The combined effects of water activity (aw) and temperature on mycotoxin production by Penicilium commune (cyclopiazonic acid — CPA) and Aspergillus flavus (CPA and aflatoxins — AF) were studied on maize over a 14-day period using a statistical experimental design. Analysis of variance showed a highly significant interaction (P 0.001) between these factors and mycotoxin production. The minimum aw/temperature for CPA production (2264 ng g–1 P. commune, 709 ng g–1 A. flavus) was 0.90 aw/30 °C while greatest production (7678 ng g–1 P. commune, 1876 ng g–1 A. flavus) was produced at 0.98 aw/20 °C. Least AF (411 ng g–1) was produced at 0.90 aw/20 °C and most (3096 ng g–1) at 0.98 aw/30 °C.  相似文献

Epicoccum nigrum conidia were produced by solid fermentation on wheat grains (cv. Rendeveaux and Brigadier) at different water activities (aw). Conidial production was highest at high aw(0.996) than at reduced aw (0.98). However, conidial production at reduced aw was improved when the aw of the substrate was adjusted with a mixture of glycerol and water. Maximum levels ofconidiation were 7–11 × 106 conidia g−1 grain. The aw of the solid substrate affected the pattern of accumulation of compatible solutes in the conidia. Mannitol was the main polyol in all conidialtypes. However, the amounts of mannitol were higher in conidia produced at high aw. At reduced aw the conidia of E. nigrum accumulated moreglycerol, which is more efficient in the osmorregulation proccess than mannitol. Arabitol accumulated in low amounts, specifically in conidia produced at the lower aw, on cv. Rendeveaux but not on cv. Brigadier. Trehalose was detected in higher amounts in cv. Rendeveaux than in cv. Brigadier, andthe amounts were higher in conidia produced at high aw. A significant amount of endogenous solutes was detected in the washing liquid used for the separation of the conidia. The effects of growth phase, reductions in the water activity (a w) of the growth medium and mild desiccation on the composition and the degree of unsaturation of cellular fatty acids (CFA) of Sinorhizobium meliloti, Bradyrhizobium elkaniiand Bradyrhizobium japonicum were studied. During the course of growth, an interchange of cis-vaccenic with lactobacillic acid and a slight increase in palmitic acid were observed while other fatty acids remained constant. The degree of unsaturation was significantly higher in the exponential phase of growth. Reductions in the a w of the medium led to an increase in lag phase, a reduction in growth rate and maximal optical densities (OD) in stationary phase cells. A decrease in the degree of unsaturation of CFA was also observed as the a w was reduced from 0.999 to 0.969 and after desiccation to 83.5% relative humidity (R.H.). The changes in the degree of unsaturation of CFA observed after growth at reduced a w may be one of the pre-adaptation steps to endure more severe desiccation.  相似文献

Transesterification activity and the industrial potential of a novel lipase prepared from Acinetobacter ventiatus RAG-1 were evaluated. Purified lipase samples were dialyzed against pH 9.0 buffer in a single optimization step prior to lyophilization. The enzyme and organic phase were pre-equilibrated (separately) to the same thermodynamic water activities (a w) ranging from a w 0.33 to 0.97. Production of 1-octyl butyrate by lipase-catalyzed transesterification of vinyl butyrate with 1-octanol in hexane was monitored by gas chromatography. Production of 1-octyl butyrate and initial rate of reaction depended on water activity. Product synthesis and rate of transesterification increased sharply with increase from a w 0.33 to 0.55. Highest product concentration (218 mM) and rate of reaction (18.7 μmol h−1 · 10 μg protein) were measured at a w 0.86. Transesterification activity in hexane represented 32% of comparable hydrolytic activity in aqueous buffer.  相似文献

Effect of water activity (a w) adjusted with NaCl or glycerol on the growth and metabolism ofLactobacillus helveticus var.pragensis at 40°C was demonstrated by growth curves (generation time) and changes in the degree of acidity of a milk medium expressed as lactic acid content. NaCl-regulateda w of 0.970 inhibited growth and acid production completely. The same, glycerol-regulateda w of 0.970 increased the generation time only from 33 min (a w=0.994) to 67 min which was less than the generation time at the highera w of 0.982 adjusted with NaCl (103 min). Glycerol-regulateda w of 0.970 did not decrease the acid production (2.6% lactic acid). Ata w of 0.951, the acid production was decreased by 39% compared with the values found in milk media with the original, unadjusteda w of 0.994, after the same time of incubation. Media witha w adjusted to the same values with NaCl or glycerol do not influence the growth and acid production ofL. helveticus in the same way. In contrast to glycerol, NaCl has a strong effect.  相似文献

Summary The effect of water activity (aw) on the growth and end-product formation of Lactobacillus viridescens SMRICC 174, Lactobacillus SMRICC 173 (homofermentative) and Brochothrix thermosphacta ATCC 11509T was studied. All strains orginated from meat or meat products. The aw was adjusted in the range 0.94–0.99 with NaCl or glycerol. A greater reduction in growth rates was found for L. viridescens and B. thermosphacta when aw was regulated with NaCl rather than with glycerol, the opposite was true for Lactobacillus 173. L. viridescens grew at aw >-0.94. At 0.94 aw B. thermosphacta was totally inhibited when NaCl was the solute and Lactobacillus 173 when glycerol was the solute. Only minor variations in the end-product formation of the Lactobacillus spp. were found at different aw values. In aerobic culture B. thermosphacta produced less l-lactic acid and more acetic acid as the aw was decreased with NaCl, while the yields were unaffected when glycerol was used.  相似文献

Stomatal responses to leaf temperature (Tl) and to the mole fractions of water vapour in the ambient air (wa) and the leaf intercellular air spaces (wi) were determined in darkness to remove the potential effects of changes in photosynthesis and intercellular CO2 concentration. Both the steady‐state and kinetic responses of stomatal conductance (gs) to wa in darkness were found to be indistinguishable from those in the light. gs showed a steep response to the difference (Δw) between wa and wi when wa was varied. The response was much less steep when wi was varied. Although stomatal apertures responded steeply to Tl when Δw was held constant at 17 mmol mol?1, the response was much less steep when Δw was held constant at about zero. Similar results were obtained in the light for Δw = 15 mmol mol?1 and Δw ≈ 0 mmol mol?1. These results are discussed in the context of mechanisms for the stomatal response to humidity.  相似文献

 Leaf features were examined in three Quercus species (Q. coccifera, Q. ilex and Q. faginea) along a steep rainfall gradient in NE Spain. The analyzed leaf traits were area, thickness, density, specific mass, leaf concentration of nitrogen, phosphorous, lignin, cellulose and hemicellulose, both on a dry weight basis (Nw, Pw, Lw, Cw, Hw) and on an area basis (Na, Pa, La, Ca, Ha). These traits were regressed against annual precipitation and correlated with each other, revealing different response patterns in the three species. Q. faginea, a deciduous tree, did not show any significant correlation with rainfall. In Q. coccifera, an evergreen shrub, Nw, Na, Lw, La and Ca increased with higher annual rainfall, while Hw decreased. In Q. ilex, an evergreen tree, leaf area, Pw and Lw increased with precipitation, whereas specific leaf mass, thickness and Ha showed the reverse response. Correlations between the leaf features revealed that specific mass variation in Q. faginea and Q. coccifera could be explained by changes in leaf density, while in Q. ilex specific leaf mass was correlated with thickness. Specific leaf mass in the three species appeared positively correlated with all the chemical components on a leaf area basis except with lignin in Q. ilex and with P in Q. ilex and Q. faginea. In these two tree species Pw showed a negative correlation with specific leaf mass. It is suggested that each species has a different mechanism to cope with water shortage which is to a great extent related to its structure as a whole, and to its habit. Summary Glycerol and arabitol were the main polyols accumulated by Zygosaccharomyces rouxii in continuous culture but the intracellular and extracellular concentrations of the polyols varied with the dilution rate and osmoticum used to adjust the water activity (aw) to 0.960. When the aw was adjusted with NaCl, glycerol was the main polyol accumulated intracellularly whereas glycerol and arabitol were accumulated when polyethylene glycol (PEG) 400 was used. The extracellular glycerol and arabitol concentrations at 0.960 aw (NaCl or PEG 400) were similar or decreased relative to cultures at 0.998 aw. Compared to steady-state cultivation at 0.998 aw, the yeast retained at 0.960 aw (NaCl or PEG 400) a greater proportion of the total glycerol intracellularly against an increased concentration ratio without significantly greater production of glycerol. Arabitol was only significant in osmoregulation when cultivated at 0.960 aw (PEG 400). The intracellular glycerol concentration was insufficient to balance the aw across the membrane, but an equilibrium could be achieved under certain conditions if arabitol was also osmotically active. The effect of the rapid reduction of the water activity (aw) on the extracellular protein and amylolytic activity of Aspergillus niger was studied. An aw value gradient from 0.90 to 0.99 in KCl solutions was applied for the mycelium treatment. It was found that the aw reduction considerably influenced the protein secretion. This phenomenon was dependent on the age of the treated mycelium and the range of the aw gradient. The highest protein and enzyme secretion yields were obtained at aw = 0.98 using a 72-h old mycelium. In comparison with the non-treated mycelium, the increase in the secretion amounted to about 60% for the amylolytic activity and 37% for the soluble protein, respectively. It was shown that the mycelium incubated in KCl solutions of an aw value from 0.90 to 0.99 had the ability for regeneration in fresh CZAPEK-DOX medium. The effect of the osmotic shock on the protein secretion was limited only for the treated cell population and declined in the mycelium which was regenerated after the transfer into the culture medium.  相似文献

Water-use strategies of Populus tremula and Tilia cordata, and the role of abscisic acid in these strategies, were analysed. P. tremula dominated in the overstorey and T. cordata in the lower layer of the tree canopy of the temperate deciduous forest canopy. Shoot water potential (), bulk-leaf abscisic acid concentration ([ABA]leaf), abscisic acid concentration in xylem sap ([ABA]xyl), and rate of stomatal closure following the supply of exogenous ABA (v) decreased acropetally through the whole tree canopy, and foliar water content per area (w), concentration of the leaf osmoticum (c), maximum leaf-specific hydraulic conductance of shoot (L), stomatal conductance (gs), and the threshold dose per leaf area of the exogenous ABA (da) required to reduce stomatal conductance increased acropetally through the tree canopy (from the base of the foliage of T. cordata to the top of the foliage of P. tremula) in non-stressed trees. The threshold dose per leaf dry mass of the exogenous ABA (dw) required to reduce stomatal conductance, was similar through the tree canopy. After a drought period (3 weeks), the , w, L, gs, da and dw had decreased, and c and v had increased in both species. Yet, the effect of the drought period was more pronounced on L, gs, da, dw and v in T. cordata, and on , w and c in P. tremula. It was concluded that the water use of the species of the lower canopy layer—T. cordata, is more conservative than that of the species of the overstorey, P. tremula. [ABA]leaf had not been significantly changed in these trees, and [ABA]xyl had increased during the drought period only in P. tremula. The relations between [ABA]leaf, [ABA]xyl and the stomatal conductance, the osmotic adjustment and the shoot hydraulic conductance are also discussed.  相似文献

This study examined six strains of Beauveria bassiana s.l. and Isaria farinosa, one strain of Isaria fumosorosea and five strains of Metarhizium anisopliae s.l. to identify the ability for (1) growth and (2) sporulation under interacting environmental factors of water activity (aw) and temperature stress. Growth on Sabouraud Dextrose Agar (SDA; water activity, aw = 0.995) or SDA modified with glycerol to 0.98, 0.96 and 0.94 aw was measured at four different temperatures (25, 30, 35 and 37°C). All M. anisopliae strains grew at 25–35°C and 0.995 aw while only two strains tolerated extreme water stress at 0.94 aw.Three strains of B. bassiana were able to grow at 25–37°C and 0.995 aw. Only one strain of I. farinosa was able to grow at 25–37°C and 0.995 aw. Aw and temperature interactions resulted in different strain-dependent responses, in terms of growth and sporulation. Only one strain of I. farinosa and three of M. anisopliae grew at 0.94 aw and none of the B. bassiana strains tolerated such water stress. At 0.96 and 0.94 aw and 35–37°C, sporulation by all the strains of the three species were significantly affected. Under elevated temperatures and drought stress, very few of these strains of entomopathogenic fungi are able to grow and sporulate. Indeed, the B. bassiana strains were unable to tolerate the extreme conditions examined. Resilience to such abiotic interactions is critical for selecting strains for formulations. Tolerance to water and temperature stress could be good criteria for selection of strains with secondary spread potential for use as part of an integrated pest management system where secondary cycling may be important, especially in sub-tropical and tropical environments.  相似文献

The effect of water availability (water activity,aw; 0.995–0.90) and temperature (18–30 °) on in vitro interactions between an ochratoxin producing strain of Aspergillus ochraceus and six other spoilage fungi was assessed in dual culture experiments on a maize meal-based agar medium. Inprimary resource capture of nutrient substrate, A. ochraceus was dominant against many of the interacting species, being able to overgrow and replace A. candidus, and sometimes A. flavus and the Eurotium spp. regardless of aw or temperature. However, with freely available water (0.995 aw) A. alternata and A. niger were dominant, with mutual antagonism between A. ochraceus and A. flavus at 25–30 °C. In the driest conditions tested (0.90 aw) there was also mutual antagonism between A. ochraceus and the two Eurotium spp. Overall, under allconditions tested the Index of Dominance for A. ochraceus was much higher than for other competing species combined suggesting that A. ochraceus wasa good competitive colonist able to replace a numberof other species. However, the growth rate ofA. ochraceus was modified and decreased by the interaction with competitors. Interaction between A. ochraceus and species such as A. alternata (18°C/0.995) and Eurotium spp. (0.995–0.95 and 25–30 °C) resulted in a significant stimulation of ochratoxin production. Theresults are discussed in relation to the effect that environmental factors have on the possible competitiveness of A. ochraceus in the maizegrain ecosystem and the role of ochratoxin in nicheexclusion of competitors. Colonisation of barley grain has been studied during storage at different water contents and with and without restriction of the air supply to simulate conditions in unsealed silos. Grain stored with a water activity >0·9 aw (20% water content) heated spontaneously when aeration was unrestricted, the maximum temperature attained increasing with aw to 58 °C at 1·0 aw (39% water content). The presence of many unripe grains increased the tendency to heat at a given mean water content. Although heating was prevented by sheeting to restrict the air supply, it could occur subsequently when the sheeting was removed. Both heating and restriction of the air supply were associated with increased carbon dioxide (to >25%) and decreased oxygen concentrations (to <5%). Germination of grain after 6·9 months storage was correlated with aw; germination levels approaching 100% were retained only at about 0·7 aw (13·5% water content). Colonisation by Aspergillus species was correlated with aw and temperature and similar correlations with Penicillium species were also found, with P. verrucosum var. cyclopium abundant at 0·85-0·90 aw (17·20% water content) and P. piceum, P. funiculosum and P. capsulatum at 0·90-0·95 aw (20–25% water content). In sealed containers P. roquefortii occurred at 1·00 aw (39% water content) and P. hordei at 0·90-0·92 aw (20–22% water content). Spores of fungi and actinomycetes formed during spontaneous heating of grain survived 6 months sealed storage although Absidia corymbifera and Micropolyspora faeni may have declined in numbers. Fungicides applied to the ripening grain had only limited effect on the colonisation of the grain during storage.  相似文献

Summary During tempeh fermentation, Rhizopus oligosporus produced polysaccharidases to degrade soya bean cell walls; the maximum activity for all polysaccharidases tested occurred 20–30 h after inoculation. R. oligosporus was also grown in a soya bean extract model medium to which glycerol was added to control water activity (a w). The overall activities of the major enzymes produced by the fungus, polygalacturonase, endocellulase and xylanase, appeared to be strongly influenced by a w. The production of enzymes as well as their specific activities were affected by a w. The optimum a w for polygalacturonase and xylanase activity coincided with that for mycelial growth, namely 0.99–1.00. Summary The Vmax and KM of various forms of lipase from Pseudomonas cepacia (powder, adsorbed onto Celite or covalently linked to polyethylene glycol) were determined in organic solvents preequilibrated to water activities (a w) from <0.1 to 0.84. The model reaction was the transesterification between n-octanol and vinyl butyrate. It was found that KM for the nucleophile increased with increasing a w for all three lipase forms. Vmax increased with increasing a w for polyethylene glycol-lipase, whereas there was an optimum at intermediate a w values (0.11 – 0.38) for lipase powder and Celite-immobilized lipase.  相似文献

Aims: To determine the effect of water activity (aw = 0·880–0·960) and temperature (15–35°C) on the percentage of viable conidia and mycelial growth of three biocontrol agents effective against water hyacinth in Mali: Alternaria sp. isolate Mlb684, Fusarium sacchari isolate Mln799 and Cadophora malorum isolate Mln715. Methods and Results: The fungi were grown in vitro on plates containing potato dextrose agar medium at different aw values (glycerol being added to adjust the aw). The percentage of viable conidia and radial growth rate decreased with decreasing water activity. Statistical analysis showed a significant effect of aw, temperature and the aw × temperature interaction on mycelial growth (P < 0·0001). Water activity emerged as the factor exerting the greatest influence. Differences were observed between the fungi tested, the C. malorum appearing more tolerant to low aw and the F. sacchari more tolerant to high temperature (35°C). Growth models predicting the combined effect of aw and temperature were developed and response surfaces generated, showing fairly good agreement with the experimental values. Conclusions: Our results confirm the previous finding that aw has a greater influence than temperature on fungal growth. Under most conditions, variation of environmental factors has a detrimental influence on the percentage of viable conidia and mycelial growth rate of fungal isolates. Significance and Impact of the Study: The developed models may contribute to predicting the best environmental conditions for use of these fungi as effective biocontrol agents against water hyacinth.  相似文献

Controlling water activity (a w) can significantly impact the growth of Salmonella in poultry litter and manure — a phenomenon that was studied quantitatively using two common serotypes of Salmonella. The quantitative effect of changes in levels of a w on Salmonella populations was determined using inoculated, frosted glass rectangles placed in closed chambers (microcosms). Glass rectangles with known concentrations of Salmonella enteritidis and S. brandenburg were placed in microcosms maintained at an a w level of 0.893 for 24 h at room temperature (RT) and then transferred to other microcosms maintained at the same temperature but with higher a w levels (0.932 and 0.987). Salmonella populations on the slides were quantified at 4, 18, 24, and 48 h. Slightly elevated levels of a w (<0.1, i.e., 10% equilibrium relative humidity) for 24 h resulted in a 100-fold increase in counts of Salmonella. The data also suggested that in vitro adaptation to dry environments may occur when the organisms are exposed to alternating levels of relatively high and low (0.987 and 0.893) levels of a w. Any increased tolerance of Salmonella to reduced levels of a w could be the result of physico-chemical changes in the organism due to selective environmental pressure, formation of a protective biofilm, and/or entry into a dormant state. Results from this study are compatible with those from previously reported on-farm surveys, reinforcing the contention that maintaining a w below 0.85 in and around litter/manure surfaces in poultry or livestock bedding areas may be a critical factor in safe production of food. Abstract

The potential of three atoxigenic strains from different geographical origins in Africa were examined for in vitro and in situ competitiveness against two toxigenic strains of Aspergillus flavus and subsequent inhibition of aflatoxin B1 (AFB1) production under different environmental conditions. Temperature, water activity (a w) and substrate influenced the types of interaction between the three AFL? and two AFL+ strains. The competitiveness and AFB1 reduction ability of the three atoxigenic strains when interacting with the two toxigenic strains were evaluated by inoculation of 100, 25:75, 50:50, 75:25 and 100% ratios of mixed spore suspensions in vitro on malt extract and milled maize agars over 28 days and in situ on stored maize grain for 14 days, respectively at 0.99, 0.96 and 0.90 a w. For all the treatments, the effect of a w and inoculum ratio and their interaction was highly significant. Toxin inhibition was >80% in vitro at both 0.99 and 0.96 a w. In situ AFB1 reduction was influenced by the toxigenic strain assayed, a w and the inoculum ratio. Where control was achieved, it was more variable at 0.96 a w, while with more stringent water stress conditions (0.90 a w) the percentage inhibition was up to 77.2%. The study shows the importance of including environmental factors in screening and identifying effective atoxigenic strains for control of AFs (aflatoxins).  相似文献
