Triglochin maritima had only 40 % germination in distilled water, but germination was substantially enhanced when seeds were exposed to ethephon, fusicoccin, proline, kinetin, and thiourea. Also, the effect of low salinity on germination was alleviated by fusicoccin, kinetin, nitrate and thiourea, whereas, the reduction in germination at high salinity was partially countered by ethephon, kinetin, thiourea and nitrate.  相似文献

All seeds of Aeluropus lagopoides and Urochondra setulosa germinated under non-saline conditions except for Sporobolus ioclados which showed only 40 % germination. Increase in salinity substantially inhibited germination and few seeds germinated at 400 mM NaCl. Germination at 200 mM NaCl was alleviated in U. setulosa by the application of gibberellic acid and fusicoccin, in A. lagopoides by thiourea, betaine, kinetin, fusicoccin and ethephon, and in S. ioclados by gibberellin and ethephon. High salinity (400 mM NaCl) induced germination inhibition was alleviated by proline, kinetin, fusicoccin and ethephon only in A. lagopoides.  相似文献

The indigenous forage grasses Lasiurus scindicus and Panicum turgidum are candidate species for the restoration of degraded desert rangelands. The impact of five dormancy regulating chemicals on overcoming salinity-induced germination inhibition was assessed under the best germination conditions in the two species. Seeds were germinated in a series of NaCl concentrations: 0–200 mM NaCl for P. turgidum, and 0–300 mM NaCl for L. scindicus. Lasiurus scindicus seeds were more tolerant to salinity than those of P. turgidum. Twenty percent of P. turgidum seeds germinated in 100 mM NaCl and none in the higher levels, but 47.5% and 8.8% of L. scindicus seeds germinated in 100 and 200 mM NaCl, respectively. The five studied chemicals (fusicoccin, GA3, kinetin, nitrate and thiourea) did not succeed in improving germination of non-saline treated seeds of the two species, compared to the control, except thiourea in P. turgidum. The salinity-induced germination inhibition in P. turgidum was completely alleviated by the application of gibberellic acid (GA3), partially alleviated by the application of fusicoccin, kinetin and thiourea, but not affected by nitrate. In L. scindicus, the germination inhibition was completely alleviated by fusicoccin, GA3, nitrate and thiourea, but partially alleviated by kinetin. For using the two grass species in restoration of degraded rangelands affected by higher salinity, the results suggest using fusicoccin, GA3, nitrate and thiourea with L. scindicus and GA3 with P. turgidum seeds as a preseeding treatment can overcome the problem of reduced germination.  相似文献

Ethylene is invariably produced during seed germination but its role in regulating seed dormancy and germination is poorly understood. Seeds of 22 halophytic species having different life forms – salt secreting dicots, salt secreting monocots, stem succulents and leaf succulents were germinated in Petri dishes kept in a growth chamber set at 20/30 °C (night/day) temperature and a 12 hr light period. Sodium chloride and ethephon were added to the medium from the beginning of the experiment. Seed germination was recorded every other day for twenty days. Application of ethylene did not have any significant effect on releasing seeds from innate dormancy. However, it appeared to have a role in alleviation of salinity effects which varied from negative in certain species to almost complete alleviation of high salinity effects in others.Our data indicates that ethylene appears to have little role in breaking innate seed dormancy however, in most halophyte seeds studied, application of ethylene alleviate the salinity effect to various degrees. Halophyte seeds which could germinate under saline conditions approaching twice the salinity of seawater may offer clues to understand management of seed germination under highly saline conditions. Seeds of the halophyte Crithmum maritimum L. were exposed to salt stress and chemical pretreatments in order to improve germination. Seeds submitted to salt stress did not germinate but they recovered rapidly upon transfer to distilled water, recovery being higher after a low salinity pretreatment. Chemical treatments resulted in differential effects on seed germination. Known dormancy breakers such as potassium nitrate and thiourea had no effect on sea fennel seed germination. Conversely, l-ascorbic acid (40 or 60 mM) and ethanol (96%) significantly improved germination rate by 10, 30 and 30%, respectively. Pretreatment of seeds with l-ascorbic acid at 40 mM was shown to alleviate the negative effects of low NaCl concentration on germination. These findings indicate that the application of ascorbic acid may be used to improve sea fennel seed germination, which is of great interest for cultivating this plant.  相似文献

Zygophyllum simplexL. is a succulent annual that grows on thecoastal and inland saline flats around Karachi, Pakistan. Theseeds are moderately salt tolerant during germination. GerminationofZygophyllum simplexseeds under various salinity, proline,betaine, GA and kinetin treatments was determined. Proline (0.1and 1 mM) and betaine (0.1 and 1 mM) alleviated the innate dormancyof seeds, and germination reached 60–70% compared to 12%in the control set. At low salinity compatible osmotica alleviatedsome effects of salinity, but at higher NaCl concentrationsboth proline and betaine were ineffective. Gibberellic acid(0.3 and 3 mM) and kinetin (0.05 and 0.5 mM) substantially alleviatedboth innate as well as salinity-induced seed dormancy. At highersalinity (125 mM), low concentrations of kinetin (0.05 mM) andhigh concentrations of GA (3 mM) were more effective. GA completelyalleviated the effect of salinity at all concentrations used. The effects of red light, far-red light, Gibberellin A3, andethephon were studied on the germination of lettuce seeds cv.Grand Rapids harvested at different stages of development. Seeds did not become capable of germination until 8 days afteranthesis. Red light promoted seed germination from the age of8–9 days following anthesis up to the newly mature stage.Ten or 11 days following anthesis, a large percentage of seedsbecame capable of germination in the dark and therefore couldbe considered not dormant. They were affected by far-red light,but less so than the mature seeds. The effect of light on the germination of developing seeds appearedto be similar to the known light effect on mature lettuce seedgermination. Fusicoccin, a toxin stimulating cell enlargement and inducing proton extrusion in various plant tissues, has been shown to replace kinetin, gibberellic acid and red light in breaking seed dormancy. It also removes the inhibitory effect of abscisic acid. The present data also show that the stimulating effect of fucicoccin on embryo growth of decoated radish (Raphanus sativus L.) and maize (Zea mays) seeds and on the development of maize embryos is accompanied by an early, significant acidification of the medium. Acidification of the medium is also observed when fusicoccin reverses the abscisic acid-induced inhibition of germination. These results support the hypothesis that the mode of action of fusicoccin in promoting germination involves, as in stimulation of cell enlargement, the activation at the cell membrane level of proton extrusion processes. The physiological significance of fusicoccin-induced release of protons at the onset of germination is discussed in comparison with the results concerning the mechanism of action of fusicoccin on cell enlargement in other plant materials.  相似文献

Germination of Planlago lanceolata seeds buried in a chalk grasslandwas higher in bare soil than in vegetated soil, and measurementof soil nitrate concentrations showed that they were high enoughto account for this stimulation. When seeds of P. lanceolatawere sown in pots of soil with or without plants, and wateredwith nutrient solution containing either no nitrate, or 14 mMnitrate (sufficiently high that not all nitrate was absorbedby the plants), the presence of plants inhibited germinationonly when the nutrient solution contained no nitrate. Flow cytometric analyses of nuclear DNA levels were carriedout during development, stratification and germination of dormantseeds from three tree species with contrasting characteristics.Norway maple (Acer platanoides) and sycamore (Acer pseudoplatanus)have orthodox (desiccation-tolerant) and recalcitrant (desiccation-sensitive)storage behaviours, respectively, and require only a periodof cold to break dormancy, whereas, orthodox cherry (Prunusavium) seeds require an initial warm period before cold stratificationto fully stimulate germination. Whole embryos and radicle tipsof both Norway maple and sycamore were found to have stablehigh levels of 4C DNA during the latter stages of developmentand both contained nuclei arrested at the 2C and 4C levels atmaturity. Mature cherry embryos had nuclei predominantly arrestedat the 2C level. This suggests that the acquisition of desiccationtolerance is not correlated with the arrest of the cell cycleat any particular nuclear DNA level. Neither DNA replicationin radicle cells nor germination occurred when seeds were maintainedmoist at a constant 20 °C. However, in the late stages ofcold treatment during stratification, nuclear DNA levels inradicle cells changed in advance of radicle emergence in theorthodox Norway maple and cherry, whereas in the recalcitrantsycamore, change was not recorded until after radicle emergence.These results show that DNA replication has potential use asan indicator of the progress of tree seeds through stratificationtreatments used to break some types of dormancy. The action of phytohormone producing bacteria and plant growth regulators on germination and seedling growth of wheat under saline conditions were studied. Seed dormancy enforced by salinity (100 mM NaCl) was substantially alleviated and the germination was promoted by gibberellin, auxin, zeatin, and ethephon from 54 to 97%. The IAA producing bacterial strains Pseudomonas aureantiaca TSAU22, Pseudomonas extremorientalis TSAU6 and Pseudomonas extremorientalis TSAU20 significantly increased seedling root growth up to 25% in non-salinated conditions and up to 52% at 100 mM NaCl, compared to control plants. It is concluded that growth regulators considerably alleviated salinity-induced dormancy of wheat seeds. The facts mentioned above make it possible to recommend root colonizing bacteria that produce phytohormone to alleviate salt stress of wheat grown under conditions of soil salinity.  相似文献

The effects of fusicoccin on the germination of dormant, light-requiring or abscisic acid-inhibited seeds has been investigated. (1) Fusicoccin (10?6M) induces germination in dormant wheat seeds (Triticum durum cv. Cappelli; 1972 crop) and stimulates it in seeds already relieved from dormancy (1971 crop), with an effect similar to that of gibberellic acid. (2) Fusicoccin (1.5 × 10?6M) is more active than the two phytohormones gibberellic acid and benzyladenine and than white light in stimulating light-requiring lettuce seeds (Lactuca sativa cv. Grand Rapids) to germinate. Germination of radish seeds (Raphanus sativus) is also accelerated by fusicoccin, while benzyladenine and gibberellic acid are less active in this material. (3) Fusicoccin (1.5 × 10?5M) removes almost completely the inhibitory effect of abscisic acid on germination of radish and lettuce seeds, whereas benzyladenine (10?4M) and gibberellic acid (3 × 10?4M) remove the inhibition only partially. The possible relationship between these results and previous information on growth by cell enlargement is discussed in terms of the mechanism of action of fusicoccin as compared with natural hormones.  相似文献

The effect of different types and concentrations of dormancy regulating chemicals (DRCs) on innate and induced dormancy was evaluated under optimal germination conditions in the invasive Prosopis juliflora shrub. Lower concentrations of gibberellic acid (0.3 mM) and kinetin (0.05 mM) were more effective in enhancing germination % and rate at higher concentrations of NaCl, but the reverse was true for thiourea. None of the DRCs alleviated innate dormancy of P. juliflora. Germination % and rate decreased as salinity increased. Percent final germination of non-treated seeds was significantly reduced at 500 mM NaCl and virtually inhibited in 600 mM NaCl. Germination reduction in 500 mM NaCl was not alleviated by any of the DRCs, but inhibition induced at 600 mM NaCl was partially alleviated by all the DRCs. Gibberellic acid had a significantly greater effect than kinetin in alleviating germination inhibition. For restoration of saline soil through the use of P. juliflora, results suggest using DRCs, particularly gibberellic acid and thiourea, as a preseeding treatment can overcome the problem of reduced germination.  相似文献

The interactive effects of phytochrome, potassium nitrate andthiourea on the germination response to alternating temperaturesin achenes (seeds) of Ranunculus sceleratus L. were studied.Using thermogradient bars, high levels of germination were recordedover a broad range of alternating temperatures providing seedsreceived daily irradiations. Reduced germination in temperaturecycles with a relatively long warm phase was related to thelevel of the active form of phytochrome (Pfr). Dose-responseexperiments to red light (R) and temperature shifts showed thatthe actions of Pfr and alternating temperatures were interdependent.Maximum germination was recorded when intermittent pulses ofR were combined with daily 4 h temperature shifts from 16°Cto 26°C. Whilst probit analysis showed that potassium nitrateand thiourea both increased population sensitivity to temperatureshifts, thiourea was a more potent stimulant. Although the effectof both chemicals was dependent on phytochrome photo-equilibriumthe threshold level of Pfr required for thiourea action wasclearly much lower than that required for nitrate action. Thioureapotentiated a response to daily temperature shifts even whenPfr was at a low, normally inhibitory level. These results indicatedifferent mechanisms of action for potassium nitrate and thioureain relation to phytochrome controlled seed germination. Seeds ofTaxus maireiare known for their deep dormancy whichcan only be broken by a procedure involving warm stratificationfollowed by cold stratification. Treatments with alternatingtemperatures of 25/15 or 23/11 °C (12 h light) for 6 monthsfollowed by 5 °C for 3 months were successful in overcomingseed dormancy. After 6 months of warm stratification, cytologicalchanges observed included: enlargement of the embryo; a decreasein the number of lipid bodies; appearance of ER; and increasesin mitochondria, plastids, dictyosomes, vacuoles and microbodiesin the shoot apical meristem. Cold stratification followingthe warm treatment induced cell division, and one or two distinctnucleoli in the shoot apical meristem cells were observed. Bothwarm and cold stratification reduced endogenous ABA concentrationsfrom the original 8888 pg per freshly harvested seed to 392and 536 pg, respectively. Treatment with exogenous gibberellinsafter seeds had been warm-stratified showed that GA4and GA7wereeffective at promoting seed germination, but GA3was not. Theseresults suggest that the strong seed dormancy ofT. maireicouldbe caused by a high ABA content and underdevelopment of theembryos in freshly shed seeds. We conclude that warm stratificationwith alternating temperatures increases the growth of embryosby cell expansion and enlargement and decreases ABA content,but seeds still remain ungerminated. The surface features of seed coat, hilum and strophiole, histologicalaspects of hilum and strophiole, and mechanism of seed dormancyinVigna minima, V. umbellata, V. dalzelliana and V. calcarataauct. pi. have been investigated. Scanning electron microscopyhas revealed a substantial variability in seed coat microtopography,hilum and strophiole, which is of taxonomic and evolutionarysignificance. The histology of the hilum is uniform in all fourtaxa, but the structure of the strophiole shows differencesbetween wild and cultivated species. All four taxa have hardseeds; those ofV. umbellata have no dormancy but those of wildspecies have water-impermeable dormancy. Seed germination experimentsusing araldite-coated, and hilum-micropyle- and strophiole-pluggedhard seeds of V. umbellata and water-impermeable seeds of V.minima, which were pin-pricked through the strophiolar groove,demonstrate that the zone of weakness to the permeability ofwater in hard seeds of these species is the strophiolar region,and not the seed coat, hilum or micropyle. The parenchymatoustissue of the strophiolar plug in the seeds ofV. umbellataisassociated with the lack of dormancy, whereas the sclerenchymatoustissue of the strophiolar plug in the seeds ofV. minima is responsiblefor the water-impermeable seed dormancy. Under natural conditions,the water-impermeable dormancy of wild species breaks down asa result of the degradation of the hard seed coat, by microbialaction, at the strophiolar groove.  相似文献

Nutrient and salt (NaCI) levels were examined in fruits of twostrand species, Cakile maritima and Arctotheca populifolia,collected near Perth, Western Australia. Nutrient levels inseeds of C. maritima and achenes of A. populifolia were relativelyhigh when compared to those in seeds of Australian native speciesor introduced crop plants, despite the very low nutrient statusof the strand habitat. The high levels of organic and essentialinorganic nutrients in seeds would be of especial value in seedlingestablishment. The upper seed of the two-segmented fruit ofCakile was heavier than the lower seed, but both seeds had verysimilar levels of nutrients. Concentrations of N, P, and micronutrientswere very much higher in seeds of Cakile and Arctotheca thanin other parts of the fruit. Cakile leaves had similar levelsof N and P to seeds, but higher levels of micronutrients, especiallyNa and Cl. In Cakile, concentration ratios for Na levels inleaves, silicule and seeds were 359: 74: 1 respectively, thosefor Cl were 200: 30: 1, suggesting very restricted loading ofNa and Cl into the phloem stream destined for fruits, and selectiveuptake by developing embryos. Cakile seeds germinate within their fruit segments, and levelsof NaCl in recently-matured fruits would be high enough to suppressgermination. The top segment of the fruit had a higher concentrationof NaCl than the bottom, and it is likely that this is responsiblefor the lower germination of seeds within top segments. Theliterature on seed dormancy in Cakile is reviewed briefly, andit is concluded that salt levels in the fruit wall and strandsoil are the major determinants of dormancy in Cakile seeds. Various daily photon doses (daily net number of photons perunit area), provided by varying both photon flux density andphotoperiod, were applied to dormant seed populations of fourgraminaceous species tested for germination in alternating-temperatureregimes on filter papers moistened with either water or a solutionof potassium nitrate. In all four species germination was dependentupon daily photon dose, the product of photon flux density andduration, and quantified by linear relations between the probitof percentage germination and the logarithm of the daily photondose determined by probit analysis. Photon flux density andphotoperiod only influenced germination indirectly as factorsof daily photon dose, except in Echinochloa turnerana (Domin)J.M. Black at high daily photon doses where lower germinationwas observed in continuous light than in 8 h d–1 photoperiod.The low energy reaction, in which increasing dose promotes seedgermination, was observed in Echlnochloa turnerana and Eragrostistef(Zucc.) Trotter but not in the populations of Brachiariahumidicola (Rendle) Schwcickerdt and Panicum maximum Jacq. usedin this study. The high irradiance reaction, in which increasingthe photon dose reduces germination, was, however, observedin all four species. In Eragrostis tef potassium nitrate influencedthe response of seed germination to light, but in the remainingspecies its influence on this relationship was either additive(Panicum maximum) or nil (Brachiaria humidicola and Echinochloaturnerana) in the alternating-temperature regimes used in thisstudy. The implications of the quantal response to photon dosefor devising optimal light regimes to be applied in laboratorytests intended to maximize seed germination are discussed. The effects of humidification, storage in liquid nitrogen (1or 30 d) and rewarming rate on seed germination were studiedin two Spanish endemics. Humidification resulted in higher germinationpercentages only in the species with hard covers, especiallyin slowly rewarmed seeds.
