Chemotropic responses by pearl millet pollen tubes   总被引:6,自引:0,他引:6  
Summary The possible existence of a chemotropic factor controlling pearl millet pollen-tube directionality within the ovary was investigated using three approaches: cytochemical analysis of water-soluble components at the micropyle, in vitro testing of various chemicals for chemotropic activity, and an attempt to isolate and characterize an ovarian chemotropic factor. Observations of pollinated pearl millet ovaries by fluorescence microscopy revealed that pollen tubes enroute to the embryo sac exhibit specific directional turns in the regions of the style base/ovary juncture, the basal placenta, and the micropyle. The placenta and the micropylar regions have water-soluble periodic acid Schiff positive-substances as well as protein; this extracellular material and the walls of the micropylar nucellar cells appear to have associated calcium (Chaubal and Reger 1991). In vitro assays of pollen-tube behavior in response to a range of external stimuli revealed that pearl millet pollen tubes exhibit directional turns in response to: (1) diffusate from excised pistil tissues; (2) glucose, but not to several other carbohydrates; (3) calcium (unwashed polygalacturonic acid-calcium gel); (4) an ovarian water soluble, low molecular weight, acidic protein. These results which apparently suggest the presence of at least three different potential chemotropic factors in pearl millet ovaries are discussed in relation to angiosperm chemotropism in general and the difficulties involved in the search for possible in vivo chemotropic factors.On Specific Cooperative Agreement 58-43YK-8-0026 with the Department of Biochemistry, University of Georgia, Athens, GA 30602, USA  相似文献

Illus. 1961.—Pollen tubes of several species and hybrids of Lilium are attracted to certain pistil parts of Lilium in vitro. Positive chemotropism is observed with a variety of pistil-pollen combinations and is independent of the compatibility of these combinations in vivo. The chemotropic factor can be extracted by steeping the pistil parts in water. Chemotropic activity is retained following freeze-drying of pistil parts or extracts. The active material from L. longiflorum is heat-labile, dialyzable and not precipitated by acetone. Under certain conditions it causes distention of the pollen tube wall. Chemotropic activity of pistil parts is not simulated by a variety of known compounds or by extracts from various sources other than pistils. A possible mechanism of action of the chemotropic factor is discussed.  相似文献

A study of the pollen tube pathway in Lilium leucanthum var. centifolium and in L. regale reveals that the entire pathway from stigma to ovule is lined with cytologically unique stigmatoid cells. Assays for chemotropic activity of tissues and exudates along the pathway of pollinated or unpollinated pistils showed that onset of chemotropic activity progressed basipetally (and, when pollinated, in advance of the pollen tubes), commencing at the stigma 3-5 days before anthesis and appearing in the ovules 1-2 days after anthesis. Activity persists about 10 days in ovules of pollinated pistils and for 14-16 days in ovules of non-pollinated pistils. Attempts to localize the source of the chemotropic factor showed that gynoecial tissues bearing stigmatoid cells are chemo-tropically active while slices of style or ovary wall lacking stigmatoid cells are inactive. When ovules were sliced transversely and the micropylar and chalazal halves assayed, only the micropylar half showed activity. We suggest that the ovules and the stigmatoid tissue along the pollen tube pathway are the sources of the chemotropic factor responsible for the directional growth of the pollen tube.  相似文献

The expansion of both root hairs and pollen tubes occurs by a process known as tip growth. In this report, an Arabidopsis thaliana mutant (tip1) is described that displays defects in both root-hair and pollen-tube growth. The root hairs of the tip1 mutant plants are shorter than those of the wild-type plants and branched at their base. The tip1 pollen-tube growth defect was identified by the aberrant segregation ratio of phenotypically normal to mutant seeds in siliques from self-pollinated, heterozygous plants. Homozygous mutant seeds are not randomly distributed in the siliques, comprising only 14.4% of the total seeds, 5.3% of the seeds from the bottom half, and 2.2% of the seeds from the bottom quarter of the heterozygous siliques. Studies of pollen-tube growth in vivo showed that mutant pollen tubes grow more slowly than wild-type pollen through the transmitting tissue of wild-type flowers. Cosegregation studies indicate that the root-hair and pollen-tube defects are caused by the same genetic lesion. Based on these findings, the TIP1 gene is likely to encode a product involved in a fundamental aspect of tip growth in plant cells.  相似文献

The relationship between pollen and progeny performance has been a subject of many studies but the evidence for pollen-tube growth rate as an indicator of progeny fitness is equivocal. We used an anemophilous tree, Betula pendula, to examine the relationship between pollen-tube growth rate and seed and seedling performance. We crossed nine maternal plants with pollen from six pollen donors in a clonal B. pendula seed orchard, measured the pollen-tube growth rates for every cross, and analyzed the performance of the resulting seeds and seedlings. The only significant positive correlation was found between pollen-tube growth rate and seed mass when we controlled for seed number per inflorescence. Using seed mass as a covariate, we found that only maternal parent had a significant effect on the number of seeds per inflorescence, the percentage of germinable and embryonic seeds, and early seedling growth. Both maternal and paternal parents had significant effects on seedling height after 85 d of growth. These results are in concordance with the general view that maternal effects are usually most apparent in seed characters and during early plant growth. This study does not provide strong evidence for the theory of pollen-tube growth rate as an indicator of progeny quality.  相似文献

The pollen tube is the most rapidly growing cell in the plant kingdom and has the function to deliver the sperm cells for fertilization. The growing tip region of the cell behaves in a chemotropic manner to respond to the guidance cues emitted by the pistil and the female gametophyte, but how it perceives and responds to these directional triggers is virtually unknown. Quantitative assessment of chemotropic behavior can greatly be enhanced by the administration of pharmacological or other biologically active agents at subcellular precision, which is a technical challenge when the target area moves as it grows. We developed a laminar flow based microfluidic device that allows for continuous administration of two different solutions with a movable interface that permits the dynamic targeting of the growing pollen tube apex over prolonged periods of time. Asymmetric administration of calcium revealed that rather than following the highest calcium concentration as would be expected with simple chemotropic behavior, the pollen tube of Camellia targets an optimal concentration suggesting the presence of two superimposed mechanisms. Subcellular application of pectin methyl esterase (PME), an enzyme that modifies the growth behavior by rigidifying the pollen tube cell wall, caused the tube to turn away from the agent – providing important evidence for a previously proposed conceptual model of the growth mechanism.  相似文献

The influence of arbuscular mycorrhizal (AM) fungus Glomus deserticola (Trappe and John) on plant growth, nutrition, flower yield, water relations, chlorophyll (Chl) contents and water-use efficiency (WUE) of snapdragon (Antirhinum majus cv. butterfly) plants were studied in potted culture under well-watered (WW) and water-stress (WS) conditions. The imposed water stress condition significantly reduced all growth parameters, nutrient contents, flower yield, water relations, and Chl pigment content and increased the electrolyte leakage of the plants comparing to those of nonstressed plants. Regardless of the WS level, the mycorrhizal snapdragon plants had significantly higher shoot and root dry mass (DM), WUE, flower yield, nutrient (P, N, K, Mg, and Ca) and Chl contents than those nonmycorrhizal plants grown both under WW or WS conditions. Under WS conditions, the AM colonization had greatly improved the leaf water potential (??w), leaf relative water content (RWC) and reduced the leaf electrolyte leakage (EL) of the plants. Although the WS conditions had markedly increased the proline content of the leaves, this increase was significantly higher in nonmycorrhizal than in mycorrhizal plants. This suggests that AM colonization enhances the host plant WS tolerance. Values of benefit and potential dry matter for AM-root associations were highest when plants were stressed and reduced under WW conditions. As a result, the snapdragon plants showed a high degree of dependency on AM fungi which improve plant growth, flower yield, water relations particularly under WS conditions, and these improvements were increased as WS level had increased. This study confirms that AM colonization can mitigate the deleterious effect of water stress on growth and flower yield of the snapdragon ornamental plant.  相似文献

We studied whether the differences between genetically different pollen donors (Betula pendula Roth clones) with respect to pollen-tube growth rate were consistent under different thermal conditions during pollen germination in vivo and in vitro. We conducted a single-donor hand-pollination experiment with same pollen donors and recipients in a plastic house seed orchard and at an outdoor clone collection. The prevailing daily mean temperature during pollen germination was 13°C higher in the plastic house than outdoors. The pollen-tube growth rate of each pollen donor was additionally determined in vitro on agar medium at five temperatures (10°, 15°, 22°, 30° and 35°C). A significant interaction between paternal clone and pollination site as well as between paternal clone and temperature was found, which provides evidence for genotype-environment interactions. Genotype-environment interactions can have evolutionary significance in maintaining the variation in pollen-tube growth rates. At seed orchards, genotype-environment interactions can cause deviations from the expected genetic composition of the seed crop depending on the prevailing environmental conditions during pollen-tube growth. The upward gravitropic bending of cut snapdragon, lupinus and anemone flowering shoots was inhibited by salicylic acid (SA) applied at 0.5 mM and above. This effect was probably not due to acidification of the cytoplasm, since other weak acids did not inhibit bending of snapdragon shoots. In order to study its mode of inhibitory action, we have examined in cut snapdragon shoots the effect of SA on three processes of the gravity-signaling pathway, including: amyloplast sedimentation, formation of ethylene gradient across the stem, and differential growth response. The results show that 1 mM SA inhibited differential ethylene production rates across the horizontal stem and the gravity-induced growth, without significantly inhibiting vertical growth or amyloplast sedimentation following horizontal placement. However, 5 mM SA inhibited all three gravity-induced processes, as well as the growth of vertical shoots, while increasing flower wilting. It may, therefore, be concluded that SA inhibits bending of various cut flowering shoots in a concentration-dependent manner. Thus, at a low concentration SA exerts its effect in snapdragon shoots by inhibiting processes operating downstream to stimulus sensing exerted by amyloplast sedimentation. At a higher concentration SA inhibits bending probably by exerting general negative effects on various cellular processes.  相似文献

BACKGROUND AND AIMS: It is generally known that fertilization is delayed for more than a few weeks after pollination in Fagales. Recent studies showed that, during that period, pollen tubes grew in pistils in close association with the development of the ovule in a five-step process in Casuarina (Casuarinaceae) and a four-step process in Alnus (Betulaceae). The number of pollen tubes was reduced from many to one, a fact suggesting that delayed fertilization plays a role for gametophyte selection. Myrica (Myricaceae) also shows delayed fertilization for >2 weeks after pollination, but nothing is known of how pollen tubes grow in the pistil during that period. METHODS: Pollen-tube growth and the development of the ovule in pistils was investigated by fluorescent and scanning electron microscopy and analysis of microtome sections of the pistils. KEY RESULTS: Developmental study of the pollen-tube growth in the pistil of M. rubra showed that the tip of the pollen tube was branched or lay in a zigzag pattern in the upper space of the ovarian locule or near the tip of the integument, and subsequently was swollen on the nucellar surface. Such morphological changes indicate that the pollen-tube growth was temporarily arrested before fertilization. The pollen-tube growth in M. rubra can therefore be summarized as occurring in three steps: (1) from the stigma to the ovarian locule; (2) from the ovarian locule to the nucellar surface; and (3) from the nucellar surface to the embryo sac. CONCLUSION: Myrica differs from other families in that the pollen tubes arrest their growth on the nucellar surface, probably digesting nutrient from nucellar cells. There is little information on five other families of Fagales. An extensive study is needed to better understand the diversity and function of the mode of pollen-tube growth within the order.  相似文献

TIP1 is a gene defined by an X-ray induced allele tip1–2 and a previously described EMS-induced allele tip1−1 . TIP1 is involved in plant cell growth. tip1–2 plants display growth defects throughout the plant and exhibit defects in both root-hair and pollen-tube growth. tip1–2 plants are partly male sterile resulting from a combination of pollen germination and pollen-tube defects; their root-hairs are short, exhibit a tendency to branch and 2–4 hairs can initiate from each hair cell. They are also slightly dwarf in stature as a result of a general decrease in cell growth indicating that TIP1 activity is required for general cell growth. We propose a role for TIP in both the initiation and maintenance of growth in tip-growing cells. In addition TIP1 activity is required for normal cell expansion (non-tip cell growth) indicating that TIP1 is not exclusively involved in tip-growth.  相似文献

Previously it has been shown that the floral scent of snapdragon flowers consists of a relatively simple mixture of volatile organic compounds (VOCs). These compounds are thought to be involved in the attraction of pollinators; however, little is known about their effect on other organisms, such as neighboring plants. Here, we report that VOCs from snapdragon flowers inhibit Arabidopsis root growth. Out of the three major snapdragon floral volatiles, myrcene, (E)-β-ocimene, and methyl benzoate (MB), MB was found to be primarily responsible for the inhibition of root growth. Ten micromoles MB reduced root length by 72.6%. We employed a microarray approach to identify the MB target genes in Arabidopsis that were responsible for the root growth inhibition phenotype in response to MB. These analyses showed that MB treatment affected 1.33% of global gene expression, including cytokinin, auxin and other plant-hormone-related genes, and genes related to seed germination processes in Arabidopsis. Accordingly, the root growth of cytokinin (cre1) and auxin (axr1) response mutants was less affected than that of the wild type by the volatile compound: roots of the treated mutants were reduced by 45.1 and 56.2%, respectively, relative to untreated control mutants. In this work, we report the occurrence of chemotropism in the arbuscular mycorrhizal (AM) fungus Glomus mosseae. Fungal hyphae were able to respond to host-derived signals by reorienting their growth towards roots and to perceive chemotropic signals at a distance of at least 910 microm from roots. In order to reach the source of chemotropic signals, hyphal tips crossed interposed membranes emerging within 1 mm from roots, eventually establishing mycorrhizal symbiosis. The specificity of chemotropic growth was evidenced by hyphal growth reorientation and membrane penetration occurring only in experimental systems set up with host plants. Since pre-symbiotic growth is a critical stage in the life cycle of obligate AM fungal symbionts, chemotropic guidance may represent an important mechanism functional to host root location, appressorium formation and symbiosis establishment.  相似文献

The protein NaGSL1 (Nicotiana alata glucan synthase-like 1) is implicated in the synthesis of callose, the 1,3-beta-glucan that is the major polysaccharide in the walls of N. alata (flowering tobacco) pollen tubes. Here we examine the production, intracellular location and post-translational processing of NaGSL1, and relate each of these to the control of pollen-tube callose synthase (CalS). The 220 kDa NaGSL1 polypeptide is produced after pollen-tube germination and accumulates during pollen-tube growth, as does CalS. A combination of membrane fractionation and immunoelectron microscopy revealed that NaGSL1 was present predominantly in the endoplasmic reticulum and Golgi membranes in younger pollen tubes when CalS was mostly in an inactive (latent) form. In later stages of pollen-tube growth, when CalS was present in both latent and active forms, a greater proportion of NaGSL1 was in intracellular vesicles and the plasma membrane, the latter location being consistent with direct deposition of callose into the wall. N. alata CalS is activated in vitro by the proteolytic enzyme trypsin and the detergent CHAPS, but in neither case was activation associated with a detectable change in the molecular mass of the NaGSL1 polypeptide. NaGSL1 may thus either be activated by the removal of a few amino acids or by the removal of another protein that inhibits NaGSL1. These findings are discussed in relation to the control of callose biosynthesis during pollen germination and pollen-tube growth.  相似文献

Aurones, a class of plant flavonoids, provide bright yellow color on some important ornamental flowers, such as cosmos, coreopsis, and snapdragon (Antirrhinum majus). Recently, it has been elucidated that aureusidin synthase (AUS), a homolog of plant polyphenol oxidase (PPO), plays a key role in the yellow coloration of snapdragon flowers. In addition, it has been shown that AUS is a chalcone-specific PPO specialized for aurone biosynthesis. AUS gene has been successfully demonstrated as an attractive tool to engineer yellow flowers in blue flowers. Despite these biological studies, the structural basis for the specificity of substrate interactions of AUS remains elusive. In this study, we performed homology modeling of AUS using Grenache PPO and Sweet potato catechol oxidase (CO). An AUS-inhibitor was then developed from the initial homology model based on the CO and subsequently validated. We performed a thorough study between AUS and PTU inhibitor by means of interaction energy, which indicated the most important residues in the active site that are highly conserved. Analysis of the molecular dynamics simulations of the apo enzyme and ligand-bound complex showed that complex is relatively stable than apo and the active sites of both systems are flexible. The results from this study provide very helpful information to understand the structure-function relationships of AUS.  相似文献

Flavonoid metabolons (weakly‐bound multi‐enzyme complexes of flavonoid enzymes) are believed to occur in diverse plant species. However, how flavonoid enzymes are organized to form a metabolon is unknown for most plant species. We analyzed the physical interaction partnerships of the flavonoid enzymes from two lamiales plants (snapdragon and torenia) that produce flavones and anthocyanins. In snapdragon, protein–protein interaction assays using yeast and plant systems revealed the following binary interactions: flavone synthase II (FNSII)/chalcone synthase (CHS); FNSII/chalcone isomerase (CHI); FNSII/dihydroflavonol 4‐reductase (DFR); CHS/CHI; CHI/DFR; and flavonoid 3′‐hydroxylase/CHI. These results along with the subcellular localizations and membrane associations of snapdragon flavonoid enzymes suggested that FNSII serves as a component of the flavonoid metabolon tethered to the endoplasmic reticulum (ER). The observed interaction partnerships and temporal gene expression patterns of flavonoid enzymes in red snapdragon petal cells suggested the flower stage‐dependent formation of the flavonoid metabolon, which accounted for the sequential flavone and anthocyanin accumulation patterns therein. We also identified interactions between FNSII and other flavonoid enzymes in torenia, in which the co‐suppression of FNSII expression was previously reported to diminish petal anthocyanin contents. The observed physical interactions among flavonoid enzymes of these plant species provided further evidence supporting the long‐suspected organization of flavonoid metabolons as enzyme complexes tethered to the ER via cytochrome P450, and illustrated how flavonoid metabolons mediate flower coloration. Moreover, the observed interaction partnerships were distinct from those previously identified in other plant species (Arabidopsis thaliana and soybean), suggesting that the organization of flavonoid metabolons may differ among plant species.  相似文献

The prevalence of nonrandom fertilization due to postpollination events has rarely been studied in natural populations, despite important implications for outcrossing rates, mate choice, and plant fitness. Nonrandom paternity within fruits can be caused by both unequal fertilization and unequal embryo abortion. Using self-compatible Hibiscus moscheutos, we studied the potential for nonrandom fertilization by comparing growth rates of pollen-tubes from different donors. The branched style of Hibiscus allowed within-flower comparisons between pollen donors. Relative pollen-tube growth rates were determined by applying pollen from pairs of donors to different stigmas on adjacent stylar branches. We then measured the number of callose plugs per tube in cross-sectional transects across the style after 3 hr. We demonstrate that rates of callose plug formation can be used as a sensitive indicator of relative pollen-tube growth rate. Differences between pollen donors were common and repeatable. Self-pollen-tubes grew slower than outcross pollen-tubes in some crosses and faster in others. Allozyme variation in glucose phosphate isomerase was used to show that individuals with fast-growing pollen-tubes sired a disproportionate number of seeds following mixed pollinations (up to 72%). Since seed abortion was negligible, we conclude that variation in pollen-tube growth rates leads to nonrandom paternity within fruits.  相似文献
