Photosynthetic activity, the content of various photosynthetic pigments, and the chloroplast ultrastructure were examined in the leaves of cucumber (Cucumis sativus L.) and pea (Pisum sativum L.) plants of different ages grown under red light (600–700 nm, 100 W/m2). In pea leaves tolerant to red-light irradiation, chloroplast ultrastructure did not essentially change. In the first true leaves of cucumber plants susceptible to red-light irradiation, we observed a considerable increase in the number and size of plastoglobules, the appearance of chloroplasts lacking grana or containing only infrequent grana, and stromal thylakoids. In the upper leaves of 22-day-old cucumber plants, the chloroplast structure was essentially similar to that of the control chloroplasts in white light, and we therefore suppose that these plants have acclimated to red light.  相似文献

Abstract. The ultrastructure of chloroplasts from palisade and spongy tissue was studied in order to analyse the adaptation of chloroplasts to the light gradient within the bifacial leaves of pea. Chloroplasts of two nuclear gene mutants of Pisum sativum (chlorotica-29 and chlorophyll b-less 130A), grown under normal light conditions, were compared with the wild type (WT) garden-pea cv. ‘Dippes Gelbe Viktoria’. The differentiation of the thylakoid membrane system of plastids from normal pea leaves exhibited nearly the same degree of grana formation in palisade and in spongy tissue. Using morphometrical measurements, only a slight increase in grana stacking capacity was found in chloroplasts of spongy tissue. In contrast, chloroplasts of mutant leaves differed in grana development in palisade and spongy tissue, respectively. Their thylakoid systems appeared to be disorganized and not developed as much as in chloroplasts from normal pea leaves. Grana contained fewer lamellae per granum, the number of grana per chloroplast section was reduced and the length of appressed thylakoid regions was decreased. Nevertheless, chloroplasts of the mutants were always differentiated into grana and stroma thylakoids. The structural changes observed and the reduction of the total chlorophyll content correlated with alterations in the polypeptide composition of thylakoid membrane preparations from mutant chloroplasts. In sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE), polypeptide bands with a relative molecular mass of 27 and 26 kilodalton (kD) were markedly reduced in mutant chloroplasts. These two polypeptides represented the major apoproteins of the light harvesting chlorophyll a/b complex from photosystem II (LHC-II) as inferred from a comparison with the electrophoretic mobility of polypeptides isolated from the LHC-II.  相似文献

Summary Primary leaves of intact bean plants (Phaseolus vulgaris) were treated with benzyladenine (BA) at different stages of growth. Changes in the ultrastructure of chloroplasts and the contents of chlorophyll, carotenoid, and protein (soluble and insoluble) in leaves with different treatments were followed and compared. When BA was applied from an early stage, it increased the chloroplast size and the number of grana per chloroplast without any pronounced effect on the grana size. When BA treatment was stopped at the early stage, these effects remained for a while and then diminished. When BA treatment was begun at a late stage, such marked effects were not observed, suggesting that only young leaves could respond to BA in that manner. However, the late treatment efficiently prevented the process of the last stage of leaf senescence characterized by disintegration of thylakoids with concomitant increase in the plastoglobule size. Chlorophyll, carotenoid, and insoluble protein contents per leaf followed similar changes in chloroplast length and the number of grana per chloroplast section.  相似文献

We examined morphological and ultrastructural differences in chloroplasts of cotton leaves and the fruiting organs, bract, and capsule wall to advance our understanding of their commonly observed differences in photosynthetic efficiency. Chloroplasts from leaves were large (7.1 μm long in cross section), lens shaped with a well developed membrane system differentiated into grana and stroma lamellae that occupied the large cross-sectional area (12.3 μm2) of the organelle. A few small plastoglobuli and starch grains were scattered in the stroma region. The bract chloroplasts were correlative of leaf chloroplasts in size (6.8 μm in length) and shape with the exception that the bract chloroplasts exhibited greater thylakoid number per granum (15.8) than the leaf chloroplasts (10.5). In contrast to leaf and bract, the capsule wall chloroplasts were smaller in size (4.3 μm) and cross sectional area (6.8 μm2) than either the leaf or bract. The most intriguing feature of the capsule wall chloroplasts was its domination by large starch granules (5.3 μm2) in the stroma which filled the whole chloroplast coercing the membrane system to move towards the periphery of the organelle. Grana number and thylakoids per granum were lowest in the capsule wall chloroplasts. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

Protoplasts were prepared from pea (Pisum sativum L.) leaves throughout development and their contents spread in a monolayer to determine the number of chloroplasts per cell. This approach permitted the rapid analysis of more than 100 cells at each stage of development. The average number of chloroplasts per cell increased from 24±10 to 64±20 during greening and expansion of the first true foliage leaves; all cells containing chloroplasts apparently increase their chloroplast number. A parallel increase in the amount of DNA per nucleus was not observed. As the leaves senesced the chloroplast number gradually decreased to 44±12. We have correlated these changes with our previous results on the percentage of chloroplast DNA per cell. Chloroplast multiplication resulted in a 2.7-fold dilution (from 272 to 102) of the number of copies of the chloroplast DNA molecule per plastid.  相似文献

The root system of potato (Solanum tuberosum L. cv. Favorita) plants was treated with different O2 and CO2 concentrations for 35 d in aeroponic culture. Under 21 or 5 % O2 in the root zones, the thickness of leaves and palisade parenchyma significantly increased at 3 600 μmol(CO2) mol−1 in the root zone, compared with CO2 concentration 380 μmol mol−1 or low CO2 concentration (100 μmol mol−1). In addition, smaller cells of palisade tissue, more intercellular air spaces and partially two layers of palisade cells were observed in the leaves with root-zone CO2 enrichment. Furthermore, there was a significant increase in the size of chloroplasts and starch grains, and the number of starch grains per chloroplast due to elevated CO2 only under 21 % O2. In addition, a significant decline in the thickness of grana and the number of lamellas, but no significant differences in the number of grana per chloroplast were observed under elevated CO2 concentration. The accumulation of starch grains in the chloroplast under elevated CO2 concentration could change the arrangement of grana thylakoids and consequently inhibited the absorption of sun radiation and photosynthesis of potato plants.  相似文献

Soybean plants grown in controlled environment cabinets under light intensities of 220 w/m2 or 90 w/m2 (400–700 nm) and day to night temperatures of 27.5–22.5 C or 20.0–12.5 C in all combinations, exhibited differences in growth rate, leaf anatomy, chloroplast ultrastructure, and leaf starch, chlorophyll, and chloroplast lipid contents. Leaves grown under the lower light intensity at both temperatures had palisade mesophyll chloroplasts containing well-formed grana. The corresponding leaves developed under the higher light intensity had very rudimentary grana. Chloroplasts from high temperature and high light had grana consisting of two or three appressed thylakoids, while grana from the low temperature were confined to occasional thylakoid overlap. Spongy mesophyll chloroplasts were less sensitive to growth conditions. Transfer experiments showed that the ultrastructure of chloroplasts from mature leaves could be modified by changing the conditions, though the effect was less marked than when the leaf was growing.  相似文献

Abstract The chloroplast ultrastructure, especially the thylakoid organization, the polypeptide composition of the thylakoid membranes and photosynthetic O2 evolution rate, chlorophyll (Chl) content and Chi a/b ratio were studied in leaves of nine plants growing in contrasting biotopes in the wild in South Finland. All the measurements were made at the beginning of the period of main growth on leaves approaching full expansion, when the CO2-saturated O2 evolution rate (measured at 20°C and 1500 μmol photons m?2s?1) was at a maximum, ranging from 19.2 to 6.9 μmol O2 cm?2 h?1. Among the species, the Chi a/b ratio varied between 3.75 and 2.71. In the mesophyll chloroplasts, the ratio of the total length of appressed to non-appressed thylakoid membranes varied between 1.07 and 1.79, the number of partitions per granum varied between 2.8 and 12.0 and the grana area between 21 and 42% of the chloroplast area. There was a significant relationship between the rate of O2 evolution of the leaf discs and the thylakoid organization in the mesophyll chloroplasts. The higher the O2 evolution rate, the lower was the ratio of the total length of appressed to non-appressed thylakoid membranes and also the lower the grana area. Although the relationship of the photosynthetic rate with the Chi content and the Chi a/b ratio of the leaves was not as clear, a significant negative correlation existed between the Chi a/b ratio and the ratio of appressed to non-appressed thylakoid membranes, indicating lateral heterogeneity in the distribution of different Chl- protein complexes.  相似文献

Summary The amounts of chlorophyll and nitrogen and the numbers of cells per unit area change as the green leaves of spinach plants grow and increase in size in the light. The changes in the numbers of chloroplasts per cell were measured by a new method. A 5-fold increase in the numbers of chloroplasts per cell took place in both palisade and mesophyll cells over a growing period of 10 days during which time the area of the leaves increased from 1 to 50 cm2. Proplastids were not present in the young green leaves but electron-microscope and phase-contrast observations showed the presence of grana-containing chloroplasts, many of which appeared to be undergoing division by constriction. It is suggested that the large increase in chloroplast numbers as leaf cells grow and expand in the light is from the division of differentiated chloroplasts containing grana.  相似文献

Question: What are the edge effect responses of epiphytic lichen communities in Mediterranean Quercus pyrenaica forest? Location: Central Spain. Methods: We established ten transects perpendicular to a road dissecting a well conserved remnant of Q. pyrenaica forest into two sections. Transects extended from the forest/road edge to 100 m into the forest. Data were collected from seven plots in each transect at different distances from the edge. Variables were grouped into stand scale variables (distance to edge, number of trees per plot, mean diameter per plot, irradiance) and tree scale variables (diameter and height of sampled trees, aspect of the sampled square and relative height of the square). We used General Mixed Linear Models and constrained ordination techniques to test the hypothesis that the spatio‐temporal heterogeneity of light and water controls the occurrence of lichens and bryophytes along the edge‐interior gradient in the Q. pyrenaica forest. Results: Microclimatic parameters vary in a non‐linear way; edge and interior stands showed the most divergent and extreme values. Although the micro‐environment within Mediterranean forests is heterogeneous, interior conditions are apparently suitable for the performance of some specific forest epiphytes. Consequently, species richness does not show significant differences along the gradient. Total epiphytic cover increases towards the forest interior, but distance to the edge together with other predictors at the tree scale (aspect and height of the square) are the most relevant predictors for the composition and structure of these communities. Conclusions: Composition and structure of epiphytic communities in a Mediterranean semi‐deciduous forest are affected by the edge between the forest and the road constructed. Since some extremely rare lichens only occur at interior stands, the conservation of these threatened elements requires urgent conservation measures because well preserved and unmanaged forests in the Mediterranean region are very rare.  相似文献

As part of an investigation into the control of chloroplast replication the number and size of chloroplasts in mesophyll cells was examined in relation to the size of the cells. In first leaves of Triticum aestivum L. and T. monococcum L. the number of chloroplasts in fully expanded mesophyll cells is positively correlated with the plan area of the cells. The linear relationship between chloroplast number per cell and cell plan area is also consistent over a fivefold range of cell size in isogenic diploid and tetraploid T. monococcum. In T. aestivum the chloroplast number per unit cell plan area varies among cells in relation to the size of the chloroplasts. Those cells containing chloroplasts with a relatively small face area have a correspondingly higher density of chloroplasts, and consequently, the total chloroplast area per unit cell plan area is very similar in all the cells. The results indicate that the proportion of the cell surface area covered by chloroplasts is precisely regulated, and that this is achieved during cell development by growth and replication of the chloroplasts.  相似文献

Illus. 1961.—The grana of starch-free chloroplasts of fully expanded leaves of Nicotiana rustica are distinct, compartmented, subplastid entities. They vary in size, shape, orientation and in the distinctness with which their compartments are delineated. It has not been possible to equate accurately their micro and ultramicro appearances. At the ultramicro level, the grana are connected with each other at irregular intervals by a system of anastomosing channels. The partitions forming the compartments of the grana may be coarse or very fine but are constant in appearance in any given chloroplast. The loculi enclosed by the partitions may vary in size with a granum, depending upon their location or upon the physiological activity of the chloroplast. The stroma does not penetrate the grana; it may be relatively fluid and the grana-fretwork system may move within it. A double envelope, which may have pores connecting stroma and hyaloplasm, surrounds the chloroplasts. Materials may collect between the surfaces of the envelope. There is considerable variation in the ultramicro details of chloroplast structure of Nicotiana rustica. It is not yet possible to distinguish accurately between those variations which may be of physiological significance and those which may be induced by processing.  相似文献

Summary The size ranges of chloroplasts in living mesophyll cells of Spinacia oleracea, Allium cepa, Beta vulgaris (Swiss chard and red beet) and Nicotiana glutinosa are extremely wide, e.g., ranging from about 6 µ2 to 103 µ2 in face area for spinach. Moreover, the size distributions are positively skewed. We interpret the size range and skewed size distributions primarily to reflect an enormous growth of the bulk of the chloroplasts from small, equal-sized chloroplasts produced by fission of a small sub-population of constricted mature chloroplasts. While actual fission has never been observed, a slow division rate of the constricted chloroplasts in N. glutinosa can account for the increase in chloroplast numbers per cell during leaf development and for the presence of small, non-constricted chloroplasts after the small chloroplasts which developed during the initial meristem activity have enlarged. Chloroplast numbers and total amount of chloroplast material per cell face were positively correlated with mesophyll-cell face size. However, the fraction of the cell face occupied with chloroplasts was essentially constant and independent of cell size and cell age while being markedly different for different species of plants. There appear to be some family characteristics in that closely related species have similar size-distributions and ranges of chloroplast sizes. The observations are discussed with respect to the ontogeny of chloroplasts in higher plants.  相似文献

The fine structure of mesophyll chloroplasts in green islands and in adjacent chlorotic areas of barley leaves infected with Erysiphe graminis (DC.) was compared with healthy non-inoculated tissue. Chloroplasts in green islands were persistent. Green-island chloroplast grana were enlarged and fewer in number than in healthy tissue. In contrast, cells in chlorotic areas had fewer chloroplasts and their lamellae showed progressive degeneration and fragmentation. The lamellae often resembled aberrant prolamellar bodies. As lamellar degeneration progressed there was a marked increase in the amount of osmophilic material within the chloroplasts.  相似文献

Effects of the desA gene from the cyanobacterium Synechocystis sp. encoding Δ12 acyl-lipid desaturase and increasing the level of unsaturated fatty acids (linoleic acid (18:2) primarily) in membrane lipids, which was inserted into potato (Solanum tuberosum L., cv. Desnitsa) plants, on chloroplast ultrastructure and plant tolerance to low temperatures were studied. The main attention was focused on modifications in the chloroplast structure and their possible relation to potato plant tolerance to oxidative and low-temperature stresses under the influence to their transformation with the Δ12 acyl-lipid desaturase gene from cyanobacterium (desA-licBM3-plants). Morphometric analysis showed that, in comparison with wild-type (WT) plants, in desA-licBM3-plants the number of grana in chloroplasts increased substantially. The total number of thylakoids in transformant chloroplasts was almost twice higher than in WT plants. The number of plastoglobules per chloroplast of transformed plants increased by 25%. A marked increase in the number of grana, total number of thylakoids, and the number of plastoglobules in chloroplasts of desA-licBM3-plants indicates their more intense lipid metabolism, as compared with WT plants, and this resulted in the conservation of some part of lipids in plastoglobules. In addition, the expression of heterological desA gene encoding Δ12 acyl-lipid desaturase positively influenced stabilization of not only structure but also functioning of chloroplast membranes, thus preventing a transfer of electrons from the ETR to oxygen and subsequent ROS generation at hypothermia. This was confirmed by the analysis of the rate of superoxide anion generation in tested genotypes.  相似文献

Liquidambar styraciflua L. seedlings and tissue-cultured plantlets were grown under high, medium, or low (315, 155, or 50 microeinsteins per square meter per second photosynthetically active radiation) quantum flux densities. Net photosynthesis, chlorophyll content, and chloroplast ultrastructure of leaves differentiated from these conditions were investigated. Seedling photosynthetic rates at light saturation were positively related to light pretreatments, being 6.44, 4.73, and 2.75 milligrams CO2 per square decimeter per hour for high, medium, and low light, respectively. Cultured plantlets under all light conditions had appreciably higher photosynthetic rates than noncultured seedlings; corresponding rates were 12.14, 13.55, and 11.36 milligrams CO2 per square decimeter per hour. Chlorophyll in seedlings and plantlets was significantly higher in low light-treated plants. Seedling leaves had chloroplasts with abundant starch regardless of light pretreatment. In high light, starch granules were predominant and associated with disrupted granal structure. Low light seedling chloroplasts had smaller starch grains and well-formed grana. In contrast, tissue culture-differentiated leaves were devoid of starch; grana were well organized in higher quantum flux density treatments, but disorganized at low flux densities.  相似文献

In transgenic (TG) tomato (Lycopersicon esculentum Mill.) overexpressed ω-3 fatty acid desaturase gene (LeFAD7) was identified, which was controlled by the cauliflower mosaic virus 35S promoter and induced increased contents of unsaturated fatty acids in thylakoid membrane. Under chilling stress at low irradiance (4 °C, 100 μmol m−2 s−1) TG plants with higher linolenic acids (18: 3) content maintained a higher O2 evolution rate, oxidizable P700 content, and maximal photochemical efficiency (Fv/Fm) than wild type (WT) plants. Low temperature treatment for 6 h resulted in extensive changes of chloroplast ultrastructure: in WT plants most chloroplasts became circular, the number of amyloids increased, appressed granum stacks were dissolved, grana disappeared, and the number of grana decreased, while only a few grana were found in leaves of TG plants. Hence the overexpression of LeFAD7 could increase the content of 18: 3 in thylakoid membrane, and this increase alleviated the photoinhibition of photosystem (PS) 1 and PS2 under chilling at low irradiance.  相似文献

The effect of dark-chilling and subsequent photoactivation on chloroplast structure and arrangements of chlorophyll–protein complexes in thylakoid membranes was studied in chilling-tolerant (CT) pea and in chilling-sensitive (CS) tomato. Dark-chilling did not influence chlorophyll content and Chl a/b ratio in thylakoids of both species. A decline of Chl a fluorescence intensity and an increase of the ratio of fluorescence intensities of PSI and PSII at 120 K was observed after dark-chilling in thylakoids isolated from tomato, but not from pea leaves. Chilling of pea leaves induced an increase of the relative contribution of LHCII and PSII fluorescence. A substantial decrease of the LHCII/PSII fluorescence accompanied by an increase of that from LHCI/PSI was observed in thylakoids from chilled tomato leaves; both were attenuated by photoactivation. Chlorophyll fluorescence of bright grana discs in chloroplasts from dark-chilled leaves, detected by confocal laser scanning microscopy, was more condensed in pea but significantly dispersed in tomato, compared with control samples. The chloroplast images from transmission-electron microscopy revealed that dark-chilling induced an increase of the degree of grana stacking only in pea chloroplasts. Analyses of O-J-D-I-P fluorescence induction curves in leaves of CS tomato before and after recovery from chilling indicate changes in electron transport rates at acceptor- and donor side of PS II and an increase in antenna size. In CT pea leaves these effects were absent, except for a small but irreversible effect on PSII activity and antenna size. Thus, the differences in chloroplast structure between CS and CT plants, induced by dark-chilling are a consequence of different thylakoid supercomplexes rearrangements.
