The in vitro growth rates under continuous light of the four dominant blue-green algae in Lough Neagh, Anabaena flos-aquae Bréb., Aphanizomenon flos-aquae Ralfs fa. gracile Lemm., Oscillatoria agardhii Gom. and Oscillatoria redekei van Goor were slower than in situ rates from Lough Neagh that had been corrected for hours of light received by the algae. However, by culturing on a 6: 18 light-dark cycle in vitro growth rates were obtained that were similar to the in situ rates. Under continuous light small species showed the fastest growth with Oscillatoria redekei the dominant species. However, this pattern was almost completely reversed under the light-dark cycle with Oscillatoria redekei only exhibiting the fastest growth rate under low light conditions. This observation showed agreement with Lough Neagh field data which showed that Oscillatoria redekei reached its maximum crop in April while the other three species were dominant during the summer months. Compared to the generally assumed high thermal tendency of blue-green algae the temperature maxima of the four species were low. No growth was observed at 35°C for any species while Anabaena flos-aquae was severely inhibited at 25°C.  相似文献

The cyanobacterium Oscillatoria brevis (Kütz.) Gom., strain NIVA CYA 7, was used to investigate how geosmin production is related to the synthesis of chlorophyll a, phycobiliproteins and β-carotene under nitrogen (NH 4 + ) and light limiting conditions. Chemostat samples were used to inoculate batch cultures that were treated with inhibitors of isoprenoid synthesis, norflurazon and dimethazone, and gabaculine that inhibits tetrapyrrole synthesis. Dimethazone decreased and norflurazon increased geosmin production under light limited conditions, as was expected due to their sites of action in the isoprenoid pathway. This effect was not so pronounced in nitrogen limited cultures due to the additional effect of increasing nitrogen deficiency during the experimental period. Norflurazon was the only inhibitor that uncoupled geosmin production completely from β-carotene formation which indicates a strikt coupling between geosmin and β-carotene biosynthesis. From the observed increase of geosmin production relative to pigment synthesis after norflurazon treatment it was suggested that isoprenoid precursors are directed to geosmin synthesis when the demand for pigment precursors is very low. Within the framework of this study the data strongly support the hypothesis of geosmin formation via the isoprenoid pathway in Oscillatoria brevis as was found for actinomycetes. Populations of Limnothrix redekei, Oscillatoria lanceaeformis, Planktothrix agardhii and Pseudanabaena limnetica were found in a hypertrophic, gravel-pit lake near Madrid (Spain), throughout a one year sampling at weekly intervals. Physico-chemical factors, phytoplankton biomass and net growth rates were measured. Oscillatoria lanceaeformis was only observed a few weeks, probably being related to phosphorus limitation. Planktothrix agardhii biomass was related to PhAR irradiances, light attenuation coefficient in the water, Brunt-Väisäla frequency and decrease of soluble reactive phosphorus. Limnothrix redekei and Pseudanabaena limnetica biomass values were related to a decrease of inorganic nitrogen and temperature. A different lag response of populations was observed in relation to the environmental features.  相似文献

A light and electron microscopic re-investigation of the pancreatic islet organ of the hagfish, Myxine glutinosa, has confirmed earlier observations that agranular and granular β-cells are the predominant cell types. However, in addition a rare second granular parenchymal cell was observed in which the granules are spherical in shape with a mean diameter of 213 nm, contrasting with the predominantly ellipsoid granules which characterise the β-cell. At present, this allegedly second granular parenchymal cell cannot be unequivocally equated with a named islet cell type and its nature remains somewhat problematic.  相似文献

Effect of light intensity on macromolecular synthesis in cyanobacteria   总被引:3,自引:0,他引:3  
The light-dependent incorporation of NaH14CO3 into low molecular weight compounds, polysaccharide, or protein was determined in cultures of the cyanobacteriumMerismopedia tenuissima incubated at a series of light intensities. There was an inverse relationship between incorporation into polysaccharide and protein. At light intensities of 90 E/m2/sec or above, relative incorporation of radioisotope into polysaccharide was greatest and relative incorporation into protein was lowest. Optimal relative protein accumulation occurred in samples incubated at 20 E/m2/sec. A broader optimum of light intensity for maximal protein accumulation was found if ammonia rather than nitrate was the nitrogen source. Physiological adaptation of cultures to growth at a particular light intensity did not alter the pattern of macromolecular incorporation when those cultures were tested over the series of light intensities. The response of cultures ofOscillatoria rubescens to light intensity was similar to that ofM. tenuissima, although incorporation into low molecular weight compounds was significantly greater.The effect of light intensity on macromolecular synthesis in a natural population ofOscillatoria rubescens was also determined. A pattern similar to that observed in batch cultures ofO. rubescens was occasionally found, but in other experiments there was no increase in relative protein incorporation when light intensity was decreased.  相似文献

The rate of C14O2 uptake in a bacterized isolate of Oscillatoria rubescens indicates that within the ranges studied temperature caused the greatest variation followed by light intensity and nutrient concentration. The variation within interaction effects of light, temperature, and nutrients was higher than that within any other combination of interactions. High temperatures (25°C) shifted the light optimum of O. rubescens growing in low to moderate nutrient levels from 1950 lux to 800 lux.  相似文献

Oscillatoria terebriformis, occurring from 54 to about 35 C in eastern Oregon hot springs, forms dense reddish-brown surface mats which are held together by the interwoven nature of the trichomes and not by a gel-like matrix, thus retaining the flexibility inherent in free trichome motility. The trichomes demonstrate gliding, flexional, and contractile movements. When the trichomes are interconnected, the mat contracts or retracts essentially as a unit in response to high light intensity (ca. 100 cm/hr) or supraoptimal temperature (ca. 2 cm/hr). Under lower light intensity and optimal temperature, the mat expands by the gliding movements of individual trichomes (1-3 cm/hr). Probable positive and negative phototopotactic responses of trichomes to low and high light intensities, respectively, also influence the density of the surface mat. Thus, the population of Oscillatoria is able to adjust its position and density rapidly by migrations and contractions, remaining almost always within the full optimal temperature range and fairly well xuithin the optimal light conditions as well.  相似文献

以3种不同退化程度的温带典型草原(大针茅轻度退化、中度退化和重度退化)为研究对象,研究植被退化对温带典型草原土壤及根系碳氮含量及储量的影响。结果显示:(1)植被退化对地下根系碳含量影响不显著(P0.05),而对地下根系氮含量的影响显著(P0.05),中度退化样地根系氮含量显著高于轻度退化和重度退化样地(P0.05)。(2)植被退化对根系碳氮储量影响显著(P0.05),根系碳氮储量随着土层深度增加而减少,总根系碳氮储量随退化程度加剧而降低。(3)土壤有机碳、总碳和总氮含量及储量均受退化程度和采样深度的影响显著(P0.05),其含量随着土壤深度的增加而显著减少,随退化程度加剧而显著降低(P0.05)。(4)土壤是根系-土壤系统碳氮储存的最主要场所,储量占比90%以上。虽然土壤碳氮储量均存在表层聚集现象,但表层储量所占比例在各样地间差异显著(P0.05)。  相似文献

The photosynthetic activity of dominant phytoplankton in eutrophic shallow lake was investigated by the autoradiographic method in 1979 and 1980. It was shown by light and dark field microscopy that all species of Cyanophyta (Oscillatoria redekei, Oscillatoria agardhii, Aphanizomenon flos-aquae) were characterized by a continuously high uptake of NaH14CO3. Similarly high photosynthetic activity was observed during the occurrence of Cryptomonas sp. and nanoplankton. Contrary to these observations, diatoms showed remarkably high portions of photosynthetically inactive biomass when their development was abundant. The reasons for this discrepancy between high biomass of diatoms and relatively low primary production (measured by the 14C-method and autoradiography) are discussed.  相似文献

In early stages of primary succession, colonizing plants can create resource patches that influence the abundance and distribution of other species. To test whether different colonizing shrubs generate contrasting patches on coastal sand dunes, we compared soil characteristics and light availability under the nitrogen-fixing shrub Lupinus arboreus, under the non-nitrogen-fixing shrub Artemisia pycnocephala, and between shrubs on dunes at a site in northern California. Concentrations of inorganic nitrogen and net nitrogen mineralization rates were generally 1–10 times greater in soil under Lupinus than under Artemisia or between shrubs. Soil water content was mostly lower under shrubs. Mean photon flux density near ground level was reduced by at least 80% at 35 cm inside shrub canopies. Topography appeared to have more effect on soil moisture but less direct effect on nitrogen availability than did Lupinus. However, Lupinus probably increases nitrogen levels more on higher, drier dunes. Microhabitats under and between nitrogen-fixing shrubs constitute a mosaic of individually poor but complementary patches in which high levels of light and moderate levels of soil nitrogen are present but tend not to occur together.  相似文献

Summary In a human medullary carcinoma of thyroid gland containing calcitonin in light microscopic demonstration by the avidin biotin complex (ABC) method characteristic secretory granules were found electron microscopically in the cytoplasm of the tumour cells. They consisted in so-called type I granules (270±25 nm) and type II granules (135±17 nm). By the immuno gold staining (IGS) method the content of many secretory granules measuring 85–270 nm (152±18 nm) in diameter could be identified as calcitonin. These granules seemed to be predominantly of type II because of their nearly corresponding size and feature. The type I grnaules were less frequent in number and they showed no or little immunoreactivity. The results indicate that the IGS-method is practicable to demonstrate the ultrastructural localization of calcitonin and to identify clearly the nature of intracytoplasmic granules in electron microscopy.  相似文献

The combined effects of light intensity and nitrogen (NO3?) on growth rate, pigment content, and biochemical composition of Gracilaria foliifera v. angustissima (Harvey) Taylor was investigated using outdoor continuous cultures. Growth of Gracilaria increased linearly with increasing light to 0.43 doublings d?1 at high light levels (383 ly d?1 of in situ light), suggesting that light may often limit growth of this plant in nature. Chlorophyll a and phycoerythrin contents were inversely proportional to light level and growth rate. However, pigment content did not affect the growth capacity of Gracilaria. There was no increase in growth or pigment content with increasing additions of nitrogen. The low nitrogen treatment was unenriched seawater that had higher NO3? levels than most coastal waters (influent = 8.61 μM; residual = 0.94 μM). When growing near its maximum rate under high light intensities, Gracilaria had a significantly (P < 0.001) lower phycoerythrin: chlorophyll a ratio (phyco: Chl a) than did Gracilaria growing more slowly under lower light (Phyco:Chl a of 2.8 ± 0.2 vs. 3.8 ± 0.3). Faster growing plants also had C:N ratios above 10, indicating N- limitation. In addition to harvesting light the phycobiliproteins of Gracilaria may store nitrogen. Growth rates of Gracilaria correlated negatively with ash (r =–0.85) and positively with the carbon: phycoerythrin ratio (r = 0.85), suggesting that these two indices can be used to estimate growth in the field.  相似文献

ABSTRACT The storage carbohydrate granules from Euglena and Pavlova were compared by light and electron microscopy. Freezeetch studies demonstrated that while both types of granules are crystalline, they have different structures. The elemental microfibril of the euglenoid granule measures 4 nm, and the elemental striation of the granule from Pavlova is 22 nm. The granules each have a unique X-ray diffraction pattern. The storage carbohydrate granules from Pavlova are not the same as paramyton, and the term “paramylon” should be reserved for the euglenoid storage carbohydrate.  相似文献

Variation in the photosynthetic function ofAbies amabilis foliage within a canopy was examined and related to three different processes that affect foliage function: foliage aging, sun-shade acclimation that occurred while foliage was expanding, and reacclimation after expansion was complete. Foliage produced in the sun had higher photosynthesis at light saturation (A max, mol·m-2·s-1), dark respiration (mol·m-2·s-1), nitrogen content (g·m-2), chlorophyll content (g·m-2), and chlorophylla:b ratio, and a lower chlorophyll to nitrogen ratio (chl:N), than foliage produced in the shade. As sun foliage becomes shaded, it becomes physiologically similar to shade foliage, even though it still retains a sun morphology. Shaded sun foliage exhibited lowerA max, dark respiration, nitrogen content, and chlorophylla:b ratio, and a higher chl:N ratio than sun foliage of the same age remaining in the open. However, shaded sun foliage had a higher chlorophyll content than sun foliage remaining in the open, even though true shade foliage had a lower chlorophyll content than sun foliage. This anomaly arises because as sun foliage becomes shaded, it retains a higher nitrogen content than shade foliage in a similar light environment, but the two forms have similar chl:N ratios. Within the canopy, most physiological indicators were more strongly correlated with the current light environment than with foliage age or leaf thickness, with the exception of chlorophyll content.A max decreased significantly with both decreasing current light environment of the foliage and increasing foliage age. The same trend with current light and age was found for the chlorophylla:b ratio. Foliage nitrogen content also decreased with a decrease in current light environment, but no distinct pattern was found with foliage age. Leaf thickness was also important for predicting leaf nitrogen content: thicker leaves had more nitrogen than thinner leaves regardless of light environment or age. The chl:N ratio had a strong negative correlation with the current light environment, and, as with nitrogen content, no distinct pattern was found with foliage age. Chlorophyll content of the foliage was not well correlated with any of the three predictor variables: current light environment, foliage age or leaf thickness. On the other hand, chlorophyll content was positively correlated with the amount of nitrogen in a leaf, and once nitrogen was considered, the current light environment was also highly significant in explaining the variation in chlorophyll content. It has been suggested that the redistribution of nitrogen both within and between leaves is a mechanism for photosynthetic acclimation to the current light environment. Within theseA. amabilis canopies, both leaf nitrogen and the chl:N ratio were strongly correlated with the current light environment, but only weakly with leaf age, supporting the idea that changing light is the driving force for the redistribution of nitrogen both within and between leaves. Thus, our results support previous theories on nitrogen distribution and partitioning. However,A max was significantly affected by both foliage age and the current light environment, indicating that changes in light alone are not enough to explain changes inA max with time.  相似文献

目的:探讨类根瘤对烟草叶片中的叶绿体及其淀粉粒含量的影响。方法:无菌培养烟草再生植株,其中一部分经过一定浓度的2,4-D与豇豆根瘤菌512快生型突变株菌液诱导处理,另外一部分为对照。运用常规电镜技术制作叶片厚切片,通过显微观察,对两组烟草叶片细胞中的叶绿体及其淀粉粒含量进行对比分析。结果:结瘤植株叶片细胞内的叶绿体含量虽然与对照之间无明显差异,但叶片栅栏组织和海绵组织细胞内淀粉粒的含量却均较对照减少,其中前者减少更为明显(P<0.05)。结论:结瘤烟草叶片内光合产物的大量同化可能与类根瘤的形成有关。  相似文献

Five different artificial associations of cyanobacterial cells with the cells or tissues of nightshade and rauwolfia were studied. The associations grown on nitrogen-containing media produced heterocysts. Cyanobacterial cells in the associations retained their ability to take up combined nitrogen from the medium, to store it in the form of cyanophycin granules, and to use them in the process of symbiotic growth. The synthesis and degradation of cyanophycin granules in cyanobacterial cells were more active in the associations than in monocultures. In the symbiotic associations of Chlorogloeopsis fritschii ATCC 27193 with Solanum laciniatum cells and of Nostoc muscorum CALU 304 with the Rauwolfia serpentina callus, heterocysts were produced with a 3- to 30-fold higher cyanophycin content than in pure cyanobacterial cultures. In contrast, in the association of N. muscorum CALU 304 with the Solanum dulcamara callus, heterocysts were produced with a lower cyanophycin content than in the N. muscorum CALU 304 pure culture. The degradation of cyanophycin granules in N. muscorum CALU 304 cells grown in associations with plant tissues or cells was subjected to mathematical analysis. The activation of cyanophycin degradation and heterocyst differentiation in the associations N. muscorum CALU 304–R. serpentinaand C.fritschii–S. laciniatum was accompanied by an enhanced synthesis of the nitrogen-containing alkaloids in plant cells. The data obtained suggest that an integrated system of nitrogen homeostasis can be formed in symbiotic associations. Depending on the growth stage of an association, its plant member can either stimulate the accumulation of combined nitrogen in vegetative cyanobacterial cells in the form of cyanophycin granules, activate their degradation, or initiate the formation of heterocysts independently of the cyanobacterial combined nitrogen deprivation sensing-signaling pathway.  相似文献

The qualitative and quantitative carotenoid composition is reported for (i) a red and a green strain of Oscillatoria limnetica and a green strain of Spirulina platensis cultivated under identical conditions and (ii) a red and a green strain of Spirulina subsalsa grown under identical conditions. No correlation between colour and carotenoid content was obtained. However, differences in carotenoid composition within the Oscillatoria and Spirulina strains were observed. Both oscillol diglycoside ex Oscillatoria limnetica and myxol glycoside ex Spirulina platensis were mixed α-glycosides with chinovose-fucose present in ca 7:2 ratio. Analytical procedures are given. The chemosystematic implications for the current taxonomy of the genera Oscillatoria and Spirulina are discussed.  相似文献

Summary Polysaccharides like dextrine and starch are shown to be the best carbon sources for the growth ofAureobasidium pullulans although growth is good upon a variety of other carbon sources. Light increases growth markedly when polysaccharides are the carbon source but not when other sugars are used. Variation in cell morphology is described in response to sugars and light. Extracellular granules, whose properties resemble those of melanin, are produced when dextrine is the carbon source in a defined medium containing asparagine as the source of nitrogen. The dark pigment was extracted from the walls of thick-walled brown cells ofA. pullulans and characterized as a melanin on the basis of several tests, including solubility and absorption spectrum.A. pullulans was grown on several defined and undefined media and the response of the fungus to light is shown to be determined by the medium, and the temperature at which the cultures are grown.  相似文献

The structure forming in the area of contact between the oocyte and the germinal epithelium in the course of oocyte maturation of the scyphozoan Aurelia aurita is termed the contact plate. This study traces the successive stages of contact plate formation in the course of oocyte maturation at the light microscopic and ultrastructural levels. At early stages of oocyte development, the appearance of granules is observed in the peripheral cytoplasm of the oocyte; these granules accumulate at the pole, which retains its connection with the germinal epithelium of the gonads. Two types of these granules are recognized: (1) granules with homogeneous content and (2) granules containing loose shapeless material in the form of thick cords. The transformation of type two granules into larger structures, as well as the consolidation of type one and type two granules at later stages of oocyte development, are probably the processes that lead to the formation of the characteristic structure and contact plate, visible in paraffin and semithin sections. It remains unclear where exactly the contact plate is localized at the moment of fertilization: inside or outside the oocyte. The content of granules and components of the plate specifically bind the antibodies (RA47) against mesoglein, the ZP domain-containing protein of the mesoglea of A. aurita. The contact plate, covering only the anomalous pole of the oocyte but detected by the presence of ZP domain-containing proteins, may prove to be the simplest egg membrane of the Zona Pellucida type.  相似文献
