Crude extracts from cotyledons of 2-day-old seedlings were separated by electrophoresis on polyacrylamide gels and stained for acid phosphatase. At least 22 bands were recognised, distributed over seven zones. Analysis of zone 1 allowed estimation of sib frequencies in 12 out of 19 hybrid cultivars. By applying either seedling zone 1 or acp-1 seed acid phosphatase analysis, sib estimates could be made in 15 of these hybrids. It is suggested that this technique offers a practical and rapid means of estimating sib frequencies in commercial seed stocks.  相似文献

Summary. Milk samples from 189 Merinoland Sheep, 145 Black Faced Mutton Sheep, 89 East Friesian Milk Sheep, 36 Rhön, 36 Pleven, 23 Tsigaja, 25 Black Razka and 86 Hungarian Merino x Pleven (F1) sheep were analysed by polyacrylamide gel electrophoresis under acid conditions and isoelectric focusing in ultrathin layer polyacrylamide gels with carrier ampholytes. Six different β-lactoglobulin (β-Lg) phenotypes (A, AB, B, AC, BC and C) were observed by both methods. The occurrence of three codominant alleles (β-LgA, β-LgB, β-Lgc) at an autosomal locus (β-Lg) was supported by family and population data on genetic equilibrium. Genotypes at the gliadin loci Gli-A1, Gli-B1, Gli-D1 and the high-molecular-weight glutenin subunit loci Glu-A1, Glu-B1, Glu-D1 were identified in 77 winter common wheat cultivars developed in the Central Forest Steppe of Ukraine in different periods of time. The highest level of variation was observed at the Gli-A1 locus. Predominant alleles (one or two per locus) were revealed. The comparison of allele frequencies in groups of cultivars developed in different periods of time (before 1996 and in 1996–2007) has demonstrated appearance of new alleles and change of frequencies of existing alleles at the storage protein loci. The high frequency of cultivars with the wheat-rye 1BL/1RS translocation was detected (about 40%). The wheat rye 1AL/1RS translocation was identified in six cultivars developed in the last decade. Four gliadin alleles, Gli-A1w (a marker for the 1AL/1RS translocation), Gli-A1x, Gli-A1y and Gli-B1x, were proposed for cataloging. The article is published in the original.  相似文献

Linkage analyses of simulated quantitative trait data were performed using the Haseman-Elston (H-E) sib pair regression test to investigate the effects of inaccurate allele frequency estimates on the type I error rates of this test. Computer simulations generating a quantitative trait in nuclear families were performed using GASP [1]. Assuming no linkage, several data sets were simulated; they differed in marker allele numbers and frequencies, number of sib pairs and number of sibships. Each set of simulated data was analyzed using (1) all parental marker data, (2) half of the parental marker data, and (3) no parental marker data, using both correct and incorrect allele frequencies in the latter 2 cases. The H-E sib pair linkage method was found to be robust to misspecification of marker allele frequencies regardless of the number of alleles.  相似文献

A field experiment was conducted to observe the effect of TS amendments on soil enzymes and phytoremediation potential of two economically important cultivars of geranium. Different doses of TS were applied in soil to examine threshold limit of HMs where geranium cultivars can be grown successfully in contaminated sites. Treatment variation significantly affected pH, EC, OC, N, P, K and HM content in soil after 50 days of incubation. After harvest, both cultivars were examined to assess the impact of various treatments on their fresh herb, dry matter, essential oil yield and HM accumulation. C/G ratio close to 1 was observed at 50 tha?1 sludge treatment in both cultivars. Urease and β-glucosidase activities in soil were maximum at 50 tha?1 whereas dehydrogenase and phosphatase activities were maximum at 100 tha?1 in both cultivars. β-glucosidase, acid and alkaline phosphatase, urease and dehydrogenase activities were relatively high after 85 days over 45 days in both cultivars. Maximum metal uptake was found in roots of cv. Bourbon followed by leaves. Geranium was observed to be a good candidate for phytoremediation as it mitigates metal toxicity by root absorption and cv. Bourbon is better candidate for the same.  相似文献

Peroxidase, alkaline phosphatase, acid phosphatase, β-glucuronidase, α-naphthyl acetate esterase (ANAE), α-naphthyl butyrate esterase, naphthol AS-D chloroacetate esterase, acetyl-L-tyrosine-α-naphthyl esterase (ATNE), tosyl-L-lysine-α-naphthyl esterase (TLNE) and periodic acid-Schiff (PAS) were studied in 17 species of elasmobranchs in which granulocytes had previously been identified at the ultrastructural level.
Eosinophils, eosinophilic and neutrophilic granulocytes contained variable acid phosphatase, esterases and PAS, but they were strongest in neutrophilic granulocytes; particularly ANAE. Esterases were released into surrounding plasma and therefore probably function as ectoenzymes. In eosinophils and some neutrophilic granulocytes there were indications of weak peroxidase, but this could not be conclusively demonstrated. Alkaline phosphatase was diffuse between granules in some eosinophils of Pavoraja , and (β-glucuronidase was diffuse in neutrophilic granulocytes of Etmopterus baxteri , otherwise granulocytes lacked these enzymes. Neutrophilic granulocytes stained moderately to strongly for ATNE and weakly and inconsistently for TLNE in Squalus acanthias and Dalatias licha . with a similar reaction in granular lymphocytoid and thrombocytoid cells of Galeorhinus ausiralis and Raja nasuta . The enzyme composition of these granulocytes is discussed.  相似文献

Genetic diversity in modern sunflower (Helianthus annuus L.) cultivars (elite oilseed inbred lines) has been shaped by domestication and breeding bottlenecks and wild and exotic allele introgressionthe former narrowing and the latter broadening genetic diversity. To assess single nucleotide polymorphism (SNP) frequencies, nucleotide diversity, and linkage disequilibrium (LD) in modern cultivars, alleles were resequenced from 81 genic loci distributed throughout the sunflower genome. DNA polymorphisms were abundant; 1078 SNPs (1/45.7 bp) and 178 insertions-deletions (INDELs) (1/277.0 bp) were identified in 49.4 kbp of DNA/genotype. SNPs were twofold more frequent in noncoding (1/32.1 bp) than coding (1/62.8 bp) sequences. Nucleotide diversity was only slightly lower in inbred lines (θ = 0.0094) than wild populations (θ = 0.0128). Mean haplotype diversity was 0.74. When extraploted across the genome (~3500 Mbp), sunflower was predicted to harbor at least 76.4 million common SNPs among modern cultivar alleles. LD decayed more slowly in inbred lines than wild populations (mean LD declined to 0.32 by 5.5 kbp in the former, the maximum physical distance surveyed), a difference attributed to domestication and breeding bottlenecks. SNP frequencies and LD decay are sufficient in modern sunflower cultivars for very high-density genetic mapping and high-resolution association mapping.  相似文献

Y. ODA AND K. TONOMURA. 1996. β-Mannanase and β-mannosidase were purified from the culture fluid of the yeast Trichosporon cutaneum JCM 2947 (= T. beigelii CBS 5790). The molecular weights of the two enzymes were estimated to be 49 900 and 114000 by SDS-PAGE and 4500 and 193 000 by gel filtration, respectively. β-Mannanase contained 43% molecular weight as carbohydrate. The K m and V max values of β-mannanase for konjac glucomannan were 2.7 (mg ml-1) and 10.6 (U mg protein-1), and those of β-mannosidase for p -nitrophenyl β-D-mannopyranoside were 0.25 (mmol l-1) and 91.7 (U mg protein-1). Maximal activities were observed between pH 4.0 and 6.5 at 50°C for β-mannanase and around 6.5 at 40°C for β-mannosidase.  相似文献

The allelic diversity of high-moleculat-weght glutenin subunits (H WIGS) in Russian and Ukrainian bread wheat cultivars was analyzed. The diversity of spring wheat cultivars for alleles of the Glu-1 loci is characterized by medium values of the polymorphism index (polymorphism information content, PlC), and in winter wheats it varies from high at the Glu-A1 locus to low at the Glu-D1 locus. The spring and winter cultivars differ significantly in the frequencies of alleles of the glutenin loci. The combination of the Glu-A1b, Glu-B1c, and Glu-D1a alleles prevails among the spring cultivars, and the combination of the Glu-A1a, Glu-B1c, and Glu-D1d alleles prevails among the winter cultivars. The distribution of the Glu-1 alleles significantly depends on the moisture and heat supply in the region of origin of the cultivars. Drought resistance is associated with the Glu-D1a allele in the spring wheat and with the Glu-B1b allele in the winter wheat. The sources of the Glu-1 alleles were identified in the spring and wheat cultivars. The analysis of independence of the distribution of the spring and winter cultivars by the market classes and by the alleles of the HMWGS loci showed a highly significant association of the alleles of three Glu-1 loci with the market classes in foreign cultivars and independence or a weak association in the Russian and Ukrainian cultivars. This seems to be due to the absence of a statistically substantiated system of classification of the domestic cultivars on the basis of their quality.  相似文献

A total of 20990 Finnish Ayrshire cows were phenotyped for the major milk proteins by isoelectric focusing in polyacrylamide gels. The predominant alleles in the Finnish Ayrshire were αS1-casein B (0.999), αS2-casein A (0.991), β-casein A1 (0.509) and α2 (0.490), α22-casein A (0.612) and β-lactoglobulin B (0.716). The K-casein E allele (0.307) was also rather common in the Finnish Ayrshire. A new K-casein variant (K-casein F) was demonstrated in two Finnish Ayrshire cows, a dam and a daughter.  相似文献

Abstract: Studies of processing of the Alzheimer β-amyloid precursor protein (βAPP) have been performed to date mostly in continuous cell lines and indicate the existence of two principal metabolic pathways: the "β-secretase" pathway, which generates β-amyloid (Aβ1–40/42; ∼4 kDa), and the "α-secretase" pathway, which generates a smaller fragment, the "p3" peptide (Aβ17–40/42; ∼3 kDa). To determine whether similar processing events underlie βAPP metabolism in neurons, media were examined following conditioning by primary neuronal cultures derived from embryonic day 17 rats. Immunoprecipitates of conditioned media derived from [35S]methionine pulse-labeled primary neuronal cultures contained 4- and 3-kDa Aβ-related species. Radiosequencing analysis revealed that the 4-kDa band corresponded to conventional Aβ beginning at position Aβ(Asp1), whereas both radio-sequencing and immunoprecipitation-mass spectrometry analyses indicated that the 3-kDa species in these conditioned media began with Aβ(Glu11) at the N terminus, rather than Aβ(Leu17) as does the conventional p3 peptide. Either activation of protein kinase C or inhibition of protein phosphatase 1/2A increased soluble βAPPα release and decreased generation of both the 4-kDa Aβ and the 3-kDa N-truncated Aβ. Unlike results obtained with continuously cultured cells, protein phosphatase 1/2A inhibitors were more potent at reducing Aβ secretion by neurons than were protein kinase C activators. These data indicate that rodent neurons generate abundant Aβ variant peptides and emphasize the role of protein phosphatases in modulating neuronal Aβ generation.  相似文献

Allelic diversity of the gliadin-coding loci Gli-1 and Gli-2 was compared with the genealogical profiles of common wheat cultivars developed in Saratov. Allele tracking through their pedigrees and hierarchic cluster analysis associated 31 Gli alleles with groups of original ancestors. The cultivars Poltavka (12 alleles of six loci) and Selivanovskii Rusak (six alleles of six loci) were identified as sources of the majority of alleles. The results of the cluster analysis fully coincided with the results of allele tracking for alleles occurring at high frequencies. For rare alleles, the resolution of the cluster analysis was somewhat lower and depended on the similarity/distance measure. Thus, it proved possible to indirectly identify the donors of gene alleles by multidimensional statistics even when data on alleles identified in ancestors are unavailable. This approach to the analysis of inheritance has two limitations: detailed pedigree data should be known, and relatively high frequencies (no less than 15–20%) should be observed for the alleles in a sample under study. Cluster analysis was used to study the association of gliadin alleles with commercial quality classes. The most important gliadin-coding alleles, which mark strong cultivars, were identified. In the Saratov cultivars, such alleles include Gli-A1f, GliB1e, Gli-D1a, Gli-A2q, Gli-B2s, and Gli-D2e, which were inherited from the landrace Poltavka, and Gli-A1i, Gli-A2s, and Gli-B2q, which were inherited from the landrace Selivanovskii Rusak.  相似文献

Hepatic lysosomes were exposed in vitro to microwave radiation (2450 MHz) either prior to or simultaneously with treatment with retinol (vitamin A), and the release of the lysosomal enzymes, β-glucuronidase, acid phosphatase, and cathepsin D, determined. A 60-min microwave exposure (10 or 100 mW/g) of retinol-treated lysosomes had no effect on the amount of release of β-glucuroni-dase, cathepsin D, or acid phosphatase. In addition, 10 and 100 mW/g irradiation of lysosome fractions for 40 min prior to a 20-min retinol and microwave treatment, had no influence on the release of these enzymes. Finally, the effect of microwave radiation on the loss of latency of acid phosphatase and β-glucuronidase from retinol-treated lysosomes was determined. Microwave radiation had no influence on the rate of appearance of these enzymes in the suspending medium. The results indicate that microwave radiation had no effect on the retinol-induced lysosomal enzyme release.  相似文献

Fungi are elements of the ontocenosis of the oral cavity and causal factors of inflammatory lesions in its mucous membrane. The objective of the study was to find differences in the activity of hydrolytic enzymes of Candida albicans isolated from patients with diseases of the periodontium and mucous membrane of the oral cavity. Of 235 patients examined, 31 were diagnosed with gingivitis, 38 with glossitis, 28 with leucoplakia, 37 with adult periodontitis, 25 with juvenile periodontitis, 36 with stomatitis prothetica and 40 with stomatitis atrophica. In 196 patients (83.4 ± 2.4%), fungi belonging to Candida species were detected. In the evaluation of Candida albicans strains (146) properties, bioMerieux API ZYM tests containing substrates for the detection of 19 hydrolases were used. All the investigated strains were characterized by the activity of 14 enzymes, i.e. phosphatase alcaline, esterase (C4), esterase lipase (C8), leucine and valine arylamidase, phosphatase acid, naphthol-AS-BI-phosphohydrolase, α galactosidase, β galactosidase, α glucosidase, β glucosidase, N-acetyl-β-glucosaminidase, α mannosidase and α fucosidase. Strains isolated from the oral cavity of patients with diseases of periodontium and mucous membrane are characterised by the highest phosphatase acid activity. The greatest enzymatic activity is characteristic of Candida albicans isolated from patients with stomatitis atrophica or stomatitis prothetica, and the lowest in strains from gingivitis or juvenile periodontitis cases. Differences in the activity of hydrolases are statistically significant (p<0.01) for: esterase (C4), leucine and valine arylamidase, phosphatase acid, naphthol-AS-BI-phosphohydrolase, β glucosidase, N-acetyl-β-glucosaminidase, of fungi isolated from patients with particular clinical diagnoses. Bacteroides gingivalis was grown in continuous culture in the presence of chlorhexidine. Maximum specific growth rates and biomass levels initially increased but then decreased as the chlorhexidine level increased from 0 to 30 μg/ml. Total inhibition of growth occurred when the chlorhexidine concentration reached 60 μg/ml. The steady-state levels of cell-bound, extracellular vesicle and extracellular soluble enzymes, trypsin-like protease, alkaline phosphatase and N -acetyl-β-glucosaminidase were measured. With increasing sub-lethal concentrations of chlorhexidine, levels of alkaline phosphatase increased noticeably in all three fractions of culture, whilst cell-bound and extracellular vesicle levels of N -acetyl-β-glucosaminidase remained approximately constant. Extracellular soluble levels of alkaline phosphatase and N -acetyl-β-glucosaminidase increased with increasing levels of chlorhexidine. The levels of trypsin-like protease decreased significantly in all fractions of the culture when cells were grown in the presence of chlorhexidine. Thus, chlorhexidine has a differential effect on the production of B. gingivalis hydrolytic enzymes.  相似文献

Studies on Persea americana have been addressed in different ways with biochemical and molecular techniques. Microsatellites are able to detect multiple alleles for particular loci and are therefore a useful tool to study genealogical relationships, population structures and genetic mapping. Ninety-six samples from 49 cultivars including three horticultural groups and hybrids were collected from the avocado germplasm bank at INIA-CENIAP (Venezuela). A modified DNA extraction protocol was performed. Forty microsatellites were selected from previous references, PCR amplifications were performed, and presence/absence, size, and number of alleles were evaluated on polyacrylamide gels. Attributes for polymorphic alleles were analyzed with POPGENE, and genetic diversity was calculated by effective sample size, number of alleles per locus (Na), effective number of alleles (Ne), Shannon information index (In), observed heterozygosis (H), expected heterozygosity (He), Wright’s fixation index (Fis), and allele frequencies. Only 14 primers were amplified, and AVT106 primer resulted monomorphic. Unique genotypes for each sample were obtained. Nine loci showed allele patterns that can be useful for taxonomic identification of cultivars or varieties. Comparing values of Fis with Ho and He, we found a direct relationship where low heterozygosis alleles identified in the population may affect the expected level. Allele frequencies ranged from 0.5632 to 0.0105. For all loci, at least one rare allele was observed. With the available information from genetic analysis, an identifying system was implemented for selected avocado cultivars maintained at the INIA-CENIAP Venezuelan germplasm bank on the basis of molecular data.  相似文献

The polysaccharidic effect of a purified 1,3- β -glucanase, a purified β -glucosidase, and of partially purified endo-1,3- β -glucanase from autolysed Penicillium oxalicum cultures on cell wall isolate fractions from the same fungus were studied.
Fractionation of 5-day-old cell wall gave rise to a series of fractions that were identified using infrared spectrophotometry. The fractions used were: F1, an α -glucan; F3, a β -glucan; F4, a chitin-glucan; and F4b, a β -glucan. The fractions were incubated with each of the enzymes and with a mixture of equal parts of the three enzymes and the products of the enzymatic hydrolysis were analyzed after 96 h incubation.
The enzymes were found to degrade fraction F4b ( β -glucan); the greatest degree of hydrolysis was reached when the three enzymes were used together, suggesting the need for synergic action by these enzymes in the cell wall degradation process.  相似文献

Abstract: The accumulations by axoplasmic transport of selected enzyme activities proximal and distal to a ligature placed on the sciatic nerve were monitored in rats exposed in utero to maternal antibodies to nerve growth factor (NGF) and in control rats. Littermates of the animals exposed to anti-NGF were shown elsewhere to have had a 70% reduction in the number of sensory neurons in dorsal root ganglia and a 90% reduction in number of neurons in superior cervical (sympathetic) ganglion. The accumulation of F--sensitive acid phosphatase activity was depressed 75% both proximal and distal to the tie. Accumulation of F--resistant acid phosphatase activity was depressed nearly 50% proximal to the tie. Distal accumulation of this activity did not occur in either group of rats. Accumulation of acetylcholinesterase activity was not affected. Proximal accumulation of glutamic dehydrogenase activity was depressed 30%. Distal accumulation of the activities of β-glucuronidase and hexokinase was depressed 50%. In the lumbar dorsal root ganglia, dry weight was reduced 40%, and the activities of peroxide-sensitive, F--resistant acid phosphatase and of the mitochondrial enzymes hexokinase, glutamic dehydrogenase, glutamic-oxalacetic transaminase, and NAD-dependent isocitric dehydrogenase were all reduced a little more, 45–50% per ganglion. However, the activities of the lysosomal enzymes, F--sensitive acid phosphatase and β-glucuronidase, of the peroxide-resistant, F--resistant acid phosphatase, and of the mitochondrial enzyme glutaminase were all reduced about 60% per ganglion. The results of these measurements were interpreted to suggest that much, and perhaps all, of the F--sensitive acid phosphatase activity in motion in peripheral nerve in rat is confined to sensory axons.  相似文献

Identifying annual ryegrass contamination in perennial ryegrass seed lots has been of major interest in seed-testing laboratories and for seed regulatory agencies in the USA for many years. This study was conducted to characterize a superoxide dismutase locus (Sod-1) and determine its potential to distinguish cultivated ryegrass species. The inheritance of Sod-1 was evaluated in a three-generation annual 2 perennial ryegrass mapping population and segregation fitted an expected 1:2:1 ratio for a single locus with two alleles. The molecular form of the Sod-1 locus was determined by H2O2 and KCN inhibitor assays which indicated that the Sod-1, and a second independently segregating Sod-2, locus were both Cu/ZnSod enzymes. The common alleles at the Sod-1 locus were scored in 13 annual and 24 perennial ryegrass cultivars to determine the potential of using this locus for species separation. The Sod-1b allele was homozygous in 98% of perennial ryegrass individuals from 24 cultivars, but those not 100% homozygous for Sod-1b were seed lots with unknown contamination from annual ryegrass. These results indicate that the Sod-1b allele in the homozygous condition is a good indicator of perenniality. All eight annual ryegrass cultivars originating in Europe or Asia had a low frequency of Sod-1b homozygous individuals or none at all. The five cultivars originating in the Western Hemisphere, however, had genotype frequencies for homozygous Sod-1b of up to 56%. The potential of the Sod-1 locus to serve as a test to separate the two growth forms depends on the source of the annual-type contamination.  相似文献
