The following results were obtained using etiolated Alaska pea epicotyls. Gibberellic acid (GA) had the remarkable effect on the elongation in part I (elongating region) of epicotyls, whereas it had little effect on that in part II (mature region) of epicotyls. In cortex of part I and II of epicotyls, the cell number in longitudinal direction was hardly affected by GA. On the increase in width of epicotyls, GA was hardly effective in any parts of epicotyls. In both part I and II GA enhanced the incorporation of 32P into all nucleic acid fractions prepared by methylated albumin kieselguhr (MAK) columns, i.e. sRNA, DNA and rRNA + mRNA. In part I the net increase in DNA and RNA content during the incubation period was slightly promoted by GA, whereas in part II the net decrease in both nucleic acids content was slightly promoted by GA. The relationship between GA-induced growth and nucleic acid synthesis is discussed.  相似文献

We developed a three-dimensional (3-D) clinostat to simulate a microgravity environment and studied the changes in plant growth processes under this condition. The rate of germination of cress (Lepidium sativum), maize (Zea mays), rice (Oryza sativa), pea (Pisum sativum), or azuki bean (Vigna angularis) was not affected on the clinostat. The clinostat rotation did not influence the growth rate of their roots or shoots, except for a slight promotion of growth in azuki roots and epicotyls. On the contrary, the direction of growth of plant organs clearly changed on the 3-D clinostat. On the surface of the earth, roots grow downward while shoots upward in parallel to the gravity vector. On the 3-D clinostat, roots of cress elongated along the direction of the tip of root primordia after having changed the direction continuously. Rice roots also grew parallel to the direction of the tip of root primordia. On the other hand, roots of maize, pea, and azuki bean grew in a random fashion. The direction of growth of shoots was more controlled even on the 3-D clinostat. In a front view of embryos, shoots grew mostly along the direction of the tip of primordia. In a side view, rice coleoptiles showed an adaxial (toward the caryopsis) while coleoptiles of maize and epicotyls of pea and azuki bean an abaxial curvature. The curvature of shoots became larger with their growth. Such an autotropism may have an important role in regulation of life cycle of higher plants under a microgravity environment.  相似文献

The 8 days old seedlings of pea (cv. Ilowiecki) and maize (cv. Alma F1) were subjected to differentiated aeration conditions (control — with pore water tension about 15 kPa and flooded treatment) for 12 days at three soil temperatures (7, 15 and 25 °C). The shoots were grown at 25 °C while the soil temperature was differentiated by keeping the cylinders with the soil in thermostated water bath of the appropriate temperature. Lowering the root temperature with respect to the shoot temperature caused under control (oxic) conditions a decrease of the root penetration depth, their mass and porosity as well as a decrease of shoot height, their mass and chlorophyll content; the changes being more pronounced in maize as compared to the pea plants. Flooding the soil diminished the effect of temperature on the investigated parameters; the temperature effect remaining significant only in the case of shoot biomass and root porosity of pea plants. Root porosity of pea plants ranged from 2 to 4 % and that of maize plants — from 4 to 6 % of the root volume. Flooding the soil caused an increase in the root porosity of the pea plants in the entire temperature range and in maize roots at lower temperatures by about 1 % of the root volume. Flooding the soil caused a decrease of root mass and penetration depth as well as a decrease of plant height, biomass and leaf chlorophyll content.  相似文献

Summary Tobacco leaves were homogenized and separated by differential centrifugation into four subcellular fractions. Each fraction was characterized by the presence of certain marker substances and enzymes. It was found that nuclei and chloroplasts were highly enriched in the 2500 ×g sediment. Mitochondria were especially enriched in the 6500 × g g sediment, but were also found in the 20000 ×g sediment. The activity of a marker enzyme for cytomembranes (inosinediphosphatase) was high in the 6500 ×g and the 20000 ×g sedi-ments. In TMV infected material complementary viral R~qA (minus strands) was enriched in the 6500 ×g and the 20000 ×g sediments. Also, after labeling infected leaves with tritiated uridine, RNase resistant radioactivity was enriched in these two fractions. The results suggest that the TMV RNA replication complex is bound to cytoplasmic membranes.  相似文献

Increased expression of the auxin-inducible gene PsIAA4/5 was observed in the elongated side of epicotyls in early growth stages of etiolated pea (Pisum sativum L. cv. Alaska) seedlings grown in a horizontal or an inclined position under 1 g conditions. Under simulated microgravity conditions on a 3D clinostat, accumulation of PsIAA4/5 mRNA was found throughout epicotyls showing automorphosis. Polar auxin transport in the proximal side of epicotyls changed when the seedlings were grown in a horizontal or an inclined position under 1 g conditions, but that under clinorotation did not, regardless of the direction of seed setting. Accumulation of PsPIN1 and PsPIN2 mRNAs in epicotyls was affected by gravistimulation, but not by clinorotation. Under 1 g conditions, auxin-transport inhibitors made epicotyls of seedlings grown in a horizontal or inclined position grow toward the proximal direction to cotyledons. These inhibitors led to epicotyl bending toward the cotyledons in seedlings grown in an inclined position under clinorotation. Polar auxin transport, as well as growth direction, of epicotyls of the agravitropic mutant ageotropum did not respond to various gravistimulation. These results suggest that alteration of polar auxin transport in the proximal side of epicotyls regulates the graviresponse of pea epicotyls.  相似文献

The distribution of two of the enzymes involved in the biosynthesisof aromatic compounds in higher plants, namely dehydroshikimicreductase and dehydroquinase, was studied. The two enzymes werefound to occur mainly in the supernatant fraction of cauliflowerbuds. In extracts of pea epicotyls, 30 per cent. of the dehydroshikimicreductase activity was associated with the mitochondria. Theactivities of the two enzymes in various organs of germinatingpea seedlings were measured over an 8-day period. Synthesisof both enzymes was indicated in the roots, shoots, and cotyledons.The two enzymes were found to occur in a variety of plant materials.  相似文献

Summary Root initiation in etiolated pea epicotyls induced by NAA and 2,4-D is inhibited by application of 5-BU in the same way as root initiation induced with IAA. The time of 5-BU action is regulated by auxin concentration. Binding of auxins to carefully extracted nucleoproteins from pea epicotyls at pH 8 resulted in a decrease of the melting point (T m) of the nucleoproteins. From the results it is concluded that auxins act by direct binding to the nucleoprotein (probably by ionic bonds) and that this binding induces a decrease in binding capacity of histone to DNA, which represents the first step in the initiation of RNA synthesis.  相似文献

The effect of phosphorus deficiency on acid phosphatase activity in the apical, middle and basal parts of the root of maize plants was followed. The supernatant obtained by centrifuging the homogenate of plant tissue at 1500 ×g was further centrifuged at 18 000 ×g, the sediment being marked as fraction II and the supernatant as fraction III. The results obtained document the fact that acid phosphatase activity of the two fractions of all analyzed root segments was higher in plants cultured in nutrient medium without phosphate than in those containing phosphorus in nutrient medium. In most cases this difference was significant to highly significant. The results of experiments proved unambiguously a higher enzymatic activity in all root segments in fraction III than in fraction II. In fraction III the highest acid phosphatase activity was found in the apical part, in fraction II in the basal part of the root.  相似文献

Hypergravity caused by centrifugation inhibits elongation growth of shoots by decreasing the cell wall extensibility via suppression of xyloglucan breakdown as well as by the thickening of cell walls. The mechanism of graviperception in hypergravity-induced growth inhibition was investigated in Arabidopsis [A. thaliana (L.) Heynh.] hypocotyls and azuki bean (Vigna angularis Ohwi et Ohashi) epicotyls. Hypergravity caused growth suppression in both sgr1-1 and pgm1, which are Arabidopsis mutants deprived of gravitropism, as in wild-type plants, suggesting that the graviperception in hypergravity-induced growth inhibition of shoots is independent of that in gravitropism. Hypergravity had no effects on growth of azuki bean epicotyls or Arabidopsis hypocotyls in the presence of lanthanum or gadolinium, which are blockers of mechanoreceptors. Moreover, lanthanum or gadolinium at the same concentration had no influence on gravitropism of azuki bean epicotyls and Arabidopsis hypocotyls. Hypergravity had no effects on cell wall extensibility and affected neither xyloglucan metabolism nor the thickness of cell walls in the lanthanum- or gadolinium-treated azuki bean epicotyls. Lanthanum or gadolinium inhibited the hypergravity-induced increase in the pH of the apoplastic fluid in the epicotyls, which is involved in the processes of the suppression of xyloglucan breakdown due to hypergravity. These findings suggest that plants perceive the hypergravity stimuli by mechanoreceptors in the plasma membrane, and utilize the perceived signal to regulate the growth rate of their shoots.Abbreviations HC-I Hemicellulose-I - HC-II Hemicellulose-II  相似文献

The turgor of pea (Pisum sativum) leaves was reduced by exposing excised pea shoots to a stream of 23°C air for 20 min. Poly(A)+ RNA was isolated from control and wilted shoots, translated in vitro and radiolabeled translation products separated by electrophoresis on two-dimensional (isoelectric focusing-sodium dodecyl sulfate) polyacrylamide gels. This analysis showed that the levels of several poly(A)+ RNAs increased in wilted plants. Most of the poly(A)+ RNAs induced in wilted plants did not accumulate in response to heat shock or exogenously applied ABA even though endogenous ABA levels were found to increase in shoots 30 min after wilting and by 4 h had increased 50-fold (1 versus 0.02 microgram per gram fresh weight). A λgt10 cDNA library was constructed using poly(A)+ RNA from wilted shoots which had been incubated for 4 hours. Differential screening of the library identified four clones corresponding to poly(A)+ RNAs which are induced in wilted shoots.  相似文献

Strawberry mottle virus (SMoV) (three isolates: HJ, 3E and N) were transmitted to Chenopodium quinoa plants by sap inoculation. All three isolates induced very similar symptoms consisting of chlorotic spots and ringspots in inoculated leaves, and vein chlorosis, mottling, and dwarfing of the upper leaves. SMoV isolate HJ was purified from infected C. quinoa by homogenisation with 10 mM phosphate buffer, pH 7.2 containing 5% Triton X-100, followed by differential, sucrose density-gradient and CsCl equilibrium density-gradient centrifugations. A fraction with a buoyant density of 1.42g- cm-3 after CsCl density-gradient centrifugation was highly infectious to C. quinoa and contained many isometric virus-like particles c. 37 nm in diameter. These virus-like particles were never found in fractions from uninfected preparations. Electrophoretic analysis of a fraction containing virus-like particles revealed that these particles might have a single coat protein subunit with the apparent molecular mass of 26 K daltons and one nucleic acid of 6.6 kilobases. Double-stranded RNA analysis of isolate HJ-infected or uninfected C. quinoa and Fragaria vesca var. semperflorens seedling line ‘Alpine’ plants showed that both infected plants had two infection-specific dsRNA bands of mol. wts 4.5 and 3.9 × 106.  相似文献

Transformation and high efficient regeneration of transgenic plants from the trimmed etiolated shoot/root region (TESRR) of Anliucheng sweet orange [Citrus sinensis (L.) Osb.] seedling was reported. A visual green fluorescent protein (GFP) marker gene was introduced to evaluate transformation efficiency by using the explants from TESRR and epicotyls. The transformation protocol was: infection 20 min, co-culture 3 d, selection culture 30 d, and rooting 15 d. Out of a total of 288 sprouted shoots obtained from TESRR, 34 shoots (11.8 %) yielded GFP expression. In contrast, only 2 (3.0 %) of the 67 sprouted shoots from epicotyl transformation yielded GFP expression. In all plants showing the green fluorescence an expected 500 bp GFP fragment was proved by PCR analysis. Southern blot analysis further confirmed the integration of GFP gene into citrus genome. Transgenic plantlets were obtained within 80 d using the TESRR, compared within 150 d by using epicotyls.  相似文献

An isolate of Australian lucerne latent virus (ALLV) from lucerne in New Zealand was mechanically transmitted to a few herbaceous hosts. It induced diagnostic symptoms in several species of the Chenopodiaceae, but was symptomless in most other hosts including lucerne and Trifolium subterraneum. It was seed transmitted in lucerne. When assayed to Chenopodium quinoa, infective C. quinoa sap lost infectivity after diluting to 10-4, heating for 10 min at 55°C and storage for 4 days at 4°C. ALLV was purified from infected C. quinoa or pea plants by extracting sap in 0.1 m borate buffer (pH 7) containing 0.2% 2-mercaptoethanol and clarifying with 15% bentonite suspension, high and low speed centrifugation and sucrose density gradient centrifugation. Purified virus preparations contained isometric particles about 25 nm in diameter and sedimented as three virus components with sedimentation coefficients (s20-w0) of 56 S, 128 S and 133 S. The 56 S component appeared to consist of nucleic acid-free protein shells. Polyacrylamide gel electrophoresis of virus preparations showed that ALLV contained a single protein species of mol. wt 55 000 and two RNA species of mol. wt 2.1 × 106 and 2.4 × 106. An antiserum to ALLV had an homologous titre of 1/256 to purified virus but failed to detect ALLV in infective sap of C. quinoa, pea or lucerne. Purified ALLV failed to react to antisera to 28 distinct isometric plant viruses including those to 10 nepoviruses.  相似文献

The dependence of the nitrogen fixing system in the root nodules of pea plants (Pisum sativum) L. cv. Torsdag II) on light induced reactions was studied. The pots of the inoculated pea plants, after the nolules had fixed nitrogen for a fornight, were transferred to a dark room. The control plants were kept under normal lighting conditions. The decay of leghemoglobin was measured after photosynthesis had ceased. In the dark the red nodules turned green in three days, when about half of the haem had been broken down. The plants in normal lighting conditions had maintained the red nodules. The appearence of leghemoglobin and bacteroids was simultaneouos. In normal lighting conditions the number of bacteroids was about 1.6 × 108 per g fresh nodules. The appearance of leghemoglobin and bacteroids was simultaneous. In normal lighting conditons the number of bacteroids was bout 1.6 × 108 per g fresh nodules. At the same time as the nodules turned green in the dark most of the bacteroids disappeared and the number of rod-shaped bacteria increased. After five days int the dark thenumber of bacteria of the green nodules was 2.2 × 108 per g fresh nodules. A large increase of of bacteria in the nodules is one of the results after the termination of effective symbiosis. Quantitative estimations were made with an automatic amino acid analysator of the amino acid composition in the root nodules of pea plants grown in the light and of pea plants grown in the dark. Altogether 27 amino acids and amides and 3 unknown ninhydrin positive compounds were found in the free amino acid fraction. In the red N-fixing nodules asparagine, the amide of aspartic acid, was the most prominent (more than 50 per cent of the total amino acid fraction), indicating the energy charge of the nitrogen fixation. 5 days in the dark affected the proportions of the amino acids as follows. Asparagine, homoserine, γ-aminobutyric acid and ethanolamine were decreased and the most of the others increased. In the hydrolysate of the non-soluble protein fraction 25 amino acids could be detected. The proportions of the amino acids in the root nodules of light-grown and dark-grown pea plants were very similar. Hydroxyproline and α, γ-diaminopimelic acid (DAP) were found in these fraction. Most of the DAP was contained in the peptide fraction. Also hydroxyproline was found to a small extent. It was assumed that the amino acids in this fraction were derived from the peptides of both plant cells and rhizobia.  相似文献

Maize and pea plants were treated with 0.0 (control), 0.01 or0.05 mM Cd in the growing medium for 11 d. Although the totalCd concentration was similar in shoot and root tissues of bothspecies, pea plants showed more severe toxic symptoms. The freshweight and percentage of water content of root and shoot decreasedconcomitantly to Cd supply. High Cd levels were found in thecell-wall fraction (Fraction I) and in Fraction IV (soluble)of maize plants, whereas Cd-treated pea accumulated more Cdin the soluble fraction. The protein concentration of FractionIV of pea shoot and root significantly increased upon treatmentwith 0.05 mM Cd, whereas maize showed no effect. Furthermore,a previously not visible protein ({small tilde}12 kDa), appearedin Fraction IV of pea root grown with the highest Cd supply.Cadmium treatment, in general, notably enhanced the concentrationsof 2-thiobarbituric acid reactive material (lipid peroxidationproducts) in pea fractions, presumably due to Cd-induced oxidativestress. We have tested the hypothesis that wall-bound calcium is redistributed from the lower to the upper sides of horizontal stems and coleoptiles during the reaction phase of gravicurvature. We used atomic absorption spectrometry to measure wall-bound calcium in the epidermal and internal layers of sunflower (Helianthus annum L.) hypocotyls and pea (Pisum sativum L.) epicotyls, and total calcium in maize (Zea mays L.) coleoptiles at the time of maximal gravireaction. In every case, we found that there was no measurable redistribution of wall-bound calcium either from the lower to the upper sides, or between epidermal and inner tissues in response to the gravistimulus. These results indicate that a redistribution of wall-bound calcium does not play a role during the gravireaction of stems and coleoptiles.  相似文献

Tobacco leaves were inoculated with tobacco mosaic virus labeled with 32P or 35S. After various intervals, extracts of the leaves were prepared. In extracts from leaves infected for 5 to 360 min, about 40 to 60% of the virus retained on leaves was recovered in the pellet of the homogenate centrifuged at 12 000 × g. The virus associated with the 12 000 × g pellet was dissociable by treatment with pancreatic RNase, alkali or sodium dodecyl sulfate (SDS). The parental virus extracted by SDS from the pellet at 12 000 × g had a large amount of partially uncoated virus possessing naked RNA. Analysis by density gradient centrifugation suggested that, in addition to partially uncoated virus, some fragmented RNA was also associated with the 12 000 × g pellet. This fragmented RNA seemed to be derived from partially uncoated virus. Density gradient analysis of SDS extracts from the 12 000 × g pellet suggested that some of the virus underwent uncoating at the internal regions of the virus particle.  相似文献

The effect of phosphorus deficiency on the activity of acid phosphatase of the first, second and third leaves of maize plants was followed. The supernatant obtained by centrifuging the homogenate of plant tissue at 1500 ×g was further centrifuged at 18 000 ×g, the sediment marked as fraction II and the supernatant as fraction III. Acid phosphatase activity of fraction II of the first to third leaves was for the whole period of culture higher in plants grown in the nutrient solution without phosphate. In fraction III this relation was established in the first leaf, after 3 days of culture in the second leaf and after 5 days in the third leaf. In all leaves higher enzyme activity was unambiguously determined in fraction III when compared with fraction II. Higher acid phosphatase activity was established in those leaves which were younger in their development, particularly in the first days of culture. With the ageing of leaves the enzyme activity decreased.  相似文献

In dark grown pea (Pisum sativum) seedlings ethylene causes the triple response in which elongation growth is inhibited, radial growth is promoted, and orientation of shoots to gravity is altered. The distribution of extensin and peroxidase activity in pea epicotyls upon ethylene treatment was studied by tissue printing on nitrocellulose paper. It was found that the localization of extensin and peroxidase activity changes after 72 and 96 hours of ethylene treatment. In untreated plants, peroxidase activity is detected only in the vascular bundles. Nonetheless, after 72 and 96 hours of ethylene treatment peroxidase activity is hardly detected in the vascular system but present in the epidermal and cortical cells. Extensin increases in the epidermal and cortical cells upon ethylene treatment but it also appears in the vascular system when peroxidase activity is no longer detected.  相似文献
