Cloning cattle     
Over the past six years, hundreds of apparently normal calves have been cloned worldwide from bovine somatic donor cells. However, these surviving animals represent less than 5% of all cloned embryos transferred into recipient cows. Most of the remaining 95% die at various stages of development from a predictable pattern of placental and fetal abnormalities, collectively referred to as the "cloning-syndrome." The low efficiency seriously limits commercial applicability and ethical acceptance of somatic cloning and enforces the development of improved cloning methods. In this paper, we describe our current standard operating procedure (SOP) for cattle cloning using zona-free nuclear transfer. Following this SOP, the output of viable and healthy calves at weaning is about 9% of embryos transferred. Better standardization of cloning protocols across and within research groups is needed to separate technical from biological factors underlying low cloning efficiency.  相似文献

Genetics has the potential to provide a novel layer of information pertaining to the origins and relationships of domestic cattle. While it is important not to overstate the power of archeological inference from genetic data, some previously widespread conjectures are inevitably contradicted with the addition of new information. Conjectures regarding domesticated cattle that fall into this category include a single domestication event with the development of Bos indicus breeds from earlier Bos taurus domesticates; the domestication of a third type of cattle in Africa having an intermediate morphology between the two taxa; and the special status of the Jersey breed as a European type with some exotic influences. In reality, a wideranging survey of the genetic variation of modern cattle reveals that they all derive from either zebu or taurine progenitors or are hybrids of the two. The quantitative divergence between Bos indicus and Bos taurus strongly supports a predomestic separation; that between African and European taurines also suggests genetic input from native aurochsen populations on each continent. Patterns of genetic variants assayed from paternally, maternally, and biparentally inherited genetic systems reveal that extensive hybridization of the two subspecies is part of the ancestry of Northern Indian, peripheral European, and almost all African cattle breeds. In Africa, which is the most extensive hybrid zone, the sexual asymmetry of the process of zebu introgression into native taurine breeds is strikingly evident. © 1998 Wiley-Liss, Inc.  相似文献

A case control study was conducted to evaluate the significance of Neospora caninum infections in cattle in England and Wales. The prevalence of N. caninum in normally calving cattle (the control group; n = 418) and aborting cattle (n = 633) was estimated using a commercial antibody-detection ELISA. Prevalence estimates for bovine virus diarrhoea virus, infectious bovine rhinotracheitis virus and Leptospira hardjo were also obtained by serology. The prevalence of N. caninum was significantly higher (P < 0.0001) in the aborting group (18%; 95% confidence interval: 15%, 21%) than in the control group (6%; 95% confidence interval: 4%, 8%); the latter is the first estimate, to date, of the national seroprevalence of N. caninum in dairy cattle in England and Wales. Prevalence estimates for bovine virus diarrhoea virus, infectious bovine rhinotracheitis virus and L. hardjo were not found to be higher in the aborting cattle than in the control group. With N. caninum, a strong association between seropositivity and abortion was found, with seropositive cows being 3.5-times more likely to abort than seronegative cows (odds ratio = 3.49; 95% confidence interval: 2.16, 5.69). Furthermore, 12.5% of abortions in dairy cattle in England and Wales may be attributable to N. caninum, as indicated by estimation of the population aetiological fraction.  相似文献

The epidemiology of clinical cases of campylobacter in temperate climates shows a striking seasonality. In the search for a seasonal environmental reservoir changes in the carriage rate and population size of campylobacters in bovine hosts with time have been measured. Most probable number (MPN) methodology was used to enumerate thermophilic campylobacters in samples taken from the small intestines of beef cattle at slaughter and the fresh faeces of four dairy herds and new-born calves. Statistical analyses revealed significant evidence for seasonal periodicity in the data from dairy herds ( P = 0·044). Not only was there a departure from constancy within a 12-month interval but these data revealed a true seasonality, that is, the same periodicity in numbers from one year to the next. Each herd had two peaks per year, in approximately spring and autumn. Peaks coincided in herds on neighbouring farms but those on farms in the north preceded those on farms in the south by 2 and 1 months, respectively ( P = 0·0057). Intestinal carriage by beef cattle at slaughter was 89·4% ( n = 360) with an average MPN campylobacters per gram fresh weight (MPN gfw−1) of 6·1 × 102. Average MPN gfw−1 in faeces from the dairy herds and calves were 69·9 ( S.D. 3) and 3·3 × 104 ( S.D. 1·7 × 102). There was no evidence of seasonal periodicity in the size of the campylobacter population in beef cattle at slaughter. Calves were campylobacter free at birth but became colonized within a few days.  相似文献

Since their domestication in the Neolithic, cattle have belonged to our cultural heritage. The reconstruction of their history is an active field of research 1 that contributes to our understanding of human history. Archeological data are now supplemented by analyses of modern and ancient samples of cattle with DNA markers of maternal, paternal, or autosomal inheritance. The most recent genetic data suggest that maternal lineages of taurine cattle originated in the Fertile Crescent with a possible contribution of South‐European wild cattle populations, while zebu cattle originate from the Indus Valley. Subsequently, cattle accompanied human migrations, which led to the dispersal of domestic cattle of taurine, indicine, or mixed origin over Asia, Africa, Europe, and the New World. This has resulted in their adaptation to different environments and considerable variation in appearance and performance. More recently, rational management of breeding led to international movements of sires, which again changed the global patterns of genetic diversity.  相似文献

Although cattle are basically not seasonal breeders, several physiological changes occur in response to two major climatic variables of season: ambient temperature and photoperiod. Ambient temperatures above 27°C lengthen the estrous cycle, decrease duration and intensity of estrus, decrease fertility and increase embryonic mortality. Some of these effects may be mediated via decreased secretion of luteinizing hormone and increased secretion of progesterone. Heat stress also suppresses growth rate, milk production and feed intake. In comparison with cattle exposed to less than 12 h of light per day, 14 to 16 h of light has relatively little effect on secretion of gonadotropins and fertility; however, growth rates, feed intake and milk yield are stimulated. Of all hormones measured in cattle, prolactin is the most responsive to changes in seasons, ambient temperature and photoperiod. Prolactin is greatest in summer when ambient temperatures are highest and photoperiods are longest.  相似文献

Homozygous transferrins of adult cattle are made up of two strong pairs and one weak pair of protein bands on starch gel electrophoresis. Foetal transferrins have only the slower band of each pair with the fastest band of the three being much stronger than in the adult type. Before term the second band of each pair begins to develop and at the same time the fastest pair becomes weaker – attaining the adult type by term or soon after. The ai protein, which is present in early foetal life and almost disappears by 250 days of embryonic development, shows individual variation. Its relationship to fetuin is discussed.  相似文献

Owing to the complex interactions among climate, plants, cattle grazing, and land management practices, the impacts of climate change on cattle have been hard to predict. Predicting future grassland ecosystem functioning relies on understanding how changes in climate alter the quantity of forage produced, but also forage quality. Plant protein, which is a function of plant nitrogen concentrations, and digestible energy limit the performance of herbivores when in short supply; moreover, deficiencies can be expensive to mitigate. To better understand how changes in temperature and precipitation would affect forage protein and energy availability, we analyzed over 21 000 measurements of cattle fecal chemistry acquired over 14 years in the continental US. Our analysis of patterns in forage quality among ecologically defined regions revealed that increasing temperature and declining precipitation decreased dietary crude protein and digestible organic matter for regions with continental climates. Within regions, quality also declined with increased temperature; however, the effects of precipitation were mixed. Any future increases in precipitation would be unlikely to compensate for the declines in forage quality that accompany projected temperature increases. As a result, cattle are likely to experience greater nutritional stress in the future. If these geographic patterns hold as a proxy for future climates, agriculture will require increased supplemental feeds or the consequence will be a decrease in livestock growth.  相似文献

Background

The control of bovine tuberculosis (bTB) remains a priority on the public health agenda in Great Britain, after launching in 1998 the Randomised Badger Culling Trial (RBCT) to evaluate the effectiveness of badger (Meles meles) culling as a control strategy. Our study complements previous analyses of the RBCT data (focusing on treatment effects) by presenting analyses of herd-level risks factors associated with the probability of a confirmed bTB breakdown in herds within each treatment: repeated widespread proactive culling, localized reactive culling and no culling (survey-only).

Methodology/Principal Findings

New cases of bTB breakdowns were monitored inside the RBCT areas from the end of the first proactive badger cull to one year after the last proactive cull. The risk of a herd bTB breakdown was modeled using logistic regression and proportional hazard models adjusting for local farm-level risk factors. Inside survey-only and reactive areas, increased numbers of active badger setts and cattle herds within 1500 m of a farm were associated with an increased bTB risk. Inside proactive areas, the number of M. bovis positive badgers initially culled within 1500 m of a farm was the strongest predictor of the risk of a confirmed bTB breakdown.

Conclusions/Significance

The use of herd-based models provide insights into how local cattle and badger populations affect the bTB breakdown risks of individual cattle herds in the absence of and in the presence of badger culling. These measures of local bTB risks could be integrated into a risk-based herd testing programme to improve the targeting of interventions aimed at reducing the risks of bTB transmission.  相似文献

In order to facilitate the on-going cattle conservation and improvement programs in Korea, we examined genetic relationships among East Asian cattle, focusing on Korean native cattle, using complete mitochondrial DNA control region sequences. In total, 67 haplotypes were discriminated by 97 variable sites in East Asian cattle. Of the variable sites, 35 represented singleton variable sites and 62 were parsimony informative sites. For Korean cattle, 30 haplotypes were discriminated by 40 variable sites. The variable sites identified in this study correspond to transition or transversion point mutations. Inter-populations genetic distance varied from 0.004 to 0.052 for East Asian cattle populations. The genetic divergence observed between Korean Brown and Jeju Black (0.004) was among the lowest. The Mongolian cattle were slightly divergent from other East Asian cattle populations studied. The network analysis uncovered that Jeju Black and Yunbian samples represented the possible ancestral Haplotype within T3 Bos taurus main lineage. Thus, we hypothesized that the Jeju and Yunbian cattle may represent one of the original native North East Asian cattle in the region.  相似文献
