ABSTRACT

The annual reproductive cycle comprises steady gametogenic activities that synchronize gonadal maturation and spawning rhythms, which are important for aquatic organisms including marine echinoderms (Echinodermata, Echinoidea). In this study, we report the annual reproductive cycle, gonadal development, and changes in nutritive phagocytes (NPs, which accumulate nutrients in germ cells) in relation to gametogenesis of the Atlantic sea urchin (Arbacia punctulata, an edible echinoid) in the Gulf of Mexico. Monthly changes in gonadal development and maturation were observed morphologically and histologically. We calculated gonadosomatic index (GSI) and compared the stages of gonadal development in order to determine the NPs index, and characteristics of germ cells (eggs and sperm) during the annual reproductive cycle. According to GSI and histological analyses, gametogenic activities were classified into four stages of both sexes: mature (June–August), spent (September–November), recovery (December–March), and growing (April–May). The GSI values in both sexes were high during summer months. In males, testicular lobules were densely packed with sperm from June to August. In females, however, mature eggs first appeared in some ovaries in May, numerically increased from June to July, and decreased in August. During gametogenesis, on the other hand, NPs in both testes and ovaries were depleted from June to August. Collectively, our results suggest that the Atlantic sea urchin spawns during summer months in the Gulf of Mexico. This is the first report, to the best of our knowledge, on gonadal development and changes in NPs during the annual reproductive cycle of any Arbacia species in the Gulf of Mexico.  相似文献

Here we characterize the germinal epithelia of both sexes of Strongylocentrotus droebachiensis, the green sea urchin, throughout its annual gametogenic cycle, using light and electron microscopy and cytochemistry. In both sexes, germinal epithelia include two interacting cellular populations: nutritive phagocytes (NPs) and germ cells. After spring spawning, NPs accumulate nutrients; amitotic oogonia and often mitotic spermatogonia occur in clusters beneath NPs; and subsequent gametogenic stages are residual or absent. During the summer, NP nutrients are mobilized for use in vitellogenesis by residual primary oocytes or to support limited spermatogenesis. In addition, some residual primary oocytes may degenerate and be phagocytized by NPs. Significant nutrient mobilization from NPs and substantial gonial cell mitoses (indicative of new gametogenesis) occur in the fall. In both sexes, all of these changes are facilitated by NPs that form basal incubation chambers near the gonadal wall and within which germ cells are surrounded by nutrients released from the NPs. In females, germ cells at several stages of gametogenesis may be housed in separate chambers in the same NP. Primary oocytes also carry out jelly coat formation, meiosis, and cortical granule translocation within NP incubation chambers. In males, many NPs cooperate to provide large continuous chambers that contain spermatogenic cells at diverse stages. In both sexes these chambers persist throughout the year and isolate gametogenesis from the gonadal lumen. NPs become slender and shorten as their nutrients are depleted. Ova or spermatozoa are stored in the gonadal lumen. Post-spawning, NPs phagocytize differentiated germ cells while simultaneously enclosing intact gonial and residual gametogenic cells in basal chambers near the gonadal wall. In light of our observations, we suggest investigating proteins that may be important in the structural, phagocytic, and nutritive functions of NPs and for which corresponding genes have already been identified in the genome of S. purpuratus, the closely related purple sea urchin.  相似文献

Abstract. Used singly, the fluorochrome tags tetracycline and calcein have yielded important insights into sea urchin biology, especially regarding growth. We present a new method of tagging using sequential fluorochrome markers, as well as a more precise method of quantifying growth. Such polyfluorochromes enable repeated markings that allow measurement of multiple growth points and unique identification of individuals or groups. We marked sea urchins, Strongylocentrotus droebachiensis , with four fluorochromes: alizarin complexone, calcein, calcein blue, and tetracycline. All fluorochromes marked both by injection and immersion. We examined the clarity of the mark produced with low, metabolically scaled doses, and higher doses similar to those that have been previously used. We tested the effect of fluorochromes on survival, growth, jaw size, and gonad size by marking a size range (3.9–44.3 mm in diameter) of urchins with either one or all four fluorochromes. We quantified growth using a nominal diameter, that is a fitted constant, times the cube root of weight, which increased the precision of measurements by a factor of six relative to measured diameter. Growth rate was a decreasing function of diameter except for a growth lag in the smallest urchins. Growth rate data for all sizes were fitted using: gamma distributions; Tanaka functions; and, for larger sizes, straight lines (von Bertalanffy model). All treatments produced clear marks, with higher doses producing more reliably clear marks. Tetracycline marking did not affect growth; other treatments produced only transient slowing of growth in the marking month. Growth rate, survival, gonad production, and jaw weight did not differ between control and treatments during the following 6 months. Thus, polyfluorochromes produce reliable marks that do not significantly affect growth or gonad production.  相似文献

Abstract. Settlement timing and differential settlement for the larval stage of the green sea urchin, Strongylocentrotus droebachiensis , in the southern Gulf of Maine was studied during the summer of 1996. Settlement densities on astroturf panels were highest in June and early July (13 to 37 m −2 d−1), and peaked in mid-June (199 m−2 d−1). Settlement was low to nonexistent from mid-July through August (0 to 2 m−2 d−1). During the peak in settlement, no selection for substrate type was observed. In the remainder of the settlement period, differential settlement occurred, with a preference for substrate covered with live coralline algae. Test diameter of newly settled urchins varied among the substrates, with urchins settling on live coralline algae having the largest test diameter (0.43 ± 0.01 mm). There were no differences in test diameter among the different weeks in which sampling was done. Sustained onshore winds occurred only during peak settlement, suggesting that wind drift currents may concentrate larvae and influence patterns of larval settlement.  相似文献

The content of total and free amino acids (FAA) in green sea urchin (Strongylocentrotus droebachiensis) gonads varied with the season and feeding on an artificial diet. Glycine was the dominant amino acid in each season contributing 12.9-16.6% to the total amino acid (TAA) content, peaking in the spring. In the FAA profile, glycine accounted for 30.3-61.4% in different seasons. A grain-based artificial diet had noticeable effects on the total and FAA compositions of S. droebachiensis. Although, glycine was the dominant amino acid in the TAA profile during early harvesting, tyrosine in gonads became more dominant on week 9 of feeding. Furthermore, glycine was the dominant amino acid in the FAA pool after feeding the artificial diet. The total FAA content in the gonads increased significantly (P<0.05) from 20.6 on week 0 to 180.6 mg/g dry mass tissue on week 3. There were no significant (P<0.05) changes between week 6 and week 9. Deoxyribonucleic acid (DNA) content exceeded that of ribonucleic acid (RNA) in each season, while in cultured urchins, RNA content exceeded that of DNA only on week 6. The RNA/DNA ratio was significantly increased in the summer, whereas this ratio was increased up to week 6 followed by a decrease on week 9 in cultured counterparts.  相似文献

Morphological changes in the mitotic cytoskeleton (MC) that occurred through the course of the first cleavage of Strongylocentrotus droebachiensis, cultured at 8°C or at 0°C, temperatures within the natural range for this species, have been investigated. Electron microscopy of MCs isolated from zygotes has revealed that they consisted largely of microtubules (mts). Thus, the morphology of these MCs is derived from the arrangement of the mts which form them. During anaphase, astral rays elongated while kinetochore fibers shortened. Asters enlarged during anaphase as a result of two events: astral ray lengthening and centrosphere enlargement. At the end of anaphase, asters of 8°C MCs filled the entire cell volume. The pattern of changes that occurred in 8°C mitotic apparatuses (MAs) also occurred in 0°C MCs. The observation of asters in 0°C MCs is contradictory to that of Stephens ('72b), who reported that 0°C MCs in this species were anastral. However, in 0°C metaphase MCs, the astral rays and spindle fibers were not as long as those in 8°C MCs. Also in 0°C MCs, the centrosphere was largely filled with dense material, whereas the centrosphere in 8°C MCs was larger and contained little dense material. Asters of 0°C MCs did not attain a large enough size to fill the egg volume completely, as did asters of 8°C MCs.  相似文献

Recruitment of Strongylocentrotus droebachiensis (Muller) was monitored for four successive years following a mass mortality of this species on a subtidal boulder bottom in St. Margaret's Bay, Nova Scotia. Recently recruited juveniles (1–2 mm diameter) were observed in the fall (September–November) between (1982) and (1984), but not in (1985). Juveniles grew to 6–8 mm diameter at 1-yr post-settlement and 19 mm diameter ((1983) cohort) at 2-yr post-settlement. Individuals > 18 mm diameter had macroscopic gonads in the spring of their third year and spawning probably occurred at 2.75-yr post-settlement. The growth rate of juveniles (3–6 mm initial diameter) maintained in cages with coralline-covered rocks was similar to that of the natural population. Growth and gonad index increased significantly when kelp (Laminaria longicruris (Pylaie)) was added to cages, suggesting that juveniles were food limited in St. Margaret's Bay. Despite an abundance of kelp and other fleshy algae, they remained cryptic under rocks and were not observed grazing on attached plants. The effects of temperature and food supply on juvenile growth and survival were examined in laboratory experiments using factorial designs. Recently-metamorphosed juveniles (from laboratory cultures), maintained at three temperature regimes (5°C, ambient, 16 °C) and two feeding regimes (no added food, coralline algae added), had similar growth rates (over 9 mth) among treatment combinations. However, growth was asymptotic at 16 °C and ambient temperatures, suggesting food limitation. Survival of these juveniles was higher in treatments with coralline algae, but survival was not affected by temperature. Field-collected juveniles (3–6 mm diameter), maintained at three temperature regimes (5°C, ambient, 16 °C) and three feeding regimes (no added food, coralline algae added, and kelp and coralline algae added), grew only in treatments with kelp added as food. In these treatments, growth rates increased with temperature. However, the growth curve at variable ambient temperature regimes was linear, suggesting seasonal acclimation. Survival of the field-collected juveniles was highest in treatments at 5 °C and/or with added kelp.  相似文献

Isolated cortices from unfertilized sea urchin eggs sequester calcium in an ATP-dependent manner when incubated in a medium containing free calcium levels characteristic of the resting cell (approximately 0.1 microM). This ATP-dependent calcium uptake activity was measured in the presence of 5 mM Na azide to prevent mitochondrial accumulation, was increased by oxalate, and was blocked by 150 microM quercetin and 50 microM vanadate (known inhibitors of calcium uptake into the sarcoplasmic reticulum). Cortical regions preloaded with 45Ca in the presence of ATP were shown to dramatically increase their rate of calcium efflux upon the addition of (a) the calcium ionophore A23187 (10 microM), (b) trifluoperazine (200 microM), (c) concentrations of free calcium that activated cortical granule exocytosis, and (d) the calcium mobilizing agent inositol trisphosphate. This pool of calcium is most likely sequestered in the portion of the egg's endoplasmic reticulum that remains associated with the cortical region during its isolation. We have developed a method for obtaining a high yield of purified microsomal vesicles from whole eggs. This preparation also demonstrates ATP-dependent calcium sequestering activity which increases in the presence of oxalate and has similar sensitivities to calcium transport inhibitors; however, the isolated microsomal vesicles did not show any detectable release of calcium when exposed to inositol trisphosphate.  相似文献

Summary

Studies of juvenile recruitment of the green sea urchin Strongylocentrotus droebachiensis in the Gulf of Maine were conducted during the summer of 1995. These experiments confirmed 12 years of previous observations that settlement only occurs during the months of June and July. Settlement panels were placed at a series of sites along the Maine and New Hampshire coastline to compare recruitment in the northeastern and southwestern regions of the Gulf of Maine. The densities of urchins recruiting in Casco Bay and at the Isles of Shoals were two orders of magnitude higher than those from Eastport and Winter Harbor. There was a discontinuity in settlement densities at Penobscot Bay. Experiments conducted at the Isles of Shoals showed a positive relationship between water motion and larval supply, but neither parameter correlated with recruitment density over eight stations. Contrary to previous results, recuitment was greater within natural, as well as, artificial kelp beds compared to urchin barren areas and control panels outside the experimental kelp beds. The impact of changing community structure due to urchin harvesting was discussed as an factor influencing differences in juvenile urchin recruitment.  相似文献

The respiration of mitochondria from gonads of Strongylocentrotus droebachiensis has been studied in the presence of 2-oxoglutarate, glutamate and succinate. The rate of oxygen consumption increased with temperature between 4 and 20 degrees C. Removal of gametes from ripe gonads was necessary to obtain good respiring mitochondria. Equilibration of respiration media with various petroleum fractions reduced the rate of oxygen consumption and the coupling of O2 consumption to ADP utilization. Lighter fractions (leaded gasolines, unleaded gasoline and related fractions) were more deleterious than medium fractions (kerosene and fuel oil). Heavy fraction (bunker C, vacuum bottoms and crude oil) were least deleterious. Tetraethyl lead contributed to the toxicity of the leaded fractions.  相似文献

The adult body plan of Strongylocentrotus purpuratus is established within the imaginal rudiment during the larval stages. To facilitate the study of these stages, we have defined a larval staging scheme, which consists of seven stages: Stage I, four-arm stage; Stage II, eight-arm stage; Stage III, vestibular invagination stage; Stage IV, rudiment initiation stage; Stage V, pentagonal disc stage; Stage VI, advanced rudiment stage; and Stage VI, tube-foot protrusion stage. Each stage is characterized by significant morphological features observed for the first time at that stage. This scheme is intended as a guide for determining the degree of larval development, and for identifying larval and adult structures. Larval anatomy was visualized using light and confocal microscopy as required on living material, whole mount fixed specimens, and serial sections. Antibody staining to localize specific gene products was also used. Detailed analysis of these data has furthered our understanding of the morphogenesis of the rudiment, and has suggested provocative questions regarding the molecular basis for these events. We intend this work to be of use to investigators studying gene expression and morphogenesis in postembryonic larvae.  相似文献

Reproductive cycle of seasonally breeding fish is synchronized with changes of photoperiod and temperature in environment. We hypothesize that arginine vasotocin (AVT) and isotocin (IT) are involved in timing and synchronization of seasonal reproductive activity in the round goby (Neogobius melanostomus). To verify this hypothesis, we examined the annual profiles of brain AVT and IT in round goby males and females in relation to their reproductive cycle. Wild round gobies were exposed to annual environmental changes in their natural habitats from where they were sampled monthly over a year. AVT and IT were measured using HPLC with fluorescence detection preceded by solid-phase extraction. This study shows seasonal variations in brain AVT and IT levels. Profiles of changes were similar in males and females: the peak of AVT was observed before spawning in March-April, whereas that of IT during spawning in May–June. Furthermore, the lowest AVT level was noted out of breeding season from November to January, while the level of IT decreased immediately at the end of the spawning. The results show that high AVT levels correlate with pre-spawning period whereas the highest IT levels correspond to spawning. A significant decline in AVT and IT in non-spawning season coincided with the quiescent phase of gametogenesis in both sexes.  相似文献

Biomineralization, the biologically controlled formation of mineral deposits, is of widespread importance in biology, medicine, and engineering. Mineralized structures are found in most metazoan phyla and often have supportive, protective, or feeding functions. Among deuterostomes, only echinoderms and vertebrates produce extensive biomineralized structures. Although skeletons appeared independently in these two groups, ancestors of the vertebrates and echinoderms may have utilized similar components of a shared genetic "toolkit" to carry out biomineralization. The present study had two goals. First, we sought to expand our understanding of the proteins involved in biomineralization in the sea urchin, a powerful model system for analyzing the basic cellular and molecular mechanisms that underlie this process. Second, we sought to shed light on the possible evolutionary relationships between biomineralization in echinoderms and vertebrates. We used several computational methods to survey the genome of the purple sea urchin Strongylocentrotus purpuratus for gene products involved in biomineralization. Our analysis has greatly expanded the collection of biomineralization-related proteins. We have found that these proteins are often members of small families encoded by genes that are clustered in the genome. Most of the proteins are sea urchin-specific; that is, they have no apparent homologues in other invertebrate deuterostomes or vertebrates. Similarly, many of the vertebrate proteins that mediate mineral deposition do not have counterparts in the S. purpuratus genome. Our findings therefore reveal substantial differences in the primary sequences of proteins that mediate biomineral formation in echinoderms and vertebrates, possibly reflecting loose constraints on the primary structures of the proteins involved. On the other hand, certain cellular and molecular processes associated with earlier events in skeletogenesis appear similar in echinoderms and vertebrates, leaving open the possibility of deeper evolutionary relationships.  相似文献

Apoptosis provides metazoans remarkable developmental flexibility by (1) eliminating damaged undifferentiated cells early in development and then (2) sculpting, patterning, and restructuring tissues during successive stages thereafter. We show here that apoptotic programmed cell death is infrequent and not obligatory during early embryogenesis of the purple sea urchin, Strongylocentrotus purpuratus. During the first 30 h of urchin development, fewer than 20% of embryos exhibit any cell death. Cell death during the cleavage stages consists of necrotic or pathological cell death, while cell death during the blastula and gastrula stages is random and predominantly caspase-mediated apoptosis. Apoptosis remains infrequent during the late blastula stage followed by a gradual increase in frequency during gastrulation. Even after prolonged exposure during the cleavage period to chemical stress, apoptosis occurs in less than 50% of embryos and always around the pre-hatching stage. Embryonic suppression of apoptosis through caspase inhibition leads to functionally normal larvae that can survive to metamorphosis, but in the presence of inducers of apoptosis, caspase inhibition leads to deformed larvae and reduced survival. Remarkably, however, pharmacological induction of apoptosis, while reducing overall survival, also significantly accelerates development of the survivors such that metamorphosis occurs up to a week before controls.  相似文献

Major yolk protein (MYP), the predominant component of yolk granules in sea urchin eggs, is also contained in the coelomic fluid and nutritive phagocytes of the gonad in both sexes. MYP is stored in ovarian and testicular nutritive phagocytes prior to gametogenesis and is used during gametogenesis as material for synthesizing proteins and other components necessary for eggs and sperm. To reveal the expression profile and the main production site of MYP, we analyzed MYP mRNA expression in immature and maturing Pseudocentrotus depressus. Real‐time reverse‐transcribed polymerase chain reaction analysis showed that MYP mRNA was expressed predominantly in the digestive tract (stomach, intestine and rectum) and the gonad of both sexes. The total amounts of MYP mRNA in the whole digestive tract and in the whole gonad were at similar levels in both immature and maturing sea urchins. MYP mRNA was also detected in white morula cells and vibratile cells separated from the coelomic fluid by density gradient centrifugation, but the expression levels in these cells were very low compared with those in the digestive tract and the gonad. Using in situ hybridization analysis, MYP mRNA was detected in the inner epithelium of the digestive tract and in nutritive phagocytes of the ovary and testis, but was not detected in the germ cells. We conclude that the adult sea urchin has two predominant production sites for MYP regardless of sex and reproductive stage: the inner epithelium of the digestive tract and the nutritive phagocytes of the gonad.   • 4.4. Radioactivity in proteins and lipids increased with increasing incubation time. No significant differences were found in the distribution patterns of radiolabel during the reproductive cycle.
  • 5.5. The data obtained are discussed in terms of current knowledge on the translocation of nutrients in echinoderms.
