A periodic acid-Schiff's substance present in the micropylar end of the ovules of Paspalum orbiculare and P. longifolium was further studied by light and electron microscopy of glutaraldehyde-osmium-fixed and freeze-substituted, osmium-fixed tissues. The PAS substance is water soluble and is found in intercellular spaces between the nucellus and inner integument, the inner and outer integuments, the outer integument and ovary wall, and in the micropyle. Structurally the substance consists of fibrils embedded in a dense, amorphous matrix and may be associated with membranous structures in special layers between the plasmalemma and the cell wall in nucellar and integumentary cells. Part of the water soluble substance is believed to be secreted from the nucellar and integumentary cells. A large amount of this substance may be formed as a result of the dissolution of about one third of the distal micopylar portion of the outer integument prior to anthesis. Many of the electron-dense fibrils seem to be fibrillar intercellular substances and others appear to originate from the cell walls, including the cuticle. Both the matrix and the fibrils may be chemically heterogeneous and together form a mucilagenous substance which may facilitate the final growth of pollen tubes in these two species.  相似文献

The micropyle and the integuments of sugar beet (Beta vulgaris) ovules have been investigated by light and electron microscopy during differentiation and maturation of the ovule. The micropyle itself is formed by the inner integument which is surrounded by the outer integument at its base. The micropyle containts a fibrillar PAS+ substance and is often covered by a thin sheet or hymen. Both integuments are cuticle-covered thin sheets, each 2-few cell layers in thickness. In the outer integument an increase in starch accumulation occurs during ovule maturation and probably functions as nutrient storage for embryo development. The inner epidermis of the inner integument differentiates as the most conspicuous cell layer of the beet ovule. During growth and maturation of the ovule a system of small perinuclear vacuoles containing dense material increases steadily in these cells. At maturity this system fills up more than half of each cell and very dense material has accumulated in each vacuole. This vacuole content is highly refractive and contains tannins and/or polyphenols.  相似文献

Differentiation betweenPityrosporum ovale andPityrosporum orbiculare is mainly based upon the morphological features of cells and on physiological characters such as the ability ofP. ovale of utilizing oleic acid (Marples, 1965). Our studies in submerged culture, however, have not shown remarkable differences between the two species: growth curves, nutritional requirements, lipid utilization and optimal temperature are practically the same. In some strains of both species (1 : 4) we did observe a limited growth capacity, associated with a lower utilization rate of lipids. But for the notably different morphological features of cells,P. ovale andP. orbiculare could be ascribed, as regards their cultural characteristics, to the same species. The search forP. orbiculare was successful in all the cases of Pityriasis versicolor, while all our attempts to isolateP. ovale from its usual areas (scalp, upper back and chest) from both healty and seborrhoic subjets, gave no results.P. orbiculare was found in the same areas in the greatest part (85 %) of these subjects. In order to explain these results, one could assume thatP. orbiculare is the only species ofPityrosporum widespread in our region.These studies were supported by grant of the Consiglio Nazionale delle Ricerche of Italy under contract n. 70.01023/115.2798.  相似文献

Two homologues of INNER NO OUTER (INO) in Nymphaea alba and N. colorata (Nymphaeaceae) were isolated and the expression pattern of the N. alba INO homologue NaINO was examined by in situ hybridization. The INO homologues obtained have a portion similar to INO in the predicted amino acid sequences between the conserved zinc finger-like and YABBY domains. In an in situ hybridization analysis, NaINO is expressed in the outer epidermis of the outer integument, inner integument, and the tip of the nucellus. The pattern observed in the outer integument is very similar to that of Arabidopsis thaliana, while the expression in the inner integument and nucellus is not observed in A. thaliana.Edited by G. The penetration process and defence reactions (hypersensitive response, oxidative burst and cell wall fortification) of Colletotrichum orbiculare were studied histochemically on pepper cultivar ‘A11’ (non‐host) and susceptible cucumber cultivar ‘Changchun Thorn’ (host). The results indicate that C. orbiculare could hardly penetrate the non‐host pepper leaves. It was papillae rather than hypersensitive response and H2O2 that played an important role in resisting the colonization and development of C. orbiculare on the non‐host pepper. The depolymerization of the actin microfilament weakened the papilla deposition of pepper and allowed successful penetration of the non‐adapted C. orbiculare, suggesting that the actin cytoskeleton of pepper is significant in preventing the invasion of the non‐host pathogen C. orbiculare.  相似文献

Area of lesions produced by Colletotrichum orbiculare on watermelon (Citrullus lanatus) seedlings was not affected by amount of culture medium on which the fungus was grown, and decreased as length of time the fungus was in culture increased. Internal integrity of C. orbiculare spores appeared degraded, and this was coincident with decreases in lesion area.  相似文献

Cetacean integument serves many functional roles, including contribution to whole body buoyancy. The blubber of the integument of different cetacean species contains varying concentrations of triacylglycerols (TAG) and wax esters (WE); generally, these lipid classes have different densities. Integument can also experience a wide range of temperatures during a dive, so its density may change with depth. The goals of this study were to measure integument density and isolated blubber lipid density in three deep‐diving odontocete species (n = 3–4)—short‐finned pilot whales (Globicephala macrorhynchus), pygmy sperm whales (Kogia breviceps), and Gervais' beaked whales (Mesoplodon europeaus)—at different temperatures (6°C–35°C), and to relate these densities to lipid content and composition. Kogia and Mesoplodon integument and isolated lipids had high WE content (78.7–99.5 wt%) and were less dense (by 1.7%–9.3%) than those of Globicephala, which were composed predominately of TAG. Generally, densities increased as temperature decreased. Changes in integument densities mirrored those of isolated lipid densities, suggesting that blubber lipids are largely responsible for the buoyant properties of cetacean integument. These data demonstrate that the contribution of the integument to whole body density depends on lipid class and temperature, and therefore may provide useful, species‐specific correction factors for diving energetics models.  相似文献

The southern African crab Hymenosoma orbiculare was recently split into five distinct species, of which three are estuarine/coastal and have peripatric distributions that are linked to temperature-defined marine bioregions. This suggests that the species’ ranges may be limited by physiological adaptations to their thermal environment. We explored this hypothesis by rearing the larvae of the warm-temperate lineage of H. orbiculare and the warm-temperate/subtropical H. longicrure at a range of water temperatures, and found clear temperature-dependent differences in the duration of larval development. Our study contributes to the growing body of evidence that stresses the importance of adaptation to regional environmental conditions, rather than physical dispersal barriers on their own, in limiting the mixing of marine species between temperature-defined biogeographic regions.  相似文献

The integument and podia of the sea cucumber Thyone briareus were examined by bright field and electron microscopy. The epidermal surface was found to be covered by an acellular, PAS positive cuticle which appeared to be secreted by the underlying epidermal cells. Although the superficial portion of the cuticle contains numerous fine filaments, their ultrastructure bears no resemblance to collagen fibers. The epidermal cells are widely spaced and have long apical processes that extend along the under surface of the cuticle forming a contiguous epithelium. The apical expansions of the epidermal cells are attached to one another by means of septate desmosomes which may run continuously around all epidermal cells. Special attachment structures within these apical expansions appear to bind the cuticle to the dermis. The epidermal cells and their apical expansions are separated from the dermis by an 800 Å thick basement membrane. Granule containing cells in the upper dermis send processes up to the cuticle where they are bound to the typical epidermal cells by septate desmosomes. The abundant membrane bound granules of the cells enter villous-like processes which pass through the cuticle. The function of these cells may be to produce an adhesive material on the podia or they may be pigment cells. The thick dermis consists of a superficial zone, containing largely ground substance; a middle or laminated zone containing laminae of collagen fibers arranged in an orthogonal fashion; and a hypodermis consisting largely of ground substance and reticular fibers. Fibroblasts are abundant in the superficial dermis and between the collagen laminae. Wandering coelomocytes, or morula cells, accumulate between the collagen laminae and in the hypodermis. They may also become an integral part of the epidermis by forming septate desmosomes with epidermal cells. Morula cells contain highly specialized spherules whose tinctorial properties and electron microscopic appearance suggest that they contain protein and mucopolysaccharide.  相似文献

Genome sequencing of pathogenic fungi has revealed the presence of various effectors that aid pathogen invasion by the manipulation of plant immunity. Effectors are often individually dispensable because of duplication and functional redundancy as a result of the arms race between host plants and pathogens. To study effectors that have functional redundancy, multiple gene disruption is often required. However, the number of selection markers that can be used for gene targeting is limited. Here, we established a marker recycling system that allows the use of the same selection marker in successive transformations in the model fungal pathogen Colletotrichum orbiculare, a causal agent of anthracnose disease in plants belonging to the Cucurbitaceae. We identified two C. orbiculare homologues of yeast URA3/pyrG, designated as URA3A and URA3B, which can be used as selection markers on medium with no uridine. The gene can then be removed from the genome via homologous recombination when the fungus is grown in the presence of 5-fluoroorotic acid (5-FOA), a chemical that is converted into a toxin by URA3 activity. The ura3a/b double mutants showed auxotrophy for uridine and insensitivity to 5-FOA. Using the ura3a/b mutants, transformation with the URA3B marker and its removal were successfully applied to disrupt the virulence-related gene, PKS1. The pks1 mutants showed a reduction in virulence, demonstrating that the method can be used to study virulence-related genes in C. orbiculare. The establishment of a URA3-based marker recycling system in plant-pathogenic fungi enables the genetic analysis of multiple genes that have redundant functions, including effector genes.  相似文献

Colletotrichum orbiculare infects cucurbits, such as cucumber and melon (Cucumis melo), as well as the model Solanaceae plant Nicotiana benthamiana, by secreting an arsenal of effectors that suppress the immunity of these distinct plants. Two conserved effectors of C. orbiculare, called NLP1 and NIS1, induce cell death responses in N. benthamiana, but it is unclear whether they exhibit the same activity in Cucurbitaceae plants. In this study, we established a new Agrobacterium-mediated transient expression system to investigate the cell death-inducing activity of NLP1 and NIS1 in melon. NLP1 strongly induced cell death in melon but, in contrast to the effects seen in N. benthamiana, mutations either in the heptapeptide motif or in the putative glycosylinositol phosphorylceramide-binding site did not cancel its cell death-inducing activity in melon. Furthermore, NLP1 lacking the signal peptide caused cell death in melon but not in N. benthamiana. Study of the transient expression of NIS1 also revealed that, unlike in N. benthamiana, NIS1 did not induce cell death in melon. In contrast, NIS1 suppressed flg22-induced reactive oxygen species generation in melon, as seen in N. benthamiana. These findings indicate distinct cell death-inducing activities of NLP1 and NIS1 in these two plant species that C. orbiculare infects.  相似文献

A technique for simultaneous inoculation of cucumber cotyledons with Colletotrichum orbiculare race 1 and Cladosporium cucumerinum has been developed. The procedure permitted both resistant and susceptible plants to be recovered. Seedlings were grown at 20°C and inoculated 24 h after emergence with Colletotrichum orbiculare (200 spores in 2 μ1 of water) and Cladosporium cucumerinum (1000 spores in 5 μ1 of water) followed by 48 h of incubation in the dark at 20°C and 100% r.h., and 48 h in a 20°C lighted growth chamber. Seedlings were then moved to a growth chamber at 21°C at night and at 26°C during the day for 4 days and plants were rated as resistant or susceptible 8 days after inoculation. No interference in the expression of resistance or susceptibility of cultivars to either pathogen was detected in simultaneous inoculations.  相似文献

采用半薄切片技术和组织化学染色法对宁夏枸杞大孢子发生和雌配子体发育过程中的细胞结构变化及营养物质积累特征进行了观察。结果表明,(1)宁夏枸杞为中轴胎座,多室子房,倒生胚珠,单珠被,薄珠心类型。(2)位于珠心表皮下的孢原细胞可直接发育为大孢子母细胞,减数分裂后形成直线型大孢子四分体,合点端第一个大孢子发育为功能大孢子,胚囊发育类型为蓼型,具有珠被绒毡层。(3)初形成的胚囊外周组织中没有营养物质积累,成熟胚囊时期出现了大量的淀粉粒且呈珠孔端明显多于合点端的极性分布特征。(4)助细胞的珠孔端具有明显的丝状器结构,呈PAS正反应表现出多糖性质,成熟胚囊具有承珠盘结构。  相似文献

SYNOPSIS. Plistophora cargoi n. sp., found in the muscles of Callinectes sapidus Rathbun, is described. Spores are ellipsoidal and about 3.3 by 5.1 μ when alive. The coiled polar filament is PAS positive. When extruded the filament appears thick throughout most of its length but has a short and fine terminal portion. The sporont gives rise to many (32 to more than 100) spores. P. sp. Summary During the course of a fluorescence microscopic investigation on the extra-ovular micropylar portion of the embryo sacs ofTorenia fournieri Lind. (Scrophulariaceae) a callosic wall was found which surrounded it almost completely until the time of anthesis. In addition, the walls of young synergids and the filiform apparatus also showed callosic fluorescence. Treatments with PAS reaction revealed a PAS-positive substance filling up the locular cavity. Our attempts to induce fluorochromasia by employing fluorescein diacetate failed, indicating the low permeability of the callosic wall around the embryo sac. It is assumed that the callose wall around the embryo sac isolates the latter from the contents of the locular cavity whereas the callose in the synergid walls may represent an intermediate stage in the maturation of these walls; the filiform apparatus is mainly composed of callose.  相似文献

An a-mating-type-specific substance responsible for sexual agglutination was purified to 397-times in specific activity (units/mg protein) from the cytoplasm of a-mating type cells. The purified substance gave a single band stained with PAS reagent but not with both Coomassie brilliant blue and silver staining reagent by polyacrylamide gel electrophoresis in the presence of 8 M urea. However, incorporation of [35S]methionine and Lowry reaction clearly indicate that the substance is a glycoprotein. The substance specifically masked sexual agglutinability of cells of the opposite mating type , indicating univalent action. The substance is a glycoprotein with a carbohydrate content of 90%, a pI of 4.5, and a molecular weight of 130,000. The substance was inactivated by 2-mercaptoethanol and proteolytic enzymes but not by glycolytic enzymes. Petrified seeds of Upper Mississippian age (Chester Series) are described from sediments from northwestern Arkansas. Specimens of Rhynchosperma quinnii gen. et sp. n. are radially symmetrical and range in length from 1.2 to 2.2 cm and 1.0 to 1.3 cm at the greatest diameter. The seeds are ovate with a tapered apex having 8–10 integumentary ribs and a broadly rounded chalaza. The integument is two-parted, consisting of an outer presumably soft tissue with secretory canals and an inner layer constructed of longitudinally oriented thick-walled cells. Nucellus and integument are confluent, but they diverge near the seed midlevel with the free portion of the nucellus forming a dome-shaped pollen-receiving structure. Additional features such as vasculature and the structure of well-preserved megagametophytes are described. The petrified specimens are compared with taxa instituted for casts and impressions which include Rhynchogonium and Boroviczia. The ovulate versus cupulate nature of these genera which has remained uncertain until now appears to be settled in favor of an ovular identity.  相似文献

Summary Localization of polysaccharides in the freeze-substituted, Eponembedded ovaries of Paspalum longifolium prior to pollination was carried out by periodic acid-Schiff's (PAS), periodic acid-thiosemicarbazide-silver proteinate (PA-TSC-AgPr) and periodic acid-thiosemicarbazide-osmium (PA-TSC-OsO4) reactions. The specificities of these three reactions were also studied. These three reactions are all effective for light microscopic demonstration of polysaccharides in the filiform apparatus, starch grains in the cells and PAS substance in the micropylar region. Nonspecific staining of the nucleoli of the egg and polar nuclei was observed in the PAS reaction. The PA-TSC-AgPr reaction is very specific for polysaccharides but its overall reaction takes a much longer period of time than the PAS reaction. The PA-TSC-OsO4 reaction colors the cytoplasm and nuclei of most cells and therefore stains of the cell walls, especially those of the egg cell and synergids, do not stand out clearly. The synergid cytoplasm contains some amorphous polysaccharides and thus it colors even in PAS and PA-TSC-AgPr preparations. In the mature embryo sac, the egg and central cell as well as antipodals are vacuolated but the two synergids have no visible vacuoles under light microscope. Each synergid has a prominent filiform apparatus at the micropylar end, which stains intensely in all three preparations. The walls of the central cell and antipodals adjacent to the nucellar cells have many inward papillae which are also intensely stained in all three preparations. Starch grains are abundant in the ovary wall and usually absent in the nucellus and integuments. They are present in the egg, central cell and antipodals, but not in the two synergids.  相似文献

The embryo position in a seed is stable in most plant species, indicating the existence of a strict regulatory mechanism that specifies the embryo position in the seed. To elucidate this mechanism, we analyzed the gypsy embryo (gym) mutant of rice, in which the position of the mature embryo in the seed is altered at a low frequency. Analyses of early embryogenesis and ovule development showed that the ectopic embryo was derived from an ill-positioned egg cell, which resulted from the incomplete curvature of the ovule. Although the development of both the inner and outer integuments was impaired, the ovule curvature was associated closely with the extent of inner integument growth. Therefore, inner integument development controls ovule curvature in rice. The expression patterns of OSH1 and OsMADS13 indicated that, in gym, a small number of indeterminate cells are maintained on the style side of the ovule and then in the integument primordium at a low frequency. The prolonged survival of these indeterminate cells disturbs normal integument development. The gym fon2 double mutant suggests that GYM and FON2 are involved redundantly in floral meristem determinacy. Possible functions of the GYM gene and the ovule developmental mechanism are discussed.  相似文献

The embryology ofStegnosperma halimifolium andS. watsonii has been studied in detail. The tapetum is of the secretory type and its cells become multinucleate. Simultaneous cytokinesis in the pollen mother cells follows meiosis. The ripe pollen grains are 3-celled. The ovule is crassinucellate, bitegmic and amphitropous, with the micropyle formed by the inner integument alone. The female archesporium is one celled, and the parietal tissue 3–5 layered. The embryo sac development conforms to thePolygonum type. A central strand, 6 or 7 cells thick, differentiates inside the nucellus and extends from the base of the embryo sac to the chalazal region. The endosperm is nuclear. The embryogeny conforms to the Caryophyllad type. The seed coat is formed by the outer epidermis of the outer integument and the inner epidermis of the inner integument. Based on this evidence and other data, the status of the genus as an independent family,Stegnospermataceae (Stegnospermaceae) is confirmed. Apparently, it forms a connecting link betweenPhytolaccaceae andCaryophyllaceae.  相似文献
