Two novel homoleptic ruthenium (II) complexes containing asymmetric tridentate ligands, 2-(benzimidazole-2-yl)-1,10-phenanthroline (PHBI) and 2-(naphtho[2,3-d]imidazole-2-yl)-1,10-phenanthroline (PHNI) have been synthesized and characterized by elemental analysis, mass spectra, (1)H NMR, and electronic spectroscopy. The DNA-binding properties of the complexes have been investigated by spectroscopic methods and viscosity measurements. The results indicate that the two complexes interact with DNA via electrostatic interaction, and the mechanisms of DNA binding with the complexes have also been discussed.  相似文献

Two copper(II) complexes, 1 and 2 with L1 and L2 [L1 = 2-hydroxybenzyl(2-(pyridin-2-yl)ethylamine); L2 = 2-hydroxybenzyl(2-(pyridin-2-yl)methylamine)] ligands, respectively, have been synthesized and characterized. The interaction of both the complexes with DNA has been studied to explore their potential biological activity. The DNA binding properties of the complexes with calf thymus (CT) DNA were studied by spectroscopic titration. The complexes show binding affinity to CT DNA with binding constant (Kb) values in the order of 105 M−1. Thermal denaturation and circular dichroism studies suggest groove binding of the complexes to CT DNA. Complexes also exhibit strong DNA cleavage activity in presence of reducing agents like 3-mercaptopropionic acid and β-mercaptoethanol. Mechanistic studies reveal the involvement of reactive hydroxyl radicals for their DNA cleavage activity.  相似文献

Four ruthenium(II) complexes with the formula [Ru(η5-C5H5)(PP)L][CF3SO3], being (PP = two triphenylphosphine molecules), L = 1-benzylimidazole, ; (PP = two triphenylphosphine molecules), L = 2,2′bipyridine, ; (PP = two triphenylphosphine molecules), L = 4-Methylpyridine, ; (PP = 1,2-bis(diphenylphosphine)ethane), L = 4-Methylpyridine, , were prepared, in view to evaluate their potentialities as antitumor agents. The compounds were completely characterized by NMR spectroscopy and their crystal and molecular structures were determined by X-ray diffraction. Electrochemical studies were carried out giving for all the compounds quasi-reversible processes. The images obtained by atomic force microscopy (AFM) suggest interaction with pBR322 plasmid DNA. Measurements of the viscosity of solutions of free DNA and DNA incubated with different concentrations of the compounds confirmed this interaction. The cytotoxicity of compounds 1234 was much higher than that of cisplatin against human leukemia cancer cells (HL-60 cells). IC50 values for all the compounds are in the range of submicromolar amounts. Apoptotic death percentage was also studied resulting similar than that of cisplatin.  相似文献

Two asymmetric tridentate copper(II) complexes, [Cu(dppt)Cl(2)].0.25H(2)O (1) (dppt=3-(1,10-phenanthrolin-2-yl)-5,6-diphenyl-as-triazine) and [Cu(pta)Cl(2)] (2) (pta=3-(1,10-phenanthrolin-2-yl)-as-triazino[5,6-f]acenaphthylene), have been prepared and characterized by elemental analysis, IR and Fast atomic bombardment mass spectra. Complex 1 has also been structurally characterized. The complexes exist as distorted square pyramid with five co-ordination sites occupied by the tridentate ligand and the two chlorine anions. DNA interaction studies suggest that the ligand planarity of the complex has a significant effect on DNA binding affinity increasing in the order [Cu(dppt)Cl(2)]< [Cu(pta)Cl(2)]. In the presence of ascorbate or glutathione, the two complexes are found to cause significant cleavage of double-strand pBR 322 DNA and [Cu(pta)Cl(2)] exhibited the higher cleaving efficiency.  相似文献

The experimental results indicate that the size and shape of the intercalated ligand have a marked effect on the binding affinity of complexes to CT-DNA. Complexes 2 and 3 have also been found to promote cleavage of plasmid pBR322 DNA from the supercoiled form I to the open circular form II upon irradiation.  相似文献

Acenaphtheno[1,2-b]-1,4,8,9-tetraazatriphenylene (atatp) and its complexes [Ru(L)2atatp](ClO4)2 x nH2O (L = 2,2'-bipyridine (bpy), n=2 (1); 1,10-phenanthroline (phen), n=2 (2); and 2,9-dimethyl-1,10-phenanthroline (dmp), n=1 (3)) have been synthesized and characterized by elemental analyses and 1H NMR. The spectral and electrochemical properties of these complexes are also examined. Complexes 1 and 2 display bright luminescence in acetonitrile but very weak luminescence in water solution. However, complex 3 is not luminescent in either solvent. The interaction of the complexes with calf thymus DNA (CT-DNA) has been studied by absorption, emission and viscosity measurements. The intrinsic binding constants of complexes 1 and 2 are 7.6 x 10(4) and 8.8 x 10(4) M(-1) respectively. The relatively low affinities of complexes 1 and 2 with DNA may arise from the atatp ligand, indicating that the size and shape of the intercalated ligand have a marked effect on the strength of interaction. Complexes 1 and 2 bind with CT-DNA in an intercalative mode but complex 3 in a non-intercalative one, showing that changing the ancillary ligand affects not only the binding magnitude, but also the binding mode of the interaction.  相似文献

Ruthenium(II) bis(2,2″-pyridyl) complexes with bridging ligands: 6,7-dichloro-2,3-di(2-pyridyl)quinoxaline; 2,3-di(2-pyridyl)-quinoxaline; 5-methyl-2,3-di(2-pyridyl) quinoxaline; 6,7-dibenzo-2,3-di(2-pyridyl)quinoxaline have been prepared. The electrochemical and spectroscopic properties of these complexes are reported. The resonance Raman spectroelectrochemical results indicate the presence of oxidation state sensitive marker bands in the resonance Raman spectra of the oxidized complexes. The spectroscopic data for the reduced complexes is similar for all four species. The resonance Raman data for the reduced species are dominated by 2,2″-bipyridyl vibrations.  相似文献

Neutral Ru(II) complexes with the formula trans-[Ru(trpy*)(L2)(pcyd)] have been prepared, where trpy* = 4,4′,4″-tri-tert-butyl-terpyridine, L2 = 2-pyrazinecarboxylato (pca), 2-pyridinecarboxylato (pic), acetylacetonato (acac) and pcyd = 3-chlorophenylcyanamido (3-Clpcyd), 2,3-dichlorophenylcyanamido (2,3-Cl2pcyd), 2,4,6-trichlorophenylcyanamido (2,4,6- Cl3pcyd), 2,3,4,5-tetrachlorophenylcyanamido (2,3,4,5-Cl4pcyd) and 3,4,5-trimethoxyphenylcyanamido (3,4,5-(OMe)3pcyd). Spectroelectrochemistry was performed on these Ru(II) complexes to obtain the visible absorption spectrum of the Ru(III)–cyanamide ligand-to-metal charge transfer chromophore. The Ru(III)–cyanamide metal–ligand coupling elements of these complexes were compared to other Ru(III)–cyanamide complexes.  相似文献

The two complexes containing bioactive ligands of the type and [Fe(L)] (PF(6))(2) (1) (where L = [1-{[2-{[2-hydroxynaphthalen-1-yl)methylidine]amino}phenyl)imino] methyl}naphthalene-2-ol]) and [Co(L(1)L(2))] (PF(6))(3) (2) (where L(1)L(2) = mixed ligand of 2-seleno-4-methylquinoline and 1,10-phenanthroline in the ratio 1:2, respectively) were synthesized and structurally characterized. The DNA binding property of the complexes with calf thymus DNA has been investigated using absorption spectra, viscosity measurements, and thermal denaturation experiments. Intrinsic binding constant K(b) has been estimated at room temperature. The absorption spectral studies indicate that the complexes intercalate between the base pairs of the CT-DNA tightly with intrinsic DNA binding constant of 2.8 × 10(5) M(-1) for (1) and 4.8 × 10(5) M(-1) for (2) in 5 mM Tris-HCl/50 mM NaCl buffer at pH 7.2, respectively. The oxidative cleavage activity of (1) and (2) were studied by using gel electrophoresis and the results show that complexes have potent nuclease activity.  相似文献

A series of enantiomerically pure polypyridyl ruthenium(II) complexes, delta- and lambda-[Ru(bpy)2 (HPIP)](PF6)2 (delta-1 and lambda-1; bpy=2,2'-bipyridine, HPIP = 2-(2-hydroxyphenyl)imidazo[4,5-f][1,10]phenanthroline), delta and lambda-[Ru(bpy)2(HNAIP)](PF6)2 (delta-2 and lambda-2; HNAIP = 2-(2-hydroxy-1-naphthyl)imidazo[4,5-f][1,10]phenanthroline), delta- and lambda-[Ru(bpy)2 (HNOIP)](PF6)2 (delta-3 and lambda-3; HNOIP = 2-(2-hydroxy-5-nitrophenyl)imidazo[4,5-f][1,10]phenanthroline), and delta- and lambda-[Ru(bpy)2(DPPZ)](PF6)2 (delta-4 and lambda-4; DPPZ= dipyridophenazine), have been synthesized. Binding behavior of these chiral complexes to calf thymus DNA (CT-DNA) has been investigated by electronic absorption, steady-state emission, and circular dichroism spectroscopies, as well as by viscosity measurements and equilibrium dialysis binding studies. Several points came from the results. (1) The DNA-binding properties were distinctly different for the [Ru(bpy)2L]2+ (L=HPIP, HNAIP, HNOIP) series of ruthenium(II) complexes, which indicates that the photophysical behavior of the complexes on binding to DNA can be modulated through ligand design. (2) Different binding rates of individual enantiomers of complexes 1 and 4 to DNA were observed through dialysis experiments. The lambda enantiomer bound more rapidly than the lambda enantiomer and their different intercalative binding geometries were suggested to be responsible. (3) Both delta-2 and lambda-2 bound weakly to CT-DNA; delta-2 may bind through a partial intercalation mode, whereas lambda-2 may bind in the DNA groove. (4) There was no noticeable enantioselectivity for complexes 1, 3, and 4 on binding to CT-DNA. Both of their enantiomers can intercalate into DNA base pairs. It is noted that delta-3 and lambda-3 exhibited almost identical spectral changes on addition of CT-DNA, and a similar binding manner of the isomers to the double helix was proposed.  相似文献

Two mixed ligand complexes [Ru(bpy)(2)(DMHBT)]Cl(2)(1) and [Ru(phen)(2)(DMHBT)]Cl(2) (2) (where DMHBT is 11,13-dimethyl-13H-4,5,9,11,14-hexaaza-benzo[b]triphenylene-10,12-dione) have been synthesized and characterized by electrospray ionization (ESI) mass, (1)H-(1)H correlation spectroscopy (COSY), electronic spectroscopy, fluorescence spectroscopy and cyclic voltammetry. Spectroscopic titration and viscosity changes of calf thymus (CT)-DNA in the presence of incremental amount of complexes 1 and 2 clearly demonstrate that both these complexes bind intercalatively to DNA, with binding constant 2.87+/-0.20 x 10(4)M(-1) and 1.01+/-0.20 x 10(5)M(-1) for complexes 1 and 2, respectively. All the experimental evidences suggest that the ancillary ligand 2,2'-bipyridine (bpy) and 1,10-phenanthroline (phen) influences the intercalative binding of these complexes to DNA.  相似文献

The complexes Pt(pq)Cl2(1) and Pt(pq)(bdt) (2) (where pq = 2-(2'pyridyl)quinoxaline and bdt=benzene-1,2-dithiolate) have been synthesized and fully characterized by UV-visible (UV-Vis), Fourier Transformer Infrared Spectra (FTIR), 1 and 2D NMR and cyclic voltammetry (CV). Interactions of the tested systems (the aforementioned complexes 1 and 2) and the free ligands pq and bdt with double stranded calf thymus DNA (CT-DNA) were studied by UV-spectrophotometric (melting curves) and circular dichroism (CD) measurements. The results suggest that both complexes 1 and 2, are able to form adducts with DNA and to distort the double helix by changing the base stacking. Complex 2 forms stronger adducts to CT-DNA than complex 1 and this is probably due to the substitution of the chlorine atoms of 1 by the 1,2-dithiolate ligand (bdt) in 2. The latter induces an extensive distortion in the planarity of 2 as density functional theory (DFT) calculations reveal. Besides, the light absorbing complex 2 possess intense mixed metal ligand to ligand charge transfer (MM'LLCT) transition in the visible region of the spectrum and could act as photoluminescent metal-based probe for the study of DNA binding. Thus, the photocleavage of DNA by 2 has been studied by UV-Vis and CD spectra and monitored by agarose gel electrophoresis. Under our experimental conditions, it is unclear that complex 2 can photocleave DNA. Furthermore, the ability of 2 to inhibit proliferation of human tumor cell lines was tested and the results indicate some cytoxytic effect on the SF-286 cells.  相似文献

We describe the synthesis, characterization, and reactivity of several Ru(II) complexes of the type cis-L2Ru(Z)n+, where L is an α-diimine [e.g. 2,2′-bipyridine (bpy) or 1,10-phenanthroline (phen)] ligand and Z is a bis-coordinated scorpionate ligand such as tris-(1-pyrazolyl)methane (HC(pz)3, PZ=1-pyrazolyl; n=2) or tetrakis-(1-pyrazolyl)borate anion (B(pz)4; n=1). The complexes each exhibit strong visible absorption assigned as a π*(L)←dπ(Ru) metal-to-ligand charge-transfer (MLCT) transition characteristic of the cis-L2Ru2+ kernel. A corresponding MLCT excited state emission is observed in room temperature CH3CN solution, although emission energies, lifetimes, and quantum yields are reduced relative to Ru(bpy)3 2+. Electronic spectra and cyclic voltammetry measurements indicate that the relative π-acceptor abilities of the coordinated Z are: Z=(1H-pyrazolyl)2(pz)2B(pz)2<(pyridine)2<(pz)2CH(pz). Uncoordinated pz groups of cis-(bpy)2Ru(pz)2B(pz)2 + can be reacted to form a sterically hindered, localized-valence (Kcom33 l mol−1) cis,cis-(bpy)2RuII(pz)2B(pz)2RuII(bpy)2 3+ dimer. The dimer properties are interpreted by comparison to the known cis-(bpy)2RuII(pz)2RuII(bpy)2 2+ analog. The dimer is photoreactive and undergoes an asymmetrical photocleavage in CH3CN (yielding cis-(bpy)2RuIII(pz)2B(pz)2 2+ and cis-(bpy)2RuII(CH3CN)2 2+), similar to the corresponding thermal reaction observed for the mixed-valence cis-(bpy)2RuII(pz)2RuIII(bpy)2 3+ system.  相似文献

The aim of this study was to synthesize and evaluate plasmid DNA interaction of new platinum(II) complexes with some 2-substituted benzimidazole derivatives as carrier ligands which may have potent anticancer activity and low toxicity. Twelve benzimidazole derivatives carrying indole, 2-/or 3-/or 4-methoxyphenyl, 4-methylphenyl, 3,4-dimethoxyphenyl, 3,4,5-trimethoxyphenyl, 4-methoxybenzyl, 3,4,5-trimethoxybenzyl, 3,4,5-trimethoxystyryl, 3,4,5-trimethoxybenzylthio or dimethylamino ethyl groups in their position 2 and twelve platinum(II) complexes with these carrier ligands were synthesized. The chemical structure of the platinum complexes have been characterized by their elemental analysis and FIR, 1H NMR and mass spectra and their 1H NMR and FIR spectra were interpreted by comparison with those of the ligands. The interaction of all the ligands and their complexes with plasmid DNA and their restriction endonuclease reactions by BamHI and HindIII enzymes were studied by agarose gel electrophoresis. It was determined that complex 1 [dichloro-di(2-(1H-indole-3-yl)benzimidazole)platinum(II)·2H2O] has stronger interaction than carboplatin and complex 10 [dichloro-di(2-(3,4,5-trimethoxystyryl)benzimidazole)platinum(II)·2H2O] has stronger interaction than both carboplatin and cisplatin with plasmid DNA.  相似文献

Mixed-ligand ruthenium(II) complexes of three photoactive ligands, viz., (E)-1-[2-(4-methyl-2-pyridyl)-4-pyridyl]-2-(1-naphthyl)-1-ethene (mppne), (E)-1-(9-anthryl)-2-[2-(4-methyl-2-pyridyl)-4-pyridyl]-1-ethene (mppae) and (E)-1-[2-(4-methyl-2-pyridyl)-4-pyridyl]-2-(1-pyrenyl)-1-ethene (mpppe), in which a 2,2′-bipyridyl unit is linked via an ethylinic linkage to either a naphthalene, an anthracene or a pyrene chromophore and three electroactive ligands, viz., 4-(4-pyridyl)-1,2-benzenediol (catpy), 5,6-dihydroxy-1,10-phenanthroline (catphen) and 1,2-benzenediol (cat), were synthesized in good to moderate yields. Complexes [Ru(bpy)2(mppne)]2+ (bpy is 2, 2′–bipyridyl), [Ru(bpy)2(mppae)]2+, [Ru(bpy)2(mpppe)]2+, [Ru(bpy)2(sq-py)]+, [Ru(bpy)2(sq-phen)]+ and [Ru(phen)2(bsq)]+ (phen is 1,10-phenanthroline) were fully characterized by elemental analysis, IR, 1H NMR, fast-atom bombardment or electron-impact mass, UV–vis and cyclic voltammetric methods. In the latter three complexes, the ligands catpy, catphen and cat are actually bound to the metal center as the corresponding semiquinone species, viz., 4-(4-pyridyl)-1,2-benzenedioleto(+I) (sq-py), 1,10-phenanthroline-5,6-dioleto(+I) (sq-phen) and 1,2-benzenedioleto(+I) (bsq), thus making the overall charge of the complexes formally equal to + 1 in each case. These three complexes are electron paramagnetic resonance active and exhibit an intense absorption band between 941 and 958 nm owing to metal-to-ligand charge transfer (MLCT, d Ruπ*sq) transitions. The other three ruthenium(II) complexes containing three photoactive ligands, mppne, mppae and mpppe, exhibit MLCT (d Ruπ*bpy ) bands in the 454–461-nm region and are diamagnetic. These can be characterized by the 1H NMR method. [Ru(bpy)2(mppne)]2+, [Ru(bpy)2(mppae)]2+ and [Ru(bpy)2(mpppe)]2+ exhibit redox waves corresponding to the RuIII/RuII couple along with the expected ligand (bpy and substituted bpy) based ones in their cyclic and differential pulse voltammograms (CH3CN, 0.1 M tetrabutylammonium hexafluorophosphate)—corresponding voltammograms of [Ru(bpy)2(sq-py)]+, [Ru(bpy)2(sq-phen)]+ and [Ru(phen)2(bsq)]+ are mainly characterized by waves corresponding to the quinone/semiquinone (q/sq) and semiquinone/1,2-diol (sq/cat) redox processes. The results of absorption and fluorescence titration as well as thermal denaturation studies reveal that [Ru(bpy)2(mppne)]2+ and [Ru(bpy)2(mppae)]2+ are moderate-to-strong binders of calf thymus DNA with binding constants ranging from 105 to 106 M−1. Under the identical conditions of drug and light dose, the DNA (supercoiled pBR 322) photocleavage activities of these two complexes follow the order:[Ru(bpy)2(mppne)]2+>[Ru(bpy)2(mppae)]2+, although the emission quantum yields follow the reverse order. The other ruthenium(II) complexes containing the semiquinone-based ligands are found to be nonluminescent and inefficient photocleavage agents of DNA. However, experiments shows that [Ru(bpy)2(sq)]+-based complexes oxidize the sugar unit and could be used as mild oxidants for the sugar moiety of DNA. The preparation of a series of tridentate ligands of formulae X(CH2C7H5N2)2 (X = NH, S, O, S2) is described. The ligands contain two benzimidazole moieties and one of NH, S, O, or S2 as the donor groups. Cu(I) and Cu(II) complexes of these ligands are prepared and characterized. Spectroscopic and X-ray data imply that the geometric constraints of these ligands impose a distorted coordination geometry at copper. The implications and relevance of this chemistry to copper proteins is discussed.  相似文献

Treatment of [(η6-p-cymene)RuCl(μ-Cl)]2 with Lawesson’s reagent [ArP(S)(μ-S)]2 (Ar = p-C6H4OMe) in the presence of ammonium hydroxide afforded the dinuclear complex [(η6-p-cymene)Ru{μ-η1(S),η2(S,S′)-ArP(O)S2}]2 (1) in which the tripodal [ArP(O)S2]2− ligands bind to the ruthenium atom in both bridging and chelating modes with two non-coordinating PO groups. Interaction of [RuHCl(CO)(PPh3)3] with [ArP(S)(μ-S)]2 and bis(diphenylphosphino)methane (dppm) in the presence of ammonium hydroxide gave the dinuclear complex [Ru(CO){μ3-η1(O),η2(S,S′)-ArP(O)S2}(dppm)]2 (2) in which the tripodal [ArPOS2]2− ligands bind the two Ru atoms via both sulfur and oxygen atoms. Treatment of [Ru(PPh3)3Cl2] with [ArP(S)(μ-S)]2 at reflux in the presence of ammonium hydroxide led to the formation of the dinuclear mixed valence complex [Ru2Cl2(μ-S){μ3-η1(O),η1(S),-η2(S,S′)-ArP(O)S2}(PPh3)3] (3), which contains a [RuII(PPh3)2Cl]+ and [RuIV(PPh3)Cl]3+ moieties by the tripodal [ArPOS2]2− ligand in a μ3-η1(O),η1(S),η2(S,S′) coordination mode and the μ-S2− anion. The crystal structures of 1, 2, and 3·CH2Cl2 along with their spectroscopic and electrochemical properties are reported.  相似文献
