Summary Embryogenic callus induced from mature caryopses of perennial ryegrass (Lolium perenne L.) were placed in liquid half-strength Murashige and Skoog (MS) basal medium and supplemented with 6.0 mg/l 2,4-dichlorophenoxy acetic acid (2,4-D), 3 g/l (w/v) casein hydrolysate (CH), and B5 vitamins, to initiate fast-growing highly embryogenic cell suspension cultures. Newly initiated suspension cultures contained a high level of large non-embryogenic cells (NE) with relatively few embryogenic (E) cells. Cell types were separated by discontinuous Percolls gradients or by filtering the newly initiated cultures through 31-μm nylon mesh. The growth conditions of the E cell were optimized by testing various media components including 2,4-D and sucrose, and subculture diluton ratio. Optimal shoot formation occurred after pretreatment of the embryogenic cells on solidified callus maintenance medium supplemented with 60 mg/l cefotaxime for 4 weeks prior to transfer to regeneration medium Regeneration media consisted of half-strength MS basal medium supplemented with B5 vitamins, 0.5 mg/l fluridone, and 0.5 mg/l BA. Most plants regenerated were albino with only a few green plants. Journal Paper number MAES 2959 of the Massachusetts Agricultural Experiment Station.  相似文献

Autotoxicity and allelopathy affect the yield of GL66 and GL72, two populations of perennial ryegrass (Lolium perenne L. cv. S23) that were originally selected for contrasting rates of mature leaf dark respiration, but under conditions where allelopathic effects could not occur and autotoxic effects were minimal. To investigate if the respiration properties are also affected, the populations were subjected to two conditions (monoculture and mixed culture) and two treatments (grown in `renewed', i.e. solution completely replaced every week, and `replenished', i.e. nutrients added to the solution every week, to compensate for the nutrients that had been absorbed from the solution). When plants were grown in mixed culture with a `renewed' nutrient solution, the rate of mature leaf respiration in the absence of inhibitors was higher in GL66 than in GL72. In GL66, this rate was unaffected by condition or treatment, whereas in GL72 it increased in the `replenished' cultures, due to autotoxic and allelopathic effects. In contrast with GL66, for GL72 the effect of an inhibitor of the alternative path (salicylhydroxamic acid, SHAM) differed between conditions and treatments. Effects of an inhibitor of the cytochrome path (KCN) were affected by treatment in both populations. The increase in the rate of root respiration of GL66 due to autotoxic and allelopathic effects was accompanied by a higher rate of the SHAM-resistant component, i.e. maximum activity of the cytochrome path. Roots of GL72 showed a higher control respiration rate and a higher rate of the SHAM-resistant component when long-term accumulation of root exudates was allowed to occur in mixed cultures (allelopathy). We conclude that allelochemicals can modify both leaf and root respiration and that there is no consistent correlation between yield and respiration for GL66 and GL72 in the present study. This shows that selection for low rates of mature leaf respiration is not an appropriate method to select for high-yielding cultivars in perennial ryegrass.  相似文献

Our study examined the influence of elevated ozone levels on the growth and mycorrhizal colonization of two populations of Elymus glaucus L. (blue wildrye). We hypothesized that ozone would reduce carbon availability to the plants, particularly below ground, and would affect mycorrhizal colonization. Because of the wide geographic range of E. glaucus, two populations of plants were selected from areas of contrasting ozone histories to examine intraspecies variation in response to ozone. Two populations of E. glaucus (southern California versus northern California) exposed in a factorial experiment involving ozone, mycorrhizal inoculation with Glomus intraradices Schenck and Smith, and plant source population. Ozone had a subtle effect on leaf area and number of tillers but did not affect overall root:shoot ratio in either population. The impact of ozone on above-ground growth characteristics was most pronounced in the southern population that came from a high-ozone environment, while below-ground responses such as reduced arbuscular colonization was most pronounced in the northern population which originated in a low-ozone environment. Further analysis of soil characteristics from the northern population of plants revealed a significant reduction in active soil bacterial biomass and an increase in total fungi per gram dry weight soil, suggesting a possible role for ozone in altering soil processes. Whether or not population differences in response to ozone were due to genetic shifts resulting from prior ozone remains to be determined. However, these subtle but important differences in population response to ozone above- and below-ground have significant implications in any attempt to generalize plant response, even within a species. Future research efforts need to include better characterization of intraspecific variation in response to ozone as well as possible adaptive strategies that may result from chronic ozone exposure.  相似文献

Tissue culture techniques, medium composition, pH value and targeted tissues, agroinfection and co-culture conditions, selection process were optimized for efficient turfgrass transformation. A highly regenerable callus lines were produced in callus induction medium modified from N6 basal medium. Six-week-old calluses were cultured on Pre-regeneration medium I for 4 days and then subjected to Agrobacterium tumefaciens. After co-cultivation at 20±1 °C in a 16 h light/8 h darkness for 3 days, the calluses were cultured on non-selective Pre-regeneration medium II supplemented with 400 mg l−1 l-cysteine for 7 days. Plantlets were regenerated on the Regeneration medium without selection pressure. A selection pressure was given to the regenerated plantlets when they were rooted on the Plantlet rooting medium. Roots appeared within 8–12 days in putative transformed plantlets. Resistant plants obtained were phenotypically normal and fully fertile. Chemical and molecular analyses confirmed that foreign genes were successfully introduced into the genome of perennial ryegrass or tall fescue. The transformation efficiency can attain 23.3% in perennial ryegrass.  相似文献

A field study was carried out near Zürich (Switzerland) to determine the yield of symbiotically fixed nitrogen (15N dilution) from white clover (Trifolium repens L.) grown with perennial ryegrass (Lolium perenne L) and from red clover (Trifolium pratense L.) grown with Italian ryegrass (Lolium multiflorum Lam.). A zero N fertilizer treatment was compared to a 30 kg N/ha per cut regime (90 to 150 kg ha−1 annually). The annual yield of clover N derived from symbiosis averaged 131 kg ha−1 (49 to 227 kg) without N fertilization and 83 kg ha−1 (21 to 173 kg) with 30 kg of fertilizer N ha−1 per cut in the seeding year. Values for the first production year were 308 kg ha−1 (268 to 373 kg) without N fertilization and 232 kg ha−1 (165 to 305 kg) with 30 kg fertilizer N ha−1 per cut. The variation between years was associated mainly with the proportion of clover in the mixtures. Apparent clover-to-grass transfer of fixed N contributed up to 52 kg N ha−1 per year (17 kg N ha−1 on average) to the N yield of the mixtures. Percentage N derived from symbiosis averaged 75% for white and 86% for red clover. These percentages were affected only slightly by supplemental nitrogen, but declined markedly during late summer for white clover. It is concluded that the annual yield of symbiotically fixed N from clover/grass mixtures can be very high, provided that the proportion of clover in the mixtures exceeds 50% of total dry mass yield.  相似文献

A laboratory incubation experiment with 15N labeled root and leaf residues of 3 agroforestry species (Leucaena leucocephala, Dactyladenia barteri and Flemingia macrophylla) was conducted under controlled conditions (25 C) for 56 days to quantify residue C and N mineralization and its relationship with residue quality. No uniform relation was found between the chemical composition of the above and below residues. The leucaena and dactyladenia roots contained more lignin (8 and 26% respectively) and less N (2.0 and 1.0% respectively) than the respective leaves (2 and 13% lignin and 2.9 and 1.4% N, respectively), whereas the differences between the lignin and N contents of the flemingia leaves and roots were not significant (4.6 and 3.0% lignin and 2.63 and 2.68% N, respectively). The leucaena leaves contained more polyphenols than the roots (6.4 and 3.6%), while the polyphenol content of the leaves and roots of the other residues was similar (5.0 and 5.1% for dactyladenia and 4.0 and 3.5% for flemingia).Three patterns of N mineralization could be distinguished. A first pattern, followed by residues producing the highest amounts of CO2, showed an initial immobilization of soil derived N, followed by a net release of both soil and residue derived N after 7 days of incubation. A second pattern, followed by the flemingia leaf residues which produced intermediate amounts of CO2 and had an intermediate quality, showed no significant immobilization of soil derived N, and significant mineralization of residue N. A third pattern, followed by both low quality dactyladenia residues, showed a low release of residue derived N and a continued inmobilization of soil derived N.Residue C mineralization was significantly (p<0.05) correlated with the residue lignin content, C-to-N ratio, and polyphenol-to-N ratio. The proportion of residue N mineralized (immobilized) after 56 days of incubation was significantly correlated with the residue N content (p<0.01) and the C-to-N ratio (p<0.05). The relations were quadratic, rather than linear. The ratio of the proportion of residue N mineralized (immobilized) over the proportion of residue C mineralized after 56 days was highly significantly correlated with the lignin content (p<0.01) and C-to-N (p<0.001), lignin-to-N (p<0.01), polyphenol-to-N (p<0.01) and (lignin+polyphenol)-to-N ratios (p<0.01) in a linear way. This indicates that due to the low availability of the residue C, relatively less N is immobilized for the very low quality residues ((lignin+polyphenol)-to-N ratio: 29.7) than for the residues with a relatively higher quality ((lignin+polyphenol)-to-N ratios between 3.3 and 12.5).  相似文献

A method is evaluated that employs variation in stable C and N isotopes from fractionations in C and N acquisition and growth to predict root biomasses of three plant species in mixtures. Celtis laevigata Willd. (C3), Prosopis glandulosa Torr. (C3, legume) and Schizachyrium scoparium (Michx.) Nash (C4), or Gossypium hirsutum L. (C3), Glycine max (L.) Merr. (C3 legume), and Sorghum bicolor (L.) Moench (C4) were grown together in separate, three-species combinations. Surface roots (0–10 cm depth) of each species from each of the two combinations were mixed in various proportions, and the relative abundances of 15N and 14N and 13C and 12C in prepared mixtures, surface roots of single species, and roots extracted from the 80-cm soil profile in which each species combination was grown were analyzed by mass spectrometry. An algebraic determination which employed the δ 13C, % 15N, and C and N concentrations of root subsamples of individual species accounted for more than 95% of the variance in biomass of each species in prepared mixtures with G. max, G. hirsutum, and S. bicolor. A similar analysis demonstrated species-specific differences in rooting patterns. Root biomasses of the C4 monocots in each combination, S. scoparium and S. bicolor, were concentrated in the upper 20 cm of soil, while those of G. hirsutum and the woody P. glandulosa were largest in lower soil strata. Analyses of stable C and N isotopes can effectively be used to distinguish roots of species which differ in ratios of 15N to 14N and 13C to 12C and thus to study belowground competition between or rooting patterns of associated species with different C and N isotope signatures. The method evaluated can be extended to quantify aboveground and belowground biomasses of component species in mixtures with isotopes of other elements or element concentrations that differ consistently among plants of interest.  相似文献

Summary The uptake of Na and K by perennial ryegrass from flowing solution culture with monitored concentrations of Na and K was followed in two experiments. In the first, when only 50 and 10 per cent of the K uptake by one set of plants, grown with K held constant at 2.5 μeq 1−1, was supplied to two other linked sets of plants and the balance supplied as Na, there was a rapid decrease in K, and an increase in Na, concentration in the shoots over a 20-day period. However, when compared with the plants grown in K in solution held constant, there was not a complete replacement of Na for K. In the second experiment the concentration of K in the culture solution was held constant at 2 μeq 1−1 and Na at 0, 5, 25, 50 and 100 μeq 1−1. Although uptake of Na increased with increasing concentration in solution the contents in the plants were low,i.e. less than 0.19 per cent and decreased with time. There was an increase in the yield of both shoots and roots with increasing Na in the solution; it was suggested that, during the early stages of growth there may have been an inadequate supply of K and that Na may have substituted for K in some of the non-specific roles of K in the plants. There was evidence in both experiments that a flux of H-ions was involved in the uptake of Na.  相似文献

Microautoradiographs showed that [14C]sucrose taken up in the xylem of small and intermediate (longitudinal) vascular bundles of Zea mays leaf strips was quickly accumulated by vascular parenchyma cells abutting the vessels. The first sieve tubes to exhibit 14C-labeling during the [14C]sucrose experiments were thick-walled sieve tubes contiguous to the more heavily labeled vascular parenchyma cells. (These two cell types typically have numerous plasmodesmatal connections.) With increasing [14C]sucrose feeding periods, greater proportions of thick- and thin-walled sieve tubes became labeled, but few of the labeled thin-walled sieve tubes were associated with labeled companion cells. (Only the thin-walled sieve tubes are associated with companion cells.) When portions of leaf strips were exposed to 14CO2 for 5 min, the vascular parenchyma cells-regardless of their location in relation to the vessels or sieve tubes-were the most consistently labeled cells of small and intermediate bundles, and label (14C-photosynthate) appeared in a greater proportion of thin-walled sieve tubes than thick-walled sieve tubes. After a 5-min chase with 12CO2, the thin-walled sieve tubes were more heavily labeled than any other cell type of the leaf. After a 10-min chase with 12CO2, the thin-walled sieve tubes were even more heavily labeled. The companion cells generally were less heavily labeled than their associated thin-walled sieve tubes. Although all of the thick-walled sieve tubes were labeled in portions of leaf strips fed 14CO2 for 5 min and given a 10-min 12CO2 chase, only five of 72 vascular bundles below the 14CO2-exposed portions contained labeled thick-walled sieve tubes. Moreover, the few labeled thick-walledsieve tubes of the transport region always abutted 14C-labeled vascular parenchyma cells. The results of this study indicate that (1) the vascular parenchyma cells are able to retrieve at least sucrose from the vessels and transfer it to the thick-walled sieve tubes, (2) the thick-walled sieve tubes are not involved in long-distance transport, and (3) the thin-walled sieve tubes are capable themselves of accumulating sucrose and photosynthates from the apoplast, without the companion cells serving as intermediary cells.  相似文献

Partitioning of 14C was assessed in sweet chestnut seedlings (Castanea sativa Mill.) grown in ambient and elevated atmospheric [CO2] environments during two vegetative cycles. The seedlings were exposed to 14CO2 atmosphere in both high and low [CO2] environments for a 6-day pulse period under controlled laboratory conditions. Six days after exposure to 14CO2, the plants were harvested, their dry mass and the radioactivity were evaluated. 14C concentration in plant tissues, root-soil system respiratory outputs and soil residues (rhizodeposition) were measured. Root production and rhizodeposition were increased in plants growing in elevated atmospheric [CO2]. When measuring total respiration, i.e. CO2 released from the root/soil system, it is difficult to separate CO2 originating from roots and that coming from the rhizospheric microflora. For this reason a model accounting for kinetics of exudate mineralization was used to estimate respiration of rhizospheric microflora and roots separately. Root activity (respiration and exudation) was increased at the higher atmospheric CO2 concentration. The proportion attributed to root respiration accounted for 70 to 90% of the total respiration. Microbial respiration was related to the amount of organic carbon available in the rhizosphere and showed a seasonal variation dependent upon the balance of root exudation and respiration. The increased carbon assimilated by plants grown under elevated atmospheric [CO2] stayed equally distributed between these increased root activities. ei]H Lambers  相似文献

Chloroplasts with high rates of photosynthetic O2 evolution (up to 120 mol O2· (mg Chl)-1·h-1 compared with 130 mol O2· (mg Chl)-1·h-1 of whole cells) were isolated from Chlamydomonas reinhardtii cells grown in high and low CO2 concentrations using autolysine-digitonin treatment. At 25° C and pH=7.8, no O2 uptake could be observed in the dark by high- and low-CO2 adapted chloroplasts. Light saturation of photosynthetic net oxygen evolution was reached at 800 mol photons·m-2·s-1 for high- and low-CO2 adapted chloroplasts, a value which was almost identical to that observed for whole cells. Dissolved inorganic carbon (DIC) saturation of photosynthesis was reached between 200–300 M for low-CO2 adapted chloroplasts, whereas high-CO2 adapted chloroplasts were not saturated even at 700 M DIC. The concentrations of DIC required to reach half-saturated rates of net O2 evolution (Km(DIC)) was 31.1 and 156 M DIC for low- and high-CO2 adapted chloroplasts, respectively. These results demonstrate that the CO2 concentration provided during growth influenced the photosynthetic characteristics at the whole cell as well as at the chloroplast level.Abbreviations Chl chlorophyll - DIC dissolved inorganic carbon - Km(DIC) coneentration of dissolved inorganic carbon required for the rate of half maximal net O2 evolution - PFR photon fluence rate - SPGM silicasol-PVP-gradient medium  相似文献

A model for rhizodeposition and root respiration was developed and parameterised based on 14C pulse labelling of Lolium perenne. The plants were grown in a two-compartment chamber on a loamy Haplic Luvisol under controlled laboratory conditions. The dynamics of 14CO2 efflux from the soil and 14C content in shoots, roots, micro-organisms, dissolved organic carbon (DOC) and soil were measured during the first 11 days after labelling. Modelled parameters were estimated by fitting on measured 14C dynamics in the different pools. The model and the measured 14C dynamics in all pools corresponded well (r 2=0.977). The model describes well 14CO2 efflux from the soil and 14C dynamics in shoots, roots and soil, but predicts unsatisfactorily the 14C content in micro-organisms and DOC. The model also allows for division of the total 14CO2 efflux from the soil in 14CO2 derived from root respiration and 14CO2 derived from rhizomicrobial respiration by use of exudates and root residues. Root respiration and rhizomicrobial respiration amounted for 7.6% and 6.0% of total assimilated C, respectively, which accounts for 56% and 44% of root-derived 14CO2 efflux from the soil planted with 43-day-old Lolium perenne, respectively. The sensitivity analysis has shown that root respiration rate affected the curve of 14CO2 efflux from the soil mainly during the first day after labelling. The changes in the exudation rate influenced the 14CO2 efflux later than first 24 h after labelling.  相似文献

The effects of elevated [CO2] (700 μl l-1 CO2) and temperature increase (+3 °C) on carbon turnover in grassland soils were studied during 2.5 years at two N fertiliser supplies (160 and 530 kg N ha-1 y-1) in an experiment with well-established ryegrass swards (Lolium perenne) supplied with the same amounts of irrigation water. During the growing season, swards from the control climate (350 μl l-1 [CO2] at outdoor air temperature) were pulse labelled by the addition of 13CO2. The elevated [CO2] treatments were continuously labelled by the addition of fossil-fuel derived CO2 (13 C of -40 to -50 ‰). Prior to the start of the experimental treatments, the carbon accumulated in the plant parts and in the soil macro-organic matter (‘old’ C) was at −32‰. During the experiment, the carbon fixed in the plant material (‘new’ C) was at −14 and −54‰ in the ambient and elevated [CO2] treatments, respectively. During the experiment, the 13C isotopic mass balance method was used to calculate, for the top soil (0–15 cm), the carbon turnover in the stubble and roots and in the soil macro-organic matter above 200 μ (MOM). Elevated [CO2] stimulated the turnover of organic carbon in the roots and stubble and in the MOM at N+, but not at N−. At the high N supply, the mean replacement time of ‘old’ C by ‘new’ C declined in elevated, compared to ambient [CO2], from 18 to 7 months for the roots and stubble and from 25 to 17 months for the MOM. This resulted from increased rates of ‘new’ C accumulation and of ‘old’ C decay. By contrast, at the low N supply, despite an increase in the rate of accumulation of ‘new’ C, the soil C pools did not turnover faster in elevated [CO2], as the rate of ‘old’ C decomposition was reduced. A 3 °C temperature increase in elevated [CO2] decreased the input of fresh C to the roots and stubble and enhanced significantly the exponential rate for the ‘old’ C decomposition in the roots and stubble. An increased fertiliser N supply reduced the carbon turnover in the roots and stubble and in the MOM, in ambient but not in elevated [CO2]. The respective roles for carbon turnover in the coarse soil OM fractions, of the C:N ratio of the litter, of the inorganic N availability and of a possible priming effect between C-substrates are discussed. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Summary Mono-specific communities of the C3 sedge, Scirpus olneyi and the C4 grass, Spartina patens, were exposed to normal ambient or elevated CO2, (ca. 680 l l–1) throughout the 1987 and 1988 growing seasons in open-top field chambers located on a tidal marsh. Single stems of C3 plants grown in ambient or elevated CO2 showed an increased photosynthetic rate when tested at elevated CO2 for both seasons. This increase in photosynthetic response in the C3 species was maintained throughout the 1987 and 1988 growing season. The stimulation of photosynthesis with elevated CO2 appeared to increase as temperature increased and decreased as photosynthetic photon flux (PPF) increased. Analysis of the photosynthetic response of the C3 species during the 1988 season indicated that significant differences in light-saturated photosynthetic rate between ambient and elevated CO2 conditions continued until October. In contrast to the C3 sedge, the C4 grass showed no significant photosynthetic increase to elevated CO2 except at the beginning of the 1988 season.  相似文献

Intergeneric hybridization between Festuca and Lolium has been a long-term goal of forage and turfgrass breeders to generate improved cultivars by combining stress tolerance of Festuca and rapid establishment of Lolium. However, wide-distance hybridizations usually result in the wild genome being eliminated from the hybrid due to incomplete chromosome pairing and crossovers. In this study, random amplified polymorphic DNA (RAPD) and simple sequence repeat (SSR) markers were used to detect the parental genome composition of F1 hybrids and backcross, generated from crosses between Festuca mairei St. Yves (Fm) and Lolium perenne L. (Lp). Each of the hybrids exhibited integration of Fm and Lp genomes with varying levels of Fm/Lp genome ratios. However, cluster and principle component analyses of the progeny consistently revealed four groups depending on the amount of genome introgression from both parents. The parental genome composition and classifications of intergeneric progeny would be useful for breeding material selection. Photosystem II chlorophyll fluorescence and leaf net gas exchanges (CO2 and H2O) were measured simultaneously on bean leaves (Phaseolus vulgaris L.) submitted either to different ambient CO2 concentrations or to a drought stress. When leaves are under photorespiratory conditions, a simple fluorescence parameter F/ Fm (B. Genty et al. 1989, Biochem. Biophys. Acta 990, 87–92; F = difference between maximum, Fm, and steady-state fluorescence emissions) allows the calculation of the total rate of photosynthetic electron-transport and the rate of electron transport to O2. These rates are in agreement with the measurements of leaf O2 absorption using 18O2 and the kinetic properties of ribulose-1,5bisphosphate carboxylase/oxygenase. The fluorescence parameter, F/Fm, showed that the allocation of photosynthetic electrons to O2 was increased during the desiccation of a leaf. Decreasing leaf net CO2 uptake, either by decreasing the ambient CO2 concentration or by dehydrating a leaf, had the same effect on the partitioning of photosynthetic electrons between CO2 and O2 reduction. It is concluded that the decline of net CO2 uptake of a leaf under drought stress is only due, at least for a mild reversible stress (causing at most a leaf water deficit of 35%), to stomatal closure which leads to a decrease in leaf internal CO2 concentration. Since, during the dehydration of a leaf, the calculated internal CO2 concentration remained constant or even increased we conclude that this calculation is misleading under such conditions.Abbreviations Ca, Ci ambient, leaf internal CO2 concentrations - Fm, Fo, Fs maximum, minimal, steady-state fluorescence emission - Fv variable fluorescence emission - PPFD photosynthetic photon flux density - qp, qN photochemical, non-photochemical fluorescence quenching - Rubisco ribulose-1,5-bisphosphate carboxylase/oxygenase  相似文献

The kidney is an organ playing an important role in ion regulation in both freshwater (FW) and seawater (SW) fish. The mechanisms of ion regulation in the fish kidney are less well studied than that of their gills, especially at the level of transporter proteins. We have found striking differences in the pattern of Na+/K+/2Cl- cotransporter (NKCC) expression between species. In the killifish kidney, NKCC is apically localized in the distal and collecting tubules and basolaterally localized in the proximal tubules. However, in the SW killifish gill, NKCC is basolaterally co-localized with Na+/K+-ATPase, whereas in FW, NKCC immunoreactivity is primarily apical, although still colocalized within the same mitochondria-rich cell with basolateral Na+/K+-ATPase. Rainbow trout kidney has NKCC only in the apical membrane of the distal and collecting tubules in both environments, with no signal being detected in the proximal tubule. On the other hand, in the trout gill, NKCC is found basolaterally in both FW and SW environments. An important observation is that, in the gills of rainbow trout, the trailing edge of the filament possesses mostly Na+/K+-ATPase-positive but NKCC-negative mitochondria-rich cells, whereas in the region between and at the roots of the gill lamellae, most mitochondria-rich cells exhibit both Na+/K+-ATPase- and NKCC-positive immunoreactivity. These results suggest that the differential localization of transporters between the two species represents differences in function between these two euryhaline fishes with different life histories and strategies. The increasing CO2 concentration in Earths atmosphere is expected to cause a greater decline in the nutritional quality of C3 than C4 plants. As a compensatory response, herbivorous insects may increase their feeding disproportionately on C3 plants. These hypotheses were tested by growing the grasses Lolium multiflorum C3) and Bouteloua curtipendula C4) at ambient (370 ppm) and elevated (740 ppm) CO2 levels in open top chambers in the field, and comparing the growth and digestive efficiencies of the generalist grasshopper Melanoplus sanguinipes on each of the four plant × CO2 treatment combinations. As expected, the nutritional quality of the C3 grass declined to a greater extent than did that of the C4 grass at elevated CO2; protein levels declined in the C3 grass, while levels of carbohydrates (sugar, fructan and starch) increased. However, M. sanguinipes did not significantly increase its consumption rate to compensate for the lower nutritional quality of the C3 grass grown under elevated CO2. Instead, these grasshoppers appear to use post-ingestive mechanisms to maintain their growth rates on the C3 grass under elevated CO2. Consumption rates of the C3 and C4 grasses were also similar, demonstrating a lack of compensatory feeding on the C4 grass. We also examined the relative efficiencies of nutrient utilization from a C3 and C4 grass by M. sanguinipes to test the basis for the C4 plant avoidance hypothesis. Contrary to this hypothesis, neither protein nor sugar was digested with a lower efficiency from the C4 grass than from the C3 grass. A novel finding of this study is that fructan, a potentially large carbohydrate source in C3 grasses, is utilized by grasshoppers. Based on the higher nutrient levels in the C3 grass and the better growth performance of M. sanguinipes on this grass at both CO2 levels, we conclude that C3 grasses are likely to remain better host plants than C4 grasses in future CO2 conditions.  相似文献
