The development and biological characteristics of Haemaphysalis longicornis were investigated under field conditions in Xiaowutai National Natural Reserve Area, North China. Unfed larvae, nymphs and adults were fed on rabbits and exposed to daylight. Three free-living stages were allowed to develop in field plot selected in a tick natural habitat. The host seeking behavior and seasonal occurrence were observed. Haemaphysalis longicornis were active from mid March to mid October. The premoult period of nymphs and preoviposition of females were regulating phases of their life cycle. The developmental durations of eggs, larvae and adults were constant under field conditions regardless when the development started. The oviposition periods in May and June were statistically shorter than those in July and August. The daily oviposition patterns of females engorged in May and June demonstrated unobvious peak, which differed from those engorged in July and August. The daily oviposition peak of the latter occurred on the 4th day of oviposition. Moreover, a positive correlation was found between the mass of the laid egg and the body weight of engorged females (r = 0.62, P < 0.001). The female reproductive efficiency indices were 2.9, 6.1, 10.5 and 9.0 in May, June, July and August, respectively. The mean weight (3.33 mg) of engorged nymphs molting to females was significantly higher than that (2.35 mg) of those molting to males (P < 0.001), but the body weights of both sexes were overlapping.  相似文献

Summary The effect of water immersion on the non-parasitic stages of Boophilus microplus were investigated both under laboratory and field conditions. Survival of engorged female ticks was influenced by their age and the temperature of the water. Eggs and larvae were more resistant to submerison than were engorged females and their survival was increased at low temperatures and in water with high oxygen content. The persistence of this tick in areas prone to flooding was attributed to the survival of parasitic stages and to larvae on the pasture. Heavy rains produce favourable pasture conditions for tick reproduction so that a large increase in population size may be expected to follow such rain.  相似文献

The susceptibility of the tick Boophilus microplus to Beauveria bassiana was evaluated by inoculating eggs, larvae and engorged females of the tick with five fungal isolates at concentrations of 106, 107 and 108 conidia/ml. Tick eggs (0.25 g) were immersed in 1 ml of a suspension of the different conidial concentrations for 1 min. Similar exposure was performed by immersion of 2000 larvae and homogeneous groups of nine engorged females in 2 and 20 ml of conidial suspension, respectively. Treated eggs, larvae and adults were placed in an incubator at 27 ± 1 °C and relative humidity above 80% for evaluation of the fungal action. All fungal isolates applied at all conidial concentrations reduced the hatching rate of larvae from treated eggs by 1.36–65.58% and increased the mortality rate of inoculated larvae by 0.8–70.49%. In the bioassay with engorged females, oviposition period was reduced by 9.69–47.80%, egg mass weight by 4.71–53.87%, estimated reproduction by 8.3–60.62%, egg production index by 5.03–54.20%, percent larval hatching by 0.27–13.96%, and the mortality rate of treated females was increased by 96.60–100%. The reduction of the estimated reproduction obtained for the treated groups ranged from 8.37 to 64.52%. The sporulation of the pathogen on dead females ranged from 3.70 to 88.88% depending on the isolate and concentration used. Isolates AM 09, CB 7 and JAB 07 were the most effective and effectiveness increased with increasing concentrations of conidia in the suspensions.  相似文献

Estimates of seasonal abundance of larvae, nymphs and females of the paralysis tick, Ixodes holocyclus, were obtained by collecting ticks that engorged on small mammals and birds trapped in two localities in southeastern Queensland: Brisbane (wet sclerophyll forest) and Tamborine Mountain (cleared rain forest). The long-tailed short-nosed bandicoot, Isoodon macrourus, was the most common mammal trapped but small numbers of other marsupials, rodents and ground frequenting birds were also captured. Small numbers of five other tick species were also collected. In both habitats there was clearly one dominant generation of I. holocyclus;7er year, although the presence of all stages at most times of the year indicated overlapping of smaller cohorts. Females were most abundant in spring and early summer, larvae in summer-autumn, and nymphs in autumn-winter. I. holocyclus was abundant on I. macrourus and rare on most other mammals and birds captured. At the peak of abundance of each instar, each bandicoot dropped from 500 to 2000 engorged larvae, 100 to 200 engorged nymphs, and four to six engorged females. Life tables were compiled for the tick in both habitats and these indicate that there was relatively high survival from engorged larva to engorged nymph and thence to engorged female and that most mortality occurred between detaching of the engorged female and the detaching of the engorged larva. The tick was more abundant on bandicoots from cleared rain forest and rain forest edge, than on those from sclerophyll woodland. The survival of engorged larvae and nymphs of I. holocyclus and the larval productivity of engorged females were examined in a warm moist climate where the tick was abundant (Tamborine Mountain) and in a hotter dryer climate where the tick was rare or absent (Amberley). In both localities engorged larvae and nymphs survived to the next instar in all seasons of the year. On most occasions engorged females produced eggs which hatched. Mature bandicoots from tick infested areas showed little or no resistance to infestation with larvae or nymphs of I. holocyclus, whereas other small mammals from the same area showed an appreciable degree of resistance to the immature stages of the tick. Feeding larvae and nymphs exposed to normal light-dark cycles in the laboratory detached during the afternoon and early evening. This behaviour and host resistance are discussed in relation to the daily activity cycles of host species, their habitat preferences, and their role as hosts for I. holocyclus.  相似文献

The life cycle of Ixodes rubicundus, the Karoo paralysis tick, was studied under field conditions in the south-western Orange Free State, South Africa, by placing freshly engorged ticks in small containers. The life cycle extends over 2 years. The two regulating phases in the life cycle, which undergo a morphogenetic diapause during the hot and dry summer months, are the egg and engorged nymph. Possible behavioural diapause in adults which suppresses questing activity before the end of March. can also serve as a third regulating phase. Temperature affects the duration of the pre-oviposition period of engorged females and the period between detachment of engorged larvae from hosts and ecdysis. Commencement of larval hatch is reasonably synchronized, irrespective of the month during which oviposition occurred. Peak activity periods of larvae (April or May) occur during a period of high rainfall and decreasing daily maximum temperatures. The period between detachment of engorged nymphs from hosts and ecdysis is highly variable (8–36 weeks). All developmental stages of the tick occur mainly during autumn and winter and no ticks are active during the hot summer months of December to February. Larvae, nymphs and adults each survive for only one season.  相似文献

The physiological effects of the winter tick,Dermacentor albipictus, on moose,Alces alces, were investigated. Blood composition, weight gain, food intake and change in the hair coat of moose calves, four infested withD. albipictus larvae, and eight uninfested, were monitored. Infested moose groomed extensively, apparently in response to feeding nymphal and adult ticks, and developed alopecia. Other clinical signs included: chronic weight loss, anemia, hypoalbumenemia, hypophosphatemia, and transient decreases in serum asparate transaminase and calcium during the period of nymphal and adult female tick engorgement. Infested animals did not become anorexic. Two moose with severe hair loss had increases in gamma globulin shortly after the onset of female tick engorgement. Results suggest that alopecia is associated with tick resistance. Animals that groom and develop hair loss likely carry fewer ticks and therefore suffer less severely from blood loss.  相似文献

During experimental Rhipicephalus (Boophilus) microplus (Acari: Ixodidae) infestation on cattle, approximately 5% of engorged female ticks showed symptoms of bacterial infection. The affected ticks were unable to oviposit, secreted a distinctive yellow exudate through the genital orifice and eventually died. Microscopic analysis of tick exudate showed abundant clusters of Gram-positive cocci bacteria that were isolated and cultured on bacteriological medium. Biochemical phenotyping and 16S rRNA ribotyping analysis on cultured bacteria identified it as Staphylococcus saprophyticus. This species was also isolated from healthy tick larvae, indicating that S. saprophyticus is commonly found in ticks during different developmental stages. However, conspicuous symptoms are only found on fully engorged females. Cultured S. saprophyticus induced identical pathological symptoms when the bacteria were experimentally inoculated into healthy ticks, demonstrating it to be the causative agent of the R. microplus infectious lethal disease described in this work.  相似文献

The objective of this study was to demonstrate the occurrence of entomopathogenic fungi on Ixodes ricinus ticks in relation to the tick stage, engorgement and season. Ticks were collected from the vegetation, from small rodents and from deer. All entomopathogenic fungi found belonged to the Hyphomycetes. Paecilomyces farinosus and Verticillium lecanii were the predominant species. Other species, found only on engorged females were: Beauveria bassiana, B. brongniartii, P. fumosoroseus and V. aranearum. Eight out of 1833 ticks collected from the vegetation and three out of 269 engorged nymphs were infected with fungi. Thirty-three out of 149 engorged females were infected, whereas males and engorged larvae were not infected. Throughout the season, a significantly higher proportion of ticks collected in autumn were infected. Entomopathogenic fungi may have a significant impact on the size of the I. ricinus population, since females were the most frequently infected stage.  相似文献

Earlier studies demonstrated that Metarhizium brunneum, usually a broad-host pathogen of arthropods, is unable to complete its pathogenic life cycle when inoculated on the fungus-resistant tick, Hyalomma excavatum engorged females. While the fungus penetrates the cuticle of fungus-susceptible tick, Rhipicephalus annulatus females, it is unable to penetrate the cuticle of fungus-resistant tick, and even perishes on its surface. This is probably due to high concentration of antifungal fatty acids and probably also due to a hypersensitive-like response of the tick. To understand the metabolic pathways occurring in the fungal hyphae upon encountering the cuticles, we compared the response of the fungus to cuticle from susceptible and resistant tick cuticles by 2D-gels. The intracellular proteomes of M. brunneum Mb7 exposed to cuticle of the fungus-susceptible tick, R. annulatus, and to the fungus-resistant tick, H. excavatum engorged females were compared after exposure to either cuticles. By means of liquid chromatography-mass spectrometry/mass spectrometry we identified in both proteomes common proteins involved in biological processes as well as unique proteins identified only in the proteome of fungus exposed to fungus-resistant tick cuticle. These proteins were identified in high probability as heat shock proteins, four key enzymes of the glyoxylate cycle, and proteins associated with hypoxia, and exposure to antifungal drugs. These findings are discussed within the M. brunneum-tick pathosystem in relation to tick resistance and host resistance in general.  相似文献

Seasonal abundance and activity of all the three post-embryogenic stages of Haemaphysalis longicornis, both feeding and free-living phases, were evaluated over a period of 2 years, from February 2008 to January 2010, in North China. Feeding ticks were removed weekly from head and ears of domestic sheep and the attachment sites of this tick were assessed coinstantaneously; free-living ticks were collected weekly in four habitat types by flag-dragging. The results suggested that H. longicornis mainly resides in shrubs and completes one generation per year with population attrition between stages. Infestation of nymphs was detected from March to September with highest peak between late April and early May; adults were detected from April to September with highest peak between late June and July, and an overwintering male population was found during late September to March; infestation of larvae was observed from June to October and peaked between middle August and early September. Most of this tick (91%) attached to head and ears of hosts. Additionally, we captured rodents from April to September 2008, but only a negligible number of nymphs were detected. This result suggested that rodents are not the principal hosts for this tick in the study area.  相似文献

The brown dog tick Rhipicephalus sanguineus is one of the most important tick species in both veterinary and human medicine. Studies on the biology of this tick are pivotal in order to develop improved control strategies. The brown dog tick is a gregarious species, tending to form large clusters in highly infested environments. This is the case of engorged females of R. sanguineus that often tend to aggregate in the environment. In the present study, we assessed whether aggregation affects R. sanguineus females in the laboratory. Engorged females of R. sanguineus were separated in six groups, each with four replicates. Six groups (G1–G6) were composed of single, five, 10, 20, 40 or 60 females, for a total of 544 females. We found that aggregation (expressed as tick density within Petri dishes) affected egg production efficiency of females and incubation period, but not egg hatching. In particular, there was a strong negative correlation between tick density and the following parameters: egg mass weight, incubation period, egg production efficiency, reproduction efficiency index, and reproductive fitness index. However, for the maintenance of tick colonies, engorged females can be maintained in groups (density up to 0.4/cm2) without major prejudices in terms of egg production and hatch rate. This information might be useful for those interested in maintaining large tick colonies, as grouping engorged females might help to save space in incubators and, most importantly, to keep the genetic heterogeneity within the tick colony, across generations.  相似文献

In this study, engorged Amblyomma lepidum ticks were found to drop off in two peaks, one in the morning and one in the evening. Most larvae and females engorged during the morning hours between 06.00 h and 10.00 h with a peak around 08.00 h, whereas the majority of the nymphs dropped in the evening between 18.00 h and 24.00 h with the peak around 22.00 h. Although the effect of time on drop-off patterns of the ticks was statistically significant (p≤ 0.001), there were no significant seasonal influences. Survival of unfed stages of A. lepidum was also studied and was found to increase from larvae to adult ticks. The longest survival periods of 10, 11 and 14 weeks were recorded during the wet season for larvae, nymphs and adults, respectively. It is concluded that environmental conditions required for survival of A. lepidum are optimal only during the wet season and that during other seasons the tick depends primarily on prevailing micro-climatic conditions for its survival.  相似文献

Biological data of three generations of Amblyomma tigrinum in the laboratory are reported and the suitability of different host species for immature ticks are compared. Grouping the three generations, infestations by both the larval and nymphal stages were performed on chickens (Gallus gallus), wistar rats (Rattus norvegicus), rabbits (Oryctolagus cuniculus),wild mice (Calomys callosus), dogs (Canis familiaris) and opossums (Didelphis albiventris). Only dogs were used for infestations by adult ticks. Tick developmental periods were observed in an incubator at 27°C and RH 90%. The proportion of engorged larvae recovered from chickens (21.7% of the exposed larvae) was significantly larger (p<0.001) than those from the five mammal species used in the infestations (maximum of 3.1%). A significant larger (p<0.01) proportion of engorged larvae successfully molted after being fed on chickens than on mammal hosts. The proportion of engorged nymphs recovered from chickens (28.8% of the exposed nymphs) was significantly larger (p<0.001) than those from mammal hosts (range: 0–2.1%). Larvae showed similar feeding periods on exposure to different host species, except for those larvae fed on C. callosus, which showed significantly longer (p<0.001) feeding periods. Engorged larvae detachment peaked on the 5th feeding day, followed by the 6th day, on all hosts except for C. callosus. Larval premolt periods were similar for engorged ticks exposed to different host species, except for larvae fed on dogs, which showed significantly longer (p<0.001) premolt periods. Host detachment of engorged nymphs peaked on the 6th feeding day on chickens. Although nymphal detachment on rats peaked on the 8th day, only 15 nymphs were recovered from this host species. In a sample of 144 F3 nymphs fed on chickens no significant difference (p>0.10) was found between the feeding or premolt periods of 82 males and 62 females, but female nymphs were significantly heavier (p<0.005) than male nymphs. Sixteen engorged females (61.5% of the exposed ticks) were recovered after being fed on dogs, and all these females laid viable eggs. Chickens, the only avian host, were the most suitable host when compared with the five mammal species. Dogs were demonstrated to be a suitable host for adults of A. tigrinum, which is consistent with, several reports of adult A. tigrinum ticks parasitizing dogs in different areas of South America. Our results reinforce that in these same areas avian species are the major hosts for immature stages of this tick species. The objective of the present work was to evaluate the efficacy of a non‐sulphur‐based homeopathic preparation and a sulphur‐containing natural product derived from Allium sativum (Linnaeus) against infestation by the cattle tick Rhipicephalus (Boophilus) microplus (Canestrini) (Acari: Ixodidae). A total of 24 crossbred calves (7 : 8 Holstein : Zebu), aged 6–8 months and maintained in individual stables under tick‐free conditions, were divided into three groups. Group 1 (control group) received no treatment; Group 2 was treated with 0.01 g/day of the homeopathic preparation Fator C&MC®, and Group 3 was treated with 20 g/day of Enxofre‐Allium sativum®. After adaptation to the diet for 1 month, each calf was subjected to artificial infestation with 8000 R. (B.) microplus larvae (aged 7–14 days) twice per week over a 5‐month period. Numbers of engorged females were recorded every 14 days and samples of freely released engorged females were collected at 14‐day intervals commencing 3 months after the start of the experiment. The engorged females were weighed, incubated for 15 days under biochemical oxygen demand conditions at 27 ± 1 °C and relative humidity >85%, and the weights of the egg masses produced were recorded. Other biological parameters, including reproduction estimate, reduction in oviposition and efficiency of treatment, were determined. A significant reduction in the number of engorged females was detected on animals treated with Enxofre‐Allium sativum® (Group 3) in comparison with the other two groups. The overall efficiency of the treatment with the sulphur‐containing product was 64%, whereas that of the homeopathic preparation was 26%. Under the experimental conditions established, Enxofre‐Allium sativum® can reduce the intensity of the R. (B.) microplus infestation.  相似文献

The use of RNA interference (RNAi) to assess gene function has been demonstrated in several three-host tick species but adaptation of RNAi to the one-host tick, Boophilus microplus, has not been reported. We evaluated the application of RNAi in B. microplus and the effect of gene silencing on three tick-protective antigens: Bm86, Bm91 and subolesin. Gene-specific double-stranded (dsRNA) was injected into two tick stages, freshly molted unfed and engorged females, and specific gene silencing was confirmed by real time PCR. Gene silencing occurred in injected unfed females after they were allowed to feed. Injection of dsRNA into engorged females caused gene silencing in the subsequently oviposited eggs and larvae that hatched from these eggs, but not in adults that developed from these larvae. dsRNA injected into engorged females could be detected by quantitative real-time RT-PCR in eggs 14 days from the beginning of oviposition, demonstrating that unprocessed dsRNA was incorporated in the eggs. Eggs produced by engorged females injected with subolesin dsRNA were abnormal, suggesting that subolesin may play a role in embryonic development. The injection of dsRNA into engorged females to obtain gene-specific silencing in eggs and larvae is a novel method which can be used to study gene function in tick embryogenesis.  相似文献

We conducted laboratory and field experiments to elucidate the life history of Ixodiphagus hookeri, a parasitoid of the ixodid tick Amblyomma variegatum in Western Kenya. Ixodiphagus hookeri females oviposited in unfed host nymphs as well as engorged nymphs, but rarely in engorged larvae. While I. hookeri developed to adults in engorged nymphs, the eggs laid in unfed nymphs disappeared within 2 days after oviposition. As temperature increased, development time of I. hookeri from oviposition to adult emergence in engorged nymphs decreased from 46 days at 23 °C to 35 days at 28 °C, and their immature survival in engorged nymphs decreased from 67% at 23 °C to 22% at 28 °C. No parasitoid adult emerged from hosts at 30 °C. Individual hosts parasitized by single females produced 42–53 adult wasps, 73% of which were females. As a typical pro-ovigenic species, I. hookeri females had an average of 84 mature eggs at emergence and lived only for a few days. When laboratory-reared, unfed nymphs of A. variegatum were attached to cattle for 4–9 days in subsistence farmers’ fields in Western Kenya, 25% of the engorged nymphs and 4% of the unfed nymphs on cattle were parasitized by I. hookeri, demonstrating that I. hookeri females search for and oviposit in A. variegatum nymphs on cattle. Unlike other strains of I. hookeri that overwinter as eggs in unfed nymphs, I. hookeri could continuously reproduce throughout the year in Western Kenya.  相似文献

The one-host biological north form / ecotype of Hyalomma scupense Schulze, 1919 (Acari, Ixodidae) is reported for the first time in Turkey herein. Following the first detection of the tick, a longitudinal field study was carried out to fill gaps in the data concerning its biological features. This study also aimed to determine how the monthly activation dynamics of this relatively cold climate-adapted species is characterized under the influence of the warm summer subtype of the Mediterranean climate. During this study, which was carried out on a monthly basis in 2014, H. scupense was found on cattle from 5 out of 18 villages screened in Thrace (the European part of Turkey). The field study revealed that i) this north ecotype of H. scupense exhibits winter one-host behavior beginning in October (with larval stages) and ending in April (with engorged adults); ii) engorged females detach from the cattle, drop on the floors of barns during night and accumulate on piled bedding contaminated with slurry manure; iii) in the late spring, engorged females lay eggs, and larvae hatch in the same area; and iv) larvae become active in autumn as the weather grows cooler. The results indicated that although one-host H. scupense is known to be distinctly adapted to cold conditions, it can also be effectively established in relatively temperate regions and complete its life cycle with some modifications in the timing of its monthly activation dynamics.  相似文献

Increases were recorded of lengths of larvae, nymphs and females of the tick, Haemaphysalis (Kaiseriana) longicornis during feeding on cattle, so that categories could be established for estimating the ages of ticks in natural populations. Most larvae and nymphs engorged in 4 days and females in 5 or 6 days. Four age categories were defined for larvae and 3 for nymphs. The growth of females was variable but 4 stages of physiological development were defined based on changes in their lengths. The lengths were determined of each instar 24 h before detachment to enable estimation of the numbers maturing each day on cattle in the field.  相似文献

A colony ofAmblyomma parvum was started with engorged females collected from cattle in the Province of Salta (25°01 S, 63°56 W), Argentina. The progeny of those ticks were fed on rabbits and the non-parasitic stages maintained at 27±1°C, 83–86% RH in darkness. The life cycle (prefeeding period not evaluated) had a mean duration of 99.6 days. The mean time (days) for the different phases of the cycle were as follows: feeding period of females, 8.0; pre-oviposition period, 5.7; oviposition period, 17.5; minimum incubation period of the eggs, 31.8; feeding period of larvae, 3.2; premoult period to nymphs, 10.9; feeding period of nymphs, 4.7; premoult period to adults, 17.8. The oviposition pattern was typical of an ixodid tick, including a linear relationship between weights of engorged females and the number of eggs laid (r=0.8659). The males increased 18% in weight after feeding on hosts (P<0.01). The mean recovery rates of larvae, nymphs and females were 28.2%, 95.3% and 90.7%, respectively. The nymphs moulting to females were heavier (6.8±0.69 mg) than those moulting to males (3.2±0.29 mg) (P<0.01). An artificial membrane system was adapted to feed Ornithodoros turicata (Ixodida: Argasidae) larvae from a laboratory colony using defibrinated swine blood. Aspects related to larval feeding and moulting to the first nymphal instar were evaluated. A total of 55.6% of all larvae exposed to the artificial membrane in two experimental groups fed to repletion and 98.0% of all fed larvae moulted. Mortality rates of first instar nymphs differed significantly depending on the sorting tools used to handle engorged larvae (χ2 = 35.578, P < 0.0001): engorged larvae handled with featherweight forceps showed significantly higher mortality (odds ratio = 4.441) than those handled with a camel‐hair brush. Differences in the physical properties of the forceps and camel‐hair brush may affect the viability of fragile soft tick larvae even when care and the same technique are used to sort them during experimental manipulations. The current results represent those of the first study to quantify successful feeding to repletion, moulting and post‐moulting mortality rates in O. turicata larvae using an artificial membrane feeding system. Applications of the artificial membrane feeding system to fill gaps in current knowledge of soft tick biology and the study of soft tick–pathogen interactions are discussed.  相似文献
