The kinetics of the formation of the purple complex [FeIII(EDTA)O2]3−, between FeIII-EDTA and hydrogen peroxide was studied as a function of pH (8.22-11.44) and temperature (10-40 °C) in aqueous solutions using a stopped-flow method. The reaction was first-order with respect to both reactants. The observed second-order rate constants decrease with an increase in pH and appear to be related to deprotonation of FeIII-EDTA ([Fe(EDTA)H2O] ⇔ Fe(EDTA)OH]2− + H+). The rate law for the formation of the complex was found to be d[FeIIIEDTAO2]3−/dt=[(k4[H+]/([H+] + K1)][FeIII-EDTA][H2O2], where k4=8.15±0.05×104 M−1 s−1 and pK1=7.3. The steps involved in the formation of [Fe(EDTA)O2]3− are briefly discussed.  相似文献

A mononuclear cobalt(III)-peroxo complex bearing a macrocyclic tetradentate N4 ligand, [CoIII(TMC)(O2)]+ (TMC = 1,4,8,11-tetramethyl-1,4,8,11-tetraazacyclotetradecane), was generated in the reaction of [CoII(TMC)]2+ and H2O2 in the presence of triethylamine in CH3CN. The reactivity of the cobalt(III)-peroxo complex was investigated in aldehyde deformylation with various aldehydes and compared with that of iron(III)- and manganese(III)-peroxo complexes, such as [FeIII(TMC)(O2)]+ and [MnIII(TMC)(O2)]+. In this reactivity comparison, the reactivities of metal-peroxo species were found to be in the order of [MnIII(TMC)(O2)]+ > [CoIII(TMC)(O2)]+ > [FeIII(TMC)(O2)]+. A positive Hammett ρ value of 1.8, obtained in the reactions of [CoIII(TMC)(O2)]+ and para-substituted benzaldehydes, demonstrates that the aldehyde deformylation by the cobalt(III)-peroxo species occurs via a nucleophilic reaction.  相似文献

The oxidation of thiocyanate by iron(V) (Fe(V)) was studied as a function of pH in alkaline solutions by a premix pulse radiolysis technique. The rates decrease with an increase in pH. The rate law for the oxidation of SCN by Fe(V) was obtained as −d[Fe(V)]/dt = k10{[H+]2/([H+]2 + K2[H+] + K2K3)}[Fe(V)][SCN], where k10 = 5.72 ± 0.19 × 106 M−1 s−1, pK2 = 7.2, and pK3 = 10.1. The reaction precedes via a two-electron oxidation, which converts Fe(V) to Fe(III). Thiocyanate reacts approximately 103× faster with iron(V) than does with iron(VI).  相似文献

An arabinoglucuronoxylan was extracted from the holocellulose of sugi (Cryptomeria japonica) wood with 10% KOH and subjected to hydrolysis by partially purified xylanase fraction from a commercial cellulase preparation “Meicelase”. Neutral sugars liberated were analyzed by size exclusion chromatography showing the presence of xylooligosaccharides up to xylohexaose. Aldouronic acids liberated were purified by preparative anion exchange chromatography. Their structures were identified by monosaccharide analysis, comparison of their volume distribution coefficients (Dvs) with those of the authentic samples in anion exchange chromatography and 1H and 13C NMR spectroscopy, resulting in the characterization of eight aldouronic acids including acids consisting of two 4-O-Me-α-D-GlcAp residues and 3-5 D-Xyl residues.

The heme-based oxygen-sensor enzyme from Escherichia coli (Ec DOS) is a heme-regulated phosphodiesterase with activity on cyclic-di-GMP and is composed of an N-terminal heme-bound sensor domain with the PAS structure and a C-terminal functional domain. The activity of Ec DOS is substantially enhanced by the binding of O2 to the Fe(II)-protoporphyrin IX complex [Fe(II) complex] in the sensor domain. The binding of O2 to the Fe(II) complex changes the structure of the sensor domain, and this altered structure becomes a signal that is transduced to the functional domain to trigger catalysis. The first step in intra-molecular signal transduction is the binding of O2 to the Fe(II) complex, and detailed elucidation of this molecular mechanism is thus worthy of exploration. The X-ray crystal structure reveals that Phe113 is located close to the O2 molecule bound to the Fe(II) complex in the sensor domain. Here, we found that the O2 association rate constants (>200 × 10−3 μM−1 s−1: F113L; 26 × 10−3 μM−1 s−1: F113Y) of the Fe(II) complexes of Phe113 mutants were markedly different from that (51 × 10−3 μM−1 s−1) of the wild-type enzyme, and auto-oxidation rates (0.00068 min−1: F113L; 0.039 min−1: F113Y) of the Phe113 mutants also differed greatly from that (0.0062 min−1) of the wild-type enzyme. We thus suggest that Phe113, residing near the O2 molecule, has a critical role in optimizing the Fe(II)-O2 complex for effective regulation of catalysis by the oxygen-sensor enzyme. Interactions of CO and cyanide anion with the mutant proteins were also studied.  相似文献

Four new mononuclear iron(III) complexes with the substituted-salicylaldimine ligands, [Fe(L1)(TCC)] (1), [Fe(L2)(TBC)] (2), [Fe(L3)(TBC)] (3) and [Fe(L4)(TCC)](CH3CN) (4) (HL1 = N′-(5-OH-salicylaldimine)-diethylenetriamine, HL2 = (N′-(5-Cl-salicylaldimine)-diethylenetriamine, HL3 N′-(5-Br-salicyl-aldimine)-dipropylenetriamine, HL4 = (N′-3,5-Br-salicylaldimine)-dipropylenetriamine, H2TCC = tetrachlorocatechol, and H2TBC = tetrabromocatechol), were prepared and characterized by XRD, EPR, and Mössbauer spectroscopy. The coordination sphere of the Fe(III) in complexes 1-4 is a distorted octahedral with N3O3 donors set which constructed by the Schiff-base ligands and the catecholate substrates of TBC or TCC. The in situ prepared Fe(III) complexes [Fe(L1)Cl2], [Fe(L2)Cl2], [Fe(L3)(Cl2)], and [Fe(L4)Cl2] in absence of TBC or TCC show a high catecholase-like activity for the oxidation of 3,5-DTBC to the corresponding quinone 3,5-DTBQ.  相似文献

Evaluation of the cytotoxicity of an ethanolic root extract of Sideroxylonfoetidissimum subsp. gaumeri (Sapotaceae) revealed activity against the murine macrophage-like cell line RAW 264.7. Systematic bioassay-guided fractionation of this extract gave an active saponin-containing fraction from which four saponins were isolated. Use of 1D (1H, 13C, DEPT135) and 2D (COSY, TOCSY, HSQC, and HMBC) NMR, mass spectrometry and sugar analysis gave their structures as 3-O-(β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranosyl)-28-O-(α-l-rhamnopyranosyl-(1 → 3)[β-d-xylopyranosyl-(1 → 4)]-β-d-xylopyranosyl-(1 → 4)-α-l-rhamnopyranosyl-(1 → 2)-α-l-arabinopyranosyl)-16α-hydroxyprotobassic acid, 3-O-β-d-glucopyranosyl-28-O-(α-l-rhamnopyranosyl-(1 → 3)[β-d-xylopyranosyl-(1 → 4)]-β-d-xylopyranosyl-(1 → 4)-α-l-rhamnopyranosyl-(1 → 2)-α-l-arabinopyranosyl)-16α-hydroxyprotobassic acid, 3-O-(β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranosyl)-28-O-(α-l-rhamnopyranosyl-(1 → 3)-β-d-xylopyranosyl-(1 → 4)[β-d-apiofuranosyl-(1 → 3)]-α-l-rhamnopyranosyl-(1 → 2)-α-l-arabinopyranosyl)-16α-hydroxyprotobassic acid, and the known compound, 3-O-β-d-glucopyranosyl-28-O-(α-l-rhamnopyranosyl-(1 → 3)[β-d-xylopyranosyl-(1 → 4)]-β-d-xylopyranosyl-(1 → 4)-α-l-rhamnopyranosyl-(1 → 2)-α-l-arabinopyranosyl)-protobassic acid. Two further saponins were obtained from the same fraction, but as a 5:4 mixture comprising 3-O-(β-d-glucopyranosyl)-28-O-(α-l-rhamnopyranosyl-(1 → 3)-β-d-xylopyranosyl-(1 → 4)[β-d-apiofuranosyl-(1 → 3)]-α-l-rhamnopyranosyl-(1 → 2)-α-l-arabinopyranosyl)-16α-hydroxyprotobassic acid and 3-O-(β-d-apiofuranosyl-(1 → 3)-β-d-glucopyranosyl)-28-O-(α-l-rhamnopyranosyl-(1 → 3)[β-d-xylopyranosyl-(1 → 4)]-β-d-xylopyranosyl-(1 → 4)-α-l-rhamnopyranosyl-(1 → 2)-α-l-arabinopyranosyl)-16α-hydroxyprotobassic acid, respectively. This showed greater cytotoxicity (IC50 = 11.9 ± 1.5 μg/ml) towards RAW 264.7 cells than the original extract (IC50 = 39.5 ± 4.1 μg/ml), and the saponin-containing fraction derived from it (IC50 = 33.7 ± 6.2 μg/ml).  相似文献

Recently, a series of Fe(II) complexes have been published by our group with 3 N-donor 1,3-bis(2′-Ar-imino)isoindoline ligands containing various Ar-groups (pyridyl, 4-methylpyridyl, thiazolyl, benzimidazolyl and N-methylbenzimidazolyl). The superoxide scavenging activity of the compounds showed correlation with the Fe(III)/Fe(II) redox potentials. Analogous, electroneutral chelate complexes with Mn(II) and Ni(II) in 2:1 ligand:metal composition are reported here. Each Mn(II) complex exhibits one reversible redox wave that is assigned as the Mn(III)/Mn(II) redox transition. The E1/2 spans a 180 mV range from − 98 (Ar = 3-methylpyridyl) to 82 mV (Ar = thiazolyl) vs. the Fc+/Fc depending on the Ar-sidearm. The SOD-like (SOD=superoxide dismutase)activity of all complexes was determined according to the McCord-Fridovich method. The Mn(II) isoindolinates have IC50 values - determined with 50 μM cytochrome c Fe(III) - that range from (3.22 ± 0.39) × 10− 6 (Ar = benzimidazolyl) to (10.80 ± 0.54) × 10− 6 M (Ar = N-methylbenzimidazolyl). In contrast with the Fe(II) complexes, the IC50 concentrations show no significant dependence on the E1/2 values in this narrow potential range emphasizing that the redox potential is not the governing factor in the Mn(II)-containing scavengers. The analogous Ni(II) compounds show no redox transitions in the thermodynamically relevant potential range (− 0.40 to 0.65 V vs. SCE) and accordingly, their superoxide scavenging activity (if any) is below the detection level.  相似文献

The two isostructural anions, [CeIII(α-PW11O39)2]11− and [CeIV(α-PW11O39)2]10−, both composed of CeIII/IV sandwiched by two mono-lacunary Keggin units [α-PW11O39]7−, were isolated as dimethylammonium salts and structurally characterized by single-crystal X-ray diffraction analysis. The structures of the [CeIII/IV(α-PW11O39)2]11−/10− anions exhibit two enantiomers d- and l-forms due to C2 (2) symmetry from the square-antiprismatic coordination of the central CeIII/IVO8 polyhedron. The CeIII compound crystallizes in achiral space group P21/n, while the CeIV analog in chiral space group P21 shows spontaneous resolution without any chiral auxiliaries. The correlation between the ionic radius of the central metal (M) and twist angle of the two mono-lacunary units was studied for a series of [M(α-PW11O39)2]n (M = CeIII/IV, EuIII, ZrIV, HfIV) anions and explained in terms of steric repulsion between the O atoms in the mono-lacunary units.  相似文献

A series of mononuclear iron(III) complexes with containing phenolate donor of substituted-salicylaldimine based ligands [Fe(L1)(TCC)] · CH3OH (1), [Fe(L2)(TCC)] · CH3OH (2), [Fe(L3)(TCC)] (3), and [Fe(L4)(TCC)] (4) have been prepared and studied as functional models for catechol dioxygenases (H2TCC = tetrachlorocatechol, or HL1 = N′-(salicylaldimine)-N,N-diethyldiethylenetriamine, HL2 = N′-(5-Br-salicylaldimine)-N,N-diethyldiethylenetriamine, HL3 = N′-(4,6-dimethoxy-salycyl-aldimine)-N,N-diethyl-diethylenetriamine, HL4 = N′-(4-methoxy-salicylaldimine)-N,N-diethyl-diethylenetriamine). They are structural models for inhibitors of enzyme-substrate adducts from the reactions of catechol 1,2-dioxygenases. Complexes 1-4 were characterized by spectroscopic methods and X-ray crystal structural analysis. The coordination sphere of Fe(III) atom of 1-4 is distorted octahedral with N3O3 donor set from the ligand and the substrate TCC occupying cis position, and Fe(III) is in high-spin (S = 5/2) electronic ground state. The in situ prepared iron(III) complexes without TCC, [Fe(L1)Cl2], [Fe(L2)Cl2], [Fe(L3)Cl2], and [Fe(L4)Cl2] are reactive towards intradiol cleavage of the 3,5-di-tert-butylcatechol (H2DBC) in the presence of O2 or air. The reaction rate of catechol 1,2-dioxygenase depends on the redox potential and acidity of iron(III) ions in complexes as well as the substituent effect of the ligands. We have identified the reaction products and proposed the mechanism of the reactions of these iron(III) complexes with H2DBC with O2.  相似文献

A straightforward synthetic method to prepare mononuclear croconato-containing iron (III) complexes, (A)3[Fe(C5O5)3] [A = tetrabutylammonium = n-Bu4N+ (1) and along with their crystal structures and magnetic properties, are reported. The Fe(III) atom adopts a pseudo-octahedral geometry while magnetic susceptibility measurements, in the 2-300 K temperature range, show the occurrence of a high spin state (S = 5/2) in both complexes.  相似文献

Reaction of [Ru2(O2CMe)4]Cl with K3[Cr(CN)5NO] in water forms Hx[RuII/III2(O2CMe)4]3−x-[Cr(CN)5NO]·zH2O (x = 0.2) that magnetically orders at 4.0 K and possesses an interpenetrating body centered cubic [a = 13.2509(2) Å] structure with random locations of the bridging nitrosyl ligands, and x/3 vacant cation sites. Similarly, the aqueous reaction of [Ru2(O2CMe)4]Cl with Na2[Fe(CN)5NO] forms paramagnetic [Ru2(O2CMe)4]2[Fe(CN)5NO]·H2O, which has a similar tetragonal interpenetrating structure [a = 13.0186(1) Å, c = 13.0699(2) Å] where the NO ligands are presumably nonbridging and 1/3 of the expected cation sites are unoccupied. The presence of uncoordinated NO sites in addition to missing neighboring [Ru2(O2CMe)4]+ units, results in significant vacancies (or holes) in the lattice.  相似文献

The aim of the present study was to investigate the effect of HIF1α on Foxp3 expression in CD4+CD25? T lymphocytes. CD4+CD25? T lymphocytes were sorted from PBMC using a CD4+CD25+ regulatory T cell isolation kit. Lentivirus containing lentiviral vector that overexpressed HIF1α (HIF‐lenti) and those containing empty expression vector (control‐lenti) were produced. Meanwhile, lentivirus that contained lentiviral vector that suppressed HIF1α expression (siHIF‐lenti) and those containing control vector (sicontrol‐lenti) were also generated. The sorted CD4+CD25? T lymphocytes were infected with HIF‐lenti, control‐lenti, siHIF‐lenti, and sicontrol‐lenti, respectively. Approximately 72 hr after transduction, real‐time PCR and Western blot were carried out to analyze the RNA and protein expression level of HIF1α and Foxp3. CD4+CD25? T lymphocytes cultured under 21% O2, 5% CO2 (normoxia) and 1% O2, 5% CO2 (hypoxia) were used as control. Our results showed that overexpression of HIF1α increased both mRNA and protein expression of Foxp3 and, meanwhile, suppression of HIF1α expression by RNAi could reverse high Foxp3 expression in CD4+CD25? T lymphocytes caused by hypoxic culture. These results suggested that hypoxia could stimulate Foxp3 expression by increasing HIF1α expression in CD4+ T lymphocytes which may promote CD4+ T lymphocytes to convert to Treg.

Salts of the Fe(III) spin crossover cation [FeIII(qsal)2]+ (qsalH = N-(8-quinolyl)salicylaldimine) and monoanions [MIII(pds)2] (M = Cu, Au; pds = pirazine-2,3-diselenolate) with formula [FeIII(qsal)2][MIII(pds)2] were prepared and characterized by single crystal X-ray diffraction and magnetic measurements. The heterogeneity of gold sites in various Au/TiO2 catalysts was studied by means of state-sensitive Au L3-edge X-ray absorption fine structure (XAFS) combined with high energy-resolution X-ray fluorescence spectrometry. A series of Au/TiO2 catalysts were prepared via deposition-precipitation method on anatase-type or mesoporous (amorphous) TiO2 added with NaOH (lower Au loading) or urea (higher Au loading). The mean Au particle size ranged between 29 and 87 Å based on high-resolution TEM (transmission electron microscope) measurements. The Au Lα1 emission peak energy for Au/mesoporous-TiO2 in air and Au/anatase-TiO2 in CO (5%) corresponded to Au0 state. The emission peak energy for Au/anatase-TiO2 in air shifted toward that of AuI state. For relatively greater Au particles (average 87 Å) dispersed on mesporous TiO2, the major valence state discriminated by Au Lα1-selecting XANES (X-ray absorption near-edge structure) spectrum tuned to Au Lα1 emission peak top was Au0, but the Auδ state could be successfully monitored by Au Lα1-selecting XANES tuned to the emission energy at 9707.6 eV, of which population was relatively small compared to the case of smaller Au particles (average 29 Å) on anatase-type TiO2. On the other hand, negative charge transfer from Au 5d to support was demonstrated in Auδ+-state sensitive XANES tuned to 9718.3-9718.7 eV. The Auδ+-state sensitive XANES spectra resembled theoretically generated XANES for interface Auδ+ sites model on TiO2 in contact with surface Ti sites. Further charge transfer was demonstrated from Au to adsorbed O2 for Au/anatase-TiO2 catalyst.  相似文献

Compounds FeIII(3-CH3O-qsal)2PF6 · nH2O (n = 0, 2) (1, 1 · 2H2O) were synthesized and characterized: the structure of 1 and the magnetic properties of both compounds were determined. Compound 1 · 2H2O presents properties characteristic of high-spin Fe(III), while 1 presents properties of low-spin Fe(III) with an onset of a gradual spin crossover at ca. 300 K.  相似文献

Four flavone glycosides isolated from extracts of the leaves of Robinia pseudoacacia (Leguminosae) were characterised by spectroscopic and chemical methods as the 7-O-β-d-glucuronopyranosyl-(1 → 2)[α-l-rhamnopyranosyl-(1 → 6)]-β-d-glucopyranosides of acacetin (5,7-dihydroxy-4′-methoxyflavone), apigenin (5,7,4′-trihydroxyflavone), diosmetin (5,7,3′-trihydroxy-4′-methoxyflavone) and luteolin (5,7,3′,4′-tetrahydroxyflavone). Assignment of glycosidic 1H and 13C resonances in their NMR spectra was facilitated by 2JHC correlations detected using the H2BC (heteronuclear two-bond correlation) pulse sequence. Spectroscopic analysis of two known triglycosides, acacetin 7-O-β-d-glucopyranosyl-(1 → 2)[α-l-rhamnopyranosyl-(1 → 6)]-β-d-glucopyranoside (previously unrecorded from this species) and acacetin 7-O-β-d-xylopyranosyl-(1 → 2)[α-l-rhamnopyranosyl-(1 → 6)]-β-d-glucopyranoside (‘acacetin trioside’), enabled inconsistencies in the literature relating to these structures to be resolved. Comparison of the flavonoid chemistry of leaves and flowers of R. pseudoacacia using LC-UV and LC-MS showed that flavone 7-O-glycosides, particularly of acacetin, predominated in the former, whereas the latter comprised mainly flavonol 3,7-di-O-glycosides, including several examples new to this species. Tissue dependent differences in flavonoid chemistry were also evident from the glycosylation patterns of the compounds.  相似文献

The reaction of [FeII(H2O)6](BF4)2 with tris(2-pyridylmethyl)amine (TPyA) and triethylamine in methanol under aerobic conditions forms [(TPyA)FFeIIIOFeIIIF(TPyA)](BF4)2 · 0.5MeOH (1), in which each Fe(III) ion is coordinated to a TPyA and an F ion as well as an oxo ion (O2−) linking two Fe(III) ions. 1 has offset face-to-face π-π interactions between the dimers, and possesses a supramolecular network structure. The magnetic susceptibility of 1 can be fit with g = 2.0, J/kB = − 153 K (106 cm−1), and θ = − 0.3 K [H = − 2JSa · Sb]. These indicate that very strong antiferromagnetic interactions occur via the oxo bridge within the Fe(III) dimer and weak antiferromagnetic interactions between the dimers.  相似文献
