Flangas , A. L., and J. G. Dickson . (U. Wisconsin, Madison.) The genetic control of pathogenicity, serotypes and variability in Puccinia sorghi. Amer. Jour. Bot. 48(4): 275–285. Illus. 1961.—Four urediospore clones of P. sorghi were inbred through the alternate host, Oxalis, and the recombinant lines were compared with the mother clones for changes in pathogenicity and serological reaction. The aecial, inbred recombinant lines were scored for pathogenicity on 6 inbred lines of maize, 4 of which contained single, dominant alleles isolated from a single gene locus each conditioning differential rust reaction and Pop 35 and Pop 36, each with 2 recessive genes. The inbred, recombinant lines from each of the 4 mother clones reflected similar dimensional changes from original pathogenicity, that is, an ultimate gain or loss in host range or specificity. Based on reaction type incited on the 4 allelic differential hosts, continued inbreeding through 3 generations did not segregate homozygous pathogenic complements into distinct pathogenic biotypes. A similar array of pathogenic types was produced after each generation of inbreeding. The suppressed pathogenicity types in the dicaryotic line were revealed only through recombination. Hybridization between dicaryotic lines of selected pathogenic types of sibling and nonsibling pedigree confirmed the indeterminate nature of the pathogenic locus. The genetic basis of the commonly occurring mesothetic-X reaction (necrotic flecks and uredia of varying size surrounded by chlorotic and necrotic areas) is explained by the heterologous complementarity of fit of the interacting genic systems (genetic units). The heterologous mesothetic-X reactions offer ambiguous definition to the specificity state of the interaction and cannot be explained on the discrete gene-for-gene basis of rust reaction. Antisera produced from the pathogenic types differentiated the homologous from the heterologous combinations and revealed cross-reactions with P. graminis avenae. The comparative antigen constitution of the urediospore clones and the sibling recombinants thus far reveal a possible parallel existing between the modes of inheritance of pathogenic specificity and antigenic specificity.  相似文献

Five levels of disease expression were scored in a cross-inoculation study of 120 host and 60 pathogen lines of wild flax Linum marginale and its rust fungus Melampsora lini sampled from six natural populations. Patterns of partial resistance showed clear evidence of gene-for-gene interactions, with particular levels of partial resistance occurring in specific host-pathogen combinations. Sympatric and putatively more highly coevolved host-pathogen combinations had a lower frequency of partial resistance types relative to allopatric combinations. Sympatric host-pathogen combinations also showed a lower diversity of resistance responses, but there was a trend toward a greater fraction of this variance being determined by pathogen-genotype × host-genotype interactions. In this system, there was no evidence that partial resistances slow host-pathogen coevolution. The analyses show that if variation is generated by among population host or pathogen dispersal, then coevolution occurs largely by pathogens overcoming the partial resistances that are generated.  相似文献

Illus. 1963.—A series of micrographs showing intercellular dikaryotic mycelium, haustorial mother cells, stages in haustorial formation, and haustoria within host cells are presented in the present report. Of special interest and potential significance in a study of obligate parasitism is an encapsulation ranging from 800 to 3400 A in thickness which surrounds the haustorium, but which is not present around the intercellular hyphae. The encapsulation completely encases the haustorium proper; it is bounded on the inside by the cell wall of the haustorium, and its thin membranous outer margin abuts directly on the protoplast of the host cell. The nature of the material composing the encapsulation is uncertain, but it appears to originate from the haustorial protoplast, and at least a portion of it may be fungal cytoplasm. This newly described structure represents the actual interface between the host and pathogen. Small vesicles which seem to originate from the outer margin of the encapsulation are sometimes found in the host cytoplasm surrounding apparently vigorous haustoria. The vesicles are bounded by a membrane and contain particulate material.  相似文献

The effect of the presence of Erysiphe graminis tritici on the reactions of each of five wheats, Malakoff, Democrat, Mediterranean, Hussar and Webster, to Puccinia triticina was studied. Under normal environmental conditions, mildew increased the susceptibility to rust of Malakoff and Democrat, though not, except in isolated instances, that of Mediterranean or Hussar. High light intensities enhanced the effect of mildew on the reaction of Democrat to P. triticina. The susceptibility of Webster to rust was unaffected or decreased by the presence of mildew.
In all varieties there was a decided tendency for rust and mildew to develop on different parts of the leaf, and when a leaf was heavily mildewed the development of P. triticina was sometimes almost entirely inhibited. The reason for this is obscure, since spores of P. triticina and E. g. tritici germinate well in each others presence, and cytological studies revealed several cases of the presence of both mildew and rust haustoria in a single living host cell. Anatomical investigations also showed that the effect of mildew on rust infection was confined to rust pustules within 1 mm. of mildew colonies.
The results obtained are in accordance with Gassner's theory that susceptibility to rust is due to the presence of certain specific proteins in the host leaf.  相似文献

Genetic variation in plant populations for resistance to pathogens and herbivores might be maintained by parasite-mediated negative frequency-dependent selection (FDS). But it is difficult to observe the time-lagged oscillations between host and parasite genotypes that should result from FDS. To evaluate the potential for FDS, we tested for local adaptation of parasites to common clones, the role of host genetic diversity in resistance to parasites, and genetic correlations among fitness, parasitism, and the frequency of host clones. We studied three populations of Arabis holboellii, a short-lived apomictic (asexual by seed) plant attacked by rust fungi and insect herbivores. To estimate clone frequency, we used polymorphic allozyme markers on 200 individuals in each population in 1990 and in 2000. We also recorded levels of parasitism and host fitness (fruit production). Only the rust fungi showed evidence for local host adaptation; they usually increased in incidence as a function of clone frequency, and they tracked temporal change in clone frequency. In further support of FDS, parasitism was lower in populations with higher genetic diversity. However, total parasitism (herbivory and disease combined) decreased as host clone frequency and fitness increased. Thus, although the highly virulent rust pathogen showed potential for driving the cycles that result from FDS, this apparently does not occur in the populations studied because the host clones were also attacked by herbivores.Co-ordinating editor: J.F. 南方锈病是玉米生产上的重要病害。2013-2015年在广西南宁和北京昌平对903份玉米种质资源进行了抗南方锈病的初步鉴定与评价,并利用SSR标记对筛选出的部分抗性材料进行了遗传多样性分析。结果表明,在903份种质中,8份自交系在广西南宁和北京昌平均对南方锈病表现高抗(HR),占总鉴定种质的0.9%;29份材料表现为抗病(R),占比3.2%,包括27份自交系和2份农家种;中抗种质(MR)100份,占比11.1%;感病(S)和高感(HS)种质分别为181和585份,占鉴定材料的20.0%和64.8%。由此可见,玉米资源中高抗南方锈病的种质较为匮乏,在不同地点均表现高抗的材料是难得的抗源。不同地理来源的玉米种质对南方锈病的抗性水平存在较大差异,其中抗性资源较为丰富的是源自内蒙古和山西的种质。42对多态性SSR引物在50份抗锈病材料中,共扩增出141个条带,多态性条带139个,多态位点百分率(PPB)为98.58%。平均等位基因数(Na)1.98,平均有效等位基因数(Ne)1.59,平均Nei's基因多样性(H)0.34,平均多态性信息含量(PIC)0.78,平均Shannon's信息指数(I)0.51;通过UPGMA聚类分析,50份抗病材料被划分为2个类群,其中,第Ⅰ类群又可划分为5个亚类,表现出较高的遗传多样性,为抗病育种中抗源的选择和利用提供参考信息。  相似文献

The resistance structure of a Turkish population of the clonal, apomictic composite Chondrilla juncea and the pathotypic structure of a co-occurring population of its obligate rust pathogen, Puccinia chondrillina, was determined by sequential inoculation of 19 host lines with 15 pathogen isolates each derived from single pustules collected from separate plants among the host population. The resultant matrix of resistant and susceptible reactions provides strong circumstantial evidence for a gene-for-gene interaction. Seven distinct pathotypes were detected in the pathogen population. One of these comprised 53% of the population, a second comprised 13%, while the remaining five pathotypes were each detected only once. The host population was similarly diverse, being composed of eight resistance phenotypes, only two of which were represented by more than one host line. Although C. juncea is apomictic, there was only 58% congruence between host resistance and multi-locus isozyme phenotype categories within this population. Pathotypic phenotypes of 13 other isolates of P. chondrillina collected from ten other Turkish and three more distant populations of C. juncea were markedly different from those found in the population studied in detail. There was no obvious relationship between the degree of geographic separation of pathotypes and their ability to attack particular C. juncea lines in this or three other populations represented by single host lines. Rust fungi, obligate biotrophs that cause disease and yield losses in crops such as cereals and soybean (Glycine max), obtain nutrients from the host through haustoria, which are specialized structures that develop within host cells. Resistance of flax (Linum usitatissimum) to flax rust (Melampsora lini) involves the induction of a hypersensitive cell death response at haustoria formation sites, governed by gene-for-gene recognition between host resistance and pathogen avirulence genes. We identified genes encoding haustorially expressed secreted proteins (HESPs) by screening a flax rust haustorium-specific cDNA library. Among 429 unigenes, 21 HESPs were identified, one corresponding to the AvrL567 gene. Three other HESPs cosegregated with the independent AvrM, AvrP4, and AvrP123 loci. Expression of these genes in flax induced resistance gene-mediated cell death with the appropriate specificity, confirming their avirulence activity. AvrP4 and AvrP123 are Cys-rich proteins, and AvrP123 contains a Kazal Ser protease inhibitor signature, whereas AvrM contains no Cys residues. AvrP4 and AvrM induce cell death when expressed intracellularly, suggesting their translocation into plant cells during infection. However, secreted AvrM and AvrP4 also induce necrotic responses, with secreted AvrP4 more active than intracellular AvrP4, possibly as a result of enhanced formation of endoplasmic reticulum-dependent disulfide bonds. Addition of an endoplasmic reticulum retention signal inhibited AvrM-induced necrosis, suggesting that both AvrM and AvrP4 can reenter the plant cell after secretion in the absence of the pathogen.  相似文献

1. This review is concerned with the obligate parasitism of the downy mildew, powdery mildew and rust fungi. 2. Mechanisms which determine resistance and susceptibility are incompletely understood. The three possibilities so far advanced, namely, hypersensitivity, phyto-alexins and nutrition are all inadequate to explain specificity. It is postulated that compounds carrying specificity are released by the invading pathogens and the interaction of the compounds with specific sites in the host cell determines whether a successful relationship is established. This hypothesis is based on the observation that host and pathogen protoplasts do not come in direct contact with each other and cells removed from the invading pathogen are affected. It should be possible to investigate this question now that some rust fungi have been cultured axenically. 3. Invasion of host tissue by these pathogens leads to alteration in host metabolism. Respiration rates and the activity of the pentose phosphate pathway are enhanced. Possible mechanisms for the reoxidation of NADPH by molecular oxygen are discussed. In the later stages of infection, photosynthetic activity is decreased and this decrease is coupled to the enhanced activity of the pentose phosphate pathway through NADPH availability. It is still not clear whether photosynthetic activity is increased or decreased during the early stages of infection. Nutrients from other parts of the plant accumulate at sites of infection, resulting in retarded growth of the host. Possible roles of hormones in nutrient mobilization and ‘green island’ formation are discussed. Changes in intermediary metabolism can be correlated with changes in amounts of nucleic acids in infected host cells. Chloroplast polysomes and ribosomes decrease during the early stages of infection but cytoplasmic ribosomes increase during patho-genesis. This increase more than compensates for the decrease in chloroplast ribosomes resulting in an overall increase in cellular RNA. 4. Some members of the Pucciniaceae and one member of the family Melamp-soraceae have now been cultured axenically. Nutritional studies indicate that these fungi are relatively non-specific with respect to their carbohydrate and amino-acid nitrogen sources but do have a specific requirement for either cysteine, methionine or homocysteine. Essential metabolites such as cysteine and cysteine-containing peptides move from hyphae into the medium and these may be important in amino-acid transport. Two stages in the development of axenic rust cultures are still imperfectly understood - the transition from germ tube to saprophytic mycelium and the fate of the colonies once formed. Further knowledge of the factors which control these stages may well suggest approaches to the axenic culture of other groups of obligately parasitic fungi.  相似文献

A trade‐off between a pathogen's ability to infect many hosts and its reproductive capacity on each host genotype is predicted to limit the evolution of an expanded host range, yet few empirical results provide evidence for the magnitude of such trade‐offs. Here, we test the hypothesis for a trade‐off between the number of host genotypes that a fungal pathogen can infect (host genotype range) and its reproductive capacity on susceptible plant hosts. We used strains of the oat crown rust fungus that carried widely varying numbers of virulence (avr) alleles known to determine host genotype range. We quantified total spore production and the expression of four pathogen life‐history stages: infection efficiency, time until reproduction, pustule size, and spore production per pustule. In support of the trade‐off hypothesis, we found that virulence level, the number of avr alleles per pathogen strain, was correlated with significant delays in the onset of reproduction and with smaller pustule sizes. Modeling from our results, we conclude that trade‐offs have the capacity to constrain the evolution of host genotype range in local populations. In contrast, long‐term trends in virulence level suggest that the continued deployment of resistant host lines over wide regions of the United States has generated selection for increased host genotype range.  相似文献

条锈菌与慢锈小麦品种互作的超微结构研究表明,随着慢锈性的表达,条锈菌的胞间菌丝发育受抑,细胞器泡囊化解体。吸器母细胞和吸器发育受阻,最终解体、坏死。吸器的受抑、坏死主要表现在吸器体形成后期。在锈菌与寄主互作的交界面,吸器外质膜皱褶、电子致密度增高,吸器外间质加宽,并有大量电子致密度加深的物质沉积。与抗病品种相比, 慢锈品种中病菌受抑、坏死程度轻,寄主细胞的过敏性坏死机率也较低。  相似文献

Phakopsora pachyrhizi is an obligate pathogen that causes Asian soybean rust. Asian soybean rust has an unusually broad host range and infects by direct penetration through the leaf cuticle. In order to understand the early events in the infection process, it is important to identify and characterize proteins in P. pachyrhizi. Germination of the urediniospore is the first stage in the infection process and represents a critical life stage applicable to studies with this obligate pathogen. We have applied a 2‐DE and MS approach to identify 117 proteins from the National Center of Biotechnology Information nonredundant protein database and a custom database of Basidiomycota EST sequences. Proteins with roles in primary metabolism, energy transduction, stress, cellular regulation and signaling were identified in this study. 活性氧的释放在动植物-病原菌互作过程中有着非常重要的作用.一般认为互作中的活性氧来源于动植物细胞生物膜中的氧化还原体系.但近年来随着互作研究的深入,发现动植物病原菌自身也有活性氧的释放以及复杂的调控系统,它们的活性氧释放能力很有可能与其致病性有一定的联系,并可能参与了互作,这些发现对深入了解动植物-病原菌的互作机制具有重要意义.概述了在细菌、真菌等多种动物病原菌中存在的活性氧释放现象,这些微生物活性氧产生的位点、相关功能分子以及调控机制,介绍了目前研究仍然较少但其潜在意义重大的植物病原菌中的活性氧释放现象、可能的调节机制和病理学意义.  相似文献

Computer models of disease take a systems biology approach toward understanding host-pathogen interactions. In particular, data driven computer model calibration is the basis for inference of immunological and pathogen parameters, assessment of model validity, and comparison between alternative models of immune or pathogen behavior. In this paper we describe the calibration and analysis of an agent-based model of Leishmania major infection. A model of macrophage loss following uptake of necrotic tissue is proposed to explain macrophage depletion following peak infection. Using Gaussian processes to approximate the computer code, we perform a sensitivity analysis to identify important parameters and to characterize their influence on the simulated infection. The analysis indicates that increasing growth rate can favor or suppress pathogen loads, depending on the infection stage and the pathogen's ability to avoid detection. Subsequent calibration of the model against previously published biological observations suggests that L. major has a relatively slow growth rate and can replicate for an extended period of time before damaging the host cell.  相似文献

Severity of peanut rust caused by Puccinia arachidis was reduced by 15 edible oils tested. Flaxseed oil was the best suppressing the disease completely. Peanut oil, wheat germ oil, brown rice oil, aloe oil, olive oil and corn germ oil also caused more than 75% reduction in disease incidence. Flaxseed oil reduced the rust to a negligible level in the greenhouse and was nearly as effective as the fungicide chlorothalonil in peanut field trials. The control of peanut rust by flaxseed oil did not result from activation of the host defence mechanisms. Flaxseed oil did not affect urediniospore germination, but reduced the germ tube length and completely suppressed appressorium formation which is essential for the pathogen to form an infection peg to pass through the stomatal aperture and infect the host tissue. Although the pathogen had penetrated, flaxseed oil still exerted some inhibitory effect against the growth of the pathogen. The advantages of using flaxseed oil to control peanut rust are that it is relatively inexpensive, easy to prepare, and friendly to the environment and human health.  相似文献

A genetic analysis of parasitic ability in the soybean cyst nematode Heterodera glycines was performed. To identify and characterize genes involved in parasitism, we developed three highly inbred H. glycines lines, OP20, OP25 and OP50, for use as parents for controlled crosses. Through these crosses, we have identified genes in the inbred parents that control reproduction of the nematode on hosts that carry resistance genes. These genes, designated as ror-* for reproduction on a resistant host, segregate in a normal Mendelian fashion as independent loci. Host range tests of F(1) generation progeny indicated that at least one parasitism gene in both the OP20 and OP50 lines for host PI 88788 was dominant. Parasitism genes in OP50 for hosts ``Peking' and PI 90763 are recessive. Two types of single female descent populations, a single backcrossed BC(1)F(2)-derived and a double backcrossed BC(2)F(1)-derived, were established on the susceptible soybean cultivar ``Lee 68.' Host range tests for parasitism in these lines demonstrated the presence of two independent genes in OP50, one for host PI 88788 designated ror-1 and one for host PI 90763 designated ror-2. OP20 carries two independent genes for parasitism on PI 88788, designated as alleles kr3 and kr4.  相似文献

Parasitism genes from phytoparasitic nematodes are thought to be essential for nematode invasion of the host plant, to help the nematode establish feeding sites, and to aid nematodes in the suppression of host plant defenses. One gene that may play several roles in nematode parasitism is chorismate mutase (CM). This secreted enzyme is produced in the nematode's esophageal glands and appears to function within the plant cell to manipulate the plant's shikimate pathway, which controls plant cell growth, development, structure, and pathogen defense. Using degenerate polymerase chain reaction primers, we amplified and cloned a chorismate mutase (Hg-cm-1) from Heterodera glycines, the soybean cyst nematode (SCN), and showed it had CM activity. RNA in situ hybridization of Hg-cm-1 cDNA to SCN sections confirms that it is specifically expressed in the nematodes' esophageal glands. DNA gel blots of genomic DNA isolated from SCN inbred lines that have differing virulence on SCN resistant soybean show Hg-cm-1 is a member of a polymorphic gene family. Some Hg-cm family members predominate in SCN inbred lines that are virulent on certain SCN resistant soybean cultivars. The same polymorphisms and correlation with virulence are seen in the Hg-cm-1 expressed in the SCN second-stage juveniles. Based on the enzymatic activity of Hg-cm-1 and the observation that different forms of the mutase are expressed in virulent nematodes, we hypothesize that the Hg-cm-1 is a virulence gene, some forms of which allow SCN to parasitize certain resistant soybean plants.  相似文献
