Soybean plants grown in controlled environment cabinets under light intensities of 220 w/m2 or 90 w/m2 (400–700 nm) and day to night temperatures of 27.5–22.5 C or 20.0–12.5 C in all combinations, exhibited differences in growth rate, leaf anatomy, chloroplast ultrastructure, and leaf starch, chlorophyll, and chloroplast lipid contents. Leaves grown under the lower light intensity at both temperatures had palisade mesophyll chloroplasts containing well-formed grana. The corresponding leaves developed under the higher light intensity had very rudimentary grana. Chloroplasts from high temperature and high light had grana consisting of two or three appressed thylakoids, while grana from the low temperature were confined to occasional thylakoid overlap. Spongy mesophyll chloroplasts were less sensitive to growth conditions. Transfer experiments showed that the ultrastructure of chloroplasts from mature leaves could be modified by changing the conditions, though the effect was less marked than when the leaf was growing.  相似文献

Statistically significant differences in both HRA and MC chloroplast ultrastructure were observed between different stages of leaf ontogeny. Growth of plastoglobuli was the most striking characteristic of chloroplast maturation and senescence. The chloroplasts in mature and senescing leaves had a more developed system of thylakoids compared to the young leaves. Illus. 1962.—Development of the form and anatomy of leaves was studied in plants of Pisum sativum grown in vermiculite under constant conditions and exposed daily to red or white light or kept in continuous darkness. The red light used had an intensity in the morphogenetically active red region of the spectrum of 70–75% that of the white light. Light had no effect on the manner of initiation or early development of leaf primordia. Quantitative data from older leaves showed that light has no effect on the pattern of later development but does affect the rate and extent of development. Under all light conditions, the length of the leaflet is closely correlated with the state of its internal anatomy. “Mature” etiolated leaves duplicate young stages of light-grown leaves. Mature leaves grown in red light duplicate not-quite-mature leaves grown in white light. The difference between white-light and red-light leaves is attributed here to light intensity and resembles that between sun and shade leaves.  相似文献

Leaf mesostructure, photochemical activity, and chloroplast photophosphorylation (PP) in the fourth true leaf of 28-day-old Chinese cabbage (Brassica chinensis L.) plants were investigated. Plants were grown under a light source based on red (650 nm) and blue (470 nm) light-emitting diodes (LED) with red/blue photon flux ratio of 7: 1 and under illumination with high-pressure sodium lamp (HPSL) at photon flux densities of 391 ± 24 μmol/(m2 s) (“normal irradiance”) and 107 ± 9 μmol/(m2 s) (“low irradiance”) in photosynthetically active range. At normal irradiance, the leaf area in plants grown under HPSL was twofold higher than in LED-illuminated plants; other parameters of leaf mesostructure were little affected by spectral quality of incident light. The lowering of growth irradiance reduced the majority of leaf mesostructure parameters in plants grown under illumination with HPSL, whereas in LED-illuminated plants the lowered irradiance reduced only specific leaf weight but increased the leaf thickness and dimensions of mesophyll cells and chloroplasts. The photochemical activity of isolated chloroplasts was almost independent of growth irradiance and light spectral quality. Light quality and intensity used for plant growing had a considerable impact on PP in chloroplasts. At normal light intensity, the highest activity of noncyclic PP in chloroplasts was observed for plants grown under HPSL; at low light intensity the highest rates of PP were noted for plants grown under LED. The P/2e ratio, which characterizes the degree of PP coupling to electron transport in the chloroplast electron transport chain, showed a similar pattern. Thus, the narrow-band spectrum of the light source had little influence on leaf mesostructure and electron transport rates. However, this spectrum significantly affected the chloroplast PP activity. The PP patterns at low and normal light intensities were opposite for plants grown under LED and HPSL light sources. We suppose that growing plants under LED array at normal light intensity disturbed the chloroplast coupling system, thus preventing the effective use of light energy for ATP synthesis. At low light intensity, chloroplast PP activity was significantly higher under LED illumination, but plant growth was suppressed because of impaired adaptation to low light intensity.  相似文献

The influence of excess irradiance on resistance of wheat (Triticum aestivum L.) photosynthetic apparatus to heating in darkness and in the light was investigated and compared with changes in leaf cell ultra-structure and composition of cell lipids and fatty acids. The leaves of 14- to 16-day-old plants grown at low irradiance (about 20 W/m2) were exposed for 1 h to irradiance of 370 or 600 W/m2 PAR. Using infrared gas analysis, we found that the preexposure of leaves to excess irradiation elevated resistance of apparent photosynthesis to 10-min heat treatment at 40–45°C. The rate of Hill reaction (reduction of 2,6-dichlorophenolindophenol by isolated chloroplasts) was higher for leaves heated at high irradiance than for leaves heated in darkness. During illumination of leaves with strong light, mesophyll cells became more abundant in mitochondria and peroxysomes, as well as in cisternae of endoplasmic reticulum and Golgi complex. The chloroplast thylakoids and grana became more extensive and numerous. At the same time, the leaf content of main classes of membrane glycerolipids increased in parallel with the increase in the phospholipid/glycolipid and lipid/chlorophyll ratios. The unsaturation index of fatty acids of membrane lipids increased because of the elevated content of linolenic acid. Thus, excessive light (not fully utilized in photosynthesis) induced in wheat leaves a series of nonspecific adaptive changes that were similar to those occurring under the action of other environmental factors, such as heat shock, cooling, salinity, and osmotic stresses.  相似文献

Cyanidium caldarium M-8 type grown in the dark was illuminatedfor 3 days, and changes of its cell and cell organelle structuresand of photosynthetic activity were observed quantitatively.Dark grown (DG) cells showed no photosynthetic activity andno phycocyanin. During 3 day illumination they fully recoveredtheir photosynthetic activities as measured by Hill reaction,and also synthesized chlorophyll a and phycocyanins. Sizes ofcell, cell nucleus, chloroplast and its nucleoid observed byfluorescence microscopy after staining with DAPI increased simultaneouslyupon illumination. The chloroplast and its ring shaped nucleoidsizes increased especially rapidly, concomitant with the recoveryof Hill activity. In fully recovered cells after 3 days, a goodcorrelation was found among the sizes of cells, chloroplastsand chloroplast nucleoids. ( Revision received June 28, 1986. The effect of benzyladenine (BA) on the diurnal changes in DNAand Chl contents per chloroplast and chloroplast replicationin primary leaves of bean plants (Phaseolus vulgaris L.) grownunder a 16 h light/8 h dark cycle was studied. Experiments weremade on primary leaves in the early expansion phase, where celldivision had been completed but chloroplasts were replicating.In untreated controls, chloroplast number, Chl content and freshweight per leaf showed daily periodic changes. Chl content perchloroplast increased in the light period every day, and freshweight per leaf increased most rapidly in the early dark period.Chloroplast number per leaf increased rapidly in the early darkperiod on day 9, though the increase began a little earlierand was less sharp on days 8 and 10. During these periods, DNAcontent per chloroplast was decreasing due to chloroplast divisionas chloroplast DNA (ctDNA) per leaf remained unchanged throughoutthe experimental period. BA induced increases in Chi contentper chloroplast, ctDNA content and fresh weight per leaf within6 h of its application, regardless of whether it was appliedat or 10 h after the beginning of the light period. Applicationof BA at 10 h in the light period shifted the start of chloroplastreplication by 6 h compared to that in untreated controls. However,when BA was applied at the beginning of illumination, the startof chloroplast replication showed the same relative change intime as above. 5-Fluorodeoxyuridine (5-FdU) promptly preventedBA-induced increase in Chl content and chloroplast number perleaf as well as ctDNA content per leaf.  相似文献

Hoflacher, H. and Bauer, H. 1982. Light acclimation in leaves of the juvenile and adult life phases of ivy (Hedera helix). – Physiol. Plant. 56: 177–182. Light acclimation was investigated during the juvenile and adult life phases of the whole-plant-development in Hedera helix L. For this purpose, cuttings of the juvenile and adult parts of one single parent plant were grown under low-light (PAR 30–50 μmol photons m?2 s?1) and high-light (PAR 300–500 μmol m?2 s?1) conditions: CO2 exchange, chloroplast functions, and specific anatomy of fully developed leaves differentiated under these conditions were determined. In juvenile plants the leaves formed under low and high light had light-saturated rates of net photosynthesis of 6.5 and 11.1 mg CO2 (dm leaf area)?2 h?1, respectively. In adult plants the rates were 9.4 and 22.2 mg dm?2 h?1, indicating a more pronounced capacity for acclimation to strong light in the adult life phase. Higher photosynthetic capacities were accompanied by higher conductances for the CO2 transfer through the stomata, leading to almost the same CO2 concentration in the intercellular spaces. Thus, stomatal conductances were not primarily responsible for the different photo-synthetic capacities. The higher rates in adult and high-light grown leaves were mainly the result of formation of thicker leaves with more chloroplasts per unit leaf area. Expressed per chloroplast, the photosynthetic capacity, the Hill reaction, and the activity of ribulose bisphosphate carboxylase were almost identical in plants grown in low-light and high-light. Measurements of photosynthetic capacity and thickness of leaves of Hedera sampled from field habitats with contrasting light regimes confirm the results of growth chamber studies. It is, therefore, concluded that both life phases of Hedera are capable of acclimating to strong light, but that during the juvenile phase this capacity is not fully developed.  相似文献

The effect of natural shading on photosynthetic capacity and chloroplast thylakoid membrane function was examined in soybean (Glycine max. cv Young) under field conditions using a randomized complete block design. Seedlings were thinned to 15 plants per square meter at 20 days after planting. Leaves destined to function in the shaded regions of the canopy were tagged during early expansion at 40 days after planting. To investigate the response of shaded leaves to an increase in available light, plants were removed from certain plots at 29 or 37 days after tagging to reduce the population from 15 to three plants per square meter and alter the irradiance and spectral quality of light. During the transition from a sun to a shade environment, maximum photosynthesis and chloroplast electron transport of control leaves decreased by two- to threefold over a period of 40 days followed by rapid senescence and abscission. Senescence and abscission of tagged leaves were delayed by more than 4 weeks in plots where plant populations were reduced to three plants per square meter. Maximum photosynthesis and chloroplast electron transport activity were stabilized or elevated in response to increased light when plant populations were reduced from 15 to three plants per square meter. Several chloroplast thylakoid membrane components were affected by light environment. Cytochrome f and coupling factor protein decreased by 40% and 80%, respectively, as control leaves became shaded and then increased when shaded leaves acclimated to high light. The concentrations of photosystem I (PSI) and photosystem II (PSII) reaction centers were not affected by light environment or leaf age in field grown plants, resulting in a constant PSII/PSI ratio of 1.6 ± 0.3. Analysis of the chlorophyll-protein composition revealed a shift in chlorophyll from PSI to PSII as leaves became shaded and a reversal of this process when shaded leaves were provided with increased light. These results were in contrast to those of soybeans grown in a growth chamber where the PSII/PSI ratio as well as cytochrome f and coupling factor protein levels were dependent on growth irradiance. To summarize, light environment regulated both the photosynthetic characteristics and the timing of senescence in soybean leaves grown under field conditions.  相似文献

The responses of young tomato plants to infection with tomato spotted wilt virus were studied at constant temperatures. The plants were grown in nutrient solutions and given three daylength treatments.
The length of the incubation period was positively correlated with temperature and a regression equation for this relationship is given. The calculated temperature at which this period becomes infinite is 6.7°C. The temperature coefficient ( Q 10) varied from 1.7 to 2.9.
Dry-weight determinations were made of stems, leaves and roots. In healthy plants the leaf/stem ratio generally fell with rise of temperature and the shoot/root ratio rose. Infection changed these ratios, which is interpreted as indicating that it interfered with supplies of carbohydrate to the growing regions.
Systemic symptoms are described for plants raised at 11.4°, 18.3°, 23.9°, and 29.4°C.
Changes in daylength did not affect the systemic symptoms or incubation period. With a 9 hr. day at 29.4°C., the virus content of sap was significantly lower than with 12 or 15 hr., and with a 12 hr. day was significantly higher than with a 15 hr. day.  相似文献

The composition of free fatty acids (FFA) in relation to Hill reaction activity and photoperoxidation of lipids was studied in chloroplasts isolated from fresh, cold and dark-stored as well as illuminated leaves of Lycopersicon esculentum Mill., Phaseolus vulgaris L. and Cucumis sativus L. Following the cold and dark-storage of leaves the loss of Hill reaction activity is accompanied by approximately a 5-fold increase in the amount of FFA and by an increase in the percentage of unsaturated FFA, particularly that of linolenic acid. Illumination of the cold- and dark-stored leaves restores both Hill reaction activity and the content and composition of chloroplast FFA. Following the second and third cycles of cold storage and illumination of leaves the percentage of unsaturated fatty acids in chloroplasts increases while that of saturated ones decreases despite of the significant restoration of Hill reaction activity. Since the illumination of cold-stored leaves results in peroxidation of inhibitory fatty acids it seems likely that this phenomenon could, at least partially, be responsible for the restoration of Hill reaction activity. Inhibition of Hill reaction activity by exogenous linolenic acid in chloroplasts of fresh, cold-stored as well as cold-stored and illuminated leaves could be reversed following the incubation of chloroplast suspension with BSA, however only to a value measured in the absence of unsaturated fatty acid. Effect of 4-thiouridine, which was proved to inhibit selectively and “light-reversibly” the synthesis of chloroplast ribosomal RNAs in radish cotyledons, on the photo-induced development of photosystem I, II and a complete electron transport chain was investigated with plastids obtained from 4-thiouridine treated dark-grown radish cotyledons after various times of development in the light. It was demonstrated that the 4-thioridine treated chloroplasts showed a higher activity of photoreduction than the control untreated chloroplasts in every system on a chlorophyll basis during the development after 24 hr illumination. This specific activity decreased in both chloroplasts, as the chloroplasts matured with the time of illumination. The activity per g of fresh cotyledons treated with 4-thiouridine, especially in the early stage of development, was lower than that of ones untreated with the drug because total chlorophyll content was poor, but the activity of the former was enhanced with the increase of total chlorophyll content upon illumination while the activity of the latter decreased on 24 hr illumination. Moreover, Hill reaction measurements showed that 4-thiouridine treated chloroplasts were saturated at lower light intensity than untreated ones inspite of the same content of chlorophyll in both the chloroplasts: photoreduction of NADP+ was saturated at 3000 lux for the former and at 5000 lux for the latter. Based upon these results, specific development of the chloroplast is discussed.  相似文献

Data from 3 lines of investigation were presented indicating that chlorophyll is not necessary for the increase in the triphosphopyridine nucleotide-requiring triosephosphate dehydrogenase accompanying the illumination of etiolated pea plants. These include A) the kinetics of the development of chlorophyll and enzyme activity, B) the presence of enzyme activity in leaves grown in the dark on normal plants and C) the high specific enzyme activity in leaves of a chlorophyll-less mutant.It was also shown that the light-initiated increase of enzyme activity continues for several days after removal from the light and that illumination with far-red light before the dark period inhibited, but did not abolish, this increase. The ability of green plants to continue to produce the enzyme in the dark was eventually lost with time, for after 7 days in the dark a stimulation in leaf protein formation was not accompanied by an increase in enzyme activity.  相似文献

Equisetum scirpoides Michx., propagated from a single clone, was grown in a controlled growth chamber at 24 ± 1 C under a photoperiod of 16 hr light/8 hr darkness. The apical cell of aerial vegetative shoots gives rise to derivatives (merophytes) in a helical sequence. Each newly formed merophyte divides anticlinally to form two superposed cells that are parallel to a lateral face of the apical cell. Radial longitudinal divisions then take place in the two superposed cells. Shoot tips were fixed every 2 hr for 24 hr to determine the mitotic index of the apical cell, six subjacent cells, and the remaining cells above the level of leaf initiation. Average mitotic indices for the 24-hr period were 3.9%, 3.9%, and 7.0%, respectively. The results indicate that the apical cell is quite active mitotically; there was no clear evidence of endopolyploidy in cells of the shoot apex, young leaves or in the developing cortex, based upon cytophotometric measurements of DNA content.  相似文献

Reducing the light intensity under which plants were grown in summer to one-third increased their susceptibility to infection with tobacco necrosis, tomato bushy stunt, tobacco mosaic and tomato aucuba mosaic viruses. With the first two viruses shading increased the average number of local lesions per leaf by more than ten times and by more than five times with the second two.
Reducing the light intensity increased the virus content of sap from leaves inoculated with Rothamsted tobacco necrosis virus by as much as twenty times. As it also reduced the total solid content of sap by about one-half, purification was greatly facilitated; crystalline preparations of the virus were readily made from shaded plants but not from unshaded controls.
Reducing the light intensity also increased the virus content of systemically infected leaves; the greatest effect was with tomato bushy stunt virus with which increases of up to ten times were obtained, but with tobacco mosaic and aucuba mosaic viruses there were also significant increases.
The importance of controlled illumination in raising plants for virus work and the possible mechanisms responsible for the variations in susceptibility are discussed.  相似文献

Levels of chloroplast antioxidants and enzymes that scavenge oxygen racidals were followed in the leaves of pea plants (Pisum sativum L. cv. Meteor) grown under glasshouse conditions between April 1984 and May 1985. While little variation in pigment levels or superoxide dismutase activity was detected during this period, plants grown in early summer (May–June) contained appreciably higher levels of ascorbate, ascorbate peroxidase and glutathione reductase than plants grown in winter (Dec–Jan.). The role of light intensity in regulating levels of chloroplast antioxidants was examined further using pea plants grown in a constant environment chamber under 100 or 400 μmol m−2 s −1 photon flux density. Chloroplasts isolated from plants grown at the higher light intensity contained significantly higher levels of ascorbate, ascorbate peroxidase, glutathione reductase and dehydroascorbate reductase. These data suggest that light intensity may have an important influence on the level and activity of chloroplast antioxidants and oxygen radical scavenger enzymes.  相似文献

Plants have evolved various photoprotective mechanisms to mitigate photodamage. Here we report the diurnal movement of chloroplasts in the leaves of succulent crassulacean acid metabolism (CAM) plants under combined light and water stress. In leaves of water-stressed plants, the chloroplasts became densely clumped in one or sometimes two areas in the cytoplasm under light and dispersed during darkness. The chloroplast clumping resulted in leaf optical changes, with a decrease in absorptance and an increase in transmittance. The plant stress hormone abscisic acid induced chloroplast clumping in the leaf cells under light. We suggest that the marked chloroplast movement in these CAM plants is a photoprotective strategy used by the plants subjected to severe water stress.Abbreviations ABA Abscisic acid - CAM Crassulacean acid metabolism  相似文献
