The ultrastructure of the oval, fusiform and triradiate morphotypes of Phaeodactylum tricornutum Bohlin is described. The organization and structure of the cytoplasmic organelles is similar in all three morphotypes, except that the vacuoles occupy the extra volume created by the arms of the fusiform and triradiate cells. The frustule in fusiform and triradiate cells is organic; in the oval type it may be organic or one of the valves may have a silica frustule surrounded by an organic wall. In all cells, the organic cell wall has up to 10 silica bands (13 nm wide) embedded in its surface in the girdle region, lacks girdle bands, and has an outer corrugated cell wall layer, except in the girdle region. Cell division, organic wall formation and silica deposition are described in detail. Four types of oval cells are also described. The relation to other diatoms is discussed.  相似文献

A rare Phaeodactylum tricornutum cruciform morphotype was obtained and stabilized with a proportion of more than 31.3% in L1 medium and is reported for the first time. Long-term culture and observation showed that the cruciform morphotype was capable of transforming to the oval form following the degeneration of arms by two processes. After three months of culture, four morphotypes existed in a relatively stable proportion in culture for six months (10.5% for oval, 11.3% for fusiform, 37.2% for triradiate and 41.0% for cruciform). Low temperature was particularly beneficial for cruciform cell formation. As the culture temperature decreased from 25°C to 10°C, the percentage of the cruciform morphotype increased from 39.1% to 55.3% approximately. The abundant cruciform cells endowed this strain with unique fatty acid characteristics. The strain cultured at 15°C showed both maximum content of neutral lipid in a single cell and total yield. The maximum content of fatty acid methyl esters was C16:1 for Phaeodactylum tricornutum cultured at four temperatures (43.82% to 50.82%), followed by C16:0 (20.47% to 22.65%). Unique fatty acid composition endowed this strain with excellent quality for biodiesel production.  相似文献

In the last few years, genome‐based studies in diatoms have received a major boost following the genome sequencing of the centric species Thalassiosira pseudonana Hasle et Heimdal and the pleiomorphic raphid pennate diatom Phaeodactylum tricornutum Bohlin. In addition, molecular tools, such as genetic transformation, have been developed for both species. Despite these molecular advances, relatively little is known regarding the genetic diversity of the available strains of these diatoms. In this study, we have compiled a historical summary of the known P. tricornutum species resources and have provided a genetic and phenotypic overview of 10 different axenic strains. Examination of intraspecies genetic diversity based on internal transcribed spacer 2 (ITS2) sequence and amplified fragment length polymorphism (AFLP) analyses indicate four different genotypes. Seven strains are predominantly fusiform, whereas one strain is predominantly oval, and another is predominantly triradiate. Another is defined as a tropical strain because it appears better acclimated to growth at higher temperatures. Observations in the natural environment indicate that P. tricornutum is a coastal marine diatom that is able to adapt to unstable environments, such as estuaries and rock pools. Because it has rarely been noted in nature, we have developed specific primers to amplify ITS2 sequences and have successfully identified it in environmental samples. These resources should become useful tools for the diatom community when combined with the whole genome sequence and will open up a range of new possibilities for experimental investigations that can exploit the genotypic and phenotypic characteristics described.  相似文献

 The ultrastructure of the vascular cambium of Robinia pseudoacacia L. was examined in trunk tissues collected over a 2 1/2 year period. During dormancy, fusiform cells are densely cytoplasmic with many small vacuoles and centrally located nuclei. Mitochondria are round to oval in sectional view. The plastids are variable in shape, have few internal membranes, and generally lack starch grains. The plasmalemma is smooth in outline. Proteinaceous material occurs in the vacuoles and many lipid droplets are scattered throughout the ground substance. Smooth tubular ER, often highly dilated, predominates, but short segments of rough ER are also present. Abundant free ribosomes are evenly distributed throughout the ground substance and the dictyosomes are inactive. Microtubules are parietal and have various orientations. During reactivation, the plasmalemma becomes irregular in outline and begins to form invaginations. Concurrently, the proteinaceous material disappears, the vacuoles begin to fuse, polysomes appear, and the dictyosomes begin to produce vesicles. During the period of cambial activity, fusiform cells are highly vacuolate, and the nuclei are centrally located. The mitochondria are round, oval, or elongate. Now the plastids contain phytoferritin, starch grains, or both. Many large invaginations of the plasmalemma intrude into the vacuole, pushing the tonoplast inward and pinching off into the vacuole, which lacks proteinaceous material. Lipid droplets are scarce. Most ER is rough, and ribosomes are generally aggregated as polysomes. Dictyosomes are actively producing vesicles. During the transition to dormancy, the fusiform cells gradually assume the appearance typical of the dormant cambium.  相似文献

The diatom Phaeodactylum tricornutum Bohlin is known to be pleomorphic. Previously it was considered that growth on medium solidified with agar produced ovate cells, whereas growth in liquid medium gave rise to fusiform cells. We have shown that ovate cells can be propagated in liquid medium if the calcium content of the medium is below 15 mg/liter. If the concentration of calcium is raised, fusiform cells develop.  相似文献

Torrubeilla pruinosa, a teliomorph of an anamorphic entomopathogenic fungus Hirsutella versicolor on mango hopper (Idioscopus clypealis) was observed. On the infected mango hopper, tiny pin head signs of ascomata were observed. Ascomata of T. pruinosa is pale brown or tawny brown, crowded, immersed in stroma, wall dark golden brown, 260–320?×?230–260?μm, asci clavate, hyaline and thin walled. The ascospores of T. pruinosa are fusiform, distoseptate with a faint tint of pigmentation, 17.5–25.0?×?5.0–7.5?μm. The anamorph stage of H. versicolor fungal hyphae is hyaline, septate and profusely branched, and conidiogenous (phialides) cells are hyaline, pear-shaped and smooth-walled with single or double sterigmata and rarely, multiple sterigmata. Each sterigmata bears single conidia which are hyaline, oval to pear-shaped.  相似文献

Summary The midgut of Blaberus craniifer is principally made up of columnar epithelial cells which are derived from small regenerative cells found grouped in nidi. Between them, small sparsely granulated cells with clear cytoplasm can be observed lying on the basal lamina. Mainly based on the size, shape and texture of their secretory granules, at least ten types of such endocrine cells have been identified. Five cell types contain a uniform population of dense granules: (1) medium-sized, round to oval granules; (2) small elongated granules; (3) large irregular granules; (4) oval granules with a highly osmiophilic core; (5) oval, haloed granules. Five others are characterized by a heterogeneous population of granules: (6) small, round to oval, variably electron-dense granules; (7) oval medium-sized granules of variable electron density; (8) large irregular granules of variable electron density; (9) small dense granules and large vesicles with filamentous material; (10) small dense granules and very large pale vesicles.In addition, near the regenerative cells, large cells characterized by very large, irregular, dense granules (up to 4 m), lack contact with the lumen, and reach the basal lamina only by slender cytoplasmic processes.Several antisera raised against mammalian peptides and amine were used to reveal axonal fibers and endocrine cells. Serotonin-like immunoreactivity is localized in a profuse innervation of the muscle layers that surround the epithelium, whereas cholecystokinin and methionine-enkephalin antisera stain a more moderate number of axonal fibers. Cholecystokinin-, methionine-enkephalin-, substance P-, vasoactive intestinal peptide-, somatoliberin-, and gonadoliberin-like immunoreactivities were detected in endocrine cells of the epithelium. While most of the cells appear pyramidal, oval, fusiform or bowl-shaped, and seem to lack contact with the lumen, cells reaching it have been detected reacting with antisera to cholecystokinin, substance P, vasoactive intestinal peptide, somatoliberin and gonadoliberin.  相似文献

Benthic diatoms live in photoautotrophic/heterotrophic biofilm communities embedded in a matrix of secreted extracellular polymeric substances. Closely associated bacteria influence their growth, aggregation, and secretion of exopolymers. We have studied a diatom/bacteria model community, in which a marine Roseobacter strain is able to grow with secreted diatom exopolymers as a sole source of carbon. The strain influences the aggregation of Phaeodactylum tricornutum by inducing a morphotypic transition from planktonic, fusiform cells to benthic, oval cells. Analysis of the extracellular soluble proteome of P. tricornutum in the presence and absence of bacteria revealed constitutively expressed newly identified proteins with mucin‐like domains that appear to be typical for extracellular diatom proteins. In contrast to mucins, the proline‐, serine‐, threonine‐rich (PST) domains in these proteins were also found in combination with protease‐, glucosidase‐ and leucine‐rich repeat‐domains. Bioinformatic functional predictions indicate that several of these newly identified diatom‐specific proteins may be involved in algal defense, intercellular signaling, and aggregation.  相似文献

Cell development and starch granule formation in seeds of three pea (Pisum sativum L.) genotypes, R/R Rb/Rb, r/r Rb/Rb, and R/R rb/rb, affecting cotyledon starch were compared. Cotyledon cells at 10 days after flowering were highly vacuolated and contained small protein bodies in the vacuoles and small oval starch granules in the cytoplasm in all three genotypes. Gradients of cell development from the center to the periphery of the cotyledon and toward the cotyledon-hypocotyl axis persisted through the cell enlargement, reserve synthesis, and into the maturation stages of cotyledon development. By 14 days after flowering, many small vacuoles lined with protein deposits had been formed. Vacuoles were only observed in peripheral and basal cells by 18 days after flowering. Starch granules were oval and birefringent in all three genotypes at 10 days. Starch granules in R/R Rb/Rb and R/R rb/rb cotyledons expanded regularly remaining nearly oval and birefringent throughout development. In contrast, starch granules from r/r Rb/Rb cotyledons began to fragment by 14 days after flowering. This process began as a single fissure, followed by a second fissure usually at or near right angles. Finally, because of the fragmentation, the granules appeared compound, and only a portion of the granule was birefringent. All genotypes contained nearly equal volumes of liquid endosperm and embryo at 10 days after flowering. In addition, a layer of parenchyma tissue (ovular and/or endospermic) inside the seed coat was observed. Although, thin walled and poorly defined cytologically, the parenchyma cells contained large numbers of starch granules. These granules were a mixture of simple and compound types in all genotypes. By 18 days after flowering, the parenchyma tissue was reduced to a small layer of cell walls and all starch granules had been mobilized.  相似文献

On the basis of LM, we isolated strains of two species of fusiform green flagellates that could be assigned to former Chlorogonium (Cg.) Ehrenb. One species, “Cg.”heimii Bourr., lacked a pyrenoid in its vegetative cells and required organic compounds for growth. The other was similar to Cg. elongatum (P. A. Dang.) Francé and “Cg.”acus Nayal, but with slightly smaller vegetative cells. Their molecular phylogeny was also studied based on combined 18S rRNA, RUBISCO LSU (rbcL), and P700 chl a‐apoprotein A2 (psaB) gene sequences. Both species were separated from Chlorogonium emend., Gungnir Nakada and Rusalka Nakada, which were formerly assigned to Chlorogonium. They were accordingly assigned to new genera, Tabris Nakada gen. nov. and Hamakko (Hk.) Nakada gen. nov. as T. heimii (Bourr.) Nakada comb. nov. and Hk. caudatus Nakada sp. nov., respectively. Tabris is differentiated from other genera of fusiform green flagellates by its vegetative cells, which only have two apical contractile vacuoles and lack a pyrenoid in the chloroplast. Hamakko, on the other hand, is distinguishable by the fact that its pyrenoids in vegetative cells are penetrated by flattened thylakoid lamellae.  相似文献

One of the most common marine dinophytes is a species known as Heterocapsa triquetra. When Stein introduced the taxon Heterocapsa, he formally based the type species H. triquetra on the basionym Glenodinium triquetrum. The latter was described by Ehrenberg and is most likely a species of Kryptoperidinium. In addition to that currently unresolved nomenclatural situation, the thecal plate composition of H. triquetra sensu Stein (1883) was controversial in the past. To clarify the debate, we collected material and established the strain UTKG7 from the Baltic Sea off Kiel (Germany, the same locality as Stein had studied), which was investigated using light and electron microscopy, and whose systematic position was inferred using molecular phylogenetics. The small motile cells (18–26 μm in length) had a biconical through fusiform shape and typically were characterized by a short asymmetrically shaped, horn‐like protuberance at the antapex. A large spherical nucleus was located in the episome, whereas a single pyrenoid laid in the lower cingular plane. The predominant plate pattern was identified as apical pore complex (Po, cp?, X), 4′, 2a, 6′′, 6c, 5s, 5′′′, 2′′′′. The triradiate body scales were 254–306 nm in diameter, had 6 ridges radiating from a central spine, 9 peripheral and 3 radiating spines, and 12 peripheral bars as well as a central depression in the basal plate. Our work provides a clarification of morphological characters and a new, validly published name for this important but yet formally undescribed species of Heterocapsa: H. steinii sp. nov.  相似文献

Four strains of heterotrophic, fusiform caulobacters have been isolated from freshwater sources. A single prostheca extends from one pole of mature cells, and cells attach to various substrata by means of a holdfast located at the distal tip of the appendage. Thus, superficially these bacteria bear a strong resemblance to bacteria in the genus Caulobacter. However, unlike Caulobacter these bacteria do not exhibit a dimorphic life cycle of motile, non-stalked daughter cells and immotile, stalked mother cells. Instead both mother and daughter cells are immotile, and at the time of cell separation the daughter cells are essentially identical mirror-image replicas of the mother cell. In addition, the prosthecae of these fusiform caulobacters do not have crossbands, they are somewhat wider than the stalks of Caulobacter and the pseudostalks of Asticcacaulis, and they terminate in a bulbous tip. The deoxyribonucleic acid (DNA) base composition ranges from 54.6–60.1, well below the 62–67 range for the genus Caulobacter. Based upon these and other differences, a new genus and species, Prosthecobacter fusiformis, is proposed for the fusiform caulobacters.  相似文献

The fine structure of dividing cambial cells of Ulmus americana and Tilia americana has been studied in material fixed in glutaraldehyde followed by osmium tetroxide. The cambia examined consisted of 7–9 rows of unexpanded fusiform cells, all of which had similar ultrastructural components. The fine structure and sequence of events of mitosis and cytokinesis in the dividing cambial cells apparently are similar to those of dividing cells in root tips and leaves. Of special interest was the observation that during cytokinesis, a broad cytoplasmic plate or phragmosome precedes the developing phragmoplast and cell plate through the dividing cambial cell. Smooth and coated vesicles derived from dictyosomes are associated with cell plate formation in these cells, smooth vesicles primarily with earlier stages of plate formation, and coated vesicles in later stages.  相似文献

The activities of several different phase I and phase II drug-metabolizing enzymes were measured in freshly isolated oval cells from rats fed a choline-deficient/DL-ethionine-supplemented diet for 6 weeks and alsoin vitro in the established oval cell line OC/CDE 6. No cytochrome P450 was spectrophotometrically measurable in both preparations and two cytochrome P450-dependent monoxygenase activities, aminopyrineN-demethylase and ethoxyresorufinO-deethylase, could not be detected in the oval cells of both sources. However, cytosolic glutathione transferase, microsomal expoxide hydrolase and UDP-glucuronosyltransferase activities were clearly measurable in oval cells. Similar enzyme activities were found in freshly isolated and cultured oval cells. The highest activities of these three enzymes were detected during the exponential growth phase of the cultured cells; thereafter the activities decreased until the cells reached confluency. Changes in phenol UDP-glucuronosyltransferase (UGT1A1) mRNA levels paralleled the variations in UDP-glucuronosyltransferase activity, i.e. they were high in exponentially growing oval cells and low in confluent cell cultures. Taking into account that oval cells are able to proliferate in the livers of rats continuously fed a choline-deficient/DL-ethionine-supplemented diet and that none of the analyzed drug metabolizing enzymes are involved in the activation or detoxication ofDL-ethionine, the described pattern might be part of a more general, nonspecific, protection mechanism enabling these cells to overcome the cytotoxic effects of a variety of carcinogens and to proliferate even in their presence. A new filamentous bluegreen alga Starria zimbabweënsis gen. nov. et sp. nov. isolated from a soil sample collected near Zimbabwe, Southern Rhodesia, has a unique triradiate morphology. In transverse section each narrow cell possesses three arm-like projections, separated by 120° and in which pigment is concentrated. Filaments may be straight or twisted and various clonable biradiate forms have originated in cultures initiated from the triradiate type. Cell ultra-structure is typical of the Oscillatoriaceae except that 70 nm pit-like pores occur throughout the L-II layer of the longitudinal walls. Wall structure of deviant forms is identical to that of the triradiate “wild type.” The organism is assigned to the Oscillatoriaceae rather than to the Gomontiellaceae or to a new family.  相似文献

A study was made of the number of plastids and mitochondria present in generative cells of Solanum immediately after microspore mitosis, and the fate of these organelles during development of the pollen was determined. Changes were followed via electron microscopy of anthers of S. chacoense and S. tuberosum Group Phureja × S. chacoense. In earliest stages the generative cells were oval and had one surface along the intine and other surfaces in contact with the vegetative cell. As the pollen matured the generative cells elongated, became spindle-shaped, and were completely engulfed in the vegetative cells. At the earliest stages studied, both mitochondria and plastids were present in the generative cell. Plastids of the generative cell were, in contrast to those of the vegetative cells, fewer, smaller, and lacking in starch. Through the maturation stages the content of these organelles in the vegetative cells remained unchanged. While the generative cells retained mitochondria until anthesis, their plastids disappeared completely during maturation. This selective loss during generative cell maturation could lead to transmission of those characteristics encoded in plastid DNA through the pistillate parent only. The mechanism could explain earlier genetic evidence that plastid characters of Solanum were transmitted uniparentally.  相似文献

The ultrastructure and mechanical properties of the fusiform, triradiate and ovoid morphotypes of Phaeodactylum tricornutum were investigated using atomic force microscopy. Using topographic imaging, we showed that the surface of the ovoid form is rougher than those of the two other specimens, and coated with an outer layer of extracellular polymers. Using spatially resolved force–indentation curves, we found that the valve of the ovoid form is about five times stiffer (Young modulus of ∼500 kPa) than those of the other forms (∼100 kPa), a finding fully consistent with the fact that only the ovoid form has a silica valve, whereas the valves in the other two consist mostly of organic material. Notably, the girdle region of both fusiform and ovoid forms was five times softer than the valve, suggesting that this region is poor in silica and enriched in organic material. For the triradiate form, we showed the arms to be softer than the core region, presumably as a result of organelle localization. Last, we observed mucilaginous footprints of moderate stiffness (∼100 kPa) in the vicinity of ovoid diatoms, which we believe are secreted extracellular polymers.  相似文献

A new order, Parmales, in the Chrysophyceae has cells with siliceous walls made up of round, triradiate and sometimes oblong plates all fitting edge to edge. In the new family, Octolaminaceae, cell walls have eight plates. Cell walls in the new genus Tetraparma have four round plates and four triradiate plates. Cell walls in the new genus Triparma have three round plates of equal size, one larger round plate, one triradiate plate and three oblong plates. In the new family, Pentalaminaceae, cell walls have three round and two triradiate plates. A total of seven new species and four subspecies are described from subarctic Pacific and Antarctic waters.  相似文献

Specimens of the new species Pinuxylon woolardii have been collected from Miocene strata near Durkee, OR. These petrified specimens are unique in that, due to weathering, individual tracheids and ray cells can be separated for detailed SEM studies. This species is characterized by its high number of ray cells; numerous rays; large resin ducts which are commonly paired; primarily uniseriate pitting, two to five (usually three) small, oval or circular pinoid pits per crossfield; and smooth walls on its transverse (ray) tracheids. The latter character relates this species to taxa in the subgenus Haploxylon of Pinus. Pinuxylon woolardii is most similar to the fossil species Pinuxylon parryoides and the extant species Pinus aristata, P. cembroides, P. edulis, P. monophylla and P. balfouriana. These modern species live in dryer environments and at higher elevations than that suggested by the Miocene leaf floras which occur near the locality of P. woolardii.  相似文献

A pair of pluteus skeletal spicules arises from a pair of calcareous granules via the triradiate form. In polarized light, each spicule behaves as though carved out of a single crystal of magnesian calcite. The optic axis lies perpendicular to the plane of the triradiate and parallel to the body rod of the pluteus. However, in the scanning electron microscope, the spicule surface appeared smooth or somewhat spongy and manifested no crystal faces. Neither etching nor fracturing revealed underlying crystalline texture. Nevertheless, rhombohedral calcite crystals could be grown epitaxially onto isolated spicules immersed in a medium containing CaCl2 and NaHCO3. The optic axes of all crystals coincided with the optic axis of the spicule on which they were grown. Corresponding faces of the crystals were all aligned parallel to each other despite the complex shape of each spicule. Where the left and right spicules joined, two mutually tilted sets of crystals were observed but not crystals of intermediate orientation. Thus, the sea urchin larval spicule is built from a stack of molecularly contiguous microcrystals but its overall shape is generated by the mesenchyme cells independent of the magnesian calcite crystal habit.  相似文献
