Use of stream-centered dielectrophoresis (1–4) produced continuous separations on three cell mixtures (1)Chlorella vulgaris withNetrium digitus, (2)Ankistrodesmus falcatus withStaurastrum gracile, and (3)Saccharomyces cerevisiae withNetrium digitus. Maximal separations were obtained for these mixtures of live cells at 100 kHz, 600 kHz, and 2.0 MHz, respectively. The technique was restricted to a frequency range of 0.01–32 MHz, and to suspensions of low conductivity in which microorganisms such as these algae and yeast are tolerant. Extension, however, to cellular organisms requiring higher osmolarity is readily feasible through the use of nonionic solutes such as sucrose, mannose, glycine, etc.  相似文献

The unicellular, simply shaped desmid Netrium digitus inhabiting acid bog ponds grows in two phases. Prior to division, the cell elongates at its central zone, whereas in a second phase, polar tip growth occurs. Electron microscopy demonstrates that Netrium is surrounded by a morphologically homogeneous cell wall, which lacks pores. Immunocytochemical and biochemical analyses give insight into physical wall properties and, thus, into adaptation to the extreme environment. The monoclonal antibodies JIM5 and JIM7 directed against pectic epitopes with different degrees of esterification label preferentially growing wall zones in Netrium. In contrast, 2F4 marks the cell wall only after experimental de-esterification. Electron energy loss spectroscopy reveals Ca-binding capacities of pectins and gives indirect evidence for the degree of their esterification. An antibody raised against Netrium mucilage is not only specific to mucilage but also recognizes wall components in transmission electron microscopy and dot blots. These results indicate a smooth transition between mucilage and the cell wall in Netrium.  相似文献

Aucuba japonica varieties are common evergreen understory shrubs in Japan.Aucuba japonica var.borealis is distributed on the Sea of Japan side of Honshu and Hokkaido where heavy snow cover lasts for more than 3 months in winter.Aucuba japonica var.japonica is distributed in areas with shallow or no snow on the Pacific Ocean side of Honshu and Shikoku. The ecophysiological characteristics of var.borealis were compared with those of var.japonica to examine the effects of heavy and long-term snow cover on the life cycle of var.borealis. Shoots of both varieties were shaded in crushed ice for 110 days, but their photosynthetic activities, chlorophyll contents and the chlorophylla/b ratio was not affected. The leaves of var.borealis were no less frost tolerant than those of var.japonica. In spite of the difference in environmental factors, both varieties had similar characteristics in seasonal changes of photosynthesis, respiration and chlorophylla/b ratio. These results suggest that var.japonica could survive in areas with heavy snow where it does not normally occur. Leaf net production (LNP) was estimated based on the microclimatic data and seasonal photosynthetic and respiration rates. The difference in the annual LNP between the two varieties was equivalent to the difference in the LNP during the snow season. One of the major effects of snow cover is to interrupt and reduce the production period of var.borealis.  相似文献

Pichia amethionina varieties have different sensitivity to the acidity of the medium: P. amethionina var. amethionina has the optimum pH 3 for agglutination and 5 for conjugation; P. amethionina var. pachycereana has the optimum pH 4-6 for agglutination and 6 for conjugation. The optimum temperature for the both organisms is 24 degrees C. Under the optimum conditions, P. amethionina var. amethionina has a 98% agglutination and a 28% conjugation; for P. amethionina var. pachycereana, these values are 83 and 55%, respectively. The agglutination of the two varieties does not depend on the presence of glucose. The conjugation of P. amethionina var. pachycereana does not occur in a medium without glucose while the conjugation of P. amethionina var. amethionina is reduced twofold under these conditions. Inositol, biotin and folic acid produce positive effect on the conjugation of P. amethionina var. pachycereana, but only folic acid favours the conjugation of P. amethionina var. amethionina. No differences have been found between the varieties by staining their cells and zygotes with FITC-ConA.  相似文献

The levitation of lone live cells by means of dielectrophoretic force provides a means of determining the relative polarization of the cells and their aqueous support medium. When done over a range of frequencies, a spectrum of dielectric (polarization) responses is obtained which serves to characterize a single living ell. In this manner, individual cells of several microorganisms,Saccharomyces cerevisiae andNetrium digitus, were investigated in the frequency range 102 to 106 Hz. These and related prior studies showed that the positive dielectrophoresis, i.e. where the suspended particle has a greater net polarization than the suspending medium, can be — used to show subtle differences between species, and even between cells of the same culture when using this technique.  相似文献

Field observations and inoculation experiments revealed that Uromyces appendiculatus var. azukicola has an autoecious and macrocyclic life cycle and produces spermogonia, aecia, uredinia, and telia on Vigna angularis var. angularis and V. angularis var. nipponensis. From inoculation experiments, it was suggested that this rust fungus has different host relationships from other varieties. Conditions are described whereby suspensions of Chlorella pyrenoidosa and Netrium digitus photosynthetically biosynthesize and excrete glycolate continuously in high yields. Aminooxyacetic acid, an inhibitor of pyridoxal phosphate-linked enzymes, increased the excretion of glycolate approximately 4-fold in 1 hour (8 millimolar) and 20-fold in 4 hours (40 millimolar) in the presence of 0.2% CO2 in air. The amount of glycolate excreted in the presence of aminooxyacetate and an atmosphere of 0.2% CO2 in air equaled or exceeded the amount excreted in 0.2% CO2 in O2 minus aminooxyacetate. CO2 and light were required for glycolate excretion. Aminooxyacetate also stimulated photosynthetic glycolate excretion in an atmosphere of 0.2% CO2 in nitrogen or helium, although the stimulation was not as great as when air or O2 was present.

The excreted glycolate was converted to H2 and CO2 by the combined action of glycolic oxidase and the formic hydrogenlyase complex found in Escherichia coli in total conversion yields of 80%.

Aim Ponderosa pine (Pinus ponderosa Douglas ex Lawson & C. Lawson) is an economically and ecologically important conifer that has a wide geographic range in the western USA, but is mostly absent from the geographic centre of its distribution – the Great Basin and adjoining mountain ranges. Much of its modern range was achieved by migration of geographically distinct Sierra Nevada (P. ponderosa var. ponderosa) and Rocky Mountain (P. ponderosa var. scopulorum) varieties in the last 10,000 years. Previous research has confirmed genetic differences between the two varieties, and measurable genetic exchange occurs where their ranges now overlap in western Montana. A variety of approaches in bioclimatic modelling is required to explore the ecological differences between these varieties and their implications for historical biogeography and impending changes in western landscapes. Location Western USA. Methods We used a classification tree analysis and a minimum‐volume ellipsoid as models to explain the broad patterns of distribution of ponderosa pine in modern environments using climatic and edaphic variables. Most biogeographical modelling assumes that the target group represents a single, ecologically uniform taxonomic population. Classification tree analysis does not require this assumption because it allows the creation of pathways that predict multiple positive and negative outcomes. Thus, classification tree analysis can be used to test the ecological uniformity of the species. In addition, a multidimensional ellipsoid was constructed to describe the niche of each variety of ponderosa pine, and distances from the niche were calculated and mapped on a 4‐km grid for each ecological variable. Results The resulting classification tree identified three dominant pathways predicting ponderosa pine presence. Two of these three pathways correspond roughly to the distribution of var. ponderosa, and the third pathway generally corresponds to the distribution of var. scopulorum. The classification tree and minimum‐volume ellipsoid model show that both varieties have very similar temperature limitations, although var. ponderosa is more limited by the temperature extremes of the continental interior. The precipitation limitations of the two varieties are seasonally different, with var. ponderosa requiring significant winter moisture and var. scopulorum requiring significant summer moisture. Great Basin mountain ranges are too cold at higher elevations to support either variety of ponderosa pine, and at lower elevations are too dry in summer for var. scopulorum and too dry in winter for var. ponderosa. Main conclusions The classification tree analysis indicates that var. ponderosa is ecologically as well as genetically distinct from var. scopulorum. Ecological differences may maintain genetic separation in spite of a limited zone of introgression between the two varieties in western Montana. Two hypotheses about past and future movements of ponderosa pine emerge from our analyses. The first hypothesis is that, during the last glacial period, colder and/or drier summers truncated most of the range of var. scopulorum in the central Rockies, but had less dramatic effects on the more maritime and winter‐wet distribution of var. ponderosa. The second hypothesis is that, all other factors held constant, increasing summer temperatures in the future should produce changes in the distribution of var. scopulorum that are likely to involve range expansions in the central Rockies with the warming of mountain ranges currently too cold but sufficiently wet in summer for var. scopulorum. Finally, our results underscore the growing need to focus on genotypes in biogeographical modelling and ecological forecasting.  相似文献

SYNOPSIS. Relationships between the cell cycle and the beginning of conjugation were analyzed for 3 hypotrichs: Diophrys scutum, Oxytricha bifaria, and Euplotes crassus. The first 2 species enter conjugation with micronuclei in G1; the latter species with a micronucleus in G2. The 1st micronuclear division of conjugating E. crassus is mitotic. Thus meiotic DNA replication occurs when the cells of each species have already entered the mating process. Cells from asynchronous populations start conjugation with their macronuclei primarily in G1 or more rarely at the beginning of the S stage in a percentage significantly different from that expected on the basis of random mating among all cells in the population. Also, macronuclear replication, when already begun, was blocked in cells undergoing conjugation. Therefore only the G1 or the very early S stages of the cell cycle are compatible with conjugation in the 3 analyzed species.  相似文献

Random amplified polymorphic DNA (RAPD) analysis was used to assess the level of genetic diversity in an endangered plant Cerastiumfischerianum var. molle and the widespread conspecific C. fischerianum var. fischerianum. At both the variety and the population level, C. fischerianum var. molle had a lower level of RAPD variation than var. fischerianum. Analysis of molecular variance ( AMOVA ) showed that most of the genetic diversity exists among populations for both varieties. Genetic differentiation among populations probably resulted from the highly selfing habits of both varieties. To conserve C. fischerianum var. molle genetically, preservation of only a part of the population is insufficient.  相似文献

Using the ELISA method, the development of Pseudocercosporella herpotrtchoides var. herpotrichoides and var. acuformis in a susceptible cultivar of winter wheat was compared under controlled and held conditions. In the greenhouse, var. acufornis grew less vigorously, was slower in penetrating the coleoptile and the successive leaf sheaths and in colonizing the stem tissue than var. herpotrichoides. In the field, these differences were confirmed on the last leaf sheaths and the stem. At ripening stage, however, identical ELISA values were measured for both varieties. Moreover, a significant variation was observed between the individual isolates of each variety. Comparison of the effect of both varieties of P. herpotrichoides on 20 wheat cultivars characterized by different resistance levels showed significant interactions. The cultivars carrying the Pch-1 gene always remained the less diseased genotypes. In general, var. acuformis developed less antigen in the cultivars than var. herpotrichoides. It is concluded, that in tests for resistance to P. herpotrichoides mixtures of many highly pathogenic isolates of both fungus varieties should be used. Less complex mixtures or single isolates may result in wrong estimates of resistance.  相似文献

Abstract Although rice has long been recognized to be uniquely adapted for growth in low oxygen environments of flooded rice fields, rice weeds of the Echinochloa crus-galli complex appear to be at least as well specialized for germination and growth under such unusual biological conditions. Seeds of two varieties of E. crus-galli germinate and grow for prolonged periods in a totally oxygen-free environment. E. crus-galli germinates as well as rice (Oryza sativa) under a total nitrogen atmosphere and produces as large a seedling in spite of its much smaller seed size. Like rice, the seedlings of E. crus-galli are unpigmented, the primary leaves do not emerge from the coleoptile and no root growth occurs without oxygen. Of particular interest is the ultrastructure of mitochondria from anaerobically-grown seedlings. Mitochondrial profiles from the primary leaf of seedlings grown continuously in nitrogen are very similar to those grown aerobically. The size and shape of the mitochondria are similar and the cristae are numerous and normal in appearance. This is in sharp contrast to previous studies of other species which have reported that mitochondria were vesiculate and tended to lose their normal fine-structure after similar periods without oxygen. Finally, based on ultrastructure and 14C labeling studies, anaerobically-grown seedlings are highly active metabolically, which may explain, at least for E. crus-galli var. oryzicola, its ability to germinate and emerge from flooded rice fields.  相似文献

Abstract

Fusarium moniliforme Sheld. var. subglutinans Wr. et Rg. causes malformations in young inflorescences of mango and seriously affects fruit formation. Quantitative studies of total, soluble and protein nitrogen contents of malformed inflorescence of two cultivars, ‘Himsagar’ and ‘Bombai’ were made. Healthy inflorescences of the former cultivar showed slightly higher nitrogen content than that of the latter. After infection, considerable increases in both soluble and protein fractions of nitrogen were observed in both host varieties. The proportion of these two nitrogen fractions, however, showed some changes in ‘Bombai’ but in ‘Himsagar’ it remained more or less unaltered.  相似文献

我们从江苏、浙江两省采到的一些样品中分离得到六株毛霉菌种并鉴定为分别属于三个变种的同一个新种,即单孢共头霉原变种(Syncephalastrum monosporum Zheng et al.sp. nov. var. monosporum),单孢共头霉冠囊变种(Syncephalastrum monosporum Zheng et al. var. cristatum Zheng et al. var. nov.),以及单孢共头霉多重生变种(Syncephalastrum monosporum Zheng et al. var. pluriproliferum Zheng et al. var. nov.).它们与共头霉属的唯一已知种总状共头霉(Syncephalastrum racemosum Cohn ex Schroeter)的主要区别在于全部孢子囊都是单孢的小型孢子囊(柱孢囊)而不是像后者一样孢子成单行排列的多袍柱孢囊.我们把这些菌归人共头霉属而没有为它们专门成立一个新属的理由除因它们与总状共头霉有明显的亲缘关系外,主要还因我们认为在不具备其它重要区别特征的情况下,在毛霉目的分类中,无论在科级或属级的水平上,都不应过分强调单孢孢子囊的作用.尽管单孢共头霉是毛霉目内唯一具单孢子柱孢囊的种,我们仍然把它们归入共头霉属内.至于我们为什么要把这六株菌鉴定为一个种的三个变种而不是三个各自独立的种,则是由于我们承认共头霉属的唯一已知种总状共头霉是一个变异性很大的种而并没有把它分成许多不同的种,因此,我们对这个属的另外一个种,即单孢共头霉同样采用较大的种概念.  相似文献

Five sympatric colour varieties of the hoplonemertean Tetrastemma nigrifrons (var. pallidum, var. bicolor, var. purpureum, var. punctata, var. albino) were found in Vostok Bay (the Sea of Japan). The taxonomic status of the varieties, some of which have been known for almost a century, was uncertain. Starch gel electrophoresis was used to compare allele frequencies of these varieties at 19 isozyme loci. Neither fixed allelic differences nor statistically significant differences in allele frequencies between different colour varieties was revealed. It was concluded therefore that all the colour varieties studied are conspecific. As estimated from the electrophoretic screening of 24 isozyme loci, the species is highly variable at the molecular level. It has high mean values of observed and expected heterozygosity estimates, H o =0.313 ± 0.049 and H e =0.323 ± 0.050, and is one of the most variable species among invertebrates.  相似文献

The annual hemiparasiteEuphrasia stricta occurs on Gotland in two early-flowering meadow varieties.E. stricta var.suecica is on the Swedish red-list as endangered, occurring in Sweden only on the Baltic island of Gotland. It probably has near relatives east of the Baltic proper. The other variety,E. stricta var.tenuis, has a wider distribution occurring almost all over Sweden, but is declining in abundance. Both varieties have close morphological similarities and habitat preferences, raising questions about which level conservation efforts should be focused on, varieties or populations. In this study we describe the genetic structure between and within these two varieties using amplified fragment length polymorphism, AFLP.F ST between varieties is 0.14, and between populations within the varietiesE. stricta var.suecica and var.tenuis F ST is 0.60 and 0.26 respectively. The partitioning of gene diversity to different levels shows that 14% of the genetic diversity occurs between varieties, 42% between populations within varieties, and 44% within populations. Significant genetic differentiation was detected between varieties, populations and subpopulations within populations using a constrained principal coordinate analysis. We suggest that all of the existing populations of these two varieties on Gotland should be preserved, since they are few (6E. stricta var.suecica and 5E. stricta var.tenuis) and much of the genetic diversity is partitioned between populations.  相似文献

Argania spinosa includes two varieties, var. apiculata and var. mutica. These argan varieties were introduced into Tunisia in ancient times and are actually cultivated in some botanic gardens. Little is known about the chemical differentiation among these argan varieties. Hence, the aim of this study was to determine the fatty‐acid composition, the total phenolic and flavonoid contents, and the antioxidant and α‐amylase‐inhibitory activities of leaf, seed, and pulp extracts of both argan varieties harvested during the months of January to April. The fatty‐acid distribution was found to depend on the argan variety, the plant organ, and the harvest time. Significant variations in the phenolic contents were observed between the investigated varieties as well as between leaves, pulps, and seeds of each variety. As expected, phenolic compounds were found to be contributors to the antioxidant and α‐amylase‐inhibitory activities of both argan varieties. The chemical differentiation observed among the two argan varieties, based mainly on the fatty‐acid composition, might have some chemotaxonomic value.  相似文献

The evolutionary and functional relationships among breeding systems and floral morphology were investigated in the Turnera ulmifolia complex. Predictions of a model of breeding system evolution among distylous and homostylous varieties were tested. Chromosome counts of 73 accessions revealed an association between breeding system and chromosome number. Diploid and tetraploid populations of five taxonomic varieties are distylous and self-incompatible, whereas hexaploid populations of three varieties are homostylous and self-compatible. The latter occur at different margins of the geographical range of the complex. Crossing studies and analyses of pollen and ovule fertility in F1's revealed that the three homostylous varieties are intersterile. To test the prediction that, homostylous varieties are long homostyles that have originated by crossing over within the distyly supergene, a crossing program was undertaken among distylous and homostylous plants. Residual incompatibility was observed in styles and pollen of each homostylous variety with patterns consistent with predictions of the cross-over model. The intersterility of hexaploid varieties suggests that long homostyly has arisen on at least three occasions in the complex by recombination within the supergene controlling distyly. Deviation from expected compatibility behavior occurs in populations of var. angustifolia that have the longest styles. These phenotypes displayed the greatest separation between anthers and stigmas (herkogamy) and set little seed in crosses with long- or short-styled plants. This suggests that they are derived from long homostyles with shorter length styles. It is proposed that selection for increased outcrossing has favored the evolution of herkogamy in long homostyles. Estimates of outcrossing rate in a distylous population using allozyme markers confirmed that dimorphic incompatibility enforces complete outcrossing. Significant genetic variation for floral traits likely to influence the mating system, such as stigma-anther separation, occurs within and among homostylous populations of var. angustifolia on Jamaica. Estimates of the mating system of families from a population with varying degrees of stigma-anther separation, using five isozyme loci, were heterogeneous and ranged from t = 0.04–0.79. Families exhibiting the largest mean stigma-anther separation have higher outcrossing rates than those with little separation.  相似文献

Floral traits such as color and size are highly diversified in lilies, but their adaptive significance remains uncertain. In the present study, we compared pollination processes between Lilium japonicum var. abeanum and var. japonicum to clarify how the two varieties are adapted to different pollinators. Lilium japonicum var. japonicum is known to be pollinated by moths, and we hypothesized from its flower traits that var. abeanum is pollinated by diurnal insects. Using waterproof digital cameras set to a recording interval of 10–30 s, we recorded flower visitors for 7–9 flowers of var. japonicum and 4–6 flowers of var. abeanum over 24‐h periods. We also recorded the number of versatile (T‐shaped) and rigid (I‐shaped) anthers per flower. For var. abeanum, we observed flowers at 05.00 and 17.00 hours to determine flower opening time and measured the intensity of floral scent using a metal‐oxide semi‐conductor odor sensor. Both diurnal and nocturnal insects visited flowers of the two varieties, but visitation of diurnal insects was more frequent in var. abeanum. Anthers of var. abeanum are usually rigid, as in many bee‐pollinated flowers, whereas those of var. japonicum are mostly versatile. Although flowers of var. japonicum are known to open in the evening, 32% of the flowers of var. abeanum started to flower during the day. Lilium japonicum var. abeanum emitted scent at night, with a maximum intensity at 20.00 hours, just as in var. japonicum. These findings suggest that the floral traits of the two varieties are adapted to the different relative availabilities of nocturnal versus diurnal pollinators. The flower of var. abeanum is more adapted to diurnal pollinators, but retains adaptations to nocturnal pollinators, particularly with regard to its scent.  相似文献
