrec 6 + in crosses recessive homozygous forrec-4 is not established at the present. The effect ofrec-6 + is recognised only with onehis-3 allele but not with another. Interaction ofrec-4 + orrec-4 with other genetic factors can give approximately ten fold variation in the prototrophic frequencies obtained with a pair of alleles. It is suggested that the control of the rate of mutations during meiosis might be one of the roles of the recombination genes.  相似文献

Batch cultures and continuous flow cultures were used to study the growth rates of zooplankton species from Shira lake, the rotifer Brachionus plicatilis Muller and calanoid copepod Arctodiaptomus salinus Daday, which were fed on phytoplankton and bacterioplankton from the lake. Analyses of the birth and survival rates were used to demonstrate that the lake phytoplankton, consisting mostly of cyanobacteria and diatomaceous algae, is inadequotes for optimal realisation of the reproductive potential of B. plicatilis when compared with the bacterial diet. The study revealed that the kinetic growth characteristics of the two zooplankters were similar: B. plicatilis r max, 0.120 d–1; S 0, 0.253; and K s, 0.114 mg dry mass l–1; and for A. salinus r max, 0.129 d–1; S 0, 0.240; and K s, 0.171 mg dry mass l–1. Fluctuations in natural food concentration reduced the growth rate of both species. Even though the threshold concentration of food for B. plicatilis and A. salinus were quite similar, the copepods were less sensitive to food limitation.  相似文献

Escherichia coli (2492/pJB4JI) matings with Erwinia chrysanthemi produced kanamycin resistant (Kmr) transconjugants, a majority of which were gentamicin sensitive (Gms). A small proportion (about 0.8%) of the Kmr Gms clones were either auxotrophic or failed to catabolize galacturonate (Gtu). The R plasmid (pJB4JI) DNA was detected in the parent E. coli strain and in a Kmr Gmr transconjugant, but not in Kmr GmsE. chrysanthemi strains carrying Tn5-induced mutations. In Hfr crosses, Kmr (Tn5) was found linked with most mutations. A majority (>95%) of prototrophic recombinants were Kms, except for Leu+ and Arg+ recombinants which were 30 to 50% Kms. Spontaneous revertants were obtained for all markers except car, gtu, lys, thr, and trp. Prototrophic revertants, with the exception of Met+, Leu+, or His+ clones, were Kms. We conclude from both genetic and physical data that Tn5 transposed from pJB4JI into different sites on the chromosome of E. chrysanthemi.  相似文献

The ability of hyporheic sediments to exchange water and retain ammonium and phosphate in the Riera Major stream ,North-East Spain, under different discharge conditions was measured by conducting short-term nutrient and chloride additions. 2. The mean exchange coefficients from free-flowing water to the storage zone (k1) and vice versa (k2) were 0.82 × 10–4 s??1 and 7 × 10??3 s??1, respectively. The ratio of storage zone cross-sectional area to stream cross-sectional area (AS/A) averaged 2.8 × 10–2 and was negatively correlated with discharge (r = –0.85, d.f. = 13, P < 0.001). 3. The percentage of hyporheic zone water which came from surface water varied as a function of discharge and hyporheic depth, ranging between 33% and 95% at 25 cm depth, and between 78% and 100% at 10 cm depth. 4. The nutrient retention efficiency in the hyporheic zone at 10 cm depth measured as uptake length (Swh) was less than 3.3 cm for ammonium and 37 cm for phosphate. Higher nutrient retentions were measured in the sediments at 10 cm depth than at 25 cm, indicating that near-surface sediments were involved more actively in phosphate retention than the deeper hyporheic sediments. The lack of ammonium at any depth of the hyporheic zone showed that ammonium was very rapidly taken up in the surfacial sediments.  相似文献

Summary Crosses were performed between homokaryons of Agrocybe aegerita having the same allele at the A incompatibility gene but different B alleles. Heterokaryotic mycelia originating from crosses between two complementary auxotrophs were characterized by their instability on complete medium and extensive anastomosis between hyphae. Diploid mycelia were selected by plating oidia recovered from these heterokaryons onto minimal medium. These mycelia were characterized by the production of larger oidia than those of homokaryons, the release of a brown pigment when growing on complete medium and extensive hyphal anastomoses. Diploids retained the two B incompatibility functions of their homokaryotic parents and gave rise to a diploid/haploid dikaryon when crossed with a compatible homokaryon. Nearly 1% of the oidia recovered from heterokaryons were diploid. These nuclear fusion frequencies as well as the production of brown pigments enabled the identification of diploid strains on complete medium. In this way, crosses between wild prototrophic strains were successfully performed. Somatic recombination was induced following the treatment of diploid mycelia with haploidizing compounds. Selection based on the inability of mycelia to produce the brown pigments on complete medium led to selection of strains homoallelic at the B locus.  相似文献

In hybrid maize (Zea mays L.) breeding, doubled haploids (DH) are increasingly replacing inbreds developed by recurrent selfing. Doubled haploids may be developed directly from S0 plants in the parental cross or via S1 families. In both these breeding schemes, we examined 2 two-stage selecting strategies, i.e., considering or ignoring cross and family structure while selection among and within parental crosses and S1 families. We examined the optimum allocation of resources to maximize the selection gain ΔG and the probability P(q) of identifying the q% best genotypes. Our specific objectives were to (1) determine the optimum number and size of crosses and S1 families, as well as the optimum number of test environments and (2) identify the superior selection strategy. Selection was based on the evaluation of testcross progenies of (1) DH lines in both stages (DHTC) and (2) S1 families in the first stage and of DH lines within S1 families in the second stage (S1TC-DHTC) with uniform and variable sizes of crosses and S1 families. We developed and employed simulation programs for selection with variable sizes of crosses and S1 families within crosses. The breeding schemes and selection strategies showed similar relative efficiency for both optimization criteria ΔG and P (0.1%). As compared with DHTC, S1TC-DHTC had larger ΔG and P (0.1%), but a higher standard deviation of ΔG. The superiority of S1TC-DHTC was increased when the selection was done among all DH lines ignoring their cross and family structure and using variable sizes of crosses and S1 families. In DHTC, the best selection strategy was to ignore cross structures and use uniform size of crosses.  相似文献

Summary The influence of variations in the boundary air layer thickness on transpirtion due to changes in leaf dimension or wind speed was evaluated at a given stomatal resistance (r s) for various combinations of air temperature (T a) and total absorbed solar energy expressed as a fraction of full sunlight (S ffs). Predicted transpiration was found to either increase or decrease for increases in leaf size depending on specific combinations of T a, S ffs, and r s. Major reductions in simulated transpiration with increasing leaf size occurred for shaded, highly reflective, or specially oriented leaves (S ffs=0.1) at relatively high T a when r s was below a critical value of near 500 s m-1. Increases in S ffs and decreases in T a lowered this critical resistance to below 50 s m-1 for S ffs=0.7 and T a=20°C. In contrast, when r s was above this critical value, an increase in leaf dimension (or less wind) resulted in increases in transpiration, especially at high T a and S ffs. For several combinations of T a, S ffs, and r s, transpiration was minimal for a specific leaf size. These theoretical results were compared to field measurements on common desert, alpine, and subalpine plants to evaluate the possible interactions of leaf and environmental parameters that may serve to reduce transpiration in xeric habitats.  相似文献

In the sexual reproduction of the green alga Closterium ehrenbergii, two sexually competent cells that are morphologically indistinguishable from the vegetative cells first come close to each other to form a sexually interacting pair. Each then divides into two gametangial cells. Isogamous conjugation occurs between nonsister gametangial cells of the two resulting pairs. With unusual selfing clones derived from a certain cross of heterothallic strains, we dissected apart a pair of gametangial cells that had already been united together by a delicate transparent tube, into which each gametangial cell was going to develop its conjugation papilla. In spite of such a degree of differentiation, when each was cultured in fresh medium, individual gametangial cells could dedifferentiate into vegetative cells and form subclones. By crossing such subclones with standard stable heterothallic mating-type strains, we show that each selfing clone of this alga actually produces both stable mt + and stable mt - cells, in addition to unstable mt - cells with selfing potency, during its mitotic vegetative growth. Although the selfing in C. ehrenbergii studied here differs in certain points from true homothallism, the results of the present study provide insight into how homothallism might have evolved from heterothallism.  相似文献

We quantified metabolic power consumption as a function of wind speed in the presence and absence of simulated solar radiation in rock squirrels, Spermophilus variegatus, a diurnal rodent inhabiting arid regions of Mexico and the western United States. In the absence of solar radiation, metabolic rate increased 2.2-fold as wind speed increased from 0.25 to 4.0 m·s-1. Whole-body thermal resistance declined 56% as wind speed increased over this range, indicating that body insulation in this species is much more sensitive to wind disruption than in other mammals. In the presence of 950 W·m-2 simulated solar radiation, metabolic rate increased 2.3-fold as wind speed was elevated from 0.25 to 4.0 m·s-1. Solar heat gain, calculated as the reduction in metabolic heat production associated with the addition of solar radiation, increased with wind speed from 1.26 mW·g-1 at 0.25 m·s-1 to 2.92 mW·g-1 at 4.0 m·s-1. This increase is opposite to theoretical expectations. The over-expression of Arabidopsis CAX1 and CAX2 causes transgenic tomato plants to reveal severe Ca2+ deficiency-like symptoms such as tip-burn and/or blossom end rot, despite there being sufficient Ca2+ in each plant part. To correct the symptoms and to moderately enhance the calcium level, a worldwide vegetable tomato was genetically engineered using a modified Arabidopsis cation/H+ antiporter sCAX2A, a mutant form of Arabidopsis CAX2. Compared with the wild-type, the sCAX2A-expressing tomato plants demonstrated elevated Ca2+ levels in the fruits with almost no changes in the levels of Mn2+, Cu2+, and Fe2+. Moreover, expression of sCAX2A in tomato plants did not show any significant alterations in their morphological phenotypes. Unlike 35S::sCAX1 construct, sCAX2A antiporter gene driven by 35S promoter can be a valuable tool for enriching Ca2+ contents in the tomato fruit without additional accumulation of the undesirable cations.  相似文献

Summary Streptomyces lividans arginine auxotrophs which show amplification of a 5.7-kb DNA sequence, arose at a very high frequency, varying from 10% to 25% of Cmls spores. The amplifiable DNA sequence was shown to be stable over many generations. However, treatment of Cmls arg mutants with subinhibitory concentrations of antibiotics such as spectinomycin, streptomycin, chloramphenicol, thiostrepton and kanamycin, either during sporulation or during vegetative growth of mycelia, led to the deletion of the entire amplified DNA sequence, including the left and right junction sequences. Depending upon the method of antibiotic treatment a reduction in the copy number of the amplified DNA was also observed. This reduction in copy number apparently occurred without drastically affecting the basic structure of the amplifiable unit of DNA. This phenomenon appears to be universal since deamplification and deletion were observed also in S. fradiae. Further, spontaneous arg mutants arose at much lower frequency from spectinomycin-pretreated Cmls cells compared to untreated cells. These arg mutants isolated in the presence of spectinomycin did not show amplification of the 5.7-kb sequence. Southern blot analysis using the 5.7-kb probe showed that the entire DNA sequence homologous to the amplifiable DNA sequence had been deleted. Offprint requests to: K. Dharmalingam  相似文献

To study meiosis, synchronous cultures are often indispensable, especially for physical analyses of DNA and proteins. A temperature-sensitive allele of the Pat1 protein kinase (pat1-114) has been widely used to induce synchronous meiosis in the fission yeast Schizosaccharomyces pombe, but pat1-114-induced meiosis differs from wild-type meiosis, and some of these abnormalities might be due to higher temperature needed to inactivate the Pat1 kinase. Here, we report an ATP analog-sensitive allele of Pat1 [Pat1(L95A), designated pat1-as2] that can be used to generate synchronous meiotic cultures at physiological temperature. In pat1-as2 meiosis, chromosomes segregate with higher fidelity, and spore viability is higher than in pat1-114 meiosis, although recombination is lower by a factor of 2–3 in these mutants than in starvation-induced pat1+ meiosis. Addition of the mat-Pc gene improved chromosome segregation and spore viability to nearly the level of starvation-induced meiosis. We conclude that pat1-as2 mat-Pc cells offer synchronous meiosis with most tested properties similar to those of wild-type meiosis.  相似文献

Relationship between heterozygosity at allozyme loci, chromosomal interchanges and fitness was analyzed in a rye cultivar showing a polymorphism for such rearrangements. Nine allozyme systems (ACO, ACPH, GOT, GPI, LAP, MDH, PER, PGD and PGM) and five components of fitness (number of fertile tillers, total offspring, egg cell fertility, flowers/ear and seeds/ear) were studied. The estimated selection coefficients against interchange heterozygotes ranged from s = 0.12 to s = 0.34. A significant effect of the genic heterozygosity on some fitness components was observed in interchange heterozygotes (tillering and total offspring), in their standard homozygous sibs (flowers/ear and seeds/ear) and in the descendants of the crosses between standard karyotypes (flowers/ear, seeds/ear and egg cell fertility). However, the main effect was linked to genetic background associated to different crosses. Significant differences for Acph-1, Gpi-1, Lap-1, Mdh-1, Mdh-4, Pgd-2 and Pgm-1 loci were also found in some of these crosses although these differences were inconsistent. This suggests that probably the allozyme loci analyzed were not directly contributing to the fitness and that they are linked, in some cases, to different deleterious alleles depending on both cross and locus. This fact could support the local effect hypothesis as explanation although we do not discard the existence of some inbreeding level (general effect hypothesis) since all crosses and loci studied show a overall consistent trend of increased fitness with increased heterozygosity.  相似文献
