Abstract. The effects of intrinsic and extrinsic factors on locomotor activity in crowd‐reared first‐stadium nymphs of the migratory locust Locusta migratoria are investigated under continuous light conditions using an actograph apparatus. Nymphs show monomodal or bimodal patterns of locomotor activity with respect to the time after the start of measurements, depending on the age. Locomotor activity is suppressed by the presence of grass in nymphs aged 0–2 days old but a peak of activity observed shortly after hatching is not suppressed. The results suggest that newly‐hatched nymphs may try to disperse from the hatching site. Nymphs show higher locomotor activity levels under moist conditions than under dry conditions during the first 5‐h period of measurements. This enhanced locomotor activity may constitute attempts to avoid high humidity. Under dim‐light conditions (2 × 10?2 Wm?2), locomotor activity is suppressed during the first half day and increases to a high level thereafter in both grass‐fed and unfed individuals. This increased activity might indicate a possible involvement of circadian rhythms. Background colour has no significant effect on the locomotor activity. The present study provides new aspects of behaviour in nymphs as well as baseline data for behavioural analysis of locust locomotion in relation to phase polyphenism.  相似文献

Varieties resistant to powdery mildew (PM; caused by Podosphaera leucotricha) are a major component of sustainable apple production. Resistance can be achieved by knocking‐out susceptibility S‐genes to be singled out among members of the MLO (Mildew Locus O) gene family. Candidates are MLO S‐genes of phylogenetic clade V up‐regulated upon PM inoculation, such as MdMLO11 and 19 (clade V) and MdMLO18 (clade VII). We report the knock‐down through RNA interference of MdMLO11 and 19, as well as the complementation of resistance with MdMLO18 in the Arabidopsis thaliana triple mlo mutant Atmlo2/6/12. The knock‐down of MdMLO19 reduced PM disease severity by 75%, whereas the knock‐down of MdMLO11, alone or in combination with MdMLO19, did not result in any reduction or additional reduction of susceptibility compared with MdMLO19 alone. The test in A. thaliana excluded a role for MdMLO18 in PM susceptibility. Cell wall appositions (papillae) were present in both PM‐resistant and PM‐susceptible plants, but were larger in resistant lines. No obvious negative phenotype was observed in plants with mlo genes knocked down. Apparently, MdMLO19 plays the pivotal role in apple PM susceptibility and its knock‐down induces a very significant level of resistance.  相似文献

The vector of Chagas disease, Triatoma infestans, is largely controlled by the household application of pyrethroid insecticides. Because effective, large‐scale insecticide application is costly and necessitates numerous trained personnel, alternative control techniques are badly needed. We compared the residual effect of organophosphate‐based insecticidal paint (Inesfly 5A IGR? (I5A)) to standard deltamethrin, and a negative control, against T. infestans in a simulated natural environment. We evaluated mortality, knockdown, and ability to take a blood meal among 5th instar nymphs. I5A paint caused significantly greater mortality at time points up to nine months compared to deltamethrin (Fisher's Exact Test, p < 0.01 in all instances). A year following application, mortality among nymphs in the I5A was similar to those in the deltamethrin (χ2 = 0.76, df=1, p < 0.76). At months 0 and 1 after application, fewer nymphs exposed to deltamethrin took a blood meal compared to insects exposed to paint (Fisher's Exact Tests, p < 0.01 and p < 0.01, respectively). Insecticidal paint may provide an easily‐applied means of protection against vectors of Chagas disease.  相似文献

Mutants in lipid metabolism often show a lethal phenotype during reproduction that prevents investigating a specific role of the lipid during different developmental processes. We focused on two non‐specific phospholipases C, NPC2 and NPC6, whose double knock‐out causes a gametophyte‐lethal phenotype. To investigate the role of NPC2 and NPC6 during vegetative growth, we produced transgenic knock‐down mutant lines that circumvent the lethal effect during gametogenesis. Despite no defect observed in leaves, root growth was significantly retarded, with abnormal cellular architecture in root columella cells. Furthermore, the short root phenotype was rescued by exogenous supplementation of phosphocholine, a product of non‐specific phospholipase C (NPC) ‐catalyzed phosphatidylcholine hydrolysis. The expression of phospho‐base N‐methyltransferase 1 (PMT1), which produces phosphocholine and is required for root growth, was induced in the knock‐down mutant lines and was attenuated after phosphocholine supplementation. These results suggest that NPC2 and NPC6 may be involved in root growth by producing phosphocholine via metabolic interaction with a PMT‐catalyzed pathway, which highlights a tissue‐specific role of NPC enzymes in vegetative growth beyond the gametophyte‐lethal phenotype.  相似文献

The simultaneous expression of costly immune effectors such as multiple antimicrobial peptides is a hallmark of innate immunity of multicellular organisms, yet the adaptive advantage remains unresolved. Here, we test current hypotheses on the evolution of such defence cocktails. We use RNAi gene knock‐down to explore, the effects of three highly expressed antimicrobial peptides, displaying different degrees of activity in vitro against Staphylococcus aureus, during an infection in the beetle Tenebrio molitor. We find that a defensin confers no survival benefit but reduces bacterial loads. A coleoptericin contributes to host survival without affecting bacterial loads. An attacin has no individual effect. Simultaneous knock‐down of the defensin with the other AMPs results in increased mortality and elevated bacterial loads. Contrary to common expectations, the effects on host survival and bacterial load can be independent. The expression of multiple AMPs increases host survival and contributes to the control of persisting infections and tolerance. This is an emerging property that explains the adaptive benefit of defence cocktails.  相似文献

Maize Iranian mosaic virus (MIMV, family Rhabdoviridae) causes an important disease in cereal crops in Iran. It is transmitted by the planthopper Laodelphax striatellus in a persistent, propagative manner. In the present study, the effect of MIMV on the energy reserves of L. striatellus was studied by comparing energy contents in viruliferous and non‐viruliferous insects. Results showed that MIMV‐infected male and female adults, and nymphs stored 1.82, 2.24 and 1.7‐fold more total energy reserves than non‐viruliferous individuals. This is consistent with a 2.55‐fold increase of the total energy (sum of energy sources in nymphs and adults) of viruliferous compared to non‐viruliferous specimens. A significant increase in glycogen (2.26‐fold), carbohydrate (2.13‐fold), lipid (1.63‐fold) and protein (1.96‐fold) was documented in viruliferous insects compared to non‐viruliferous insects. Based on these results, we conclude that MIMV enhances the energy reserves of its vector and therefore, may play a vital role in the ecology of L. striatellus.  相似文献

Glycogen synthase kinase/SHAGGY‐like kinases (SKs) are a highly conserved family of signaling proteins that participate in many developmental, cell‐differentiation, and metabolic signaling pathways in plants and animals. Here, we investigate the involvement of SKs in legume nodulation, a process requiring the integration of multiple signaling pathways. We describe a group of SKs in the model legume Lotus japonicus (LSKs), two of which respond to inoculation with the symbiotic nitrogen‐fixing bacterium Mesorhizobium loti. RNAi knock‐down plants and an insertion mutant for one of these genes, LSK1, display increased nodulation. Ηairy‐root lines overexpressing LSK1 form only marginally fewer mature nodules compared with controls. The expression levels of genes involved in the autoregulation of nodulation (AON) mechanism are affected in LSK1 knock‐down plants at low nitrate levels, both at early and late stages of nodulation. At higher levels of nitrate, these same plants show the opposite expression pattern of AON‐related genes and lose the hypernodulation phenotype. Our findings reveal an additional role for the versatile SK gene family in integrating the signaling pathways governing legume nodulation, and pave the way for further study of their functions in legumes.  相似文献

Oviposition preferences of herbivorous insects affect offspring performance. Both positive and negative links between oviposition preference and offspring performance have been reported for many species. A gall‐inducing leafhopper, Cicadulina bipunctata Melichar (Hemiptera: Cicadellidae), feeds on various Poaceae plants and induces galls of enhanced nutritional value for their offspring. Although gall induction by C. bipunctata improves nymphal performance, the oviposition preference of females between galled and non‐galled host plants is still unclear. In this paper, the nymphal performance and oviposition and feeding‐site preference of C. bipunctata were investigated using galled wheat, Triticum aestivum L., and non‐galled barley, Hordeum vulgare L., as host plants. The survival rate of C. bipunctata on wheat was significantly higher than on barley. In the choice test, significantly more eggs were laid into barley, whereas the number of eggs deposited on both hosts was not significantly different in the no‐choice test. The number of settling individuals per leaf area was not significantly different between wheat and barley, suggesting no clear preference for oviposition between these plants. Experience as a nymph with a growing host did not affect oviposition preference as adult female. The inconsistent correspondence between offspring performance and oviposition preference of C. bipunctata may reflect the high mobility of nymphs and/or differences in leaf area between host plants. The results indicate that the previous finding that oviposition preference and offspring performance are not always positively correlated in herbivorous insects is applicable to gall‐inducing insects.  相似文献

Halyomorpha halys Stål (Hemiptera: Pentatomidae) has recently become a major orchard pest in the Mid‐Atlantic, USA. Large H. halys populations can develop on wild tree hosts adjacent to orchards, posing an ongoing threat to fruit. Adults and nymphs feed on tree fruit, causing economic injury. Understanding the seasonal patterns of nymphal host use among trees at the orchard‐woodland interface may aid the development of integrated pest management strategies for this pest. In laboratory and field experiments, modified versions of published trap designs – ‘Circle’, ‘Hanula’, ‘M&M’ (after Moeed & Meads) traps – were compared for their effectiveness for capturing H. halys nymphs walking up and down tree trunks. In the laboratory, second instars were released at the top and bottom of ailanthus (tree of heaven), Ailanthus altissima (Mill.) Swingle (Simaroubaceae), logs and captures were recorded after 24 h. Circle and M&M traps, respectively, were most effective for capturing nymphs walking up and down. In the field, traps were deployed on ailanthus trees next to apple orchards and captures were recorded weekly from 24 July to 11 September 2013. As in the laboratory, Circle and M&M traps captured the greatest number of upward‐ and downward‐walking nymphs. Hanula traps were least effective in both experiments. In the field, 88% of total captures were of nymphs walking up trees. This was at least partially explained by behavioral assays in the laboratory demonstrating that nymphs exhibited negative gravitaxis and positive phototaxis. Stage‐specific trends in captures of instars walking up during field sampling were observed. These results suggest that trunk traps can be used to address important ecological questions about seasonal patterns of host use by H. halys nymphs.  相似文献

The impact of deltamethrin‐impregnated cloth targets on Stegomyia polynesiensis (= Aedes polynesiensis) (Marks) (Diptera: Culicidae) was assessed under laboratory and semi‐field settings in French Polynesia. Stegomyia polynesiensis females were released into small laboratory cages and large field cages containing either a deltamethrin‐treated or an untreated navy blue cloth, and mosquito knock‐down and mortality were assessed. The 24‐h mortality rate in mosquitoes exposed to the insecticide‐treated target in small cages was 98.0%. These mosquitoes also demonstrated significantly higher levels of knock‐down than those exposed to the untreated target. Mortality in field cages was assessed at 24 and 48 h. The 24‐h mortality rate in mosquitoes exposed to the control target was 31.2%, whereas that in those exposed to the deltamethrin‐treated target was 54.3%. The 48‐h mortality rate was also elevated in mosquitoes exposed to the deltamethrin‐treated target, but this result did not differ significantly from that observed in mosquitoes exposed to the control target. The significant suppression of female S. polynesiensis by deltamethrin‐treated resting targets in this study indicates that these targets could play a role in the control of an important disease vector in the South Pacific region.  相似文献

Adult Dermestes maculatus were tested for resistance by exposing them to films of lindane in oil on filter papers. Insects which survived a discriminating dose, designed to knock down 100% of susceptible insects, were classed as resistant. Of 42 strains intercepted on imports or collected from infested poultry houses, 34 were found to be resistant when first tested. The F1 progeny of survivors from 19 of these resistant strains were also exposed to the discriminating dose. Significant increases in survival confirmed the presence of resistance in 18 strains.  相似文献

Although dietary restriction (DR) is known to extend lifespan across species, from yeast to mammals, the signalling events downstream of food/nutrient perception are not well understood. In Caenorhabditis elegans, DR is typically attained either by using the eat‐2 mutants that have reduced pharyngeal pumping leading to lower food intake or by feeding diluted bacterial food to the worms. In this study, we show that knocking down a mammalian MEKK3‐like kinase gene, mekk‐3 in C. elegans, initiates a process similar to DR without compromising food intake. This DR‐like state results in upregulation of beta‐oxidation genes through the nuclear hormone receptor NHR‐49, a HNF‐4 homolog, resulting in depletion of stored fat. This metabolic shift leads to low levels of reactive oxygen species (ROS), potent oxidizing agents that damage macromolecules. Increased beta‐oxidation, in turn, induces the phase I and II xenobiotic detoxification genes, through PHA‐4/FOXA, NHR‐8 and aryl hydrocarbon receptor AHR‐1, possibly to purge lipophilic endotoxins generated during fatty acid catabolism. The coupling of a metabolic shift with endotoxin detoxification results in extreme longevity following mekk‐3 knock‐down. Thus, MEKK‐3 may function as an important nutrient sensor and signalling component within the organism that controls metabolism. Knocking down mekk‐3 may signal an imminent nutrient crisis that results in initiation of a DR‐like state, even when food is plentiful.  相似文献

The pace‐of‐life syndrome (POLS) hypothesis means that animal behavior is correlated with life history strategies. Studies have reported that the free‐running period of the circadian rhythm (length of the period) is correlated with life history strategies in some animals. Although the length of the circadian rhythm may be associated with the POLS hypothesis, few studies have investigated the relationships among animal behavior, life history traits, and circadian rhythm. We tested the POLS hypothesis in the assassin bug, Amphibolus venator, which shows individual variation in locomotor activity. We found higher repeatability of differences in locomotor activity between individuals. Moreover, we found a trade‐off between locomotor activity and developmental period such that active individuals developed faster. However, locomotor activity was not correlated with the length of the circadian rhythm in Avenator. Therefore, this study suggests that the length of the circadian rhythm in Avenator does not support the POLS hypothesis.  相似文献

Thermal‐stress selection can affect multiple fitness components including mating success. Reproductive success is one of the most inclusive measures of overall fitness, and mating success is a major component of reproduction. However, almost no attention has been spent to test how mating success can be affected by thermal‐stress selection. In this study, we examine the mating success in the cactophilic Drosophila buzzatii Patterson & Wheeler (Diptera: Drosophilidae) derived from two natural populations that nearly represent the ends of an altitudinal cline for heat knock‐down resistance. Furthermore, we extended the analysis using laboratory lines artificially selected for high and low heat knock‐down resistance. Mating success at high temperature was found to be higher in the lowland than the highland population after a heat pre‐treatment. Moreover, individuals selected for heat knock‐down resistance showed higher mating success at high temperature than did individuals selected for low knock‐down resistance. These results indicate that adaptation to thermal stress can confer an advantage on fitness‐related traits including mating success and highlight the benefits of earlier heat exposure as an adaptive plastic response affecting mating success under stress of higher temperature.  相似文献
