Mediterranean junipers are of special ecological importance as key components of resource islands in semi-arid mountain ecosystems of the Mediterranean basin. The fragmentation of their habitat, which was primarily natural and driven by climatic drought conditions, is currently being aggravated by anthropogenic pressure. In the framework of this concern, the present work aims to contribute establishing a genomic profile of Juniperus in its western Mediterranean range, with a special emphasis placed on J. thurifera. DNA contents were assessed by flow cytometry in 43 populations of nine taxa within their Mediterranean range (first reports for J. navicularis, J. thurifera subsp. africana and J. thurifera subsp. thurifera). Chromosome numbers were determined by orcein staining in eight taxa (first counts for J. oxycedrus subsp. badia, J. phoenicea subsp. phoenicea, J. phoenicea subsp. turbinata, of 2n?=?2x?=?22, and for J. thurifera subsp. thurifera, of 2n?=?4x?=?44). Tetraploid cytotypes have been the only ones found in the 19 populations of J. thurifera studied, this being the first report of a Juniperus species exclusively polyploid. In J. thurifera, C-value does not respond to habitat fragmentation, in the same way that genetic diversity within populations was previously shown to be unaltered, suggesting that this factor has not had, at least to date, a significant impact on populations at genomic and genetic levels. Habitat fragmentation leads to deeply age-biased populations with a male-biased imbalanced sex ratio (lack of females), indicating an urgent need to improve regeneration within the populations of this species.  相似文献

We previously reported on the importance of osteoactivin (OA/Gpnmb) in osteogenesis. In this study, we examined the role of OA in osteoclastogenesis, using mice with a nonsense mutation in the Gpnmb gene (D2J) and wild-type controls (D2J/Gpnmb+). In these D2J mice, micro-computed tomography and histomorphometric analyses revealed increased cortical thickness, whereas total porosity and eroded surface were significantly reduced in D2J mice compared with wild-type controls, and these results were corroborated by lower serum levels of CTX-1. Contrary to these observations and counterintuitively, temporal gene expression analyses supported up-regulated osteoclastogenesis in D2J mice and increased osteoclast differentiation rates ex vivo, marked by increased number and size. The finding that MAPK was activated in early differentiating and mature D2J osteoclasts and that survival of D2J osteoclasts was enhanced and mediated by activation of the AKT-GSK3β pathway supports this observation. Furthermore, this was abrogated by the addition of recombinant OA to cultures, which restored osteoclastogenesis to wild-type levels. Moreover, mix and match co-cultures demonstrated an induction of osteoclastogenesis in D2J osteoblasts co-cultured with osteoclasts of D2J or wild-type. Last, in functional osteo-assays, we show that bone resorption activity of D2J osteoclasts is dramatically reduced, and these osteoclasts present an abnormal ruffled border over the bone surface. Collectively, these data support a model whereby OA/Gpnmb acts as a negative regulator of osteoclast differentiation and survival but not function by inhibiting the ERK/AKT signaling pathways.  相似文献

Studies were made of the dependence of the rate of oxygen consumption, Jr, on the electrical potential difference, Δψ, across the frog skin. After the abolition of sodium transport by ouabain the basal oxygen consumption was independent of Δψ. In fresh skins Jr was a linear function of Δψ over a range of at least ±70 mv. Treatment with aldosterone stimulated the short-circuit current, Io, and the associated rate of oxygen consumption, Jro, and increased their stability; linearity was then demonstrable over a range of ±160 mv. Brief perturbations of Δψ (±30–200 mv) did not alter subsequent values of Io. Perturbations for 10 min or more produced a "memory" effect both with and without aldosterone: accelerating sodium transport by negative clamping lowered the subsequent value of Io; positive clamping induced the opposite effect. Changes in Jro were more readily detectable in the presence of aldosterone; these were in the same direction as the changes in Io. The linearity of Jr in Δψ indicates the validity of analysis in terms of linear nonequilibrium thermodynamics—brief perturbations of Δψ appear to produce no significant effect on either the phenomenological coefficients or the free energy of the metabolic driving reaction. With randomized complete block design, long-term water use efficiency (WUEL), photosynthetic parameters (net photosynthetic rate Pn, transpiration rate Tr, stomatal conductance Gs, instantaneous water use efficiency WUEi, maximum photosynthetic rate P max, light saturation pointLSP, carboxylic efficiency Ce), stomatal properties (frequency, size, superior/inferior stoma amount ratio) and root/shoot ratio of twelve Populus deltoids clones were studied under water stress in a greenhouse. Four water treatments were designed, namely, well-watered condition, slight water stress, moderate water stress, and severe water stress. Although the volumes of irrigated water for different treatments were the same, the irrigation intervals were strictly controlled. The results showed that long-term water use efficiency (WUEL) differences among tested clones were obvious and became greater with the increase in water stress. J2, J6, J7, J8, and J9 were excellent clones with high WUEL. Stomatal frequency, stomatal size, Gs, Tr, Pn, and root/shoot ratio of J2et al were moderate, whereas the superior/inferior ratio, Pmax, LSP, Ce of J2et al were higher than other clones. The interrelations between WUEL and the physiological parameters were analyzed. The results revealed that the WUEL differences were induced by a series of physiological parameters, and clones with higher WUEL always had strong photosynthetic capacity, higher WUEi, and optimum root/shoot ratio.  相似文献

The absorption characteristics of artepillin C (AC), an active ingredient of Brazilian propolis, were examined by measuring permeation across Caco-2 cell monolayers. The permeation rate in the basolateral-to-apical direction, Jbl → ap, in the presence of proton gradient was 0.14 nmol/min/mg protein, whereas Jbl → ap in the absence of proton gradient was 1.14 nmol/min/mg protein. The latter value is nearly the same as the permeation rate in the apical-to-basolateral direction, Jap → bl, both in the presence and absence of proton gradient. In the presence of proton gradient, Jap → bl was almost constant, irrespective of NaN3 or benzoic acid. However, Jbl → ap dramatically increased upon the addition of NaN3 or benzoic acid specifically to the apical side. In both the presence and absence of proton gradient, Jap → bl also appeared to be constant irrespective of the paracellular permeability of Caco-2 cells. After AC was loaded apically in the presence of proton gradient, the intracellular AC increased with time. This accumulation was inhibited by apically loaded NaN3. These indicate that AC transport occurs mainly via transcellular passive diffusion, although a considerable amount of AC was taken up intracellularly by monocarboxylic acid transporter (MCT) on the apical side and not transported out across the basolateral membrane, suggesting that different subtypes of MCT are involved.  相似文献

The Levene model is the simplest mathematical model to describe the evolution of gene frequencies in spatially subdivided populations. It provides insight into how locally varying selection promotes a population’s genetic diversity. Despite its simplicity, interesting problems have remained unsolved even in the diallelic case.In this paper we answer an open problem by establishing that for two alleles at one locus and J demes, up to 2J−1 polymorphic equilibria may coexist. We first present a proof for the case of stable monomorphisms and then show that the result also holds for protected alleles. These findings allow us to prove that any odd number (up to 2J−1) of equilibria is possible, before we extend the proof to even numbers. We conclude with some numerical results and show that for J>2, the proportion of parameter space affording this maximum is extremely small.  相似文献

Tumor necrosis factor alpha (Tnf) plays a pleiotropic role in murine malaria. Some investigations have correlated Tnf with hypothermia, hyperlactatemia, hypoglycemia, and a suppression of the erythropoietic response, although others have not. In this study, we have evaluated parasitemia, survival rate and several pathological features in C57BL/6JTnf−/− and C57BL/6JTnf+/+ mice after infection with Plasmodium chabaudi adami 408XZ. Compared to the C57BL/6JTnf+/+ mice, C57BL/6JTnf−/− mice showed increased parasitemia and decreased survival rate, whereas blood glucose, blood lactate and body weight were not significantly different. However, C57BL/6JTnf−/− mice suffered significantly more from severe anemia and hypothermia than C57BL/6JTnf+/+ mice. These results suggest that Tnf is an important mediator of parasite control, but not of anemia development. We hypothesize that the high mortality observed in the Tnf knock-out mice is due to increased anemia and pathology as a direct result of increased levels of parasitemia.  相似文献

The exchange of cell K with K42, J K, has been measured in cat right ventricular papillary muscle under conditions of a steady state with respect to intracellular K concentration. Within the limits of the measurement, all of cell K exchanged at a single rate. Cells from small cats are smaller and have larger surface/volume ratios than cells from large cats. The larger surface/volume ratio results in larger flux values. J K increases in an approximately linear manner as the external K concentration is increased twentyfold, from 2.5 to 50 mM, at constant intracellular K concentration. The permeability for K ions, P K, calculated from the influx and membrane potential, remains very nearly constant over this range of external K concentrations. J K is not affected by replacement of O2 by N2, or by stimulated contractions at 60 per minute, but K influx decreases markedly in 10-5 M and 10-8 M ouabain.  相似文献

In this study, we examined steady-state and dynamic photosynthetic performance and leaf nitrogen (N) partitioning in the typical shade-demanding herb Panax notoginseng grown along a light gradient. Gas exchange on a leaf area basis was significantly reduced under low irradiance, with gas exchange on a leaf mass basis reaching a maximum value and then decreasing along the light gradient. Specific leaf area significantly increased with decreasing irradiance levels (P < 0.001), whereas carboxylation efficiency was decreased (P < 0.001). In addition, decreasing growth irradiance levels led to declines in maximum carboxylation rate (V cmax) and maximum electron transport rate (J max), although V cmax/mass and J max/mass were relatively less affected than V cmax/area and J max/area. Slow photosynthetic response to simulated sunflecks was observed under low levels of growth irradiance, with stomatal limitations only detected in leaves grown under low-light conditions. Chlorophyll content increased significantly with decreasing irradiance levels. N content on a leaf mass basis apparently increased, while N content on a leaf area basis markedly decreased. The fraction of leaf N allocated to light-harvesting components increased significantly with decreasing growth irradiance levels, whereas the fraction allocated to carboxylation and bioenergetics was significantly reduced. As an adaptation strategy to growth irradiance, we conclude that adjustments in specific leaf area may be more important than changes in leaf physiology and biochemistry in typical shade-demanding species such as P. notoginseng.  相似文献

Precursors of the tail of bacteriophage λ have been detected by measurements of in vitro complementation activities and serum blocking activity in sucrose gradients of lysates defective in tail genes.On the basis of these measurements, a pathway for the assembly of the λ tail is proposed:The morphogenesis of the λ tail starts from the tail fiber (product of gene J) located at the distal end of the tail, and proceeds to the proximal end. Gene J by itself produces a 15 S structure with serum blocking activity but without any detectable in vitro complementation activity, which may be the least advanced precursor of the λ tail or an abortive product. Functions of genes J, I, K, L are required for the formation of a 15 S precursor that has in vitro complementation activities with J, I, K and L lysates and serum blocking activity. If the products of genes G and H act on the latter 15 S precursor, a 25 S precursor is made, but this precursor seems either to be in equilibrium with the 15 S precursor or to degrade easily into the 15 S precursor. Gene M has a function of stabilizing the 25 S precursor. After the action of gene M product, the 25 S precursor is ready to serve as a nucleus on which the product of gene V (the major tail protein) assembles. However, gene U product is also necessary at this step for the correct assembly of the major tail protein on the 25 S precursor. Without gene U product the assembly of the major tail protein does not stop at the correct length and a polytail is formed instead of a morphologically normal tail. Finally, gene Z product acts on the morphologically normal tail and makes it a biologically active tail. Without the action of gene Z product, the defective tail binds to a head and forms a phage-like particle which is only very weakly infectious. (The position of gene T in the pathway is not determined, because no sus mutant is available in gene T.)Two abnormal, less efficient pathways are also present in vitro. (1) If gene U product acts on a polytail in an U lysate, the polytail finally binds to a head and forms a phage particle with an extra long tail which is infectious to a small extent. (2) The function of gene K seems to be bypassed to some extent: K lysates accumulate particles which sediment as fast as normal phage and which are complemented by other tail lysates.  相似文献

Volume (Jv) and solute (Js) fluxes through Phaseolus root systems were observed over a 24-hour period. The volume flux was varied in a pressure chamber by altering the hydrostatic pressure in 10 steps, from 0 to 0.41 megapascals. All root systems showed strong diurnal peaks in volume flux. The five transport coefficients (σ, ω, Js*, Lp, and π*) were estimated from a nonlinear least squares algorithm. Analysis of the data revealed that all the coefficients exhibited a diurnal rhythm. When the total differential of the volume flux was considered it was possible to show that the diurnal changes in volume flux were due to a complex interaction between the diurnally shifting coefficients with the role of each highly dependent on the level of volume flux. At low volume fluxes, ω, Js*, and π* accounted for nearly all the diurnal change in volume flux. At high volume fluxes, however, the major influence shifted to Lp and π*, while ω and Js* became relatively unimportant. Thus, π* was the only coefficient of interest across the entire range of Jv and appeared to be the single most important one in determining the diurnal rhythm of Jv under conditions of a constant applied pressure.  相似文献

Discussions of active transport usually assume stoichiometry between the rate of transport J+ and the metabolic rate Jr. However, the observation of a linear relationship between J+ and Jr does not imply a stoichiometric relationship, i.e., complete coupling. Since coupling may possibly be incomplete, we examine systems of an arbitrary degree of coupling q, regarding stoichiometry as a limiting case. We consider a sodium pump, with J+ and Jr linear functions of the electrochemical potential difference, -X+, and the chemical affinity of the metabolic driving reaction, A. The affinity is well defined even for various complex reaction pathways. Incorporation of a series barrier and a parallel leak does not affect the linearity of the composite observable system. The affinity of some region of the metabolic chain may be maintained constant, either by large pools of reactants or by regulation. If so, this affinity can be evaluated by two independent methods. Sodium transport is conveniently characterized by the open-circuit potential (Δψ)I=0 and the natural limits, level flow (J+)X+=0, and static head X0+ = (X+)J+=0. With high degrees of coupling -X0+/F approaches the electromotive force ENa (Ussing); -X0+/F cannot be identified with ((RT/F) ln f)X+=0, where f is the flux ratio. The efficiency η = -J+X+/JrA is of significance only when appreciable energy is being converted from one form to another. When either J+ or -X+ is small η is low; the significant parameters are then the efficacies εJ+ = J+/JrA and εX+ = -X+/JrA, respectively maximal at level flow and static head. Leak increases both J+ and εJ+ for isotonic saline reabsorption, but diminishes -X0+ and εX. Electrical resistance reflects both passive parameters and metabolism. Various fundamental relations are preserved despite coupling of passive ion and water flows.  相似文献

DNA was examined by RAPD banding for Junipers chinensis, J.c. var. sargentii, J.c. var. tsukusiensis, J. communis, J.c. var. nipponica, J.c. var. saxatilis, J. conferta, J. formosana, J. procumbens, J. rigida, J. taxifolia and J.t. var. lutchuensis. The DNA data readily separated junipers of section Sabina from section Juniperus. J.c. var. tsukusiensis from Taiwan was found to be sufficiently different from J.c. var. tsukusiensis (Yakushima) to warrant the recognition of a new variety: J. chinensis var. taiwanensis R.P. Adams and C-F. Hsieh nov. var. Juniperus formosana from mainland China was found to be different from J. formosana from Taiwan and a new variety is recognized: J. formosana var. mairei (Lemee and Lev.) R.P. Adams and C-F. Hsieh comb. nov. Juniperus communis var. nipponica was found to be very distinct from J. communis and this supports its recognition as a variety. The recognition of J. conferta as a variety of J. rigida [J. rigida var. conferta (Parl.) Patschke] is supported by the data. The data also supports the recognition of J. lutchuensis Koidz. [=J. taxifolia var. lutchuensis (Koldz.) Satake] and J. morrisonicola Hayata [=J. squamata var. morrisonicola (Hayata) H.L. Li and H. Keng] at the specific levels.  相似文献

Azo dyes are recalcitrant and xenobiotic nature makes these compounds a challenging task for continuous biodegradation up to satisfactorily levels in large-scale. In the present report, the biodegradation efficiency of alginate immobilized indigenous Aeromonas sp. MNK1 on Methyl Orange (MO) in a packed bed reactor was explored. The experimental results were used to determine the external mass transfer model. Complete MO degradation and COD removal were observed at 0.20 cm bead size and 120 ml/h flow rate at 300 mg/l of initial dye concentration. The degradation of MO decreased with increasing bead sizes and flow rates, which may be attributed to the decrease in surface of the beads and higher flux of MO, respectively. The experimental rate constants (k ps) for various beads sizes and flow rates were calculated and compared with theoretically obtained rate constants using external film diffusion models. From the experimental data, the external mass transfer effect was correlated with a model J D = K Re ?(1 ? n). The model was tested with K value (5.7) and the Colburn factor correlation model for 0.20, 0.40 and 0.60 bead sizes were J D = 5.7 Re ?0.15, J D = 5.7 Re ?0.36 and J D = 5.7 Re ?0.48, respectively. Based on the results, the Colburn factor correlation models were found to predict the experimental data accurately. The proposed model was constructive to design and direct industrial applications in packed bed reactors within acceptable limits.  相似文献

The evolution of color vision is often studied through the lens of receptor gain relative to an ancestor with fewer spectral classes of photoreceptor. For instance, in Heliconius butterflies, a genus-specific UVRh opsin duplication led to the evolution of UV color discrimination in Heliconius erato females, a rare trait among butterflies. However, color vision evolution is not well understood in the context of loss. In Heliconius melpomene and Heliconius ismenius lineages, the UV2 receptor subtype has been lost, which limits female color vision in shorter wavelengths. Here, we compare the visual systems of butterflies that have either retained or lost the UV2 photoreceptor using intracellular recordings, ATAC-seq, and antibody staining. We identify several ways these butterflies modulate their color vision. In H. melpomene, chromatin reorganization has downregulated an otherwise intact UVRh2 gene, whereas in H. ismenius, pseudogenization has led to the truncation of UVRh2. In species that lack the UV2 receptor, the peak sensitivity of the remaining UV1 photoreceptor cell is shifted to longer wavelengths. Across Heliconius, we identify the widespread use of filtering pigments and co-expression of two opsins in the same photoreceptor cells. Multiple mechanisms of spectral tuning, including the molecular evolution of blue opsins, have led to the divergence of receptor sensitivities between species. The diversity of photoreceptor and ommatidial subtypes between species suggests that Heliconius visual systems are under varying selection pressures for color discrimination. Modulating the wavelengths of peak sensitivities of both the blue- and remaining UV-sensitive photoreceptor cells suggests that Heliconius species may have compensated for UV receptor loss.  相似文献

Butterfly wing color patterns often contain eyespots, which are developmentally determined at the late larval and early pupal stages by organizing activities of focal cells that can later form eyespot foci. In the pupal stage, the focal position of a future eyespot is often marked by a focal spot, one of the pupal cuticle spots, on the pupal surface. Here, we examined the possible relationships of the pupal focal spots with the underneath pupal wing tissues and with the adult wing eyespots using Junonia butterflies. Large pupal focal spots were found in two species with large adult eyespots, J. orithya and J. almana, whereas only small pupal focal spots were found in a species with small adult eyespots, J. hedonia. The size of five pupal focal spots on a single wing was correlated with the size of the corresponding adult eyespots in J. orithya. A pupal focal spot was a three-dimensional bulge of cuticle surface, and the underside of the major pupal focal spot exhibited a hollowed cuticle in a pupal case. Cross sections of a pupal wing revealed that the cuticle layer shows a curvature at a focal spot, and a positional correlation was observed between the cuticle layer thickness and its corresponding cell layer thickness. Adult major eyespots of J. orithya and J. almana exhibited surface elevations and depressions that approximately correspond to the coloration within an eyespot. Our results suggest that a pupal focal spot is produced by the organizing activity of focal cells underneath the focal spot. Probably because the focal cell layer immediately underneath a focal spot is thicker than that of its surrounding areas, eyespots of adult butterfly wings are three-dimensionally constructed. The color-height relationship in adult eyespots might have an implication in the developmental signaling for determining the eyespot color patterns.  相似文献

Water flux through excised roots (Jv) is determined by root hydraulic conductance (Lp) and the ion flux to the xylem (Ji) that generates an osmotic gradient to drive water movement. These properties of roots are strongly temperature dependent. Abscisic acid (ABA) can influence Jv by altering Lp, Ji, or both. The effects of root temperature on responses to ABA were determined in two species differing in their temperature tolerances. In excised barley (Hordeum vulgare L.) roots, Jv was maximum at 25°C; 10 micromolar ABA enhanced Jv, primarily by increasing Lp, at all temperatures tested (15-40°C). In sorghum (Sorghum bicolor L.) roots, Jv peaked at 35°C; ABA reduced this optimum temperature for Jv to 25°C by increasing Lp at low temperatures and severely inhibiting Ji (dominated by fluxes of K+ and NO3) at warm temperatures. The inhibition of K+ flux by ABA at high temperature was mostly independent of external K+ availability, implying an effect of ABA on ion release into the xylem. In sorghum, ABA enhanced water flux through roots at nonchilling low temperatures but at the expense of tolerance of warm temperatures. These effects imply that ABA may shift the thermal tolerance range of roots of this heat-tolerant species toward cooler temperatures.  相似文献

Apoptosis is essential for complex multicellular organisms and its failure is associated with genome instability and cancer. Interactions between apoptosis and genome-maintenance mechanisms have been extensively documented and include transactivation-independent and -dependent functions, in which the tumor-suppressor protein p53 works as a ‘molecular node’ in the DNA-damage response. Although apoptosis and genome stability have been identified as ancient pathways in eukaryote phylogeny, the biological evolution underlying the emergence of an integrated system remains largely unknown. Here, using computational methods, we reconstruct the evolutionary scenario that linked apoptosis with genome stability pathways in a functional human gene/protein association network. We found that the entanglement of DNA repair, chromosome stability and apoptosis gene networks appears with the caspase gene family and the antiapoptotic gene BCL2. Also, several critical nodes that entangle apoptosis and genome stability are cancer genes (e.g. ATM, BRCA1, BRCA2, MLH1, MSH2, MSH6 and TP53), although their orthologs have arisen in different points of evolution. Our results demonstrate how genome stability and apoptosis were co-opted during evolution recruiting genes that merge both systems. We also provide several examples to exploit this evolutionary platform, where we have judiciously extended information on gene essentiality inferred from model organisms to human.  相似文献

Methylmercury(II) and mercury(II) complexes of imidazole (1), 1-methylimidazole (2), and the 1,3-dimethylimidazolium ion (3) have been prepared in aqueous or ethanolic solution. Elemental analysis and 1H nmr spectroscopy have been used to characterize the complexes. The MeHg (Me = methyl) binding sites have been identified as N1, N3 (1), N3, C2 (2), and C2 (3). Reaction with HgO leads to the formation of Hg-bridged complexes of the type Im-Hg-Im, (Im = imidazole), where bonding occurs through N1 (1) and C2 (3); the latter is also formed as a result of symmetrization of the C2-bound MeHg complex. The formation of the C2-bound (carbene) complexes is discussed in terms of the increased acidity of the C2 proton resulting from coordination of an electrophilic species at N3. Based on electrostatic considerations, there appears to be a “minimum degree of activation” required before C2 bonding can occur, which explains the lack of this coordination mode in 1. 199Hg-1H spin-spin coupling (4J) is observed for C-bound mercury, but not for N-bound mercury, which is interpreted in terms of a decreased ligand exchange rate in the former case, due to the greater stability of the Hg-C bond. 2J coupling constants measured in (CD3)2SO for a number of MeHg complexes of heterocyclic ligands (including the imidazoles of the present study) correlate well with the ligand pKa (25°C, aqueous solution), according to 2J = ?3.88 pKa + 248.5. Results in the present work are discussed in relation to our previous work with nucleosides. The significance of the results to biological systems is considered.  相似文献

The cut ends of excised Zea mays roots were sealed to a pressure transducer and their root pressures recorded. These rose approximately hyperbolically to a maximum value of 4.21 ± 0.34 bar after 30 to 40 minutes. Xylem exudate could not be collected at this pressure since the flow rate was zero. Samples of exudate were collected at lower applied pressures (ΔP), however, and Δπ, the osmotic pressure difference between them and the solution bathing the root, was measured by freezing point depression. A plot of ΔP/Δπ against Jv/Δπ, where Jv is the volume flux, proved to be a straight line whose intercept, equal to σ, the reflection coefficient, was 0.853 ± 0.016. The maximum xylem concentrations of various chemical species were found by a similar extrapolative method and compared with those in the cell sap. This indicated that (a) Ca2+, Mg2+, NO32−, SO42−, and most amino acids move from the cells to the xylem down an electrochemical potential gradient; (b) relative to these ions H+, NH4+, glutamine and asparagine are actively transported into the xylem; and (c) H2PO4, and K+ are actively retained in the symplasm.  相似文献
