Root extracts of leek (Allium porrum L.) and soybean (Glycine max L. Merr.) showed trehalase activity which was inhibited by phloridzin and was several times higher than the activity of general -glucosidase. The activity had an acidic optimum. Trehalase activity in extracts of sporocarps and extraradical mycelium of the arbuscular mycorrhizal fungus Glomus mosseae Nicol. & Gerd. (Trappe & Gerd.) was higher than in root extracts and had an optimum at pH 7. Following inoculation with G. mosseae, trehalase activity increased in mycorrhizal roots above the levels observed in nonmycorrhizal roots. Irrespective of fungal colonization, root trehalase activity increased in the presence of Mg2+, decreased in the presence of Mn2+ and Zn2+, and was unaffected by Na2EDTA.  相似文献

The extent to which trehalose is accumulated in the vegetative mycelium of strains of Neurospora crassa is significantly affected by conidiation. In heavily conidiating strains a rapid decrease in mycelial trehalose occurs following the initiation of conidiation. Meanwhile, trehalase activity in the vegetative mycelium of heavily conidiating strains increases rapidly following the initiation of conidiation, although apparently it is not directly caused by the sporulation process. High levels of both trehalase and trehalose appear concomitantly in the newly formed conidia.  相似文献

Lyophilized cells of the non-pathogenic yeast Saccharomyces boulardii are used in many countries for the treatment of several types of diarrhoea and other gastrointestinal diseases. Although the cells must be viable, their mechanism of action is unknown. The disaccharide trehalose is a protectant against several forms of environmental stress in yeast and is involved in maintaining cell viability. There is no information on the enzymes involved in degradation of trehalose in S. boulardii. The aim of the present study was to characterize trehalase activity in this yeast. Cells of S. boulardii grown in glucose exhibited neutral trehalase activity only in the exponential phase. Acidic trehalase was not detected in glucose medium. Cells grown in trehalose exhibited acid and neutral trehalase activities at all growth stages, particularly in the exponential phase. The optimum pH and temperature values for neutral trehalase activity were determined as 6.5 and 30 °C respectively, the half-life being approximately 3 min at 45 °C. The relative molecular mass of neutral trehalase is 80 kDa and the K m 6.4 mM (±0.6). Neutral trehalase activity at pH 6.5 was weakly inhibited by 5 mM EDTA and strongly inhibited by ATP, as well as the divalent ions Cu++, Fe++ and Zn++. Enzyme activity was stimulated by Mg++ and Ca++ only in the absence of cAMP. The presence of cAMP with no ion additions increased activity by 40%. Mycelium ofBeauveria bassiana can be grown in liquid culture, filtered, and the mycelium dried. After rehydration the mycelium sporulates. Two carbohydrate sources (sucrose and maltose), and one nitrogen/vitamin source (yeast extract) were tested for mycelium growth and subsequent conidial production. Maximum mycelium growth (12.31 mg/ml), in liquid culture, was in the sucrose (3.5%)/yeast extract (3.5%) medium, but mycelium from a maltose (2%)/yeast extract (0.75%) medium produced the maximum of 4.62×106 conidia/mg dry mycelium after incubation in moist Petri dishes. Using the data on mycelium yield (in liquid culture) and conidial production (by dry mycelium) it is calculated that the sucrose (3.5%)/yeast extract (3.5%) and the maltose (2%)/yeast extract (0.75%) media produce most conidia per media volume (an equivalent of 3.52–3.72×107 conidia/ml).   相似文献

An electrophoretically homogenous aggregate of acid trehalase, invertase and an unidentified 37–41 kDa protein was purified from Saccharomyces cerevisiae. N-terminal analysis of the protein revealed an amino acid sequence identical to that of Bgl2p (endo-β-l,3-glucanase) of S. cerevisiae. Acid trehalase activity with co-eluted glucanase activity was observed from late growth phase through early stationary phase. Pools with high percentage of Bgl2p corresponded with high acid trehalase activity. A BGL2 deletion strain had lower acid trehalase activity. The 37–41 kDa protein represents Bgl2p which, besides imparting glucanase activity, could also be acting as a regulator for the acid trehalase activity by association in the enzyme aggregate.  相似文献

A toxic effect of α,α-trehalose in an angiospermic plant, Cuscuta reflexa (dodder), is described. This disaccharide and its analogs, 2-aminotrehalose and 4-aminotrehalose, induced a rapid blackening of the terminal region of the vine which is involved in elongation growth. From the results of in vitro growth of several angiospermic plants and determination of trehalase activity in them, it is concluded that the toxic effect of trehalose in Cuscuta is because of the very low trehalase activity in the vine. As a result, trehalose accumulates in the vine and interferes with some process closely associated with growth. The growth potential of Lemna (a duckweed) in a medium containing trehalose as the carbon source was irreversibly lost upon addition of trehalosamine, an inhibitor of trehalase activity. It is concluded that, if allowed to accumulate within the tissue, trehalose may be potentially toxic or inhibitory to higher plants in general. The presence of trehalase activity in plants, where its substrate has not been found to occur, is envisaged to relieve the plant from the toxic effects of trehalose which it may encounter in soil or during association with fungi or insects.  相似文献

Morchella conica Pers. strains of the study were isolated from fruit bodies collected in ash-mixed forests. At first, the strains were cultured on potato dextrose agar (PDA), then on modified Murashige and Skoog (MS) solid agar media. A normal-growing strain was chosen for the trehalase induction experiments. During the trehalase induction treatment, mycelia were grown in liquid culture containing different concentrations of trehalose. After the induction period of trehalase enzymes, physiological state of the mycelium and the oxidative stress were monitored in the vegetative mycelia by measuring the change of the malondialdehyde content, superoxide dismutase enzyme activity, the fresh and dry weight. The examined Morchella conica strain utilized the trehalose properly. The rising amount of the trehalose triggered the increase of the mycelial trehalase enzyme activity. Our results clearly proved that both neutral and acidic trehalase isoenzyme activity of the Morchella conica mycelium are inducible and are playing important role in the utilization of external trehalose.  相似文献

Trehalases play a central role in the metabolism of trehalose and can be found in a wide variety of organisms. A periplasmic trehalase (α,α-trehalose glucohydrolase, EC 3.2.1.28) from the thermophilic bacterium Rhodothermus marinus was purified and the respective encoding gene was identified, cloned and overexpressed in Escherichia coli. The recombinant trehalase is a monomeric protein with a molecular mass of 59 kDa. Maximum activity was observed at 88°C and pH 6.5. The recombinant trehalase exhibited a K m of 0.16 mM and a V max of 81 μmol of trehalose (min)−1 (mg of protein)−1 at the optimal temperature for growth of R. marinus (65°C) and pH 6.5. The enzyme was highly specific for trehalose and was inhibited by glucose with a K i of 7 mM. This is the most thermostable trehalase ever characterized. Moreover, this is the first report on the identification and characterization of a trehalase from a thermophilic bacterium.  相似文献

Summary The production of protease and mycelium byNeurospora sitophila cultured on solid and liquid potato dextrose media was studied. Maximal activity of protease extracted from 4-day-old cultures occurred at pH 6.5 when an unfractionated peanut (groundnut) protein substrate was used. The greatest protease activity and mycelium production occurred during the first day of the 4-day test period. Potato dextrose media containing more than 0.2 M NaCl resulted in decreased protease and mycelium production, while tapioca starch was without affect at concentrations up to 1.4%. Addition of up to 0.3 M sucrose to growth media greatly stimulated protease production and mycelial growth. Maximal proteolytic activity was observed in extracts from mycelium cultured in potato dextrose media adjusted from pH 6.0 to 7.5. Activity was greater when soluble peanut protein was used as a substrate, compared to unfractionated or globulin protein substrates.  相似文献

Biochemical characterization of a trehalase, detected in the mid-exponential growth phase of Candida utilis NCIM Y500, has indicated that it was a neutral trehalase and possibly the only trehalase present in this strain. Unlike Saccharomyces cerevisiae and other C. utilis strains, this strain without acid trehalase grew quite well in minimal or complete medium containing trehalose as the sole source of carbon. Both these observations were contradictory to the findings reported for acid trehalase mutants of S. cerevisiae and C. utilis. The trehalase system of the strain is suggested to be similar to that of fungi.  相似文献

The onion white rot pathogen Sclerotium cepivorum was cultured on agar media containing 2% malt extract and one of the antifungal antibiotics, endomycin, griseofulvin, venturicidin and cycloheximide at concentrations that reduced but did not prevent growth of mycelium. When onion seeds or agar discs impregnated with diffusates from onion bulbs were placed on the antibiotic media, radial growth of the fungus was greatly increased, and there was a profuse development of aerial mycelium. Gaseous diffusates from onion tissue and from impregnated agar discs were also effective. On the antibiotic media, tomato, cabbage and radish seeds did not stimulate the growth of S. cepivorum and the onion exudates did not stimulate the growth of four other fungi. This and other evidence is considered to show that the stimulation of growth of S. cepivorum was not caused by any direct effect on the antibiotics but by a tolerance of the fungus to them, which was specifically induced by an exudate from its host plant. The phenomenon may be related to the reported reversal by onion extracts of the inhibitory effects of soil mycostasis on germination of sclerotia of the fungus.  相似文献

Aspergillus nidulans was grown on media with added amounts of manganese ranging from 0–2.5 μM. Manganese deficiency prevented cleistothecium development, although good vegetative growth was retained. Subsequent analysis of the mycelium produced under Mn2+ deficient growth revealed that α-1,3 glucan, the main carbon and energy source for fructification, was virtually absent from the cell wall. Several enzymes related to cell wall composition were investigated. β-1,3 glucanase, and very remarkably, α-1,3 glucanase reached about the same activity on the Mn2+ deficient and sufficient media, but amylase and protease were about 60 and 75% lower respectively on the Mn2+ deficient media and the correlation of these findings is discussed.  相似文献

Summary The BS-21 strain of Streptomyces rimosus can utilize adenine, and guanine, as sole nitrogen sources, and in small concentrations, can increase the mycelium production in media containing threonine, ornithine and cysteine, further, it initiates growth in media containing valine, isoleucine, leucine and oxyproline. Pyrimidine bases are unsuitable as exclusive nitrogen sources, and unlike purine bases, do not increase growth in the presence of the nitrogen sources listed before. The yield of oxytetracycline follows the production rate of mycelium production.  相似文献

When improvingStreptomyces coeruleorubidus JA 10092, a producer of antibiotics of the daunomycinone complex, the most active variants were found among isolates of morphological typesbld-1 (with a suppressed production of the aerial mycelium on organic media containing glucose) andwhi (with an asporogenic aerial mycelium on glucose media and with the bald phenotype on media containing starch). Submerged cultures of thewhi mutants produced increased quantities of daunomycinone glycosides in the antibiotic complex, the amount of free anthracyclinones being simultaneously decreased. Thewhi strains differed from the wild type also in higher demands for aeration, concentration of glucose and in an increased production capacity in starch media. The overall antibiotic activity increased more than 40 times after a six-step selection (application of UV light, γ-radiation, nitrous acid and natural spreads) combined with an altered fermentation technology.  相似文献

Agaricus bisporus grew well in submerged culture in a medium containing malt extract, phosphate, and casein. Moderate growth occurred in defined media containing glucose, asparagine, phenylalanine, vitamins and minerals. Other amino acids did not stimulate growth. Growth was stimulated by vegetable oils, partly due to utilization of the oils, and partly to a more complex mechanism. Oleates had the same effect as vegetable oils; palmitates a lesser one. In shaken flasks maximum yield was reached after 22–24 days and in stirred and aerated fermentors after 8–10 days. Besides dry weight of mycelium, laccase activity was determined. The latter determination is suitable for a rapid estimation of the growth in routine experiments. The flavour of the mycelium was like that of mushrooms but weaker. It was strongest in standing liquid cultures and on solid media. The mycelium grown in submerged culture was suitable as spawn for mushroom culture. Summary Heat treatment of Phycomyces sporangiospores, which breaks dormancy, causes a very rapid 10- to 15fold increase in trehalase activity; soon after the heat shock the enzyme activity decays. This phenomenon can be repeated several times by repeating the heat shocks. Prolonging the heat treatment over the minimum required time delays the decay of enzyme activity. Cycloheximide does not prevent the rise in enzyme activity. It is suggested that heat treatment converts temporarily an inactive form of trehalase into an active one. Optimal enzyme activity is obtained at pH 7.5 and the enzyme requires metal ions for maximal activity. The possible role of trehalase in the spore-activation process is discussed.  相似文献

An assessment of the influence of water nanosilver suspension was made at a concentration of 10 mg·L?1 on biological material (i.e., vegetative mycelium and conidia of Isaria fumosorosea entomopathogen) on a background of the silver nitrate ionic form used. Conidia of I. fumosorosea treated with silver nitrate for more than 168 h were completely deactivated. The application of nanosilver on Isaria hyphae resulted in a quantitative limitation of mycelium growth and its weaker sporulation after culturing compared to the control. The pathogenicity of a conidial suspension obtained from such culturing towards test insects did not diverge from that observed in the standard culture. No obvious toxic effects of nanosilver were observed on I. fumosorosea conidia. Isaria conidia, after exposure to nanosilver over a period between 1 and 800 h, initially demonstrated weaker vegetative mycelium formation in culture on solid medium and, as a consequence, this mycelium often sporulated in a poorer manner. In one case, there was a significant stimulation of the sporulation process for nanosilver treatment before culture for 168 h. Concurrently, conidial suspensions obtained from the culture after exposure of over 168 h to nanosilver exhibited enhanced pathogenicity towards test insects, which may be considered a beneficial phenomenon in terms of the function played by Isaria in the whole environment. The reaction of conidia with nanosilver indicates the deactivation of conidia cells in suspensions and a possibility of selection in increased pathogenicity.  相似文献

Application of ethephon slightly increased the growth of hyphae of Botrytis cinerea. A competitive inhibitor of ethylene binding, 2,5-norbornadiene (NBD), inhibited growth of hyphae and mycelium and retarded the development of Botrytis cinerea. Transfer of the mycelium from an atmosphere containing NBD to air relieved the inhibition, indicating that the NBD effects were non-toxic and reversible. Addition of exogenous ethylene to an atmosphere containing NBD (20 ml 1-1) effectively reduced the inhibition. Inhibition due to 40 ml 1–1 NBD was not relieved by ethylene at any of the concentrations tested; however, a positive effect of ethylene appeared following transfer of the mycelia to air. The trehalase I of Dictyostelium discoideum exhibits characteristics of a typical lysosomal enzyme. The enzyme is glycosylated and carries a number of negatively charged components which cause it to be a very acidic protein. Strain M31, bears a recessive mutation mod A which alters the post-translational modification of several lysosomal enzymes including trehalase. A direct consequence of this mutation is a reduction of the negatively charged components on lysosomal enzymes. This reduction in negativity is observed in the altered chromatographic and electrophoretic behaviour of M31 trehalase.Trehalase I is synthesized during spore germination. Tunicamycin prevents the formation of recoverable trehalase from germinating spores but does not interfere with the germination process. These results indicate that the trehalase I synthesized during spore germination is not required for the successful completion of spore germination. Minor modification in the glycosylation, as seen in strain M31, does not affect the enzymatic activity. However, when glycosylation is greatly reduced by tunicamycin the enzyme is inactive.  相似文献

We cloned the Kluyveromyces lactis KlNTH1 gene, which encodes neutral trehalase. It showed 65.2% and 68.5% identity at nucleotide and amino acid sequence level, respectively, with the Saccharomyces cerevisiae NTH1 gene. Multiple alignment of the predicted trehalase protein sequences from yeasts, bacteria, insects, and mammals revealed two major domains of conservation. Only the yeast trehalases displayed in an N-terminal extension two consensus sites for cAMP-dependent protein phosphorylation and a putative Ca2+-binding sequence. Gene disruption of the KlNTH1 gene abolished neutral trehalase activity and clearly revealed a trehalase activity with an acid pH optimum. It also resulted in a high constitutive trehalose level. Expression of the KlNTH1 gene in an S. cerevisiae nth1Δ mutant resulted in rapid activation of the heterologous trehalase upon addition of glucose to cells growing on a nonfermentable carbon source and upon addition of a nitrogen source to cells starved for nitrogen in a glucose-containing medium. In K. lactis, the same responses were observed except that rapid activation by glucose was observed only in early-exponential-phase cells. Inactivation of K. lactis neutral trehalase by alkaline phosphatase and activation by cAMP in cell extracts are consistent with control of the enzyme by cAMP-dependent protein phosphorylation.
